OBJETIVO: Investigar o estresse oxidativo durante a regeneração hepática em ratos submetidos à hepatectomia (HP) e, ao mesmo tempo, avaliar a função hepática enquanto em regeneração. MÉTODO: 36 ratos Wistar machos jovens foram aleatoriamente distribuídos em dois grupos de 18 animais: submetidos somente à laparotomia (controle, Grupo G1) e parcialmente hepatectomizados (experimento, Grupo G2). Nos tempos 36h (T1), 168h (T2) e 336h (T3) pós-HP, GSH foi medida no plasma e no tecido hepático, enquanto Gli e BT foram aquilatados no sangue. A massa do fígado residual foi utilizada para estimar a evolução da regeneração hepática. RESULTADOS: Houve diferença estatisticamente significativa no crescimento dos lobos residuais nos grupos controle e experimento. GSH hepático e plasmático se mostraram significantemente maior nos animais parcialmente hepatectomizados.,em todos os tempos. Hiperglicemia estatisticamente significativa ocorreu nos ratos pós-HP nos tempos T2 e T3. A BT não apresentou qualquer alteração entre os grupos. CONCLUSÃO: Durante a regeneração hepática pós-HP em ratos há um aumento do estresse oxidativo e o fígado residual permanece apto na manutenção da homeostase orgânica.

The environmental impacts of a single mine often remain local, but acidic and metal-rich acid mine drainage (AMD) from the waste materials may pose a serious threat to adjacent surface waters and their ecosystems. Testate amoebae (thecamoebian) analysis was used together with lake sediment geochemistry to study and evaluate the ecological effects of sulphidic metal mines on aquatic environments. Three different mines were included in the study: Luikonlahti Cu-mine in Kaavi, eastern Finland, Haveri Cu-Au mine in Ylöjärvi, southern Finland and Pyhäsalmi Zn-Cu-S mine in Pyhäjärvi, central Finland. Luikonlahti and Haveri are closed mines, but Pyhäsalmi is still operating. The sampling strategy was case specific, and planned to provide a representative sediment sample series to define natural background conditions, to detect spatial and temporal variations in mine impacts, to evaluate the possible recovery after the peak contamination, and to distinguish the effects of other environmental factors from the mining impacts. In the Haveri case, diatom analyses were performed alongside thecamoebian analysis to evaluate the similarities and differences between the two proxies. The results of the analyses were investigated with multivariate methods (direct and indirect ordinations, diversity and distance measure indices). Finally, the results of each case study were harmonized, pooled, and jointly analyzed to summarize the results for this dissertation. Geochemical results showed broadly similar temporal patterns in each case. Concentrations of ions in the pre-disturbance samples defined the natural baseline against which other results were compared. The beginning of the mining activities had only minor impacts on sediment geochemistry, mainly appearing as an increased clastic input into the lakes at Haveri and Pyhäsalmi. The active mining phase was followed by the metallic contamination and, subsequently, by the most recent change towards decreased but still elevated metal concentrations in the sediments. Because of the delay in the oxidation of waste material and formation of AMD, the most intense, but transient metal contamination phase occurred in the post-mining period at Luikonlahti and Haveri. At Pyhäsalmi, the highest metal contamination preceded effluent mitigation actions. Spatial gradients were observed besides the temporal evolution in both the pre-disturbance and mine-impacted samples from Luikonlahti and Pyhäsalmi. The geochemical gradients varied with distance from the main source of contaminants (dispersion and dilution) and with water depth (redox and pH). The spatial