The present study regards an applied social research (qualitative) performed in two institutions which lead children s cancer treatment in Natal, Rio Grande do Norte. The main goal of this study is, as of a literature review at works which characterizes the first generations of study about substantive rationality, to detect Decision Making process related aspects that may serve as a basis to elaborate analysis categories from decision making process, aggregating them into a new study that may provide an advance to the theme in administrative science. The academic works based on the analysis model created by Mauricio Serva served as a basis to deep research into such theme, which verifies the predominant rationality in eleven administrative internal processes in productive organizations. This dissertation intends to go beyond the identification of the predominant rationality by elaborating new categories of analysis, and making possible the continuity of the subject in administrative science. Based on Guerreiro Ramos s work, which sees a kind of ideal organization, as known as isonomies, this study still calls upon Karl Polanyi s thoughts, which with the objective of comprehending the independent economic phenomenon of the value that allows considering non-market economies, find that the economy of the men is submerged in his social relations; it also rescues the studies from Max Weber who investigates the meaning of social action to better understand the rationality, and refers to the study of Jürgen Habermas, who proposes a broader conception about rationality, within the theory of communicative action. As a result of this theme s review, seven analysis categories of the decision making process have been formulated. They were applied in the institutions that had been chosen and helped to detect the type of predominant rationality in the categories of the decision making process. The results confirm that, although the decisions making process involves rational elements, such as information, identification of alternatives, there are also specific values of each individual with his experience and view oh the world, permeated not only by instrumental rationality, but also by substantive rationality. The study has verified that two similar institutions may show different types of rationality in the decision making process, when decision factors may tend to instrumental rationality, according to administration classic way, as well as they may emerge from substantive rationality, thus contributing to the process of emancipation of the human being in his sphere of work

Perturbation of natural ecosystems, namely by increasing freshwater use and its degradative use, as well as topsoil erosion by water of land-use production systems, have been emerging as topics of high environmental concern. Freshwater use has become a focus of attention in the last few years for all stakeholders involved in the production of goods, mainly agro-industrial and forest-based products, which are freshwater-intensive consumers, requiring large inputs of green and blue water. This thesis presents a global review on the available Water Footprint Assessment and Life Cycle Assessment (LCA)-based methods for measuring and assessing the environmental relevance of freshwater resources use, based on a life cycle perspective. Using some of the available midpoint LCA-based methods, the freshwater use-related impacts of a Portuguese wine (white ‘vinho verde’) were assessed. However, the relevance of environmental green water has been neglected because of the absence of a comprehensive impact assessment method associated with green water flows. To overcome this constraint, this thesis helps to improve and enhance the LCA-based methods by providing a midpoint and spatially explicit Life Cycle Impact Assessment (LCIA) method for assessing impacts on terrestrial green water flow and addressing reductions in surface blue water production caused by reductions in surface runoff due to land-use production systems. The applicability of the proposed method is illustrated by a case study on Eucalyptus globulus conducted in Portugal, as the growth of short rotation forestry is largely dependent on local precipitation. Topsoil erosion by water has been characterised as one of the most upsetting problems for rivers. Because of this, this thesis also focuses on the ecosystem impacts caused by suspended solids (SS) from topsoil erosion that reach freshwater systems. A framework to conduct a spatially distributed SS delivery to freshwater streams and a fate and effect LCIA method to derive site-specific characterisation factors (CFs) for endpoint damage on aquatic ecosystem diversity, namely on algae, macrophyte, and macroinvertebrates organisms, were developed. The applicability of this framework, combined with the derived site-specific CFs, is shown by conducting a case study on E. globulus stands located in Portugal as an example of a land use based system. A spatially explicit LCA assessment was shown to be necessary, since the impacts associated with both green water flows and SS vary greatly as a function of spatial location.

