BACKGROUND: Intervention studies have shown an increased mortality in patients who received beta-carotene. However, the mechanisms involved in this phenomenon are still unknown. OBJECTIVE: Evaluate the influence of beta-carotene on oxidative stress and the expression of connexin 43 in rat hearts. METHODS: Wistar rats, weighing approximately 100 g, were allocated in two groups: Control Group (n=30), that received the diet routinely used in our laboratory, and Beta-Carotene Group (n = 28), which received beta-carotene (in crystal form, added and mixed to the diet) at a dose of 500 mg of beta-carotene/kg of diet. The animals received the treatment until they reached 200-250g, when they were sacrificed. Samples of blood, liver and heart were collected to perform Western blotting and immunohistochemistry for connexin 43; morphometric studies, dosages of beta-carotene by high-performance liquid chromatography as well as reduced glutathione, oxidized glutathione and lipids hydroperoxides were performed by biochemical analysis. RESULTS: Beta-carotene was detected only in the liver of Beta-Carotene Group animals (288 ± 94.7 µg/kg). Levels of reduced/oxidized glutathione were higher in the liver and heart of Beta-Carotene Group animals (liver - Control Group: 42.60 ± 1.62; liver - Beta-Carotene Group: 57.40 ± 5.90; p = 0.04; heart: - Control Group: 117.40 ± 1.01; heart - Beta-Carotene Group: 121.81 ± 1.32 nmol/mg protein; p = 0.03). The content of total connexin 43 was larger in Beta-Carotene Group. CONCLUSION: Beta-carotene demonstrated a positive effect, characterized by the increase of intercellular communication and improvement of anti-oxidizing defense system. In this model, mechanism does not explain the increased mortality rate observed with the beta-carotene supplementation in clinical studies. (Arq Bras Cardiol. 2013; [online].ahead print, PP.0-0)

We extend the Miles mechanism of wind-wave generation to finite depth. A beta-Miles linear growth rate depending on the depth and wind velocity is derived and allows the study of linear growth rates of surface waves from weak to moderate winds in finite depth h. The evolution of beta is plotted, for several values of the dispersion parameter kh with k the wave number. For constant depths we find that no matter what the values of wind velocities are, at small enough wave age the beta-Miles linear growth rates are in the known deep-water limit. However winds of moderate intensities prevent the waves from growing beyond a critical wave age, which is also constrained by the water depth and is less than the wave age limit of deep water. Depending on wave age and wind velocity, the Jeffreys and Miles mechanisms are compared to determine which of them dominates. A wind-forced nonlinear Schrodinger equation is derived and the Akhmediev, Peregrine and Kuznetsov-Ma breather solutions for weak wind inputs in finite depth h are obtained.

Background: Autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy (APECED) syndrome is a complex immunologic disease caused by mutation of the autoimmune regulator (AIRE) gene. Autoimmunity in patients with APECED syndrome has been shown to result from deficiency of AIRE function in transcriptional regulation of thymic peripheral tissue antigens, which leads to defective T-cell negative selection. Candidal susceptibility in patients with APECED syndrome is thought to result from aberrant adaptive immunity. Objective: To determine whether AIRE could function in anticandidal innate immune signaling, we investigated an extrathymic role for AIRE in the immune recognition of beta-glucan through the Dectin-1 pathway, which is required for defense against Candida species. Methods: Innate immune signaling through the Dectin-1 pathway was assessed in both PBMCs from patients with APECED syndrome and a monocytic cell line. Subcellular localization of AIRE was assessed by using confocal microscopy. Results: PBMCs from patients with APECED syndrome had reduced TNF-alpha responses after Dectin-1 ligation but in part used a Raf-1-mediated pathway to preserve function. In the THP-1 human monocytic cell line, reducing AIRE expression resulted in significantly decreased TNF-a release after Dectin-1 ligation. AIRE formed a transient complex with the known Dectin-1 pathway components phosphorylated spleen tyrosine kinase and caspase recruitment domain-containing