Este trabalho descreve a construção e a determinação de parâmetros físico-químicos com um eletrodo de segunda ordem do tipo, Pt|Hg|Hg2(PAB)2|Grafite, sensível ao íon p-aminobenzoato (PAB). O eletrodo é construído facilmente, apresenta um rápido tempo de resposta, é de baixo custo e tem um tempo de vida útil superior a 12 meses. Utilizando o referido eletrodo foi possível estimar os coeficientes de atividade iônica individuais do PAB, a 25ºC, em força iônica ajustada entre 0,700-3,000 mol L-1 com NaClO4, em solução aquosa. O potencial molal padrão do eletrodo determinado a 25ºC é de (445,5 ± 0,5) mV. A constante termodinâmica do produto de solubilidade, T Kps (I=0) do Hg2(PAB)2 determinada com o eletrodo é de (T Kps = 2,50 x 10-12 mol³ L-3 , a 25ºC). Uma característica favorável consiste no fato de que o eletrodo pode ser utilizado para determinar a concentração de íons p-aminobenzoato livres em sistemas complexos de íons metálicos.

Carbon monoxide was detected and determined by a piezoelectric quartz crystal sensor coated with nickel(II)-phthalocyanine 50 % (v/v) solution in glycerine. Studies on the effect of temperature, flow rate, and some possible interferents were carried out. Calibration curves, sensor stability (lifetime) and the precision of measurements were also verified. The resulting selectivity is probably due to the coordinative binding between the electronically unsatured metal complexes and the analyte. The analytical curve is linear in the concentration range 0.10 to 1.0 % (v/v).

The number of autonomous wireless sensor and control nodes has been increasing rapidly during the last decade. Until recently, these wireless nodes have been powered with batteries, which have lead to a short life cycle and high maintenance need. Due to these battery-related problems, new energy sources have been studied to power wireless nodes. One solution is energy harvesting, i.e. extracting energy from the ambient environment. Energy harvesting can provide a long-lasting power source for sensor nodes, with no need for maintenance. In this thesis, various energy harvesting technologies are studied whilst focusing on the theory of each technology and the state-of-the-art solutions of published studies and commercial solutions. In addition to energy harvesting, energy storage and energy management solutions are also studied as a subsystem of a whole energy source solution. Wireless nodes are also used in heavy-duty vehicles. Therefore a reliable, long-lasting and maintenance-free power source is also needed in this kind of environment. A forestry harvester has been used as a case study to study the feasibility of energy harvesting in a forestry harvester’s sliding boom. The energy harvester should be able to produce few milliwatts to power the target system, an independent limit switch.

Semiempirical calculations at the level of PM3 of theory were carried out to study the structural and electronic properties of C80 and some of its doped derivatives with the elements of group III and V at the level of PM3 of theory. We have selected these elements to be substituted in the fullerene-C80 cage in order to show the effect of such structural change on the electronic properties of the molecules studied. The theoretical IR spectra, some of physical and chemical properties of the molecules studied are obtained and discussed.

National Health Surveillance Agency (ANVISA) established in the decree number 54 maximum allowed levels for Ni and Pb in mineral and natural waters at 20 µg L-1 and 10 µg L-1, respectively. For screening analysis purposes, the high-resolution continuum source flame atomic absorption spectrometry technique (HR-CS FAAS) was evaluated for the fast-sequential determination of nickel and lead in mineral waters.Two atomic lines for Ni (232.003 nm - main and 341.477 nm - secondary) and Pb (217.0005 nm - main and 283.306 nm - secondary) at different wavelength integrated absorbance (number of pixels) were evaluated. Sensitivity enhanced with the increase of the number of pixels and with the summation of the atomic lines absorbances. The main figures of merit associated to the HR-CS FAAS technique were compared with that obtained by line-source flame atomic absorption spectrometry (LS FAAS). Water samples were pre-concentrated about 5-fold by evaporation before analysis. Recoveries of Pb significantly varied with increased wavelength integrated absorbance. Better recoveries (92-93%) were observed for higher number of pixels at the main line or summating the atomic lines (90-92%). This influence was irrelevant for Ni, and recoveries in the 92-104% range were obtained in all situations.

Voltammetric technique was used to study the binary and ternary complexes of cadmium with L-amino acids and vitamin-C (L-ascorbic acid) at pH =7.30 ± 0.01, µ = 1.0M KNO3 at 25ºC and 35ºC. Cd (II) formed 1:1:1, 1:1:2 and 1:2:1 complexes with L-lysine, L-ornithine, L-threonine, L-serine, L-phenylglycine, L-phenylalanine, L-glutamic acid and L-aspartic acid used as primary ligands and L-ascorbic acid used as secondary ligand. The trend of stability constant of complexes was L-lysine < L-ornithine < L-threonine < L-serine < L-phenylglycine < L-phenylalanine < L-glutamic acid < L-aspartic acid which can be explained on the basis of size, basicity and steric hindrance of ligands. The values of stability constant (log β) varied from 2.23 to11.33 confirm that these drugs i.e. L-amino acids or in combination with L-ascorbic acid or their complexes could be used against Cd (II) toxicity. The study has been carried out at 35ºC also to determine the thermodynamic parameters such as enthalpy change (ΔH), Free energy change (ΔG) and entropy change (ΔS) respectively.

