La resistencia de las plantas a los hongos necrótrofos como Plectosphaerella cucumerina es genéticamente compleja y depende de la activación coordinada de distintas rutas de señalización (Llorente et al, 2005; Sanchez-Vallet et al, 2010). Entre éstas se encuentran las mediadas por la proteína G heterotrimérica, un complejo formado por tres subunidades (Gα, Gβ y Gγ) que regula tanto la respuesta de inmunidad a diferentes patógenos como distintos procesos de desarrollo (Temple and Jones, 2007). En esta Tesis hemos demostrado que, en Arabidopsis, el monómero funcional formado por las subunidades Gβ y Gγ1/Gγ2 es el responsable de la regulación de la respuesta de defensa, ya que mutantes nulos en estas subunidades (agb1 y agg1 agg2) presentan una alta susceptibilidad al hongo P. cucumerina. Además, hemos identificado varios aminoácidos (Q102, T188 y R235) de la proteína AGB1 esenciales en la interacción con los efectores correspondientes para la regulación de la respuesta inmune (Jiang et al, enviado). Para determinar las bases moleculares de la resistencia mediada por la proteína G heterotrimérica, llevamos a cabo un análisis transcriptómico comparativo entre los genotipos agb1 y Col-0, el cual reveló que la resistencia mediada por AGB1 no depende de rutas defensivas implicadas en la resistencia a hongos necrotrofos, como las mediadas por el ácido salicílico (SA), etileno (ET), jasmónico (JA) o ácido abscísico (ABA), o la ruta de biosíntesis de metabolitos derivados del triptófano. Este estudio mostró que un número significativo de los genes desregulados en respuesta a P. cucumerina en el genotipo agb1 respecto a las plantas silvestres codificaban proteínas con funciones relacionadas con la pared celular. La evaluación de la composición y estructura de la pared de los mutantes de las subunidades de la proteína G heterotrimérica reveló que los genotipos agb1 y agg1 agg2 presentaban alteraciones similares diferentes de las observadas en plantas silvestres Col-0, como una reducción significativa en el contenido de xilosa en la pared. Estos datos sugieren que la proteína G heterotrimérica puede modular la composición/estructura de la pared celular y contribuir, de esta manera, en la regulación de la respuesta inmune (Delgado- Cerezo et al, 2011). La caracterización del interactoma de la proteína G heterotrimérica corroboró la relevancia funcional que presenta en la regulación de la pared celular, ya que un número significativo de las interacciones identificadas estaban comprendidas por proteínas relacionadas directa o indirectamente con la biogénesis y remodelación de la pared celular (Klopffleisch et al, 2011). El papel en inmunidad de algunos de estos potenciales efectores ha sido validado mediante el análisis de la resistencia a P. cucumerina de los mutantes de pérdida de función correspondientes. Con el objetivo de caracterizar las rutas de señalización mediadas por AGB1 e identificar efectores implicados en esta señalización, llevamos a cabo una búsqueda de mutantes supresores de la susceptibilidad de agb1 a P. cucumerina, identificándose varios mutantes sgb (supressor of Gbeta). En esta Tesis hemos caracterizado en detalle el mutante sgb10, que presenta una activación constitutiva de las rutas de señalización mediadas por SA y JA+ET y suprime el fenotipo de susceptibilidad de agb1. SGB10 y AGB1 forman parte de rutas independientes en la regulación de la respuesta inmune, mientras que interaccionan de forma compleja en el control de determinados procesos de desarrollo. La mutación sgb10 ha sido cartografiada entre los genes At3g55010 y At3g56408, que incluye una región con 160 genes. ABSTRACT Plant resistance to necrotrophic fungi Plectosphaerella cucumerina is genetically complex and depends on the interplay of different signalling pathways (Llorente et al, 2005; Sanchez-Vallet et al, 2010). Among others, the heterotrimeric G protein complex has a relevant role. The G protein that is formed by three subunits (Gα, Gβ and Gγ) is a pleiotropic regulator of immune responses to different types of pathogens and developmental issues (Temple and Jones, 2007). Throughout the Thesis, we have demonstrated that Arabidopsis’ functional monomer formed by the Gβ and Gγ1/Gγ2 subunits is a key regulator of defense response, as null mutants (agb1 and agg1 agg2) are equally hypersusceptible to P. cucumerina infection. In addition we have identified several AGB1 aminoacids (Q102, T188 y R235) essentials to interact with specific effectors during the regulation of immune response (Jiang et al, sent).To determine the molecular basis of heterotrimeric G protein mediated resistance we have performed a microarray analysis with agb1-1 and wild type Col-0 plants before and after P. cucumerina challenge. A deep and exhaustive comparative transcriptomical analysis of these plants revealed that AGB1 mediated resistance does not rely on salicilic acid (SA), ethylene (ET), jasmonates (JA), abscisic acid (ABA) or triptophan derived metabolites biosynthesis. However the analysis revealed that a significant number of cell wall related genes are misregulated in the agb1 mutant after pathogen challenge when compared to wild-type plants. The analysis of cell wall composition and structure showed similar cell wall alterations between agb1 and agg1 agg2 mutants that are different from those of wild-type plants, so far the mutants present a significant reduction in xylose levels. All these results suggest that heterotrimeric G protein may regulate immune response through modifications in the cell wall composition/structure (Delgado-Cerezo et al, 2011). The characterization of Heterotrimeric G protein interactome revealed highly connected interactions between the G-protein core and proteins involved in cell wall composition or structure (Klopffleisch et al, 2011). To test the role in immunity of several effectors identified above, we have performed resistance analysis of corresponding null mutants against P. cucumerina. In order to characterize AGB1 mediated signalling pathway and identify additional effectors involved in AGB1-mediated immune response against P. cucumerina, we have performed a screening to isolate mutants with suppression of agb1 phenotype. One of the mutants, named sgb10, has been characterized during the Thesis. The mutant shows constitutive expression of SA, JA+ET-mediated defense signaling pathways to suppres agb1 hypersusceptibility. SGB10 and AGB1 proteins seem to be part of independent pathways in immunity, however its function during development remains unclear. At present, we have mapped the sgb10 mutation between At3g55010 and At3g56408 genes. This region contains 160 genes.