protein 9 after receptor ligation and localized with Dectin-1 at the cell membrane. Conclusion: AIRE can participate in the Dectin-1 signaling pathway, indicating a novel extrathymic role for AIRE and a defect that likely contributes to fungal susceptibility in patients with APECED syndrome. OBJECTIVE: The preservation of biological samples at a low temperature is important for later biochemical and/or histological analyses. However, the molecular viability of thawed samples has not been studied sufficiently in depth. The present study was undertaken to evaluate the viability of intact tissues, tissue homogenates, and isolated total RNA after defrosting for more than twenty-four hours. METHODS: The molecular viability of the thawed samples (n = 82) was assessed using the A260/A280 ratio, the RNA concentration, the RNA integrity, the level of intact mRNA determined by reverse transcriptase polymerase chain reaction, the protein level determined by Western blotting, and an examination of the histological structure. RESULTS: The integrity of the total RNA was not preserved in the thawed intact tissue, but the RNA integrity and level of mRNA were perfectly preserved in isolated defrosted samples of total RNA. Additionally, the level of beta-actin protein was preserved in both thawed intact tissue and homogenates. CONCLUSION: Isolated total RNA does not undergo degradation due to thawing for at least 24 hours, and it is recommended to isolate the total RNA as soon as possible after tissue collection. Moreover, the protein level is preserved in defrosted tissues.

We discuss the gravitational collapse of a spherically symmetric massive core of a star in which the fluid component is interacting with a growing vacuum energy density. The influence of the variable vacuum in the collapsing core is quantified by a phenomenological beta parameter as predicted by dimensional arguments and the renormalization group approach. For all reasonable values of this free parameter, we find that the vacuum energy density increases the collapsing time, but it cannot prevent the formation of a singular point. However, the nature of the singularity depends on the value of beta. In the radiation case, a trapped surface is formed for beta <= 1/2, whereas for beta >= 1/2, a naked singularity is developed. In general, the critical value is beta = 1-2/3(1 + omega) where omega is the parameter describing the equation of state of the fluid component.

Marine soft bottom systems show a high variability across multiple spatial and temporal scales. Both natural and anthropogenic sources of disturbance act together in affecting benthic sedimentary characteristics and species distribution. The description of such spatial variability is required to understand the ecological processes behind them. However, in order to have a better estimate of spatial patterns, methods that take into account the complexity of the sedimentary system are required. This PhD thesis aims to give a significant contribution both in improving the methodological approaches to the study of biological variability in soft bottom habitats and in increasing the knowledge of the effect that different process (both natural and anthropogenic) could have on the benthic communities of a large area in the North Adriatic Sea. Beta diversity is a measure of the variability in species composition, and Whittaker’s index has become the most widely used measure of beta-diversity. However, application of the Whittaker index to soft bottom assemblages of the Adriatic Sea highlighted its sensitivity to rare species (species recorded in a single sample). This over-weighting of rare species induces biased estimates of the heterogeneity, thus it becomes difficult to compare assemblages containing a high proportion of rare species. In benthic communities, the unusual large number of rare species is frequently attributed to a combination of sampling errors and insufficient sampling effort. In order to reduce the influence of rare species on the measure of beta diversity, I have developed an alternative index based on simple probabilistic considerations. It turns out that this probability index is an ordinary Michaelis-Menten transformation of Whittaker's index but behaves more favourably when species heterogeneity increases. The suggested index therefore seems appropriate when comparing patterns