Neste estudo, procurou-se desenvolver e avaliar filmes para a captação e quantificação de vapores de amônia no ar, através de um sensor piezelétrico de quartzo. Muitas substâncias e suas misturas, em diferentes proporções, foram investigadas como possíveis filmes captores. Em etapas seguintes, verificaram-se alguns parâmetros importantes como o efeito da vazão do poluente, da temperatura de trabalho e da massa da película, otimizando-se as condições experimentais para a montagem do método. Concluiu-se que o filme mais promissor, sob as condições ajustadas, seria uma mistura 2:1 (v/v) de solução comercial (A) de ácido glicólico (70 % m/m) em água, adicionada a tetrakis(hidroxietil)etilenodiamina - THEED (3:4 v/v), com uma solução saturada (B) de ácido tânico em acetona. Estudos de repetibilidade, do tempo de contacto (poluente-sensor) e de alguns possíveis interferentes completaram os trabalhos. As curvas analíticas resultantes mostraram faixas lineares na região de trabalho (2,0 a 11 ppm v ou 1,4 a 7,7 mg/m³ de NH3), para quatro diferentes tempos de exposição (0,5; 1; 2 e 3 min), com coeficientes de correlação (r) variando entre 0,9994 e 0,9980, e respectivas sensibilidades de 13,5 a 42,0 Hz / ppm v.

Adequate supply of oxygen is essential for the survival of multicellular organisms. However, in several conditions the supply of oxygen can be disturbed and the tissue oxygenation is compromised. This condition is termed hypoxia. Oxygen homeostasis is maintained by the regulation of both the use and delivery of oxygen through complex, sensitive and cell-type specific transcriptional responses to hypoxia. This is mainly achieved by one master regulator, a transcription factor called hypoxiainducible factor 1 (HIF-1). The amount of HIF-1 is under tight oxygen-dependent control by a family of oxygen-dependent prolyl hydroxylase domain proteins (PHDs) that function as the cellular oxygen sensors. Three family members (PHD1-3) are known to regulate HIF of which the PHD2 isoform is thought to be the main regulator of HIF-1. The supply of oxygen can be disturbed in pathophysiological conditions, such as ischemic disorders and cancer. Cancer cells in the hypoxic parts of the tumors exploit the ability of HIF-1 to turn on the mechanisms for their survival, resistance to treatment, and escape from the oxygen- and nutrient-deprived environment. In this study, the expression and regulation of PHD2 were studied in normal and cancerous tissues, and its significance in tumor growth. The results show that the expression of PHD2 is induced in hypoxic cells. It is overexpressed in head and neck squamous cell carcinomas and colon adenocarcinomas. Although PHD2 normally resides in the cytoplasm, nuclear translocation of PHD2 was also seen in a subset of tumor cells. Together with the overexpression, the nuclear localization correlated with the aggressiveness of the tumors. The nuclear localization of PHD2 caused an increase in the anchorage-independent growth of cancer cells. This study provides information on the role of PHD2, the main regulator of HIF expression, in cancer progression. This knowledge may prove to be valuable in targeting the HIF pathway in cancer treatment.

Wireless sensor networks and its applications have been widely researched and implemented in both commercial and non commercial areas. The usage of wireless sensor network has developed its market from military usage to daily use of human livings. Wireless sensor network applications from monitoring prospect are used in home monitoring, farm fields and habitant monitoring to buildings structural monitoring. As the usage boundaries of wireless sensor networks and its applications are emerging there are definite ongoing research, such as lifetime for wireless sensor network, security of sensor nodes and expanding the applications with modern day scenarios of applications as web services. The main focus in this thesis work is to study and implement monitoring application for infrastructure based sensor network and expand its usability as web service to facilitate mobile clients. The developed application is implemented for wireless sensor nodes information collection and monitoring purpose enabling home or office environment remote monitoring for a user.

Since the introduction of automatic orbital welding in pipeline application in 1961, significant improvements have been obtained in orbital pipe welding systems. Requirement of more productive welding systems for pipeline application forces manufacturers to innovate new advanced systems and welding processes for orbital welding method. Various methods have been used to make welding process adaptive, such as visual sensing, passive visual sensing, real-time intelligent control, scan welding technique, multi laser vision sensor, thermal scanning, adaptive image processing, neural network model, machine vision, and optical sensing. Numerous studies are reviewed and discussed in this Master’s thesis and based on a wide range of experiments which already have been accomplished by different researches the vision sensor are reported to be the best choice for adaptive orbital pipe welding system. Also, in this study the most welding processes as well as the most pipe variations welded by orbital welding systems mainly for oil and gas pipeline applications are explained. The welding results show that Gas Metal Arc Welding (GMAW) and its variants like Surface Tension Transfer (STT) and modified short circuit are the most preferred processes in the welding of root pass and can be replaced to the Gas Tungsten Arc Welding (GTAW) in many applications. Furthermore, dual-tandem gas metal arc welding technique is currently considered the most efficient method in the welding of fill pass. Orbital GTAW process mostly is applied for applications ranging from single run welding of thin walled stainless tubes to multi run welding of thick walled pipes. Flux cored arc welding process is faster process with higher deposition rate and recently this process is getting more popular in pipe welding applications. Also, combination of gas metal arc welding and Nd:YAG laser has shown acceptable results in girth welding of land pipelines for oil and gas industry. This Master’s thesis can be implemented as a guideline in welding of pipes and tubes to achieve higher quality and efficiency. Also, this research can be used as a base material for future investigations to supplement present finding.

