Les nombreuses études cliniques sur les maladies affectant les noyaux gris centraux (NGC) ont montré l’importance des déficits moteurs engendrés par un dysfonctionnement de ces structures. La locomotion est particulièrement affectée chez ces patients, ce qui suggère un rôle important des NGC dans le contrôle de celle-ci. En revanche, très peu d’études fondamentales ont enregistré l’activité des neurones des NGC durant la locomotion chez des modèles intacts. Le globus pallidus, en tant qu’une des principales structures de sortie des NGC, constitue une cible idéale pour étudier le signal transmis par les NGC au thalamus et au tronc cérébral. L’objectif de notre étude est de mieux comprendre la contribution que joue le segment externe du globus pallidus (GPe) dans le contrôle de la locomotion sous guidage visuel. Cette étude constitue le premier rapport d’enregistrements de l’activité des neurones du GPe lors de modifications volontaires de la marche sous guidage visuel. Notre hypothèse générale est que le GPe contribue au contrôle de la locomotion, et particulièrement lors des modifications de la marche. Nous avons enregistré l’activité des neurones du GPe de chats au repos, puis marchant sur un tapis et enjambant des obstacles. Nos résultats ont révélé que la plupart des cellules montraient peu de changements lors de la locomotion non-obstruée par rapport au repos. En revanche, la moitié des neurones enregistrés présentaient une modulation significative de leur décharge durant l’enjambement de l’obstacle. Nous avons observé une majorité de ces cellules qui diminuaient leur fréquence et un tiers de neurones qui l’augmentaient pendant le pas modifié, suggérant que le GPe exerce sur le mouvement un contrôle dépendant du contexte, permettant sélectivement l’augmentation ou la diminution de l’activité motrice. De plus, presque toutes les cellules déchargeaient durant la totalité de la phase de balancement du membre antérieur controlatéral, ce qui semble indiquer une contribution du GPe dans le timing et la durée de cette phase. Finalement, la moitié des neurones présentaient un changement réciproque entre la condition de lead et la condition de trail. Ce résultat suggère que le GPe participerait à la sélection d’action du membre lors des modifications de la marche. Nos résultats illustrent la complexité de l’activité du GPe lors de la locomotion sous guidage visuel et ils sont compatibles avec une contribution plus générale au mouvement que celle exercée par le cortex moteur.

L’incidence constante des maladies liées à l’âge reflète un réel enjeu dans nos sociétés actuelles, principalement lorsqu’il est question des cas de cancers, d’accidents cérébraux et de maladies neurodégénératives. Ces désordres sont liés à l’augmentation de l’espérance de vie et à un vieillissement de la population. Les coûts, estimés en milliards de dollars, représentent des sommes de plus en plus importantes. Bien que les efforts déployés soient importants, aucun traitement n’a encore été trouvé. Les maladies neurodégénératives, telles que la maladie d’Alzheimer, de Parkinson, d’Huntington ou la sclérose latérale amyotrophique (SLA), caractérisées par la dégénérescence d’un type neuronal spécifique à chaque pathologie, représentent un défi important. Les mécanismes de déclenchement de la pathologie sont encore nébuleux, de plus il est maintenant clair que certains de ces désordres impliquent de nombreux gènes impliqués dans diverses voies de signalisation induisant le dysfonctionnement de processus biologiques importants, tel que le métabolisme. Dans nos sociétés occidentales, une problématique, directement lié à notre style de vie s’ajoute. L’augmentation des quantités de sucre et de gras dans nos diètes a amené à un accroissement des cas de diabètes de type II, d’obésité et de maladies coronariennes. Néanmoins, le métabolisme du glucose, principale source énergétique du cerveau, est primordial à la survie de n’importe quel organisme. Lors de ces travaux, deux études effectuées à l’aide de l’organisme Caenorhabditis elegans ont porté sur un rôle protecteur du glucose dans un contexte de vieillissement pathologique et dans des conditions de stress cellulaire. Le vieillissement semble accéléré dans un environnement enrichi en glucose. Cependant, les sujets traités ont démontré une résistance importante à différents stress et aussi à la présence de protéines toxiques impliquées dans la SLA et la maladie de Huntington. Dans un deuxième temps, nous avons démontré que ces effets peuvent aussi être transmis à la génération suivante. Un environnement enrichi en glucose a pour bénéfice de permettre une meilleure résistance de la progéniture, sans pour autant transmettre les effets néfastes dû au vieillissement accéléré.

