Although silicon is not recognized as a nutrient, it may benefit rice plants and may alleviate the Mn toxicity in some plant species. The dry matter yield (root, leaf, sheaths and leaf blade) and plant architecture (angle of leaf insertion and leaf arc) were evaluated in rice plants grown in nutrient solutions with three Mn doses, with and without Si addition. The treatments were arranged in a 2 x 3 factorial [with and without (2 mmol L-1) Si; three Mn doses (0.5; 2.5 and 10 µmol L-1)], in a randomized block design with 4 replications. The experimental unit was a 4 L plastic vase with 4 rice (Metica-1 cultivar) plants. Thirty nine days after keeping the seedlings in the nutrient solution the plant dry matter yield was determined; the angle of leaf insertion in the sheath and the leaf arc were measured; and the Si and Mn concentrations in roots, sheaths and leaves were determined. The analysis of variance (F test at 5 and 1 % levels) and the regression analysis (for testing plant response to Mn with the Si treatments) were performed. The Si added to the nutrient solution increased the dry matter yield of roots, sheaths and leaf blades and also decreased the angle of leaf blade insertion into the sheath and the foliar arc in the rice plant. Additionally, it ameliorated the rice plant architecture which allowed an increase in the dry matter yield. Similarly, the addition of Mn to the solution improved the architecture of the rice plants with gain in dry matter yield. As Si was added to the nutrient solution, the concentration of Mn in leaves decreased and in roots increased thus alleviating the toxic effects of Mn on the plants.

Rapport de synthese :Comparaison des effets vasculaires et tubulaires rénaux de plusieurs antagonistes des récepteurs de |'angiotensine II en combinaison avec un diurétique thiazidique chez l'humainObjectif : Le but de ce travail était d'investiguer si les antagonistes des récepteurs AT1 de l'angiotensine II (ARA2) entraînent un blocage équivalent des récepteurs au niveau vasculaire et au niveau rénal, en particulier lorsque le système rénine- angiotensine est stimulé par l'administration d'un diurétique thiazidique. Méthode : trente volontaires masculins en bonne santé ont participé à cette étude randomisée, contrôlée, en simple insu. Nous avons mesuré les variations de pression artérielle, d'hémodynamique rénale ainsi que la réponse tubulaire rénale à une perfusion d'angiotensine II 3ng/kg/min administrée sur 1 heure. Ceci avant traitement puis après sept jours d'administration, 24 heures après la dernière dose de médicament. Nous avons comparé l'irbésartan 300 mg seul ou en association avec 12.5 ou 25 mg d'hydrochlorothiazide. (irbésartan 300/12.5 ; irbésartan 300/25). Nous avons également comparé les effets de l'irbésartan 300/25 au losartan 100 mg, au valsartan 160 mg ainsi qu'à l'olmésartan 20 mg, tous administrés avec 25 mg d'hydrochlorothiazide. Chaque participant a été randomisé pour recevoir 2 traitements de 7 jours espacés d'une période d'une semaine sans traitement. Résultats: La réponse de la pression artérielle à |'angiotensine II exogène était bloquée >90% avec l'irbésartan 300 mg seul ou en association avec le diurétique. Il en était de même avec l'olmésartan 20/25. Par contre le blocage n'était que de 60% environ dans les groupes valsartan 160/25 et losartan 100/25. Au niveau rénal, |'angiotensine II exogène réduisait le flux plasmatique rénal de 36% en pré- traitement. Dans les groupes recevant l'irbésartan 300 mg et l'olmésartan 20 mg associés à l'hydrochlorothiazide 25 mg, la vasoconstriction rénale était bloquée presque entièrement alors qu'el|e ne |'était que partiellement avec le valsartan 160/25 et le losartan 100/25 (34 et 45%, respectivement). En pré-traitement, au niveau tubulaire, l'angiotensine II exogène réduisait le volume urinaire de 84% et l'excrétion urinaire de sodium de 65 %. Les effets tubulaires n'étaient que partiellement bloqués par l'administration d'ARA2. Conclusion: Ces résultats démontrent que les ARA; aux doses maximales recommandées ne bloquent pas aussi efficacement les récepteurs ATI au niveau tubulaire qu'au niveau vasculaire. Cette observation pourrait constituer une justification à l'hypothèse selon laquelle des doses plus importantes d'ARA2 seraient nécessaires afin d'obtenir une meilleure protection d'organe. De plus, nos résultats confirment qu'i| y a d'importantes différences entre les ARA2, relatives à leur capacité d'induire un blocage prolongé sur 24 heures des récepteurs AT1 au niveau vasculaire et tubulaire.

