Zusammenfassung: Freizeitaktivitäten wie Mountainbiking, Sportklettern, Drachen- und Gleitschirmfliegen oder Windsur-fen sind relativ neue Erscheinungen der letzten fünfzehn bis fünfundzwanzig Jahre. Ihre Entwicklung und Ausdifferenzierung beschränkt sich vor allem auf Gesellschaften westlich-industrieller Prägung, deren Grundlage ein funktional differenziertes Gesellschaftssystem ist. Mit Hilfe des Beobachtungsin-strumentes der neueren soziologischen Systemtheorie wird zunächst das Aufkommen und die immer fortschreitende Ausdifferenzierung von Trend- und Natursportarten analysiert und gedeutet. Es zeigt sich, daß diese neuen Freizeitaktivitäten über den Zugriff auf die eigene Körperlichkeit in hohem Ma-ße zur Identitätsfindung der Individuen solcher Gesellschaftssysteme beitragen.Aus geographischer Sicht stellt sich die Frage nach der Bedeutung dieser Entwicklung für die Land-schaft. Dabei wird Landschaft explizit nicht als etwas verstanden, daß 'an sich' existiert, sondern als etwas, das 'erlebt' oder 'begriffen', also immer wieder aufs Neue im Auge des Betrachters konstru-iert wird. Für die Sporttreibenden von Trend- und Natursportarten ist der Blick auf die Welt durch ihre Sportart strukturiert. Jedenfalls immer dann, wenn es darum geht, eine Sportstätte für ihre jeweilige Sportausübung auszuwählen. Die Naturlandschaft (oder auch Kulturlandschaft) wird damit zur 'Sportlandschaft'. Der Begriff der 'Landschaft' erscheint dann im Sinne Werlens 'alltäglichen Geo-graphie-Machens' als eine permanent neu zu erstellende und damit sich auch ständig verändernde Perspektive auf die Objektwelt. So kann sich im Auge der Sporttreibenden von Trend- und Natur-sportarten jedes Objekt und jede Landschaft, wenn man so will: die Welt, als ein einziger großer Sportplatz darstellen.Wenn also alles potentiell eine Sportstätte sein kann, stellt sich die Frage, wie es zur Herausbildung sogenannter Top Spots kommt, also zu jenen Sportstätten, die innerhalb der jeweiligen Sportart von herausragender Bedeutung sind - sozusagen die oberste Sprosse der Karriereleiter erklommen haben - und an der jeder gewesen sein muß, der innerhalb der sportartspezifischen Szene etwas gelten will. Dieser Frage wird anhand eines Fallbeispiels nachgegangen. Als Grundlage wurde ein Untersu-chungsgebiet gewählt, das sich bereits in einem fortgeschrittenen Stadium der Entwicklung zu einem Top Spot befindet - eine Landschaft also, die bereits eine 'Karriere als Sportlandschaft' aufzuweisen hat. Die peripher gelegene US-amerikanische Kleinstadt Moab in Südost-Utah (ca. 5.000 Einwohner) hat sich seit Anfang der 1990er Jahre zu einem internationalen Szene-Treffpunkt für Mountainbiking entwickelt. Zu dem wichtigsten Mountainbike-Trail (dem Slickrock Bike Trail) kommen pro Jahr al-lein mehr als 200.000 Mountainbike-TouristInnen. Neben dem Mountainbiking spielt River Rafting eine bedeutende Rolle im Tourismus und es hat sich dort in den letzten Jahren außerdem eine kleine Sportkletter-Szene etabliert, die in den nächsten Jahren sicherlich noch an Bedeutung zunehmen wird.

