The objective of this study is to use different in vitro culture systems of preantral follicles from Nelore breed bovine fetuses in the last gestation quarter. The evaluation of treatments considered the time of growth of isolated follicles. Preantral follicles were mechanically isolated and submitted to the individual culture, for 9 days, in media no supplemented or supplemented with fetal calf serum (FCS), bovine serum albumin (BSA) or synthetic defined supplement substitute of serum KnockoutSR (KNO). We have also evaluated the effects of collagen gel or fetal calf fibroblast monolayer as substratum for in vitro cultures. The increase on the follicular diameter was followed in the first day (0 h), at the 72 h, 144 h and 216 h. Considering cultures of isolated follicles, the results have shown that the association between media supplemented with FCS and collagen gel was significantly more efficient on the increase of the follicular diameter than other treatments. It is not still established a system of appropriate cultivation that sustains the differentiation and multiplication of the granular cells and that maintains the contact of the same ones with the oocyte to provide molecules and factors that supply the metabolic demand. We also understand that our results also represent another promising step on the search for the ultimate system of in vitro culture of preantral follicles from bovines.

The high rate of mineralization of organic matter on savannah soils, which is reached fi ve times faster than in temperate regions, leads us to the challenge of electing the best system of management that maintains and/or increase it in soil, guaranteeing its quality and sustainability. In this sense the present research aimed to study the effects of green, organic and mineral manure on the chemical properties on the chemical properties of an Oxisol, on Savannah area, cultivated with cotton (Gossypium hirsutum) under conventional tillage and no-tillage in the pasture fi eld (Brachiaria decumbens) for 20 years. The experimental design was a randomized block design with split plots. The main plots consisted of two treatments: conventional tillage and no-tillage, and the subplots of six treatments: control (no fertilizer), mineral fertilizer recommended for the crop, according to the soil chemical analysis, organic fertilizer (cattle manure - 20 t ha-1), organic fertilizer (cattle manure - 20 t ha-1) + 1/2mineral fertilization recommended according to the analysis of soil, green manure-1 (Crotalaria juncea) and green manure-2 (Pennisetum americanum). There were studied the following soil chemical properties: P, OM, pH, K, Ca, Mg, Al, Al + H, S, exchange capacity cations and base saturation. The soil samples for the analysis were performed on layers of 0,00-0,05 m, 0,05-0,10 and 0,10-0,20 m. Then it came the following conclusions: the fertilization interfere in soil chemical properties and the preparation did not interfere, the cattle manure and its association with the mineral fertilizer caused increasing level elements in the soil, Crotalaria juncea and Pennisetum americanum did not infl uence on soil chemical properties.

Sugarcane is one of the most important crops of the Brazilian agribusiness and this importance justifies the use of techniques that allow the implementation of cultivation systems capable of reducing the variability of soil characteristics and the establishment of efficient agricultural planning. The aim of the present work was the planning and implementation of cultivation systems for sugarcane using geostatistical techniques, in the Pereira Barreto region, SP. An area of 505 ha was mapped using the global positioning system (GPS) and a Digital Elevation Model was elaborated (MDE). Soil samples were collected for 0-0.25 m depth, in each 7 ha, for their chemical attributes and texture characterization. Data were analyzed by descriptive statistics and geostatistics. The determination of the spatial distribution of soil granulometric and chemical attributes allowed the allocation of the studied sugarcane varieties according to soil fertility and clay content. The kriging maps of soil granulometric and chemical attributes brought useful information to the establishment of production environments with different soil and crop managements. The identification of different environments by means of geostatistical techniques allowed the precise planning of the sugarcane cultivation, as well as the adequacy of fertilization practices and the allocation of suitable sugarcane varieties adapted to the conditions imposed by differences in the soil attributes.

Aiming to evaluate the association of hydroponic lettuce cultivation with residues from a fish intensive breeding system, a project was carried out in the Aquaculture Center in the FCAV-UNESP at Jaboticabal, SP, Brazil. A closed system was designed in order to allow the water to circulate through the fish tanks, a clarifier tank for removal of residues, a reservoir for biological conversion of ammonia into nitrate, and the hydroponic system. After this process, water returned back to the fish tanks. Three varieties of lettuce, constituting the treatments with four repetitions were evaluated. The results of the chemical analysis of the residual water from the fish tanks indicated the presence of the majority of the mineral nutrients necessary for vegetable development. Their concentration was close to that found in nutrient solutions, used for lettuce hydroponic cultivation, except for potassium and magnesium. The low concentration of magnesium in the water did not prevent lettuce development, although the plants presented visual symptoms of deficiency of this nutrient. Differences were not found between the varieties produced, regarding productivity and the mean weight of fresh plants.