The blood pressure, heart rate and humoral responses to single intravenous doses of the angiotensin converting enzyme inhibitor captopril were evaluated in 5 volunteers on a free salt intake. Each subject was given at one-week intervals a 1, 5 and 25 mg intravenous dose of captopril as well as the vehicle of captopril. The study was conducted in a single-blind fashion and the order of treatment phases was randomized. Captopril was found to inhibit the renin-angiotensin system in a dose-dependent fashion. A fall in circulating angiotensin II was observed with doses of 1 and 5 mg. Plasma angiotensin II was not detectable 15 min after the 25 mg dose. Acute inhibition of angiotensin converting enzyme with intravenous captopril had no effect on blood pressure and heart rate.

We developed a mathematical model of Ca transport along the late distal convoluted tubule (DCT2) and the connecting tubule (CNT) to investigate the mechanisms that regulate Ca reabsorption in the DCT2-CNT. The model accounts for apical Ca influx across transient receptor potential vanilloid 5 (TRPV5) channels and basolateral Ca efflux via plasma membrane Ca-ATPase pumps and type 1 Na/Ca exchangers (NCX1). Model simulations reproduce experimentally observed variations in Ca uptake as a function of extracellular pH, Na, and Mg concentration. Our results indicate that amiloride enhances Ca reabsorption in the DCT2-CNT predominantly by increasing the driving force across NCX1, thereby stimulating Ca efflux. They also suggest that because aldosterone upregulates both apical and basolateral Na transport pathways, it has a lesser impact on Ca reabsorption than amiloride. Conversely, the model predicts that full NCX1 inhibition and parathyroidectomy each augment the Ca load delivered to the collecting duct severalfold. In addition, our results suggest that regulation of TRPV5 activity by luminal pH has a small impact, per se, on transepithelial Ca fluxes; the reduction in Ca reabsorption induced by metabolic acidosis likely stems from decreases in TRPV5 expression. In contrast, elevations in luminal Ca are predicted to significantly decrease TRPV5 activity via the Ca-sensing receptor. Nevertheless, following the administration of furosemide, the calcium-sensing receptor-mediated increase in Ca reabsorption in the DCT2-CNT is calculated to be insufficient to prevent hypercalciuria. Altogether, our model predicts complex interactions between calcium and sodium reabsorption in the DCT2-CNT.

Soils of the coastal plains of Rio Grande do Sul, Brazil, are affected by salinization, which can hamper the establishment and development of crops in general, including rice. The application of high doses of KCl may aggravate the crop damage, due to the high saline content of this fertilizer. This study aimed to evaluate the effect of K fertilizer management on some properties of rice plant, grown in soils with different sodicity levels, and determine which attribute is best related to yield. The field study was conducted in four Albaqualfs with exchangeable Na percentages of 5.6, 9.0, 21 and 32 %. The management of KCl fertilizer consisted of the application of 90 kg ha-1 K2O broadcast, 90 kg ha-1 K2O in the row and 45 kg ha-1 K2O in the row + 45 kg ha-1 K2O at panicle initiation (PI). Plant density, dry matter evolution, height, SPAD (Soil Plant Analysis Development value indicating relative chlorophyll contents) index, tiller mass, 1,000-grain weight, panicle length and grain yield were evaluated. The plant density was damaged by application of K fertilizer in the row, especially at full dose (90 kg ha-1), at three sodicity levels, resulting in loss in biomass accumulation in later stages, affecting the crop yield, even at the lowest level of soil sodicity (5.6 %). All properties were correlated with yield; the highest positive correlation was found with plant density and shoot dry matter at full flowering, and a negative correlation with panicle length.

A cana-de-açúcar possui grande importância econômico-social e política para o Brasil. Este trabalho teve como objetivo avaliar o efeito de doses e fontes de manganês nessa cultura. O experimento foi realizado no sítio Fujimoto, área administrada pela Destilaria Vale do Paraná S/A Álcool e Açúcar, no município de Suzanápolis, SP. O delineamento experimental utilizado foi o de blocos ao acaso no esquema fatorial 5 x 3, sendo cinco doses de Mn (0; 2,5; 5,0; 7,5; e 10,0 kg ha-1) e três fontes (quelato, FTE e sulfato de manganês), aplicadas no sulco de plantio, em quatro repetições. As parcelas foram constituídas por quatro linhas de 5 m de comprimento e espaçadas 1,5 m. A variedade de cana-de-açúcar utilizada foi a RB 86-7515, realizando-se dois cortes. As fontes de Mn proporcionaram semelhantes produtividades de colmos, tanto da cana-planta quanto da primeira cana-soca. O quelato de Mn proporcionou maior número de colmo por metro de sulco na cana-soca. As doses de Mn não influenciaram a produtividade de colmos da cana-planta e da cana- soca, porém aumentaram o número de internódios e o diâmetro de colmo na cana-planta até as doses de 6,9 e 6,6 kg ha-1 de Mn, respectivamente.

