O objetivo deste estudo foi estimar a prevalência da brucelose bovina nos 22 municípios que compõem a região denominada Estrato 1 do Estado de Mato Grosso do Sul, e identificar os fatores de risco associados à infecção. A região amostrada constitui uma área de 70.214,1 km², que representa 19,7% do Estado. O rebanho de região estudada é de, aproximadamente, 5,7 milhões de cabeças, correspondente a 23% do efetivo de 24,9 milhões de bovinos de Mato Grosso do Sul. Nas 210 propriedades amostradas, no período de dezembro de 2003 a março de 2004, foram colhidas 2.376 amostras de sangue de fêmeas com idade igual ou superior a 24 meses, submetidas a testes diagnósticos em série. A triagem, realizada por meio do teste do antígeno acidificado tamponado, foi seguida pelo teste confirmatório 2-mercaptoetanol. Na mesma ocasião da colheita das amostras, foi preenchido um questionário com informações de identificação, tipo de criação e práticas de manejo. Em animais, a prevalência real foi estimada em 5,6%, e em rebanhos, 37,3%. As variáveis que apresentaram associação, por meio da análise univariada odds ratio (OR) e intervalo de confiança (IC) de 95%, com a soropositividade à brucelose foram: o tipo de exploração corte (OR = 2,82, IC 95% = 1,49-5,34), a raça Zebu (OR = 2,62, IC 95% = 1,40-4,88) e o aborto (OR= 1,83, IC 95% = 1,01-3,33). Os resultados demonstram que, além da brucelose ser prevalente no estrato estudado em Mato Grosso do Sul, o controle da doença pode consistir na adoção de programa com especial atenção à exploração do tipo corte, à raça Zebu e à presença do aborto.

Foram identificados fatores de risco associados à leptospirose em fêmeas bovinas em idade reprodutiva no Estado da Bahia. Foram amostradas aleatoriamente 10.823 fêmeas bovinas com idade igual ou superior a 24 meses procedentes de 1.414 propriedades. Para o diagnóstico sorológico da infecção por Leptospira spp., foi utilizada a Soroaglutinação Microscópica (SAM) utilizando 24 sorovares como antígenos. Um rebanho foi considerado foco quando apresentou pelo menos um animal soropositivo. Das 1.414 propriedades investigadas, 1.076 (77,9%; IC 95% = 75,7-80,0%) apresentaram pelo menos um animal reagente na SAM para qualquer sorovar. O sorovar Hardjo (Hardjoprajitno) foi o mais prevalente, com 34,49% (IC 95% = 31,97-37,14%) das propriedades positivas. Presença de mais de 28 fêmeas bovinas em idade reprodutiva no rebanho (OR=2,11; p<0,001), presença de cervídeos (OR=2,02; p=0,010), compra de animais (OR=1,57; p<0,001), abate de animais na própria fazenda (OR=1,58; p=0,030) e utilização de partos compartilhados (OR=1,63; p<0,001) foram identificados como fatores de risco para leptospirose por qualquer sorovar. Os fatores de risco para leptospirose pelo sorovar Hardjo (Hardjoprajitno) foram a presença de suínos (OR=1,28; p=0, 040) e a compra de animais (OR=1,48; p<0,001).

The aim of this study was evaluate the risk factors for Mycobacterium avium subsp. paratuberculosis (Map) seroprevalence in sheep in the North of Portugal. The effects on seroprevalence of several variables such as individual characteristics, management practices, farm characteristics, animal health, and available veterinary services were evaluated. This information was then used in a multivariable logistic regression model in order to identify risk factors for Map seropositivity. Univariable analysis was used to screen the variables used in the logistic regression model. Variables that showed p values of <0.15 were retained for the multivariable analysis. Fifteen variables were associated with paratuberculosis in univariable analysis. The multivariable logistic regression model identified a number of variables as risk factors for seropositivity like sheep pure local and/or a cross of a local breed (OR=2.02), herd size with 31-60 head (OR=2.14), culling during the Spring-Summer season (OR=1.69) and the use of an anti-parasitic treatment such as Ivermectin as the only anti-parasitic medication (OR=5.60). Potential risk factors identified in this study support current recommendations for the control of paratuberculosis.

