A cDNA clone encoding a thiol-protease (TPE4A) was isolated from senescent ovaries of pea (Pisum sativum) by reverse transcriptase-polymerase chain reaction. The deduced amino acid sequence of TPE4A has the conserved catalytic amino acids of papain. It is very similar to VSCYSPROA, a thiol-protease induced during seed germination in common vetch. TPE4A mRNA levels increase during the senescence of unpollinated pea ovaries and are totally suppressed by treatment with gibberellic acid. In situ hybridization indicated that TPE4A mRNA distribution in senescent pea ovaries is different from that of previously reported thiol-proteases induced during senescence, suggesting the involvement of different proteases in the mobilization of proteins from senescent pea ovaries. TPE4A is also induced during the germination of pea seeds, indicating that a single protease gene can be induced during two different physiological processes, senescence and germination, both of which require protein mobilization.

Low concentrations of the glucose (Glc) analog mannose (Man) inhibit germination of Arabidopsis seeds. Man is phosphorylated by hexokinase (HXK), but the absence of germination was not due to ATP or phosphate depletion. The addition of metabolizable sugars reversed the Man-mediated inhibition of germination. Carbohydrate-mediated regulation of gene expression involving a HXK-mediated pathway is known to be activated by Glc, Man, and other monosaccharides. Therefore, we investigated whether Man blocks germination through this system. By testing other Glc analogs, we found that 2-deoxyglucose, which, like Man, is phosphorylated by HXK, also blocked germination; no inhibition was observed with 6-deoxyglucose or 3-O-methylglucose, which are not substrates for HXK. Since these latter two sugars are taken up at a rate similar to that of Man, uptake is unlikely to be involved in the inhibition of germination. Furthermore, we show that mannoheptulose, a specific HXK inhibitor, restores germination of seeds grown in the presence of Man. We conclude that HXK is involved in the Man-mediated repression of germination of Arabidopsis seeds, possibly via energy depletion.

Expression of the alcohol dehydrogenase gene (ADH) of Arabidopsis is known to be induced by environmental stresses and regulated developmentally. We used a negative-selection approach to isolate mutants that were defective in regulating the expression of the ADH gene during seed germination; we then characterized three recessive mutants, aar1–1, aar1–2, and aar2–1, which belong to two complementation groups. In addition to their defects during seed germination, mutations in the AAR1 and AAR2 genes also affected anoxic and hypoxic induction of ADH and other glycolytic genes in mature plants. The aar1 and aar2 mutants were also defective in responding to cold and osmotic stress. The two allelic mutants aar1–1and aar1–2 exhibited different phenotypes under cold and osmotic stresses. Based on our results we propose that these mutants are defective in a late step of the signaling pathways that lead to increased expression of the ADH gene and glycolytic genes.

Germination of lettuce (Lactuca sativa L.) seed is regulated by phytochrome. The requirement for red light is circumvented by the application of gibberellin (GA). We have previously shown that the endogenous content of GA1, the main bioactive GA in lettuce seeds, increases after red-light treatment. To clarify which step of GA1 synthesis is regulated by phytochrome, cDNAs encoding GA 20-oxidases (Ls20ox1 and Ls20ox2, for L. sativa GA 20-oxidase) and 3β-hydroxylases (Ls3h1 and Ls3h2 for L. sativa GA 3β-hydroxylase) were isolated from lettuce seeds by reverse-transcription polymerase chain reaction. Functional analysis of recombinant proteins expressed in Escherichia coli confirmed that the Ls20ox and Ls3h encode GA 20-oxidases and 3β-hydroxylases, respectively. Northern-blot analysis showed that Ls3h1 expression was dramatically induced by red-light treatment within 2 h, and that this effect was canceled by a subsequent far-red-light treatment. Ls3h2 mRNA was not detected in seeds that had been allowed to imbibe under any light conditions. Expression of the two Ls20ox genes was induced by initial imbibition alone in the dark. The level of Ls20ox2 mRNA decreased after the red-light treatment, whereas that of Ls20ox1 was unaffected by light. These results suggest that red light promotes GA1 synthesis in lettuce seeds by inducing Ls3h1 expression via phytochrome action.

Plant growth and development are regulated by interactions between the environment and endogenous developmental programs. Of the various environmental factors controlling plant development, light plays an especially important role, in photosynthesis, in seasonal and diurnal time sensing, and as a cue for altering developmental pattern. Recently, several laboratories have devised a variety of genetic screens using Arabidopsis thaliana to dissect the signal transduction pathways of the various photoreceptor systems. Genetic analysis demonstrates that light responses are not simply endpoints of linear signal transduction pathways but are the result of the integration of information from a variety of photoreceptors through a complex network of interacting signaling components. These signaling components include the red/far-red light receptors, phytochromes, at least one blue light receptor, and negative regulatory genes (DET, COP, and FUS) that act downstream from the photoreceptors in the nucleus. In addition, a steroid hormone, brassinolide, also plays a role in light-regulated development and gene expression in Arabidopsis. These molecular and genetic data are allowing us to construct models of the mechanisms by which light controls development and gene expression in Arabidopsis. In the future, this knowledge can be used as a framework for understanding how all land plants respond to changes in their environment.