extent of the highest metal contamination associated with these mines remained rather limited. At Haveri, the metallic impact was widespread, with the upstream site in another lake basin found to be contaminated. Changes in thecamoebian assemblages corresponded well with the geochemical results. Despite some differences, the general features and ecological responses of the faunal assemblages were rather similar in each lake. Constantly abundant strains of Difflugia oblonga, Difflugia protaeiformis and centropyxids formed the core of these assemblages. Increasing proportions of Cucurbitella tricuspis towards the surface samples were found in all of the cases. The results affirmed the indicator value of some already known indicator forms, but such as C. tricuspis and higher nutrient levels, but also elicited possible new ones such as D. oblonga ‘spinosa’ and clayey substrate, high conductivity and/or alkalinity, D. protaeiformis ‘multicornis’ and pH, water hardness and the amount of clastic material and Centropyxis constricta ‘aerophila’ and high metal and S concentrations. In each case, eutrophication appeared to be the most important environmental factor, masking the effects of other variables. Faunal responses to high metal inputs in sediments remained minor, but were nevertheless detectable. Besides the trophic state of the lake, numerical methods suggested overall geochemical conditions (pH, redox) to be the most important factor at Luikonlahti, whereas the Haveri results showed the clearest connection between metals and amoebae. At Pyhäsalmi, the strongest relationships were found between Ca- and S-rich present loading, redox conditions and substrate composition. Sediment geochemistry and testate amoeba analysis proved to be a suitable combination of methods to detect and describe the aquatic mine impacts in each specific case, to evaluate recovery and to differentiate between the effects of different anthropogenic and natural environmental factors. It was also suggested that aquatic mine impacts can be significantly mitigated by careful design and after-care of the waste facilities, especially by reducing and preventing AMD. The case-specific approach is nevertheless necessary because of the unique characteristics of each mine and variations in the environmental background conditions.

OBJETIVO: avaliar as características clínicas e laboratoriais de parentes de primeiro grau do sexo masculino de pacientes com diagnóstico confirmado de síndrome de ovários policísticos (SOP) e comparar os achados com um grupo controle sem história familiar de SOP. MÉTODOS: foram selecionados aleatoriamente 28 homens com idade entre 18 e 65 anos que possuíam parentesco de primeiro grau com mulheres diagnosticadas com SOP e 28 controles pareados por idade, cintura e índice de massa corporal (IMC). RESULTADOS: homens com parentesco de 1º grau com mulheres com SOP comparados ao Grupo Controle apresentaram níveis mais elevados de triglicerídeos (189,6±103,1 versus 99,4±37,1; p<0,0001), HOMA-IR (Homeostase Model Assesment) (3,5±9,1 versus 1,0±1,0; p=0,0077) e glicemia (130,1±81,7 versus 89,5±7,8; p=0,005), além de menores níveis da globulina ligadora de hormônios sexuais (SHBG) (23,8±13,8 versus 31,1±9,1; p=0,003). Os níveis de SHBG se correlacionaram independentemente com os níveis de triglicérides. Os parentes de 1º grau também apresentavam mais sinais clínicos de hiperandrogenismo. CONCLUSÕES: parentes de primeiro grau do sexo masculino das pacientes com SOP apresentam maior grau de dislipidemia e de resistência à insulina, além de níveis mais baixos de SHBG com mais sinais clínicos de hiperandrogenismo. Esses achados sugerem que a resistência à insulina pode ter origem hereditária em indivíduos com história familiar de SOP, independentemente de parâmetros antropométricos.