Objetivou-se com este estudo calcular a prevalência e identificar os fatores de risco associados à infecção por Toxoplasma gondii em suínos criados em granjas tecnificadas no Estado de Alagoas, Brasil. Para compor a amostra do estudo de prevalência foram utilizados 342 suínos, sendo 312 matrizes e 30 varrões, oriundos de sete granjas de ciclo completo e distribuídas em cinco municípios do Estado de Alagoas. O exame sorológico para a pesquisa de anticorpos anti-Toxoplasma gondii foi realizado através da técnica de Imunofluorescência Indireta (RIFI), utilizando-se anticorpos anti-IgG-suíno conjugado ao isotiocianato de fluoresceína. A análise dos fatores de risco foi realizada por meio da aplicação de questionários constituídos por perguntas objetivas referentes ao criador, às características gerais da propriedade, ao manejo produtivo, reprodutivo e sanitário. Determinou-se uma prevalência de 26,9% (92/342) de suínos soropositivos. O fator associado à infecção foi a introdução de reprodutores nas granjas nos últimos cinco anos (p=0,014; OR=1,83; IC=1,13-2,96). Concluiu-se que a infecção por Toxoplasma gondii encontra-se disseminada em suínos criados em granjas tecnificadas no Estado de Alagoas, Brasil. Recomenda-se o controle da população de gatos, a realização de práticas de limpeza e higienização das instalações e realização de testes laboratoriais para diagnóstico da infecção por T. gondii nos animais a serem introduzidos no plantel como medidas de redução dos índices de infecção na região estudada.

The aim of this study was to determine the prevalence of anti-Leptospira spp. antibodies and the risk factors for Leptospira spp. infection in breeding cattle herds in the south central region of Paraná state. It was based on the statistic delineation/serological samples and information regarding the selected farms employed in the study of bovine brucellosis for Paraná state in the context of National Program for Control and Eradication of Brucellosis and Tuberculosis. A total of 1.880 females aged >24 months from 274 non vaccinated herds were studied. Serum samples were tested for antibodies against Leptospira spp. using microscopic agglutination test (MAT) with 22 Leptospira serovars. The epidemiological questionnaire was applied on all the selected farms and aimed to obtain epidemiological data. Hundred eighty one of 274 herds were positive for Leptospira spp./presenting prevalence of positive herds of 66.06% (IC95%=60.12-71,65%). Presence of >43 cattle (OR=3.120; IC=1.418-6.867)/animal purchase (OR=2.010; IC=1.154-3.500)/rent of pastures (OR=2.925; IC=1.060-8.068) and presence of maternity paddock (OR=1.981; IC=1,068-3,676) were identified as risk factors for leptospirosis due to any serovar in the multivariate logistic regression. Risk factors for leptospirosis due to serovar Hardjo were presence of >43 cattle (OR=3.622; IC=1.512-8,677)/animal purchase (OR=3.143; IC=1.557-6.342)/rent of pastures (OR=4.070; IC=1.370-12.087) and presence of horses (OR=2.981; IC=1.321-6.726). These results indicate that Leptospira spp. infection is widespread in the south central region of Paraná state and that factors related to the herd characteristic and management are associated with the infection.

Objetivou-se com este estudo analisar os fatores de risco associados à mastite bovina na microrregião de Garanhuns, Pernambuco. Para isso, foram submetidas ao exame microbiológico 1260 amostras de leite e em cada propriedade foi aplicado um questionário contendo informações relacionadas às características raciais dos animais, dados gerais sobre as propriedades, manejo do rebanho e higiênico-sanitário durante a ordenha. Para a análise dos fatores de risco foi realizada uma regressão logística considerando como variável dependente o exame microbiológico. Ao exame microbiológico observou-se 477 (37,9%) amostras positivas e 783 (62,1%) negativas. Os fatores de risco associados à mastite foram: sistema de criação semi-intensivo (OR=2,3; p<0,0001), alimentação dos animais durante a ordenha (OR=1,3; p=0,0101), rodízio de antimicrobianos (OR=1,3; p=0,0077) e o uso da terapia da vaca seca (OR=1,5; p=0,0013). Os resultados obtidos demonstram que os principais fatores de risco associados à mastite estão relacionados ao manejo higiênico-sanitário ao qual o rebanho é submetido. O conhecimento desses fatores fornece subsídios para a elaboração de medidas de prevenção e controle para a enfermidade em questão, melhorando a produtividade dos animais e, consequentemente, a lucratividade do sistema de produção.

