Neisseria meningitidis expresses a range of lipooligosaccharide (LOS) structures, comprising of at least 13 immunotypes (ITs). Meningococcal LOS is subject to phase variation of its terminal structures allowing switching between ITs, which is proposed to have functional significance in disease. The objectives of this study were to investigate the repertoire of structures that can be expressed in clinical isolates, and to examine the role of phase-variable expression of LOS genes during invasive disease. Southern blotting was used to detect the presence of LOS biosynthetic genes in two collections of meningococci, a global set of strains previously assigned to lineages of greater or lesser virulence, and a collection of local clinical isolates which included paired throat and blood isolates from individual patients. Where the phase-variable genes lgtA, lgtC or IgtG were identified, they were amplified by PCR and the homopolymeric tracts, responsible for their phase-variable expression, were sequenced. The results revealed great potential for variation between alternate LOS structures in the isolates studied, with most strains capable of expressing several alternative terminal structures. The structures predicted to be currently expressed by the genotype of the strains agreed well with conventional immunotyping. No correlation was observed between the structural repertoire and virulence of the isolate. Based on the potential for LOS phase variation in the clinical collection and observations with the paired patient isolates, our data suggest that phase variation of LOS structures is not required for translocation between distinct compartments in the host. Background: Thalamotomy has been reported to be successful in ameliorating the motor symptoms of tremor and/or rigidity in people with Parkinson's disease (PD), emphasising the bona fide contribution of this subcortical nucleus to the neural circuitry subserving motor function. Despite evidence of parallel yet segregated associative and motor cortico-subcortical-cortical circuits, comparatively few studies have investigated the effects of this procedure on cognitive functions. In particular, research pertaining to the impact of thalamotomy on linguistic processes is fundamentally lacking. Aims: The purpose of this research was to investigate the effects of thalamotomy in the language dominant and non-dominant hemispheres on linguistic functioning, relative to operative theoretical models of subcortical participation in language. This paper compares the linguistic profiles of two males with PD, aged 75 years (10 years of formal education) and 62 years (22 years of formal education), subsequent to unilateral thalamotomy procedures within the language dominant and non-dominant hemispheres, respectively. Methods & Procedures: Comprehensive linguistic profiles comprising general and high-level linguistic abilities in addition to on-line semantic processing skills were compiled up to 1 month prior to surgery and 3 months post-operatively, within perceived on'' periods (i.e., when optimally medicated). Pre- and post-operative language performances were compared within-subjects to a group of 16 non-surgical Parkinson's controls (NSPD) and a group of 16 non-neurologically impaired adults (NC). Outcomes & Results: The findings of this research suggest a laterality effect with regard to the contribution of the thalamus to high-level linguistic abilities and, potentially, the temporal processing of semantic information. This outcome supports the application of high-level linguistic assessments and measures of semantic processing proficiency to the clinical management of individuals with dominant thalamic lesions. Conclusions: The results reported lend support to contemporary theories of dominant thalamic participation in language, serving to further elucidate our current understanding of the role of subcortical structures in mediating linguistic processes, relevant to cortical hemispheric dominance.

Sonic Hedgehog is a secreted morphogen involved in patterning a wide range of structures in the developing embryo. Disruption of the Hedgehog signalling cascade leads to a number of developmental disorders and plays a key role in the formation of a range of human cancers. The identification of genes regulated by Hedgehog is crucial to understanding how disruption of this pathway leads to neoplastic transformation. We have used a Sonic Hedgehog (Shh) responsive mouse cell line, C3H/10T1/2, to provide a model system for hedgehog target gene discovery. Following activation of cell cultures with Shh, RNA was used to interrogate microarrays to investigate downstream transcriptional consequences of hedgehog stimulation. As a result 11 target genes have been identified, seven of which are induced (Thrombomodulin, GILZ, BF-2, Nr4a1, IGF2, PMP22, LASP1) and four of which are repressed (SFRP-1, SFRP-2, Mip1-gamma, Amh) by Shh. These targets have a diverse range of putative functions and include transcriptional regulators and molecules known to be involved in regulating cell growth or apoptosis. The corroboration of genes previously implicated in hedgehog signalling, along with the finding of novel targets, demonstrates both the validity and power of the C3H/10T1/2 system for Shh target gene discovery.

