The aim of this work was to develop an appropriate sample preparation procedure for the flame atomic absorption spectrometry determination of Cu, Fe, Mn, and, Zn in rocks used in organic agriculture as a source of macro- and micronutrients.Six different procedures were evaluated and are classified into three groups: (a) pressure digestion system with Teflon bombs, (b) conventional wet digestion in a digester heating block, and (c) closed microwave system with pressure and temperature control. Two standard reference materials and two commercial samples were analyzed. It was found that the closed microwave system required low reagent consumption, less time, and resulted in low contamination.

The objective of this study was to verify the effect of the passive recovery time following a supramaximal sprint exercise and the incremental exercise test on the lactate minimum speed (LMS). Thirteen sprinters and 12 endurance runners performed the following tests: 1) a maximal 500 m sprint followed by a passive recovery to determine the time to reach the peak blood lactate concentration; 2) after the maximal 500 m sprint, the athletes rested eight mins, and then performed 6 x 800 m incremental test, in order to determine the speed corresponding to the lower blood lactate concentration (LMS1) and; 3) identical procedures of the LMS1, differing only in the passive rest time, that was performed in accordance with the time to peak lactate (LMS2). The time (min) to reach the peak blood lactate concentration was significantly higher in the sprinters (12.76+/-2.83) than in the endurance runners (10.25+/-3.01). There was no significant difference between LMS1 and LMS2, for both endurance (285.7+/-19.9; 283.9+/-17.8 m/min; r= 0.96) and sprint runners (238.0+/-14.1; 239.4+/-13.9 m/min; r= 0.93), respectively. We can conclude that the LMS is not influenced by a passive recovery period longer than eight mins (adjusted according with the time to peak blood lactate), although blood lactate concentration may differ at this speed. The predominant type of training (aerobic or anaerobic) of the athletes does not seem to influence the phenomenon previously described.

In intubation experiments (643-1168 mg per animal), most of the stevioside administered to chickens was recovered unchanged in the excreta, and only about 2% was converted into steviol. Neither stevioside nor steviol could be found in the blood. In chronic studies (667 mg of stevioside/kg of feed) with laying hens and meat-type chickens, no significant differences were found in feed uptake, weight gain, and feed conversion as the result of stevioside administration. The egg production and egg composition of laying hens were not influenced. Most of the stevioside taken up was found untransformed in the excreta, and about 21.5% or 7.3% was converted to steviol by meat-type chickens or laying hens, respectively. No stevioside or steviol could be detected in the blood or in the eggs of the different groups of animals. In anaerobic incubation experiments with chicken excreta, only a 20% conversion of stevioside into steviol was found. No harmful effects were observed in the chronic stevioside supplementation experiments nor in the intubation experiments in which very high stevioside doses were given.

We performed comparative studies of the pathogenicity of six strains of Paracoccidioides brasiliensis (Bt-9, Bt-4, Pb-9, Pb-18, Bt-7 and B-1183) for young adult male ddY mice and growth rate of each strain under different oxygen atmospheres (aerobic, micro-aerobic and anaerobic atmospheres) at 37-degrees-C. 10(6) units of yeast cells were intravenously injected into each mouse. The pathogenicity of each isolate was determined by a scoring system based on organ culture and histopathological findings. The growth rates under different oxygen atmospheres were determined by a scoring system in which 300 fungal units per strain were counted. The strain Bt-9 showed the greatest pathogenicity, followed by Bt-4. Pb-9 and Pb-18 had on intermediate rank of pathogenicity. Bt-7 and B-1183 were the least pathogenic of the strains tested. Except for strain Bt-7 all strains showed an excellent growth under an aerobic atmosphere. Bt-4 and Bt-9 also showed excellent growth under a micro-aerobic atmosphere, followed by Pb-9, whereas the growth of Pb-18, Bt-7 and B-1183 was limited. There was a correlation between the growth rate under a micro-aerobic atmosphere and the pathogenicity of a strain. The growth rate of P. brasiliensis under a micro-aerobic atmosphere strongly correlated to its pathogenicity.

The aim of this investigation was to study the effect of organic acids and/or anaerobic cecal microflora (ACM) on systemic and digestive infection of broilers by Salmonella typhimurium and S. enteritidis. ACM was used without previous bacterial identification. The treatment with ACM increased the resistance to Salmonella spp infection. Infection was more evident in caeca, followed by rectum and crop and did not interfere on body weight of broilers. Treated and control groups showed the same degree of infection at the end of the experiment. The use of ACM isolated or combined with acetic acid, reduced the colonization of the chick's digestive system by S. typhimurium and S. enteritidis. Acetic acid added to ACM did not potentiate the reduction of digestive system colonization. Except for the crop, the isolated use of acetic, propionic or formic acids did not reduce S. typhimurium and S. enteritidis, in caeca and rectum. The use of organic acids and ACM had little effect on reduction of caecum pH. The treatment with ACM reduced the quantity of S. enteritidis in the faeces. The reduction of caecum pH did not reduce the quantity of S. enteritidis in faeces. S. enteritidis was much more invasive than S. typhimurium and use of organic acids and ACM had little effect on reduction of systemic infection.

