Exogenously added IL-10 rapidly inhibited Staphylococcus aureus- or LPS- induced cytokine mRNA expression in human PBMCs and monocytes, with a maximal effect observed when IL-10 was added from 20 h before until 1 h after the addition of the inducers. Nuclear run-on assays revealed that the inhibition of IL-12 p40, IL-12 p35, and TNF-α was at the gene transcriptional level and that the addition of IL-10 to S. aureus- or LPS-treated PBMCs did not affect mRNA stability. The inhibitory activity of IL-10 was abrogated by cycloheximide (CHX), suggesting the involvement of a newly synthesized protein(s). The addition of CHX at 2 h before S. aureus or LPS also inhibited the accumulation of IL-12 p40 mRNA, but did not inhibit IL-12 p35 and TNF-α mRNA. This finding suggests that p40 transcription is regulated through a de novo synthesized protein factor(s), whereas the addition of CHX at 2 h after S. aureus activation caused superinduction of the IL-12 p40, IL-12 p35, and TNF-α genes. These results indicate that in human monocytes, the mechanism(s) of IL-10 suppression of both IL-12 p40 and IL-12 p35 genes is primarily seen at the transcriptional level, and that the induction of the IL-12 p40 and p35 genes have different requirements for de novo protein synthesis.

We report the singular filtration properties of an ultrafiltration membrane made with mesoporous silica that exhibits cylindrical pores aligned mostly normal to the support. This membrane supported on tubular commercial macroporous alumina supports was prepared by the interfacial growth mechanism between stable silica-surfactant hybrid micelles made of the association of silica oligomers with polyethyleneoxide-based (PEO) surfactants and sodium fluoride, a well-known silica condensation catalyst [Boissière et al., An ultrafiltration membrane made with mesoporous MSU-X silica, Chem. Mater. 15 (2003) 460-463]. It appears that the combined effect of the silica nature of the membrane, whose surface charge can be easily adjusted by changing the pH and the non-connected cylindrical shape of the pores provides a new behavior in the retention properties, as proved by the filtration of polyoxyethylene polymers (PEO) with different molecular weights. Depending on the filtration conditions, a rejection rate of 80% and a steep cut-off at 2000 Da can be obtained or, on the reverse, polymers three times bigger than the pore diameter can diffuse through the membrane. This new filtration mechanism, which opens up new modes of separation modes, is explained in the light of both topology of the porous network and pH-dependent interactions between PEO polymers and silica porous media. © 2004 Elsevier B.V. Molecular hybridization is a new concept in drug design and development based on the combination of pharmacophoric moieties of different bioactive substances to produce a new hyrid compound with improved affinity and efficacy, when compared to the parent drugs. Additionally, this strategy can results in compounds presenting modified selectivity profile, different and/or dual modes of action and reduced undesired side effects. So, in this described several example of different strategies for drug design, discovery and pharmacomodulation focused on new innovative hybrid compounds presenting analgesic, anti-inflammatory, platelet anti-aggregating, anti-infections, anticancer, cardio- and neuroactive properties.

Anti-inflammatory drugs are known to be the most widely-marketed drugs in the world, despite their serious side effects, mainly on the gastrointestinal tract. Thus, there are constant efforts to discover new prototypes with improved therapeutic activity and safety for the patient. Since the advent of the computational chemistry, the theoretical study of the physiological behavior of a new compound and hence an understanding of its supposed mechanism of action have been made a lot more accessible. Thus, molecule-receptor mathematical modeling was applied to compound I (1-(2,6-dichlorophenyl)indolin-2-one), to predict theoretically its ability to inhibit, selectively, the COX-2 isoform of prostaglandin endoperoxide synthase (PGHS-2), and the best positions to introduce chemical groups and to make molecular modifications.

Background: The aim of this study was to evaluate the changes in testicular function of rats due to cigarette smoke exposure and the possible mechanism by which zinc protects against these alterations. Methods: MaleWistar rats (60 days old) were randomly divided into 3 groups: control (G1, n = 10); exposed to cigarette smoke (G2, n = 10; 20 cigarettes/day/9 weeks) and exposed to cigarette smoke and supplemented with zinc (G3, n = 8; 20 cigarettes/day/9 weeks; 20 mg/kg zinc chloride daily for 9 weeks, by gavage). After the treatment period, the animals were euthanized, and materials were collected for analyses. Results: G2 rats showed a reduction in body mass; impaired sperm concentration, motility, morphology and vitality; and increased malonaldehyde and thiol group levels and superoxide dismutase activity as compared to G1. Zinc prevented the reduction of sperm concentration and the excessive increase of lipid peroxidation and induced an increase in plasma testosterone levels, wet weight of testis and thiol group concentration. Conclusions: Exposure to cigarette smoke led to harmful effects on testicular function at least partially due to the exacerbation of oxidative stress. Supplementary zinc had an important modulator/protector effect on certain parameters. The mechanism of zinc protection can be through an increase of SH concentration. Thus, zinc supplementation may be a promising addition to conventional treatments for male infertility related to smoking. Copyright © 2012 by Institute of Pharmacology Polish Academy of Sciences.

