The counseling on HIV/Aids consists in a prevention strategy that contributes to increase the diagnosis of HIV and start earlier the treatment. The counseling has as pillars the emotional and educational support, risks evaluation that aim at the adoption of safe practices and the individual s responsibility for his own health. To accomplish these results, it is necessary that health workers understand counseling as a unique educational moment that stimulates the user s critical-reflection when it comes to his role as an active subject in this process. This study aimed to analyze the counseling on HIV/Aids conducted by the professionals of the Testing and Counseling Center (CTA), based on the educational perspective of Paulo Freire . This is a descriptive qualitative study with a critical reflexive design based on the principles of Action-Science. All the professionals acting as counselors in the Joao Pessoa, PB CTA, eight in total, took part in the study. Data were collected during the month of March, 2011, through non participative observation and semi-structured interviews with a critical-reflexive focus, analyzed according to the tenets of the critical-reflexive methodology, and discussed taking into consideration the Paulo Freire s pedagogy and pertinent literature. It was observed that most of the professionals expressed the work philosophy of CTA as the diagnosis and prevention of the disease, associated with the utilization and demonstration of condoms. However, upon observation of their counseling sessions, these ideas were not converted in actions. Educational themes were not covered and the condom wasn t offered at any time. The counseling actions focused on the provision of information and filling out the paper forms which are necessary for attendance. The sessions were conducted with brief dialogues and little opportunity for the users to expose or complement their thoughts and needs. The professionals mentioned as facilitating conditions for counseling, the team interaction and physical structure. The difficulties focused on the users low cognition, the large demand for attendance, aspects related to the service organization, and the counselors absences and delays. After reflecting about the actions observed in the counseling, the majority of professionals admitted the need to modify their practice in the incorporation of educational principles for the achievement of a broader prevention, and seemed to be willing to work in this perspective. In conclusion, although the counselors show ideas consistent with the purposes of CTA, these ideas are limited when it comes to the understanding of the meaning of prevention in HIV/Aids. Taking into consideration that they express a certain comprehension and act differently during the counseling, they demonstrate a lack of bond between the theories in use and the proposed ones, in accordance with the contribution of the action-science theory. The counseling, as an educative practice, doesn t materialize in the counseling itself and the orientation for reflection is not given during the attendance. These findings suggest the need to include the process of reflection in the execution of the actions of counseling, so that these practices are guided by reflexive practice, aiming at transforming the way of thinking and acting into a more educational perspective toward a more democratic and holistic assistance.

This research aimed to study competition effects of legumes intercropping and weeds on seeds yield and quality of Sorghum bicolor (L.) Moench. The experiment was carried out in a Rhodic Nitisol, under field condition in Botucatu region, São Paulo State, Brazil. Ten treatments were tested: sorghum (cv. A-6304) intercropped with Dolichos lablab L., Mucuna aterrima (Piper et Tracy) Holland, Canavalia ensiformis D. C. or Crotalaria juncea L., that were sown in the same time of sorghum or 20 days after the sorghum sowing, and sorghum weed-free and sorghum without weeding. The experimental design employed was the completely randomized blocks with four replications. There were evaluated sorghum seeds yield and some characteristcs of seed quality: purity, hectolitric weight, 1000 seeds weight, insect infestation, germination and vigor (by first counting of germination test and accelerated ageing test). The competition caused by legumes intercropping and weeds affected the yield and the physical and physiological characteristics of sorghum seeds, but the intensity of prejudicial effects depended on the time and the species of legume intercropping.