This study seeks to understand how the physiological constraints of diving may change on a daily and seasonal basis. Dive data were obtained from southern elephant seals (Mirounga leonina) from South Georgia using satellite relay data loggers. We analysed the longest (95th percentile) dive durations as proxies for physiological dive limits. A strong, significant relationship existed between the duration of these dives and the time of day and week of year in which they were performed. The depth of the deepest dives also showed a significant, but far less consistent, relationship with local time of day and season. Changes in the duration of the longest dives occurred irrespective of their depth. Dives were longest in the morning (04:00-12:00 h) and shortest in the evening (16:00-00:00 h). The size of the fluctuation varied among animals from 4.0 to 20.0 min. The daily pattern in dive depth was phase-shifted in relation to the diurnal rhythm in dive duration. Dives were deeper at midday and shallower around midnight. Greater daily changes in duration occurred in seals feeding in the open ocean than in those foraging on the continental shelf. The seasonal peak in the duration of the longest dives coincided with austral midwinter. The size of the increase in dive duration from autumn/spring to winter ranged from 11.5 to 30.0 min. Changes in depth of the longest dives were not consistently associated with particular times of year. The substantial diurnal and seasonal fluctuations in maximum dive duration may be a result of changes in the physiological capacity to remain submerged, in addition to temporal changes in the ecological constraints on dive behaviour. We speculate about the role of melatonin as a hormonal mediator of diving capability.

Induction of resistance is defined as the activation of a state of resistance against diseases which is induced systemically in plants by the use of biotic or abiotic agents without any modification of the plant genome, occurring non-specific way, by activating genes coding for various plant defense responses. Chitosan is a polymer derived from the deacetylation of chitin, which is found in large quantities in crustacean shell, and studied with the potential to control plant pathogens, both by its direct fungistatic action, as the ability to induce protection of plants, indicating the presence of molecules of elicitoras characteristics. Three experiments with objective of evaluating the potential of chitosan in the seedling resistance induction were developed, beet (Beta vulgaris) seeds, cucumber (Cucumis sativus) seeds and tomato (Solanum lycopersicum) seeds, and the control of Fusarium sp., Rhizoctonia solani K¨uhn e Pythium sp. in vitro conditions. The experimental design was completely randomized, with four replications. Beet seeds, tomato and cucumber were submerged in chitosan solution for 20 minutes, in concentrations of 0.25, 0.5, 1 and 2% in the control and distilled water. Seeds were sown in trays containing Plantmax Florestalr substrate sterilized and inoculated with Fusarium sp., Rhizoctonia solani K¨unh and Pythium sp., respectively for the three cultures. The experiment was conducted for 14 days in growth chamber with controlled temperature (25 C 2 C), light (12 hour photoperiod) and humidity (70% 10%). The evaluations were seed emergency, seedling damping-off, seedling length, fresh weight and activity of the enzymes phenylalanine amˆonia-liase (PAL), chitinase and b-1,3-glucanase. It was also rated the mycelial growth of Fusarium sp., Pythium sp. and R. solani on P.D.A. (Potato-Dextrose and Agar) culture medium containing chitosan at the same concentrations evaluated in seeds. For beet growing, seed treatment with chitosan presented higher emergence and the length of the seedlings, and reduced the percentage of tipping. Treatment with chitosan activated the systemic acquired resistance with expression of chitinase and b-1,3-glucanase enzymes. For the tomato crop in chitosan concentration of 0.25% favored the emergency of seedlings, reduced the incidence of tipping and activated the PAL enzymes, chitinase and b-1,3-glucanase. In cucumber on the concentration of up 0.5% favored seedlings emergence and reduces the incidence of tipping. Chitosan activated the PAL enzymes and b-1,3-glucanase. Chitosan also presented fungistatic action on the initial growth of Pythium sp. and R. solani in vitro conditions, however, such action did not prevail until the end of the experiment. To Fusarium sp. the concentration of chitosan resulted in the reduction of mycelial growth in vitro.

