The Ph.D. thesis deals with the conformational study of individual cylindrical polymer brush molecules using atomic force microscopy (AFM). Imaging combined with single molecule manipulation has been used to unravel questions concerning conformational changes, desorption behavior and mechanical properties of individual macromolecules and supramolecular structures. In the first part of the thesis (chapter 5) molecular conformations of cylindrical polymer brushes with poly-(N-isopropylacrylamide) (PNIPAM) side chains were studied in various environmental conditions. Also micelle formation of cylindrical brush-coil blockcopolymers with polyacrylic acid side chains and polystyrene coil have been visualized. In chapter 6 the mechanical properties of single cylindrical polymer brushes with (PNIPAM) side chains were investigated. Assuming that the brushes adopt equilibrium conformation on the surface, an average persistence length of lp= (29 3) nm was determined by the end-to-end distance vs. contour length analysis in terms of the wormlike chain (WLC) model. Stretching experiments suggest that an exact determination of the persistence length using force extension curves is impeded by the contribution of the side chains. Modeling the stretching of the bottle brush molecule as extension of a dual spring (side chain and main chain) explains the frequently observed very low persistence length arising from a dominant contribution of the side chain elasticity at small overall contour lengths. It has been shown that it is possible to estimate the true persistence length of the bottle brush molecule from the intercept of a linear extrapolation of the inverse square root of the apparent persistence length vs. the inverse contour length plot. By virtue of this procedure a true persistence length of 140 nm for the PNIPAM brush molecules is predicted. Chapter 7 and 8 deal with the force-extension behavior of PNIPAM cylindrical brushes studied in poor solvent conditions. The behavior is shown to be qualitatively different from that in a good solvent. Force induced globule-cylinder conformational changes are monitored using molecule specific force spectroscopy which is a combined AFM imaging and SMFS technique. An interesting behavior of the unfolding-folding transitions of single collapsed PNIPAM brush molecules has been observed by force spectroscopy using the so called fly-fishing mode. A plateau force is observed upon unfolding the collapsed molecule, which is attributed to a phase transition from a collapsed brush to a stretched conformation. Chapter 9 describes the desorption behavior of single cylindrical polyelectrolyte brushes with poly-L-lysine side chains deposited on a mica surface using the molecule specific force spectroscopy technique to resolve statistical discrepancies usually observed in SMFS experiments. Imaging of the brushes and inferring the persistence length from a end-to-end distance vs. contour length analysis results in an average persistence length of lp = (25 5) nm assuming that the chains adopt their equilibrium conformation on the surface. Stretching experiments carried out on individual poly-L-lysine brush molecules by force spectroscopy using the fly-fishing mode provide a persistence length in the range of 7-23 nm in reasonable accordance with the imaging results. In chapter 10 the conformational behavior of cylindrical poly-L-lysine brush-sodium dodecyl sulfate complexes was studied using AFM imaging. Surfactant induced cylinder to helix like to globule conformational transitions were observed.

Protein aggregation and formation of insoluble aggregates in central nervous system is the main cause of neurodegenerative disease. Parkinsons disease is associated with the appearance of spherical masses of aggregated proteins inside nerve cells called Lewy bodies. -Synuclein is the main component of Lewy bodies. In addition to -synuclein, there are more than a hundred of other proteins co-localized in Lewy bodies: 14-3-3 protein is one of them. In order to increase our understanding on the aggregation mechanism of -synuclein and to study the effect of 14-3-3 on it, I addressed the following questions. (i) How -synuclein monomers pack each other during aggregation? (ii) Which is the role of 14-3-3 on -synuclein packing during its aggregation? (iii) Which is the role of 14-3-3 on an aggregation of -synuclein seeded by fragments of its fibrils? In order to answer these questions, I used different biophysical techniques (e.g., Atomic force microscope (AFM), Nuclear magnetic resonance (NMR), Surface plasmon resonance (SPR) and Fluorescence spectroscopy (FS)).