The present work was conducted in a fruit tree propagation area of the Plant Production Department of the Faculdade de Ciencias Agrarias e Veterinarias, Universidade Estadual Paulista (FCAV/UNESP) in Jaboticabal, SP, and also in a commercial orchard in Araguari, MG, with the objective to verify the potential of vegetative growth (stem diameter, height of plants and leaf number) of plants of passion fruit (Passiflora alata Dryander), gotten by cutting and seed, comparing the initial development of plants in the field. This experiment was carried out from January 2002 to February 2003. The experiment using seeds was conducted at a shadow house, and the one that used cuttings in an intermitent mist. The cuttings and seeds were collected from adult plants which came from Passifloraceae Active Germoplasm Bank (BAG) of the Plant Production Department of FCAC/UNESP. For the cuttings, it was used the intermediate part of the branches in stadium of vegetative growth. The seeds, in order to obtain the seedlings, had been sown in plastic trays. Cuttings and seedlings were transplanted to plastic bags with substrate in shadow house and with daily irrigation. They were acclimatized and planted on field, after 60 days. on field, the stem diameter, plant height and number of leaves were better for cuttings than for seedlings in Jaboticabal, SP. In Araguari, MG, stem diameter was larger in the seedlings, which plant heights and number of leaves were larger on cuttings.

The aim of this research was to identify methods for breaking dormancy and promoting germination of Stryphnodendron adstringens and S. polyphyllum seeds. Seeds of both species were submitted to the following treatments for breaking dormancy: control; mechanical scarification (sandpaper number 220); hot water (initial temperature of 87 degrees C) for 5 and 15 minutes; chemical scarification with H(2)SO(4) (95%) for 30, 45, 60, 75, 90 and 105 minutes; thermal shock 1 (0 degrees C and 50% RH for 1 h and thereafter 10 minutes into water Ti=85 degrees C, Tf=57 degrees C); thermic shock 2 (40 degrees C and 40% RH for 6 h and thereafter 10 minutes into water, Ti=0 degrees C, Tf=3 degrees C); thermal shock 3 (40 degrees C and 40% RH for 6h and thereafter 10 minutes into water, Ti=25 degrees C, Tf=25 degrees C). We concluded that seeds belonging to same genus but from different species answered differently to some treatments for breaking dormancy. Chemical scarification with H(2)SO(4) (45 minutes) and mechanical scarification with sandpaper are methods that can be employed with efficiency to break dormancy and promote germination for both species.

The objective of this research was to evaluate the growth of Tabebuia heptaphylla seedlings in distinct substrates with different amounts of urban waste compost and the relation between this growth and irrigation. A completely randomized experimental design was used, with a factorial arrangement of 15 substrates and 2 irrigation levels. The substrates were composed by the combination of different materials: urban waste, tanned cattle manure, vermiculite, soil and the commercial form Plantmax (R). For the study of the seedlings development, the following characteristics were evaluated: plant height, stem diameter at soil level, number of leaves, above ground dry matter, root system dry matter, relation between plant height and stem base diameter, Dickson quality index and relation between plant height and above ground dry matter. Evaluations of plant height, stem diameter at soil level and number of leaves were made at 75, 90, 105, 120, 135 and 150 days after sowing. According to the results, it was concluded that urban waste compost does not increase plant development. Significant differences in relation to the irrigation levels were found, with better results for the 150% irrigation level compared to 100% evapotranspiration.

With the objective of evaluating the biomass production and nutrient removal by plant cover in the Agreste region of Alagoas, an experiment was conducted in the experimental field of the Federal University of Alagoas - Campus Arapiraca. Randomized block design was used, with eight treatments and four replications. The treatments were: Crotalaria juncea, Crotalaria spectabilis, Cajanus cajan (L.) Mill sp., Cajanus cajan, Canavalia ensiformis, Dolichos lablab, Mucuna aterrima and the spontaneous local vegetation (control). The green matter in an area of 1 m(2) during the flowering of each species was evaluated, and biomass was then dried in an oven at 65 degrees C until constant weight for dry matter, in which the contents of macro and micronutrients were extracted. Leguminous plant showed potential for use as green manure in the Agreste region of Alagoas, with N contents higher than the spontaneous vegetation and not being different from one for the accumulation of P, K, Ca, Mg, S, B, Mn and Zn. The spontaneous vegetation was similar to dry matter of legumes production. Among the treatments Cajanus cajan showed higher dry matter production and N accumulation in the aerial part.