The stratigraphic architecture of deep sea depositional systems has been discussed in detail. Some examples in Ischia and Stromboli volcanic islands (Southern Tyrrhenian sea, Italy) are here shown and discussed. The submarine slope and base of slope depositional systems represent a major component of marine and lacustrine basin fills, constituting primary targets for hydrocarbon exploration and development. The slope systems are characterized by seven seismic facies building blocks, including the turbiditic channel fills, the turbidite lobes, the sheet turbidites, the slide, slump and debris flow sheets, lobes and tongues, the fine-grained turbidite fills and sheets, the contourite drifts and finally, the hemipelagic drapes and fills. Sparker profiles offshore Ischia are presented. New seismo-stratigraphic evidence on buried volcanic structures and overlying Quaternary deposits of the eastern offshore of the Ischia Island are here discussed to highlight the implications on marine geophysics and volcanology. Regional seismic sections in the Ischia offshore across buried volcanic structures and debris avalanche and debris flow deposits are here presented and discussed. Deep sea depositional systems in the Ischia Island are well developed in correspondence to the Southern Ischia canyon system. The canyon system engraves a narrow continental shelf from Punta Imperatore to Punta San Pancrazio, being limited southwestwards from the relict volcanic edifice of the Ischia bank. While the eastern boundary of the canyon system is controlled by extensional tectonics, being limited from a NE-SW trending (counter-Apenninic) normal fault, its western boundary is controlled by volcanism, due to the growth of the Ischia volcanic bank. Submarine gravitational instabilities also acted in relationships to the canyon system, allowing for the individuation of large scale creeping at the sea bottom and hummocky deposits already interpreted as debris avalanche deposits. High resolution seismic data (Subbottom Chirp) coupled to high resolution Multibeam bathymetry collected in the frame of the Stromboli geophysical experiment aimed at recording seismic active data and tomography of the Stromboli Island are here presented. A new detailed swath bathymetry of Stromboli Island is here shown and discussed to reconstruct an up-to-date morpho-bathymetry and marine geology of the area, compared to volcanologic setting of the Aeolian volcanic complex. The Stromboli DEM gives information about the submerged structure of the volcano, particularly about the volcano-tectonic and gravitational processes involving the submarine flanks of the edifice. Several seismic units have been identified around the volcanic edifice and interpreted as volcanic acoustic basement pertaining to the volcano and overlying slide chaotic bodies emplaced during its complex volcano-tectonic evolution. They are related to the eruptive activity of Stromboli, mainly poliphasic and to regional geological processes involving the geology of the Aeolian Arc.

A contaminação aleatória de alimentos por micotoxinas afeta as condições de sanidade das dietas de humanos e animais. Dentre as toxinas fúngicas, deoxinivalenol (DON) se destaca pela freqüente contaminação de produtos agrícolas e alimentos e pela sua resistência a degradação pelo emprego de métodos tradicionais de processamento, o que motiva políticas de controle e a busca por técnicas de descontaminação. A descontaminação biológica empregando processsos fermentativos tem sido apontada como uma alternativa promissora, pois permite degradar micotoxinas através do sistema enzimático microbiano e melhorar características funcionais e sensoriais de matérias-primas e insumos alimentícios. Este trabalho teve por objetivo estudar condições e mecanismos de biodegradação de deoxinivalenol empregando Aspergillus oryzae e Rhizopus sp. em sistemas fermentativos submersos. Para tanto, foi necessário adequar metodologia para reação de derivação na determinação cromatográfica de DON; estudar o potencial e condições de degradação via fermentação submersa por Aspergillus oryzae e Rhizopus sp.; e avaliar a atividade de oxidoredutases e a citotoxicidade dos extratos fementados. A otimização da metodologia estabeleceu a melhor condição para a reação de derivação com 200 µL de anidrido trifluoroacético e 18 mg de bicarbonato de sódio, durante 6 minutos a 74 °C na faixa entre 7 e 21 µg de DON. A quantificação de DON residual no meio fermentado mostrou que as espécies fúngicas Rhizopus sp. e Aspergillus oryzae possuem a capacidade de degradar DON demonstrando índices médios de 87,4 e 62,4% respectivamente, principalmente quando o meio submerso foi água estéril e fermentação realizada durante 48 horas. A velocidade máxima de degradação neste intervalo foi de 10,8 e 12,4 ppb/h, observando também um aumento na atividade específica da enzima peroxidase. Os extratos dos fermentados com A. oryzae e Rhizopus sp. apresentaram efeito de inibição de proliferação celular (IC50) quando concentrados 10 vezes em 48 e 72 horas respectivamente. Os meios fermentados com Rhizopus sp. apresentaram menor efeito (1,5 vezes) quando comparado com Aspergillus oryzae.

