Aquest treball de recerca té com a objectiu la síntesi i caracterització de nous interruptors moleculars basats en cromòfors azobenzènics i amb aplicació a nivell biològic. Més concretament, es planteja el desenvolupament d’interruptors moleculars basats en el sistema MAG per a controlar el funcionament d’un canal iònic de les cèl·lules del sistema nerviós central governat pel receptor iGluR. Tot i que ja s’han descrit interruptors moleculars tipus MAG que permeten el control d’aquest canal, en aquest treball es pretenen optimitzar dues de les seves propietats: augmentar el rendiment quàntic de fotoisomerització entre els estats trans i cis de l’interruptor i augmentar la constant tèrmica d’isomerització cis-&trans. Aquestes dues propietats depenen de les característiques estructurals del cromòfor azobenzènic central del sistema MAG.

OBJECTIVE To study the molecular genetic and clinical features of cerebral cavernous malformations (CCM) in a cohort of Spanish patients. METHODS We analyzed the CCM1, CCM2, and CCM3 genes by MLPA and direct sequencing of exons and intronic boundaries in 94 familial forms and 41 sporadic cases of CCM patients of Spanish extraction. When available, RNA studies were performed seeking for alternative or cryptic splicing. RESULTS A total of 26 pathogenic mutations, 22 of which predict truncated proteins, were identified in 29 familial forms and in three sporadic cases. The repertoire includes six novel non-sense and frameshift mutations in CCM1 and CCM3. We also found four missense mutations, one of them located at the third NPXY motif of CCM1 and another one that leads to cryptic splicing of CCM1 exon 6. We found four genomic deletions with the loss of the whole CCM2 gene in one patient and a partial loss of CCM1and CCM2 genes in three other patients. Four families had mutations in CCM3. The results include a high frequency of intronic variants, although most of them localize out of consensus splicing sequences. The main symptoms associated to clinical debut consisted of cerebral haemorrhage, migraines and epileptic seizures. The rare co-occurrence of CCM with Noonan and Chiari syndromes and delayed menarche is reported. CONCLUSIONS Analysis of CCM genes by sequencing and MLPA has detected mutations in almost 35% of a Spanish cohort (36% of familial cases and 10% of sporadic patients). The results include 13 new mutations of CCM genes and the main clinical symptoms that deserves consideration in molecular diagnosis and genetic counselling of cerebral cavernous malformations.

The administration of selective serotonin reuptake inhibitors (SSRIs) typically used as antidepressants increases alcohol consumption after an alcohol deprivation period in rats. However, the appearance of this effect after the treatment with selective noradrenaline reuptake inhibitors (SNRIs) has not been studied. In the present work we examined the effects of a 15-d treatment with the SNRI atomoxetine (1, 3 and 10 mg/kg, i.p.) in male rats trained to drink alcohol solutions in a 4-bottle choice test. The treatment with atomoxetine (10 mg/kg, i.p.) during an alcohol deprivation period increased alcohol consumption after relapse. This effect only lasted one week, disappearing thereafter. Treatment with atomoxetine did not cause a behavioral sensitized response to a challenge dose of amphetamine (1.5 mg/kg, i.p.), indicating the absence of a supersensitive dopaminergic transmission. This effect is markedly different from that of SSRI antidepressants that produced both long-lasting increases in alcohol consumption and behavioral sensitization. Clinical implications are discussed.

Este estudo foi desenvolvido com o objetivo de caracterizar as quedas de pacientes internados ocorridas em hospital terciário. Foram analisados 826 Boletins de Notificação de Eventos Adversos, de um período de 30 meses, que registraram 0,30 quedas por 1000 pacientes/dia. Quedas do leito foram mais frequentes (55%), com maior prevalência na enfermaria de neurologia. Maior frequência de quedas foi verificada no período noturno (63,7%), nos primeiros cinco dias da admissão (61,7%), nos pacientes de sexo masculino (57,5%) e na faixa etária maior de 60 anos (50%). Nos casos de quedas do leito, os diagnósticos relacionaram-se a doenças infecciosas e parasitárias (18,2%), doenças do sistema nervoso (18,2%) e doenças do aparelho circulatório (13,7%). Nas quedas da própria altura, os diagnósticos relacionaram-se a neoplasias (19,4%) e doenças do aparelho geniturinário (16,1%). A caracterização desses eventos adversos auxilia no reconhecimento dos grupos de maior risco e na elaboração de propostas preventivas.

