植物已经演化出多种保护其免受强光抑制和破坏的机制,从而使植物体在自然界能够应付复杂多变的光照环境。虽然人们早就确定Cyt b-559存在于PSII反应中心内,但目前对其性质与功能的认识还不充分。本工作的目的就是研究Cyt b-559天然分子特性,探讨其生理功能和存在的意义。取得了一些有新意的结果: 1、依据PSII反应中心分离纯化的原理,应用更有效的层析介质DEAE-Sephacel,我们设计了快速高效的从菠菜和水稻中分离纯化Cyt b-559的方法和流程,获得了高纯度的样品。它们在非变性胶电泳中具有相同的泳动性。蛋白组分的HPLC结果证明,纯化的Cyt b-559的确由两个亚基组成,α亚基和β亚基的分子量用我们设计的适合于分析小蛋白的Tricine—SDS—PAGE方法准确测定为9.4kDa和4.5kDa。 2、利用HPLC技术分析了纯化的Cyt b-559样品的色素组成,结果表明Cytb-559中含有Chl α而不含类胡萝卜素分子,这一结果通过吸收光谱和共振拉曼光谱的分析得到进一步地证明。通过等电聚焦方法分析了Cyt b-559的等电点,发现其亚基的等电点相差很大,全蛋白的等电点与...更多D1、D2蛋白的等电点也不相同,推测在体内生理pH条件下它们具有相反带电性而在PSII组装中发挥作用。 3、低温荧光光谱的检测结果表明,Cyt b-559的荧光发射峰位在563nm和666nm;首次证明Cyt b-559可以发出荧光和将电子传递给结合在其上的辅助叶绿素,但传递能力比较低故而导致其荧光特性与PSII反应中心的不相同。Cytb-559的紫外荧光光谱表明Trp残基位于其内部的疏水区域,证明Cyt b-559中的芳香族氨基酸可能在其功能的发挥中起一定作用。 4、通过MCD的分析,发现Cyt b-559中血红素的MCD信号在540—580nm和400—440nm波段,而且光谱形状和强度与PSII反应中心的相一致,说明PSII反应中心该范围内的MCD信号中有Cyt b-559的贡献。FTIR光谱的测定结果证明Cyt b-559血红素的配体是组氨酸,其二级结构中α-螺旋占了一半。此外,还比较了Cyt b-559和PSII反应中心的膜脂成分,发现两者有很大的相似性。不同植物来源的Cyt b-559在许多性质上都表现出高度一致,从一个侧面证明Cyt b-559在进化中的保守性。 5、PSll反应中心发生光破坏时,原初电子供体P680己受到严重破坏。我们发现,在光抑制的最初一段时间内,Cyt b-559吸收峰值发生变化:在受体侧光抑制的条件下,其吸收峰值先略有增加而后才下降,而在供体侧光抑制条件下则相反,说明 Cyt b-559对光抑制的发生非常敏感,可能在光抑制早期保护PSll反应中心。 6、纯化的Cyt b-559的组氨酸含量在照光前后没有显著的变化,说明 PSll反应中心内被破坏的组氨酸不属于Cyt b-559。PSll反应中心所含的组氨酸中有些可被DEPC修饰,但我们的实验结果表明DEPC不能修饰Cyt b-559的组氨酸。这可能有利于Cyt b-559保护功能的发挥。 7、我们观察到,在两种光抑制条件下,LP Cyt b-559光还原和 HP Cyt b-559光氧化具有对pH值的依赖性,说明Cyt b-559在光保护中的作用不仅与其高低电势态有关,而且与其质子化程度有联系。CCCP促进HP Cyt -559释放质子,从而维持循环电子传递。DCBQ和 DCMU在很低浓度时都抑制 Cyt b-559光还原,前者不影响Cyt b-559光氧化而后者在CCCP存在时也会抑制Cyt b-559光氧化。 8、Cyt b-559有定位PSll反应中心其它蛋白的锚蛋白的作用。黄化苗转绿实验证明在 HP Cyt b-559的含量增加超过 45%以后放氧活性开始逐渐增加。Cytb-559从低电势态到高电势态的转变是放氧复合物组装到PSll反应中心的关键步骤之一。在植物正常生长时,Cyt b-559与 P680的其它电于供体发生竟争,起到安全阀门的作用。 9、在逆境条件下,Cyt b-559具有保护PSll反应中心免受强光破坏而起到“分于开关”的作用。我们的实验表明,在室温条件下存在通过Cyt b-559的环式电子流,存在从氧化态LP Cyt b-559到还原态HP Cyt b-559的一个循环,其中的氧化还原变化与质子化/去质子化反应相连。通过与其它血红素蛋白的比较,我们推测 Cyt b-559“分子开关”的关键是:光抑制情况下,铁原子与远端His之间的疏水空穴被氧自由基占据后使得铁进入叶琳中央孔中,迫使近端HIS向叶琳平面位移,从而引起 Cyt b-559构象改变,使电势态发生转变。