SummaryThe present thesis deals with the choosing of habitats and species structures of invertebrate populations in urban areas. The main emphasis is focused on the group of saltatoria. Samplings by stationary traps and also by net-catching were carried out from 1993 to 1997. The survey extended over the whole city area of Mainz, Germany, as well as several spots in the area of Frankfurt upon Main. Shifting calcerous sands received special consideration. For a better distinction of the surveying area, three different zones were established, 'Relicts of natural landscape', 'Traditional worked rural landscape' and 'Urban environment'. Partly significant differences in species and individual abundance between the zones showed the influence of the different cultivation on the fauna. To investigate population size and -structure as well as migration and habitat preference - especially in urban areas - capture-recapture experiments were performed on the two species Oedipoda caerulescens L. and Sphingonotus caerulans L. in 1998. Large secondary habitats, such as train stations, are getting more and more important for the survival of the highly endangered S. caerulans. Another centre of attention was the structure of invertebrate populations on class level. The results of the investigation show, that the importance of urban environment for the conservation of species and also nature has been highly underestimated so far.ZusammenfassungDie vorliegende Arbeit beschäftigt sich mit der Habitatwahl und Artenzusammensetzung von Arthropodenpopulationen im besiedelten Bereich. Der Schwerpunkt lag auf der Gruppe der Saltatoria. Über mehrere Jahre (1993-1996) wurden Fänge mit stationären Fallen sowie Kescherfänge durchgeführt. Das Untersuchungsgebiet umfaßte in erster Linie das Stadtgebiet von Mainz, darüber hinaus wurden einzelne Standorte in Frankfurt/M. und Umgebung untersucht. Ein Schwerpunkt der Untersuchungen lag auf Kalkflugsanddünen, besonders Restflächen im besiedelten Bereich. Sekundäre Sandstandorte oder vergleichbare anthropogen entstandene Böden wurden ebenfalls beprobt Das Stadtgebiet wurde aufgrund besserer Unterscheidung in drei Zonen aufgeteilt, die 'Naturlandschaftsrelikte', die 'Zone der Alten Kulturlandschaft' und die 'Urbane Landschaft'. Größtenteils signifikante Unterschiede in der Arten- und Individuenanzahl zwischen den Zonen zeigen den bedeutenden Einfluß der unterschiedlichen Bearbeitung und Kultivierung auf die Fauna. An den beiden Spezies Oedipoda caerulescens L. und Sphingonotus caerulans L. wurden Fang-Wiederfangexperimente zur Abschätzung der Populationsgröße, -struktur, Habitatwahl sowie Migration durchgeführt. Besonders für die stark gefährdete S. caerulans stellte sich die enorme Bedeutung von großflächigen Sekundärhabitaten im besiedelten Bereich, wie sie Bahnhöfe darstellen, heraus. Der zweite Schwerpunkt lag auf der Struktur von Invertebratenpopulationen im besiedelten Bereich. Sie wurden auf Ordnungsniveau erfaßt. Die Ergebnisse der Untersuchungen zeigen, daß die Bedeutung des besiedelten Bereiches für den Arten- und Biotopschutz bisher weit unterschätzt wurde.

The spatio-temporal variations in diversity and abundance of deep-sea macrofaunal assemblages (excluding meiofaunal taxa, as Nematoda, Copepoda and Ostracoda) from the Blanes Canyon (BC) and adjacent open slope are described. The Catalan Sea basin is characterized by the presence of numerous submarine canyons, which are globally acknowledged as biodiversity hot-spots, due to their disturbance regime and incremented conveying of organic matter. This area is subjected to local deep-sea fisheries activities, and to recurrent cold water cascading events from the shelf. The upper canyon (~900 m), middle slope (~1200 m) and lower slope (~1500 m) habitats were investigated during three different months (October 2008, May 2009 and September 2009). A total of 624 specimens belonging to 16 different taxa were found into 67 analyzed samples, which had been collected from the two study areas. Of these, Polychaeta, Mollusca and Crustacea were always the most abundant groups. As expected, the patterns of species diversity and evenness were different in time and space. Both in BC and open slope, taxa diversity and abundance are higher in the shallowest depth and lowest at -1500 m depth. This is probably due to different trophic regimes at these depths. The abundance of filter-feeders is higher inside BC than in the adjacent open slope, which is also related with an increment of predator polychaetes. Surface deposit-feeders are more abundant in the open slope than in BC, along with a decrement of filter-feeders and their predators. Probably these differences are due to higher quantities of suspended organic matter reaching the canyon. The multivariate analyses conducted on major taxa point out major differences effective taxa richness between depths and stations. In September 2009 the analyzed communities double their abundances, with a corresponding increase in richness