Background: During early steps of embryonic development the hindbrain undergoes a regionalization process along the anterior-posterior (AP) axis that leads to a metameric organization in a series of rhombomeres (r). Refinement of the AP identities within the hindbrain requires the establishment of local signaling centers, which emit signals that pattern territories in their vicinity. Previous results demonstrated that the transcription factor vHnf1 confers caudal identity to the hindbrain inducing Krox20 in r5 and MafB/Kreisler in r5 and r6, through FGF signaling [1].Results: We show that in the chick hindbrain, Fgf3 is transcriptionally activated as early as 30 min after mvHnf1 electroporation, suggesting that it is a direct target of this transcription factor. We also analyzed the expression profiles of FGF activity readouts, such as MKP3 and Pea3, and showed that both are expressed within the hindbrain at early stages of embryonic development. In addition, MKP3 is induced upon overexpression of mFgf3 or mvHnf1 in the hindbrain, confirming vHnf1 is upstream FGF signaling. Finally, we addressed the question of which of the FGF-responding intracellular pathways were active and involved in the regulation of Krox20 and MafB in the hindbrain. While Ras-ERK1/2 activity is necessary for MKP3, Krox20 and MafB induction, PI3K-Akt is not involved in that process.Conclusion: Based on these observations we propose that vHnf1 acts directly through FGF3, and promotes caudal hindbrain identity by activating MafB and Krox20 via the Ras-ERK1/2 intracellular pathway.

El projecte en el qual s’emmarca aquest treball de recerca es centra en el desenvolupament d’interruptors moleculars que permeten el control del funcionament d’un canal iònic de les cèl·lules del sistema nerviós central. En aquest treball de Màster en Experimentació Química, s’ha treballat, més concretament, en la síntesi del derivat del glutamat. S’ha dissenyat i desenvolupat una ruta sintètica del fragment de glutamat en 8 passos amb un 7% de rendiment global.

STUDY OBJECTIVES The objective of this study was to evaluate the effectiveness of a mass vaccination programme carried out in Catalonia (Spain) in the last quarter of 1997 in response to an upsurge of serogroup C meningococcal disease (SCMD). DESIGN Vaccination coverage in the 18 month to 19 years age group was investigated by means of a specific vaccination register. Vaccination effectiveness was calculated using the prospective cohort method. Cases of SCMD were identified on the basis of compulsory reporting and microbiological notification by hospital laboratories. Vaccination histories were investigated in all cases. Unadjusted and age adjusted vaccination effectiveness referred to the time of vaccination and the corresponding 95% confidence intervals (CI) were estimated at 6, 12, 18 and 24 months of follow up. SETTING All population aged 18 months to 19 years of Catalonia. MAIN RESULTS A total of seven cases of SCMD were detected at six months of follow up (one in the vaccinated cohort), 12 cases at 12 months (one in the vaccinated cohort), 19 cases at 18 months (two in the vaccinated cohort) and 24 at 24 months (two in the vaccinated cohort). The age adjusted effectiveness was 84% (95%CI 30, 97) at six months, 92% (95%CI 63, 98) at 12 months, 92% (95% CI 71, 98) at 18 months and 94% (95%CI 78, 98) at 24 months. In the target population, cases have been reduced by more than two thirds (68%) two years after the vaccination programme. In the total population the reduction was 43%. CONCLUSION Vaccination effectiveness has been high in Catalonia, with a dramatic reduction in disease incidence in the vaccinated cohort accompanied by a relevant reduction in the overall population. Given that vaccination coverage was only 54.6%, it may be supposed that this vaccination effectiveness is attributable, in part, to the herd immunity conferred by the vaccine.

