De forma a melhorar a precisão dos resultados obtidos com instrumentos doseadores de líquidos utilizados em ambiente clínico, foi submetido e aprovado um projeto internacional denominado Metrology for Drug Delivery (MeDD), financiado pela União Europeia, em que o Instituto Português da Qualidade (IPQ) é um dos sete participantes. Ao nível nacional, todo o projeto foi desenvolvido no Laboratório de Volume (LVO) do IPQ, em parceria com o Departamento de Engenharia Mecânica e Industrial (DEMI) da Faculdade de Ciências e Tecnologia da Universidade Nova de Lisboa (FCT/UNL), a B.Braun Portugal e o Hospital Garcia de Orta (HGO). A presente dissertação centra-se na elaboração de um procedimento de calibração de sistemas doseadores de fluidos, usando como ponto de partida um sistema padrão de micro caudal desenvolvido anteriormente pelo LVO em colaboração com a FCT-UNL/DEMI. Nesta dissertação são apresentados estudos de desempenho realizados em sistemas utilizados no hospital. Os instrumentos doseadores de fluidos mais usuais nos hospitais são as seringas perfusoras e as bombas peristálticas, que têm como função fornecer os fluidos necessários para hidratar, nutrir ou administrar fármacos. Com o objetivo de estudar o desempenho destes equipamentos em diferentes situações, tais como a influência da administração de fluidos mais viscosos em comparação com água, o tempo de resposta dos instrumentos e o impacto dos acessórios na linha de perfusão, realizaram-se diversos ensaios. Através dos ensaios realizados foi possível determinar o erro e a incerteza associada a cada equipamento para cada experiência realizada. A maioria dos resultados obtidos com os sistemas doseadores de fluidos permitiram verificar que os valores da incerteza dos caudais eram inferiores ao erro máximo admissível indicado pelo fabricante e serviram de base para a elaboração de um procedimento técnico.

Introduction: Paracoccidioidomycosis (PCM) is a systemic infection caused by the fungus Paracoccidioides brasiliensis. PCM is considered one of the most important systemic mycoses in Latin America. Methods: This is a clinical, epidemiological, retrospective, quantitative study of PCM cases in patients attending the National Health Service in the State of Rondônia in 1997-2012. The examined variables included sex, age group, year of diagnosis, education level, profession, place of residence, diagnostic test, prior treatment, medication used, comorbidities and case progress. Results: During the study period, 2,163 PCM cases were registered in Rondônia, and the mean annual incidence was 9.4/100,000 people. The municipalities with the highest rates were located in the southeastern region of Rondônia, and the towns of Pimenteiras do Oeste and Espigão do Oeste had the highest rates in the state, which were 39.1/100,000 and 37.4/100,000 people, respectively. Among all cases, 90.2% and 9.8% were observed in men and women, respectively, and most cases (58.2%) were observed in patients aged between 40 and 59 years. Itraconazole was used to treat 91.6% (1,771) of cases, followed by sulfamethoxazole in combination with trimethoprim (4.4% [85] of cases). One hundred thirty-one (6%) patients died. Conclusions: The State of Rondônia has a high incidence of PCM, and the municipalities in the southeastern region of the state were found to have the highest incidence rates of this disease. Our findings suggest that Rondônia is the state in the northern region with the highest mortality rate for PCM.

Este livro reúne 15 artigos escritos em tempos diferentes, agora reunidos num único volume publicado sob os auspícios do Centro de História de Além-Mar. Econtram-se, contudo, unidos por um denominador comum, o Brasil colonial, e por uma época predominante, o século XVIII. Partindo da noção de que o conhecimento é uma forma de poder, a autora pretende entender como a utilização do saber produzido ao longo de setecentos foi particularmente útil na governação da América Portuguesa, utilizado por governantes como meio de controlo do espaço, da natureza e da humanidade. Este conhecimento permitiu também o descobrimento da colónia brasileira pelos europeus de setecentos, com repercussões evidentes na formação de uma consciência europeia e na formulação de imagens sobre o Império Português e os portugueses, que acentuaram as diferenças culturais, políticas, económicas e científicas existentes, pondo em causa uma visão uniforme da Europa das Luzes.

