In this study, fluid bed granulation was applied to improve the dissolution of nimodipine and spironolactone, two very poorly water-soluble drugs. Granules were obtained with different amounts of sodium dodecyl sulfate and croscarmellose sodium and then compressed into tablets. The dissolution behavior of the tablets was studied by comparing their dissolution profiles and dissolution efficiency with those obtained from physical mixtures of the drug and excipients subjected to similar conditions. Statistical analysis of the results demonstrated that the fluid bed granulation process improves the dissolution efficiency of both nimodipine and spironolactone tablets. The addition of either the surfactant or the disintegrant employed in the study proved to have a lower impact on this improvement in dissolution than the fluid bed granulation process.

[EN]The age and growth of the sand sole Pegusa lascaris from the Canarian Archipelago were studied from 2107 fish collected between January 2005 and December 2007. To find an appropriate method for age determination, sagittal otoliths were observed by surface-reading and frontal section and the results were compared. The two methods did not differ significantly in estimated age but the surface-reading method is superior in terms of cost and time efficiency. The sand sole has a moderate life span, with ages up to 10 years recorded. Individuals grow quickly in their first two years, attaining approximately 48% of their maximum standard length; after the second year, their growth rate drops rapidly as energy is diverted to reproduction. Males and females show dimorphism in growth, with females reaching a slightly greater length and age than males. Von Bertalanffy, seasonalized von Bertalanfy, Gompertz, and Schnute growth models were fitted to length-at-age data. Akaike weights for the seasonalized von Bertalanffy growth model indicated that the probability of choosing the correct model from the group of models used was >0.999 for males and females. The seasonalized von Bertalanffy growth parameters estimated were: L? = 309 mm standard length, k = 0.166 yr?1, t0 = ?1.88 yr, C = 0.347, and ts = 0.578 for males; and L? = 318 mm standard length, k = 0.164 yr?1, t0 = ?1.653 yr, C = 0.820, and ts = 0.691 for females. Fish standard length and otolith radius are closely correlated (R2 = 0.902). The relation between standard length and otolith radius is described by a power function (a = 85.11, v = 0.906)

Habitat structure is known to influence the abundance of fishes on temperate reefs. Biotic interactions play a major role in determining the distribution and abundance of species. The significance of these forces in affecting the abundance of fishes may hinge on the presence of organisms that either create or alter habitat. On temperate reefs, for example, macroalgae are considered autogenic ecosystem engineers because they control resource availability to other species through their physical structure and provide much of the structure used by fish. On both coral and temperate reefs, small cryptic reef fishes may comprise up to half of the fish numbers and constitute a diverse community containing many specialized species. Small cryptic fishes (<100 mm total length) may be responsible for the passage of 57% of the energy flow and constitute ca. 35% of the overall reef fish biomass on coral reefs. These benthic fish exploit restricted habitats where food and shelter are obtained in, or in relation to, conditions of substrate complexity and/or restricted living space. A range of mechanisms has been proposed to account for the diversity and the abundance of small fishes: (1) lifehistory strategies that promote short generation times, (2) habitat associations and behaviour that reduce predation and (3) resource partitioning that allows small species to coexist with larger competitors. Despite their abundance and potential importance within reef systems, little is known of the community ecology of cryptic fishes. Specifically on habitat associations many theories suggested a not clear direction on this subject. My research contributes to the development of marine fish ecology by addressing the effects of habitat characteristics upon distribution of cryptobenthic fish assemblages. My focus was on the important shallow, coastal ecosystems that often serve as nursery habitat for many fish and where different type of habitat is likely to both play important roles in organism distribution and survival. My research included three related studies: (1) identification of structuring forces on cryptic fish assemblages, such as physical and biological forcing; (2) macroalgae as potential tools for cryptic fish and identification of different habitat feature that could explain cryptic fish assemblages distribution; (3) canopy formers loss: consequences on cryptic fish and relationship with benthos modifications. I found that: (1) cryptic fish assemblages differ between landward and seaward sides of coastal breakwaters in Adriatic Sea. These differences are explained by 50% of the habitat characteristics on two sides, mainly due to presence of the Codium fragile, sand and oyster assemblages. Microhabitat structure influence cryptic fish assemblages. (2) Different habitat support different cryptic fish assemblages. High heterogeneity on benthic assemblages reflect different fish assemblages. Biogenic components that explain different and diverse cryptic fish assemblages are: anemonia bed, mussel bed, macroalgal stands and Cystoseira barbata, as canopy formers. (3) Canopy forming loss is not relevant in structuring directly cryptic fish assemblages. A removal of canopy forming algae did not affect the structure of cryptic fish assemblages. Canopy formers algae on Conero cliff, does not seem to act as structuring force, probably due to its regressive status. In conclusion, cryptic fish have been shown to have species-specific associations with habitat features relating to the biological and non biological components afforded by fish. Canopy formers algae do not explain cryptic fish assemblages distribution and the results of this study and information from the literature (both from the Mediterranean Sea and elsewhere) show that there are no univocal responses of fish assemblages. Further exanimations on an non regressive status of Cystoseira canopy habitat are needed to define and evaluate the relationship between canopy formers and fish on Mediterranean sea.