Com o objetivo de determinar os aspectos epidemiológicos que envolvem a doença de Chagas (DC) canina e identificar os principais fatores de risco da enfermidade no semiárido paraibano, foi conduzido um estudo na zona rural de Patos, onde a área foi dividida em três estratos amostrais (Norte, Sul e Oeste) e, em cada estrato foram amostradas aleatoriamente 294 casas, e dessas todos os cães domiciliados representaram as unidades elementares do estudo. Em cada unidade domiciliar foi aplicado um questionário epidemiológico para se obter informações sobre indicadores que favorecem a disseminação da doença no segmento peridomiciliar. O diagnóstico sorológico para DC em cães foi baseado em três métodos (RIFI, ELISA e HAI), sendo consideradas positivas aquelas amostras que apresentassem pelo menos dois testes reagentes e ausência de reatividade cruzada. Para zona rural do município, a prevalência de cães sororreagentes para T. cruzi por estrato amostral foram: Norte 6,05%, Sul 3,59% e Oeste 2,97%, correspondendo a uma prevalência em sua totalidade de 4,08%. Os fatores de riscos (odds ratio, OR) evidenciados em análise multifatorial foram: tipo de parede (OR=2,59 [1,24-5,4]), presença de armazém (OR=1,89 [1,31-3,0]), presença de galinheiros (OR=8,31 [1,29-61,7]), contato com animais (OR=9,11 [1,12-73,9]), contato com aves (OR=9,7 [1,81-52,83]), triatomíneos capturados (OR=16,58 [3,43-80,23]) e antropismo (OR=4,35 [1,36-14,0]. Diante dos resultados foi possível se obter informações inerentes à situação epidemiológica da Doença de Chagas ressaltando características biogeográficas da zona rural do semiárido paraibano, elevando a espécie canina e os fatores de risco evidenciados, em destaque ao contato com aves e ecótopos artificiais, operacionalizando indicadores a serem assistidos e considerados na cadeia de transmissão da doença na região.

Abstract: The aim of the present study was to assess the occurrence of Mycoplasma gallisepticum (MG) infection and risk factors of this disease in three hundred serum samples from on 23 familiar agricultural properties in the semiarid region of the state of Pernambuco, Brazil. ELISA was used to study antibodies anti-Mycoplasma gallisepticum. The univariate analysis (chi-squared test or Fischer's exact test) followed by multivariate analysis (logistic regression) were used to assess the risk factors with two variables: management and sanity of the poultry. It was detected a frequence of 53.33% (157/300) of the birds were positive for MG, with 100% foci. The risk factors confirmed by multivariate analysis, in the present study, were the presence of other poultry species on the property, including Numida meleagris (OR=2.22; p=0.005), parrots (OR=1.72; p=0.027), and of passerines (OR=1.88; p=0.007). These results showed that Mycoplasma gallisepticum infection is endemic among backyard poultry in the semiarid region of the state of Pernambuco. These birds could be a source of infection for other wild or domestic poultry. . This is the first report of the occurrence of avian mycoplasmosis in backyard poultry in the state of Pernambuco in northeastern Brazil. The risk factors identified should serve as a parameter for the health authorities to seek solutions related to controlling the disease.

Os redutores de crescimento têm sido utilizados com sucesso em trigo, evitando o acamamento, mas a recomendação de uso não diferencia cultivares quanto ao porte. Visando determinar a dose e a época de aplicação mais adequadas de trinexapac-ethyl (Moddus), em cultivares de trigo, realizaram-se experimentos na Fazenda Escola da Universidade Estadual de Ponta Grossa, no ano de 2005. O delineamento experimental foi de blocos ao acaso, em esquema fatorial 2 x 2 x 6, com três repetições para cada cultivar de trigo. Os tratamentos constaram de duas doses de nitrogênio (50 e 240 kg ha-1), duas épocas de aplicação de trinexapac-ethyl (entre o 1º e o 2º nó e entre o 2º e o 3º nó perceptível) e de seis doses de trinexapac-ethyl (0, 31,2, 62,5, 93,7, 125,0 e 156,2 g ha-1). Foram utilizados oito cultivares de trigo com diferentes respostas ao acamamento: OR-1, CD-104 e CD-105 (resistentes), Alcover, Ônix e Vanguarda (moderadamente resistentes), Supera (moderadamente suscetível) e CEP24 (suscetível). Avaliaram-se características agronômicas, componentes da produção e produtividade. O clima ameno não foi favorável à ocorrência de acamamento, que foi observado somente para as menores doses de trinexapac-ethyl no cultivar CEP-24. A dose de nitrogênio afetou características da planta e a produtividade de alguns cultivares. O estádio de aplicação de trinexapac-ethyl não afetou de forma substancial a produtividade, e sim a altura das plantas, que foi menor quanto mais tardia a aplicação. Em geral, a equação de ajuste da produtividade em relação às doses de trinexapac-ethyl foi quadrática, ocorrendo aumento da produção com o aumento da dose do redutor, até um limite, variável de acordo com o cultivar.