With the increase of use of digital media the need for the methods of multimedia protection becomes extremely important. The number of the solutions to the problem from encryption to watermarking is large and is growing every year. In this work digital image watermarking is considered, specifically a novel method of digital watermarking of color and spectral images. An overview of existing methods watermarking of color and grayscale images is given in the paper. Methods using independent component analysis (ICA) for detection and the ones using discrete wavelet transform (DWT) and discrete cosine transform (DCT) are considered in more detail. A novel method of watermarking proposed in this paper allows embedding of a color or spectral watermark image into color or spectral image consequently and successful extraction of the watermark out of the resultant watermarked image. A number of experiments have been performed on the quality of extraction depending on the parameters of the embedding procedure. Another set of experiments included the test of the robustness of the algorithm proposed. Three techniques have been chosen for that purpose: median filter, low-pass filter (LPF) and discrete cosine transform (DCT), which are a part of a widely known StirMark - Image Watermarking Robustness Test. The study shows that the proposed watermarking technique is fragile, i.e. watermark is altered by simple image processing operations. Moreover, we have found that the contents of the image to be watermarked do not affect the quality of the extraction. Mixing coefficients, that determine the amount of the key and watermark image in the result, should not exceed 1% of the original. The algorithm proposed has proven to be successful in the task of watermark embedding and extraction.

O Cerrado é formado por um mosaico de fisionomias campestres, savânicas e florestais que possuem um típico ciclo fenológico. Nesse contexto, os dados do MODIS fornecem medidas diárias que permitem monitorar a sazonal fenologia da vegetação. O objetivo deste trabalho foi caracterizar formações savânicas, formações florestais e áreas de cerrado convertido pela ação antrópica, utilizando séries temporais de NDVI e EVI do sensor MODIS, após a suavização de ruídos. A metodologia adotada pode ser subdividida nos seguintes passos: (a) confecção do cubo temporal com NDVI e EVI, onde o perfil em z corresponde à assinatura temporal, (b) tratamento do ruído e (c) detecção das assinaturas temporais. A Transformação pela Fração Mínima de Ruído (MNF) foi aplicada para suavizar ruídos contidos nas séries temporais. Os resultados indicaram que os valores de NDVI foram maiores que os valores de EVI e existiu relação com as estações do ano. As formações florestais foram as que apresentaram maiores valores de NDVI e EVI, possuindo ainda as menores variações entre as estações. As áreas convertidas apresentaram os menores valores dos dois índices, tendo grande queda em seus valores no início da estação seca, provavelmente por se tratar do período de colheita. O estudo concluiu que as séries temporais NDVI e EVI são úteis na diferenciação dos tipos de vegetação.

The objective of this work was to evaluate the feasibility of vegetative propagation through cutting technique of seven tree species with strong occurrence in the riparian forest of the Lower São Francisco River in Sergipe State, under different concentrations of indolbutiric acid at 0, 2500, and 5000 mg.L-1, for potentialization of its use in soil bioengineering technique. It was used a complete random block design with three replicates, and a total of twenty-one treatments. The evaluation period was 120 days for each species, and the data collection was made in intervals of fifteen days, in a total of eight evaluations for each species. The evaluated parameters were: Survival Rate, callus formation, and Root Dry matter Weight. Among the studied species, Schinus terebinthifolius Raddi presented the best results related to cutting technique mainly under the indolbutiric acid concentration of 2500 mg.L-1.

Imagens NDVI (Índice de Vegetação por Diferença Normalizada) do sensor Modis foram utilizadas para mapear as classes de uso e cobertura da terra nas Serras do Sudeste e Campanha Meridional do Rio Grande do Sul. A metodologia compreendeu a elaboração de um banco de dados espaciais e a aplicação de técnicas de processamento digital (contraste linear, classificação digital e operações aritméticas) sobre imagens dos satélites Landsat e Terra de diversas datas. Os resultados indicaram que a cobertura florestal passou de 8,6% para 11,6% e 14,3% da área total da microrregião Serras do Sudeste, entre os anos 2000, 2004 e 2008. Na Campanha Meridional, a expansão da cobertura florestal passou de 11,1% para 11,2% e 11,5% da área total no mesmo período. Conclui-se que imagens MOD13Q1, de baixa resolução espacial (250 m), podem ser usadas em grandes áreas para mapear florestas e os demais temas adequadamente.