本文由两部分组成,一部分是关于一组新型除草剂(K-15,K-23)的抑制特性及作用位点的研究;另一部分是关于碳酸氢盐对细胞色素b-559高电势的保护作用的研究。 在第一部分,我们首先研究了抑制剂K-23对Psn放氧活性、DCIP光还原和可变荧光等光合特性的影响。研究发现,K-23在低浓度时刺激放氧活性,而在相对高浓度时抑制放氧活性。但是,K-23在低浓度时却有效地抑制了可变荧光。这些数据表明了新型抑制剂的抑制反应是基于氧化还原作用而不是猝灭作用。此外,通过采用胰蛋白酶消化类囊体膜的方式初步检测了新型抑制剂的作用部位,其结果表明:虽然新型抑制剂抑制受体侧电子传递,但它的抑制部位与DCMU不同。 其次,研究了新型抑制剂对光系统II色素蛋白复合体与多肽组分的影响及抑制剂的键合蛋白。应用SDS-PAGE技术,发现新型抑制剂主要作用于光系统II的反应中心蛋白。用温和的Deriphat-PAGE分析也证实了新型抑制剂作用于核心复合物,导致核心复合物二聚体消失。 进一步用SDS-PAGE分析新型抑制剂对Psn多肽组分的影响,发现新型抑制剂主要影响D.、D2、CP43和CP47。用荧光发射的方法确定了K-15键合在D2蛋白上。 最后,结合荧光动力学和HPLC方法,分别从定性和定量的角度,以核心复合物以及抑制剂存在下从BBY中分离的核心复合物为研究对象,详细研究了抑制剂对QA的取代作用。研究发现,在无去垢剂或低浓度去垢剂存在情况下,由于不能创造出适合于QA存在的疏水环境,我们没有得到QA被K-15取代的实验证据。而在抑制剂K-15存在下,用2.2% HTG从BBY分离的核心复合物的实验中,检测不到正的可变荧光Fv,而是得到了降低的FM,这个结果表明QA已被抑制剂在它的作用位点处所取代。 在第二部分,研究了pH5.0—7.0范围内碳酸氢盐对Cyt b-559氧化还原状态转变的影响。首先研究了pH5.0~7.0条件下碳酸氢盐对PSII Cyt b-559还原减氧化差异吸收光谱的影响,发现铁氰化钾还原的PSII Cyt b-559 HP的含量随介质pH值的降低而减少。然而,碳酸氢盐的加入阻止了由于介质pH降低而引起的Cyt b-559由高电势向低电势的转化。比如,当样品温育在pH5.0的介质中,铁氰化钾还原的Cyt b-559 HP含量占总量的25%-30%,当介质中加入2mmol/L碳酸氢盐后Cyt b-559 HP的含量上升,占总量的50%-56%。碳酸氢盐效应对氢醌还原的Cyt b-559HP含量的影响尤为显著。pH6.5时碳酸氢盐对Cyt b-559的还原氧化状态的影响不显著。其次,分别研究了PSII经Tris处理去除锰簇和三个外周蛋白及NH20H处理去除锰簇和17 kDa和23 kDa后,碳酸氢盐对Cytb一559 HP影响的pH依赖值,发现不论在pH5.0还是pH6.5的介质中碳酸氢盐效应都不存在。 综合以上实验结果,我们认为碳酸氢盐对酸化引起的Cyt b-559氧化还原状态的影响与它和锰的作用有关,但也不能排除钙离子与碳酸氢盐之间的协同作用。