OBJETIVO: investigar o efeito da exposição à fumaça de cigarro durante os períodos de prenhez e lactação de ratas sobre o ganho de peso corpóreo e tecidual, parâmetros séricos e produção láctea, bem como a repercussão na prole, desde o nascimento até o período jovem adulto. MÉTODOS: 40 ratas Wistar prenhes foram divididas em quatro grupos: CG - não expostas à fumaça de cigarro e sacrificadas ao término da gestação; CL - não expostas à fumaça de cigarro e sacrificadas ao término da lactação; FG - expostas à fumaça de cigarro e sacrificadas ao término da gestação; FL - expostas à fumaça de cigarro e sacrificadas ao término da lactação. As proles foram separadas por sexo e dividas conforme o grupo de suas mães, sendo sacrificadas na fase jovem adulto. Nas ratas e nas proles foram avaliados peso tecidual, peso corpóreo e parâmetros séricos. Foi também analisada a produção láctea por filhote. RESULTADOS: o peso corpóreo de ratas estava diminuído no grupo FL durante a lactação (CL=267,0±7,2; FL=235,5±7,2 g*,*p<0,05). Não foi detectado tecido adiposo nos grupos CL e FL, porém, em FG, esse tecido estava reduzido comparado ao CG (CG=3,3±0,3; FG=2,4±0,3 g*, *p<0,05). As ratas expostas à fumaça de cigarro apresentaram maiores valores de glicemia (CG=113±17, CL=86±16, FG=177±21*, FL=178±23 mg/dL*, *p<0,05 CG versus FG e CL versus FL). Os grupos CL e FL apresentaram menor valor de colesterol-HDL, sem alteração no colesterol total. Por fim, as ratas expostas à fumaça de cigarro tiveram a produção láctea inferior comparadas às não expostas (CL=6,7±0,4, FL=5,4±0,3 g*, *p<0,05). Nas proles das ratas FG e FL observou-se diminuição do PC desde o nascimento até a fase jovem adulto, porém não houve alteração nos pesos de gastrocnêmio, fígado e coração de todos os grupos, e o tecido adiposo não foi detectado em proles fêmeas. Houve aumento na glicemia da prole de ratas expostas à fumaça de cigarro em ambos os sexos (machos: Pcg=107±10,5, Pcl=115±8,6, Pfg=148±16,8*, Pfl=172±11,2**; fêmeas: Pcg=109±27,2, Pcl=104±9,7, Pfg=134±20,0*, Pfl=126±13,3**; p<0,05 Pcg versus Pfg e Pcl versus Pfl). CONCLUSÕES: a exposição à fumaça de cigarro durante a prenhez e a lactação acarretou prejuízos morfométricos e séricos tanto nas mães como nas proles, o que persistiu até a fase jovem adulta.

Pré-eclâmpsia é uma síndrome sistêmica caracterizada por intenso estado inflamatório e antiangiogênico. A fisiopatologia da pré-clâmpsia envolve alterações no processo de invasão trofoblástica, com consequente inadequado suprimento sanguíneo uterino e estresse oxidativo do tecido placentário. As alterações placentárias decorrentes desse processo levam à maior produção de sFlt-1, um receptor solúvel para as moléculas de VEGF e PlGF. O sFlt-1 impede com que VEGF e PlGF realizem suas funções na homeostase endotelial, culminando com disfunção dessas células. De uma maneira geral, os processos inflamatórios, de disfunção endotelial e estresse oxidativo estão interligados e agem de maneira sinérgica. Trabalhos recentes têm demonstrado que elevações nas concentrações séricas de sFlt-1 ocorrem 5 a 6 semanas antes das manifestações clínicas da pré-eclâmpsia. Concomitantemente, observa-se queda nas concentrações séricas de PlGF. Sendo assim, as dosagens séricas de sFlt-1 e PlGF têm sido sugeridas para o diagnóstico precoce de pré-eclâmpsia. Ademais, os conhecimentos adquiridos a respeito dos fatores antiangiogênicos proporcionam ainda a possibilidade de novas linhas de pesquisa sobre possíveis terapias para a pré-eclâmpsia. Neste artigo, foram revisados os aspectos inflamatórios e antiangiogênicos envolvidos na fisiopatologia da pré-eclâmpsia. Por fim, foram correlacionados esses aspectos com o risco elevado para doenças cardiovasculares apresentado por essas pacientes ao longo de suas vidas.