Chlorinated Aliphatic Hydrocarbons (CAHs) are widespread wastewater and groundwater contaminants and represent a real danger for human health and environment. This research is related to the biodegradation technologies to treat chlorinated hydrocarbons. In particular the study of this thesis is focused on chloroform cometabolism by a butane-grown aerobic pure culture (Rhodococcus aetherovorans BCP1) in continuous-flow biofilm reactors, which are used for in-situ and on-site treatments. The work was divided in two parts: in the first one an experimental study has been conducted in two packed-bed reactors (PBRs) for a period of 370 days; in the second one a fluid dynamics and kinetic model has been developed in order to simulate the experimental data concerning a previous study made in a 2-m continuous-flow sand-filled reactor. The goals of the first study were to obtain preliminary information on the feasibility of chloroform biodegradation by BCP1 under attached-cell conditions and to evaluate the applicability of the pulsed injection of growth substrate and oxygen to biofilm reactors. The pulsed feeding represents a tool to control the clogging and to ensure a long bioreactive zone. The operational conditions implemented in the PBRs allowed the attainment of a 4-fold increase of the ratio of chloroform degraded to substrate consumed, in comparison with the phase of continuous substrate supply. The second study was aimed at identifying guidelines for optimizing the oxygen/substrate supply schedule, developing a reliable model of chloroform cometabolism in porous media. The tested model led to a suitable interpretation of the experimental data as long as the ratio of CF degraded to butane consumed was ≤ 0.27 mgchloroform /mgbutane. A long-term simulation of the best-performing schedule of pulsed oxygen/substrate supply indicated the attainment of a steady state condition characterized by unsatisfactory bioremediation performances, evidencing the need for a further optimization of the pulsed injection technique.

The two Mars Exploration Rovers (MER), Spirit and Opportunity, landed on the Martian surface in January 2004 and have since collected a wealth of information about their landing sites. As part of their payload, the miniaturised Mössbauer spectrometer MIMOS II contributes to the success of the mission by identifying Iron-bearing minerals and by determining Iron oxidation states in them. The basis of this work is the data set obtained at Opportunity’s landing site at Meridiani Planum. A portion of this data set is evaluated with different methods, with the aim to thoroughly characterize lithologic components at Meridiani Planum and possible relations between them.rnMIMOS II is able to measure Mössbauer spectra at different energies simultaneously, bearing information from different sampling depths of the investigated target. The ability of depth-selective Mössbauer spectroscopy to characterize weathered surface layers is illustrated through its application to two suitable rock targets that were investigated on Mars. In both cases, an enhanced concentration of Iron oxides at the rock surface was detected, pointing to a low degree of aqueous alteration. rnThe mineral hematite (α-Fe2O3) is present in the matrix of outcrop rocks and in spherules weathering from the outcrop. Simultaneous fitting of Mössbauer spectra was applied to data sets obtained on both target types to characterize the hematite component in detail. This approach reveals that two hematite populations are present, both in the outcrop matrix as well as in spherules. The hematite component with a comparably high degree of crystallinity and/or chemical purity is present in the outcrop matrix. The investigation of hematite at Meridiani Planum has shown that simultaneous fitting is a suitable and useful method to evaluate a large, correlated set of Mössbauer spectra.rnOpportunity encountered loose, cm-sized rocks along its traverse. Based on their composition and texture, these “cobbles” can be divided into three different groups. Outcrop fragments are impact-derived ejecta from local outcrop rocks. Cobbles of meteoritic origin contain the minerals kamacite (Fe,Ni) and troilite (FeS) and exhibit high Ni contents. Melt-bearing impact breccias bear similarities to local outcrop rocks and basaltic soil, with a phase composition and texture consistent with a formation scenario involving partial melting and inclusion of small, bright outcrop clasts. rnIron meteorites on the Martian surface experience weathering through the presence of even trace amounts of water due to their metallic nature. Opportunity encountered and investigated four Iron meteorites, which exhibit evidence for physical and chemical weathering. Discontinuous coatings contain Iron oxides, pointing to the influence of limited amounts of water. rnA terrestrial analogue site for Meridiani Planum is the Rio Tinto basin in south-west Spain. With its deposits of sulfate- and iron-oxide-bearing minerals, the region provides an adequate test bed for instrumentation for future Mars missions. In-situ investigations at Rio Tinto were carried out with a special focus on the combined use of Mössbauer spectroscopy with MIMOS II and Raman spectroscopy with a field-portable instrument. The results demonstrate that the two instruments provide complementary information about investigated samples.

