Bacillus thuringiensis is a Gram-positive bacterium which main characteristic is the production of Cry proteins, that is toxic to some insects. These proteins, when ingested by susceptible insects, become active causing their death. In nature, it is possible to found B. thuringiensis strains which produce these proteins, but they differ in productivity (some of these isolates are more productive then others), and as to the toxicity levels of the produced proteins. Two B. thuringiensis strains that were highly effective against Spodoptera frugiperda larvae were chosen to verifying genetic mutation implication on Cry proteins productivity. One strain with a prolific spores production, while the other one only produced small amounts of spores. A genomic mutant library of these two isolates was, separately, constructed by genome Tn-5 transposon random insertion. Data analysis showed that mutation had a direct effect on the spores production, inducing an increase as well as a decrease in the production, according to the different strain observed. These results indicate, for the first time, that it is possible to use the described technique with B. thuringiensis, as well as the possibility to genetically breeding this bacteria. Another possibility introduced here is the possibility to do functional genetic studies mediated by mutagenesis in this bacterium.

The feeding choices of the mangrove crab Ucides cordatus for various mangrove plant leaves (Avicennia schaueriana, Laguncularia racemosa, and Rhizophora mangle) at different ages (mature, senescent pre-abscission, and decomposing leaves) were examined. In a controlled experiment set in a mangrove area, we evaluated crab selection for different plant leaves by analyzing foraging rate (number of leaves with predation marks) and leaf consumption. Crabs were housed individually in plastic containers and after a 3-day fast supplied with leaf fragments every 24 h for 72 h. Uneaten leaves were removed before each new food offering. No food selection was observed in the first day, but after this period, senescent leaves, which have a high polyphenol content, were rejected. On the third day, an interactive effect between plant species and leaf age was shown to affect leaf selection, with mature leaves of A. schaueriana and L. racemosa being more selected than the other treatments. This observation was consistent across crab sexes and ages. Our results show that food selection by this mangrove crab changes through time in fasted animals, suggesting that this variable must be controlled in food preference studies. © 2012 Springer Science+Business Media B.V.

The effects of ingested neem oil, a botanical insecticide obtained from the seeds of the neem tree, Azadirachta indica, on the midgut cells of predatory larvae Ceraeochrysa claveri were analyzed. C. claveri were fed on eggs of Diatraea saccharalis treated with neem oil at a concentration of 0.5%, 1% and 2% during throughout the larval period. Light and electron microscopy showed severe damages in columnar cells, which had many cytoplasmic protrusions, clustering and ruptured of the microvilli, swollen cells, ruptured cells, dilatation and vesiculation of rough endoplasmic reticulum, development of smooth endoplasmic reticulum, enlargement of extracellular spaces of the basal labyrinth, intercellular spaces and necrosis. The indirect ingestion of neem oil with prey can result in severe alterations showing direct cytotoxic effects of neem oil on midgut cells of C. claveri larvae. Therefore, the safety of neem oil to non-target species as larvae of C. claveri was refuted, thus the notion that plants derived are safer to non-target species must be questioned in future ecotoxicological studies. © 2012 Elsevier Ltd.

In the northwestern region of the state of São Paulo, Brazil, the freshwater crab Dilocarcinus pagei Stimpson, 1861 is usually sold as bait for several fish species for sport fishing. Thus, knowledge of the reproductive biology of this species is essential to provide information for maintaining natural stocks and implementing future farming initiatives. The objective of this study was to investigate the population dynamics of the red freshwater crab D. pagei, focusing on reproductive and juvenile recruitment periods, sex ratio, and individual growth. Sampling was performed monthly from October 2005 to September 2007 in the Marimbondo-Fumas Hydroelectric Plant reservoir, Rio Grande, municipality of Icém in northwestern São Paulo state. The individuals were sexed, carapace widths were measured, and the gonads were examined macroscopically. In total, 1002 individuals were analyzed, consisting of 568 males and 434 females, of which 4 were ovigerous and 35 carried early juveniles in the incubatory chamber. The largest numbers of reproductive individuals of both sexes were recorded during the winter and spring months before spawning, which occurred in the spring. Intense juvenile recruitment and the highest rainfall levels were observed during the Southern Hemisphere summer. The results indicate that spawning can be considered seasonal, and the simultaneous emergence of juveniles with high rainfall levels is probably the result of the higher availability of food and potential sites for protection against predators. The skewed sex ratio (♂ 1:0.76 ♀) may be associated with growth rates, mortality, food restriction, behavioral differences, migration, and differential habitat use between the sexes. The similar maximum size (CW∞) and growth constant (k) for males (k = 1.78 y-1, CW = 61.43 mm) and females (k = 1.67 y-1, CW = 60.66 mm) may be associated with different energy investment strategies between the sexes. The present study reports the use of biomarkers analyzes in mangrove root crab Goniopsis cruentata tissues to assess the environmental quality of two tropical estuarine areas. Animals from Ceará River estuary presented inhibition of ChE and GST enzymatic activities and higher rates of DNA damage with respect to those sampled in a pristine environment. G. cruentata appears to represent a proper species to monitor the quality of tropical estuaries. Since Ceará River is a legally protected area, this survey highlight the needs to implement actions to control pollution loads and improve the protection of natural ecosystems and resources. © 2013 Elsevier Ltd. The ovarian cycle histochemistry and its relationship with hepatopancreas weight in the blue crab Callinectes danae (Crustacea: Portunidae). -Acta Zoologica (Stockholm) 00:1-13. Several studies use macroscopic patterns of the ovarian development in crustaceans. Here, we examined the relationship between ovary histochemistry, changes in gonadosomatic and hepatosomatic indices against the macroscopic pattern of the ovarian development in Callinectes danae. Animals were collected in the south coast of São Paulo State, Brazil. Ovaries were macroscopically classified as juvenile, rudimentary, developing, intermediary, mature, and rudimentary ovigerous. Samples were fixed in 4% paraformaldehyde, processed for historesin, and stained with HE, protein, and neutral and acid polysaccharides detection. The juvenile oocytes are not enclosed by follicular cells and have fewer yolk nuclei being less intense in PAS reactivity than rudimentary oocytes. Developing oocytes show yolk granules and thick follicular cells. Yolk granules were positive for proteins and neutral polysaccharides. The intermediary stage is marked by a qualitative increase in yolk granules and the onset of chorion formation. In mature oocytes, cytoplasm is completely filled by yolk granules and the chorion is completely formed. Ovigerous ovaries have several atretic follicles and large quantities of hemocytes in the process of tissue reorganization. In C. danae, the changes in cell, goandosomatic and hepatosomatic indices coinciding with macroscopic observations and any combination of different macroscopic stages in a single pattern should be avoided. © 2011 The Royal Swedish Academy of Sciences.

