Extracellular iron reduction has been suggested as a candidate metabolic pathway that may explain a large proportion of carbon respiration in temperate peatlands. However, the o-phenanthroline colorimetric method commonly employed to quantitate iron and partition between redox species is known to be unreliable in the presence of humic and fulvic acids, both of which represent a considerable proportion of peatland dissolved organic matter. We propose ionic liquid extraction as a more accurate iron quantitation and redox speciation method in humic-rich peat porewater. We evaluated both o-phenanthroline and ionic liquid extraction in four distinct peatland systems spanning a gradient of physico-chemical conditions to compare total iron recovery and Fe2+:Fe3+ ratios determined by each method. Ionic liquid extraction was found to provide more accurate iron quantitation and speciation in the presence of dissolved organic matter. A multivariate approach utilizing fluorescence- and UV-Vis spectroscopy was used to identify dissolved organic matter characteristics in peat porewater that lead to poor performance of the o-phenanthroline method. Where these interferences are present, we offer an empirical correction factor for total iron quantitation by o-phenanthroline, as verified by ionic liquid extraction. In farbstoffsensibilisierten Solarzellen (DSSC) spielen Chromophore, die als Lichtsammel- und Energie-/Elektronentransfersysteme fungieren, eine zentrale Rolle. Phthalocyanine mit ihren intensiven Absorptionsbanden um 400 nm und 700 nm besitzen großes Potential für die effektive Sensibilisierung von Solarzellen. Trotz ihrer vielversprechenden physikochemischen Eigenschaften und intensiver Bemühungen erreichen Phthalocyanin-sensibilisierte Solarzellen nicht die Effizienzen, die bisher mit anderen Chromophorklassen erzielt werden konnten. In der vorliegenden Dissertation wurde die Entwicklung effizienter Lichtsammelsysteme für DSSC auf der Basis von Aza-substituierten Phthalocyaninen, sogenannten Pyrazinoporphyrazinen, verfolgt. Ein besonderer Fokus lag dabei auf einer Verbesserung der Absorptionseigenschaften der Chromophore im Bereich ihrer intrinsischen Absorptionslücke zwischen den Maxima um 400 nm und 700 nm. Um diese optische Lücke zu schließen wurden komplementär absorbierende BODIPY-Farbstoffe kovalent an synthetisch maßgeschneiderte Porphyrazine gebunden. Insgesamt wurden sechs neue Porphyrazin-Sensibilisatoren synthetisiert und photophysikalisch sowie elektrochemisch charakterisiert. Alle in dieser Arbeit untersuchten Porphyrazine tragen sterisch anspruchsvolle Tri(p-tolyl)propinyl-Gruppen um Agglomerationen zu vermeiden. Darüber hinaus wurden die Porphyrazine peripher entweder mit Hydroxy- oder Carboxygruppen als Bindungsstellen für oxidische Materialien ausgestattet sowie mit sechs BODIPY-Auxiliarfarbstoffen funktionalisiert, deren Substitutionsmuster variiert wurden. Zur Darstellung der komplexen Porphyrazine wurde eine Syntheseroute erarbeitet, die statistische Cyclisierungen unterschiedlicher Dinitril-Vorstufen beinhaltete und es ermöglichte, funktionelle Gruppen erst am vorgeformten Makrocyclus einzuführen. Die photophysikalische Untersuchung der hochfunktionalisierten Farbstoffe erfolgte über UV/Vis- und Fluoreszenzspektroskopie. Im Fall der BODIPY-Porphyrazin-Hybride schließt die zusätzliche Absorptionsbande der peripheren BODIPY-Einheiten die intrinsische Absorptionslücke der Porphyrazine. Die Hybride zeigen somit eine breite Absorption über den gesamten sichtbaren Spektralbereich mit hohen Extinktionskoeffizienten von ca. 4·10^5 M^−1cm^−1. Mittels Fluoreszenz- und Anregungsspektren wurde ein photoinduzierter Energie-transfer von den BODIPY-Einheiten auf den Porphyrazinkern nachgewiesen. Das elektrochemische Verhalten der BODIPY- und Porphyrazin-Verbindungen wurde mittels Cyclo- und Square-Wave-Voltammetrie untersucht. Die Effizienzen der Lichtenergieumwandlung wurden mit Hilfe von selbst-hergestellten und standardisierten farbstoffsensibilisierten Solarzellen bewertet. Alle Solarzellen zeigten eine messbare Photoaktivität unter Bestrahlung. Die Wirkungsgrade der Zellen lagen jedoch alle unter 1 %. Generell führten die Carboxyl-funktionalisierten Porphyrazine zu besseren Wirkungsgraden als die analogen, mit der tripodalen Ankergruppe ausgestatteten Derivate. Die mit Hilfe von Adsorptionsisothermen ermittelten Bindungskonstanten der Adsorption der Farbstoffe auf der TiO2-Oberfläche zeigten, dass beide Hafteinheiten eine feste Verankerung der Chromophore auf den TiO2-Elektroden ermöglichten. Insgesamt wirkte sich die Präsenz der peripheren BODIPY-Farbstoffe positiv auf die Wirkungsgrade der Solarzellen aus, jedoch nur in geringem Maß. Dieses Ergebnis wurde hauptsächlich auf die geringe Energiedifferenz zwischen der Leitungsbandkante des TiO2 und den LUMO-Energieniveaus der Chromophore zurückgeführt. Zusätzlich scheinen konkurrierende Prozesse wie die direkte Photoelektroneninjektion von den BODIPY-Einheiten in das TiO2 eine wichtige Rolle zu spielen. Neben der Anwendung in DSSC wurde die Wechselwirkung der Porphyrazine mit Graphen untersucht. Hierzu wurden A3B-Porphyrazine mit Pyrenyl-Seitenketten ausgestattet, die eine nicht-kovalente Verankerung des Chromophors auf Graphen ermöglichen. UV/Vis- und Fluoreszenzmessungen gaben u.a. erste Hinweise auf eine elektronische Kommunikation zwischen den beiden Hybridpartnern.