Cottonseed meals (CSM) with different protein levels (35, 39 and 40%) were evaluated to determine their chemical composition and measure apparent digestibility coefficients (ADC) of dry matter, protein, amino acids, energy, lipid and minerals for Nile tilapia. Apparent digestibility coefficients were determined using a reference purified diet containing 0.1% of chromic oxide indicator, and test diets contained 60% of reference diet and 40% of test ingredients. Results showed that apparent digestibility coefficients of cottonseed meals were different (p < 0.05). The apparent digestibility coefficients were: dry matter, 53.45-54.32%; crude protein, 71.56-74.81%; energy, 53.80-58.60%; lipid, 85.37-89.11%; phosphorus, 33.75-34.50%; calcium, 41.21-43.75%; iron, 62.02-79.85%; zinc, 0-67.41%; copper, 13.37-14.27%; essential amino acids, 71.39-79.17%; and non-essential amino acids, 77.08-82.58%. The ADC of protein did not reflect the majority of amino acids ADC showing variation in individual digestibility of amino acids and among cottonseed meals. Cottonseed meal can be used as protein succedaneum in Nile tilapia diets.

Variations in soil attributes occur based on relief forms and parent material. The objective of this research was to study the influences of geomorphic surfaces on the spatial distribution of soil attributes in a sugarcane cultivation area. First, 530,67 hectares were mapped by using a Global Positioning System. Then, a Digital Elevation Model was developed and geomorphic surfaces were identified and delimited, according to topographic and stratigraphic criteria based on detailed field investigations. Soil samples were collected every 7 ha, at 0.0-0.25 m and 0.80-1.00 m depths, totalizing 134 samples. Texture, Ca, K, Mg, SB, CEC, V%, pH, and OMS analyses were carried out as well as descriptive statistics and geostatistics analysis. It was concluded that the geostatistic techniques and digital elevation model helped to notice that soil attributes presented limits close to the geomorphic surfaces ones.

Phosphorus is considered an essential element for plants, but it is found in low amounts in Brazilian soils, mainly in areas destined to pastures, and the application of organic matter contributes to the improvement of physical, chemical and biological soil attributes. The research had the objective of evaluating the application effects of the Bokashi organic compound on soil macro and micronutrients contents and leaves, as well as on Brachiaria brizantha cv. Marandú dry mass yield, evaluating the potential of the organic fertilizer as phosphorus source, in comparison with conventional chemical fertilizers. Different organic compound doses supplied the soil with 17 kg P2O5 ha-1, 34 kg P2O5 ha-1, 68 kg P2O5 ha-1, 136 kg P 2O5 ha-1, and 204 kg P2O5 ha-1, comparing it with the conventional chemical fertilizer and the control. The results revealed that the organic fertilizer Bokashi can adequately replace the conventional chemical fertilizer of Brachiaria brizantha cv. Marandú, supplying the soil and plant with appropriate nutrients levels and maintaining the dry mass yield.

Agaricus brasiliensis is a Brazilian basidiomycete which has been cultivated and consumed around the world as a therapeutic food. Casing layer is one of the most important steps on A. brasiliensis cultivation and European peat is the most used casing layer on Agaricus bisporus cultivation. Besides the importance of it on mushroom cultivation the peat import could be a risk of exotic organism introduction. Alternative as Brazilian peat is barely used for mushroom growers in Brazil. Thus, the objective of this work was to evaluate Brazilian peat with and without pasteurization as casing layer on A. brasiliensis cultivation. The fungus was previously grown on wheat grains and transferred to a substratum prepared by composted traditional method. After mycelium colonization of the substratum a pasteurized or non pasteurized Brazilian peat (casing layer) was added. It was concluded that pasteurization of the casing layer increases in 30% yield after 65 days of cultivation. There is no difference of yield for pasteurized and non pasteurized casing layer until 30 days of cultivation. An increase of flies is observed in non pasteurized casing layer. The production flush is easily perceived with pasteurized casing layer but not with non pasteurized casing layer.