IntroductionMicrosporidia constitute the most common black fly pathogens, although the species' diversity, seasonal occurrence and transmission mechanisms remain poorly understood. Infections by this agent are often chronic and non-lethal, but they can cause reduced fecundity and decreased longevity. The objective of this study was to identify microsporidia infecting Simulium (Chirostilbia) pertinax (Kollar, 1832) larvae from Caraguatatuba, State of São Paulo, Brazil, by molecular and morphological characterization.MethodsLarvae were collected at a single point in a stream in a rural area of the city and were kept under artificial aeration until analysis. Polydispyrenia spp. infection was characterized by the presence of at least 32 mononuclear spores measuring 6.9 ± 1.0 × 5.0 ± 0.7µm in persistent sporophorous vesicles. Similarly, Amblyospora spp. were characterized by the presence of eight uninucleate spores measuring 4.5 × 3.5µm in sporophorous vesicles.ResultsThe molecular analysis confirmed the presence of microsporidian DNA in the 8 samples (prevalence of 0.51%). Six samples (Brazilian larvae) were related to Polydispyrenia simulii and Caudospora palustris reference sequences but in separate clusters. One sample was clustered with Amblyospora spp. Edhazardia aedis was the positive control taxon.ConclusionsSamples identified as Polydispyrenia spp. and Amblyospora spp. were grouped with P. simulii and Amblyospora spp., respectively, corroborating previous results. However, the 16S gene tree showed a considerable distance between the black fly-infecting Amblyospora spp. and the mosquito-infecting spp. This distance suggests that these two groups are not congeneric. Additional genomic region evaluation is necessary to obtain a coherent phylogeny for this group.

O estudo sobre os textos escritos produzidos por alunos da 7ª classe em Angola foi estabelecido por nós com base na hipótese da existência de dificuldades de escrita. Há bastantes dúvidas entre os docentes angolanos sobre a capacidade dos alunos na elaboração de textos espontâneos. Desta forma, com o objectivo de conhecermos as capacidades de produção de textos escritos pelos alunos do ensino geral no Lubango, de elaborarmos uma tipologia de análise dos erros encontrados e de propormos estratégias de superação das dificuldades apresentadas nos textos dos alunos, fomos à Escola do I Ciclo do Ensino Secundário 27 de Março do Lubango – Huíla recolher as composições da Prova Trimestral para verificar as dificuldades de escrita apresentadas nos textos dos alunos. Partimos de 314 textos, dos quais foi extraída a amostra de 10%, correspondendo a 32 textos seleccionados através da tabela de números aleatórios. Os textos foram analisados com base na tipologia de erros de Sousa (1996). Desta forma, foram detectados erros de substituição, de omissão, de divisão/aglutinação, os quais foram muito significativos e ocorreram em 66,856% dos casos no total. Os erros de substituição maiúsculas/minúsculas, os de adição, de grafias homófonas constituem 30.856% dos casos. Confirmam-se as dificuldades apresentadas pelos alunos que devem merecer tratamento através das propostas de remediação apresentadas. As dificuldades encontradas nos textos destes alunos devem servir de base para a planificação da disciplina de Língua Portuguesa no Ensino Primário de forma a dotar o aluno de competências para a melhoria da comunicação escrita na sala de aula.

Chagas disease (ChD), a neglected tropical disease caused by infection with the parasite Trypanosoma cruzi (T. cruzi), remains a serious public health issue in Latin America and is an emerging disease in several non-endemic countries, where knowledge of the condition and experience with its clinical management are limited. Regionally, the disease is the major cause of disability secondary to tropical diseases in young adults. Health-related quality of life (HRQoL) impairment is common in patients with ChD, especially in those with Chagas dilated cardiomyopathy, the most severe manifestation of the disease, which frequently leads to heart failure. The aim of this review was to conduct a literature search for studies that have evaluated the determining factors of HRQoL in ChD patients. We included cross-sectional, case-control, cohort, and experimental studies, as well as clinical trials that evaluated the HRQoL in ChD patients aged 18 to 60 years and are presenting an explicit description of statistical analysis. Using a combination of keywords based on Descriptors in Health Sciences (DeCS) and Medical Subject Headings (MeSH) for searches in PubMed and the Scientific Electronic Library Online (SciELO), 148 studies were found. After exclusions, 12 studies were selected for analysis. Three main findings were extracted from these studies: 1) cardiac involvement is associated with a worse HRQoL in ChD patients; 2) HRQoL is associated with the patients' functional capacity; and 3) simple and inexpensive therapeutic measures are effective for improving HRQoL in ChD patients. Hence, ChD patients' functional capacity, the effectiveness of non-surgical conservative treatment, and cardiac involvement are important determining factors for the HRQoL in ChD patients.