The aim of this thesis was to apply the techniques of the atomic force microscope (AFM) to biological samples, namely lipid-based systems. To this end several systems with biological relevance based on self-assembly, such as a solid-supported membrane (SSM) based sensor for transport proteins, a bilayer of the natural lipid extract from an archaebacterium, and synaptic vesicles, were investigated by the AFM. For the characterization of transport proteins with SSM-sensors proteoliposomes are adsorbed that contain the analyte (transport protein). However the forces governing bilayer-bilayer interactions in solution should be repulsive under physiological conditions. I investigated the nature of the interaction forces with AFM force spectroscopy by mimicking the adsorbing proteoliposome with a cantilever tip, which was functionalized with charged alkane thiols. The nature of the interaction is indeed repulsive, but the lipid layers assemble in stacks on the SSM, which expose their unfavourable edges to the medium. I propose a model by which the proteoliposomes interact with these edges and fuse with the bilayer stacks, so forming a uniform layer on the SSM. Furthermore I characterized freestanding bilayers from a synthetic phospholipid with a phase transition at 41C and from a natural lipid extract of the archaebacterium Methanococcus jannaschii. The synthetic lipid is in the gel-phase at room temperature and changes to the fluid phase when heated to 50C. The bilayer of the lipid extract shows no phase transition when heated from room temperature to the growth temperature (~ 50C) of the archeon. Synaptic vesicles are the containers of neurotransmitter in nerve cells and the synapsins are a family of extrinsic membrane proteins, that are associated with them, and believed to control the synaptic vesicle cycle. I used AFM imaging and force spectroscopy together with dynamic light scattering to investigate the influence of synapsin I on synaptic vesicles. To this end I used native, untreated synaptic vesicles and compared them to synapsin-depleted synaptic vesicles. Synapsin-depleted vesicles were larger in size and showed a higher tendency to aggregate compared to native vesicles, although their mechanical properties were alike. I also measured the aggregation kinetics of synaptic vesicles induced by synapsin I and found that the addition of synapsin I promotes a rapid aggregation of synaptic vesicles. The data indicate that synapsin I affects the stability and the aggregation state of synaptic vesicles, and confirm the physiological role of synapsins in the assembly and regulation of synaptic vesicle pools within nerve cells.

This thesis was driven by the ambition to create suitable model systems that mimic complex processes in nature, like intramolecular transitions, such as unfolding and refolding of proteins, or intermolecular interactions between different cell compo-nents. Novel biophysical approaches were adopted by employing atomic force mi-croscopy (AFM) as the main measurement technique due to its broad diversity. Thus, high-resolution imaging, adhesion measurements, and single-molecule force distance experiments were performed on the verge of the instrumental capabilities. As first objective, the interaction between plasma membrane and cytoskeleton, me-diated by the linker protein ezrin, was pursued. Therefore, the adsorption process and the lateral organization of ezrin on PIP2 containing solid-supported membranes were characterized and quantified as a fundament for the establishment of a biomimetic model system. As second component of the model system, actin filaments were coated on functionalized colloidal probes attached on cantilevers, serving as sensor elements. The zealous endeavor of creating this complex biomimetic system was rewarded by successful investigation of the activation process of ezrin. As a result, it can be stated that ezrin is activated by solely binding to PIP2 without any further stimulating agents. Additional cofactors may stabilize and prolong the active conformation but are not essentially required for triggering ezrins transformation into an active conformation. In the second project, single-molecule force distance experiments were performed on bis-loop tetra-urea calix[4]arene-catenanes with different loading rates (increase in force per second). These macromolecules were specifically designed to investigate the rupture and rejoining mechanism of hydrogen bonds under external load. The entangled loops of capsule-like molecules locked the unbound state of intramolecular hydrogen bonds mechanically, rendering a rebinding observable on the experimental time scale. In conjunction with Molecular Dynamics simulations, a three-well potential of the bond rupture process was established and all kinetically relevant parameters of the experiments were determined by means of Monte Carlo simulations and stochastic modeling. In summary, it can be stated that atomic force microscopy is an invaluable tool to scrutinize relevant processes in nature, such as investigating activation mechanisms in proteins, as shown by analysis of the interaction between F-actin and ezrin, as well as exploring fundamental properties of single hydrogen bonds that are of paramount interest for the complete understanding of complex supramolecular structures.