of taxa. This could be related to a mobilizing event, like the release of accumulated food-supply in a nepheloid layer associated to the arrival of autumn. The highest abundance in BC is detected in the shallowest depth and in late summer (September), probably due to higher food availability caused by stronger flood events coming from Tordera River. The effects of such events seemed to involve adjacent open slope too. The nMDS conducted on major taxa abundance shows a slight temporal difference between the three campaigns samples, with a clear clustering between samples of Sept 09. All depth and all months were dominated by Polychaeta, which have been identified to family level and submitted to further analysis. Family richness have clearly minimum at the -1200 m depth of BC, highlighting the presence of a general impact affecting the populations in the middle slope. Three different matrices have been created, each with a different taxonomic level (All Taxa “AT”, Phylum Level “PL” and Polychaeta Families “PF”). Multivariate analysis (MDS, SIMPER) conducted on PL matrix showed a clear spatial differences between stations (BC and open slope) and depths. MDSs conducted on other two matrices (AT and PF) showed similar patterns, but different from PL analysis. A 2 nd stage analysis have been conducted to understand differences between different taxonomic levels, and PL level has been chosen as the most representative of variation. The faunal differences observed were explained by depth, station and season. My research PhD work is focused on the Electrochemically Generated Luminescence (ECL) investigation of several different homogeneous and heterogeneous systems. ECL is a redox induced emission, a process whereby species, generated at electrodes, undergo a high-energy electron transfer reaction to form excited states that emit light. Since its first application, the ECL technique has become a very powerful analytical tool and has widely been used in biosensor transduction. ECL presents an intrinsically low noise and high sensitivity; moreover, the electrochemical generation of the excited state prevents scattering of the light source: for all these characteristics, it is an elective technique for ultrasensitive immunoassay detection. The majority of ECL systems involve species in solution where the emission occurs in the diffusion layer near to the electrode surface. However, over the past few years, an intense research has been focused on the ECL generated from species constrained on the electrode surface. The aim of my work is to study the behavior of ECL-generating molecular systems upon the progressive increase of their spatial constraints, that is, passing from isolated species in solution, to fluorophores embedded within a polymeric film and, finally, to patterned surfaces bearing “one-dimensional” emitting spots. In order to describe these trends, I use different “dimensions” to indicate the different classes of compounds. My thesis was mostly developed in the electrochemistry group of Bologna with the supervision of Prof Francesco Paolucci and Dr Massimo Marcaccio. With their help and also thanks to their long experience in the molecular and supramolecular ECL fields and in the surface investigations using scanning probe microscopy techniques, I was able to obtain the results herein described. Moreover, during my research work, I have established a new collaboration with the group of Nanobiotechnology of Prof. Robert Forster (Dublin City University) where I spent a research period. Prof. Forster has a broad experience in the biomedical field, especially he focuses his research on film surfaces biosensor based on the ECL transduction. This thesis can be divided into three sections described as follows: (i) in the fist section, homogeneous molecular and supramolecular ECL-active systems, either organic or inorganic species (i.e., corannulene, dendrimers and iridium metal complex), are described. Driving force for this kind of studies includes the search for new luminophores that display on one hand higher ECL efficiencies and on the other simple mechanisms for modulating intensity and energy of their emission in view of their effective use in bioconjugation applications. (ii) in the second section, the investigation of some heterogeneous ECL systems is reported. Redox polymers comprising inorganic luminophores were described. In such a context, a new conducting platform, based on carbon nanotubes, was developed aimed to accomplish both the binding of a biological molecule and its electronic wiring to the electrode. This is an essential step for the ECL application in the field of biosensors. (iii) in the third section, different patterns were produced on the electrode surface using a Scanning Electrochemical Microscopy. I developed a new methods for locally functionalizing an inert surface and reacting this surface with a luminescent probe. In this way, I successfully obtained a locally ECL active platform for multi-array application.

