We measured visual performance in achromatic and chromatic spatial tasks of mercury-exposed subjects and compared the results with norms obtained from healthy individuals of similar age. Data were obtained for a group of 28 mercury-exposed subjects, comprising 20 Amazonian gold miners, 2 inhabitants of Amazonian riverside communities, and 6 laboratory technicians, who asked for medical care. Statistical norms were generated by testing healthy control subjects divided into three age groups. The performance of a substantial proportion of the mercury-exposed subjects was below the norms in all of these tasks. Eleven of 20 subjects (55%) performed below the norms in the achromatic contrast sensitivity task. The mercury-exposed subjects also had lower red-green contrast sensitivity deficits at all tested spatial frequencies (9/11 subjects; 81%). Three gold miners and 1 riverine (4/19 subjects, 21%) performed worse than normal subjects making more mistakes in the color arrangement test. Five of 10 subjects tested (50%), comprising 2 gold miners, 2 technicians, and 1 riverine, performed worse than normal in the color discrimination test, having areas of one or more MacAdam ellipse larger than normal subjects and high color discrimination thresholds at least in one color locus. These data indicate that psychophysical assessment can be used to quantify the degree of visual impairment of mercury-exposed subjects. They also suggest that some spatial tests such as the measurement of red-green chromatic contrast are sufficiently sensitive to detect visual dysfunction caused by mercury toxicity.

Mercury is a xenobiotic metal that is a highly deleterious environmental pollutant. The biotransformation of mercury chloride (HgCl2) into methylmercury chloride (CH3HgCl) in aquatic environments is well-known and humans are exposed by consumption of contaminated fish, shellfish and algae. The objective of the present study was to determine the changes induced in vitro by two mercury compounds (HgCl2 and CH3HgCl) in cultured human lymphocytes. Short-term human leukocyte cultures from 10 healthy donors (5 females and 5 males) were set-up by adding drops of whole blood in complete medium. Cultures were separately and simultaneously treated with low doses (0.1 to 1000 µg/l) of HgCl2 and CH3HgCl and incubated at 37ºC for 48 h. Genotoxicity was assessed by chromosome aberrations and polyploid cells. Mitotic index was used as a measure of cytotoxicity. A significant increase (P < 0.05) in the relative frequency of chromosome aberrations was observed for all concentrations of CH3HgCl when compared to control, whether alone or in an evident sinergistic combination with HgCl2. The frequency of polyploid cells was also significantly increased (P < 0.05) when compared to control after exposure to all concentrations of CH3HgCl alone or in combination with HgCl2. CH3HgCl significantly decreased (P < 0.05) the mitotic index at 100 and 1000 µg/l alone, and at 1, 10, 100, and 1000 µg/l when combined with HgCl2, showing a synergistic cytotoxic effect. Our data showed that low concentrations of CH3HgCl might be cytotoxic/genotoxic. Such effects may indicate early cellular changes with possible biological consequences and should be considered in the preliminary evaluation of the risks of populations exposed in vivo to low doses of mercury.

A ligação entre as zonas urbanas e as questões ambientais ficam mais próximas na medida em que cresce a conscientização global de conservar, melhorar e valorizar os serviços ambientais prestados pela natureza para a sustentabilidade da vida, dentro e fora da cidade. Cobertura vegetal (ou cobertura verde) está dentre as principais fontes de tais serviços. Uma vez que o processo de urbanização se mostra irreversível e os problemas ambientais urbanos se alastram em tamanho e extensão, a presença do verde está diretamente relacionada aos indicadores de qualidade de vida urbana. Como reflexo do processo de urbanização, a cidade de Belém perdeu uma grande porcentagem de seus ecossistemas naturais, de modo que este trabalho se concentrou em analisar alguns serviços ecossistêmicos—qualidade do ar, poluição do ar e regulação do clima - fornecidos pela qualidade e pela quantidade de cobertura vegetal local, considerando as alterações na distribuição espaço-temporal, em três distritos administrativos. Um marco teórico foi construído e analisado; a cobertura vegetal foi calculada, utilizando-se NDVI e Cobertura Vegetal Fracional em imagens do LANDSAT 5, ao longo de um período de 23 anos. A partir de uma proposta de escala mais detalhada de NDVI, análises quantitativas e qualitativas da cobertura verde evidenciaram perda significativa de cobertura muito densa, densa, moderada e aumento de áreas de pouca ou nenhuma vegetação. Ademais, lesão das áreas verdes sinalizou tendências de aumento da poluição do ar, da poluição sonora e da temperatura. A carência de dados relacionados ao meio ambiente não deixa dúvida sobre a urgência de investimento nos serviços ambientais provenientes da cobertura vegetal, para a sustentabilidade urbana em Belém, cujos cenários previstos são de drásticas perdas de área verde. Mais pesquisas e iniciativas de instituições públicas e privadas são necessárias para a contribuição aos serviços ambientais em Belém e, consequentemente, ao bem-estar público.