En regiones en las que no se dispone de un modelo hidrológico correctamente calibrado para estimar la escorrentía, se puede recurrir a la utilización de la escorrentía directa simulada por los modelos climáticos o a la escorrentía calculada a partir de fórmulas climatológicas que emplean variables climáticas básicas como la precipitación, temperatura y radiación solar, simuladas por los modelos regionales de clima (MRC). El presente trabajo compara el comportamiento de la escorrentía directa obtenida por 10 simulaciones de los MRC del proyecto europeo PRUDENCE y la escorrentía media anual calculada a partir de la aplicación de cinco fórmulas climatológicas (Schreiber, Ol’dekop, Budyko, Turc-Pike, Zhang et al.) basadas en el índice de aridez, definido por la relación entre la evapotranspiración potencial y la precipitación. Series temporales mensuales de escorrentía, precipitación, temperatura y radiación solar son generadas a partir de las simulaciones de los MRC en 338 cuencas de España que cubren la totalidad del territorio peninsular, bajo condiciones de clima actual (periodo 1961-1990). La evapotranspiración potencial se obtiene usando el método presentado por Hargreaves. Estas formas funcionales estiman la relación entre la evapotranspiración actual y la precipitación y a través de un balance hídrico se calculan los valores de escorrentía anual. El comportamiento general de las variables climáticas simuladas por los MRC se caracteriza por presentar menor sesgo para precipitación y temperatura que para escorrentía. Empleando estadísticos de comparación se analiza la capacidad que tiene la escorrentía directa y la escorrentía anual calculada a partir de las fórmulas climáticas para reproducir los valores observados de escorrentía natural estimada por el modelo hidrológico distribuido SIMPA en las cuencas españolas. En total se generaron 10 series mensuales de escorrentía directa y 50 series de escorrentía anual basadas en el índice de aridez (cada fórmula climática aplicada a las 10 simulaciones de los MRC). Los resultados muestran que la fórmula de Schreiber produce la mejor aproximación a los valores observados y por tanto minimiza el sesgo predominante en la variable escorrentía. Adicionalmente, estos resultados se validan con las capas de escorrentía media anual de alta resolución proporcionada por la UNH/GRDC (University of New Hampshire/Global Runoff Data Centre) que preservan la exactitud de las medidas de las aportaciones observadas en las principales estaciones hidrológicas de todo el mundo, y que en la actualidad es el “mejor estimador” de la escorrentía terrestre sobre grandes extensiones. En este caso, los resultados muestran también que la fórmula de Schreiber estima mejor los valores de escorrentía anual que la escorrentía directa simulada por MRC.