Foliar diagnosis is a method for assessing the nutritional status of agricultural crops, which helps in the understanding of soil fertility and rationalized application of fertilizers taking into account economic and environmental criteria. The study aimed to use the landrelief as criteria to assist in interpreting the spatial variability of nutrient content of the citrus leaf. The leaves were collected at regular intervals of 50 m, totaling 332 sampling points. Data were analyzed by descriptive statistics, geostatistics and induction of decision tree. With the aid of digital elevation model (MDE) and the profile planaltimetric, the area was divided into three different landrelief and sub-strands. The highest values for nutrients from the leaves of citrus were observed at the top (concave area) segments on a half-slope and lower slope. The nutrients from the citrus leaves showed high values of correlation (above 0.5) with the altitude of the study area. The technique of geostatistics and the induction of decision tree show that the relief is the variable with the greatest potential to interpret the maps of spatial variability of nutrients from the citrus leaves.

An experiment was carried out in greenhouse during the period January to April 2010, at Center of Agricultural Sciences of the Federal University of Paraiba, in Areia, Paraiba State, Brazil, in order to evaluate the effects of saline water and bovine biofertilizer on the seedling growth of Indian neem. The substrate was material of a non-saline soil collected in depth of 0-20 cm. The treatments were arranged in a completely randomized design using a 5 x 2 factorial, referring to salinity levels of irrigation water of 0.5, 1.0, 2.0, 3.0 and 4.0 dS m(-1), with and without bovine biofertilizer applied to the soil only once after dilution with water (1: 1), a day before sowing, in volume corresponding to 10% of the substrate. At 86 days after emergence of seedlings the plant growth in height and principal root length, diameter of stem and root, leaf number and dry mass of roots and shoots of plants were evaluated. The salinity of irrigation water increased the salinity levels in the substrate inhibiting the growth in height, stem diameter, leaf emission by plants, diameter and length of principal root and the dry matter production of roots and aerial parts (leaves + stem) of neem, but with less pronounced decrease in plants under the treatments with bovine biofertilizer.

One of needs of modern agriculture is the prediction of spatial variability of soil properties at more detailed scales for sustainable management and optimization of management practices. The mathematical model associated with knowledge of variability of soil attributes and mapping of relief forms has helped in agricultural planning. In this regard the aim of this study was to characterize the spatial variability of physical and chemical properties of Oxisols and Ultisols using numerical classification and the digital elevation model. Two distinct landforms: convex for the Oxisol (158 ha) and linear for the Ultisol (172 ha). 53 samples from the Oxisol and 57 samples from the Ultisol were taken. Multivariate analysis of clusters of attributes studied from their euclidean distances was performed. This analysis by dendograms along with digital elevation models for different soils characterized was more homogeneous in Ultisol groups, and less homogeneous for the Oxisol in convex landform. These quantitative methods showed that the landforms conditioned the spatial pattern of soil attributes.

The bean is a nutrient demanding plant, either due to the small and shallow root system or due to the short cycle. A study with doses of lime and boron was conducted and correlations were made between two types of leaf (+ 1 and + 3) of the bean with the contents of these elements in plants and production, aiming to establish the most appropriate leaf for diagnosis of nutritional status. The experiment was conducted in greenhouse conditions employing bean seeds cv. BRS Talisma. Lime rates considering the saturation of bases (70%), were calculated as zero, half the standard dose, dose to increase V = 70% (default), one and a half and twice the standard dose, while the boron amounts used were: zero, 0.6, 1.2, 1.8 and 2.4 mg B dm(-3). At the time of flowering, diagnostic leaves + 1 and + 3 (mature leaves from the apex) were collected along with sampling of a plant (aerial part) per pot and the measurement of production. Through correlations it was verified that nutrients, calcium, magnesium and boron in the leaf + 3 showed higher correlation coefficients with levels in the plant and production data.