The ultrastructure of the sperm of the gecarcinid land crab Cardisoma guanhumi is described by scanning and transmission electron microscopy. Ultrastructural sperm morphology lends support for the monophyletic origin of the Thoracotremata, and for the placement of C. guanhumi in that clade. Additionally, it further attests the low level of variability within the thoracotreme clade. With regard to ultrastructural morphology, small differences concerning the apical button, the hyaline periopercular rim, the inner acrosome zone, and the outer two lamellations in the outer acrosome zone have been found between the spermatozoa of C. guanhumi and Cardisoma carnifex, the only other species in this genus studied for spermatozoal morphology. The main purpose of this study was to provide an estimate for the onset of morphological sexual maturity in the freshwater crab Trichodactylus fluviatilis, from a population located in south-eastern Brazil, based on the relative growth relationships, as well as the characterization of ''handedness''. Monthly collections were carried out at night, from January 2006 to January 2007, in a small water body. In the laboratory, the crabs were sexed; the following body structures were measured and the estimate of onset of sexual maturity was given by the logistic function, as y a/(1\+becx), for 50% of the population to reach the adult phase (CW50carapace width). Right propodus length and abdomen width were the best-fit relationships to represent the onset of sexual maturity for males and females, respectively. Based on the logistic function, the onset of sexual maturity was estimated to occur at around 18mm CW for both sexes. The relative growth recorded for this species is consistent with the model for brachyuran crabs (Hartnoll RG. 1982, Growth. In: Bliss, DE, editor. The biology of Crustacea: embryology, morphology and genetics. New York: Academic Press. p. 111-196), except for the abdomen growth, which is likely to be an adaptive condition related to protection of the newly hatched young. © 2013 Taylor & Francis.

Free amino acids (FAA) are used principally as substrate in protein synthesis and the source of energy in aerobic catabolism. In marine fish, embryo and larvae FAA are used to maintain body fluid osmolality during fish early development. However, there is essentially no information about FAA concentrations in early ontogeny of freshwater neotropical species in comparison to marine fishes. Therefore, the aim of this study was to evaluate the FAA concentrations in pacu, Piaractus mesopotamicus, eggs and larvae. Broodstock fish were induced to spawn and ovulated females were stripped of their eggs and immediately sampled for analysis. Larvae were sampled right after hatching (HL) and after the completion of the yolk-sac absorption (YSA). The wet weight of the HL and YSA larvae amounted to 0.5±0.1mg and 1.1±0.3mg, respectively. HL larvae showed higher levels of most of the indispensable amino acids (IAA) in comparison to eggs and YSA larvae. Exceptions were observed with His and Trp that showed higher or similar levels, respectively, in YSA larvae. The FAA Orn, Tau, Glu, Gln, Gly, and Tyr increased concentrations in both larval stages while that of Tau was found in higher concentration in all analyzed stages. Also, the concentrations of Asn, Ala, Pro, Ser, and Asp were higher in HL larvae. Both larval stages displayed a rise in total free IAA/total free DAA (dispensable amino acids) ratio. The authors conclude that the highest level of FAA in HL pacu larvae is indicative of active proteolysis of yolk reserves and a probable catabolism regulation of some FAA through spare-effect. In addition, Tau is one of the major FAA occurring during pacu ontogeny and may be performing regulation on body fluid osmolality regulation. © Copyright by the World Aquaculture Society 2013.