Cellular directional migration in an electric field (galvanotaxis) is one of the mechanisms guiding cell movement in embryogenesis and in skin epidermal repair. The epithelial sodium channel (ENaC), in addition to its function of regulating sodium transport in kidney, has recently been found to modulate cell locomotory speed. Here we tested whether ENaC has an additional function of mediating the directional migration of galvanotaxis in keratinocytes. Genetic depletion of ENaC completely blocks only galvanotaxis and does not decrease migration speed. Overexpression of ENaC is sufficient to drive galvanotaxis in otherwise unresponsive cells. Pharmacologic blockade or maintenance of the open state of ENaC also decreases or increases, respectively, galvanotaxis, suggesting that the channel open state is responsible for the response. Stable lamellipodial extensions formed at the cathodal sides of wild-type cells at the start of galvanotaxis; these were absent in the ENaC knockout keratinocytes, suggesting that ENaC mediates galvanotaxis by generating stable lamellipodia that steer cell migration. We provide evidence that ENaC is required for directional migration of keratinocytes in an electric field, supporting a role for ENaC in skin wound healing.

Apesar da grande quantidade de N acumulada em plantações de eucalipto de alta produtividade, o aumento em volume do tronco em resposta à aplicação de N não tem sido expressivo nem consistente. O objetivo deste trabalho foi verificar o efeito de doses e fontes de N sobre o crescimento e o acúmulo de N em plantas de eucalipto, na serapilheira, além do impacto nas frações da matéria orgânica do solo (MOS). O experimento foi instalado em campo, no município de Itamarandiba-MG, em blocos ao acaso com três repetições, consistindo da aplicação em cobertura de doses (0, 60, 120 e 240 kg ha-1) e fontes de N distintas (sulfato de amônio e nitrato de amônio) em clone de eucalipto (AEC1528®). O efeito dos tratamentos sobre o crescimento e acúmulo de N nas plantas foi avaliado aos 30 meses de idade, abatendo-se árvores com DAP médio e separando-as em lenho, casca, galhos e folhas, para determinação da produção de matéria seca e dos teores e conteúdos de nutrientes das plantas. Amostras de solo e de serapilheira foram coletadas para análises de nutrientes. Os teores de C e N total da matéria orgânica particulada (MOP) e da matéria orgânica associada à fração mineral (MOAM) foram determinados por espectrometria de massa de razão isotópica, após separação física da MOS. As análises estatísticas consistiram de análise de variância e de regressão. A aplicação de adubos nitrogenados promoveu aumento no crescimento volumétrico do tronco e na matéria seca da parte aérea. A dose de N como sulfato de amônio para obter 90 % da produção máxima foi de 74 kg ha-1, a qual resultou em incremento de 42,3 % no volume de tronco em relação à testemunha sem adubação nitrogenada. Na dose de 120 kg ha-1 de N, não houve diferença de resposta à aplicação de sulfato de amônio e nitrato de amônio. Não foram detectadas alterações nos estoques de C e N da MOS com a adubação nitrogenada. No entanto, houve aumento da absorção de Ca, Mg e S. A taxa de recuperação aparente de N no campo foi maior na dose de 120 kg ha-1 de N, atingindo 34,4 %.

BACKGROUND: The SCN5A gene encodes for the α-subunit of the cardiac sodium channel NaV1.5, which is responsible for the rapid upstroke of the cardiac action potential. Mutations in this gene may lead to multiple life-threatening disorders of cardiac rhythm or are linked to structural cardiac defects. Here, we characterized a large family with a mutation in SCN5A presenting with an atrioventricular conduction disease and absence of Brugada syndrome. METHOD AND RESULTS: In a large family with a high incidence of sudden cardiac deaths, a heterozygous SCN5A mutation (p.1493delK) with an autosomal dominant inheritance has been identified. Mutation carriers were devoid of any cardiac structural changes. Typical ECG findings were an increased P-wave duration, an AV-block I° and a prolonged QRS duration with an intraventricular conduction delay and no signs for Brugada syndrome. HEK293 cells transfected with 1493delK showed strongly (5-fold) reduced Na(+) currents with altered inactivation kinetics compared to wild-type channels. Immunocytochemical staining demonstrated strongly decreased expression of SCN5A 1493delK in the sarcolemma consistent with an intracellular trafficking defect and thereby a loss-of-function. In addition, SCN5A 1493delK channels that reached cell membrane showed gain-of-function aspects (slowing of the fast inactivation, reduction in the relative fraction of channels that fast inactivate, hastening of the recovery from inactivation). CONCLUSION: In a large family, congregation of a heterozygous SCN5A gene mutation (p.1493delK) predisposes for conduction slowing without evidence for Brugada syndrome due to a predominantly trafficking defect that reduces Na(+) current and depolarization force.
Resposta do abacaxizeiro 'Vitória' a doses de nitrogênio em solos de tabuleiros costeiros da Paraíba

