P>With the evidence showing the protection variability of bacille Calmette-Guerin, new potential vaccines for tuberculosis have been tested around the world. One of the general concerns in tuberculosis vaccine development is the possibility of priming the host immune system with prior exposure to environmental mycobacteria antigens, which can change the efficacy of subsequent vaccination. As there is a great homology between the species from Mycobacterium genera, the previous contact of experimental animals with environmental mycobacteria could sensitize the mice and, in this way, could influence subsequent vaccine research. The aim of our study was to investigate critical points in an animal facility to search for environmental mycobacteria that eventually could be in direct or indirect contact with the experimental animals. Samples were collected from surfaces of walls, floor, animal cages and shelves and analysed using the Ogawa-Kudoh decontamination method. Samples of drinking water, food and sawdust were collected for analysis by the NALC/NaOH decontamination method. Also, the samples were cultivated directly in broth medium, without any method for decontamination. After decontamination methods, we observed bacterial colony growth in 4.31% of the total of samples analysed. These samples were stained with Ziehl-Neelsen and we did not detect any acid-fast bacilli, suggesting that the animal facility analysed is free from contamination by environmental mycobacteria and is not a source of mycobacterial antigens. Furthermore, our study showed a new paradigm in tuberculosis vaccine development: concern about the animal facility environment in terms of immune system priming of experimental animals by nascent bacterial contaminants.

10-(Octyloxy) decyl-2-(trimethylammonium) ethyl phosphate (ODPC) is an alkylphospholipid that can interact with cell membranes because of its amphiphilic character. We describe here the interaction of ODPC with liposomes and its toxicity to leukemic cells with an ED-50 of 5.4, 5.6 and 2.9 pM for 72 h of treatment for inhibition of proliferation of NB4, U937 and K562 cell lines, respectively, and lack of toxicity to normal hematopoietic progenitor cells at concentrations up to 25 pM. The ED-50 for the non-malignant HEK-293 and primary human umbilical vein endothelial cells (HUVEC) was 63.4 and 60.7 mu M, respectively. The critical micellar concentration (CMC) of ODPC was 200 mu M. Dynamic light scattering indicated that dipalmitoylphosphatidylcholine (DPPC) liposome size was affected only above the CMC of ODPC. Differential calorimetric scanning (DCS) of liposomes indicated a critical transition temperature (T(c)) of 41.5 degrees C and an enthalpy (Delta H) variation of 7.3 kcal mol(-1). The presence of 25 mu M ODPC decreased T(c) and Delta H to 393 degrees C and 4.7 kcal mol(-1), respectively. ODPC at 250 mu M destabilized the liposomes (36.3 degrees C. 0.46 kcal mol(-1)). Kinetics of 5(6)-carboxyfluorescein (CF) leakage from different liposome systems indicated that the rate and extent of CF release depended on liposome composition and ODPC concentration and that above the CMC it was instantaneous. Overall, the data indicate that ODPC acts on in vitro membrane systems and leukemia cell lines at concentrations below its CMC, suggesting that it does not act as a detergent and that this effect is dependent on membrane composition. The electrochemical degradation of different glyphosate herbicide formulations on RuO(2) and IrO(2) DSA(A (R)) electrodes is investigated. Parameters that could influence the formation of organochloride compounds during electrolysis are studied. The effects of chloride concentration, electrodic composition, current density, and electrolysis time are reported. The influence of the oxide composition on herbicide degradation seems to be almost insignificant; however, there is a straight relationship between anode composition and organic halides formation. Commercial herbicide formulations have lower degradation rates and lead to the formation of a larger quantities of organochloride compounds. In high chloride concentrations, there is a significant increase in organic mineralization, and the relationship between chloride concentration and organic halides formation is direct. Only in low chloride medium investigated the organochloride concentration obtained was below the limit values allowed in Brazil. The determination of organic halides absorbable (AOX) during electrolysis increases significantly with the applied current. Even during long-term electrolysis, a large amount of organochloride compounds is formed.

Wolcott-Rallison syndrome (WRS, OMIM 226980) is a rare autosomal recessive disorder characterized by permanent neonatal diabetes mellitus, epiphyseal dysplasia, and other multisystemic clinical manifestations. We described two novel mutations in the EIF2AK3 gene in two consanguineous families with WRS from Brazil and Morocco. We have observed in case 1 a homozygous C > T replacement at base pair c.1192 at exon 7, generating a stop codon at position 398 (Gln398Stop). Both of his parents were found to be heterozygous for the mutation. We detected in both parents of case 2, a deceased Moroccan girl, a duplication of base pair c.851A at exon 5 (c.851dupA) leading to a frameshift and a stop codon at position 285 (p.Pro285AlafsX3). Both cases 1 and 2 had neonatal diabetes mellitus, multiple epiphyseal dysplasia, and growth delay, and presented episodes of acute hepatic dysfunction. Case 1 presented central hypothyroidism, developmental delay, and mild mental retardation. Case 2 presented a fatal episode of acute renal failure. The clinical phenotype associated with the syndrome can be variable, but a combination of infancy-onset diabetes mellitus, multiple epiphyseal dysplasia, and hepatic and/or renal dysfunction is the mainstay of diagnosis.