INTRODUCTION : Exercise-induced ventricular arrhythmia (EIVA) and autonomic imbalance are considered as early markers of heart disease in Chagas disease (ChD) patients. The objective of the present study was to verify the differences in the occurrence of EIVA and autonomic maneuver indexes between healthy individuals and ChD patients with no apparent cardiac involvement. METHODS : A total of 75 ChD patients with no apparent cardiac involvement, aged 44.7 (8.5) years, and 38 healthy individuals, aged 44.0 (9.2) years, were evaluated using echocardiography, symptom-limited treadmill exercise testing and autonomic function tests. RESULTS : The occurrence of EIVA was higher in the chagasic group (48%) than in the control group (23.7%) during both the effort and the recovery phases. Frequent ventricular contractions occurred only in the patient group. Additionally, the respiratory sinus arrhythmia index was significantly lower in the chagasic individuals compared with the control group. CONCLUSIONS : ChD patients with no apparent cardiac involvement had a higher frequency of EIVA as well as more vagal dysfunction by respiratory sinus arrhythmia. These results suggest that even when asymptomatic, ChD patients possess important arrhythmogenic substrates and subclinical disease.

Um dos maiores desafios da neurofisiologia é o de compreender a forma como a informação é transmitida através do sistema nervoso. O estudo do sistema nervoso tem várias aplicações, tanto na neurologia, permitindo avanços ao nível clínico, como noutras áreas, e.g., nos sistemas de processamento de informação baseados em redes neuronais. A transmissão de informação entre neurónios é feita por via de sinais elétricos. A compreensão deste fenómeno é ainda incompleta e há projectos a nível europeu e mundial com o objetivo de modular o sistema nervoso no seu todo de forma a melhor o compreender. Uma das teses que se desenvolve hoje em dia é a de que a transmissão de sinais elétricos no sistema nervoso é influenciada por fenómenos de sincronia. O objetivo desta dissertação é o de otimizar um protocolo de aquisição e análise de dados reais de eletroencefalograma e eletromiograma com o propósito de observar fenómenos de sincronia, baseando-se num algoritmo (análise por referência de fase, ou RPA, do inglês reference phase analysis) que deteta sincronias de fase entre os sinais de eletroencefalograma (EEG) e um sinal de referência, que é, no caso presente, o eletromiograma (EMG). A otimização deste protocolo e sua validação indicaram a existência de fenómenos significativos de sincronia no sinal elétrico, transmitido entre os músculos da mão e o córtex motor, no decorrer da ação motora.

Remote sensing - the acquisition of information about an object or phenomenon without making physical contact with the object - is applied in a multitude of different areas, ranging from agriculture, forestry, cartography, hydrology, geology, meteorology, aerial traffic control, among many others. Regarding agriculture, an example of application of this information is regarding crop detection, to monitor existing crops easily and help in the region’s strategic planning. In any of these areas, there is always an ongoing search for better methods that allow us to obtain better results. For over forty years, the Landsat program has utilized satellites to collect spectral information from Earth’s surface, creating a historical archive unmatched in quality, detail, coverage, and length. The most recent one was launched on February 11, 2013, having a number of improvements regarding its predecessors. This project aims to compare classification methods in Portugal’s Ribatejo region, specifically regarding crop detection. The state of the art algorithms will be used in this region and their performance will be analyzed.

Abstract: INTRODUCTION: Conjunctival swab PCR was evaluated as a tool to diagnose visceral leishmaniasis in dogs. METHODS: Conjunctival swab PCR was compared to indirect immunofluorescence antibody test and blood PCR. RESULTS: Indirect immunofluorescence was significantly correlated with conjunctival swab PCR (p < 0.05), but not with blood PCR (p > 0.05). In addition, conjunctival swab PCR was significantly associated with presence of clinical symptoms (p < 0.05), whereas blood PCR was associated with absence of clinical symptoms (p < 0.05). CONCLUSIONS: Results indicate that conjunctival swab PCR is useful in epidemiological surveys of canine visceral leishmaniasis.