O objetivo deste trabalho foi identificar os efeitos de sistemas de preparo do solo e de rotações de culturas sobre o rendimento de grãos e economicidade da cultura do feijoeiro (Phaseolus vulgaris L.) irrigado pelo sistema pivô central. O trabalho foi conduzido durante seis anos consecutivos, na Embrapa-Centro Nacional de Pesquisa de Arroz e Feijão, localizado no Município de Santo Antônio de Goiás, GO, em Latossolo Vermelho-Escuro, de textura argilosa. O experimento consistiu de um fatorial 4 x 6, em delineamento inteiramente casualizado, com parcelas subdivididas. Os sistemas de preparo do solo foram: P1 arado de aiveca (novembro-dezembro) alternado com grade aradora (maio-junho); P2 arado de aiveca contínuo; P3 grade aradora contínua; P4 plantio direto; e as rotações de culturas: R1 arroz-feijão; R2 milho-feijão; R3 soja-trigo, R4 soja-trigo-soja-feijão-arroz-feijão; R5 arroz consorciado com calopogônio-feijão; e R6 milho-feijão-milho-feijão-arroz -feijão. As rotações R1, R2, R3 e R5 foram anuais, e as R4 e R6, trienais. Neste trabalho analisaram-se somente as rotações que continham feijão. O arroz, o milho e a soja foram semeados em novembro-dezembro (verão), e o feijão e o trigo, em maio-junho (inverno). Houve efeito do preparo do solo e das rotações de cultura sobre o rendimento de grãos do feijoeiro. A cultura produziu mais quando se utilizou no preparo do solo a combinação de arado de aiveca, nos cultivos de verão, e grade aradora nos de inverno. Os rendimentos do feijoeiro foram maiores quando a cultura foi implantada bienalmente na mesma área, nas rotações com arroz/calopogônio e arroz, e menores, nas rotações com milho. O feijoeiro irrigado, com relação ao preparo do solo e rotações de culturas, foi economicamente viável, propiciando taxas de retorno que variaram de 67% a 97%.

Adult mammalian central nervous system (CNS) axons have a limited regrowth capacity following injury. Myelin-associated inhibitors (MAIs) limit axonal outgrowth and their blockage improves the regeneration of damaged fiber tracts. Three of these proteins, Nogo-A, MAG and OMgp, share two common neuronal receptors: NgR1, together with its co-receptors (p75(NTR), TROY and Lingo-1), and the recently described paired immunoglobulin-like receptor B (PirB). These proteins impair neuronal regeneration by limiting axonal sprouting. Some of the elements involved in the myelin inhibitory pathways may still be unknown, but the discovery that blocking both PirB and NgR1 activities leads to near-complete release from myelin inhibition, sheds light on one of the most competitive and intense fields of neuroregeneration study during in recent decades. In parallel with the identification and characterization of the roles and functions of these inhibitory molecules in axonal regeneration, data gathered in the field strongly suggest that most of these proteins have roles other than axonal growth inhibition. The discovery of a new group of interacting partners for myelin-associated receptors and ligands, as well as functional studies within or outside the CNS environment, highlights the potential new physiological roles for these proteins in processes such as development, neuronal homeostasis, plasticity and neurodegeneration.

A adubação nitrogenada é uma prática cultural normalmente utilizada pelos produtores de feijão. O objetivo deste trabalho foi avaliar o uso do clorofilômetro como instrumento indicador da necessidade de adubação nitrogenada em cobertura no feijoeiro, cultivado no inverno, irrigado por aspersão, pelo sistema pivô central. A primeira etapa, realizada em 2000, consistiu no cultivo das cultivares de feijão, Pérola e Jalo Precoce, submetidas a 0, 30, 60 e 120 kg ha-1 de N aplicados em cobertura, com a finalidade de estabelecer curvas de resposta entre as leituras do clorofilômetro, modelo Minolta SPAD-502, em cada dose de N, e a produtividade de grãos. A segunda etapa, realizada em 2001, consistiu em validar, no campo, os valores de leitura do medidor indicativos de nível adequado de N na planta. As produtividades das duas cultivares e as leituras do clorofilômetro aumentaram com o aumento da dose de nitrogênio. Na mesma dose de N, os valores de leitura foram maiores na cultivar Pérola. O clorofilômetro se mostrou eficaz como instrumento indicador da necessidade de adubação nitrogenada em cobertura no feijoeiro.

O nitrogênio é um nutriente essencial ao feijoeiro e sua carência é observada em quase todos os solos. O objetivo deste trabalho foi avaliar a resposta do feijoeiro irrigado por aspersão à adubação nitrogenada em cobertura, num Latossolo Vermelho distrófico. O cultivo foi realizado em sistema pivô central, em condições de plantio direto com sucessão de diferentes culturas. Os tratamentos constituíram-se de sete culturas: braquiária cv. Marandu, milho em consórcio com braquiária, guandu, milheto, mombaça, sorgo granífero e estilosantes cv. Mineirão. Sobre as palhadas picadas das culturas, foi semeado o feijão cv. Pérola e aplicados em cobertura 0, 30, 60 e 120 kg ha-1 de N (uréia). Houve efeito das palhadas sobre a produtividade de grãos e as maiores produtividades alcançadas foram sobre as palhadas de milheto e do guandu. O feijoeiro responde à aplicação de N em cobertura em todas as sucessões, com resposta quadrática sobre o milheto e o guandu, e linear nas demais.