Notopronocephalus peekayi gen, et sp, n. is described from the intestine of Elseya latisternum Gray, 1867, E. dentata (Gray, 1863) and Emydura signata Ahl, 1932 from rivers in Queensland. The new genus is distinguished by the absence of ventral glands, simple (neither diverticulate nor sinuous) caeca terminating at the anterior margin of the testes, excretory arms not uniting in forebody, single ovary, two opposite testes close to the posterior end of the body, intracaecal genital pore, vitelline follicles anterior to the testes, cirrus-sac orientated obliquely and not divided into two portions, and the uterus intracaecal. This is the first pronocephalid to be described from an Australian freshwater turtle and the first from the family Chelidae.

Objectives/Hypothesis: Blood supply to the Hadad-Bassagasteguy pedicled nasoseptal flap may be interrupted by surgery of the pterygopalatine fossa, posterior septectomy, or large sphenoidotomies. This would preclude its use for reconstruction of skull base defects after expanded endonasal approaches (EEA). We present a novel method to ascertain the patency of the nasoseptal artery after prior surgery, and consequently the availability of the nasoseptal flap, using acoustic Doppler sonography. Study Design: Retrospective clinical review. Methods: Four patients who underwent EEAs were evaluated intraoperatively with acoustic Doppler sonography. The mucosa that covers the inferior aspect of the rostrum of the sphenoid sinus was scanned with the tip of the probe. Reflection of sound waves representing intravascular blood flow was assessed. Results: In three patients, the artery was identified in at least one side. One remaining patient showed no acoustic signal suggesting loss of the nasoseptal artery bilaterally, therefore necessitating the use of a fat graft for the reconstruction. Conclusions: Acoustic Doppler sonography seems to be a feasible and effective way to ascertain the availability of the nasoseptal artery. It is a relatively inexpensive and simple technique that can be performed by any endoscopic surgeon.

Traumatic transanal small bowel evisceration is a rare condition usually associated with suction injuries or blunt abdominal trauma. We report the first case of intestinal evisceration through the anus caused by penetrating trauma (rectal impalement). Additionally, we performed a literature review of all English language articles since 1970 concerned with traumatic transanal small bowel evisceration. Mechanisms of injury and the surgical management are discussed.

Spotted fever is a disease caused by bacteria from the genus Rickettsia of the spotted fever group (SFG). Rickettsia rickettsii is likely the main agent of Brazilian spotted fever (BSF). With the objective of gathering information on the circulation of SFG rickettsiae in Londrina, Parana state, ticks from dogs and horses and also blood from dogs, horses and humans were collected in a neighbourhood of the city which presented potential for circulation of rickettsiae between hosts and vectors. Amblyomma cajennense, Dermacentor nitens, and Rhipicephalus sanguineus ticks were subjected to Polymerase Chain Reaction targeting a fragment of the Rickettsia gltA gene. This specific gene encodes the enzyme citrate synthase of Rickettsia spp., and results on all ticks were negative. Human and animal sera were tested by Indirect Immunofluorescence Assay in which R. rickettsii and R. parkeri were used as antigens. Sera from 4.7% human, 2.7% canine and 38.5% equine were positive for R. rickettsii. For R. parkeri, 0.9% human, 2.7% canine and 11.5% equine samples were positive. All samples reactive to R. parkeri also reacted to R. rickettsii. An epidemiological questionnaire was applied, but there were no statistically significant results. Comparison of our serological results with previous studies in Brazil, among BSF endemic and non-endemic areas, indicates that there is no established rickettsial infection in the study area, a statement corroborated with our molecular analysis. Nonetheless, as humans of the present study are highly exposed to tick infestations, health education within the population is needed to obtain efficient tick control. P>To evaluate the prevalence of antibodies to Chlamydophila psittaci 364 serum samples were collected from veterinarians, biologists, animal scientists, veterinary students, animal keepers and others employees in 20 zoos, and from veterinary practitioners in 10 Brazilian states. Subjects ranged from 15 to 64 years of age, with 268 (74%) males and 96 (26%) females. Chlamydial antibodies were determined by the complement fixation test (CFT) and specific anti-C. psittaci IgG antibodies were determined by the microimmunoflurescence (MIF) test. Complement fixation test showed 23.9% (87/364) and MIF test showed 4.7% (17/364) positive serum samples. Titres ranged from 16 to 256 in both assays, demonstrating evidence of recent or current infection. Although chlamydial antibodies were detected in workers of seventeen zoos, MIF test only detected specific C. psittaci antibodies in seven of them. Previous psittacosis infection was suspected in eight workers of two zoos, five of whom reported having pneumonia, while employed at the zoos. However, diagnosis was not established in any of these cases in the past. Results indicated the occurrence of infection and previous contact of Brazilian zoo workers with C. psittaci, as well as the zoonotic potential of psittacosis in this risk population. Other studies are necessary to evaluate the risk factors of infection in this population. This seroepidemiological survey confirmed the need to adopt preventive measures to control avian chlamydiosis and protect the health of zoo workers in the country.