This work reports the development of field-effect transistors (FETs), whose channel is based on zinc oxide (ZnO) nanoparticles (NPs). Using screen-printing as the primary deposition technique, different inks were developed, where the semiconducting ink is based on a ZnO NPs dispersion in ethyl cellulose (EC). These inks were used to print electrolyte-gated transistors (EGTs) in a staggered-top gate structure on glass substrates, using a lithium-based polymeric electrolyte. In another approach, FETs with a staggered-bottom gate structure on paper were developed using a sol-gel method to functionalize the paper’s surface with ZnO NPs, using zinc acetate dihydrate (ZnC4H6O4·2H2O) and sodium hydroxide (NaOH) as precursors. In this case, the paper itself was used as dielectric. The various layers of the two devices were characterized using X-ray diffraction (XRD), scanning electron microscopy (SEM), Fourier Transform Infrared spectroscopy (FTIR), thermogravimetric and differential scanning calorimetric analyses (TG-DSC). Electrochemical impedance spectroscopy (EIS) was used in order to evaluate the electric double-layer (EDL) formation, in the case of the EGTs. The ZnO NPs EGTs present electrical modulation for annealing temperatures equal or superior to 300 ºC and in terms of electrical properties they showed On/Off ratios in the order of 103, saturation mobilities (μSat) of 1.49x10-1 cm2(Vs)-1 and transconductance (gm) of 10-5 S. On the other hand, the ZnO NPs FETs on paper exhibited On/Off ratios in the order of 102, μSat of 4.83x10- 3 cm2(Vs)-1and gm around 10-8 S.

Tenofovir (TFV) is one of the most used antiretroviral drugs. However, it is associated with tubular damage with mitochondria as a possible target. Tubulopathy precedes glomerular dysfunction, thus classic markers of renal function like the glomerular filtration rate (GFR) do not detect early TFV damage. Prediction and management of drug induced renal injury (DIRI) rely on the mechanisms of the drug insult and in optimal animal models to explore it. Zebrafish (Danio rerio) offers unique advantages for assessing DIRI, since the pronephros is structurally very similar to its human counterpart and is fully developed at 3.5 days postfertilization. The main aim of the present work was to evaluate the effects of TFV, as well as its pro-drug, tenofovir disoproxil fumarate (TDF), on the GFR and in mitochondria morphology in tubular cells of zebrafish larvae. Lethality curves were performed to understand the relationship between drug concentration and lethality. LC10 was selected to explore the renal function using the FITC-inulin assay and to analyze the mitochondrial toxicity by electron microscopy on larvae exposed to TDF, TFV, paracetamol and gentamicin (positive controls) or water (negative control). Lethality curves showed that gentamicin was the most lethal drug, followed by TDF, TFV and paracetamol. Gentamicin and paracetamol decreased the GFR, but no differences were found for either TDF or TFV, when compared to controls (%FITC Control = 33±8; %FITC TDF = 35±10; %FITC TFV = 30±10; %FITC Gentamicin = 46±17; %FITC Paracetamol = 83±14). Tubular mitochondria from treated larvae were notably different from non-treated larvae, showing swelling, irregular shapes, decreased mitochondria network, cristae disruption and loss of matrix granules. These results are in agreement with the effects of these drugs in humans and thus, demonstrate that zebrafish larvae can be a good model to assess the functional and structural damage associated with DIRI.

Vanadium dioxide (VO2) is a promising material with large interest in construction industry and architecture, due to its thermochromic properties. This material may be used to create "smart" coatings that result in improvements in the buildings energy efficiency, by reducing heat exchanges and, consequently, the need for acclimatization. In this work, VO2 thin films and coatings were produced and tested in laboratory, to apply in architectural elements, such as glass, rooftop tiles and exterior paints. Thin films were produced by RF magnetron sputtering and VO2 nanoparticles were obtained through hydrothermal synthesis, aiming to create "smart" windows and tiles, respectively. These coatings have demonstrated the capability to modulate the transmittance of infrared radiation by around 20%. The VO2 nanoparticle coatings were successfully applied on ceramic tiles. The critical temperature was reduced to around 40ºC by tungsten doping. Ultimately, two identical house models were built, in order to test the VO2 coatings, in real atmospheric conditions during one of the hottest months of the year, in Portugal – August.