Carbon nanoscrolls (graphene layers rolled up into papyrus-like tubular structures) are nanostructures with unique and interesting characteristics that could be exploited to build several new nanodevices. However, an efficient and controlled synthesis of these structures was not achieved yet, making its large scale production a challenge to materials scientists. Also, the formation process and detailed mechanisms that occur during its synthesis are not completely known. In this work, using fully atomistic molecular dynamics simulations, we discuss a possible route to nanoscrolls made from graphene layers deposited over silicon oxide substrates containing chambers/pits. The scrolling mechanism is triggered by carbon nanotubes deposited on the layers. The process is completely general and can be used to produce scrolls from other lamellar materials, like boron nitride, for instance. © 2013 American Institute of Physics.

Amostras de sangue de índios nativos na aldeia Kararao (Kayapó) foram analisadas, usando-se métodos sorológico e molecular, para caracterizar a infecção e analisar a transmissão do HTLV-II. Observou-se reatividade específica em 3/26 indivíduos, dos quais duas amostras eram de uma mãe e de seu filho. A análise pela RFLP de regiões pX e env confirmou a infecção pelo HTLV-II. A seqüência de nucleotídios do segmento 5'LTR e a análise filogenética mostraram alta similaridade (98%) entre as três amostras e o protótipo HTLV-IIa (mot) e confirmaram a ocorrência do subtipo HTLV-IIc. Houve uma alta similaridade genética (99,9%) entre as amostras da mãe e do filho e a única diferença foi uma deleção de dois nucleotídios (TC) na seqüência materna. Estudos epidemiológicos anteriores entre índios nativos do Brasil forneceram prova da transmissão intrafamilial e vertical do HTLV-IIc. O presente estudo fornece evidência molecular da transmissão do HTLV-IIc de mãe para filho, um mecanismo que em grande parte é responsável pela endemicidade do HTLV nessas populações epidemiologicamente fechadas. Embora a verdadeira via de transmissão seja desconhecida, a amamentação materna poderia ser a mais provável.

O Virus Oropouche (VORO; Bunyaviridae, Orthobunyavirus) é um dos mais importantes arbovírus que infecta humanos na Amazônia brasileira, e é causador da febre do Oropouche. Entre 1961 e 2009, um grande número de epidemias foi registrado em diferentes centros urbanos dos Estados Brasileiros do Acre, Amapá, Amazonas, Maranhão, Pará, Rondônia e Tocantins, e também no Panamá, Peru e Trinidad & Tobago. Este trabalho teve por objetivo desenvolver um estudo retrospectivo dos aspectos epidemiológicos e moleculares do VORO enfatizando sua distribuição, a dinâmica das epidemias ocorridas no período, bem como a dispersão de diferentes genótipos na América Latina e no Brasil como contribuição à epidemiologia molecular do VORO. Para tanto 66 isolamentos do VORO pertencentes ao acervo do Instituto Evandro Chagas foram propagados em camundongos e em cultura de células VERO, seguida da extração do RNA viral e obtenção do cDNA por RTPCR; os amplicons foram purificados e submetidos ao sequenciamento nucleotídico para análises moleculares e evolução, incluindo o rearranjo genético, estudo de relógio molecular e análise de dispersão viral. Foi demonstrada a presença de quatro linhagens distintas do VORO na Amazônia brasileira (genótipos I, II, III e IV), sendo os genótipos I e II, respectivamente os mais frequentemente encontrados em áreas da Amazônia ocidental e oriental. Esses e o genótipo III estão constantemente evoluindo, mediante o mecanismo “boom and boost” que resulta na emergência seguida de substituição das sublinhagens (subgenótipos) circulantes por outras mais recentes. O genótipo III do VORO, previamente encontrado somente no Panamá, foi descrito na Amazônia e Sudeste do Brasil. Os dados obtidos pela análise filogenética comparativa das topologias para os segmentos PRNA e MRNA sugerem que o VORO utiliza o rearranjo genético como mecanismo de geração de biodiversidade viral, sendo o genótipo I o mais estável e o II o mais instável e, portanto, mais sensível às pressões evolutivas; foi reconhecido um novo genótipo do VORO neste estudo em amostras isoladas em Manaus no ano de 1980, que foi denominado de genótipo IV. O estudo do relógio molecular mostrou que a emergência do VORO se deu no Estado do Pará provavelmente há 223 anos e daí ao longo dos anos se dispersou pela PanAmazônia bem como para o Caribe, sendo que o genótipo I foi o que originou os demais genótipos do VORO.