This study documents recovery status and symptom changes in a one-year follow-up of sexually abused (SA) adolescent girls in child protection services in the province of Québec, Canada. Sixteen French-speaking participants were interviewed using the Multidimensional Trauma Recovery and Resiliency Interview (MTRR-I), which was in turn rated by interviewers using the companion rating scale, the MTRR, and completed questionnaires assessing symptoms, types of maltreatment endured and services received. Participants were between the ages of 13 and 17 years (M = 15.2 years) when first interviewed and had experienced severe sexual abuses. Analyses of one-year follow-up data revealed statistically significant changes towards better functioning on multiple domains and less symptomatology for a majority of the girls interviewed. The recovery status of a minority of research participants seems to have worsened in the interval. The discussion considers these findings and addresses relevancy of the MTRR measures in cases of SA adolescents.

This thesis covers various aspects of viral diseases affecting shrimp aquaculture. The research component of this thesis can be divided into four areas. The areas covered are: I) A study to determine the prevalence of WSSV among the crustaceans in the Vembanad estuary, the shrimp aquaculture farms surrounding the estuary, and the sea off Cochin coast, India using two , sets of nested PCR primers. 2) An investigation to compare the sequence of six major structural proteins of WSSV; vp28, vp26, vp 19, vp68, vp281, vp466 from different geographical locations with that of an isolate from India. 3) Simultaneous occurrence of HPV, IHHNV, MBV and WSSV in postlarvae of P. monodon from hatcheries in India was monitored by Polymerase Chain Reaction. 4) A real time PCR procedure was developed for the quantitative analysis of WSSV infection. The viral load of postlarvae from hatcheries in Kerala meant for aquaculture was also determined using the quantitative PCR.

There is no baseline data available at present on the nature of various diseases that occur in a orchid population, under cultivation, in any commercial orchid farm maintained by small scale entrepreneurs who invest considerable amount of money, effort and time. The available data on type of disease symptoms, causative agent, , nature of pathogens, as to bacteria or ftmgi or any other biological agents, and their source, appropriate and effective control measures could not be devised, for large scale implementation and effective management, although arbitrary methods are being practiced by very few farms. Further influence of seasonal variations and environmental factors on disease outbreak is also not scientifically documented and statistically verified as to their authenticity. In this context, the primary objective of the present study was to create a data bank on the following aspects 1. Occurrence of different disease symptoms in Dendrobium hybrid over a period of one year covering all seasons 2. Variations in the environmental parameters at the orchid farms 3. Variations in the characteristics of water used for irrigation in the selected orchid farm 4. Microbial population associated with the various disease symptoms 5. Isolation and identification of bacteria isolated from diseased plants 6. Statistical treatment of the quantitative data and evolving statistical model

In forestry, availability of healthy seeds is an important factor in raising planting stock. Initial seed health and storage conditions are the major factors governing the germinability of seeds. Like seeds of agricultural and horticultural crops, forest tree seeds are also liable to be affected by micro-organisms during storage, which affects the germination, and reduces the viability. Further introduction of seed-borne diseases into newly sown crops/areas on account of using unhealthy seeds is also not ruled out. Availability of healthy stock of seedlings is intrinsic for raising plantations and to meet this requirement elimination of nursery diseases by appropriate chemicals is of prime imortance. As exotic tree species may become susceptible to various native pathogens, it is generally considered better to select indigenous tree species for large scale plantations as they are well adapted to local environment. However, before taking up large scale afforestation progranme involving any indigenous tree species, it is essential to have knowledge about seed disorders and seedling diseases and their management. with a View to select appropriate tree species with fewer seed disorders and seedling disease problems for use in further plantation programme, four indigenous tree species such as Albizia odoratissima (L.f) Benth., Lagerstroemia microcazpa Wt., Pterocazpus marsupiwn Roxb. and Xylia xylocarpa (Roxb.) Taub. were evaluated to meet the above parameters

Bamboos are vulnerable to various diseases which affect them in nurseries, plantations as well as in natural stands. In India, rot and blight of emerging culms have already been identified as the limiting factor of the bamboo production in many bamboo growing areas, especially in the coastal belts of Orissa (Jamaluddin et a1., 1992). Similarly, foliage blight and rust have been recorded to pose threat to nursery as well as outplanted seedlings which are in the early establishnent phase (Bakshi et a1., 1972; Harsh et a1., 1989). With the increased emphasis and priority on raising multipurpose tree species, large—scale planting of bamboos has been initiated recently in the State. Limited experience in raising the bamboo seedlings together with the lack of information on bamboo diseases and their control measures often resulted in partial to complete failure of many nurseries. Also, poor handling of bareroot seedlings for outplanting affected seriously the planting programme. This was clearly reflected by the large-scale nortality of outplanted young seedlings reported from many plantations. So far, no systanatic attempt has been made to study the diseases affecting bamboos in nurseries, plantations and natural stands in the country. Hence, the present investigation was taken up to conduct a systematic study of the diseases affecting bamboos in Kerala.

