Charles Larned (1791-1834) was a lawyer and American military officer who served during the War of 1812. He was the son of Simon Larned (1753-1817), who served as a captain in the Revolutionary War and was a member of the United States Congress from Massachusetts from 1804-1805. Charles studied law in the office of Henry Clay in Kentucky, and was dining with a group of prominent citizens when word was received that General William Henry Harrison could soon be overpowered by General Henry Proctor. Colonel Owen, a member of the group, organized a regiment to reinforce Harrison’s troops. Larned became a member and subsequently survived the River Raisin Massacre and was later present at the Battle of the Thames. He was also part of a group of men who learned of General William Hull’s plan to surrender Detroit to the British and planned to overtake him should this occur. However, the plan failed, Hull did surrender and the men became prisoners of the British. After the war, Larned became a lawyer, and served as Attorney General of Michigan Territory during the Black Hawk War. During the cholera epidemic of 1834, he worked tirelessly to assist others, but was stricken with the disease and died. Our dear Father we hope & trust is now in Green Bush, where he will probably remain a month perhaps & from thence he expects to go to Sacket’s harbor—at which place you know our troops are fast collecting-- We shall hope to see him either here or there before he goes. Probably before this you have been informed of the great loss your friend Sherrill has sustained in the death of his mother—also of the revolution that has taken place in Hackbridge as it respects the religion & morality of the place that more than one hundred on the plain have become religious converts & c—indeed I am at a loss what to say that will afford your pleasure—a narrative at this time must be gloomy indeed. Diatoms are renowned for their robust ability to perform NPQ (Non-Photochemical Quenching of chlorophyll fluorescence) as a dissipative response to heightened light stress on photosystem II, plausibly explaining their dominance over other algal groups in turbulent light environs. Their NPQ mechanism has been principally attributed to a xanthophyll cycle involving the lumenal pH regulated reversible de-epoxidation of diadinoxanthin. The principal goal of this dissertation is to reveal the physiological and physical origins and consequences of the NPQ response in diatoms during short-term transitions to excessive irradiation. The investigation involves diatom species from different originating light environs to highlight the diversity of diatom NPQ and to facilitate the detection of core mechanisms common among the diatoms as a group. A chiefly spectroscopic approach was used to investigate NPQ in diatom cells. Prime methodologies include: the real time monitoring of PSII excitation and de-excitation pathways via PAM fluorometry and pigment interconversion via transient absorbance measurements, the collection of cryogenic absorbance spectra to measure pigment energy levels, and the collection of cryogenic fluorescence spectra and room temperature picosecond time resolved fluorescence decay spectra to study excitation energy transfer and dissipation. Chemical inhibitors that target the trans-thylakoid pH gradient, the enzyme responsible for diadinoxanthin de-epoxidation, and photosynthetic electron flow were additionally used to experimentally manipulate the NPQ response. Multifaceted analyses of the NPQ responses from two previously un-photosynthetically characterised species, Nitzschia curvilineata and Navicula sp., were used to identify an excitation pressure relief ‘strategy’ for each species. Three key areas of NPQ were examined: (i) the NPQ activation/deactivation processes, (ii) how NPQ affects the collection, dissipation, and usage of absorbed light energy, and (iii) the interdependence of NPQ and photosynthetic electron flow. It was found that Nitzschia cells regulate excitation pressure via performing a high amplitude, reversible antenna based quenching which is dependent on the de-epoxidation of diadinoxanthin. In Navicula cells excitation pressure could be effectively regulated solely within the PSII reaction centre, whilst antenna based, diadinoxanthin de-epoxidation dependent quenching was implicated to be used as a supplemental, long-lasting source of excitation energy dissipation. These strategies for excitation balance were discussed in the context of resource partitioning under these species’ originating light climates. A more detailed investigation of the NPQ response in Nitzschia was used to develop a comprehensive model describing the mechanism for antenna centred non-photochemical quenching in this species. The experimental evidence was strongly supportive of a mechanism whereby: an acidic lumen triggers the diadinoxanthin de-epoxidation and protonation mediated aggregation of light harvesting complexes leading to the formation of quencher chlorophyll a-chlorophyll a dimers with short-lived excited states; quenching relaxes when a rise in lumen pH triggers the dispersal of light harvesting complex aggregates via deprotonation events and the input of diadinoxanthin. This model may also be applicable for describing antenna based NPQ in other diatom species.

Each person with Autism Spectrum Disorder (ASD) comes with unique characteristics (idiosyncratic) that give clues to the world they know (Connolly, 2008). It is through their body that they (a) know the world they are experiencing, (b) make meaning, and (c) express certain behaviours. I used Laban’s Movement Analysis (LMA) to practice an attuned and appreciative approach to describing and understanding the body movement in one severe manifestation of autism in an adolescent male. LMA observes human movement across many disciplines and can be applied in many contexts providing a body honoring discourse for description (Connolly, 2008). The framework examines movement in body, space, quality, and relation. Each theme provides a detailed description of the individual’s movement, thus, giving us a richer understanding of patterns and possible triggers to self-injurious behaviours (SIB). During the summer of August 2013, I participated in Brock University’s annual Autism Camp and worked with a 15 year old male named “Aaron” who manifests with low functioning autism. The purpose of my research project was to code and analyze a series of photos taken to help gain insight into movement patterns associated with stressed embodiment and self-injury in “Aaron”. As I understood more about these embodied expressions, I uncovered valuable information on how to read patterns and discover what triggers these events, thus providing strategies on how to help people do more refined observations and make meaning of the behaviour. Laban’s movement analysis provided a sensitized discourse appropriate to the embodied expressions depicted in the photos.