Self-assembled molecular structures were investigated on insulating substrate surfaces using non-contact atomic force microscopy. Both, substrate preparation and molecule deposition, took place under ultra-high vacuum conditions. First, C60 molecules were investigated on the TiO2 (110) surface. This surface exhibits parallel running troughs at the nanometer scale, which strongly steer the assembly of the molecules. This is in contrast to the second investigated surface. The CaF2 (111) surface is atomically flat and the molecular assemblyrnwas observed to be far less affected by the surface. Basically different island structures were observed to what is typically know. Based on extensive experimental studies and theoretical considerations, a comprehensive picture of the processes responsible for the island formation of C60 molecules on this insulating surfaces was developed. The key process for the emergence of the observed novel island structures was made out to be the dewetting of molecules from the substrate. This new knowledge allows to further understand andrnexploit self-assembly techniques in structure fabrication on insulating substrate surfaces. To alter island formation and island structure, C60 molecules were codeposited with second molecule species (PTCDI and SubPc) on the CaF2 (111) surface. Depending on the order of deposition, quiet different structures were observed to arise. Thus, these are the first steps towards more complex functional arrangements consisting of two molecule species on insulating surfaces.

The concern of this work is to present the characterization of blue emitting GaN-based LED structures by means of Atomic Force Microscopy. Here we show a comparison among the samples with different dislocation densities, in order to understand how the dislocations can affect the surface morphology. First of all we have described the current state of art of the LEDs in the present market. Thereafterwards we have mentioned in detail about the growth technique of LED structures and the methodology of the characterization employed in our thesis. Finally, we have presented the details of the results obtained on our samples studied, followed by discussions and conclusions. L'obiettivo di questa tesi quello di presentare la caratterizzazione mediante Microscopia a Forza Atomica di strutture di LED a emissione di luce blu a base di nitruro di gallio (GaN). Viene presentato un confronto tra campioni con differente densit di dislocazioni, allo scopo di comprendere in che modo la presenza di dislocazioni influisce sulla morfologia della superficie. Innanzitutto, viene descritto il presente stato dell'arte dei LED. Successivamente, sono forniti i dettagli riguardanti la tecnica di crescita delle strutture dei LED e il metodo di caratterizzazione adottato. Infine, vengono mostrati e discussi i risultati ottenuti dallo studio dei campioni, seguiti dalle conclusioni.

La standardisation de la langue franaise a invitablement eu des consquences soit dans lattitude de dfense du franais, soit dans linquitude de ceux qui parlent une autre langue. Cette tude veut tablir une comparaison entre le rapport langue du pouvoir/langues non officielles et le rapport professeur/lve en milieu scolaire. La distinction entre langue officielle et dites langues minoritaires sera mis en relation avec la langue des jeunes, qui, lcole, se confrontent et entrent en collision avec la langue de leur professeur. En effet, les classes htrognes et linfluence de la langue maternelle auprs des lves, fils dimmigrs, exposent les enseignants toute une srie de difficults lies aux diffrences culturelles. Les dnonciations et les cas de violence dans les coles mettront laccent galement sur lopinion publique et sur le type de mesures mises en relief par les Institutions publiques. Laire gographique prise en considration inclut Paris et ses environs mme si quelques exemples denqutes menes dans dautres parties de la France et ltranger seront cits.

Light pseudoscalar bosons, such as the axion that was originally proposed as a solution of the strong CP problem, would cause a new spin-dependent short-range interaction. In this thesis, an experiment is presented to search for axion mediated short-range interaction between a nucleon and the spin of a polarized bound neutron. This interaction cause a shift in the precession frequency of nuclear spin-polarized gases in the presence of an unpolarized mass. To get rid of magnetic field drifts co-located, nuclear spin polarized 3He and 129Xe atoms were used. The free nuclear spin precession frequencies were measured in a homogeneous magnetic guiding field of about 350nT using LTc SQUID detectors. The whole setup was housed in a magnetically shielded room at the Physikalisch Technische Bundesanstalt (PTB) in Berlin. With this setup long nuclear spin-coherence times, respectively, transverse relaxation times of 5h for 129Xe and 53h for 3He could be achieved. The results of the last run in September 2010 are presented which give new upper limits on the scalar-pseudoscalar coupling of axion-like particles in the axion-mass window from 10^(-2) eV to 10^(-6) eV. The laboratory upper bounds were improved by up to 4 orders of magnitude.