Im Rahmen meiner Arbeit wurden erstmals die Intermediärfilament-Proteine (IF-Proteine) des Sibirischen Störs Acipenser baeri (Strahlenflosser, Knorpelganoid) kloniert und sequenziert. Aus einer cDNA-Bank konnten die Sequenzen von 13 IF-Proteine gewonnen werden. Von insgesamt zehn Keratinen codieren sieben für Typ I-Keratine und drei für Typ II. Zusätzlich konnten noch Desmin, Vimentin und ein Lamin identifiziert werden. Je einem Typ I- (K13) und einem Typ-II-Keratin (K2) fehlen wenige Aminosäuren in der Head-Domäne.Cytoskelett-Präparationen aus Epidermis, Mitteldarm, Magen und Kieme wurden mittels 2D-PAGE aufgetrennt. Durch Einsatz des CKBB-Test und Immunoblots wurden die verschiedenen Typ I und II-Keratine sowie Desmin und Vimentin identifiziert. Die gewebsspezifische Expression der Keratine ermöglichte zumeist ihre Einteilung in 'E' (epidermal) und 'S' ('simple epithelial').Die MALDI-MS-Analyse einer 2D-PAGE-Koelektrophorese von Seitenflosse und Mitteldarm zeigte, daß die 34 vorhandenen Proteinflecke auf nur 13 verschiedene IF-Proteine zurückgehen. Neun dieser Flecke konnten Sequenzen zugewiesen werden. Zusammen mit den verbleibenden vier Proteinflecken ergeben sich für den Stör nunmehr insgesamt 17 bekannte IF-Proteine. Von drei biochemisch identifizierten IS-Keratinen kommt eines nur im Mitteldarm vor und nur einem konnte eine Sequenz zugeordnet werden (K18). Dem einzigen Typ IIS-Keratin konnte keine Sequenz zugeordnet werden, wahrscheinlich handelt es sich um dabei um das K8-Orthologe. Jedem der fünf Typ IE-Proteine konnte eine Sequenz zugeordnet werden (K10 bis K14), ebenso wie dem einzigen identifizierten Typ IIE-Keratin (K2). Von den Typ III-Proteinen wurden Desmin und Vimentin ihren Proteinflecken zugeordnet. Die nicht zugeordnete Sequenz aba-k1 codiert möglicherweise für ein IIE-Keratin, während aba-k15 vermutlich die Sequenz für ein IE-Keratin enthält. Bei den Proteinflecken, denen eine Sequenz zugeordnet werden konnten, kann für Aba-K2 die Zugehörigkeit zum IIE-Typ angenommen werden, während es sich bei Aba-K10 wahrscheinlich um ein IE-Keratin handelt.Durch Datenbankvergleiche und molekulare Stammbäume konnte die Zugehörigkeit der identifizierten Lamin-Sequenz zum B3-Subtyp der Vertebraten gezeigt werden.Die Daten der Biochemie und indirekten Immunfluoreszenzmikroskopie zeigen, daß Keratine in Epithelien und Vimentin in mesenchymalen Geweben vorkommen. Es existieren starke Hinweise, daß im letzten Gewebetyp Keratine auch koexprimiert werden. Desmin kommt in großen Mengen im Magen und im Mitteldarm vor und stellt dort das prominenteste Protein.Mit den gewonnenen Sequenzdaten wurden molekulare Stammbäume und Sequenzidentitäten berechnet. Die daraus resultierenden Konsequenzen für die Verwandtschaftsverhältnisse der verschiedenen IF-Proteine sowie der Wirbeltiere werden diskutiert.

Electronic applications are nowadays converging under the umbrella of the cloud computing vision. The future ecosystem of information and communication technology is going to integrate clouds of portable clients and embedded devices exchanging information, through the internet layer, with processing clusters of servers, data-centers and high performance computing systems. Even thus the whole society is waiting to embrace this revolution, there is a backside of the story. Portable devices require battery to work far from the power plugs and their storage capacity does not scale as the increasing power requirement does. At the other end processing clusters, such as data-centers and server farms, are build upon the integration of thousands multiprocessors. For each of them during the last decade the technology scaling has produced a dramatic increase in power density with significant spatial and temporal variability. This leads to power and temperature hot-spots, which may cause non-uniform ageing and accelerated chip failure. Nonetheless all the heat removed from the silicon translates in high cooling costs. Moreover trend in ICT carbon footprint shows that run-time power consumption of the all spectrum of devices accounts for a significant slice of entire world carbon emissions. This thesis work embrace the full ICT ecosystem and dynamic power consumption concerns by describing a set of new and promising system levels resource management techniques to reduce the power consumption and related issues for two corner cases: Mobile Devices and High Performance Computing.