One of the most important advantages of database systems is that the underlying mathematics is rich enough to specify very complex operations with a small number of statements in the database language. This research covers an aspect of biological informatics that is the marriage of information technology and biology, involving the study of real-world phenomena using virtual plants derived from L-systems simulation. L-systems were introduced by Aristid Lindenmayer as a mathematical model of multicellular organisms. Not much consideration has been given to the problem of persistent storage for these simulations. Current procedures for querying data generated by L-systems for scientific experiments, simulations and measurements are also inadequate. To address these problems the research in this paper presents a generic process for data-modeling tools (L-DBM) between L-systems and database systems. This paper shows how L-system productions can be generically and automatically represented in database schemas and how a database can be populated from the L-system strings. This paper further describes the idea of pre-computing recursive structures in the data into derived attributes using compiler generation. A method to allow a correspondence between biologists' terms and compiler-generated terms in a biologist computing environment is supplied. Once the L-DBM gets any specific L-systems productions and its declarations, it can generate the specific schema for both simple correspondence terminology and also complex recursive structure data attributes and relationships.

Pili of Neisseria meningitidis are a key virulence factor, being the major adhesin of this capsulate organism and contributing to specificity for the human host. Pili are post-translationally modified by addition of either an O-linked trisaccharide, Gal (beta1-4) Gal (alpha1-3) 2,4-diacetamido-2,4,6-trideoxyhexose or an O-linked disaccharide Gal (alpha1,3) GlcNAc. The role of these structures in meningococcal pathogenesis has not been resolved. In previous studies we identified two separate genetic loci, pglA and pglBCD, involved in pilin glycosylation. Putative functions have been allocated to these genes; however, there are not enough genes to account for the complete biosynthesis of the described structures, suggesting additional genes remain to be identified. In addition, it is not known why some strains express the trisaccharide structure and some the disaccharide structure. In order to find additional genes involved in the biosynthesis. of these structures, we used the recently published group A strain Z2491 and group B strain MC58 Neisseria meningitidis genomes and the unfinished Neisseria meningitidis group C strain FAM18 and Neisseria gonorrhoeae strain FA1090 genomes to identify novel genes involved in pilin glycosylation, based on homology to known oligosaccharide biosynthetic genes. We identified a new gene involved in pilin glycosylation designated pglE and examined four additional genes pgIB/B2, pglF, pglG and pglH. A strain survey revealed that pglE and pglF were present in each strain examined. The pglG, pglH and pgIB2 polymorphisms were not found in strain C311#3 but were present in a large number of clinical isolates. Insertional mutations were constructed in pglE and pglF in N. meningitidis strain C311#3, a strain with well-defined lipopolysaccharide (LPS) and pilin-linked glycan structures. Increased gel migration of the pilin subunit molecules of pglE and pglF mutants was observed by Western analysis, indicating truncation of the trisaccharide structure. Antisera specific for the C311#3 trisaccharide failed to react with pilin from these pglE and pglF mutants. GC-MS analysis of the sugar composition of the pglE mutant showed a reduction in galactose compared with C311#3 wild type. Analysis of amino acid sequence homologies has suggested specific roles for pglE and pglF in the biosynthesis of the trisaccharide structure. Further, we present evidence that pglE, which contains heptanucleotide repeats, is responsible for the phase variation between trisaccharide and disaccharide structures in strain C311#3 and other strains. We also present evidence that pglG, pglH and pgIB2 are potentially phase variable.

The G-77, BASIC, and global climate governance: a new era in multilateral environmental negotiations

The G-77 has historically organized the participation of developing countries in multilateral environmental negotiations. This article analyses the impact of a new coalition of emerging powers - Brazil, China, India, and South Africa as BASIC - on the G-77's role in climate governance. While there are important benefits for both sides in their relationship, I argue that the G-77 is also disadvantaged in several concrete ways by the BASIC countries.