It is located in eastern Uganda, the wetland system is characterized by open water that is clear with some emergent vegetation mainly of sedges and floating vegetation dominated by the day water lily Nymphaea sp and submerged water weeds. Some patches of Cyperus papyrus exist in areas with minimal agricultural encroachment. The main inflow into the Opeta system is through River Sironko.

A field study of the invertebrate communities of the Nabugabo lakes(Nabugabo,Kayanja and Kayugi)showed the occurrence of copepoda, cladocera and rotifera(micro-invertebrates or zooplankton); Ephemeroptera and Diptera(macro-invertebrates or zoo-benthos). The most commonly encountered taxa were thermocyclops neglectus, moinamicrura,several rotiferan species(micro-invertebrates);P.adusta,chironomus, tanipodinae and trichoptera(macro- invertebrates). These organisms are assumed to be readily available as food sources for fishes in the Nabugabo lakes. Higher abundance and diversity of invertebrates occurred in Lake Nabugabo compared to Kayanja and Kayugi. There were no major differences in diversity and abundance of organisms between inshore and offshore areas of the different lakes. The highest diversity of macro-invertebrates(up to 15 taxa)was recovered from roots of macrophyte(higher water-based plants)such as Miscanthidium and Papyrus. The zooplankton of Nabugabo lakes typify a tropical assemblage with few species among genera and dominance of the communities by small-bodied organisms. Some taxa,common to many other water bodies such as Mesocyclops spp.,Calanoids(Copepoda), Caridina nilotica (Decapoda)were noticeably missing in the Nabugabo lakes community, probably due to environmental limitations including low conductivity and pH. Where they occur,these missing taxa have been shown to be key forage items for fishes and therefore their absence in Nabugabo lakes may have implications with respect to potential for fishery production. However other valuable invertebrate types such as cyclopoid copepods,ephemeroptera, chironomid and chaoborid larvae do occur in sufficiently high diversity and abundance to support viable fisheries resources. The high diversity and abundance of invertebrates associated with aquatic macrophytes such as Papyrus and Miscanthidium need to be protected through control of access and utilisation of shoreline vegetation.

Fish species diversity in Lake Nabugabo, Uganda, has declined following establishment of the introduced fish species in the lake. Most of the native fish species have disappeared and the lake is now dominated by the introduced Nile perch, Lates niloticus and the Nile tilapia Oreochromis niloticus. The dominant native fish species include Synodontis afrofischeri, Schilbe intermedius, Rastrineobola argentea and Brycinussadleri. Some of the native fish species that have disappeared from Lake Nabugabo were reported to occur in lakes Kayugi and Kayanja, which are adjacent to Lake Nabugabo but separated from it by extensive papyrus swamps. The Nabugabo lakes are satellite water bodies in the Lake Victoria basin, which is known to have experienced fish species changes due to the introduction of the Nile perch Lates niloticus during the 1960s.The Nabugabo lakes comprising of Lake Nabugabo main, and the smaller lakes Kayanja and Kayugi were investigated between 2000 and 2002 with experimental gill netting to evaluate the potential of these lakes in conservation of fish species diversity. Results show that some native fish species especially Oreochromis esculentus, and Oreochromis variabilis and the haplochromine cichlid Prognathochromis venator that have disappeared from Lake Nabugabo still occur in Lakes Kayanja and Kayugi. Inshore habitats with macrophyte cover were also found to be important habitats for the endangered native fish species in the Nabugabo lakes. These lakes and inshore habitats need to be protected to conserve the endangered native fish species and to reduce further decline in fish species diversity.