One of the key for the understanding of an orogenic belt is the characterization of the terranes involved and the identification of the suture(s) separating crustal blocks: these are essential information for large-scale paleo-reconstructions. In addition, the structural relationships between the terranes involved in the collisional processes and the eventual UHP relicts may provide first order inputs to exhumation models of subducted rocks. The structure of the Rhodope Massif (northern Greece and southern Bulgaria) results from the stacking of high-grade nappes during a continental collision, which age is comprised between Latest-Jurassic and Early-Cenozoic. UHP and HP relicts, associated with oceanic and ultramafic material, suggest the presence of a dismembered suture zone within the massif. The location of this suture remains unclear; furthermore, up to now, the UHP and eclogitic localities represent isolated spots and no synthesis on their structural position within the massif has been proposed. The first aim of this work is to define the relationships between HP-UHP relicts, crustal blocks, shear zones and amphibolitic material. To achieve this objective, we characterized the accreted blocks in terms of protoliths ages of the orthogneisses mainly along two cross sections on the Greek part of the belt. Geochemical affinities of meta-igneous rocks served as a complementary tool for terrane characterization and geodynamic interpretation. Single-zircon Pb-Pb evaporation and zircon U-Pb SHRIMP dating of orthogneiss protoliths define two groups of intrusion-ages: Permo-Carboniferous and Late Jurassic-Early Cretaceous. Structurally, these two groups correspond to distinct units: the Late Jurassic gneissic complex overthrusts the one bearing the Permo-Carboniferous orthogneisses. Mylonites, eclogites, amphibolites of oceanic affinities, and UHP micaschists, mark a “melange” zone, intensively sheared towards the SW, which separates the two units. Thus, we interpret them as two distinct terranes, the Rhodope and Thracia terranes, separated by the Nestos suture. The correlation of our findings in northern Greece to the Bulgarian part of the Massif suggests a northern rooting of the Nestos Suture. This configuration results of the closure of a marginal oceanic basin of the Tethys system by a north-directed subduction. This interpretation is supported by the geochemical affinities of the orthogneisses: the Late-Jurassic igneous rocks formed by subduction-related magmatism, pprobably the same north-directed subduction that gave rise to the UHP metamorphism of the metasediments of the “melange” zone. It is noteworthy that the UHP-HP relicts seem to be restricted to the contact between the two terranes suggesting that the UHP relicts are exhumed only within the suture zone. Furthermore, the singularity of the suture suggests that the Late-Jurassic subduction explains the occurrence of UHP and eclogite relicts in the Central Rhodope despite the large age range previously attributed the UHP and/or HP stage.

Lung ultrasound use is increasing in respiratory medicine thanks to its development in the latest years. Actually it allows to study diseases of the chest wall (traumas, infections, neoplasms), diaphragm (paralysis, ipokinesis), pleura (effusions, pneumothorax, thickenings, neoplasms) and lung parenchyma (consolidations, interstitial syndromes, peripheral lesions). One of the most useful application of chest ultrasound is the evaluation of effusions. However, no standardized approach for ultrasound-guided thoracenthesis is available. Our study showed that our usual ultrasonographic landmark (“V-point”) could be a standard site to perform thoracenthesis: in 45 thoracenthesis no pneumothorax occurred, drainage was always successful at first attempt. Values of maximum thickness at V-point and drained fluid volume showed a significative correlation. Proteins concentration of ultrasound patterns of effusions (anechoic, ipoechoic, moving echoic spots, dense moving spots, hyperechoic) were compared to those of the macroscopic features of fluids showing connection between light-yellow fluid and echoic moving spots pattern and between ipoechoic/dense moving spots and cloudy-yellow/serum-haematic fluids. These observations suggest that ultrasound could predict chemical-physical features of effusions. Lung ultrasound provides useful information about many disease of the lung, but actually there is not useful in obstructive bronchial diseases. Analysing diaphragmatic kinetics using M-mode through transhepatic scan we described a similarity between diaphragm excursion during an expiratory forced maneuver and the volume/time curve of spirometry. This allowed us to identify the M-mode Index of Obstruction (MIO), an ultrasound-analogue of FEV1/VC. We observed MIO values of normal subjects (9) and obstructed patients (9) comparing the two groups. FEV1/VC and MIO showed a significant correlation suggesting that MIO may be affected by airways obstruction; MIO values were significatively different between normal and obstructed so that it could identify an obstructive syndrome. The data show that it is possible to suspect the presence of obstructive syndrome of the airways using ultrasonography of the diaphragm.