Source of the Nile Fish farm (SON) is located at Bugungu area in Napoleon Gulf, northern Lake Victoria. The proprietors of the farm requested NaFIRRI to provide technical assistance to undertake regular environment monitoring of the cage site as is mandatory under the NEMA conditions. NAFIRRI agreed to undertake regular environment surveys in the cage area covering selected physical‐chemical factors i.e. water column depth, water transparency, water column temperature, dissolved oxygen, pH, conductivity, redox potential and turbidity; nutrient status, algal and invertebrate communities (micro‐invertebrates/zooplankton and macro‐invertebrates/macro‐benthos) as well as fish community. The first year‐round quarterly surveys were completed for the year 2011. It was decided by SON management to change the frequency of the monitoring surveys to biannual starting in the year 2012 and the first such survey, which is the subject of this report, was undertaken in June 2012. Results/observations made are presented in this technical report along with a scientific interpretation and discussion of the results with reference to possible impacts of the cage facilities to the water environment and aquatic biota. Physicochemical parameters of 31 fish pond water samples of Tripura were studied to ascertain the nutrient profile of acidic soil zone and the impact of water acidity towards aquaculture productivity. The pH was acidic (mean 6.63±0.44) with high Fe (mean1.04±0.40 mglˉ¹) and AI (mean 2.67±2.41 mglˉ¹) contents. These were mostly responsible for pond water acidity and poor productivity with low nitrogen, phosphate and total alkalinity. The study also showed strong negative relationship between water pH and redox potential (R²=0.5251). However, pH was positively significant with electrical conductivity. The roles of redox potential and electrical conductivity in water acidity were found highly important. Available calcium content was also found low (mean 2.91±2.96 mglˉ¹). Elevating level of pH of pond water could be the possible management practices in acidic water so that such unproductive water might be productive enough with higher phosphate and nitrogen levels for better biological production.

Electrical double-layer capacitors owe their large capacitance to the formation of a double-layer at the electrode/electrolyte interface of high surface area carbon-based electrode materials. Greater electrical energy storage capacity has been attributed to transition metal oxides/nitrides that undergo fast, reversible redox reactions at the electrode surface (pseudo-capacitive behavior) in addition to forming electrical double-layers. Solution Precursor Plasma Spray (SPPS) has shown promise for depositing porous, high surface area transition metal oxides. This investigation explored the potential of SPPS to fabricate a-MoO 3 coatings with micro-structures suitable for use as super-capacitor electrodes. The effects of number of spray passes, spray distance, solution concentration, flow rate and spray velocity on the chemistry and micro-structure of the a-MoO 3 deposits were examined. DTA/TGA, SEM, XRD, and electrochemical analyses were performed to characterize the coatings. The results demonstrate the importance of post-deposition heating of the deposit by subsequent passes of the plasma on the coating morphology. © ASM International.

Chemical looping combustion (CLC) is a novel combustion technology that involves cyclic reduction and oxidation of oxygen storage materials to provide oxygen for the combustion of fuels to CO2 and H2O, whilst giving a pure stream of CO2 suitable for sequestration or utilisation. Here, we report a method for preparing of oxygen storage materials from layered double hydroxides (LDHs) precursors and demonstrate their applications in the CLC process. The LDHs precursor enables homogeneous mixing of elements at the molecular level, giving a high degree of dispersion and high-loading of active metal oxide in the support after calcination. Using a Cu-Al LDH precursor as a prototype, we demonstrate that rational design of oxygen storage materials by material chemistry significantly improved the reactivity and stability in the high temperature redox cycles. We discovered that the presence of sodium-containing species were effective in inhibiting the formation of copper aluminates (CuAl2O4 or CuAlO 2) and stabilising the copper phase in an amorphous support over multiple redox cycles. A representative nanostructured Cu-based oxygen storage material derived from the LDH precursor showed stable gaseous O2 release capacity (∼5 wt%), stable oxygen storage capacity (∼12 wt%), and stable reaction rates during reversible phase changes between CuO-Cu 2O-Cu at high temperatures (800-1000 °C). We anticipate that the strategy can be extended to manufacture a variety of metal oxide composites for applications in novel high temperature looping cycles for clean energy production and CO2 capture. © The Royal Society of Chemistry 2013.