This is a histological and histochemical analysis of the terminal portion of the female reproductive system and genital ducts of the blue land crab (Cardisoma guanhumi). Animals were collected in the Jaguaribe estuary (Ceará, Brazil) and dissected. Genital duct fragments were fixed and submitted to different staining techniques. The female reproductive system consists of a pair of ovaries and a pair of genital ducts. In the mid-posterior portion of each lobe, the ovaries communicate with the genital ducts, which are subdivided into oviduct, spermatheca, vagina, and gonopore. Histologically, the spermatheca of C. guanhumi is composed of columnar secretory epithelium and is divided into a dorsal zone and a ventral zone, the latter covered internally by a cuticle layer. Both zones are enveloped by a thin layer of loose connective tissue. Histological cross sections revealed the vagina to be concave, a pattern considered phylogenetically more advanced than the simple, tubular form. The reproductive biology of a species includes factors beyond its sexual maturity, fecundity and reproductive period, and may extend to the differential distribution of individuals. The reproductive dynamics of the blue crab Callinectes ornatus was investigated through monthly collections over the course of 2 years in three bays on the southeastern coast of Brazil. For each bay, six transects were established, four of them parallel to the beach line (at depths of 5, 10, 15, and 20 m), one transect exposed to wave action, and another sheltered from waves. Females and males were classified according to the gonadal maturation stage, and were grouped as individuals with reproductive potential (mature gonads or breeding females) or not (rudimentary gonads or in development). Analyses using ordination techniques (PCA) and gradient analysis (CCA) showed that 82.13 % of environmental variations were explained by the transect arrangement, and these characteristics explained 86.70 % of the differential distribution of female crabs and 96.57 % of the distribution of males. These results indicate that females with reproductive potential were more abundant in deeper regions, while females with rudimentary or developed gonads were abundant in shallower habitats and areas sheltered from wave action. Thus, the distribution of C. ornatus in these bays was linked to their reproductive state, as part of the reproductive strategy of the population. © 2013 Senckenberg Gesellschaft für Naturforschung and Springer-Verlag Berlin Heidelberg.

A alimentação é considerada crítica em cultivo de larvas de decápodes. No entanto, algumas espécies podem apresentar reservas nutritivas suficientes para completar o desenvolvimento larval sem a necessidade de alimentação externa (conhecido como desenvolvimento larval lecitotrófico). No presente estudo, dois experimentos foram realizados para verificar se o callianassídeo Lepidophthalmus siriboia tem comportamento lecitotrófico ou se precisa de alimento externo para completar o desenvolvimento larval: Experimento 1, larvas submetidas a um período inicial de alimentação e, Experimento 2, larvas submetidas a um período inicial de inanição. Em ambos os experimentos, observou-se altas taxas de sobrevivência, com apenas 2 megalopas e 1 zoea III mortos. Estes resultados sugerem fortemente que larvas de L. siriboia são lecitotróficas, tendo reservas suficientes para completar o desenvolvimento larval, enquanto o estágio de megalopa apresenta lecitotrofia facultativa. Os períodos larvais de cada estágio nos tratamentos foram bastante semelhantes. No entanto, foram observadas algumas diferenças significantes entre alguns períodos, os quais podem estar relacionados às condições de cultivo, provavelmente devido a fatores abióticos, à variabilidade individual da condição larval, ou ainda a fatores como estresse causado às fêmeas ovígeras durante embriogênese.

Larvas do caranguejo da lama Panopeus lacustris Desbonne, 1867 foram cultivadas em laboratório a partir de fêmeas ovígeras coletadas na área estuarina do Rio Caeté na região Amazônica. O desenvolvimento completo desta espécie consistiu em 4 de zoea e um megalopa, onde cada estágio foi descrito e ilustrado em detalhes. Os resultados foram comparados com os de outros estudos anteriores sobre o desenvolvimento larval das espécies pertencentes ao gênero Panopeus e brevemente discutidos. Além da descrição das larvas do desenvolvimento completo de P. lacustris, foi descrita e ilustrada a primeira fase de zoea de treze espécies de caranguejos braquiúros coletados no mesmo estuário: P. lacustris, P. americanus Saussure, 1857, Eurytium limosum (Say, 1818), Sesarma curacaoense De Man, 1892, Sesarma rectum Randall, 1840, Armases rubripes (Rathbun, 1897), Aratus pisonii (H. Milne Edwards, 1837), Ocypode quadrata (Fabricius, 1787), Uca rapax (Smith, 1870), Uca maracoani (Latreille, 1802), Uca thayeri Rathbun, 1900, Ucides cordatus (Linnaeus, 1763) e Pachygrapsus gracilis (Saussure, 1858). Apenas a zoea I de P. lacustris não foi descrito novamente. As características morfológicas dessas espécies são comparadas com as descrições originais. Para facilitar o estudo de material coletado no plâncton, foi desenvolvida uma chave para identificação das espécies descritas neste estudo.
