A significant proportion of the human population suffers from some form of skin disorder, whether it be from burn injury or inherited skin anomalies. The ideal treatment for skin disorders would be to regrow skin tissue from stem cells residing in the individual patient's skin. Locating these adult stem cells and elucidating the molecules involved in orchestrating the production of new skin cells are important steps in devising more-efficient methods of skin production and wound healing via the ex vivo expansion of patient keratinocytes in culture. This review focuses on the structure of the skin, the identification of skin stem cells, and the role of Notch, Wnt and Hedgehog signalling cascades in regulating the fate of epidermal stem cells. © 2005 Cambridge University Press.

Liver regeneration is an orchestrated cellular response that coordinates cell activation, lipid metabolism, and cell division. We found that caveolin-1 gene - disrupted mice (cav1(-/-) mice) exhibited impaired liver regeneration and low survival after a partial hepatectomy. Hepatocytes showed dramatically reduced lipid droplet accumulation and did not advance through the cell division cycle. Treatment of cav1(-/-) mice with glucose ( which is a predominant energy substrate when compared to lipids) drastically increased survival and reestablished progression of the cell cycle. Thus, caveolin-1 plays a crucial role in the mechanisms that coordinate lipid metabolism with the proliferative response occurring in the liver after cellular injury.

In order to begin to understand how primary olfactory and vomeronasal organ (VNO) axons target specific regions of the olfactory bulb, we examined the sorting behaviour of these axons following neonatal unilateral olfactory bulbectomy. Bulbectomy induced widespread ipsilateral death of the primary olfactory and VNO neurons. After 4 weeks, many new sensory axons had re-grown into the cranial cavity and established a prominent plexus with evidence of dense tufts that were similar in gross appearance to glomeruli. Axons expressing the cell adhesion molecule OCAM, which normally innervate the ventrolateral and rostral halves of the main and accessory olfactory bulbs, respectively, sorted out and segregated from those axons not expressing this molecule within the plexus. In addition, VNO axons formed large discrete bundles that segregated from main olfactory axons within the plexus. Thus, VNO and primary olfactory axons as well as discrete subpopulations of both are able to sort out and remain segregated in the absence of the olfactory bulb. Sorting and convergence of axons therefore occur independently of the olfactory bulb and are probably attributable either to inherent properties of the axons themselves or to interactions between the axons and accompanying glial ensheathing cells.

Neonatal X-irradiation of central nervous system (CNS) tissue markedly reduces the glial population in the irradiated area. Previous in vivo studies have demonstrated regenerative success of adult dorsal root ganglion (DRG) neurons into the neonatally-irradiated spinal cord. The present study was undertaken to determine whether these results could be replicated in an in vitro environment. The lumbosacral spinal cord of anaesthetised Wistar rat pups, aged between 1 and 5 days, was subjected to a single dose (40 Gray) of X-irradiation. A sham-irradiated group acted as controls. Rats were allowed to reach adulthood before being killed. Their lumbosacral spinal cords were dissected out and processed for sectioning in a cryostat. Cryosections (10 mum-thick) of the spinal cord tissue were picked up on sterile glass coverslips and used as substrates for culturing dissociated adult DRG neurons. After an appropriate incubation period, cultures were fixed in 2% paraformaldehyde and immunolabelled to visualise both the spinal cord substrate using anti-glial fibrillary acidic protein (GFAP) and the growing DRG neurons using anti-growth associated protein (GAP-43). Successful growth of DRG neurites was observed on irradiated, but not on non-irradiated, sections of spinal cord. Thus, neonatal X-irradiation of spinal cord tissue appears to alter its environment such that it can later support, rather than inhibit, axonal regeneration. It is suggested that this alteration may be due, at least in part, to depletion in the number of and/or a change in the characteristics of the glial cells. (C) 2000 ISDN. Richard Wagner concebia a arte como uma atividade similar à religião, que deveria conduzir o ser humano à reflexão sobre as questões principais de sua existência e levá-lo ao aperfeiçoamento. Wagner sempre foi obcecado pela ideia da redenção e a preocupação do compositor com a regeneração do ser humano perpassa toda a sua obra. Os conceitos religiosos de Wagner, presentes em sua obra musical e em seus ensaios literários, reúnem tradições cristãs e budistas, ideias políticas e preceitos mitológicos que delineiam o seu credo pessoal, uma forma de religião sincrética na qual a arte tem o seu lugar como elemento de transcendência, cumprindo a função de interpretar os símbolos míticos para torna-los compreensíveis à percepção do espírito humano. Os ideais artísticos de Wagner vão ao encontro do pensamento de Paul Tillich e a sua Teologia da Cultura. Tillich afirma que a religião não está restrita aos limites dos templos religiosos ou aos domínios institucionais, mas encontra-se em qualquer expressão humana na qual se manifeste a preocupação suprema . Ela pode ser reconhecida em qualquer situação onde se encontre o elemento incondicional, nas manifestações da criatividade humana e na cultura, na busca honesta da verdade ou na procura de solução para as adversidades da existência. Portanto, o objetivo desse estudo é buscar no pensamento tillichiano uma correlação teológica para os anseios de redenção evidenciados na obra de arte wagneriana.

The Foot-and-Mouth Disease (FMD) outbreak of 2001 in the UK was completely unprecedented in its scale and severity, with over four million animals culled and a cost to the Exchequer of over £4 billion. Local authorities were at the front line in dealing with the outbreak, in coordinating the cull of livestock, the disposal of carcasses as well as attempting to deal with its aftermath and, in particular, the impact on the wider rural economy. This article examines the impacts of this crisis on three local authorities, Devon, Herefordshire and Cumbria. It examines how far the crisis acted as a catalyst in developing strategies to deal with a future outbreak as well as new local initiatives to promote regeneration in the areas most adversely affected. It focuses on developments that can be directly attributed to the crisis and shows that FMD had a considerable impact on communications and 'joined-up' activity within local authorities and with local stakeholders. © 2006, LEPU, South Bank University.

The 12-month radial growth of Parmelia conspersa thalli with isidia or with apothecia and isidia was not influenced by removal of the thalli centres. When large thalli had their centres removed and the thallus perimeter was divided into fragments of about 1.0cm in diameter, growth of the fragments was less than the controls, but recovered to near control values after four or five months growth. These results suggest first, that fixed carbon for radial growth may be made in a narrow annulus at the perimeter and second, that there may be little transfer of fixed carbon between the annulus and the centre of the thallus ar around the annulus. Fragments of the centre and the perimeter regenerated growing points, suggesting that fragmentation may be an important method of vegetative reproduction in some lichens.
