The present work shows a method for the determination of chloramphenicol (CAP) antibiotic in milk, powder milk and honey. The solid phase extraction and liquid-liquid extraction were applied as a clean-up and pre-concentration strategies followed by LC-ESI/MS/MS analysis. The recovery was studied for different fortification levels from 0.05 to 1.00 µg L-1 in milk, showing values between 91 101% and RSD bellow 8.0%, while honey was spiked with a concentration of 0.20 µg kg-1 yelding a mean recovery of 83% and RSD of 6.5%. The quantification transition 321>152 showed a LOD of 0.52 ng kg-1 and LOQ of 1.85 ng kg-1.

A simple, fast and low-cost methodology was optimized, seeking preparation of a crude pig liver esterase (PLE) concentrate. Basically, the method consisted of the following steps: liver homogenization, acetone washing, enzyme extraction and purification/concentration. Starting from 1 kg of fresh liver more than 200 kU of PLE suspension were obtained after 8 hours, at an estimated cost of US$0.21/kU. The PLE concentrate thus obtained was stable, showing 96-100% of the initial activity after 7 months in refrigerator at 4°C.

By the year 2005 the world biochemical market will reach an estimated $ 100 billion and separation processes are a vital link between lab discoveries and the fulfillment of this commercialization potential. The practical application of aqueous two-phase systems (ATPS) to extraction processes has been exploited for several years for the recovery of biological products. Unfortunately, this has not resulted in an extensive presence of the technique in commercial processes. In this paper a critical overview of the fundamental thermodynamic properties related to formation of aqueous two-phase systems and their application to extraction and purification of bioparticules is presented.

Water quality was monitored at the upper course of the Rio das Velhas, a major tributary of the São Francisco basin located in the state of Minas Gerais, over an extension of 108 km from its source up to the limits with the Sabara district. Monitoring was done at 37 different sites over a period of 2 years (2003-2004) for 39 parameters. Multivariate statistical techniques were applied to interpret the large water-quality data set and to establish an optimal long-term monitoring network. Cluster analysis separated the sampling sites into groups of similarity, and also indicated the stations investigated for correlation and recommended to be removed from the monitoring network. Principal component analysis identified four components, which are responsible for the data structure explaining 80% of the total variance of the data. The principal parameters are characterized as due to mining activities and domestic sewage. Significant data reduction was achieved.

The aim of this work is to present the new applications of membrane technology in fat and oil processing, with emphasis on development and applications of the enzymatic membrane reactor and its association with extraction and purification technology by supercritical fluids (SCF). Combining the extraction by SCFs and the separation by membranes allows the integration of extractions reactions with selective separation by membranes through filtration of the supercritical mixture (SCF + extracted solutes). This association provides important energy savings regarding the SCF recompression costs.

European naturalists explored Brazil in long scientific expeditions and published accounts that make up a rich and still largely untapped historiographic source for the understanding of the history of chemistry. The production of indigo dye, the manufacture of limestone, extraction and purification of saltpeter and the production of salt are discussed. Lime was used to whitewash walls and, mixed with whale oil, as cement to glue stones in buildings of the colonial period. It was prepared by burning seashells in specifically designed ovens. Saltpeter was produced by reacting naturally occurring calcium and magnesium nitrate with potassium-rich wood ashes to yield KNO3. NaCl was obtained by evaporating seawater under the sun. Indigo, a native plant, was cultivated and processed to produce the renowned dye, which was exported to Europe.

This study presents Pd determinations in bovine tissue samples containing palladium prepared in the laboratory, and CCQM-P63 automotive catalyst materials of the Proficiency Test, using instrumental thermal and epithermal neutron activation analysis and energy dispersive X-ray fluorescence techniques. Solvent extraction and solid phase extraction procedures were also applied to separate Pd from interfering elements before the irradiation in the nuclear reactor. The results obtained by different techniques were compared against each other to examine sensitivity, precision and accuracy.