Este trabalho descreve a síntese de novos derivados de coumarinas 3-substituídas por grupos arilo, etenilarilo e etenil-organometálicos, através de novas metodologias via reacções de Heck e de metátese (Grubbs), com controlo da regioquímica e com significativos rendimentos reaccionais. A aplicação destas metodologias permitiu a síntese dos derivados, 3-fenilcoumarina (131), 3-(4-bromofenil)coumarina, (132), 3-(4-iodofenil)­coumarina (134), 3-(4-nitrofenil)coumarina (136), 3-(4-etilfenil)coumarina (133), 4-(coumarin-3-il)benzaldeído (135), 3-(4-metoxifenil)coumarina (137), (E)-3-acrilato-[4-(coumarin-3-il)fenil] de metilo (138), 6,7-metileno­dioxi-[3-(E)-2'-feniletenil]coumarina (145), 6,7-dimetoxi-[-(E)-2'-fenil­etenil]coumarina (146), 6,7-dimetoxi-[3-(E)-2'-(6'-nitrofenil)etenil]coumarina (147), 4-[2-(E)-(6,7-dimetoxicoumarin-3-il)etenil]benzaldeído (148) e 6,7-dimetoxi-[3-(E)-2'-ferroceniletenil]coumarina (149), dos quais os últimos nove, são compostos novos, identificados e caracterizados pela primeira vez. A deslocalização do sistema de electrões  conjugados, induzida pelos diversos substituintes das coumarinas, foi igualmente avaliada através da espectroscopia de UV/Vis. This article reports a combined thermodynamic, spectroscopic, and computational study on the interactions and structure of binary mixtures of hydrogenated and fluorinated substances that simultaneously interact through strong hydrogen bonding. Four binary mixtures of hydrogenated and fluorinated alcohols have been studied, namely, (ethanol + 2,2,2-trifluoroethanol (TFE)), (ethanol + 2,2,3,3,4,4,4-heptafluoro-1-butanol), (1-butanol (BuOH) + TFE), and (BuOH + 2,2,3,3,4,4,4-heptafluoro-1-butanol). Excess molar volumes and vibrational spectra of all four binary mixtures have been measured as a function of composition at 298 K, and molecular dynamics simulations have been performed. The systems display a complex behavior when compared with mixtures of hydrogenated alcohols and mixtures of alkanes and perfluoroalkanes. The combined analysis of the results from different approaches indicates that this results from a balance between preferential hydrogen bonding between the hydrogenated and fluorinated alcohols and the unfavorable dispersion forces between the hydrogenated and fluorinated chains. As the chain length increases, the contribution of dispersion increases and overcomes the contribution of H-bonds. In terms of the liquid structure, the simulations suggest the possibility of segregation between the hydrogenated and fluorinated segments, a hypothesis corroborated by the spectroscopic results. Furthermore, a quantitative analysis of the infrared spectra reveals that the presence of fluorinated groups induces conformational changes in the hydrogenated chains from the usually preferred all-trans to more globular arrangements involving gauche conformations. Conformational rearrangements at the CCOH dihedral angle upon mixing are also disclosed by the spectra.