As lipases e os biossurfactantes são compostos produzidos por microrganismos através de fermentações em estado sólido (FES) ou sumberso (FSm), os quais são aplicáveis nas indústrias alimentícia e farmacêutica, na bioenergia e na biorremediação, entre outras. O objetivo geral deste trabalho foi otimizar a produção de lipases através de fermentação em estado sólido e fermentação submersa. Os fungos foram selecionados quanto à habilidade de produção de lipases através de FES e FSm e aqueles que apresentaram as maiores atividades lipolíticas foram utilizados na seleção de variáveis significativas e na otimização da produção de lipases nos dois modos de cultivo. Foram empregadas técnicas seqüenciais de planejamento experimental, incluindo planejamentos fracionários, completos e a metodologia de superfície de resposta para a otimização da produção de lipases. As variáveis estudadas na FES foram o pH, o tipo de farelo como fonte de carbono, a fonte de nitrogênio, o indutor, a concentração da fonte de nitrogênio, a concentração do indutor e a cepa do fungo. Na FSm, além das variáveis estudadas na FES, estudaram-se as variáveis concentração inicial de inóculo e agitação. As enzimas produzidas foram caracterizadas quanto à temperatura e pH ótimos e quanto à estabilidade a temperatura e pH. Nas condições otimizadas de produção de lipases, foi avaliada a correlação entre a produção de lipases e bioemulsificantes. Inicialmente foram isolados 28 fungos. Os fungos Aspergillus O- 4 e Aspergillus E-6 foram selecionados como bons produtores de lipases no processo de fermentação em estado sólido e os fungos Penicillium E-3, Trichoderma E-19 e Aspergillus O-8 como bons produtores de lipases através da fermentação submersa. As condições otimizadas para a produção de lipases através de fermentação em estado sólido foram obtidas utilizando-se o fungo Aspergillus O-4, farelo de soja, 2% de nitrato de sódio, 2% de azeite de oliva e pHs inferiores a 5, obtendo-se atividades lipolíticas máximas de 57 U. As condições otimizadas para a produção de lipases na fermentação submersa foram obtidas utilizando-se o fungo Aspergillus O-8, farelo de trigo, 4,5% de extrato de levedura, 2% de óleo de soja e pH 7,15. A máxima atividade obtida durante a etapa de otimização foi 6 U. As lipases obtidas por FES apresentaram atividades máximas a 35ºC e pH 6,0, enquanto que as obtidas por FSm apresentaram ótimos a 37ºC e pH 7,2. A estabilidade térmica das lipases produzidas via FSm foi superior a das lipases obtidas via FES, com atividades residuais de 72% e 26,8% após 1h de exposição a 90ºC e 60ºC, respectivamente. As lipases obtidas via FES foram mais estáveis em pH´s alcalinos, com atividades residuais superiores a 60% após 24 h de exposição, enquanto as lipases produzidas via FSm foram mais estáveis em pH´s ácidos, com 80% de atividade residual na faixa de pH entre 3,5 e 6,5. Na fermentação submersa a correlação entre a produção de lipases e a atividade emulsificante óleo em água (O/A) e água em óleo (A/O) dos extratos foi 95,4% e 86,8%, respectivamente, obtendo-se atividades emulsificantes máximas O/A e A/O de 2,95 UE e 42,7 UE. Embora a maior produção de lipases tenha sido obtida na fermentação em estado sólido, não houve produção concomitante de biossurfactantes. Os extratos da fermentação submersa apresentaram redução da tensão superficial de 50 mN m -1 para 28 mN m -1 e atividade antimicrobiana frente ao microrganismo S. aureus ATCC 25923, com potenciais antimicrobianos de 36 a 43% nos três primeiros dias de fermentação. A fermentação submersa foi a técnica que apresentou os melhores resultados de otimização da produção de lipases, bem como de produção simultânea de biossurfactantes.

Today the Ria de Aveiro of northern Portugal has a hydromorphological regime in which river influence is limited to periods of flood. For most of the annual cycle, tidal currents and wind waves are the major forcing agents in this complex coastal lagoon–estuarine system. The system has evolved over two centuries from one that was naturally fluvially dominant to one that is today tidally dominant. Human influence was a trigger for these changes, starting in 1808 when its natural evolution was halted by the construction of a new inlet/outlet channel through the mobile sand spit that isolates it from the Atlantic Ocean. In consequence, tidal ranges in the lagoon increased rapidly from ~0.1 m to >1 m and continued to increase, as a result of continued engineering works and dredging, today reaching ~3 m on spring tides. Hydromorphological adjustments that have taken place include the deepening of channels, an increase in the area of inter-tidal flats, regression of salt marsh, increased tidal propagation and increased saline intrusion. Loss of once abundant submerged aquatic vegetation (SAV), due to increased tidal flows, exacerbated by increased recreational activities, has been accompanied by a change from fine cohesive sediments to coarser, mobile sediments with reduced biological activity.