As miopatias em equinos são classificadas de acordo com aspectos clínicos, morfológicos e moleculares, em três grandes grupos: não associadas ao exercício, associadas ao exercício e devido alteração da condução elétrica do sarcolema. Apesar dos avanços no diagnóstico, a literatura ainda relata surtos de miopatia em equinos sem etiologia esclarecida em diversos países. O objetivo desse estudo foi descrever as alterações histológicas e histoquímicas de biópsias musculares de equinos acometidos por miopatia. Sete equinos da raça Quarto de Milha, com 18-24 meses de idade, apresentaram sinais clínicos de miopatia. Dentre esses animais, cinco apresentaram sinais subagudos leves a moderados e dois apresentaram sinais hiperagudos severos e decúbito lateral. Foram realizadas biópsias musculares utilizando a técnica percutânea, por agulha tipo Bergström, no músculo glúteo médio em todos os animais acometidos. As amostras foram processadas por meio de técnicas histológicas (HE, Tricrômio de Gomori modificado) e histoquímicas (PAS, NADH, ATPase). Nos quadros clínicos mais leves, a principal alteração encontrada foi a presença de fibras vermelhas rajadas do tipo I e IIA, que estão associadas às alterações do metabolismo oxidativo e das funções mitocondriais, como ocorrem nas miopatias mitocondriais. Também foram observadas fibras atróficas do tipo I e IIA, além da presença de agregados subsarcolemais. Nos quadros mais severos o tecido muscular apresentou infiltrado inflamatório, aumento de colágeno, fagocitose, necrose, calcificação e regeneração muscular. Diante dos achados morfológicos, da resposta à terapia com vitamina E e Se e da baixa mortalidade quando comparado aos relatos de miopatia atípica, conclui-se que esse surto foi desencadeado por lesões mitocondriais, caracterizadas pelas fibras musculares vermelhas rajadas, possivelmente devido uma quebra da homeostase de vitamina E e Se, sendo compatível com o diagnóstico de miopatia nutricional.

Apoptose tem um papel importante na manutenção da homeostase placentária, e o desequilíbrio desse processo pode comprometer a gestação. O objetivo do presente estudo foi avaliar a ocorrencia de apoptose em amostras de placenta de vacas em diferentes fases de gestação. Amostras de placentomos de 15 vacas saudáveis com 4 (n=5), 6 (n=5) e 9 (n=5) meses de gestação foram colhidas e processadas rotineiramente para a histologia, imuno-histoquímica e isolamento de DNA. As lâminas obtidas foram coradas em HE, ou submetidas à análise imuno-histoquímica das proteínas pró-apoptóticas caspase-3 e Bax, e da proteína anti-apoptótica Bcl-2. O DNA isolado foi submetido à eletroforese em gel de agarose para detecção da fragmentação internucleossômica do genoma. Os resultados de histomorfometria revelaram que as células apoptóticas aumentaram progressivamente com o avanço da gestação. Confirmou-se a apoptose pela fragmentação característica do DNA genômico, visualizada pelo clássico "padrão em escada" na eletroforese em gel de agarose. Adcionalmente, a imunoexpressão de caspase-3, Bax e Bcl-2 foram observadas em todas as amostras. Entretanto, a proteína caspase-3 apresentou marcação mais intensa em todos os tempos gestacionais, quando comparada com a marcação das proteínas Bcl-2 e Bax. Esses resultados confirmam e reforçam a importância da apoptose na maturação placentária. Além disto, indica que caspase-3, Bax e Bcl-2 estão envolvidas nos mecanismos de ativação da apoptose pela via intrínseca mitocondrial ao longo da gestação, contribuindo para o equilíbrio fisiológico da celularidade e renovação celular na placenta bovina.