In chapter one, the autoxidation kinetics of natural oil substrates, including, triglyceric sunflower oil, olive oil, terpenic squalene, and p-cymene were calibrated through differential oximetry methods. Calibration allows their use as reference oxidizable substrates for further studies, e.g. for quantitative testing of antioxidants under biomimetic settings. Several essential oils samples, of different botanical species or different productions of same species were studied for their antioxidant activity in inhibited autoxidation kinetics. Their antioxidant activities were matched with their composition analyzed by GC-MS. In chapter two, the molecular mechanism of the synergy between the common phenolic antioxidants such as tocopherol and catechols with widespread essential component gamma-terpinene was studied through lipid oxidation kinetics. Wherein, gamma-terpinene was able to disclose the key intermediacy HOO·, which acted as a reducing agent regenerating the phenolic antioxidant. This counterintuitive role of HOO· radicals was further investigated in detail and allowed to rationalize for the first time the purported antioxidant behavior of PDA melanin nanoparticles. It will also open to a deeper understanding of the redox biology of quinones. Regarding melanin, its role is broadly important in living organisms and its control, including its inhibition, is of great importance with several relevant applications ranging from food preservation to control of human skin pigmentation. In chapter three, an oximetry method combined with the traditional UV-Vis spectroscopy was developed to study the tyrosinase inhibition kinetics, which allowed identifying Glabridin (from G. glabra, L.), as one of the most effective natural tyrosinase inhibitors.

Deterioration phenomena occurring on outdoor cultural heritage have been the subject of several studies, but relatively few works investigated the specific role of Particulate Matter (PM) in the corrosion of metals. This topic is really complex and, besides field exposures, accelerated ageing tests are also necessary to isolate and understand deterioration mechanisms due to PM. For this reason, the development of a methodology that allows to reproduce and analyze the effect of PM on alloys through accelerated ageing in climatic chamber has been started. On quaternary bronze specimens, single salts and a mix of them were deposited via two deposition methods: dry (directly depositing the salt on the surface) and wet (dropping a salt solution and drying it), simulating the initial chemical activation of the salts by RH% variations or by raindrops, respectively. Then, to better mimic the composition of real PM, a mixture containing a soluble salts, a mineral, a black carbon and an organic fraction was formulated and spread on the samples. The samples were placed in a climatic chamber and exposed to cyclic variations of T and RH for three weeks. The ageing cycles were set according to predictions on salt deliquescence/recrystallization through E-AIM model and to the evaluation of regional climatic data. The surface evolution was followed by SEM-EDX, Raman, AT-IR and UV-Vis Spectrophotometry. At the end of the test, mass losses were determined and corroded metals removed by pickling were analyzed by AAS. On the basis of the obtained results, the tested procedures seem to be promising in accelerating and mimicking realistic corrosion phenomena, as under the selected conditions, corrosion products typically found at different exposure time (from days to years) on outdoor bronzes were able to progressively form and evolve. Moreover, the two deposition modes simulating different condition of chemical activation of PM deposits allow to obtain complementary information.