Satellite lakes and rivers in the Victoria and Kyoga basins provide a sanctuary for endangered native fish species. The structural heterogeneity of macrophyte covering these lakes has made it possible for most of the biodiversity to be kept intact. The Kyoga minor lakes have the highest fish species diversity especially of the haplochromines. Most fish communities of these satellite lakes are composed of native species.

A sociedade em que vivemos hoje encontra-se submersa pela tecnologia em todas as áreas e saberes, exigindo-se aos cidadãos a capacidade de pensar e agir racionalmente perante os desafios e problemas que vão surgindo ao longo da vida. As tecnologias da informação e comunicação (TIC) são uma presença evidente no quotidiano e diversas investigações e estudos têm vindo a enfatizar o seu potencial no processo de ensino-aprendizagem. Ademais, face aos desafios científicos e tecnológicos da nossa sociedade, é fundamental formar-se cidadãos capazes de pensar criticamente e racionalmente, de tomarem decisões focadas no que se deve fazer ou acreditar. Assim, reconhecendo a importância de se promover o pensamento crítico (PC) recorrendo-se às TIC, desenvolveu-se a presente investigação com o intuito de promover o PC dos alunos, através da exploração de diversos recursos digitais já disponíveis gratuitamente na internet. Decorrente da finalidade apresentada formularam-se as seguintes questões de estudo: (i) Quais os contributos dos recursos digitais explicitamente promotores de pensamento crítico dos alunos do 3.º ano de escolaridade?; e (ii) Quais os contributos dos recursos digitais explicitamente promotores de pensamento crítico para a construção de conhecimentos científicos, na área das Ciências, dos alunos do 3.º ano de escolaridade?. A presente investigação desenvolveu-se de acordo com um paradigma sócio-crítico, segundo uma perspetiva metodológica predominantemente qualitativa e com base num plano de Investigação-Ação. A implementação das três atividades construídas, incluindo a exploração dos recursos digitais, decorreu numa turma de 26 alunos inseridos num colégio do distrito de Aveiro, no qual se realizou a Prática Pedagógica Supervisionada da professora investigadora. Para a recolha dos dados utilizaram-se diversos instrumentos, nomeadamente testes de levantamento inicial e final das capacidades de PC, listas de verificação, diário do investigador, fichas de trabalho resolvidas pelos alunos (tendo por base a Taxonomia de Ennis) e questionários de autoavaliação do desempenho dos mesmos. Na análise de dados, privilegiou-se a análise de conteúdo recorrendo-se ao software WebQDA. Com base nos instrumentos de recolha de dados referidos verificou-se que os alunos manifestaram uma maior facilidade a responder a questões que promoviam o uso de capacidades de PC referentes à Clarificação Elementar e à Indução e uma dificuldade acentuada na dimensão das Estratégias e Táticas. No que diz respeito à construção de conhecimentos científicos, os alunos demonstraram, principalmente, dificuldades em reconhecer que o movimento é impulsionado pelas rodas dentadas e a constatar que o tamanho do fio influencia a velocidade do pêndulo. Pode-se concluir que os recursos digitais, de um modo geral, contribuíram para mobilizar o potencial de capacidades de PC e para a construção de conhecimentos científicos. O contributo deste estudo, embora modesto, prende-se com o potencial das TIC na Educação em Ciências dos 1º CEB, na promoção do pensamento crítico e de conhecimentos.