Apesar de importantes, são bastante escassas as informações sobre adubação e nutrição mineral de cultivares de abacaxi resistentes à fusariose. O objetivo deste trabalho foi avaliar a resposta do abacaxizeiro 'Vitória', em termos de nutrição mineral, produção e qualidade, à aplicação de doses crescentes de N, em Espodossolo Humilúvico da região de Tabuleiros Costeiros, Estado da Paraíba. O experimento foi conduzido em delineamento experimental de blocos casualizados, com cinco tratamentos, correspondente às doses de 100, 200, 300, 500 e 600 kg ha-1 de N (ureia aos 60, 180 e 270 dias após o plantio), e três repetições. Avaliaram-se o comprimento, a matéria fresca da folha 'D' e os teores de N, P e K nas porções clorofilada e aclorofilada aos 300 e 420 dap; e peso médio, produtividade, atributos físicos (peso, comprimento e diâmetro mediano de infrutescências; peso e comprimento de coroa) e químicos (pH, sólidos solúveis - SS, acidez titulável - AT e relação SS/AT) de infrutescências na colheita. A elevação da dose de N aumentou linearmente os valores de comprimento e matéria fresca da folha 'D'. O aumento das doses promoveu também elevação dos teores de N e diminuição dos de P e K da porção clorofilada aos 300 dap, sem alterar, entretanto, os teores desses nutrientes aos 420 dap. Na porção aclorofilada, a elevação das doses de N aumentou os teores de K aos 320 dap, mas diminuiu-os aos 420 dap. Os valores de peso de infrutescências e de produtividade aumentaram com a elevação das doses de N, estimando-se valores máximos de 1,0 kg e 37,9 t ha-1, com doses de 409,0 e 439,0 kg ha-1 de N, respectivamente. As doses de N não alteraram os atributos de qualidade das infrutescências, as quais se mostraram compatíveis com as características descritas para a cultivar.

Neuropeptide Y (NPY) is known to potentiate the pressor effect of norepinephrine. In the present work, we evaluated in unanesthetized normotensive rats the effect of NPY on blood pressure responsiveness not only to norepinephrine, but also to tyramine, a sympathomimetic agent acting indirectly to B-HT933, a selective alpha-2 adrenoceptor stimulant, to angiotensin II and vasopressin. Dose-response curves to the various pressor agents were established starting at the 45th min of an i.v. infusion with either NPY (0.025 and 0.1 microgram/min) or its vehicle. The two doses of NPY increased blood pressure by an average of approximately 6 mm Hg, which was not significantly different from the vehicle-induced blood pressure changes. NPY significantly enhanced the pressor effect of norepinephrine, tyramine and angiotensin II, but not that of B-HT933 and vasopressin. We also tested whether NPY inhibits the enzyme activity of Na, K-adenosine triphosphatase using a purified toad kidney preparation. Concentrations of NPY from 10(-14) M up to 10(-6) M had no effect on the enzyme activity. It appears therefore that the blood pressure potentiating effect of NPY is not restricted to alpha adrenoceptor stimulation with norepinephrine, but involves also the vasoconstrictor hormone angiotensin II. This NPY-induced potentiation does not seem to depend upon stimulation of alpha-2 adrenoceptors or inhibition of Na,K-adenosine triphosphatase.