This study poses as its objective the genetic characterization of the ancient population of the Great White shark, Carcharodon carcharias, L.1758, present in the Mediterranean Sea. Using historical evidence, for the most part buccal arches but also whole, stuffed examples from various national museums, research institutes and private collections, a dataset of 18 examples coming from the Mediterranean Sea has been created, in order to increase the informations regarding this species in the Mediterranean. The importance of the Mediterranean provenance derives from the fact that a genetic characterization of this species' population does not exist, and this creates gaps in the knowledge of this species in the Mediterranean. The genetic characterization of the individuals will initially take place by the extraction of the ancient DNA and the analysis of the variations in the sequence markers of the mitochondrial DNA. This approach has allowed the genetic comparison between ancient populations of the Mediterranean and contemporary populations of the same geographical area. In addition, the genetic characterization of the population of white sharks of the Mediterranean, has allowed a genetic comparison with populations from global "hot spots", using published sequences in online databases (NCBI, GenBank). Analyzing the variability of the dataset, both in terms space and time, I assessed the evolutionary relationships of the Mediterranean population of Great Whites with the global populations (Australia/New Zealand, South Africa, Pacific USA, West Atlantic), and the temporal trend of the Mediterranean population variability. This method based on the sequencing of two portions of mitochondrial DNA genes, markers showed us how the population of Great White Sharks in the Mediterranean, is genetically more similar to the populations of the Australia Pacific ocean, American Pacific Ocean, rather than the population of South Africa, and showing also how the population of South Africa is abnormally distant from all other clusters. Interestingly, these results are inconsistent with the results from tagging of this species. In addition, there is evidence of differences between the ancient population of the Mediterranean with the modern one. This differentiation between the ancient and modern population of white shark can be the result of events impacting on this species occurred over the last two centuries.

Drug abuse is a major global problem which has a strong impact not only on the single individual but also on the entire society. Among the different strategies that can be used to address this issue an important role is played by identification of abusers and proper medical treatment. This kind of therapy should be carefully monitored in order to discourage improper use of the medication and to tailor the dose according to the specific needs of the patient. Hence, reliable analytical methods are needed to reveal drug intake and to support physicians in the pharmacological management of drug dependence. In the present Ph.D. thesis original analytical methods for the determination of drugs with a potential for abuse and of substances used in the pharmacological treatment of drug addiction are presented. In particular, the work has been focused on the analysis of ketamine, naloxone and long-acting opioids (buprenorphine and methadone), oxycodone, disulfiram and bupropion in human plasma and in dried blood spots. The developed methods are based on the use of high performance liquid chromatography (HPLC) coupled to various kinds of detectors (mass spectrometer, coulometric detector, diode array detector). For biological sample pre-treatment different techniques have been exploited, namely solid phase extraction and microextraction by packed sorbent. All the presented methods have been validated according to official guidelines with good results and some of these have been successfully applied to the therapeutic drug monitoring of patients under treatment for drug abuse.

The aim of this work is to measure the stress inside a hard micro object under extreme compression. To measure the internal stress, we compressed ruby spheres (a-Al2O3: Cr3+, 150 µm diameter) between two sapphire plates. Ruby fluorescence spectrum shifts to longer wavelengths under compression and can be related to the internal stress by a conversion coefficient. A confocal laser scanning microscope was used to excite and collect fluorescence at desired local spots inside the ruby sphere with spatial resolution of about 1 µm3. Under static external loads, the stress distribution within the center plane of the ruby sphere was measured directly for the first time. The result agreed to Hertz’s law. The stress across the contact area showed a hemispherical profile. The measured contact radius was in accord with the calculation by Hertz’s equation. Stress-load curves showed spike-like decrease after entering non-elastic phase, indicating the formation and coalescence of microcracks, which led to relaxing of stress. In the vicinity of the contact area luminescence spectra with multiple peaks were observed. This indicated the presence of domains of different stress, which were mechanically decoupled. Repeated loading cycles were applied to study the fatigue of ruby at the contact region. Progressive fatigue was observed when the load exceeded 1 N. As long as the load did not exceed 2 N stress-load curves were still continuous and could be described by Hertz’s law with a reduced Young’s modulus. Once the load exceeded 2 N, periodical spike-like decreases of the stress could be observed, implying a “memory effect” under repeated loading cycles. Vibration loading with higher frequencies was applied by a piezo. Redistributions of intensity on the fluorescence spectra were observed and it was attributed to the repopulation of the micro domains of different elasticity. Two stages of under vibration loading were suggested. In the first stage continuous damage carried on until certain limit, by which the second stage, e.g. breakage, followed in a discontinuous manner.