We have examined the seed germination in Arabidopsis thaliana of wild type (wt), and phytochrome A (PhyA)- and B (PhyB)-mutants in terms of incubation time and environmental light effects. Seed germination of the wt and PhyA-null mutant (phyA) was photoreversibly regulated by red and far-red lights of 10-1,000 micromol m-2 when incubated in darkness for 1-14 hr, but no germination occurred in PhyB-null mutant (phyB). When wt seeds and the phyB mutant seeds were incubated in darkness for 48 hr, they synthesized PhyA during dark incubation and germinated upon exposure to red light of 1-100 nmol m-2 and far-red light of 0.5-10 micromol m-2, whereas the phyA mutant showed no such response. The results indicate that the seed germination is regulated by PhyA and PhyB but not by other phytochromes, and the effects of PhyA and PhyB are separable in this assay. We determined action spectra separately for PhyA- and PhyB-specific induction of seed germination at Okazaki large spectrograph. Action spectra for the PhyA response show that monochromatic 300-780 nm lights of very low fluence induced the germination, and this induction was not photoreversible in the range examined. Action spectra for the PhyB response show that germination was photoreversibly regulated by alternate irradiations with light of 0.01-1 mmol m-2 at wavelengths of 540-690 nm and 695-780 nm. The present work clearly demonstrated that PhyA photoirreversibly triggers the germination upon irradiations with ultraviolet, visible and far-red light of very low fluence, while PhyB controls the photoreversible effects of low fluence.

The codon usage of a hybrid bacterial gene encoding a thermostable (1,3-1,4)-beta-glucanase was modified to match that of the barley (1,3-1,4)-beta-glucanase isoenzyme EII gene. Both the modified and unmodified bacterial genes were fused to a DNA segment encoding the barley high-pI alpha-amylase signal peptide downstream of the barley (1,3-1,4)-beta-glucanase isoenzyme EII gene promoter. When introduced into barley aleurone protoplasts, the bacterial gene with adapted codon usage directed synthesis of heat stable (1,3-1,4)-beta-glucanase, whereas activity of the heterologous enzyme was not detectable when protoplasts were transfected with the unmodified gene. In a different expression plasmid, the codon modified bacterial gene was cloned downstream of the barley high-pI alpha-amylase gene promoter and signal peptide coding region. This expression cassette was introduced into immature barley embryos together with plasmids carrying the bar and the uidA genes. Green, fertile plants were regenerated and approximately 75% of grains harvested from primary transformants synthesized thermostable (1,3-1,4)-beta-glucanase during germination. All three trans genes were detected in 17 progenies from a homozygous T1 plant.

The etiolated germination process of oilseed plants is characterized by the mobilization of storage lipids, which serve as a major carbon source for the seedling. We found that during early stages of germination in cucumber, a lipoxygenase (linoleate: oxygen oxidoreductase, EC 1.13.11.12) form is induced that is capable of oxygenating the esterified fatty acids located in the lipid-storage organelles, the so-called lipid bodies. Large amounts of esterified (13S)-hydroxy-(9Z,11E)-octadecadienoic acid were detected in the lipid bodies, whereas only traces of other oxygenated fatty acid isomers were found. This specific product pattern confirms the in vivo action of this lipoxygenase form during germination. Lipid fractionation studies of lipid bodies indicated the presence of lipoxygenase products both in the storage triacylglycerols and, to a higher extent, in the phospholipids surrounding the lipid stores as a monolayer. The degree of oxygenation of the storage lipids increased drastically during the time course of germination. We show that oxygenated fatty acids are preferentially cleaved from the lipid bodies and are subsequently released into the cytoplasm. We suggest that they may serve as substrate for beta-oxidation. These data suggest that during the etiolated germination, a lipoxygenase initiates the mobilization of storage lipids. The possible mechanisms of this implication are discussed.

