Pore-forming toxins are known for their ability to efficiently form transmembrane pores which eventually leads to cell lysis. The dynamics of lysis and underlying self-assembly or oligomerization pathways leading to pore formation are incompletely understood. In this manuscript the pore-forming kinetics and lysis dynamics of Cytolysin-A (ClyA) toxins on red blood cells (RBCs) are quantified and compared with experimental lysis data. Lysis experiments are carried out on a fixed mass of RBCs, under isotonic conditions in phosphate-buffered saline, for different initial toxin concentrations ranging from 2.94-14.7 nM. Kinetic models which account for monomer binding, conformation and oligomerization to form the dodecameric ClyA pore complex are developed and lysis is assumed to occur when the number of pores per RBC (n(p)) exceeds a critical number, n(pc). By analysing the model in a sublytic regime (n(p) < n(pc)) the number of pores per RBC to initiate lysis is found to lie between 392 and 768 for the sequential oligomerization mechanism and between 5300 and 6300 for the non-sequential mechanism. Rupture rates which are first order in the number of RBCs are seen to provide the best agreement with the lysis experiments. The time constants for pore formation are estimated to lie between 1 and 20 s and monomer conformation time scales were found to be 2-4 times greater than the oligomerization times. Cell rupture takes places in 100s of seconds, and occurs predominantly with a steady number of pores ranging from 515 to 11 000 on the RBC surface for the sequential mechanism. Both the sequential irreversible and non-sequential kinetics provide similar predictions of the hemoglobin release dynamics, however the hemoglobin released as a function of the toxin concentration was accurately captured only with the sequential model. Each mechanism develops a distinct distribution of mers on the surface, providing a unique experimentally observable fingerprint to identify the underlying oligomerization pathways. Our study offers a method to quantify the extent and dynamics of lysis which is an important aspect of developing novel drug and gene delivery strategies based on pore-forming toxins.

This study considers linear filtering methods for minimising the end-to-end average distortion of a fixed-rate source quantisation system. For the source encoder, both scalar and vector quantisation are considered. The codebook index output by the encoder is sent over a noisy discrete memoryless channel whose statistics could be unknown at the transmitter. At the receiver, the code vector corresponding to the received index is passed through a linear receive filter, whose output is an estimate of the source instantiation. Under this setup, an approximate expression for the average weighted mean-square error (WMSE) between the source instantiation and the reconstructed vector at the receiver is derived using high-resolution quantisation theory. Also, a closed-form expression for the linear receive filter that minimises the approximate average WMSE is derived. The generality of framework developed is further demonstrated by theoretically analysing the performance of other adaptation techniques that can be employed when the channel statistics are available at the transmitter also, such as joint transmit-receive linear filtering and codebook scaling. Monte Carlo simulation results validate the theoretical expressions, and illustrate the improvement in the average distortion that can be obtained using linear filtering techniques.

Metabolism is a defining feature of life, and its study is important to understand how a cell works, alterations that lead to disease and for applications in drug discovery. From a systems perspective, metabolism can be represented as a network that captures all the metabolites as nodes and the inter-conversions among pairs of them as edges. Such an abstraction enables the networks to be studied by applying graph theory, particularly, to infer the flow of chemical information in the networks by identifying relevant metabolic pathways. In this study, different weighting schemes are used to illustrate that appropriately weighted networks can capture the quantitative cellular dynamics quite accurately. Thus, the networks now combine the elegance and simplicity of representation of the system and ease of analysing metabolic graphs. Metabolic routes or paths determined by this therefore are likely to be more biologically meaningful. The usefulness of the approach is demonstrated with two examples, first for understanding bacterial stress response and second for studying metabolic alterations that occurs in cancer cells.