The biological consequences of constitutive fibroblast growth factor-4 (fgf4) expression have been analysed during anterior CNS development of mouse chimeric embryos. Severe mutant embryos exhibit exencephaly, absence of eye development and anomalous differentiation of neuropithelium. These embryos also show ectopic limb buds resembling the early phases of limb development. Because our results show that anterior CNS in those chimeric embrios does not express shh ectopically, we suggest that malformations may be due to interference between the ectopic expression of fgf4 in the cephalic area and the receptors for the members of the FGF family that regulate brain and eye development, namely fgf8. If this is correct, the results indirectly suport the crucial role of fgf8 in patterning the anterior CNS.

Rationale Mephedrone (4-methylmethcathinone) is a still poorly known drug of abuse, alternative to ecstasy or cocaine. Objective The major aims were to investigate the pharmacokineticsa and locomotor activity of mephedrone in rats and provide a pharmacokinetic/pharmacodynamic model. Methods Mephedrone was administered to male SpragueDawley rats intravenously (10 mg/kg) and orally (30 and 60 mg/kg). Plasma concentrations and metabolites were characterized using LC/MS and LC-MS/MS fragmentation patterns. Locomotor activity was monitored for 180240 min. Results Mephedrone plasma concentrations after i.v. administration fit a two-compartment model (α=10.23 h−1, β=1.86 h−1). After oral administration, peak mephedrone concentrations were achieved between 0.5 and 1 h and declined to undetectable levels at 9 h. The absolute bioavailability of mephedrone was about 10 % and the percentage of mephedrone protein binding was 21.59±3.67%. We have identified five phase I metabolites in rat blood after oral administration. The relationship between brain levels and free plasma concentration was 1.85±0.08. Mephedrone induced a dose-dependent increase in locomotor activity, which lasted up to 2 h. The pharmacokineticpharmacodynamic model successfully describes the relationship between mephedrone plasma concentrations and its psychostimulant effect. Conclusions We suggest a very important first-pass effect for mephedrone after oral administration and an easy access to the central nervous system. The model described might be useful in the estimation and prediction of the onset, magnitude,and time course of mephedrone pharmacodynamics as well as to design new animal models of mephedrone addiction and toxicity.

Scrapie is a transmissible spongiform encephalopathy with a wide PrPres dissemination in many non-neural tissues and with high levels of transmissibility within susceptible populations. Mechanisms of transmission are incompletely understood. It is generally assumed that it is horizontally transmitted by direct contact between animals or indirectly through the environment, where scrapie can remain infectious for years. In contrast, in utero vertical transmission has never been demonstrated and has rarely been studied. Recently, the use of the protein misfolding cyclic amplification technique (PMCA) has allowed prion detection in various tissues and excretions in which PrPres levels have been undetectable by traditional assays. The main goal of this study was to detect PrPres in fetal tissues and the amniotic fluid from natural scrapie infected ewes using the PMCA technique. Six fetuses from three infected pregnant ewes in an advanced clinical stage of the disease were included in the study. From each fetus, amniotic fluid, brain, spleen, ileo-cecal valve and retropharyngeal lymph node samples were collected and analyzed using Western blotting and PMCA. Although all samples were negative using Western blotting, PrPres was detected after in vitro amplification. Our results represent the first time the biochemical detection of prions in fetal tissues, suggesting that the in utero transmission of scrapie in natural infected sheep might be possible.

Glioblastoma multiforme (GBM) is a commonly occurring brain tumor with a poor prognosis. GBM can develop both “de novo” or evolve from a previous astrocytoma and is characterized by high proliferation and infiltration into the surrounding tissue. Following treatment (surgery, radiotherapy, and chemotherapy), tumors often reappear. Glioma-initiating cells (GICs) have been identified in GBM and are thought to be responsible for tumors initiation, their continued growth, and recurrence. β-catenin, a component of the cell-cell adhesion complex and of the canonical Wnt pathway, regulates proliferation, adhesion, and migration in different cell types. β-catenin and components of the Wnt canonical pathway are commonly overexpressed in GBM. Here, we review previous work on the role of Wnt/β-catenin signalling in glioma initiation, proliferation, and invasion. Understanding the molecular mechanisms regulating GIC biology and glioma progression may help in identifying novel therapeutic targets for GBM treatment.
