The majority of patients with chronic hepatitis C fail to respond to antiviral therapy. The genetic basis of this resistance is unknown. The quasispecies nature of HCV may have an important implication concerning viral persistence and response to therapy. The HCV nonstructural 5A (NS5A) protein has been controversially implicated in the inherent resistance of HCV to interferon (IFN) antiviral therapy. To evaluate whether the NS5A quasispecies pre-treatment composition of HCV 1a/1b is related to responsiveness to combined pegylated interferon (PEG-IFN) and Ribavirin therapy, detailed analyses of the complete NS5A were performed. Fifteen full-length NS5A clones were sequenced from 11 pretreatment samples of patients infected with genotype 1 HCV (3 virological sustained responders, 4 non-responders, and 4 end-of-treatment responders). Our study could not show a significant correlation between the mean number of mutations in HCV NS5A before treatment and treatment outcome, and the phylogenetic construction of complete NS5A sequences obtained from all patients failed to show any clustering associated with a specific response pattern. No single amino acid position was associated with different responses to therapy in any of the NS5A regions analyzed, and mutations were clustered downstream the ISDR, primarily in the V3 region. We observed that the CRS and NLS regions of the NS5A protein were conflicting for some variables analyzed, although no significant differences were found. If these two regions can have antagonistic functions, it seems viable that they present different mutation profiles when compared with treatment response. The patient sample that presented the lowest genetic distance values also presented the smallest number of variants, and the most heterogeneous pattern was seen in the end-of-treatment patients. These results suggest that a detailed molecular analysis of the NS5A region on a larger sample size may be necessary for understanding its role in the therapy outcome of HCV 1a/1b infection. O teste de condutividade elétrica tem sido excelente ferramenta para avaliar a qualidade de sementes de diversas espécies e, mais recentemente, estudos têm sido conduzidos visando verificar sua aplicabilidade em sementes florestais. Assim este trabalho objetivou estabelecer metodologia específica do teste de condutividade elétrica para sementes de branquilho. Foram usados três lotes de sementes resultantes do armazenamento em embalagens de vidro (Lote I), pano (Lote II) e papel (Lote III), em câmara fria (10ºC e 60% UR), por cinco meses, os quais foram submetidos ao teste de germinação, avaliando-se a porcentagem e o índice de velocidade de germinação. No teste de condutividade elétrica foram estudadas três quantidades de sementes por repetição (25, 50 e 75 sementes), três volumes de água deionizada (50, 75 e 100mL) e 11 tempos de embebição das sementes (2, 4, 6, 12, 18, 24, 30, 36, 42, 48 e 72h), a 25ºC. O lote I apresentou maior porcentagem e velocidade de germinação. O teste de condutividade elétrica possibilitou a mesma discriminação dos lotes que o teste de germinação, em combinações variadas dos fatores estudados. Foi possível separar o lote de melhor qualidade fisiológica (Lote I) dos demais lotes, já a partir de 2 horas de embebição, utilizando-se amostras de 75 sementes embebidas em 50, 75 ou 100mL de água; ou amostras de 50 sementes embebidas em 50mL de água, a partir de 18 horas de embebição. Desta forma, pode-se recomendar o uso de 75 sementes, embebidas em 75mL de água, por 24 horas à temperatura de 25ºC, para a condução do teste de condutividade elétrica.

Com o objetivo de verificar a existência de variabilidade temporal e espacial do tamanho de amostra da temperatura mínima do ar média mensal de trinta e sete municípios do Rio Grande do Sul, utilizaram-se os dados de temperatura mínima do ar do período de 1931 a 2000. Determinou-se o tamanho de amostra da temperatura mínima do ar média mensal em cada mês e município. Realizou-se análise de agrupamento dos meses e dos municípios pelo método hierárquico vizinho mais distante. Há variabilidade do tamanho de amostra (número de anos) para a estimativa da temperatura mínima do ar média mensal no Estado do Rio Grande do Sul no tempo e no espaço. Maior tamanho de amostra, no Estado do Rio Grande do Sul, é necessário nos meses de maio, junho e julho, com diminuição gradativa em direção a janeiro e dezembro. Há variabilidade do tamanho de amostra entre os municípios do Estado do Rio Grande do Sul.

Com o objetivo de verificar a variabilidade temporal e espacial do tamanho de amostra da radiação solar global média decendial, de 22 locais do Estado do Rio Grande do Sul, utilizaram-se séries de dados de radiação solar global do período de 1956 a 2003. Determinou-se o tamanho de amostra da radiação solar global média decendial em cada decêndio e local e agruparam-se os decêndios e os locais pelo método hierárquico 'vizinho mais distante'. Há variabilidade do tamanho de amostra (número de anos) para a estimativa da radiação solar global média decendial no Estado do Rio Grande do Sul no tempo e no espaço. Maior tamanho é necessário nos decêndios dos meses de junho, julho, agosto e setembro em relação aos outros meses. Para os locais e decêndios estudados, 30 anos de observações são suficientes para estimar a média (µ) de radiação solar global média decendial, para um erro de estimação igual a 12.3%, com coeficiente de confiança de 95%.