Glucaric acid (GA) is one of the building block chemicals derived from sugar biomass with higher added value. Nowadays, GA is produced by oxidation of glucose (Glu) with either stoichiometric oxidants (HNO3), or by means of electrochemical or biochemical synthesis. However, these processes show drawbacks from either the environmental or economic viewpoint. For this reason, gold nanoparticles (Au NPs) supported on activated carbon (AC) have been studied as catalysts for the oxidation of Glu, using O2 as oxidant in the presence of a base. Using sol immobilization technique, Au NPs have been supported on AC following different experimental procedures. UV-Vis spectroscopy, XRD, TEM and TG analysis were utilized in the characterization of the catalysts. The operational conditions were optimized obtaining 24% of yield of GA, 37% to GO and 27% to byproducts in 1 h, 1000 rpm, 10 bar of O2 and Glu:Au:NaOH molar ratio of 1000:1:3000. Under such conditions, catalysts show relatively high Glu conversion (≥82%) with different GA yields. GO+GA yield is around 58-61%. Therefore, the oxidation reaction was performed at 15 min where Au/AC PVA0 reached the highest yield of GA (16%) and Au/AC PVA2.4 gave the lowest (8%). It is evident that the presence of PVA influences to a higher degree the reaction rate than the Au NPs size. Hence, the effect of different heat treatments where applied for the removal of PVA: washing with water at 60℃ or heat treatment (120-250℃) with Air/H2. Washing treatment and heat treatment at 120℃ with Air/H2 may have resulted in the mildest treatments for the removal of PVA. Finally, two different supports have been used in order to study the effect of metal-support interaction in the immobilization of Au NPs: ZrO2 and AC. Au/AC catalyst demonstrated a higher conversion of GO to GA at short reaction times (15.1% yield GA) compared to Au/ZrO2 (2.4% yield GA).

Preformed Au nanoparticles supported on activated carbon and TiO2 were synthesised by sol-immobilisation. Polyethylene glycol, polyvinyl pyrrolidone and polyvinyl alcohol were used as stabilisers for the gold nanoparticles at different polymer/Au wt/wt ratios for each polymer. The effect of polymer/Au wt/wt ratios was investigated on (i) the average nanoparticle size, (ii) catalytic activity for two reactions, 4-nitrophenol reduction and glucose oxidation to glucaric acid. 4-nitrophenol reduction is recognised as a model reaction for nanomaterial catalytic activity tests; glucose oxidation to glucaric acid is a reaction that is traditionally carried out with concentrated nitric acid, for which alternative reaction pathways are looked for in an effort to reduce its environmental impact. The catalysts were characterised from the nanoparticle synthesis by colloidal method by means of UV-vis spectroscopy and DLS analysis, to the immobilisation step by XRD and TEM. The effect of the polymer:Au wt/wt ratio on nanoparticle size depends on the polymer nature, and point out the need to optimise supported nanoparticle synthesis protocols in the future depending on the type of stabiliser. The catalytic tests revealed that the polymers interact with Au nanoparticles through different active sites. Activated carbon (AC) and TiO2 were compared as supports for Au nanoparticles stabilised by PVA at PVA/Au 0,65 wt/wt. AC-supported Au NPs were the most active for glucose oxidation while TiO2-stabilised Au NPs were five times more active in 4-nitrophenol reduction that AC-supported NPs. Hence support and stabiliser are important parameters that should be optimised in order to achieve high catalytic activity for a given reaction.