of complexity in marine benthic assemblages. Although the new index makes an important contribution to the study of biodiversity in sedimentary environment, it remains to be seen which processes, and at what scales, influence benthic patterns. The ability to predict the effects of ecological phenomena on benthic fauna highly depends on both spatial and temporal scales of variation. Once defined, implicitly or explicitly, these scales influence the questions asked, the methodological approaches and the interpretation of results. Problem often arise when representative samples are not taken and results are over-generalized, as can happen when results from small-scale experiments are used for resource planning and management. Such issues, although globally recognized, are far from been resolved in the North Adriatic Sea. This area is potentially affected by both natural (e.g. river inflow, eutrophication) and anthropogenic (e.g. gas extraction, fish-trawling) sources of disturbance. Although few studies in this area aimed at understanding which of these processes mainly affect macrobenthos, these have been conducted at a small spatial scale, as they were designated to examine local changes in benthic communities or particular species. However, in order to better describe all the putative processes occurring in the entire area, a high sampling effort performed at a large spatial scale is required. The sedimentary environment of the western part of the Adriatic Sea was extensively studied in this thesis. I have described, in detail, spatial patterns both in terms of sedimentary characteristics and macrobenthic organisms and have suggested putative processes (natural or of human origin) that might affect the benthic environment of the entire area. In particular I have examined the effect of off shore gas platforms on benthic diversity and tested their effect over a background of natural spatial variability. The results obtained suggest that natural processes in the North Adriatic such as river outflow and euthrophication show an inter-annual variability that might have important consequences on benthic assemblages, affecting for example their spatial pattern moving away from the coast and along a North to South gradient. Depth-related factors, such as food supply, light, temperature and salinity play an important role in explaining large scale benthic spatial variability (i.e., affecting both the abundance patterns and beta diversity). Nonetheless, more locally, effects probably related to an organic enrichment or pollution from Po river input has been observed. All these processes, together with few human-induced sources of variability (e.g. fishing disturbance), have a higher effect on macrofauna distribution than any effect related to the presence of gas platforms. The main effect of gas platforms is restricted mainly to small spatial scales and related to a change in habitat complexity due to a natural dislodgement or structure cleaning of mussels that colonize their legs. The accumulation of mussels on the sediment reasonably affects benthic infauna composition. All the components of the study presented in this thesis highlight the need to carefully consider methodological aspects related to the study of sedimentary habitats. With particular regards to the North Adriatic Sea, a multi-scale analysis along natural and anthopogenic gradients was useful for detecting the influence of all the processes affecting the sedimentary environment. In the future, applying a similar approach may lead to an unambiguous assessment of the state of the benthic community in the North Adriatic Sea. Such assessment may be useful in understanding if any anthropogenic source of disturbance has a negative effect on the marine environment, and if so, planning sustainable strategies for a proper management of the affected area.