The ornamental rock industry generates huge amounts of wastes during the process of extraction and sawing of rock blocks. The ornamental rock powder waste is a non-biodegradable material, which represents the increase in environmental problem. The waste was collected from a granitic rock sawing plant located in Santo Antônio de Pádua, Rio de Janeiro. The chemical-environmental characterization and classification of the waste were done according to ABNT standards. The results showed that the granitic rock powder waste should be classified as Class II A - "No Inert", because of its high concentrations of lead, chrome, iron and manganese.

Extraction and clean-up are essential points in polycyclic aromatic hydrocarbon (PAHs) analysis in a solid matrix. This work compares extraction techniques and clean-up procedures for PAH analysis. PAH levels, their toxicological significance and source were also evaluated in the waters of the Cocó and Ceará rivers. The efficiency of PAH recovery was higher for the soxhlet and ultrasonic techniques. PAH recovery varied from 69.3 to 99.3%. Total PAH concentration (ΣHPA) varied from 720.73 to 2234.76 µg kg-1 (Cocó river) and 96.4 to 1859.21 µg kg-1 (Ceará river). The main PAH sources are pyrolytic processes and the levels were classified as medium so that adverse effects are possible.

Plastic packaging materials intended for use in food packaging is an area of great interest from the scientific and economic point of view due to the irreversible internationalization and globalization process of food products. Nevertheless, a debate related to food safety aspects has emerged within the scientific community. Therefore, the development of analytical methods that allow identifying and quantifying chemical substances of toxicological potential in the packaging is considered essential. This article focuses on the main analytical methods, including validation parameters, as well as extraction and quantification techniques for determination of volatile organic compounds from food packaging materials.

A novel solventless sample preparation, stir-bar sorptive extraction (SBSE), for extraction, and sample enrichment of organic compounds from biological fluids, is described in this manuscript from principle to applications. The SBSE is based on sorptive extraction, whereby the compounds are extracted into a polymer coating, polydimethylsiloxane (PDMS), on a magnetic stirring rod. The extraction is controlled by the partitioning coefficient of drugs between the PDMS and sample matrix, and upon the sample-extraction medium phase ratio. The SBSE technique has been applied successfully, with high sensitivities, to biomedical analysis of volatiles and for semi-volatiles drugs from biological sample, including urine, plasma, and saliva. SBSE combined with in situ derivatization, drugs quite more polar (e.g. metabolites) also can be analyzed.

A technique for both extraction and activity measurement of peroxidase extracted from arazá (Eugenia stipitata Mc Vaugh) is described. Peroxidase from arazá pulp fruit was extracted using a combination of protein precipitation with acetone and extraction with 50 mM sodium buffer phosphate (pH 6.0). Optimum activity using guaiacol as H-donor was obtained at pH from 5.0 to 6.5, temperature from 60 to 75 °C, H2O2 between 10 to 15 mM and guaiacol from 80 to 160 mM. Thermal inactivation showed a first-order inactivation kinetic. Reactivation was observed when extracts were heated at 80 °C and afterwards incubated at 25 °C.

The aim of the present work was to develop and optimize a method for determination of bioavailable phosphorus in samples of feces and fish feed using ultrasound extraction and subsequent quantification by visible spectrophotometry. Using as extractor solution HNO3 0.50 mol L-1, the great conditions of extraction established were: sample mass - 100 mg, samples granulometry - < 60 µm, sonification time - five cycles of 40 s and ultrasound potency - 136 W. The proposed method was applied in studies of digestibility of this nutrient in different feeds used in diets of juvenile of Nile tilapia.

A synthesis of (+)-±-terpineol from (+)-limonene was proposed as a project for undergraduate organic laboratory course. Terpineol is a useful flavor and fragrance compound, and several aspects of this preparation are suited for experimental organic classes, including basic techniques for extraction and analyses of essential oils, different reaction types and the possibility of a high degree of student interest.

The effects of the extraction system (50% methanol or 50% methanol pH 2.0), volume/material ratio, temperature, time and extractions with 70% acetone were evaluated in the total phenolic compounds (TPC) extraction and in antioxidant activities (AA) using FRAP and ABTS assays in guava fruit. The best yield was obtained when 0.5 g of guava were extracted first with 20 mL 50% methanol and then four times with 20 mL 70% acetone during 30 min at 50 °C. Among the different trials guava fruit exhibited high levels of AA as well as TPC.