Metal nanoparticle catalysts have in the last decades been extensively researched for their enhanced performance compared to their bulk counterpart. Properties of nanoparticles can be controlled by modifying their size and shape as well as adding a support and stabilizing agent. In this study, preformed colloidal gold nanoparticles supported on activated carbon were tested on the reduction of 4-nitrophenol by NaBH4, a model reaction for evaluating catalytic activity of metal nanoparticles and one with high significance in the remediation of industrial wastewaters. Methods of wastewater remediation are reviewed, with case studies from literature on two major reactions, ozonation and reduction, displaying the synergistic effects observed with bimetallic and trimetallic catalysts, as well as the effects of differences in metal and support. Several methods of preparation of nanoparticles are discussed, in particular, the sol immobilization technique, which was used to prepare the supported nanoparticles in this study. Different characterization techniques used in this study to evaluate the materials and spectroscopic techniques to analyze catalytic activities of the catalyst are reviewed: ultraviolet-visible (UV-Vis) spectroscopy, dynamic light scattering (DLS) analysis, X-ray diffraction (XRD) analysis and transmission electron microscopy (TEM) imaging. Optimization of catalytic parameters was carried out through modifications in the reaction setup. The effects of the molar ratio of reactants, stirring, type and amount of stabilizing agent are explored. Another important factor of an effective catalyst is its reusability and long-term stability, which was examined with suggestions for further studies. Lastly, a biochar support was newly tested for its potential as a replacement for activated carbon.

Cool giant and supergiant stars are among the brightest populations in any stellar system and they are easily observable out to large distances, especially at infrared wavelengths. These stars also dominate the integrated light of star clusters in a wide range of ages, making them powerful tracers of stellar populations in more distant galaxies. High-resolution near-IR spectroscopy is a key tool for quantitatively investigating their kinematic, evolutionary and chemical properties. However, the systematic exploration and calibration of the NIR spectral diagnostics to study these cool stellar populations based on high-resolution spectroscopy is still in its pioneering stage. Any effort to make progress in the field is innovative and of impact on stellar archaeology and stellar evolution. This PhD project takes the challenge of exploring that new parameter space and characterizing the physical properties, the chemical content and the kinematics of cool giants and supergiants in selected disc fields and clusters of our Galaxy, with the ultimate goal of tracing their past and recent star formation and chemical enrichment history. By using optical HARPS-N and near-infrared GIANO-B high-resolution stellar spectra in the context of the large program SPA-Stellar Population Astrophysics: the detailed, age-resolved chemistry of the Milky Way disk” (PI L. Origlia), an extensive study of Arcturus, a standard calibrator for red giant stars, has been performed. New diagnostics of stellar parameters as well as optimal linelists for chemical analysis have been provided. Then, such diagnostics have been used to determine evolutionary properties, detailed chemical abundances of almost 30 different elements and mixing processes of a homogeneous sample of red supergiant stars in the Perseus complex.

Pollution of water bodies is one of the most common environmental problems today. Organic pollutants are one of the main drawbacks in this natural resource, among which the following stand out long-lived dyes, pharmaceuticals, and pesticides. This research aims at obtaining nanocomposites based on polycaprolactone-chitosan (PCL-CS) electrospun nanofibers (NFs) containing TiO2 nanoparticles (NPs) for the adsorption and photocatalytic degradation of organic pollutants, using Rhodamine B as a model. The fabricated hybrid materials were characterized by FT-IR, TGA, DSC, SEM, TEM, tensile properties, and the contact angle of water drops. The photoactivity of the NFs was investigated using a batch-type system by following UV-Vis absorbance and fluorescence of rhodamine B (RhB). For this purpose, TiO2NPs were successfully ex-situ incorporated into the polymer matrix promoting good mechanical properties and higher hydrophilicity of the material. The results showed that CS in the NFs increased the absorption and degradation of RhB by the TiO2NPs. CS attracted the pollutant molecules to the active sites vicinity of TiO2NPs, favoring initial adsorption and degradation. In other words, a bait-hook-and-destroy effect was evidenced. It also was demonstrated that the sensitization of TiO2 by organic dyes (e.g., perylene derivative) considerably improves the photocatalytic activity under visible radiation, allowing the use of low amounts of TiO2. (≈0.05 g/1 g of fiber). Hence, the current study is expected to contribute with an environmentally friendly green alternative solution.