ABSTRACT One of the major ecological challenges on Lake Victoria resources is the existence of “hot spots”, caused by human waste, urban runoff, and industrial effluents. The lake is tending towards eutrophication which is attributed to the increasing human population in its watershed. A report of the levels of perfluorooctane sulfonate and perfluorooctanoic acid in environmental matrices of Lake Victoria is presented, and the management implication of perfluorinated compounds and similar potential organic pollutants examined. Two widely consumed and economically important fish species namely Lates niloticus (Nile perch) and Oreochromis niloticus (Nile tilapia) were obtained from Winam gulf of Lake Victoria, Kenya, and analysed for perfluorooctane sulfonate and perfluorooctanoic acid in muscles and liver using liquid chromatography coupled with mass spectroscopy. Variability in the concentrations of perfluorooctanoic acid or perfluorooctane sulfonate in river waters (range perfluorooctanoic acid 0.4 – 96.4 ng/L and perfluorooctane sulfonate < 0.4 – 13.2 ng/L) was higher than for Lake waters (range perfluorooctanoic acid 0.4 – 11.7 ng/L and perfluorooctane sulfonate < 0.4 – 2.5 ng/L respectively). Significant correlations were tested between perfluorinated compounds levels in sediments, fish and water. Wastewater treatment plants and other anthropogenic sources have been identified as significant sources or pathways for the introduction of perfluoroalkyl compounds into Lake Victoria ecosystem. In this study, elevated concentrations of perfluorooctanoic acid and perfluorooctane sulfonate was found in two wastewater treatment plants (WWTPs) in Kisumu, City of Kenya. An alternative analytical method to liquid chromatography/ mass spectroscopy for analysis of perfluorocarboxylic acids in abiotic and biotic matrices where high concentrations are expected is also presented. Derivatisation of the acid group to form a suitable alkyl ester provided a suitable compound for mass spectroscopy detection coupled to gas chromatography instrumental analysis. The acid is esterified by an alkyl halide i.e benzyl bromide as the alkylating agent for Perfluorocarboxylic acids quantification. The study also involved degradability measurements of emerging perfluorinated surfactants substitutes. The stability of the substitutes of perfluorinated surfactants was tested by employing advanced oxidation processes, followed by conventional tests, among them an automated method based on the manometric respirometry test and standardized fix bed bioreactor [FBBR] on perfluorobutane sulfonate (PFBS), a fluoroethylene polymer, fluorosurfactant (Zonyl), two fluoraliphaticesters (NOVEC ™ FC4430 and NOVEC ™ FC4432) and 10-(trifluoromethoxy) decane-sulfonate. Most of these emmerging surfactants are well-established in the market and have been used in several applications as alternatives to PFOS and PFOA based surfactants. The results of this study can be used as pioneer information for further studies on the sources, behaviour and fate of PFOA and PFOS and other related compounds in both abiotic and biota compartments of Lake Victoria and other lakes. Further an overview in degradation of emerging perfluorinated compounds substitutes is presented. Contribution in method development especially for acid group based fluorosurfactants is presented. The data obtained in this study can particularly be considered when formulating policies and management measures for preservation and sustainability of Lake Victoria resources.