Thrust-generating flapping foils are known to produce jets inclined to the free stream at high Strouhal numbers St = fA/U-infinity, where f is the frequency and A is the amplitude of flapping and U-infinity is the free-stream velocity. Our experiments, in the limiting case of St —> infinity (zero free-stream speed), show that a purely oscillatory pitching motion of a chordwise flexible foil produces a coherent jet composed of a reverse Benard-Karman vortex street along the centreline, albeit over a specific range of effective flap stiffnesses. We obtain flexibility by attaching a thin flap to the trailing edge of a rigid NACA0015 foil; length of flap is 0.79 c where c is rigid foil chord length. It is the time-varying deflections of the flexible flap that suppress the meandering found in the jets produced by a pitching rigid foil for zero free-stream condition. Recent experiments (Marais et al., J. Fluid Mech., vol. 710, 2012, p. 659) have also shown that the flexibility increases the St at which non-deflected jets are obtained. Analysing the near-wake vortex dynamics from flow visualization and particle image velocimetry (PIV) measurements, we identify the mechanisms by which flexibility suppresses jet deflection and meandering. A convenient characterization of flap deformation, caused by fluid-flap interaction, is through a non-dimensional effective stiffness', EI* = 8 EI/(rho V-TEmax(2) s(f) c(f)(3)/2), representing the inverse of the flap deflection due to the fluid-dynamic loading; here, EI is the bending stiffness of flap, rho is fluid density, V-TEmax is the maximum velocity of rigid foil trailing edge, s(f) is span and c(f) is chord length of the flexible flap. By varying the amplitude and frequency of pitching, we obtain a variation in EI* over nearly two orders of magnitude and show that only moderate EI*. (0.1 less than or similar to EI * less than or similar to 1 generates a sustained, coherent, orderly jet. Relatively `stiff' flaps (EI* greater than or similar to 1), including the extreme case of no flap, produce meandering jets, whereas highly `flexible' flaps (EI* less than or similar to 0.1) produce spread-out jets. Obtained from the measured mean velocity fields, we present values of thrust coefficients for the cases for which orderly jets are observed.

Ho3+ (0.25-7 mol%) doped Sr2CeO4 nanophosphors were synthesized by solution combustion method using urea as fuel. The structural properties of the nanophosphors were investigated by powder X-ray diffraction studies (PXRD), scanning electron microscopy (SEM) and transmission electron microscopy (TEM) techniques. UV-Visible and photoluminescence (PL) spectroscopic techniques were used for analysing the optical properties of the nanoparticles. PXRD and TEM results revealed the formation of Sr2CeO4: Ho3+ nanocrystalline particles with orthorhombic crystal structure. From the UV-Vis studies the optical band gap energy found to decrease from 5.9 to 5.74 eV with increase in dopant concentration. The PL spectra exhibit the broad excitation band from 200 to 400 nm which concurs well with the commercial near UV LED. The PL spectra vary with the dopant content due to energy transfer from the host to the activator. In this present work we demonstrate that color tuning of phosphor can be achieved by merely varying the Ho3+ ions concentration. The CIE and CCT chromaticity coordinates suggests Sr2CeO4: Ho3+ nanophosphors may be potentially applicable as promising single - phased phosphors for lighting applications. (C) 2015 Elsevier B.V. Plants, herbivores and parasitoids affect each other directly and indirectly; however, feedback effects mediated by host plant traits have rarely been demonstrated in these tritrophic interactions. Brood-site pollination mutualisms (e.g. those involving figs and fig wasps) represent specialised tritrophic communities where the progeny of mutualistic pollinators and of non-mutualistic gallers (both herbivores) together with that of their parasitoids develop within enclosed inflorescences called syconia (hence termed brood-sites or microcosms). Plant reproductive phenology (which affects temporal brood-site availability) and inflorescence size (representing brood-site size) are plant traits that could affect reproductive resources, and hence relationships between trees, pollinators and non-pollinating wasps. Analysing wasp and seed contents of syconia, we examined direct, indirect, trophic and non-trophic relationships within the interaction web of the fig-fig wasp community of Ficus racemosa in the context of brood site size and availability. We demonstrate that in addition to direct resource competition and predator-prey (host-parasitoid) interactions, these communities display exploitative or apparent competition and trait-mediated indirect interactions. Inflorescence size and plant reproductive phenology impacted plant-herbivore and plant-parasitoid associations. These plant traits also influenced herbivore-herbivore and herbivore-parasitoid relationships via indirect effects. Most importantly, we found a reciprocal effect between within-tree reproductive asynchrony and fig wasp progeny abundances per syconium that drives a positive feedback cycle within the system. The impact of a multitrophic feedback cycle within a community built around a mutualistic core highlights the need for a holistic view of plant-herbivore-parasitoid interactions in the community ecology of mutualisms.