Die Morphogenese einer Pflanzenzelle wird in großem Maße durch die Dynamik kortikaler Mikrotubuli (MT) bestimmt, die auf die Zellwandsynthese Einfluß nehmen. In dieser Arbeit wurden die Transkriptmengen der alpha-Tubulin-Isotypen und des gamma-Tubulin während der Entwicklung des Gerstenblattes analysiert, um Zusammenhänge zu bereits beschriebenen Umwandlungen im kortikalen MT-Cytoskelett der Mesophyllzellen aufzudecken. Erstmals konnte bei einer höheren Pflanze die Genexpression auf RNA-Ebene innerhalb einer Tubulin-Multigenfamilie im Verlauf der Blattentwicklung umfassend dargestellt werden.Es wurden blattspezifische cDNA-Bibliotheken erstellt und mittels RT-PCR homologe DNA-Gensonden für die Screeningprozesse der cDNA-Bibliotheken hergestellt. cDNA-Sequenzen von alpha-, beta-, und gamma-Tubulin konnten isoliert werden. Weitere, weniger abundante alpha-Tubulin-Sequenzen wurden während zusätzlicher Screeningrunden über PCR-Ausschluß häufig vertretener, bereits bekannter Isotypen isoliert.Die cDNA-Sequenzen von insgesamt fünf verschiedenen Isotypen des alpha-Tubulin konnten aufgeklärt werden, drei Isotypen wiesen bis zu fünf im nicht kodierenden 3´-Bereich verkürzte Varianten auf, die aber in ihrer Anzahl deutlich unterrepräsentiert waren. Die abgeleiteten Aminosäuresequenzen umfassten bei drei Isotypen 451 Aminosäuren (AS), zwei Isotypen waren im C-Terminus um eine bzw. um zwei AS kürzer. Die fünf alpha-Tubulin-Isotypen wiesen charakteristische Expressionsmuster auf, die in drei Klassen unterteilbar waren. Die Isotypen HVATUB1 und HVATUB5 (MT-Band-Isotypen) hatten den maximalen Gehalt in Blattbereichen, in denen auch hauptsächlich Mesophyllzellen mit kortikalen MT-Bänderungen vorkommen, wobei HVATUB5 den am schwächsten exprimierte Isotyp darstellte. HVATUB3 (Random-MT-Isotyp) zeigte die stärksten Expressionsraten. Die im Meristem und meristemnahen Bereichen bereits recht hohe Abundanz erreichte erst nach der Zellstreckungszone in einer Blattzone das Maximum, in dem hauptsächlich Mesophyllzellen mit zerstreut angeordneten MT anzutreffen sind. Die Isotypen HVATUB2 und HVATUB4 (MImax-Isotypen) waren in mitotisch aktiven, basalen Blattbereichen dominant.Die cDNA-Sequenz vom gamma-Tubulin der Gerste, HVGTUB, wurde ermittelt; die abgeleitete Aminosäuresequenz bestand aus 469 AS. Das Auftreten einer im nicht kodierenden 3´-Bereich kürzeren Variante konnte erstmals bei pflanzlichem gamma-Tubulin beschrieben werden. Southernblot-Analysen ließen darauf schließen, daß gamma-Tubulin nur als Einzelkopie im Genom der Gerste vorkommt. gamma-Tubulin wurde im mitosereichen Meristem der Blattbasis am stärksten exprimiert. Da die Abnahme der Transkriptmenge weitaus langsamer verlief als die Abnahme der Zellteilungsaktivität, ist anzunehmen, daß gamma-Tubulin neben der Erfüllung von mitose- und zellteilungsspezifischen Funktionen auch eine Rolle im Zusammenhang mit der Dynamik des kortikalen MT-Cytoskeletts spielt. Einen ersten Schritt zur Aufklärung der Genfamilie des beta-Tubulin bei Gerste stellt die Isolierung drei verschiedener cDNA-Sequenzen von beta-Tubulin dar.

ZUSAMMENFASSUNGIn den Gehirnen von Alzheimer-Patienten werden beta-Amyloid-Plaques gefunden, deren Hauptbestandteile die neurotoxischen beta-Amyloid-Peptide (A-beta) sind. Im Verlauf des nicht-amyloidogenen Wegs wird das Amyloid-Vorläuferproteins (APP) innerhalb der A-beta-Sequenz durch die alpha-Sekretase prozessiert, wobei das neuroprotektive APPs-alpha freigesetzt und die Entstehung der A-beta-Peptide verhindert wird. Die Aktivitätserhöhung der alpha-Sekretase ADAM10 könnte eine übermäßige Produktion der A-beta-Peptide abwenden.Zum Auffinden ADAM10-stimulierender Substanzen konnte ein Testsystem entwickelt werden, das auf der Fusion der 119 C-terminalen Aminosäurereste des Amyloid-Vorläuferproteins mit einem Reporterprotein beruht. Durch seine alkalische Phosphataseaktivität kann dieses Reporterprotein stellvertretend für das freigesetzte endogene APPs-alpha photometrisch im Zellkulturüberstand quantifiziert werden. Substanzen, die aktivierend auf die alpha-Sekretase ADAM10 wirken, können somit schnell und mit einer hohen Empfindlichkeit ermittelt werden.Die alpha-Sekretasen ADAM10 und TACE werden als inaktive Zymogene synthetisiert und besitzen eine Proprotein-Konvertasen-Erkennungssequenz zwischen der Prodomäne und der Metalloproteinase-Domäne. In dieser Arbeit konnte nachgewiesen werden, dass Proprotein-Konvertasen an der Prozessierung beider Zymogene beteiligt sind. ADAM10 und TACE wurden durch die Überexpression der Proprotein-Konvertasen PC7 und Furin in HEK293-Zellen in größerem Umfang prozessiert. Dies resultierte in einer erhöhten katalytischen Aktivität. Mutiertes ADAM10 ohne Proprotein-Konvertasen-Spaltstelle konnte nicht mehr in die katalytisch aktive Form überführt werden. Diese Ergebnisse eröffnen neue Ansätze zur Stimulierung des nicht-amyloidogenen Wegs.