(Response of Dalbergia miscolobium Benth. seedlings, a cerrado tree species, to a mineral nutrient supply). The soil under the Brazilian cerrado vegetation is very poor in nutrients. The aim of this study was to investigate the effect of the addition of a nutrient solution in the early growth of D. miscolobium, a typical cerrado tree species, cultivated in cerrado soil. Seeds were germinated in Petri dishes and seven-day old seedlings were transfered to pots containing cerrado soil. These were supplied once a week with a normal, 1/2 or 1/10 strength nutrient solution. Four growth analysis were done over a period of 145 days from seed imbibition, in terms of dry mass, leaf area and carbohydrate and nitrogen contents. Other derived parameters were also analysed. The addition of a nutrient solution promoted early growth with a preferential separation of dry matter towards the tops; in the control seedlings (no nutrient addition) more dry matter accumulated in the root system. This suggests a preferential investment in root growth in poor soils such as the cerrado ones. Reliable evidence of this is given by the level of carbohydrate and protein which was higher in the roots than in the tops of plants growing in cerrado soil without the addition of nutrients.

An efficient micropropagation protocol was developed for the medicinal plant Phyllanthus stipulatus (Euphorbiaceae) using nodal segments for axillary shoot proliferation. Maximum multiplication rates (8-9 shoots per explant) was achieved on MS media supplemented with either 2.5-5.0 muM IBA. The best basal media for axillary shoot proliferation when 0.62 muM BA was supplemented were MS, MS/2 and AR (4-5 shoots per explant). Rooting was achieved with 100% of the microshoots on MS medium without growth regulators. Regenerated plants were successfully acclimatized and about 88% of plantlets survived under ex vitro conditions. Flowering was observed in 81% of the ex vitro grown plantlets after 12 weeks of acclimatization. High frequency callus initiation and growth was achieved when nodal segment explants were inoculated either in the vertical position, in the light on MS medium supplemented with 5.0 muM NAA or horizontally oriented, in the dark on MS supplemented with 5.0 muM NAA or 1.25-5.0 muM BA or 2iP. Root cultures were successfully established on MS medium containing 1.1 muM NAA. The optimized micropropagation, callus and root culture protocols offer the possibility to use cell/organ culture techniques for vegetative propagation and secondary metabolism studies.

Increasing levels of atmospheric ammonia from anthropogenic sources have become a serious problem for natural vegetation. Short-term effects of different ammoniacal sources on the N metabolism of Tillandsia pohliana, an atmospheric bromeliad, were investigated. One-year-old, aseptically grown plants were transferred to a modified Knudson medium lacking N for three weeks. Plants were subsequently transferred to Knudson media supplemented with 0.5, 1.0, or 1.5 mM of N in the forms of NH3 or NH4+ as the sole N source. The activities of glutamine synthetase (GS) and glutamate dehydrogenase (GDH-NADH) were determined after 40 h. The GS activity was stimulated significantly by increasing the levels of the gaseous form. The GDH-NADH activity increased significantly under increasing N concentrations with NH3, while no significant differences were observed with NH4+ as a N source. These results may reflect a faster NH3 absorption by T. pohliana compared to NH4+ uptake. The increased activity of GDH-NADH in NH3 treatment may play a role in protecting the cells from the toxic effects of increased endogenous level of free ammonium. A raise in the concentration of N, especially in the form of NH3, greatly increased the content of free amino acids and soluble proteins. A possible utilisation of T. pohliana to evaluate the changes of atmospheric gaseous ammonia is proposed.