Atualmente, a produtividade do feijoeiro comum (Phaseolus vulgaris L.) pode ser reduzida devido à ocorrência de doenças em todo o território nacional, destacando-se a murcha de fusário, causada por Fusarium oxysporum f. sp. phaseoli (Fop). No campo, o patógeno é disseminado a longas distâncias através das sementes infectadas e/ou contaminadas e a sua sobrevivência ocorre, principalmente, no solo. Os objetivos deste trabalho foram: avaliar a inibição do crescimento micelial de Fop por Trichoderma spp.; classificar a sensibilidade in vitro de Fop e Trichoderma spp., separadamente, a fungicidas e verificar a compatibilidade entre fungicidas químicos e biológicos para controle de Fop, presente nas sementes e no solo. Para avaliar a inibição do crescimento micelial de Fop, foram utilizados três isolados do patógeno, os quais foram confrontados, in vitro, com três isolados de Trichoderma spp. em testes de cultura pareada e produção de metabólitos voláteis a 20-22°C. Os experimentos foram conduzidos em delineamento inteiramente casualizado, com cinco repetições para cada isolado de Trichoderma. Para a classificação da sensibilidade in vitro de Fop e Trichoderma a fungicidas, foram avaliados os mesmos isolados anteriormente utilizados. Foram comparados dez fungicidas, em doses entre 0 a 100 mg L-1 que foram ajustadas de acordo com a CI50 de cada fungicida. Com base na percentagem de inibição do crescimento micelial, foram estimados os valores da concentração inibitória de 50% (CI50) e 100% (CI100) e selecionaram-se os fungicidas compatíveis com Trichoderma spp. A compatibilidade entre tratamentos químico e biológico foi avaliada através da inoculação artificial de sementes de feijão com um isolado de Fop (IAC 11.299-1) e infestação do mesmo no solo. As sementes foram tratadas com os fungicidas fludioxonil, flutriafol e tiofanato metílico, e com os três produtos biológicos, separadamente e em misturas. Avaliou-se o efeito dos tratamentos por meio dos testes de sanidade, germinação, comprimento de plântulas, massa da matéria seca em laboratório e índice de velocidade de emergência e porcentagem de emergência em estufa não climatizada. O efeito protetor dos tratamentos foi verificado através do teste de transmissão do patógeno solo-planta. Todos os isolados de Trichoderma apresentaram antagonismo in vitro contra Fop. No teste de cultura pareada foi observada uma redução de 15 a 20% no crescimento micelial do patógeno. No teste de produção de metabólitos voláteis, o isolado T12-1086G05 foi responsável pela maior inibição do crescimento micelial de Fop (10 a 48%). Os testes de sensibilidade in vitro mostraram que tiofanato metílico, flutriafol e fludioxonil foram compatíveis com Trichoderma (CI50 > 2 mg L-1). Com exceção do flutriafol e do GF 422 isolados e em mistura, todos os tratamentos foram eficientes na erradicação de Fop nas sementes, sem afetar a sua qualidade fisiológica. No teste de transmissão, verificou-se que a incidência de Fop foi de 5 a 40% no hipocótilo e de 5 a 30% nas raízes de feijoeiro provenientes de sementes tratadas com os produtos.

Resumo A presente dissertação visou avaliar o efeito da utilização do pato de Pequim no controlo de infestantes em milho biológico em comparação com o método clássico da sacha e da sacha/amontoa, avaliando a percentagem de cobertura das infestantes no solo e a produtividade do milho. Para além disso, pretendeu-se, ainda, perceber em que estádio vegetativo do milho os patos já não provocavam danos consideráveis na cultura. A investigação decorreu entre maio e outubro de 2015, em Coimbra, na superfície agrícola da ESAC, em 0,2 ha da área certificada em Agricultura Biológica, utilizando milho da variedade regional Pigarro. Os patos foram adquiridos com 4 semanas e abatidos às 12 semanas, tendo-se avaliado o seu crescimento nesse período. O pastoreio com patos, em diferentes estados vegetativos da cultura (Ve, V3 e V5 - germinação e emergência, três e cinco folhas completamente desenvolvidas, respetivamente), não permitiu afirmar com rigor qual o estádio vegetativo em que os animais já não provocavam danos. Analisando e comparando o pastoreio com patos após a 1ª sacha, após a sacha/amontoa e o método clássico, concluiu-se que tanto a cobertura do solo pelas infestantes quanto a produção de milho Pigarro (kg/ha) não variaram significativamente, para α = 0,05, após realização de análise de variância de fator único. Os resultados obtidos, embora preliminares e carecendo de continuidade de estudos, permitem, no entanto, indiciar que as opções ensaiadas podem vir a substituir o método clássico e serem alternativas válidas no combate às infestantes em milho biológico. PALAVRAS-CHAVE: Pato de Pequim; Milho biológico; Infestantes; Cobertura; Pastoreio. iii Abstract This dissertation aimed to evaluate the effect of using the Peking duck for weed control in organic corn compared to the classical method of hoeing and weeding / earthing up, assessing the percentage of coverage of weeds in soil and corn yield. Furthermore, it was also intended to realize in what corn's vegetative stage, ducks no longer caused considerable damage to the crop. The research took place between May and October 2015, in Coimbra, in the ESAC's agricultural area of 0.2 ha certified in organic farming, using corn's regional variety, Throat clearing. The ducks were purchased at 4 weeks old and slaughtered at the 12th week, while their growth in this period was estimated. The grazing with ducks in different vegetative states of culture (Ve, V3 and V5 - germination and emergence, three and five fully developed leaves, respectively), didn't allow us to say with accuracy in what vegetative stage the animals no longer caused damage. Analyzing and comparing grazing with ducks after 1st weeding, after weeding / ridging and the classical method, it was concluded that both the soil cover by weeds as the Throat clearing corn production (kg / ha) did not vary significantly, for α = 0.05, after performing single- factor analysis of variance. The results, although preliminary and lacking continuity studies allow, however, to indicate that the tested options may come to replace the traditional method and be valid alternatives to combat weeds in biological maize.