Medulloblastoma (MB) is a paediatric malignant brain tumour, sensitive to ionizing radiations (IR). However radiotherapy has detrimental effects on long-term survivors and the tumour is incurable in a third of patients, due to intrinsic radioresistance. Alterations of the Wnt pathway distinguish a molecular subgroup of MBs and nuclear beta-catenin, indicative of activated Wnt, is associated with good outcome in MB. Therefore there are increasing evidences about Wnt involvement in radio-response: IR induce activation of Wnt signalling with nuclear translocation of beta-catenin in MB cell lines. We studied effects of Wnt pathway activation in a MB cell line with p53 wild-type: UW228-1. Cells were stably transfected with a beta-catenin constitutively active and assessed for growth curves, mortality rate, invasiveness and differentiation. Firstly, activation of Wnt pathway by itself induced a slower cell growth and a higher mortality. After IR treatment, nuclear beta-catenin further inhibited cell growth, increasing mortality. Cell invasiveness was strongly inhibited by Wnt activation. Furthermore, Wnt cell population was characterized by club shaped cells with long cytoplasmic extensions containing neurofilaments, suggesting a neural differentiation of this cell line. These findings suggest that nuclear beta-catenin may leads to a less aggressive phenotype and increases radio-sensitivity in MB, accounting for its favourable prognostic value. In the future, Wnt/beta-catenin signalling will be considered as a molecular therapeutic target to develop new drugs for the treatment of MB.

Sterne mit einer Anfangsmasse zwischen etwa 8 und 25 Sonnenmassen enden ihre Existenz mit einer gewaltigen Explosion, einer Typ II Supernova. Die hierbei entstehende Hoch-Entropie-Blase ist ein Bereich am Rande des sich bildenden Neutronensterns und gilt als möglicher Ort für den r-Prozess. Wegen der hohen Temperatur T innerhalb der Blase ist die Materie dort vollkommen photodesintegriert. Das Verhältnis von Neutronen zu Protonen wird durch die Elektronenhäufigkeit Ye beschrieben. Die thermodynamische Entwicklung des Systems wird durch die Entropie S gegeben. Da die Expansion der Blase schnell vonstatten geht, kann sie als adiabatisch betrachtet werden. Die Entropie S ist dann proportional zu T^3/rho, wobei rho die Dichte darstellt. Die explizite Zeitentwicklung von T und rho sowie die Prozessdauer hängen von Vexp, der Expansionsgeschwindigkeit der Blase, ab. Der erste Teil dieser Dissertation beschäftigt sich mit dem Prozess der Reaktionen mit geladenen Teilchen, dem alpha-Prozess. Dieser Prozess endet bei Temperaturen von etwa 3 mal 10^9 K, dem sogenannten "alpha-reichen" Freezeout, wobei überwiegend alpha-Teilchen, freie Neutronen sowie ein kleiner Anteil von mittelschweren "Saat"-Kernen im Massenbereich um A=100 gebildet werden. Das Verhältnis von freien Neutronen zu Saatkernen Yn/Yseed ist entscheidend für den möglichen Ablauf eines r-Prozesses. Der zweite Teil dieser Arbeit beschäftigt sich mit dem eigentlichen r-Prozess, der bei Neutronenanzahldichten von bis zu 10^27 Neutronen pro cm^3 stattfindet, und innerhalb von maximal 400 ms sehr neutronenreiche "Progenitor"-Isotope von Elementen bis zum Thorium und Uran bildet. Bei dem sich anschliessendem Ausfrieren der Neutroneneinfangreaktionen bei 10^9 K und 10^20 Neutronen pro cm^3 erfolgt dann der beta-Rückzerfall der ursprünglichen r-Prozesskerne zum Tal der Stabilität. Diese Nicht-Gleichgewichts-Phase wird in der vorliegenden Arbeit in einer Parameterstudie eingehend untersucht. Abschliessend werden astrophysikalische Bedingungen definiert, unter denen die gesamte Verteilung der solaren r-Prozess-Isotopenhäufigkeiten reproduziert werden können.