The mercury rejected in the water system, from mining operations and lixiviation of soils after deforestation, is considered to be the main contributors to the contamination of the ecosystem in the Amazon Basin. The objectives of the present study were to examine cytogenetic functions in peripheral lymphocytes within a population living on the banks of the Tapajós River with respect to methylmercury (MeHg) contamination, using hair mercury as a biological indicator of exposure. Our investigation shows a clear relation between methylmercury contamination and cytogenetic damage in lymphocytes at levels well below 50 micrograms/gram, the level at which initial clinical signs and symptoms of mercury poisoning occur. The first apparent biological effect with increasing MeHg hair level was the impairment of lymphocyte proliferation measured as mitotic index (MI). The relation between mercury concentration in hair and MI suggests that this parameter, an indicator of changes in lymphocytes and their ability to respond to culture conditions, may be an early marker of cytotoxicity and genotoxicity in humans and should be taken into account in the preliminary evaluation of the risks to populations exposed in vivo. This is the first report showing clear cytotoxic effects of long-term exposure to MeHg. Although the results strongly suggest that, under the conditions examined here, MeHg is both a spindle poison and a clastogen, the biological significance of these observations are as yet unknown. A long-term follow-up of these subjects should be undertaken.

O mercúrio é um metal que se destaca dos demais por se apresentar líquido em temperatura e pressão normais. Este xenobiótico se apresenta como a maior fonte de poluição em várias partes do mundo e tem como característica ser altamente tóxico ao Sistema Nervoso Central (SNC). O despejo é na forma líquida diretamente no solo e leito dos rios. Este metal pesado é complexado com vários elementos presentes no solo ou sedimentos sendo convertido à metilmercúrio (MeHg) pela microbiota aquática. O MeHg apresenta a capacidade de se acumular ao longo da cadeia trófica, um evento conhecido como biomagnificação, o qual afeta diretamente a vida humana. Nesse sentido, a Região Amazônica se destaca por possuir todos os componentes necessários para a manutenção do ciclo biogeoquímico do mercúrio, além de populações cronicamente expostas a este metal pesado, sendo este fato considerado um problema de saúde pública. Tem-se conhecimento que este xenobiótico após a exposição aguda a altas doses promove desordens relacionadas ao surgimento de processos degenerativos no SNC, entretanto, os efeitos a baixas concentrações ainda não são totalmente conhecidos. Nesse sentido, se destacam as células gliais que atuam como mediadores no processo de neurotoxicidade desse metal, principalmente em baixas concentrações. Apesar de este tipo celular exibir um importante papel no processo de intoxicação mercurial, a ação deste metal sobre as células glias é pouco conhecida, principalmente sobre o genoma e a proliferação celular. Desta forma, este trabalho se propõe a avaliar o efeito da exposição a este xenobiótico em baixa concentração sobre o material genético e a proliferação celular em células da linhagem glial C6. As avaliações bioquímica (atividade mitocondrial – medida pelo ensaio de MTT –) e morfofuncional (integridade da membrana – avaliada pelo ensaio com os corantes BE e AA –) confirmaram a ausência de morte celular após a exposição ao metal pesado na concentração de 3 μM por um intervalo de 24 horas. Mesmo sem promover processos de morte celular, o tratamento com esta concentração subletal de MeHg foi capaz de aumentar significativamente os níveis dos marcadores de genotoxicidade (fragmentação do DNA, formação de micronúcleos, pontes nucleoplásmica e brotos nucleares). Ao mesmo tempo, foi possível observar uma alteração no ciclo celular através do aumento do índice mitótico e uma mudança no perfil do ciclo celular com aumento da população celular nas fases S e G2/M, sugerindo um aprisionamento nessa etapa. Esta mudança no ciclo celular, provocada por 24h de exposição ao MeHg, foi seguida de uma redução no número de células viáveis e confluência celular 24h após a retirada do MeHg e substituição do meio de cultura, além do aumento no tempo de duplicação da cultura do mesmo. Este estudo demonstrou pela primeira vez que a exposição ao metilmercúrio em concentração baixa e subletal é capaz de promover eventos genotóxicos e distúrbios na proliferação celular em células de origem glial.