O presente trabalho teve como principal objectivo a aplicação dos conceitos adquiridos nas unidades curriculares da licenciatura e do mestrado. Nesse sentido tomou-se a opção da realização de um projecto de execução de uma obra de arte para o trabalho final de mestrado, realizado num conceituado gabinete de projecto de pontes e viadutos, com vista à obtenção do grau de mestre. O projecto refere-se a uma obra de arte que se insere num nó rodoviário de ligação entre duas auto-estradas que, dados os seus condicionamentos de perfil, quer longitudinal quer transversal, e implantação no local, pode ser considerada uma obra de arte especial. O processo de análise foi feito com recurso ao programa de cálculo automático SAP2000®, tendo em conta os regulamentos nacionais aplicáveis a estruturas de pontes e os Eurocódigos em fase de implementação em Portugal. O projecto de execução, objecto deste trabalho, foi realizado em função de uma metodologia de construção simples, que é o recurso à utilização, por fases, de cimbres apoiados no solo. A optimização das fases de construção é um factor condicionante na análise e concepção da obra de arte deste projecto, pelo que se procura obter uma relação optimizada entre o rendimento de construção e o funcionamento estrutural da obra de arte, de forma a garantir a segurança da estrutura e o conforto dos utilizadores a longo prazo.

A adequada previsão do comportamento diferido do betão, designadamente da retracção, é essencial no projecto de uma obra de grandes dimensões, permitindo conceber, dimensionar e adoptar as disposições construtivas para um comportamento estrutural que satisfaça os requisitos de segurança, utilização e durabilidade. O actual momento é marcado por uma transição em termos da regulamentação de estruturas, com a eminente substituição da regulamentação nacional por regulamentação europeia. No caso das estruturas de betão, o Regulamento de Estruturas de Betão Armado e Pré-Esforçado (REBAP), em vigor desde 1983, será substituído pelo Eurocódigo 2. Paralelamente, a Federation International du Betón publicou o Model Code 2010 (MC2010), um documento que certamente terá forte influência na evolução da regulamentação das estruturas de betão. Neste contexto, o presente trabalho tem como objectivo estabelecer uma comparação entre os diferentes modelos de previsão da retracção incluídos nos documentos normativos referidos, identificando as principais diferenças e semelhanças entre eles e quantificando a influência dos diferentes factores considerados na sua formulação, de forma a avaliar o impacto que a introdução destes modelos de previsão irá ter no projecto de estruturas de betão. Com o propósito de aferir a forma como estes modelos reflectem a realidade do fenómeno em estudo, procedeu-se à aplicação destes modelos de previsão ao betão de duas obras cujo comportamento estrutural é observado pelo LNEC, concretamente a ponte Miguel Torga, sobre o rio Douro, na Régua, e a ponte sobre o rio Angueira, no distrito de Bragança. Em ambas as obras tinha sido efectuada a caracterização in situ da retracção, tendo-se comparado os valores experimentais assim obtidos com os valores provenientes da aplicação dos modelos de previsão considerados neste trabalho. Finalmente são apresentadas algumas conclusões obtidas com o trabalho desenvolvido nesta dissertação, bem como algumas sugestões para desenvolvimentos futuros.

O presente trabalho, refere-se ao projecto de estabilidade, em betão armado e pré-esforçado, da Escola Superior de Enfermagem Artur Ravara, situada na zona da EXPO em Lisboa. O edifício apresenta-se com uma implantação em “L”, tendo como dimensões máximas 38,50m x 54,80m e desenvolve-se em altura por quatro pisos, dos quais, dois são enterrados. A estrutura do edifício em causa, apresenta duas juntas de dilatação, por forma a tornar desprezáveis os efeitos devidos à retracção e diminuição de temperatura, dividindo o edifício em três blocos. As suas fundações são indirectas, constituídas por estacas moldadas no terreno e respectivos maciços de encabeçamento. As lajes são fungiformes aligeiradas de moldes perdidos, de modo a permitir vencer maiores vãos, que variam entre os 6,60m e os 10,00m, e permitindo também maior rapidez de execução e maior economia. As consolas de 3,50m de vão, em laje maciça, são suportadas por vigas pré-esforçadas de secção variável. Para o cálculo automático da estrutura e da obtenção dos respectivos desenhos das armaduras, foi utilizado o programa de cálculo automático, Tricalc 7.1. O conteúdo do projecto em questão, sendo de carácter académico, não corresponde à versão real, à qual não se teve acesso. O dimensionamento das fundações, devido à fraca resistência dos solos e o dimensionamento da estrutura, devido à geometria e dimensões do edifício, permitiram enfrentar desafios interessantes. Tais desafios, deram possibilidade de enriquecer bastante os conhecimentos sobre a engenharia de estruturas.