Background: Estimates of the performance of carbohydrate deficient transferrin (CDT) and gamma glutamyltransferase (GGT) as markers of alcohol consumption have varied widely. Studies have differed in design and subject characteristics. The WHO/ISBRA Collaborative Study allows assessment and comparison of CDT, GGT, and aspartate aminotransferase (AST) as markers of drinking in a large, well-characterized, multicenter sample. Methods: A total of 1863 subjects were recruited from five countries (Australia, Brazil, Canada, Finland, and Japan). Recruitment was stratified by alcohol use, age, and sex. Demographic characteristics, alcohol consumption, and presence of ICD-10 dependence were recorded using an interview schedule based on the AUDADIS, CDT was assayed using CDTect(TM) and GGT and AST by standard methods. Statistical techniques included receiver operating characteristic (ROC) analysis. Multiple regression was used to measure the impact of factors other than alcohol on test performance. Results: CDT and GGT had comparable performance on ROC analysis, with AST performing slightly less well. CDT was a slightly but significantly better marker of high-risk consumption in men. All were more effective for detection of high-risk rather than intermediate-risk drinking. CDT and GGT levels were influenced by body mass index, sex, age, and smoking status. Conclusions: CDT was little better than GGT in detecting high- or intermediate-risk alcohol consumption in this large, multicenter, predominantly community-based sample. As the two tests are relatively independent of each other, their combination is likely to provide better performance than either test alone, Test interpretation should take account sex, age. and body mass index.

O principal objetivo deste trabalho foi identificar e caracterizar a evolução diária da Camada Limite Atmosférica (CLA) na Região da Grande Vitória (RGV), Estado do Espírito Santo, Brasil e na Região de Dunkerque (RD), Departamento Nord Pas-de-Calais, França, avaliando a acurácia de parametrizações usadas no modelo meteorológico Weather Research and Forecasting (WRF) em detectar a formação e atributos da Camada Limite Interna (CLI) que é formada pelas brisas marítimas. A RGV tem relevo complexo, em uma região costeira de topografia acidentada e uma cadeia de montanhas paralela à costa. A RD tem relevo simples, em uma região costeira com pequenas ondulações que não chegam a ultrapassar 150 metros, ao longo do domínio de estudos. Para avaliar os resultados dos prognósticos feitos pelo modelo, foram utilizados os resultados de duas campanhas: uma realizada na cidade de Dunkerque, no norte da França, em Julho de 2009, utilizando um sistema light detection and ranging (LIDAR), um sonic detection and ranging (SODAR) e dados de uma estação meteorológica de superfície (EMS); outra realizada na cidade de Vitória – Espírito Santo, no mês de julho de 2012, também usando um LIDAR, um SODAR e dados de uma EMS. Foram realizadas simulações usando três esquemas de parametrizações para a CLA, dois de fechamento não local, Yonsei University (YSU) e Asymmetric Convective Model 2 (ACM2) e um de fechamento local, Mellor Yamada Janjic (MYJ) e dois esquemas de camada superficial do solo (CLS), Rapid Update Cycle (RUC) e Noah. Tanto para a RGV quanto para a RD, foram feitas simulações com as seis possíveis combinações das três parametrizações de CLA e as duas de CLS, para os períodos em que foram feitas as campanhas, usando quatro domínios aninhados, sendo os três maiores quadrados com dimensões laterais de 1863 km, 891 km e 297 km, grades de 27 km, 9 km e 3 km, respectivamente, e o domínio de estudo, com dimensões de 81 km na direção Norte-Sul e 63 km na Leste-Oeste, grade de 1 km, com 55 níveis verticais, até um máximo de, aproximadamente, 13.400 m, mais concentrados próximos ao solo. Os resultados deste trabalho mostraram que: a) dependendo da configuração adotada, o esforço computacional pode aumentar demasiadamente, sem que ocorra um grande aumento na acurácia dos resultados; b) para a RD, a simulação usando o conjunto de parametrizações MYJ para a CLA com a parametrização Noah produziu a melhor estimativa captando os fenômenos da CLI. As simulações usando as parametrizações ACM2 e YSU inferiram a entrada da brisa com atraso de até três horas; c) para a RGV, a simulação que usou as parametrizações YSU para a CLA em conjunto com a parametrização Noah para CLS foi a que conseguiu fazer melhores inferências sobre a CLI. Esses resultados sugerem a necessidade de avaliações prévias do esforço computacional necessário para determinadas configurações, e sobre a acurácia de conjuntos de parametrizações específicos para cada região pesquisada. As diferenças estão associadas com a capacidade das diferentes parametrizações em captar as informações superficiais provenientes das informações globais, essenciais para determinar a intensidade de mistura turbulenta vertical e temperatura superficial do solo, sugerindo que uma melhor representação do uso de solo é fundamental para melhorar as estimativas sobre a CLI e demais parâmetros usados por modelos de dispersão de poluentes atmosféricos.