Diese Arbeit stellt eine ausfhrliche Studie fundamentaler Eigenschaften der Kalzit CaCO3(10.4) und verwandter Mineraloberflchen dar, welche nicht nur durch die Verwendung von Nichtkontakt Rasterkraftmikroskopie, sondern hauptschlich durch die Messung von Kraftfeldern ermglicht wurde. Die absolute Oberflchenorientierung sowie der hierfr zugrundeliegende Prozess auf atomarer Skala konnten erfolgreich fr die Kalzit (10.4) Oberflche identifiziert werden.rnDie Adsorption chiraler Molekle auf Kalzit ist relevant im Bereich der Biomineralisation, was ein Verstndnis der Oberflchensymmetrie unumgnglich macht. Die Messung des Oberflchenkraftfeldes auf atomarer Ebene ist hierfr ein zentraler Aspekt. Eine solche Kraftkarte beleuchtet nicht nur die fr die Biomineralisation wichtige Wechselwirkung der Oberflche mit Moleklen, sondern enthlt auch die Mglichkeit, Prozesse auf atomarer Skala und damit Oberflcheneigenschaften zu identifizieren.rnDie Einfhrung eines hchst flexiblen Messprotokolls gewhrleistet die zuverlssige und kommerziell nicht erhltliche Messung des Oberflchenkraftfeldes. Die Konversion der rohen f Daten in die vertikale Kraft Fz ist jedoch kein trivialer Vorgang, insbesondere wenn Gltten der Daten in Frage kommt. Diese Arbeit beschreibt detailreich, wie Fz korrekt fr die experimentellen Bedingungen dieser Arbeit berechnet werden knnen. Weiterhin ist beschrieben, wie Lateralkrfte Fy und Dissipation erhalten wurden, um das volle Potential dieser Messmethode auszureizen.rnUm Prozesse auf atomarer Skala auf Oberflchen zu verstehen sind die kurzreichweitigen, chemischen Krfte Fz,SR von grter Wichtigkeit. Langreichweitige Beitrge mssen hierzu an Fz angefittet und davon abgezogen werden. Dies ist jedoch eine fehleranfllige Aufgabe, die in dieser Arbeit dadurch gemeistert werden konnte, dass drei unabhngige Kriterien gefunden wurden, die den Beginn zcut von Fz,SR bestimmen, was fr diese Aufgabe von zentraler Bedeutung ist. Eine ausfhrliche Fehleranalyse zeigt, dass als Kriterium die Abweichung der lateralen Krfte voneinander vertrauenswrdige Fz,SR liefert. Dies ist das erste Mal, dass in einer Studie ein Kriterium fr die Bestimmung von zcut gegeben werden konnte, vervollstndigt mit einer detailreichen Fehleranalyse.rnMit der Kenntniss von Fz,SR und Fy war es mglich, eine der fundamentalen Eigenschaften der CaCO3(10.4) Oberflche zu identifizieren: die absolute Oberflchenorientierung. Il collasso di diverse colonne, caratterizzate da danneggiamenti simili, quali ampie fessure fortemente inclinate ad entrambe le estremit dellelemento, lo schiacciamento del calcestruzzo e linstabilit dei ferri longitudinali, ha portato ad interrogarsi riguardo gli effetti dellinterazione tra lo sforzo normale, il taglio ed il momento flettente. Lo studio iniziato con una ricerca bibliografica che ha evidenziato una sostanziale carenza nella trattazione dellargomento. Il problema stato approcciato attraverso una ricerca di formule della scienza delle costruzioni, allo scopo di mettere in relazione lo sforzo assiale, il taglio ed il momento; la ricerca si principalmente concentrata sulla teoria di Mohr. In un primo momento stata considerata linterazione tra solo due componenti di sollecitazione: sforzo assiale e taglio. Lanalisi ha condotto alla costruzione di un dominio elastico di taglio e sforzo assiale che, confrontato con il dominio della Modified Compression Field Theory, trovata tramite ricerca bibliografica, ha permesso di concludere che i risultati sono assolutamente paragonabili. Lanalisi si poi orientata verso linterazione tra sforzo assiale, taglio e momento flettente. Imponendo due criteri di rottura, il raggiungimento della resistenza a trazione ed a compressione del calcestruzzo, inserendo le componenti di sollecitazione tramite le formule di Navier e Jourawsky, sono state definite due formule che mettono in relazione le tre azioni e che, implementate nel software Matlab, hanno permesso la costruzione di un dominio tridimensionale. In questo caso non stato possibile confrontare i risultati, non avendo la ricerca bibliografica mostrato niente di paragonabile. Lo studio si poi concentrato sullo sviluppo di una procedura che tenta di analizzare il comportamento di una sezione sottoposta a sforzo normale, taglio e momento: stato sviluppato un modello a fibre della sezione nel tentativo di condurre un calcolo non lineare, corrispondente ad una sequenza di analisi lineari. La procedura stata applicata a casi reali di crollo, confermando lavvenimento dei collassi.