Tässä työssä tutkittiin sähkön käyttömahdollisuuksia metsäteollisuuden jäteveden-puhdistamolla syntyvän biolietteen kuivauksessa. Lisäksi selvityksen kohteena olivat lietteen sähkökuivaukseen vaikuttavat tekijät. Työn kirjallisessa osassa perehdyttiin lietteiden syntyyn jätevedenpuhdistamolla sekä lietteiden ominaisuuksiin. Työssä tutustuttiin myös lietteiden käsittelyyn ja siinä käytettäviin tekniikoihin. Ennen työn kokeellista osaa esiteltiin sähkökuivauksen periaate sekä sähkökuivaukselle aiemmin tehtyjä tutkimuksia ja niistä saatuja kokemuksia. Myös tuotteistetut elektrokuivauksen sovellukset olivat osa työtä. Kokeellisessa osassa keskityttiin varsinaisiin sähkökuivauskokeisiin. Tutkimukset suoritettiin suodatuskokein, jotka toteutettiin imuun perustuvalla Büchner-suodattimella sekä painesuodattimella. Kokeissa tutkittiin tasavirran, virran pulssituksen ja vaihtovirran vaikutusta metsäteollisuuden primääri- ja biolietteeseen. Tasavirtatutkimuksissa sähkövirtaa johdettiin lietteeseen suodatuksen aikana, kun taas vaihtovirtaa testattaessa liete käsiteltiin ensin sähköllä ja suodatettiin erikseen. Suodatuskokeiden lisäksi tutkimuksissa tarkkailtiin lietteiden kuiva-ainepitoisuuksia, redox-potentiaalia, lämpötilaa, pH:ta ja sähkönjohtokykyä. Lietteitä tutkittiin myös tuhkapitoisuuden suhteen sekä suoritettiin solututkimuksia fluoresenssilukijan avulla. Työn laboratoriotutkimukset osoittivat sähkön vaikuttavan selkeästi biolietteen ve-denerotuskykyyn: sähkö nopeutti lietteen suodatusta parhaimmillaan yli 70 %. Säh-kökuivauksella lietteen kuiva-ainepitoisuudet pysyivät samoina, mutta suodatus nopeutui ja suodatusajat jopa puolittuivat. Myös lietteen sähkönjohtokyky kasvoi sähkökäsittelyn aikana, mikä viittasi veden vapautumiseen solujen sisältä. Lietteen kunto ja ominaisuudet, polymeeri, sekoitus sekä sähkökäsittelyn parametrit olivat tärkeässä asemassa sähkökuivauksen onnistumisen kannalta.

Blood-derived products are commonly administered to horses and humans to treat many musculoskeletal diseases, due to their potential antioxidant and anti-inflammatory effects. Nevertheless, antioxidant effects have never been shown upon horse synovial fluid cells in vitro. If proved, this could give a new perspective to justify the clinical application of blood-derived products. The aim of the present study was to investigate the antioxidant effects of two blood-derived products - plasma (unconditioned blood product - UBP) and a commercial blood preparation (conditioned blood product - CBP)¹ - upon stimulated equine synovial fluid cells. Healthy tarsocrural joints (60) were tapped to obtain synovial fluid cells; these cells were pooled, processed, stimulated with lipopolysaccharide (LPS) or phorbol 12-myristate 13-acetate (PMA), and evaluated by flow cytometry for the production of reactive oxygen species (ROS). Upon addition of any blood-derived product here used - UBP and CBP - there was a significant decrease in the oxidative burst of synovial fluid cells (P<0.05). There was no difference between UBP and CBP effects. In conclusion, treatment of stimulated equine synovial cells with either UBP or CBP efficiently restored their redox equilibrium.

No presente trabalho avaliaram-se os parâmetros hematológicos e bioquímicos de exemplares adultos de tilápias (Oreochromis niloticus) sob a influência do fator estresse fisiológico em animais submetidos à exposição ao ar durante a engorda em sistema raceway. Foram analisados o eritrograma, teor de hemoglobina, volume globular, o volume corpuscular médio (VCM), a hemoglobina corpuscular média (HCM), a concentração de hemoglobina corpuscular média (CHCM), o leucograma, contagem diferencial de leucócitos, o plaquetograma, a glicose, a proteína total, o colesterol, o triglicerídeo e os eletrólitos (cálcio, cloretos, sódio e potássio). Os resultados revelaram que houve uma homogeneidade de distribuição para hemácias, volume globular, hemoglobina, índices hemantimétricos, proteína total, glicose, colesterol, e íons séricos, indicados pelos valores relativamente baixos do coeficiente de variação. Houve correlação positiva somente para leucócitos totais, células de defesa orgânica (neutrófilos e linfócitos), glicose, colesterol, sódio e cálcio. Quanto ao leucograma, à medida que os animais foram expostos ao ar, o número de leucócitos diminuiu gradativamente (leucopenia), ocorrendo simultaneamente neutrofilia e linfopenia. O índice glicêmico constituiu um bom indicador de estresse fisiológico, devido à hiperglicemia (82,0±20,88mg/dL) demonstrada nos tratamentos. A exposição ao ar constituiu um fator de desequilíbrio na homeostase iônica, e na síntese de colesterol endógeno. Entretanto, o tempo de recuperação não ocasionou a completa reabilitação fisiológica frente ao desafio imposto.

