Este trabalho teve como objetivo investigar o efeito da manipulação neonatal sobre a resposta da prolactina (PRL) ao estresse por vapores de éter em ratas de 11, 28 e 75 dias de idade. A manipulação neonatal consistia em manipular gentilmente os filhotes durante 1 minuto do 1o ao 10o dia de vida. Os animais foram divididos em dois grupos: não-manipuladas e manipuladas e em três idades: 11, 28, e 75 dias. O último grupo foi estudado nas fases do diestro II e estro. Aos 11 e 28 dias de idade as coletas de sangue foram realizadas através de decapitação antes e aos 2, 5 e 10 minutos após o estresse de 1 minuto por vapores de éter. Nos animais adultos, a veia jugular externa direita foi canulada na tarde do proestro e diestro I e o experimento realizado as 9:00 horas da manhã do estro e do diestro II, respectivamente. Amostras de 600 µL de sangue foram coletadas antes e aos 2, 5, 10, 15 e 30 minutos após o estresse de 1 minuto por vapores de éter. As concentrações plasmáticas de PRL (antes, 2, 5, 10, 15 e 30 minutos após o estresse) e as de estradiol (antes do estresse) foram determinadas por radioimunoensaio. Os resultados mostram que as concentrações plasmáticas basais de PRL aumentaram em ambos os grupos (não-manipuladas e manipuladas) dos 11 aos 28 dias e dos 28 aos 75 dias de idade nas fêmeas em estro e diestro II, com exceção das não-manipuladas em estro, nas quais não foi observado um aumento significativo da concentração plasmática de PRL dos 28 aos 75 dias de idade. O grupo das ratas manipuladas de 28 dias de idade apresentou uma menor concentração plasmática basal de PRL e aos 5 e 10 minutos após o estresse quando comparadas ao grupo das não-manipuladas da mesma idade (28 dias). Não foi observada a resposta da PRL ao estresse em ratas de 11 e 28 dias de idade. Na manhã do estro a resposta da prolactina ao estresse não foi diferente entres os grupos (não-manipuladas e manipuladas), contudo na manhã do diestro II, o grupo das manipuladas apresentou uma resposta de menor magnitude quando comparada ao grupo das não-manipuladas. A concentração plasmática de estradiol não foi diferente entre o estro e o diestro II e nem entre as não-manipuladas e manipuladas. Em conjunto estes resultados indicam que: as concentrações plasmáticas de PRL aumentaram no decorrer das três idades (exceto no grupo das não-manipuladas dos 28 aos 75 dias de idade na manhã do estro); a menor concentração plasmática basal de PRL observada nas ratas manipuladas aos 28 dias de idade pode ser resultado do retardo da instalação da puberdade observado em ratas manipuladas no período neonatal; o período hiporresponsivo da PRL ao estresse se estende até a idade pré-puberal; a concentração plasmática de estradiol no dia do experimento parece não ser responsável pelo efeito da manipulação neonatal sobre a resposta da PRL ao éter que somente foi observado nas fêmeas na manhã do diestro II.

The course of in vivo infection of five isolates of Yersinia pseudotuberculosis was followed for three weeks in Swiss mice. The strains were isolated from diarrheic and normal feces and mesenteric lymph nodes of healthy and sick stock animals. Four strains of serogroup O:3 and one of serogroup O:1a, with and without the virulence plasmid, were inoculated intragastrically and intravenously in the mice. Groups of five animals were sacrificed at 6 h and 3, 6, 10, 15, and 21 days after inoculation, and organs and tissues were checked for possible macroscopic alterations. Development of infection was monitored at these times by performing viable bacterial counts in homogenates of selected tissues. The animals were cheked daily for clinical alterations. The results of the study showed that strains with the virulence plasmid infected organs and tissues at various times and at varying intensity by both routes of infection, the strain of type O:1a being the most invasive. Moreover, clinical and pathological alterations occurred only in animals inoculated with bacteria carrying the virulence plasmid, regardless of the route of infection.

In this work, the analysis of electroosmotic pumping mechanisms in microchannels is performed through the solution of Poisson-Boltzmann and Navier Stokes equations by the Finite Element Method. This approach is combined with a Newton-Raphson iterative scheme, allowing a full treatment of the non-linear Poisson-Boltzmann source term which is normally approximated by linearizations in other methods.

A new selective sensor based on molecularly imprinted polymers (MIPs) was developed for the determination of hexazinone (HXZ) in environmental samples. MIPs were synthesized using a non-covalent approach, and selection of the monomers employed in the polymerization reaction was carried out by molecular modeling. Three functional monomers with high (2-vinylpyridine (MP17)) and intermediate (methacrylic acid (MP12) and acrylamide (MP5)) energies of binding to the template (HXZ) were selected for preparation of the MIPs, in order to conduct comparative studies and validate the theoretical data. For sensor construction, carbon pastes were modified with each MIP or NIP (non-imprinted polymer), and HXZ determination was performed using differential pulse adsorptive cathodic stripping voltammetry (DPAdCSV). All parameters affecting the sensor response were optimized. In HCl at pH 2.5, the sensor prepared with MP17 (5% w/w in the paste) showed a dynamic linear range between 1.9 × 10−11 and 1.1 × 10−10 mol L−1, and a detection limit of 2.6 × 10−12 mol L−1, under the following conditions: accumulation time of 200 s at a potential of −0.5V, scan rate of 50 mVs−1, pulse amplitude of 60 mV, and pulse width of 50 ms. The sensor was selective in the presence of other similar compounds, and was successfully applied to the analysis of HXZ in river water samples.