The construction of protein-based photoelectrochemical cells that produce a variety of alternating currents in response to discontinuous illumination is reported. The photovoltaic component is a protein complex from the purple photosynthetic bacterium Rhodobacter sphaeroides which catalyses photochemical charge separation with a high quantum yield. Photoelectrochemical cells formed from this protein, a mobile redox mediator and a counter electrode formed from cobalt disilicide, titanium nitride, platinum, or multi-walled carbon nanotubes (MWCNT) generate a direct current during continuous illumination and an alternating current with different characteristics during discontinuous illumination. In particular, the use of superhydrophobic MWCNT as the back electrode results in a near symmetrical forward and reverse current upon light on and light off, respectively. The symmetry of the AC output of these cells is correlated with the wettability of the counter electrode. Potential applications of a hybrid biological/synthetic solar cell capable of generating an approximately symmetrical alternating current are discussed. Copyright © 2013 WILEY-VCH Verlag GmbH & Co. Microorganisms play an important role in removing pollutants from constructed wetlands. We investigated the microbial characteristics in a novel integrated vertical-flow constructed wetland (IVCW), which has been in operation in Wuhan, China since 1998. We used phospholipid fatty acid (PLFA) and amoA gene to analyze the structure and diversity of the microbial community within the IVCW. PLFA results suggested that the amount of bacterial PLFA was significantly higher than that of fungal PLFA, but the total microbial biomass represented by PLFA index was low in the system. Microbial spatial distribution showed significantly higher bacterial (both G(+) and G(-)) and fungal biomass in the surface than in the subsurface layers. The ratios of monounsaturated to branched PLFA demonstrated that an anaerobic layer sandwiched by two aerobic layers existed in the IVCW, consistent with the redox potential results. Analysis of the amoA revealed the presence of Nitrosomonas-like sequences in the surface substrate of the downflow chamber and apparent diversities of ammonia-oxidizing bacteria in the system. These results suggest that microorganisms, despite their relatively low biomass, have inhabited the IVCW, and the results will offer some valuable information on microbe to system designers and managers.

A fluorescence immunoassay for human IgG (Ag) was developed using a pH-sensitive polymer prepared by thermal initiation or redox initiation polymerization as a carrier. In the competitive immunoassay, appropriate quantity of Ag was immobilized on the polymer and the standard Ag (or sample) solution, and a constant amount of fluorescein isothiocyanate labeled goat anti-human IgG antibody (Ab-FITC) was added. Immobilized Ag and the standard (or sample) Ag competed for binding to the Ab-FITC in 37 C in homogeneous format. After changing the pH to separate the polymer-immune complex precipitate, it was re-dissolved and determined by fluorescence method. The results showed that the immobilization efficiency, immunological reaction activities of immobilized Au and phase transition pH range were improved as Ag was immobilized by thermal initiation instead of redox initiation polymerization. Under optimum conditions, the calibration graphs for the Ag in both methods, thermal initiation and redox initiation, were linear over the concentration range of 0.0-1000 ng mL(-1), with detection limits 8 (thermal initiation) and 12 ng mL(1) (redox initiation), respectively. Moreover, some pH-sensitive polymer prepared only in organic solvent or under high temperature could also be used as an immunoreaction carrier by thermal initiation polymerization. Goal, Scope and Background. In some cases, soil, water and food are heavily polluted by heavy metals in China. To use plants to remediate heavy metal pollution would be an effective technique in pollution control. The accumulation of heavy metals in plants and the role of plants in removing pollutants should be understood in order to implement phytoremediation, which makes use of plants to extract, transfer and stabilize heavy metals from soil and water. Methods. The information has been compiled from Chinese publications stemming mostly from the last decade, to show the research results on heavy metals in plants and the role of plants in controlling heavy metal pollution, and to provide a general outlook of phytoremediation in China. Related references from scientific journals and university journals are searched and summarized in sections concerning the accumulation of heavy metals in plants, plants for heavy metal purification and phytoremediation techniques. Results and Discussion. Plants can take up heavy metals by their roots, or even via their stems and leaves, and accumulate them in their organs. Plants take up elements selectively. Accumulation and distribution of heavy metals in the plant depends on the plant species, element species, chemical and bioavailiability, redox, pH, cation exchange capacity, dissolved oxygen, temperature and secretion of roots. Plants are employed in the decontamination of heavy metals from polluted water and have demonstrated high performances in treating mineral tailing water and industrial effluents. The purification capacity of heavy metals by plants are affected by several factors, such as the concentration of the heavy metals, species of elements, plant species, exposure duration, temperature and pH. Conclusions. Phytoremediation, which makes use of vegetation to remove, detoxify, or stabilize persistent pollutants, is a green and environmentally-friendly tool for cleaning polluted soil and water. The advantage of high biomass productive and easy disposal makes plants most useful to remediate heavy metals on site. Recommendations and Outlook. Based on knowledge of the heavy metal accumulation in plants, it is possible to select those species of crops and pasturage herbs, which accumulate fewer heavy metals, for food cultivation and fodder for animals; and to select those hyperaccumulation species for extracting heavy metals from soil and water. Studies on the mechanisms and application of hyperaccumulation are necessary in China for developing phytoremediation.