The aim of this thesis, included within the THESEUS project, is the development of a mathematical model 2DV two-phase, based on the existing code IH-2VOF developed by the University of Cantabria, able to represent together the overtopping phenomenon and the sediment transport. Several numerical simulations were carried out in order to analyze the flow characteristics on a dike crest. The results show that the seaward/landward slope does not affect the evolution of the flow depth and velocity over the dike crest whereas the most important parameter is the relative submergence. Wave heights decrease and flow velocities increase while waves travel over the crest. In particular, by increasing the submergence, the wave height decay and the increase of the velocity are less marked. Besides, an appropriate curve able to fit the variation of the wave height/velocity over the dike crest were found. Both for the wave height and for the wave velocity different fitting coefficients were determined on the basis of the submergence and of the significant wave height. An equation describing the trend of the dimensionless coefficient c_h for the wave height was derived. These conclusions could be taken into consideration for the design criteria and the upgrade of the structures. In the second part of the thesis, new equations for the representation of the sediment transport in the IH-2VOF model were introduced in order to represent beach erosion while waves run-up and overtop the sea banks during storms. The new model allows to calculate sediment fluxes in the water column together with the sediment concentration. Moreover it is possible to model the bed profile evolution. Different tests were performed under low-intensity regular waves with an homogeneous layer of sand on the bottom of a channel in order to analyze the erosion-deposition patterns and verify the model results.

La tesi contiene uno studio sperimentale sul comportamento di una sabbia limosa del sottosuolo della laguna veneta e propone un'interpretazione dei risultati sperimentali ottenuti alla luce dei presupposti teorici di un approccio costitutivo avanzato noto come "Plasticità Generalizzata". Il programma sperimentale è consistito nella realizzazione di prove edometriche e prove triassiali su campioni di sabbia provenienti dal sito di Treporti, situato in prossimità della bocca di Lido. La risposta sperimentale, in termini di modulo volumetrico, è stata messa a confronto con i risultati di alcuni studi di letteratura, con particolare riferimento a quelli condotti da Jefferies & Been (2000). La disponibilità di prove di compressione edometrica realizzate nella cella K0 e la conseguente possibilità di valutare il coefficiente di spinta a riposo ha permesso di interpretare le prove in termini di tensione media efficace p' e di verificare l'applicabilità al caso in esame degli approcci di letteratura disponibili, spesso sviluppati a partire da prove di compressione isotropa effettuate in cella triassiale. Il comportamento tenso-deformativo osservato è stato successivamente simulato con un modello costitutivo per sabbie sviluppato nell'ambito della Plasticità Generalizzata. In particolare sono state utilizzate tre diverse formulazioni, che costituiscono un avanzamento dell'iniziale modello costitutivo proposto da Pastor, Zienkiewicz e Chan (1990), basate sull'uso di un parametro di stato del materiale definito rispetto alle condizioni di Stato Critico. Dal confronto tra previsioni del modello e risposta sperimentale è stato possibile individuare la formulazione che meglio simula il comportamento meccanico osservato sia in compressione edometrica sia in prove di taglio ed è stato proposto un set di parametri costitutivi ritenuti rappresentativi del terreno studiato.

The effect of zeolite amendment for enhanced sorption capacity on the consolidation behavior and hydraulic conductivity, k, of a typical soil-bentonite (SB) backfill for vertical cutoff walls was evaluated via laboratory testing. The consolidation behavior and k of test specimens containing fine sand, 5.8 % (dry wt.) sodium bentonite, and 0, 2, 5, or 10 % (dry wt.) of one of three types of zeolite (clinoptilolite, chabazite-lower bed, or chabazite-upper bed) were measured using fixed-ring oedometers, and k also was measured on separate specimens using a flexible-wall permeameter. The results indicated that addition of a zeolite had little impact on either the consolidation behavior or the k of the backfill, regardless of the amount or type of zeolite. For example, the compression index, Cc, for the unamended backfill specimen was 0.24, whereas values of Cc for the zeolite amended specimens were in the range 0.19 ≤ Cc ≤ 0.23. Similarly, the k for the unamended specimen based on flexible-wall tests was 2.4 x 10-10 m/s, whereas values of k for zeolite amended specimens were in the range 1.2 x 10-10 ≤ k ≤ 3.9 x 10-10 m/s. The results of the study suggest that enhancing the sorption capacity of typical SB backfills via zeolite amendment is not likely to have a significant effect on the consolidation behavior or k of the backfill, provided that the amount of zeolite added is small (≤ 10 %).