In questo lavoro di tesi è stata investigata la sintesi di compositi a base di nano particelle di biossido di titanio rivestite da un bio-tensioattivo naturale. Il noto fotocatalizzatore (Nano-TiO2) è stato accoppiato ad un bio-tensioattivo dalle riconosciute proprietà antibatteriche, antivirali e anti-tumorali per ottenere un materiale composito multifunzionale. Diverse opzioni di design sono state investigate e la sintesi ottimizzata attraverso una caratterizzazione sistematica dei materiali prodotti, sia sulle sospensioni (DLS, ELS, TEM) sia sui prodotti granulati e calcinati (XRD, FT-IR, SEM, UV-Vis., BET). Per comprendere il ruolo del bio-tensioattivo e i potenziali effetti sinergici che il materiale composito potesse generare, si sono effettuate diverse caratterizzazioni funzionali testando il materiale per la realizzazione di nano-fasi fotocatalitiche da impiegare in processi di adsorbimento/degradazione di inquinanti acquosi, per la realizzazione di rivestimenti tessili antibatterici e come composito utile per l’assorbimento di metalli pesanti.

The present paper describes a novel, simple and reliable differential pulse voltammetric method for determining amitriptyline (AMT) in pharmaceutical formulations. It has been described for many authors that this antidepressant is electrochemically inactive at carbon electrodes. However, the procedure proposed herein consisted in electrochemically oxidizing AMT at an unmodified carbon nanotube paste electrode in the presence of 0.1 mol L(-1) sulfuric acid used as electrolyte. At such concentration, the acid facilitated the AMT electroxidation through one-electron transfer at 1.33 V vs. Ag/AgCl, as observed by the augmentation of peak current. Concerning optimized conditions (modulation time 5 ms, scan rate 90 mV s(-1), and pulse amplitude 120 mV) a linear calibration curve was constructed in the range of 0.0-30.0 μmol L(-1), with a correlation coefficient of 0.9991 and a limit of detection of 1.61 μmol L(-1). The procedure was successfully validated for intra- and inter-day precision and accuracy. Moreover, its feasibility was assessed through analysis of commercial pharmaceutical formulations and it has been compared to the UV-vis spectrophotometric method used as standard analytical technique recommended by the Brazilian Pharmacopoeia.

Silver nanoparticles have attracted considerable attention due to their beneficial properties. But toxicity issues associated with them are also rising. The reports in the past suggested health hazards of silver nanoparticles at the cellular, molecular, or whole organismal level in eukaryotes. Whereas, there is also need to examine the exposure effects of silver nanoparticle to the microbes, which are beneficial to humans as well as environment. The available literature suggests the harmful effects of physically and chemically synthesised silver nanoparticles. The toxicity of biogenically synthesized nanoparticles has been less studied than physically and chemically synthesised nanoparticles. Hence, there is a greater need to study the toxic effects of biologically synthesised silver nanoparticles in general and mycosynthesized nanoparticles in particular. In the present study, attempts have been made to assess the risk associated with the exposure of mycosynthesized silver nanoparticles on a beneficial soil microbe Pseudomonas putida. KT2440. The study demonstrates mycosynthesis of silver nanoparticles and their characterisation by UV-vis spectrophotometry, FTIR, X-ray diffraction, nanosight LM20 - a particle size distribution analyzer and TEM. Silver nanoparticles obtained herein were found to exert the hazardous effect at the concentration of 0.4μg/ml, which warrants further detailed investigations concerning toxicity.

This study addressed the use of conventional and vegetable origin polyurethane foams to extract C. I. Acid Orange 61 dye. The quantitative determination of the residual dye was carried out with an UV/Vis absorption spectrophotometer. The extraction of the dye was found to depend on various factors such as pH of the solution, foam cell structure, contact time and dye and foam interactions. After 45 days, better results were obtained for conventional foam when compared to vegetable foam. Despite presenting a lower percentage of extraction, vegetable foam is advantageous as it is considered a polymer with biodegradable characteristics.