Os levantamentos epidemiológicos são importantes para o conhecimento da prevalência e tipologia das doenças bucais, podendo-se a partir dos dados coletados, planejar, executar e avaliar ações de saúde. É necessário, no entanto, que haja rigor metodológico que garanta reprodutibilidade, validade e confiabilidade, e que haja uniformidade de procedimentos para permitir comparações nacionais e internacionais. A iniciativa da Organização Mundial da Saúde (OMS) na assessoria à realização de levantamentos tem se mostrado extremamente útil, estimulando o estabelecimento de padrões de procedimentos que podem ser utilizados em todos os países. em 1991 foi publicada a edição em português da terceira edição do Oral Health Surveys - basic methods, de 1987, um manual que objetiva fornecer instruções para a realização de levantamentos epidemiológicos e tal publicação passou a servir de base a estudos realizados em diversos locais do Brasil e do mundo. O objetivo deste trabalho, é analisar criticamente a metodologia para Levantamento Epidemiológico em Saúde Bucal da OMS, na tentativa de contribuir para o aperfeiçoamento da mesma. de acordo com a presente análise, foram encontrados pontos relevantes para consideração, referentes à amostragem, à calibração dos examinadores e aos critérios para a avaliação de saúde bucal e necessidades de tratamento. Concluiu-se, em nível de recomendação, que, devido ao caráter dinâmico do conhecimento científico e, levando-se em consideração as diferenças regionais com relação ao padrão de desenvolvimento das doenças bucais, as propostas de padronização de levantamentos devem ser periodicamente revisadas. É provável, ainda, que outros pontos importantes não tenham sido detectados nesta análise, tornando-se premente ampliar esta discussão para toda a comunidade odontológica.

A body of research has developed within the context of nonlinear signal and image processing that deals with the automatic, statistical design of digital window-based filters. Based on pairs of ideal and observed signals, a filter is designed in an effort to minimize the error between the ideal and filtered signals. The goodness of an optimal filter depends on the relation between the ideal and observed signals, but the goodness of a designed filter also depends on the amount of sample data from which it is designed. In order to lessen the design cost, a filter is often chosen from a given class of filters, thereby constraining the optimization and increasing the error of the optimal filter. To a great extent, the problem of filter design concerns striking the correct balance between the degree of constraint and the design cost. From a different perspective and in a different context, the problem of constraint versus sample size has been a major focus of study within the theory of pattern recognition. This paper discusses the design problem for nonlinear signal processing, shows how the issue naturally transitions into pattern recognition, and then provides a review of salient related pattern-recognition theory. In particular, it discusses classification rules, constrained classification, the Vapnik-Chervonenkis theory, and implications of that theory for morphological classifiers and neural networks. The paper closes by discussing some design approaches developed for nonlinear signal processing, and how the nature of these naturally lead to a decomposition of the error of a designed filter into a sum of the following components: the Bayes error of the unconstrained optimal filter, the cost of constraint, the cost of reducing complexity by compressing the original signal distribution, the design cost, and the contribution of prior knowledge to a decrease in the error. The main purpose of the paper is to present fundamental principles of pattern recognition theory within the framework of active research in nonlinear signal processing.

OBJECTIVE: To study whether antioxidant supplementation will reduce the incidence of preeclampsia among patients at increased risk.METHODS: A randomized, placebo-controlled, double-blind clinical trial was conducted at four Brazilian sites. Women between 12 0/7 weeks and 19 6/7 weeks of gestation and diagnosed to have chronic hypertension or a prior history of preeclampsia were randomly assigned to daily treatment with both vitamin C (1,000 mg) and vitamin E (400 International Units) or placebo. Analyses were adjusted for clinical site and risk group (prior preeclampsia, chronic hypertension, or both). A sample size of 734 would provide 80% power to detect a 40% reduction in the risk of preeclampsia, assuming a placebo group rate of 21% and alpha=.05. The a level for the final analysis, adjusted for interim looks, was 0.0458.RESULTS: Outcome data for 707 of 739 randomly assigned patients revealed no significant reduction in the rate of preeclampsia (study drug, 13.8% [49 of 355] compared with placebo, 15.6% [55 of 352], adjusted risk ratio 0.87 [95.42% confidence interval 0.61-1.25]). There were no differences in mean gestational age at delivery or rates of perinatal mortality, abruptio placentae, pre-term delivery, and small for gestational age or low birth weight infants. Among patients without chronic hypertension, there was a slightly higher rate of severe preeclampsia in the study group (study drug, 6.5% [11 of 170] compared with placebo, 2.4% [4 of 168], exact P=.11, odds ratio 2.78, 95% confidence interval 0.79-12.62).CONCLUSION: This trial failed to demonstrate a benefit of antioxidant supplementation in reducing the rate of preeclampsia among'patients with chronic hypertension and/or prior preeclampsia.