The aim of this study was to obtain membrane-bound alkaline phosphatase from osteoblastic-like cells of human alveolar bone. Cells were obtained by enzymatic digestion and maintained in primary culture in osteogenic medium until subconfluence. First passage cells were cultured in the same medium and at 7, 14, and 21 days, total protein content, collagen content, and alkaline phosphatase activity were evaluated. Bone-like nodule formation was evaluated at 21 days. Cells in primary culture at day 14 were washed with Tris-HCl buffer, and used to extract the membrane-bound alkaline phosphatase. Cells expressed osteoblastic phenotype. The apparent optimum pH for PNPP hydrolysis by the enzyme was pH 10.0. This enzyme also hydrolyzes ATP, ADP, fructose-1-phosphate, fructose-6-phosphate, pyrophosphate and beta-glycerophosphate. PNPPase activity was reduced by typical inhibitors of alkaline phosphatase. SDS-PAGE of membrane fraction showed a single band with activity of similar to 120 kDa that could be solubilized by phospholipase C or Polidocanol. (c) 2007 International Federation for Cell Biology. Background: Sugarcane is an increasingly economically and environmentally important C4 grass, used for the production of sugar and bioethanol, a low-carbon emission fuel. Sugarcane originated from crosses of Saccharum species and is noted for its unique capacity to accumulate high amounts of sucrose in its stems. Environmental stresses limit enormously sugarcane productivity worldwide. To investigate transcriptome changes in response to environmental inputs that alter yield we used cDNA microarrays to profile expression of 1,545 genes in plants submitted to drought, phosphate starvation, herbivory and N-2-fixing endophytic bacteria. We also investigated the response to phytohormones (abscisic acid and methyl jasmonate). The arrayed elements correspond mostly to genes involved in signal transduction, hormone biosynthesis, transcription factors, novel genes and genes corresponding to unknown proteins. Results: Adopting an outliers searching method 179 genes with strikingly different expression levels were identified as differentially expressed in at least one of the treatments analysed. Self Organizing Maps were used to cluster the expression profiles of 695 genes that showed a highly correlated expression pattern among replicates. The expression data for 22 genes was evaluated for 36 experimental data points by quantitative RT-PCR indicating a validation rate of 80.5% using three biological experimental replicates. The SUCAST Database was created that provides public access to the data described in this work, linked to tissue expression profiling and the SUCAST gene category and sequence analysis. The SUCAST database also includes a categorization of the sugarcane kinome based on a phylogenetic grouping that included 182 undefined kinases. Conclusion: An extensive study on the sugarcane transcriptome was performed. Sugarcane genes responsive to phytohormones and to challenges sugarcane commonly deals with in the field were identified. Additionally, the protein kinases were annotated based on a phylogenetic approach. The experimental design and statistical analysis applied proved robust to unravel genes associated with a diverse array of conditions attributing novel functions to previously unknown or undefined genes. The data consolidated in the SUCAST database resource can guide further studies and be useful for the development of improved sugarcane varieties.

Cells from rat bone marrow exhibit the proliferation-differentiation sequence of osteoblasts, form mineralized extracellular matrix in vitro and release alkaline phosphatase into the medium. Membrane-bound alkaline phosphatase was obtained by method that is easy to reproduce, simpler and fast when compared with the method used to obtain the enzyme from rat osseous plate. The membrane-bound alkaline phosphatase from cultures of rat bone marrow cells has a MWr of about 120 kDa and specific PNPP activity of 1200 U/tng. The ecto-enzyme is anchored to the plasma membrane by the GPI anchor and can be released by PIPLC (selective treatment) or polidocanol (0.2 mg/mL protein and 1% (w/v) detergent). The apparent optimum pH for PNPP hydrolysis by the enzyme was pH 10. This fraction hydrolyzes ATP (240 U/mg), ADP (350 U/ mg), glucose 1-phosphate (1100 U/mg), glucose 6-phosphate (340 Wing), fructose 6-phosphate (460 U/mg), pyrophosphate (330 U/mg) and (3glycerophosphate (600 U/mg). Cooperative effects were observed for the hydrolysis of PPi and beta-glycerophosphate. PNPPase activity was inhibited by 0.1 mM vanadate (46%), 0.1 mM ZnCl2 (68%), 1 mM levamisole (66%), 1 mM arsenate (44%), 10 mM phosphate (21%) and 1 mM theophylline (72%). We report the biochemical characterization of membrane-bound alkaline phosphatase obtained from rat bone marrow cells cultures, using a method that is simple, rapid and easy to reproduce. Its properties are compared with those of rat osseous plate enzyme and revealed that the alkaline phosphatase obtained has some kinetics and structural behaviors with higher levels of enzymatic activity, facilitating the comprehension of the mineralization process and its function.