O objetivo deste trabalho foi utilizar diferentes técnicas químicas (dimetilsulfóxido, ácido clorídrico, acético e lático), técnicas mecânicas (banho ultrassônico, abrasão com pérolas de vidro, maceração com terra diatomácea, ruptor ultrassônico e imersão em nitrogênio líquido) e técnica enzimática (preparado enzimático comercial Glucanex®) para a recuperação de carotenoides a partir da ruptura da parede celular das leveduras Sporidiobolus pararoseus e Rhodotorula mucilaginosa isoladas de amostras ambientais. Para isso a obtenção de biomassa foi realizada através de cultivos submersos no meio YM, a 25 °C, 180 rpm por 168 h. Para a ruptura celular, a operação de congelamento da biomassa (-18°C por 48 h) foi estudada. Os métodos de secagem convencional por ar forçado (35°C/48h) e liofilização (-80°C/48h, em ultrafreezer, seguido de liofilizador até alcançar 2% de umidade da amostra) também foram avaliados. Nas técnicas químicas aplicadas, o dimetilsulfóxido apresentou os melhores resultados para as duas leveduras, porém o seu uso é limitado devido a sua toxicidade. Para S. pararoseus, os maiores valores encontrados foram para o ácido clorídrico, seguido do acético e do lático, sendo detectada diferença entre eles quando aplicado o congelamento. Com R. mucilaginosa, os maiores valores foram encontrados para os ácidos acético e lático, seguido do ácido clorídrico, no qual o congelamento da biomassa também não influenciou a recuperação dos carotenoides. Dentre as técnicas mecânicas estudadas, para a levedura S. pararoseus, o banho ultrassônico e a abrasão com pérolas de vidro apresentaram os resultados mais promissores comparados ao DMSO (84,79±2,34 e 76,87±2,06 μg/g respectivamente), onde o processo de congelamento da biomassa não influenciou positivamente no percentual de extratibilidade e na concentração específica dos carotenoides quando utilizada estas técnicas. Com Rhodotorula mucilaginosa, o banho ultrassônico propiciou a recuperação da maior concentração específica de carotenoides (193,5±25,8 μg/g), sendo que o processo de congelamento também não influenciou positivamente no percentual de extratibilidade e na concentração específica dos carotenoides. Através do Delineamento Central Composto Rotacional (DCCR) 23 foi possível avaliar que a levedura S. pararoseus não demonstrou nenhum efeito sob as variáveis pH, temperatura e concentração de enzima. Assim, a melhor condição de trabalho escolhida foi pH 7,4, 30 ºC e concentração de enzima de 1,0 g/gcs, onde apresentou a concentração específica de 42,6 μg/g e volumétrica de 308 μg/L de carotenoides. Para R. mucilaginosa, a condição ótima foi definida como 1,0 g/gcs, pH 5,0 e temperatura de 30 ºC, onde foi encontrado 115,1±8,1 μg/g e 470,1±38,8 μg/L para a concentração específica e volumétrica de carotenoides, respectivamente. A utilização de técnicas combinadas empregando banho ultrassônico e lise enzimática não proporcionou melhorias nos resultados para ambas as leveduras. A liofilização provocou um ganho de 20% e 13,7% na concentração específica dos carotenoides das leveduras S. pararoseus e R. mucilaginosa, respectivamente, onde o congelamento da biomassa não influenciou significativamente (p<0,05) a recuperação de carotenoides provenientes das duas leveduras, podendo ser eliminada do processo. Assim, para S. pararoseus o banho ultrassônico e as pérolas de vidro apresentaram os melhores resultados na recuperação de carotenoides, e para R. mucilaginosa o melhor resultado foi alcançado com o banho ultrassônico.

Cabomba caroliniana is a submersed macrophyte that has become a serious invader. Cabomba predominantly spreads by stem fragments, in particular through unintentional transport on boat trailers ('hitch hiking'). Desiccation resistance affects the potential dispersal radius. Therefore, knowledge of maximum survival times allows predicting future dispersal. Experiments were conducted to assess desiccation resistance and survival ability of cabomba fragments under various environmental scenarios. Cabomba fragments were highly tolerant of desiccation. However, even relatively low wind speeds resulted in rapid mass loss, indicating a low survival rate of fragments exposed to air currents, such as fragments transported on a boat trailer. The experiments indicated that cabomba could survive at least 3 h of overland transport if exposed to wind. However, even small clumps of cabomba could potentially survive up to 42 h. Thus, targeting the transport of clumps of macrophytes should receive high priority in management. The high resilience of cabomba to desiccation demonstrates the risk of continuing spread. Because of the high probability of fragment viability on arrival, preventing fragment uptake on boat trailers is paramount to reduce the risk of further spread. These findings will assist improving models that predict the spread of aquatic invasive macrophytes.