This thesis focuses on the molecular mechanisms regulating the photosynthetic electron transfer reactions upon changes in light intensity. To investigate these mechanisms, I used mutants of the model plant Arabidopsis thaliana impaired in various aspects of regulation of the photosynthetic light reactions. These included mutants of photosystem II (PSII) and light harvesting complex II (LHCII) phosphorylation (stn7 and stn8), mutants of energy-dependent non-photochemical quenching (NPQ) (npq1 and npq4) and of regulation of photosynthetic electron transfer (pgr5). All of these processes have been extensively investigated during the past decades, mainly on plants growing under steady-state conditions, and therefore many aspects of acclimation processes may have been neglected. In this study, plants were grown under fluctuating light, i.e. the alternation of low and high intensities of light, in order to maximally challenge the photosynthetic regulatory mechanisms. In pgr5 and stn7 mutants, the growth in fluctuating light condition mainly damaged PSI while PSII was rather unaffected. It is shown that the PGR5 protein regulates the linear electron transfer: it is essential for the induction of transthylakoid ΔpH that, in turn, activates energy-dependent NPQ and downregulates the activity of cytochrome b6f. This regulation was shown to be essential for the photoprotection of PSI under fluctuations in light intensity. The stn7 mutants were able to acclimate under constant growth light conditions by modulating the PSII/PSI ratio, while under fluctuating growth light they failed in implementing this acclimation strategy. LHCII phosphorylation ensures the balance of the excitation energy distribution between PSII and PSI by increasing the probability for excitons to be trapped by PSI. LHCII can be phosphorylated over all of the thylakoid membrane (grana cores as well as stroma lamellae) and when phosphorylated it constitutes a common antenna for PSII and PSI. Moreover, LHCII was shown to work as a functional bridge that allows the energy transfer between PSII units in grana cores and between PSII and PSI centers in grana margins. Consequently, PSI can function as a quencher of excitation energy. Eventually, the LHCII phosphorylation, NPQ and the photosynthetic control of linear electron transfer via cytochrome b6f work in concert to maintain the redox poise of the electron transfer chain. This is a prerequisite for successful plant growth upon changing natural light conditions, both in short- and long-term.

RESUMO O Brasil se destaca no cenário americano como um dos países com maior potencial para a piscicultura, principalmente a dulcícola. A tilápia do Nilo (Oreochromis niloticus) se destaca sendo a espécie mais cultivada em nosso país. Metodologias para diminuir a interferência de agentes estressores nas funções vitais e fisiológicas dos peixes são importantes durante o manejo. O presente trabalho teve como objetivo avaliar o efeito do eugenol nas respostas metabólicas e iônicas de juvenis de tilápia do Nilo, submetidos ao transporte em sacos plásticos, em diferentes densidades, a fim de verificar a eficiência do produto como agente mitigador do estresse. O eugenol foi utilizado na concentração de 15mg/L em água. As densidades avaliadas foram 4, 7 e 10 peixes L-1, equivalente a 140, 245 e 350g L-1. Após quatro horas de transporte foram avaliados os parâmetros metabólicos (glicose e lactato) e iônicos (cloreto, magnésio e cálcio), bem como a qualidade da água nos sacos plásticos. Em relação aos dois parâmetros metabólicos, o uso do eugenol com o intuito de diminuir as respostas do estresse não foi satisfatório. Houve elevação no nível de glicose nas densidades 140 e 350g L-1 imediatamente ao término do transporte, e o teor de lactato dos peixes na densidade 245g L-1 aumentou 24 horas depois, indicando que os animais não conseguiram manter a homeostase inicial. Dentre as concentrações de íons avaliados, o magnésio foi o que sofreu maior variação. Podemos concluir que a adição de 15mg L-1 de eugenol na água durante o transporte de juvenis de tilápia do Nilo nas densidades de 140, 245 e 350g L-1 não foi capaz de minimizar as respostas ao estresse.