We report on structural, electronic, and optical properties of boron-doped, hydrogenated nanocrystalline silicon (nc-Si:H) thin films deposited by plasma-enhanced chemical vapor deposition (PECVD) at a substrate temperature of 150 degrees C. Film properties were studied as a function of trimethylboron-to-silane ratio and film thickness. The absorption loss of 25% at a wavelength of 400 nm was measured for the 20 nm thick films on glass and glass/ZnO:Al substrates. By employing the p(+) nc-Si:H as a window layer, complete p-i-n structures were fabricated and characterized. Low leakage current and enhanced sensitivity in the UV/blue range were achieved by incorporating an a-SiC:H buffer between the p- and i-layers.

Pretendemos neste trabalho abordar o desenvolvimento da atenção conjunta no contexto de interacções entre a criança e seus cuidadores pela importância que lhe é atribuída para o desenvolvimento sóciocognitivo e, em particular, o desenvolvimento da linguagem. O interesse por este tema advém também do relativo atraso que a investigação reporta no caso de crianças surdas filhas de pais ouvintes que se associa ao designado fenómeno de atenção dividida (Gallaway, 1998; Lederberg, 1993; Wood, Wood Griffiths, & Howarth, 1986). Por causa deste fenómeno, as crianças surdas têm de aprender a coordenar a sua atenção em relação à pessoa com quem interagem, bem como em relação a objecto ou eventos no seu ambiente. Pelas potenciais implicações para as práticas, analisaremos as estratégias de interacção usadas por mães surdas que, com mais facilidade do que mães ouvintes, andaimam o desenvolvimento das capacidades de coordenação da atenção visual das crianças e facilitam a ocorrência de episódios de atenção conjunta e a influencia que os resultados destas investigações têm tido em programas de intervenção precoce direccionados para o apoio a crianças surdas e suas famílias.

Numa primeira abordagem a A Lady’s Visit to Manilla and Japan (1863), de Anna D’Almeida, os leitores não deverão esperar encontrar a narrativa de uma experiência que poderia ter sido produzida por um desses “Etonnants voyageurs! Quelles nobles histoires / Nous lisons dans vos yeux profonds comme les mers!”, citando o último poema de Les Fleurs du Mal de Baudelaire. Nem deverão esperar ser confrontados com o relato superficial de uma turista indolente sobre a diversão convencional ou o previsível choque moral experimentados durante as várias etapas do seu grand tour pessoal, tão em voga, e que são característicos deste tipo de literatura, particularmente popular no campo emergente do turismo do final do século xix. Neste artigo, proponho-me analisar a escrita feminina occidental no contexto dos encontros culturais, mais precisamente, as imagens que uma viajante ocidental do século xix cria a partir da sua breve exposição a vários espaços e práticas da Ásia. A família D’Almeida viajou pelo Extremo Oriente entre Março e Julho de 1862. O título A Lady’s Visit to Manilla and Japan induz em erro, pois a narrativa começa em Singapura e termina em Hong Kong, mas a família visitou também Macau, Xangai, Nagasáqui, Yokohama, Xiamen (Hokkien) e Cantão, entre outros lugares, atestando assim o profundo desejo dos D’Almeida de explorar in loco todas as potencialidades dos países visitados Neste estudo tenciono demonstrar as complexidades que existem dentro de / entre as histórias, experiências e actividades interculturais de mulheres, e como estas alargam o âmbito do estudo dos sistemas sociais e culturais. Ao examinar as diferenças e semelhanças de género, podemos elaborar construções teóricas que analisam as variações entre mulheres; como elas são influenciadas pela classe, raça, etnia e religião; e como estas moldam a forma como entendemos a posição da mulher na cultura e na sociedade. O preconceito de classe da elite ocidental considera a mulher não-ocidental como sendo ‘a outra’, alguém que representa aquilo que o escritor ocasional não é. A questão da representação feminina das suas congéneres como ‘mulheres-outras’, com base numa ampla variedade de diferenças, é definitivamente um desafio para os estudos interculturais e de género.