Objective: This case-control study analyzed mass spectrometry fingerprinting patterns of culture media samples used for embryo culture to predict embryo implantation. Methods: The culture medium harvested after embryo transfer of 22 embryos from 13 patients was used for the experiments. After embryo transfer, the remaining culture media were collected and samples were split in positive (n=8) and negative (n=14) implantation groups according to implantation outcomes (100% or 0% of implantation). Samples were individually diluted and injected directly to the Electrospray ionization (ESI) MS coupled to a Quadrupole Time-of-flight MS (Q-ToF-MS).Ions relative intensities of each spectrum were considered. Data analysis was conducted in MatLab 7.0 version using Partial Least Squares - Discriminant Analysis toolbox. Results: There were 3027 observed ions at 100% and 0% implantation groups by ESI-Q-ToF-MS. The statistical model could categorize the samples in two clusters, based on their positive and negative implantation outcomes. Less intense ions present in the mass spectra with statistical significance have contributed to the major differences to group distinction. Conclusions: Positive and negative implantation embryos showed a specific biochemical pattern present in culture media, which could be detected as a fast, simple and non-invasive way. This biochemical profile could help the selection of the most viable embryo, improving single embryo transfer and thus eliminating the risk and undesirable outcomes of multiple pregnancies. © Todos os direitos reservados a SBRA - Sociedade Brasileira de Reprodução Assistida.

A laboratory culture of Ankistrodesmus gracilis algae was evaluated by studying the biology of the species and its chemical composition in a traditional medium (CHU 12) and in two alternative culture media, NPK (20-5-20) and macrophyte (Eichhornia crassipes) + NPK, in three different types of recipients (fiberglass, carboy and plastic bag). First peak in the growth curve of Ankistrodesmus gracilis occurred on the ninth day in macrophyte + NPK medium (74.16 x 10 5 cells mL -1) in a fiberglass recipient. However, highest density (p < 0.01) was reported in medium CHU 12 (122.87 x 10 5 cells mL -1) in a plastic bag on the twelfth day. Cell density was over 70 x 10 5 cells mL -1 starting on the twelfth day. Growth rate of A. gracilis was similar (p > 0.05) in culture media in the three recipients. Protein and fiber were similar (p > 0.05) in the treatments, but lipids were higher (p < 0.05) in NPK. Nitrate, ammonia, total phosphorus and orthophosphate contents were over 1 mg L -1 in NPK (p < 0.01). Results show that alternative media, such as NPK and macrophyte + NPK, are possible for large-scale culture of A. gracilis cultured in three types of recipients. Costs are low, occupying less space when cultured in plastic bags and in the laboratory.

The aim of the study was to evaluate the serum biochemical parameters of the Nile tilapia (Oreochromis niloticus), Thai Chitralada lineage, raised under an intensive pisciculture system and captured by the cast net technique. Blood samples from 40 individuals were obtained by caudal venopuncture. Mean body weight was 453 ± 52 g. The serum biochemical parameters evaluated were total protein, albumin, globulins, acid uric, creatinine, urea, calcium, inorganic phosphorus, Ca/P ratio, alkaline phosphatase, and magnesium. In general, the data showed similar parameters in comparison to those established for scale fish, exception of the acid uric.

O consumo de águas salinas pelas plantas na maioria dos casos afetam o rendimento das culturas em suas diversas fases prejudicando desenvolvimento e produção. O trabalho avaliou o consumo hídrico da rúcula (Eruca sativa) em sistema hidropônico NFT com águas salinas. Foram analisados seis níveis crescentes de salinidade da água (CEa), quais sejam: 0,2; 1,2; 2,2; 3,2; 4,2 e 5,2 (dS m-1) utilizaram-se duas fontes de sais: águas salobras (AS) e NaCl. Foi determinado o volume evapotranspirado por planta (VETc) no sistema durante o cultivo. O consumo hídrico nos primeiros dias após transplantio foi de 250 mL dia-1 aproximadamente para ambas as testemunhas e decresceram com o aumento da salinidade, a eficiência de utilização da água foi reduzida revelando uma resposta linear decrescente em função da salinidade ocorrendo uma resposta negativa das plantas que apresentaram uma redução na quantidade de folhas de 3,34%.

Forty-eight pigs with 63 day olds and average weight of 23.40±1.3 kg, were allotted in a randomized block design with three treatments(0%, 10% and 20% of cottonseed meal (FA) in diets) and eight replicates with two pigs (one barrow and one gilt). The blocks were used to control changes in initial body weight. Experimental period was the growth phase (63 to 106 days old) and growing-finishing (63 to 147 days old). In each phase the diets were isonutritives, varying however the levels of crude fiber. The levels of cottonseed meal (FA) did not affect (p>0.05) the performance of animals during the growth phase or during the total period of evaluation (growingfinishing). The qualitative parameters of manure indicated that the FA determined decrease in the nitrogen and phosphorus content, but only in the growth phase. In the finishing phase the increase in the FA inclusion promoted an increase in dry matter content. Although FA can be used in up to 20% in diets for pigs in the growing and finishing phase, it is necessary to include L-lysine and soybean oil which causes an increase in cost per kg produced pig.