Polymeric materials that conduct electricity are highly interesting for fundamental studies and beneficial for modern applications in e.g. solar cells, organic field effect transistors (OFETs) as well as in chemical and bio‐sensing. Therefore, it is important to characterize this class of materials with a wide variety of methods. This work summarizes the use of electrochemistry also in combination with spectroscopic methods in synthesis and characterization of electrically conducting polymers and other π‐conjugated systems. The materials studied in this work are intended for organic electronic devices and chemical sensors. Additionally, an important part of the presented work, concerns rational approaches to the development of water‐based inks containing conducting particles. Electrochemical synthesis and electroactivity of conducting polymers can be greatly enhanced in room temperature ionic liquids (RTILs) in comparison to conventional electrolytes. Therefore, poly(para‐phyenylene) (PPP) was electrochemically synthesized in the two representative RTILs: bmimPF6 and bmiTf2N (imidazolium and pyrrolidinium‐based salts, respectively). It was found that the electrochemical synthesis of PPP was significantly enhanced in bmimPF6. Additionally, the results from doping studies of PPP films indicate improved electroactivity in bmimPF6 during oxidation (p‐doping) and in bmiTf2N in the case of reduction (n‐doping). These findings were supported by in situ infrared spectroscopy studies. Conducting poly(benzimidazobenzophenanthroline) (BBL) is a material which can provide relatively high field‐effect mobility of charge carriers in OFET devices. The main disadvantage of this n‐type semiconductor is its limited processability. Therefore in this work BBL was functionalized with poly(ethylene oxide) PEO, varying the length of side chains enabling water dispersions of the studied polymer. It was found that functionalization did not distract the electrochemical activity of the BBL backbone while the processability was improved significantly in comparison to conventional BBL. Another objective was to study highly processable poly(3,4‐ethylenedioxythiophene) poly(styrenesulfonate) (PEDOT:PSS) water‐based inks for controlled patterning scaled‐down to nearly a nanodomain with the intention to fabricate various chemical sensors. Developed PEDOT:PSS inks greatly improved printing of nanoarrays and with further modification with quaternary ammonium cations enabled fabrication of PEDOT:PSS‐based chemical sensors for lead (II) ions with enhanced adhesion and stability in aqueous environments. This opens new possibilities for development of PEDOT:PSS films that can be used in bio‐related applications. Polycyclic aromatic hydrocarbons (PAHs) are a broad group of π‐conjugated materials consisting of aromatic rings in the range from naphthalene to even hundred rings in one molecule. The research on this type of materials is intriguing, due to their interesting optical properties and resemblance of graphene. The objective was to use electrochemical synthesis to yield relatively large PAHs and fabricate electroactive films that could be used as template material in chemical sensors. Spectroscopic, electrochemical and electrical investigations evidence formation of highly stable films with fast redox response, consisting of molecules with 40 to 60 carbon atoms. Additionally, this approach in synthesis, starting from relatively small PAH molecules was successfully used in chemical sensor for lead (II).