Questo lavoro di tesi ha avuto come obiettivo la preparazione di catalizzatori attivi nella reazione di ossidazione parziale catalitica, CPO, del metano per produrre gas di sintesi. I catalizzatori sono stati preparati tramite sintesi elettrochimica di composti di tipo idrotalcite a base di Rh/Mg/Al utilizzando come supporto schiume metalliche costituite da FeCrAlY. L’impiego di questo tipo di supporto comporta una serie di vantaggi, dallo sviluppo del catalizzatore all’ottimizzazione del processo catalitico in termini di prestazioni catalitiche, diminuzione degli “hot spots” termici, diminuzione delle perdite di carico e costi del catalizzatore. La sintesi del catalizzatore è stata effettuata per mezzo di una cella elettrochimica innovativa, che lavora in flusso, e quindi permette la continua rigenerazione della soluzione di sintesi, a differenza di quanto avviene in una cella elettrochimica standard a singolo comparto. La precipitazione dei composti di tipo idrotalcite si ottiene grazie alla tecnica di elettrogenerazione di basi, ovvero grazie alla generazione di un pH basico all’interno della cella elettrochimica a seguito dell’applicazione di un potenziale catodico. Il pH generato è il parametro più importante e determina la natura e la qualità del materiale depositato. È sorta quindi la necessità di sviluppare un sensore potenziometrico miniaturizzato per la determinazione istantanea del pH durante la sintesi, da installare all’interno della schiuma stessa. È possibile correlare le prestazioni catalitiche dei catalizzatori sintetizzati con la cella elettrochimica in flusso, alle loro caratteristiche di morfologia superficiale ed alla composizione chimica, e confrontare le stesse prestazioni catalitiche con quelle ottenute sintetizzando i catalizzatori con la cella elettrochimica standard a singolo comparto.

The brown rot fungi belong to a group of fungal pathogens that causes considerable damage to cultivated fruits trees, particularly stone fruits and apples in the temperate regions of the World and during the postharvest with an important economic impact. In particular in Italy, it is important to monitor the Monilinia population to control economic losses associated to the peach and nectarine market. This motivates the research steps presented in this dissertation on Monilinia Italian isolates. The Monilinia species collected from stone fruits have been identified using molecular analysis based on specific primers. The relevant role of M. fructicola was confirmed and, for the first time, it was found also on apple fruits. To avoid the development of resistant strains and implement valid treatment strategies, the understanding of the fruit natural resistance during different developmental stages and the assessment of the Monilinia sensitivity/resistance to fungicides are required. The relationship between the inhibition spots and the phenolic compounds in peach fruit peel was highlighted in this research. Three methods were used to assess isolate resistance/sensitivity, the amended medium, the Spiral Gradient Endpoint Method (SGD) and the Alamar Blue method. The PCR was used to find possible mutation points in the b-tubulin gene that is responsible for fungicide resistance. Interestingly, no mutation points were observed in resistant M. laxa isolates, suggesting that the resistance could be stimulated by environmental factors. This lead to the study of the effect of the temperature on the resistance and the preliminary results of in vitro tests showed that maximum inhibition was observed at 30°C.

Air quality represents a key issue in the so-called pollution “hot spots”: environments in which anthropogenic sources are concentrated and dispersion of pollutants is limited. One of these environments, the Po Valley, normally experiences exceedances of PM10 and PM2.5 concentration limits, especially in winter when the ventilation of the lower layers of the atmosphere is reduced. This thesis provides a highlight of the chemical properties of particulate matter and fog droplets in the Po Valley during the cold season, when fog occurrence is very frequent. Fog-particles interactions were investigated with the aim to determine their impact on the regional air quality. Size-segregated aerosol samples were collected in Bologna, urban site, and San Pietro Capofiume (SPC), rural site, during two campaigns (November 2011; February 2013) in the frame of Supersito project. The comparison between particles size-distribution and chemical composition in both sites showed the relevant contribution of the regional background and secondary processes in determining the Po Valley aerosol concentration. Occurrence of fog in November 2011 campaign in SPC allowed to investigate the role of fog formation and fog chemistry in the formation, processing and deposition of PM10. Nucleation scavenging was investigated with relation to the size and the chemical composition of particles. We found that PM1 concentration is reduced up to 60% because of fog scavenging. Furthermore, aqueous-phase secondary aerosol formation mechanisms were investigated through time-resolved measurements. In SPC fog samples have been systematically collected and analysed since the nineties; a 20 years long database has been assembled. This thesis reports for the first time the results of this long time series of measurements, showing a decrease of sulphate and nitrate concentration and an increase of pH that reached values close to neutrality. A detailed discussion about the occurred changes in fog water composition over two decades is presented.