In the present study, fifteen typee of diseases occurring in twenty one species of finfishes end shellfishes are documented and the causes studied. All these are being reported from India for the first time which adds considerably to our existinq knowledge on the subject.In fact, the candidate's studies embodied in this Thesis has also an applied significance aimed at the proper management_of finfish and shellfish aquaculture systems in this country.The thesis is presented in seven main chapters.

The prevalence and genetic susceptibility of autoimmune diseases (ADs) may vary depending on latitudinal gradient and ethnicity. The aims of this study were to identify common human leukocyte antigen (HLA) class II alleles that contribute to susceptibility to six ADs in Latin Americans through a meta-analysis and to review additional clinical, immunological, and genetic characteristics of those ADs sharing HLA alleles. DRB1∗03:01 (OR: 4.04; 95%CI: 1.41–11.53) was found to be a risk factor for systemic lupus erythematosus (SLE), Sjogren’s syndrome (SS), and type 1 diabetes mellitus (T1D). DRB1 ¨ ∗04:05 (OR: 4.64; 95%CI: 2.14–10.05) influences autoimmune hepatitis (AIH), rheumatoid arthritis (RA), and T1D; DRB1∗04:01 (OR: 3.86; 95%CI: 2.32–6.42) is a susceptibility factor for RA and T1D. Opposite associations were found between multiple sclerosis (MS) and T1D. DQB1∗06:02 and DRB1∗15 alleles were risk factors for MS but protective factors for T1D. Likewise, DQB1∗06:03 allele was a risk factor for AIH but a protective one for T1D. Several common autoantibodies and clinical associations as well as additional shared genes have been reported in these ADs, which are reviewed herein. These results indicate that in Latin Americans ADs share major loci and immune characteristics.

The age at onset refers to the time period at which an individual experiences the first symptoms of a disease. In autoimmune diseases (ADs), these symptoms can be subtle but are very relevant for diagnosis. They can appear during childhood, adulthood or late in life and may vary depending on the age at onset. Variables like mortality and morbidity and the role of genes will be reviewed with a focus on the major autoimmune disorders, namely, systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), multiple sclerosis (MS), type 1 diabetes mellitus (T1D), Sjögren's syndrome, and autoimmune thyroiditis (AITD). Early age at onset is a worst prognostic factor for some ADs (i.e., SLE and T1D), while for others it does not have a significant influence on the course of disease (i.e., SS) or no unanimous consensus exists (i.e., RA and MS).

Epigenetics is defined as the study of all inheritable and potentially reversible changes in genome function that do not alter the nucleotide sequence within the DNA. Epigenetic mechanisms such as DNA methylation, histone modification, nucleosome positioning, and microRNAs (miRNAs) are essential to carry out key functions in the regulation of gene expression. Therefore, the epigenetic mechanisms are a window to understanding the possible mechanisms involved in the pathogenesis of complex diseases such as autoimmune diseases. It is noteworthy that autoimmune diseases do not have the same epidemiology, pathology, or symptoms but do have a common origin that can be explained by the sharing of immunogenetic mechanisms. Currently, epigenetic research is looking for disruption in one or more epigenetic mechanisms to provide new insights into autoimmune diseases. The identification of cell-specific targets of epigenetic deregulation will serve us as clinical markers for diagnosis, disease progression, and therapy approaches.

Polyautoimmunity is one of the major clinical characteristics of autoimmune diseases (ADs). The aim of this study was to investigate the prevalence of ADs in spondyloarthropathies (SpAs) and vice versa. This was a two-phase cross-sectional study. First, we examined the presence of ADs in a cohort of patients with SpAs (). Second, we searched for the presence of SpAs in a well-defined group of patients with ADs () including rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), and Sjögren’s syndrome (SS). Among patients with SpAs, ankylosing spondylitis was observed in the majority of them (55.6%). There were two patients presenting with SS in the SpA group (1.4%) and 5 patients with autoimmune thyroiditis (3.5%). The global prevalence of ADs in SpAs was 4.86%. In the ADs group, there were 5 patients with SpAs (0.46%). Our results suggest a lack of association between SpAs and ADs. Accordingly, SpAs might correspond more to autoinflammatory diseases rather than to ADs.