Homogeneous temperature regions are necessary for use in hydrometeorological studies. The regions are often delineated by analysing statistics derived from time series of maximum, minimum or mean temperature, rather than attributes influencing temperature. This practice cannot yield meaningful regions in data-sparse areas. Further, independent validation of the delineated regions for homogeneity in temperature is not possible, as temperature records form the basis to arrive at the regions. To address these issues, a two-stage clustering approach is proposed in this study to delineate homogeneous temperature regions. First stage of the approach involves (1) determining correlation structure between observed temperature over the study area and possible predictors (large-scale atmospheric variables) influencing the temperature and (2) using the correlation structure as the basis to delineate sites in the study area into clusters. Second stage of the approach involves analysis on each of the clusters to (1) identify potential predictors (large-scale atmospheric variables) influencing temperature at sites in the cluster and (2) partition the cluster into homogeneous fuzzy temperature regions using the identified potential predictors. Application of the proposed approach to India yielded 28 homogeneous regions that were demonstrated to be effective when compared to an alternate set of 6 regions that were previously delineated over the study area. Intersite cross-correlations of monthly maximum and minimum temperatures in the existing regions were found to be weak and negative for several months, which is undesirable. This problem was not found in the case of regions delineated using the proposed approach. Utility of the proposed regions in arriving at estimates of potential evapotranspiration for ungauged locations in the study area is demonstrated.

The proposed work discusses different parameters which are considered to improve the performance of a tin oxide-based thin film gas sensor. This includes analysing and deducing suitable catalytic additives to enhance the performance of the sensor in terms of selectivity and sensitivity. Chemical sensitization and electronic sensitization are performed to improve the rate of response of the sensor.

We prepose a 5-bit lattice Boltzmann model for KdV equation. Using Chapman-Enskog expansion and multiscale technique, we obtained high order moments of equilibrium distribution function, and the 3rd dispersion coefficient and 4th order viscosity. The parameters of this scheme can be determined by analysing the energy dissipation.

Approximate Bayesian computation (ABC) is a popular technique for analysing data for complex models where the likelihood function is intractable. It involves using simulation from the model to approximate the likelihood, with this approximate likelihood then being used to construct an approximate posterior. In this paper, we consider methods that estimate the parameters by maximizing the approximate likelihood used in ABC. We give a theoretical analysis of the asymptotic properties of the resulting estimator. In particular, we derive results analogous to those of consistency and asymptotic normality for standard maximum likelihood estimation. We also discuss how sequential Monte Carlo methods provide a natural method for implementing our likelihood-based ABC procedures.

Resumen: En este trabajo se estudia la fi gura del judío como posible inspirador de miedo para la sociedad hispanocristiana medieval, lo que parece estar en íntima relación con la imagen que de él se conformó a lo largo de la Edad Media en los derechos civil y canónico. Al analizar las distintas causas de temor inducidas por los judíos, se distingue entre los factores que causaban miedo físico, que muy probablemente tenían un componente más popular e irracional, aun cuando con frecuencia pudieran ser temores inducidos desde medios intelectuales, y los factores generadores de miedo espiritual, de naturaleza más intelectual y racional.

La Guerra de la Independencia, según la denominación tradicional de la historiografía española o The Peninsular Wars, como se suele denominar ese acontecimiento por los historiadores de habla inglesa, se perfila como uno de esos momentos en que las relaciones hispano-británicas adquieren una intensidad especial. De esa colaboración, de ese acto de comunicación intensa entre combatientes de un mismo enemigo, surgen unas manifestaciones literarias y muchos más relatos y diarios escritos por oficiales e incluso simples soldados del ejército de su majestad que contienen todo un tesoro de anécdotas, descripciones, y, principalmente, valoraciones dignas de analizar y hasta de disfrutar. Se trata, en este caso, de una recopilación de vivencias muy personales, en la mayoría de las ocasiones muy dolorosas, como cabría esperar de una guerra, y que a menudo ofrecen juicios y opiniones de España y Portugal que revelan una actitud paternalista y crítica.

The formation of shear bands in plane sheet is studied, both analytically and experimentally, to enhance the fundamental understanding of this phenomenon and to develop a capability for predicting material failure. The evolution of voids is measured and its interaction with the process of shear banding is examined. The evolving dilatancy in plasticity is shown to have a vital role in analysing the shear-band type of bifurcation, and tremendously reduces the theoretical value of critical stresses. The analyses, referring to both localized and diffuse modes of bifurcation, fairly explain the corresponding observations obtained through testing a dual-phase steer sheet and provide a justification of the constitutive model used.

This paper is concerned with some mathematical aspects of the Van Dyke method inperturbation theory, i.e. the singularity criteria of perturbation series. The author suggestsa sign criterion and a Domb-syke plot for the cases with complex conjugate singularities, thussucceeding in extending the conclusions of Van Dyke's. Subsequently. effects of singularitiesof the lower order upon the criteria are taken into account. In addition, a method of locat-ing singular points is developed by analysing the new perturbation series derived by the Eulertransformation.