OBJECTIVES Creation of an atraumatic, hearing-preservation cochleostomy is integral to the future of minimally invasive inner ear surgery. The goal of this study was to develop and characterize a novel chemical approach to cochleostomy. STUDY DESIGN Prospective animal study. SETTING Laboratory. METHODS Experimental animal study in which phosphoric acid gel (PAG) was used to decalcify the otic capsule in 25 Hartley guinea pigs. Five animals in each of 5 surgical groups were studied: (1) mechanically opening the auditory bulla alone, (2) PAG thinning of the basal turn otic capsule, leaving endosteum covered by a layer of bone, (3) micro-pick manual cochleostomy, (4) PAG chemical cochleostomy, exposing the endosteum, and (5) combined PAG/micro-pick cochleostomy, with initial chemical thinning and subsequent manual removal of the last osseous layer. Preoperative and postoperative auditory brainstem responses and otoacoustic emissions were obtained at 2, 6, 10, and 16 kHz. Hematoxylin and eosin-stained paraffin sections were compared. RESULTS Surgical and histologic findings confirmed that application of PAG provided reproducible local bone removal, and cochlear access was enabled. Statistically significant auditory threshold shifts were observed at 10 kHz (P = .048) and 16 kHz (P = .0013) following cochleostomy using PAG alone (group 4) and at 16 kHz using manual cochleostomy (group 3) (P = .028). No statistically significant, postoperative auditory threshold shifts were observed in the other groups, including PAG thinning with manual completion cochleostomy (group 5). CONCLUSION Hearing preservation cochleostomy can be performed in an animal model using a novel technique of thinning cochlear bone with PAG and manually completing cochleostomy.

Although the permanently to seasonally ice-covered Arctic Ocean is a unique and sensitive component in the Earth's climate system, the knowledge of its long-term climate history remains very limited due to the restricted number of pre-Quaternary sedimentary records. During Polarstern Expedition PS87/2014, we discovered multiple submarine landslides over a distance of >350 km along Lomonosov Ridge. Removal of younger sediments from steep headwalls has led to exhumation of Miocene to early Quaternary sediments close to the seafloor, allowing the retrieval of such old sediments with gravity cores. Multi-proxy biomarker analyses of these gravity cores reveal for the first time that the late Miocene central Arctic Ocean was relatively warm (4-7°C) and ice-free during summer, whereas sea ice occurred during spring and autumn/winter. A comparison of our proxy data with Miocene climate simulations seems to favour relatively high late Miocene atmospheric CO2 concentrations. These new findings from the Arctic region provide new benchmarks for groundtruthing global climate reconstructions and modeling.

Fluid flow through the axial hydrothermal system at fast spreading ridges is investigated using the Sr-isotopic composition of upper crustal samples recovered from a tectonic window at Pito Deep (NE Easter microplate). Samples from the sheeted dike complex collected away from macroscopic evidence of channelized fluid flow, such as faults and centimeter-scale hydrothermal veins, show a range of 87Sr/86Sr from 0.7025 to 0.7030 averaging 0.70276 relative to a protolith with 87Sr/86Sr of ~0.7024. There is no systematic variation in 87Sr/86Sr with depth in the sheeted dike complex. Comparison of these new data with the two other localities that similar data sets exist for (ODP Hole 504B and the Hess Deep tectonic window) reveals that the extent of Sr-isotope exchange is similar in all of these locations. Models that assume that fluid-rock reaction occurs during one-dimensional (recharge) flow lead to significant decreases in the predicted extent of isotopic modification of the rock with depth in the crust. These model results show systematic misfits when compared with the data that can only be avoided if the fluid flow is assumed to be focused in isolated channels with very slow fluid-rock exchange. In this scenario the fluid at the base of the crust is little modified in 87Sr/86Sr from seawater and thus unlike vent fluids. Additionally, this model predicts that some rocks should show no change from the fresh-rock 87Sr/86Sr, but this is not observed. Alternatively, models in which fluid-rock reaction occurs during upflow (discharge) as well as downflow, or in which fluids are recirculated within the hydrothermal system, can reproduce the observed lack of variation in 87Sr/86Sr with depth in the crust. Minimum time-integrated fluid fluxes, calculated from mass balance, are between 1.5 and 2.6 * 10**6 kg/m**2 for all areas studied to date. However, new evidence from both the rocks and a compilation of vent fluid compositions demonstrates that some Sr is leached from the crust. Because this leaching lowers the fluid 87Sr/86Sr without changing the rock 87Sr/86Sr, these mass balance models must underestimate the time-integrated fluid flux. Additionally, these values do not account for fluid flow that is channelized within the crust.

Using the sea ice proxy IP25 and phytoplankton-derived biomarkers (brassicasterol and dinosterol) Arctic sea-ice conditions were reconstructed for Marine Isotope Stage (MIS) 3 to 1 in sediment cores from the north of Barents Sea continental margin across the Central Arctic to the Southern Mendeleev Ridge. Our results suggest more extensive sea-ice cover than present-day during MIS 3, increasing sea-ice growth during MIS 2 and decreased sea-ice cover during the last deglacial. The summer ice edge sustained north of the Barents Sea even during extremely cold (i.e., Last Glacial Maximum (LGM)) as well as warm periods (i.e., Bølling-Allerød). During the LGM, the western Svalbard margin and the northern Barents Sea margin areas were characterized by high concentrations of both IP25 and phytoplankton biomarkers, interpreted as a productive ice-edge situation, caused by the inflow of warm Atlantic Water. In contrast, the LGM high Arctic proper (north of 84°N) was covered by thick permanent sea ice throughout the year with rare break up, indicated by zero or near-zero biomarker concentrations. The spring/summer sea-ice margin significantly extended southwards to the southern Lomonosov Ridge and Mendeleev Ridge during the LGM. Our proxy reconstructions are very consistent with published model results based on the North Atlantic/Arctic Ocean Sea Ice Model (NAOSIM).