A flow-injection (FI) system to match concentrations was used as an auto-diluter in multielement determination by inductively coupled plasma-mass spectrometry (ICP-MS). The flow system comprised loop-based injection or a timed valve that introduced a variable sample volume info a spray chamber through a standard Meinhard nebulizer of an ICP-MS. Routinely analyzed samples such as water, plant, and steel were selected. The accuracy of multielement determination was checked against water standard reference material from the National Institute of Standards and Technology (1643d), plant standard reference material from the National Bureau of Standards (1572 citrus leaves), and steel standard reference material from the National Bureau of Standards (AISI 4340). The measuring system was calibrated with a multielement solution, yielding a linear plot with good precision [relative standard deviation (RSD) < 3%, n = 12]. The results were in agreement at a 95% confidence level with the certified values for the reference materials and also with those obtained by continuous aspiration and by (FI) with a discrete volume. A rapid and economical method is described for the determination of deltamethrin in wheat, rice, peanuts and corn. It is based on simultaneous extraction and clean-up on a column packed with alumina and silica gel using n-hexane-ethyl ether (8:2, v/v), followed by a derivatization step and gas-chromatographic analysis. Recoveries from fortified cereal and peanut samples were determined at four concentration levels and ranged from 73 to 109%. The detection limits were 0.01 to 0.03 mg/ kg. This method simplifies the traditional procedures in terms of sample size, solvent consumption and analysis time. © Springer-Verlag 1998.

The formation of silica on core yttrium iron garnet presents a variety of different applications as corrosion resistance and stabilization of magnetic properties. Well-defined magnetic particles were prepared by heterocoagulating silica on yttrium iron garnet to protect the core. Yttrium iron garnet was obtained using a homogeneous nucleation process by controlling the chemical routes from cation hydrolysis in acid medium. The heterocoagulation was induced by tetraethyl orthosilicate hydrolysis in appropriate yttrium iron garnet dispersion medium. The presence of silica on yttrium iron garnet was characterized by vibrating sample magnetometry, X-ray photoemission spectroscopy, transmission electron microscopy, small area electron diffraction and differential thermal analysis. © 2001 Elsevier Science B.V. Autism spectrum disorders are severe psychiatric diseases commonly identified in the population. They are diagnosed during childhood and the etiology has been much debated due to their variations and complexity. Onset is early and characterized as communication and social interaction disorders and as repetitive and stereotyped behavior. Austistic disorders may occur together with various genetic and chromosomal diseases. Several chromosomal regions and genes are implicated in the predisposition for these diseases, in particular those with products expressed in the central nervous system. There are reports of autistic and mentally handicapped patients with submicroscopic subtelomeric alterations at the distal end of the long arm of chromosome 2. Additionally, there is evidence that alterations at 2q37 cause brain malformations that result in the autistic phenotype. These alterations are very small and not identified by routine cytogenetics to which patients are normally submitted, which may result in an underestimation of the diagnosis. This study aimed at evaluating the 2q37 region in patients with autistic disorders. Twenty patients were studied utilizing the fluorescence in situ hybridization technique with a specific probe for 2q37. All of them were also studied by the GTC banding technique to identify possible chromosomal diseases. No alterations were observed in the 2q37 region of the individuals studied, and no patient presented chromosomal diseases. This result may be due to the small sample size analyzed. The introduction of routine analysis of the 2q37 region for patients with autistic disorders depends on further studies. ©FUNPEC-RP.

The epidemiological surveys are important to the deployment, implementation and evaluation of projects and health actions in a community. The planning, goals, samples, team training/calibration, carrying out and publication of results are extremely important in the epidemiological surveys. Thus, the care with the sample and statistical analysis is fundamental for the results to be consistent and trustworthy in order to be able to be inferred for all the population. The aims of this study is to investigate the statistical methodology used in papers on dental caries epidemiological surveys published from 1960 to 2001. A bibliographical survey was carried out in BBO, MEDLINE and SCIELO databases. The papers found were analyzed with regards to the statistical methodology applied in the whole study, from the sampling to the tabling of data. Most studies (72.6%) only presented the number of elements that composed the sample, without explaining the planning involved in obtaining it.