Nowadays biomass transformation has a great potential for the synthesis of value-added compounds with a wide range of applications. Terpenoids, extracted from biomass, are inexpensive and renewable raw materials which often have a biological activity and are widely used as important organic platform molecules in the development of new medicines as well as in the synthesis of fine chemicals and intermediates. At the same time, special attention is devoted to the application of gold catalysts to fine chemical synthesis due to their outstanding activity and/or selectivity for transformations of complex organic compounds. Conversion of renewable terpenoids in the presence of gold nanoparticles is one of the new and promising directions in the transformation of biomass to valuable chemicals. In the doctoral thesis, different kinds of natural terpenoids, such as α-pinene, myrtenol and carvone were selected as starting materials. Gold catalysts were utilized for the promising routes of these compounds transformation. Investigation of selective α-pinene isomerization to camphene, which is an important step in an industrial process towards the synthesis of camphor as well as other valuable substrates for the pharmaceutical industry, was performed. A high activity of heterogeneous gold catalysts in the Wagner-Meerwein rearrangement was demonstrated for the first time. Gold on alumina carrier was found to reach the α-pinene isomerization conversion up to 99.9% and the selectivity of 60-80%, thus making this catalyst very promising from an industrial viewpoint. A detailed investigation of kinetic regularities including catalyst deactivation during the reaction was performed. The one-pot terpene alcohol amination, which is a promising approach to the synthesis of valuable complex amines having specific physiological properties, was investigated. The general regularities of the one-pot natural myrtenol amination in the presence of gold catalysts as well as a correlation between catalytic activity, catalyst redox treatment and the support nature were obtained. Catalytic activity and product distribution were shown to be strongly dependent on the support properties, namely acidity and basicity. The gold-zirconia (Au/ZrO2) catalyst pretreated under oxidizing atmosphere was observed to be rather active, resulting in the total conversion of myrtenol and the selectivity to the corresponding amine of about 53%. The reaction kinetics was modelled based on the mechanistic considerations with the catalyst deactivation step incorporated in the mechanism. Carvone hydrogenation over a gold catalyst was studied with the general idea of investigating both the activity of gold catalysts in competitive hydrogenation of different functional groups and developing an approach to the synthesis of valuable carvone derivatives. Gold was found to promote stereo- and chemoselective carvone hydrogenation to dihydrocarvone with a predominant formation of the trans-isomer, which generally is a novel synthetic method for an industrially valuable dihydrocarvone. The solvent effect on the catalytic activity as well as on the ratio between trans- and cis-dihydrocarvone was evaluated.

I studied the associations between migration-related physiological regulation (corticosterone) and body condition of barn swallows (Hirundo rustica L.). An additional purpose was to determine whether oxidative stress and biotransformation activity vary seasonally. Since physiological regulation, biotransformation activity and the stress involved may be important factors for body condition during migration; they may have direct effects on migration success. This in turn may influence other important life history stages, such as breeding and moult. In the thesis I used barn swallow data of the Finnish Ringing Centre (1997–2009), consisting of all juveniles ringed in the nests and recaptured from night roosts later the same autumn. Before the autumn migration in Finland I also captured, ringed and sampled barn swallows from night roosts in 2003, 2006, 2007 and 2011. Samples preceding spring migration in South Africa were collected in 2007. Juvenile barn swallows started to migrate southward in mid-August (first broods). Second broods started their migration at a younger age and almost a month later than first broods (mid-September). Barn swallows increased body mass and accumulated fat for the autumn migration. In the course of the autumn they seemed to be able to prevent the loss of energy already accumulated, since the proportional overnight mass loss, fat loss and faecal production decreased. Surprisingly, corticosterone, the major energy-regulating hormone in birds, seemed not to be involved in the fuelling process. Previous studies with warblers, sparrows and shorebirds had shown that during migration, the baseline levels of corticosterone were elevated in order to facilitate fuelling. It is possible that for Finnish barn swallows the most important fuelling place is in southern Europe, since northern and eastern populations migrate via the Balkan Peninsula. However, the adrenocortical stress response of Finnish barn swallows in good body condition was lower than that of those in poor body condition. Birds clearly suppressed the response, probably to prevent the catabolic effects of excessive corticosterone levels; birds cannot afford to lose muscle mass before migration. South African barn swallows had high levels of baseline corticosterone, but this may have been associated with the high oxidative damage and biotransformation activity of those birds. Barn swallows in spring and summer had low biotransformation activity and intermediate oxidative stress, which was probably related to breeding. Autumn birds had low biotransformation activity and oxidative stress but high redox enzyme activities in some migration-related enzymes.