A quantum chemistry based Green's function formulation of long-range charge transfer in deoxyribose nucleic acid (DNA) double helix is proposed. The theory takes into account the effects of DNA's electronic structure and its incoherent interaction with aqueous surroundings. In the implementation, the electronic tight-binding parameters for unsolvated DNA molecules are determined at the HF/6-31G* level, while those for individual nucleobase-water couplings are at a semiempirical level by fitting with experimental redox potentials. Numerical results include that: (i) the oxidative charge initially at the donor guanine site does hop sequentially over all guanine sites; however, the revealed rates can be of a much weaker distance dependence than that described by the ordinary Ohm's law; (ii) the aqueous surroundings-induced partial incoherences in thymine/adenine bridge bases lead them to deviate substantially from the superexchange regime; (iii) the time scale of the partially incoherent hole transport through the thymine/adenine pi stack in DNA is about 5 ps. Poly(3,4-ethylenedioxythiopliene):poly(styrene sulfonate) (PEDOT:PSS) films have been electrochemically polymerized in situ on ITO glass substrate in boron trifluoride diethyl etherate electrolyte (BFEE). Cyclic voltammograms show good redox activity and stability of the PEDOT films. These films had been directly used to fabricate organic-inorganic hybrid solar cells with the structure of ITO/PEDOT/ZnO:MDMC-PPV/Al. The solar cells made of electrochemically polymerized films exhibit higher energy conversion efficiencies compared with that prepared by the spin-coating method, and the highest value is 0.33%. This in-situ electropolymerized method effectively simplifies fabricating procedures and may blaze a facile and economical route for producing high-efficiency solar cells.

Autothermal reforming of methanol for hydrogen production was investigated over ZnO-ZnCr2O4 supported on a series of metal oxides (Al2O3, CeO2, ZrO2 and CeO2-ZrO2)CeO2-ZrO2 mixed oxides with Ce /Zr molar ratio of 4/1 was found to be the optimal support which showed significant effect on the catalytic activity and selectivity. The ZnO-ZnCr2O4/CeO2-ZrO2 and ZnO-ZnCr2O4 catalysts were characterized by XRD, TEM, H-2-TPR and XPS. The results show that CeO2-ZrO2 mixed oxides have significant effect on the catalytic performance and the supported catalyst shows more uniform temperature distribution in the catalyst bed which was mainly due to its reasonable redox properties.

本论文系统地研究了La_(1-x)Sr_xNiO_3, La_(1-1.333x)Th_xNiO_3,La_(2-x)Sr_xNiO_4, La_(2-x)Sr_xCuO_4 (0 ≤ x ≤ 1)四系列催化剂的固体化学性质和用于氨氧化制硝酸的催化活性之间的关系。为了更深入地探讨多元复合氧化物的固体化学性质与催化行为之间的关系,本论文还开展了对稀土A_2BO_4型复合氧化物:La_(2-x)Sr_xNiO_4和La_(2-x)Sr_xCuO_4 (0 ≤ x ≤ 1)两系列催化剂的固化性质与催化性能的研究。ABO_3, A_2BO_4两类复合氧化物用于氨氧化,都可能遵循氧化-还原(Redox)机理,高价B离子为主要活性离子,高温区脱出的氧可能为主要活性氧种。比较ABO_3和A_2BO_4两类复合氧化物,ABO_3能稳定较高含量的高价B离子,但结构不如A_2BO_4稳定。A_2BO_4结构稳定,能在较宽掺杂范围内保持结构不变,但高价B离子含量不如ABO_3中高。

CdS clusters in zeolite-Y have been prepared by the exchange of Cd2+ into the zeolite following by sulfurization with Na2S in solution. Blue-shifts from the bulk caused by quantum size effect and the change of absorption upon CdS loading are observed. Two absorption bands are detected for one of the sample and are assigned to the 1s-1s band and exciton transition, respectively. The exciton feature is more pronounced in the excitation spectrum than in the absorption spectrum, and the luminescence excited at the exciton band is stronger than that at the 1s-1s band.