Organic semiconductor technology has attracted considerable research interest in view of its great promise for large area, lightweight, and flexible electronics applications. Owing to their advantages in processing and unique physical properties, organic semiconductors can bring exciting new opportunities for broad-impact applications requiring large area coverage, mechanical flexibility, low-temperature processing, and low cost. In order to achieve highly flexible device architecture it is crucial to understand on a microscopic scale how mechanical deformation affects the electrical performance of organic thin film devices. Towards this aim, I established in this thesis the experimental technique of Kelvin Probe Force Microscopy (KPFM) as a tool to investigate the morphology and the surface potential of organic semiconducting thin films under mechanical strain. KPFM has been employed to investigate the strain response of two different Organic Thin Film Transistor with active layer made by 6,13-bis(triisopropylsilylethynyl)-pentacene (TIPS-Pentacene), and Poly(3-hexylthiophene-2,5-diyl) (P3HT). The results show that this technique allows to investigate on a microscopic scale failure of flexible TFT with this kind of materials during bending. I find that the abrupt reduction of TIPS-pentacene device performance at critical bending radii is related to the formation of nano-cracks in the microcrystal morphology, easily identified due to the abrupt variation in surface potential caused by local increase in resistance. Numerical simulation of the bending mechanics of the transistor structure further identifies the mechanical strain exerted on the TIPS-pentacene micro-crystals as the fundamental origin of fracture. Instead for P3HT based transistors no significant reduction in electrical performance is observed during bending. This finding is attributed to the amorphous nature of the polymer giving rise to an elastic response without the occurrence of crack formation.

This study deals with the determination of the retentive force between primary and secondary telescopic crowns under clinical conditions. Forty-three combined fixed-removable prostheses with a total of 140 double crowns were used for retention force measurement of the telescopic crowns prior to cementation. The crowns had a preparation of 1-2. A specifically designed measuring device was used. The retentive forces were measured with and without lubrication by a saliva substitute. The measured values were analyzed according to the type of tooth (incisors, canines, premolars, and molars). Additionally, a comparison between lubricated and unlubricated telescopic crowns was done. As maximum retention force value 29.98N was recorded with a telescopic crown on a molar, while the minimum of 0.08N was found with a specimen on a canine. The median value of retention force of all telescopic crowns reached 1.93N with an interquartile distance of 4.35N. No statistically significant difference between lubricated and unlubricated specimens was found. The results indicate that retention force values of telescopic crowns, measured in clinical practice, are often much lower than those cited in the literature. The measurements also show a wide range. Whether this proves to be a problem for the patient's quality of life or not can however only be established by a comparison of the presented results with a follow-up study involving measurement of intraoral retention and determination by e.g. oral health impact profile.

The delicate anatomy of the ear require surgeons to use great care when operating on its internal structures. One example for such an intervention is the stapedectomy, where a small crook shaped piston is placed in the oval window of the cochlea and connected to the incus through crimping thus bypassing the diseased stapes. Performing the crimp process with the correct force is necessary since loose crimps poorly transmit sound whereas tight crimps will eventually result in necrosis of the incus. Clinically, demand is high to reproducibly conduct the crimp process through a precise force measurement. For this reason, we have developed a fiber Bragg grating (FBG) integrated microforceps for use in such interventions. This device was calibrated, and tested in cadaver preparations. With this instrument we were able to measure for the first time forces involved in crimping a stapes prosthesis to the incus. We also discuss a method of attaching and actuating such forceps in conjunction with a robot currently under development in our group. Each component of this system can be used separately or combined to improve surgical accuracy, confidence and outcome.