Ionic liquids, ILs, have recently been studied with accelerating interest to be used for a deconstruction/fractionation, dissolution or pretreatment processing method of lignocellulosic biomass. ILs are usually utilized combined with heat. Regarding lignocellulosic recalcitrance toward fractionation and IL utilization, most of the studies concern IL utilization in the biomass fermentation process prior to the enzymatic hydrolysis step. It has been demonstrated that IL-pretreatment gives more efficient hydrolysis of the biomass polysaccharides than enzymatic hydrolysis alone. Both cellulose (especially cellulose) and lignin are very resistant towards fractionation and even dissolution methods. As an example, it can be mentioned that softwood, hardwood and grass-type plant species have different types of lignin structures leading to the fact that softwood lignin (guaiacyl lignin dominates) is the most difficult to solubilize or chemically disrupt. In addition to the known conventional biomass processing methods, several ILs have also been found to efficiently dissolve either cellulose and/or wood samples – different ILs are suitable for different purposes. An IL treatment of wood usually results in non-fibrous pulp, where lignin is not efficiently separated and wood components are selectively precipitated, as cellulose is not soluble or degradable in ionic liquids under mild conditions. Nevertheless, new ILs capable of rather good fractionation performance have recently emerged. The capability of the IL to dissolve or deconstruct wood or cellulose depends on several factors, (e.g. sample origin, the particle size of the biomass, mechanical treatments as pulverization, initial biomassto-IL ratio, water content of the biomass, possible impurities of IL, reaction conditions, temperature etc). The aim of this study was to obtain (fermentable) saccharides and other valuable chemicals from wood by a combined heat and IL-treatment. Thermal treatments alone contribute to the degradation of polysaccharides (e.g. 150 °C alone is said to cause the degradation of polysaccharides), thus temperatures below that should be used, if the research interest lies on the IL effectiveness. On the other hand, the efficiency of the IL-treatment can also be enhanced to combine other treatment methods, (e.g. microwave heating). The samples of spruce, pine and birch sawdust were treated with either 1-Ethyl-3-methylimidazolium chloride, Emim Cl, or 1-Ethyl-3-methylimidazolium acetate, Emim Ac, (or with ionized water for comparison) at various temperatures (where focus was between 80 and 120 °C). The samples were withdrawn at fixed time intervals (the main interest treatment time area lied between 0 and 100 hours). Double experiments were executed. The selected mono- and disaccharides, as well as their known degradation products, 5-hydroxymethylfurfural, 5-HMF, and furfural were analyzed with capillary electrophoresis, CE, and high-performance liquid chromatography, HPLC. Initially, even GC and GC-MS were utilized. Galactose, glucose, mannose and xylose were the main monosaccharides that were present in the wood samples exposed to ILs at elevated temperatures; in addition, furfural and 5-HMF were detected; moreover, the quantitative amount of the two latter ones were naturally increasing in line with the heating time or the IL:wood ratio.

In order to examine the relationship between anxiety and reinforcing effects of alcohol, drug-naive male Wistar rats weighing 250-300 g were classified as "anxious" and "non-anxious" in the elevated plus-maze test. A conditioned place preference test was then used to investigate the reinforcing effects of ethanol (EtOH) on these animals. On 2 alternate days, groups of "anxious", "non-anxious" and "normal" rats received intraperitoneal (ip) injections of EtOH (0.5, 1.0 or 1.5 g/kg) immediately before a 15-min confinement to the white compartment. On the 2 intervening days the same rats received ip injections of saline before confinement to the opposite compartment. On day 5, a 15-min free-choice test was carried out with no injections. Rats classified as "anxious" showed a significant, though not dose-dependent preference for all doses of ethanol compared to saline-treated animals. These data demonstrate that rats regarded as "anxious" are more sensitive to the reinforcing effects of EtOH than "non-anxious" and "normal" Wistar rats and emphasize the relevance of the basal levels of anxiety of rats when trying to detect the reinforcing effects of EtOH.

Brain ischemia followed by reperfusion causes neuronal death related to oxidative damage. Furthermore, it has been reported that subjects suffering from ischemic cerebrovascular disorders exhibit changes in circulating platelet aggregation, a characteristic that might be important for their clinical outcome. In the present investigation we studied tert-butyl hydroperoxide-initiated plasma chemiluminescence and thiol content as measures of peripheral oxidative damage in naive and preconditioned rats submitted to forebrain ischemia produced by the 4-vessel occlusion method. Rats were submitted to 2 or 10 min of global transient forebrain ischemia followed by 60 min or 1, 2, 5, 10 or 30 days of reperfusion. Preconditioned rats were submitted to a 10-min ischemic episode 1 day after a 2-min ischemic event (2 + 10 min), followed by 60 min or 1 or 2 days of reperfusion. It has been demonstrated that such preconditioning protects against neuronal death in rats and gerbils submitted to a lethal (10 min) ischemic episode. The results show that both 2 and 10 min of ischemia cause an increase of plasma chemiluminescence when compared to control and sham rats. In the 2-min ischemic group, the effect was not present after reperfusion. In the 10-min ischemic group, the increase was present up to 1 day after recirculation and values returned to control levels after 2 days. However, rats preconditioned to ischemia (2 + 10 min) and reperfusion showed no differences in plasma chemiluminescence when compared to controls. We also analyzed plasma thiol content since it has been described that sulfhydryl (SH) groups significantly contribute to the antioxidant capacity of plasma. There was a significant decrease of plasma thiol content after 2, 10 and 2 + 10 min of ischemia followed by reperfusion when compared to controls. We conclude that ischemia may cause, along with brain oxidative damage and cell death, a peripheral oxidative damage that is reduced by the preconditioning phenomenon.