We evaluated the effectiveness of supplementation with high dose of oral vitamin D3 to correct vitamin D insufficiency. We have shown that one or two oral bolus of 300,000 IU of vitamin D3 can correct vitamin D insufficiency in 50% of patients and that the patients who benefited more from supplementation were those with the lowest baseline levels. INTRODUCTION: Adherence with daily oral supplements of vitamin D3 is suboptimal. We evaluated the effectiveness of a single high dose of oral vitamin D3 (300,000 IU) to correct vitamin D insufficiency in a rheumatologic population. METHODS: Over 1 month, 292 patients had levels of 25-OH vitamin D determined. Results were classified as: deficiency <10 ng/ml, insufficiency ≥10 to 30 ng/ml, and normal ≥30 ng/ml. We added a category using the IOM recommended cut-off of 20 ng/ml. Patients with deficient or normal levels were excluded, as well as patients already supplemented with vitamin D3. Selected patients (141) with vitamin D insufficiency (18.5 ng/ml (10.2-29.1) received a prescription for 300,000 IU of oral vitamin D3 and were asked to return after 3 (M3) and 6 months (M6). Patients still insufficient at M3 received a second prescription for 300,000 IU of oral vitamin D3. Relation between changes in 25-OH vitamin D between M3 and M0 and baseline values were assessed. RESULTS: Patients (124) had a blood test at M3. Two (2%) had deficiency (8.1 ng/ml (7.5-8.7)) and 50 (40%) normal results (36.7 ng/ml (30.5-5.5)). Seventy-two (58%) were insufficient (23.6 ng/ml (13.8-29.8)) and received a second prescription for 300,000 IU of oral vitamin D3. Of the 50/124 patients who had normal results at M3 and did not receive a second prescription, 36 (72%) had a test at M6. Seventeen (47%) had normal results (34.8 ng/ml (30.3-42.8)) and 19 (53%) were insufficient (25.6 ng/ml (15.2-29.9)). Of the 72/124 patients who receive a second prescription, 54 (75%) had a test at M6. Twenty-eight (52%) had insufficiency (23.2 ng/ml (12.8-28.7)) and 26 (48%) had normal results (33.8 ng/ml (30.0-43.7)). At M3, 84% patients achieved a 25-OH vitamin D level >20 ng/ml. The lowest the baseline value, the highest the change after 3 months (negative relation with a correlation coefficient r = -0.3, p = 0.0007). CONCLUSIONS: We have shown that one or two oral bolus of 300,000 IU of vitamin D3 can correct vitamin D insufficiency in 50% of patients.

Nos preparos conservacionistas de solo, a eficácia dos resíduos culturais na redução da erosão hídrica diminui se os resíduos forem transportados pela enxurrada e, ou, se esta sulcar o solo. A esse fenômeno denomina-se falha dos resíduos, caracterizando a existência de comprimento crítico de declive, ou seja, estabelece a distância no terreno a partir da qual a enxurrada adquire tensão cisalhante suficiente para superar a resistência do resíduo ao transporte e, ou, a resistência do solo ao sulcamento. O objetivo deste trabalho foi averiguar a existência de comprimentos críticos de declive em duas direções de semeadura direta, sob diferentes tipos e doses de resíduos culturais. O trabalho foi desenvolvido em campo, entre maio de 2009 e maio de 2011, sobre um Nitossolo Bruno aluminoférrico húmico, em São José do Cerrito (SC), usando unidades experimentais com dimensões de 3,5 x 11 m, com declividade média de 0,144 m m-1. Foram testadas as formas de execução de semeadura no sentido transversal (em contorno) e paralelo ao declive (na pendente), sobre os resíduos culturais de milho (Zea mays), trigo (Triticum aestivum) e soja (Glycine max), nas doses de 50 e 100 % dos resíduos produzidos. Foram realizados testes de chuva simulada, com o uso de simulador de chuva de braços rotativos, na intensidade de chuva planejada de 65 mm h-1 e com duração de 90 min cada um - tempo esse necessário para se obter a taxa constante de enxurrada. Ao final dos 90 min de chuva, sem desligar o aparelho simulador de chuva, foram aplicados níveis crescentes de fluxo extra de água limpa na cabeceira das parcelas, simulando a enxurrada provinda de declives mais longos. Para identificação da falha dos resíduos, utilizou-se a teoria de erosão, efetuando as relações entre taxa de erosão (Dp) x taxa de enxurrada (q m) e concentração de sedimentos na enxurrada (C) x taxa de enxurrada (q m). Detectou-se comprimento crítico de declive em todos os tratamentos; de modo geral, a semeadura direta em contorno e a dose maior dos resíduos de milho, trigo e soja aumentaram o comprimento crítico de declive em relação à semeadura direta na direção da pendente e à menor dose de resíduos, respectivamente. Esses comprimentos variaram de 35 a 155 m. Da teoria de erosão, o ponto de falha foi mais bem identificado analisando conjuntamente as relações Dp x q m e C x q m.