The aims of this study were to estimate the changes in total bacterial counts (TBC) in poultry litter samples, consisting of rice hulls, after storage, and to identify pathogenic bacteria. For the countings Plate Count agar (Difco) was used. Enrichment and selective media such as blood agar, MacConkey, Baird Parker, brain and heart agar, and egg yolk solid media, and cooked meat and brain and heart infusion, incubated under aerobic or anaerobic conditions were used to isolate Staphylococcus aureus, Salmonella sp, Clostridium perfringens, C. botulinum, C. chauvoei, Campylobacter sp, Escherichia coli, and Corynebacterium sp. Litter samples were collected from the houses of the Veterinary School experimental aviary. A fully randomized experimental design was used with four treatments and four replications, for a total of 16 samples. A decrease in TBC was detected when treatment T1 (zero days of storage) was compared with treatments T2 (14 days of storage). on the other hand the treatments T3 (28 days of storage) and T4 (42 days of storage) presented significantly superior counting in relation to treatment T1. Some pathogenic bacteria of Enterobacteriaceae such as Escherichia coil, Proteus, Arizona, Providencia, Edwardsiella, as well as Staphylococcus aureus, S. epidermidis, different species of genus Clostridium as C. perfringens, C. sordelli, C. chauvoei, C. tetani and C. novyi as well as some strains of Corynebacterium pyogenes were isolated.

The stabilization of swine wastewaters from swine confined housing by the combination of a upflow anaerobic sludge blanket (UASB) reactor and waste stabilization ponds is a viable alternative to minimize the environmental impact caused by inadequate disposal of swine wastewaters. In the present study, the polluting load of pre-decanted swine wastewater treated with a series of two 0.705 m(3) UASB reactors and then in parallel in aerated and non-aerated stabilization tanks was investigated from January to July, 2000. Physicochemical and microbiological analyses were made adopting standard methods (Standard Methods for Examination of Water and Wastewater, 19th ed., American Public Health Association, Washington, DC, 1995). COD values decreased as the wastewater ran through the integrated biodigestion system dropping from about 3492 +/- 511-4094 mg l(-1) +/- 481 to 124 +/- 52-490 mg l(-1) +/- 230, while nitrate and nitrite levels increased in stabilization tanks, ranging respectively from 4 +/- 0 to 20 mg l(-1) +/- 3 and 3 +/- 1 to 11 mg l(-1) +/- 24. Although the removal of Escherichia coli was more than 97% +/- 6, the effluents of the treatment system still contained unacceptable levels of E. coli (1.6 x 10(3)-1.2 x 10(6) 100 ml(-1)) according to WHO guidelines for use of wastewater in agriculture and aquaculture. These results indicate the necessity of changes on operational characteristics of the treatment system such as an increase of the hydraulic retention time in UASB reactors or in stabilization tanks. Biofertilizers are organic products usually obtained from an anaerobic process, evaluated mainly as phytoprotectors for agriculture, although they can improve fruit production. By this way, a field experiment was carried out in Remigio County, Paraiba State, Brazil, to evaluate the effects of two bovine biofertilizers on yield, medium mass and quality of papaya fruits (Carica papaya L.), 'Baixinho de Santa Amalia cultivar. Treatments were distributed in a randomized complete blocks design with three replications and 15 plants in each parcel, in a 2 x 5 factorial arrangement, with two biofertilizers: simple (water and fresh bovine manure) and supermagro (water, fresh bovine manure, macro and micronutrients and a protein mix), applied in soil under liquid form at five volumes, as follows: 0.0; 0.5; 1.0; 1.5; e 2.0 L cave(-1). There was no statistical difference between biofertilizers for fruit mass, yield and external and internal fruit quality variables, while between volumes differences were registered for both biofertilizers. The results, except for fruit firmness, registered positive effects of the biofertilizers on 'Baixinho de Santa Amalia' papaya cultivar in relation to yield and physical and chemical fruit quality.

To determine the inhibitory capacity of lactic acid bacteria due to the action of antagonistic substances, we tested 474 isolates of Lactobacillus from the crop and cecum of chickens against gram-positive and gram-negative indicator microorganisms by the spot-on-the-lawn and well-diffusion antagonism methods. of the 474 isolates, 265 demonstrated antimicrobial activity against the indicator microorganisms. Isolates identified as L. reuteri, L. salivarius, or Lactobacillus spp. inhibited Enterococcus faecalis, E. faecium, Listeria monocytogenes, and Salmonella spp. but not L. casei, L. delbrueckii, L. fermentum, or L. helveticus by the well-diffusion simultaneous antagonism method under anaerobic incubation conditions. The antagonistic substances produced by some of the Lactobacillus isolates were inactivated after treatment by proteolytic enzymes, which suggested that the substances could be antimicrobial peptides or bacteriocins.

Despite improvements in connectology, peritoneal dialysis (PD) - associated peritonitis contributes significantly to morbidity and modality failure in patients maintained on PD therapy. A broad spectrum of organisms -gram-positive, gram-negative, fungal, anaerobic - are involved in this complication. In addition, a significant percentage of episodes involve polymicrobial and culture-negative infection. Technological advances are being developed to minimize the incidence of access-related complications such as peritonitis. Many traditional factors such as exit-site infection and poor technique have been already identified. In the present review, we discuss the geographic, patient selection, and clinical issues that can affect peritonitis rates in different areas of the world and in different centers in the same area.