Watersheds are complex systems involving social, economic, and ecological dimensions that are constantly interacting and influencing each other, and governance of these systems involve a large and diverse cast of actors that add to the complexity and difficulty in deciding what is best for the watershed and people. Resilience thinking offers a way to understand and navigate the uncertainty, dynamics and complexity of watershed governance. This primer describes key ideas associated with resilience: more inclusive participation; building a shared understanding; inclusion of ecosystem services and functions in long-term planning; strong leadership; institutional and decision making flexibility; and, a decentralized system. This primer is an initial effort to translate the scholarly understanding of these key ideas and initiate a dialogue about their application in the context of watershed governance.

L'angiotensine-II (Ang-II), synthétisée à partir de sources extracardiaques et intracardiaques, régule l'homéostasie cardiaque en favorisant des effets mitogéniques et en promouvant la croissance cellulaire résultant d’une altération de l'expression génique. Dans cette étude, nous avons évalué la possibilité que les récepteurs de l'angiotensine-1 (AT1) ou les récepteurs de l'angiotensine-2 (AT2) situés sur l'enveloppe nucléaire régulent l’expression génique des cardiomyocytes. En analysant les noyaux cellulaires retenus des fractions de cœur de rat par immunobuvardage Western, nous avons détecté une co-purification préférentielle des protéines AT1 et AT2 avec un marqueur de la membrane nucléaire (Nup 62), par rapport aux marqueurs de la membrane plasmique (Calpactin I), de l’appareil de Golgi (GRP 78) ou du réticulum endoplasmique (GM130). La microscopie confocale a permis de démontrer la présence des AT1 et AT2 dans les membranes nucléaires. La microinjection de l’Ang-II-FITC sur des cardiomyocytes a provoqué une liaison de préférence aux sites nucléaires. Les enregistrements de transients calciques ont illustré que les AT1 nucléaires régulent le relâchement du Ca2+. L’incubation des ligands spécifiques d’AT1 et d’AT2 avec l’UTP [α32P] a résulté en une synthèse de novo d’ARN (par exemple, 16,9 ± 0,5 cpm/ng ADN contrôle vs 162,4 ± 29,7 cpm/ng ADN-Ang II, 219,4 ± 8,2 cpm/ng ADN L -162313 (AT1) et 126,5 ± 8,7 cpm/ng ADN CGP42112A (AT2), P <0,001). L’incubation des noyaux avec Ang-II augmente de façon significative l’expression de NFκB, une réponse qui est réprimée partiellement par la co-administration de valsartan ou de PD123177. Les expériences dose-réponse avec Ang-II administrée à l'ensemble des noyaux purifiés vs. aux cardiomyocytes seuls a montré une augmentation plus importante dans les niveaux d'ARNm de NFκB avec une affinité de ~ 3 fois plus grande (valeurs d’EC50 = 9 contre 28 pmol/L, respectivement), suggérant un rôle préférentiel nucléaire dans la signalisation. Par conséquent, nous avons conclu que les membranes cardiaques nucléaires possèdent des récepteurs d’Ang-II couplés à des voies de signalisation et à la transcription génique. La signalisation nucléaire pourrait jouer un rôle clé dans les changements de l'expression de gènes cardiaques, entraînant ainsi des implications mécanistiques et thérapeutiques diverses.

L’endothéline-1 (ET-1) est un puissant agent vasoconstricteur dont la production est dérégulée dans plusieurs maladies inflammatoires où l’expression des cyclooxygénases-1/2 (COX-1/2) est augmentée. Puisqu’il est connu que la voie p38 MAPK est impliquée dans la régulation de l’ET-1 au niveau de l’ARNm, nous avons étudié le rôle de l’un de ses substrats, la kinase MK2 dans la régulation post-transcriptionnelle de l’ET-1 et des COX. Pour ce faire, nous avons utilisé des souris MK2-déficientes (MK2-/-) ainsi que des contrôles (MK2+/+) issus de la même portée. Des paramètres de la fonction cardiaque ont été mesurés sous anesthésie à l’aide d’un cathéter Millar et la réactivité vasculaire de l’artère fémorale a été mesurée par myographe. L’expression de ET-1, COX-1 et COX-2 a été quantifiée dans la cellule endothéliale aortique (CE) par qPCR. En réponse à l’ET-1 (100 nM), l’expression de la préproET-1 dans les CE augmente en fonction du temps (p<0.05) : cette variation est accentuée chez les souris MK2-/-. Bien que la pression artérielle soit similaire entre les souris MK2+/+ et MK2-/-, l’inhibition de COX (indométacine, 1 μM) augmente (p<0.05) la contraction à l’ET-1 des vaisseaux isolés provenant de souris MK2+/+ mais pas des MK2-/-. Ces données suggèrent un rôle de MK2 dans la réponse vasculaire à l’ET-1 et possiblement dans la signalisation post-récepteur de l’ET-1 en général.