This paper presents preliminary findings for a metallomics study of mercury in the muscle of the fish from Amazonas region - Brazil, after protein separation by two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) and subsequent evaluation of mercury by Thermal Decomposition and Amalgamation Coupled with Atomic Absorption (TTA-CAAS). It was found that mercury is present in 18 protein spots of dourada muscle (Brachyplatystoma rousseauxii) and 9 protein spots of pacu muscle (Myleus sp.). The protein spots in which they were determined the presence of mercury present molecular weight of 13.5 and 21.4 kDa and isoelectric point of 3.8 and 9.2, respectively.

This experiment aim was to evaluate the contamination of the trace elements (TE) arsenic, barium, cadmium, chromium, copper, mercury, molybdenum, nickel, lead, selenium, and zinc considered in the CONAMA resolution 375 after 13 years long using sewage sludge (SS) as fertilizer in two soils: an Eutroferric Clayed Red Latosol (Rhodic Eutrudox - RE) and a Dystrophic Red Latosol (Typic Haplorthox - TH). Experiment in the field under maize cultivation had four treatments (0, 5, 10, and 20 t of SS ha(-1), dry weight), five replications and an experimental design in randomized blocks. The agrochemicals (dolomitic limestone, single superphosphate, and potassium chloride), SS, soils, and the certified reference materials were digested according to the USEPA 3051A method and the chemical elements were quantified by ICP OES. The TE contents found in the agrochemicals used should not cause immediate environmental impact. The higher TE values were found in the RE and they did not reach the agricultural (more stringent) Investigation Level (IL) yet, according to 420 CONAMA resolution. Persisting the actual SS fertilization amount applied in the soil and the TE concentration in the SS is foreseen that Ba, Cd, Cr, Cu, Ni, and Zn will be the first elements to reach the IL in the RE.

To verify the levels of concentration of some heavy metals in fishes from Sorocaba river (São Paulo, Brazil) and evaluate if this contamination offers health risks to the fishermen, 63 samples of fishes collected from four points along the river were studied for cadmium, lead, chromium, nickel and mercury, with emphasis in this last, since it is the most toxic and most probable as a fish contaminant. Analyzing muscle samples by cold vapor atomic absorption spectrometry it was shown that the fishes are not contaminated. None of the five metals studied were present in prohibitive level and the fishes could be judged secure for human consume. It was also analyzed data from four years of cadmium, lead, chromium, nickel and mercury monitoring made by CETESB, from 1997 to 2000, in water from the main rivers of the State of São Paulo. The study pointed out that the majority of the monitored rivers still present contamination by those metals in a level that requires an improvement of the pollution control actions.

Fish bioassays are valuable tools that can be used to elucidate the toxicological potential of numerous substances that are present in the aquatic environment. In this study, we assessed the antagonistic action of selenium (Se) against the toxicity of mercury (Hg) in fish (Oreochromis niloticus). Six experimental groups with six fish each were defined as follows: (1) control, (2) mercury (HgCl2), (3) sodium selenite (Na2Se4O3), (4) sodium selenate (Na2Se6O4), (5) mercury + sodium selenite (HgCl2 + Na2Se4O3), and (6) mercury + sodium selenate (HgCl2 + Na2Se6O4). Hematological parameters [red blood cells (RBC), white blood cells (WBC), and erythroblasts (ERB)] in combination with cytogenotoxicity biomarkers [nuclear abnormalities (NAs) and micronuclei (MN)] were examined after three, seven, ten, and fourteen days. After 7 days of exposure, cytogenotoxic effects and increased erythroblasts caused by mercury, leukocytosis triggered by mercury + sodium selenite, leukopenia associated with sodium selenate, and anemia triggered by mercury + sodium selenate were observed. Positive correlations that were independent of time were observed between WBC and RBC, ERB and MN, and NA and MN. The results suggest that short-term exposure to chemical contaminants elicited changes in blood parameters and produced cytogenotoxic effects. Moreover, NAs are the primary manifestations of MN formation and should be included in a class characterized as NA only. Lastly, the staining techniques used can be applied to both hematological characterization and the measurement of cytogenotoxicity biomarkers.