Uno de los principales problemas a los que se enfrentan las comunidades rurales en los países en vías de desarrollo en el ámbito agrícola es la falta de acceso a la asistencia técnica y la capacitación entre de los pequeños agricultores. La disponibilidad de alimentos en Nicaragua es el 72% de la media del Suministro de Energía Alimentaria (SEA), aunque en las zonas de montaña esta cifra baja al 65% del SEA nacional. Este trabajo analiza el impacto que las escuelas de campo para agricultores (ECAs) están teniendo sobre la seguridad alimentaria y nutricional, así como su relación con el margen bruto agrario y la diversificación de cultivos y usos del suelo en las comunidades rurales de montaña de San José de Cusmapa. En 2010 se realizaron 80 encuestas semiestructuradas y se midieron indicadores ecológicos, económicos y sociales. La formación y capacitación proporcionada por las ECAs a lo largo del tiempo, ha resultado tener un efecto positivo sobre el incremento de la agrobiodiversidad, que se asociada a una mejora en el suministro de energía y nutrientes en la dieta. También se ha observado un efecto positivo en el margen bruto agrario de las familias en ECAs con más de cuatro años de actividad.

El Sistema de Seguros Agrarios con el Seguro de cobertura de los daños por sequía en los pastos aprovechados por el ganado en régimen extensivo (línea de seguro 133) aplica la teledetección mediante un índice de vegetación (NDVI), con el fin de solucionar los problemas de peritación que surgen cuando se tiene que determinar la cantidad y calidad del pasto afectado por la sequía. Por ello el seguro de cobertura de los daños por sequía en pastos es el principal instrumento para hacer frente al gasto que supone la necesidad de suplemento de alimentación del ganado reproductor debido a la sequía. En las comarcas de Vitigudino, Trujillo y Valle de los Pedroches (España) se comparó la evolución del seguro de sequía en pastos desde 2006 a 2010 con un modelo matemático de crecimiento del pasto en función de las variables ecofisiológicas y ambientales. Sumadas las decenas de sequía extrema y sequía leve, el modelo matemático contabilizó un número mayor de decenas que las proporcionadas por Agroseguro. La recomendación es comparar las curvas de crecimiento del pasto con las curvas de evolución del NDVI, para ajustar ambos modelos

La sequía es un término meteorológico que significa un periodo seco prolongado. El objetivo de este trabajo es caracterizar el fenómeno estacional de la sequía en pastos anuales de dehesa. Durante 2010 y 2011 se realizó un seguimiento del pasto herbáceo en El Cubo de Don Sancho (Salamanca), Trujillo (Cáceres) y Pozoblanco (Córdoba). Se midió la producción herbácea y se caracterizó botánicamente cada zona, además se midió mensualmente la variación del contenido de agua en el suelo mediante un TDR y la precipitación. Los datos de campo de precipitación, humedad del suelo y cantidad de pasto en pie, y los datos estimados de evaporación se han comparado con la evolución del índice de vegetación para seguros de sequía por teledetección determinado por Agroseguro. Los resultados mostraron un retardo entre la acumulación de agua en el suelo y el crecimiento del pasto, que se transfiere a las medidas del índice de vegetación por teledetección. En los dos años de estudio los periodos de sequía sucedieron al inicio de crecimiento, justo después de la sequía estacional típica del verano.

Tablas de Daimiel National Park is located in the Upper Guadiana Basin and represents one of the largest and most important wetlands in Europe. The long term ecological integrity of this wetland is inherently associated with the maintenance of a shallow groundwater table, namely the Western Mancha aquifer (WMA) or Aquifer 23. The intensive use of groundwater, mainly for irrigation, has led over the last decades to deep socio‐economic changes. Such intensive use has also lowered the water table of Aquifer 23, drastically reducing the flooded area of the wetland and threatening its ecological integrity. A number of plans and measures have been developed and implemented since the declaration of overexploitation of Aquifer 23 in the year 1987. The most recent one is the Special Plan for the Upper Guadiana (SPUG), approved in 2008. This Plan is the main measure to comply with achieving the objective of good quantitative and qualitative status required under the Water Framework Directive (2000). This paper offers a new type of integrated analysis which allows assessing under a common lens the physical, economic and social dimensions of groundwater use in the area. The first objective is to calculate the groundwater footprint of agricultural production in the Upper Guadiana basin and its evolution during 2000‐2008. For this purpose, we have applied the Extended Water Footprint (EWF) methodology ‐a novel approach based on the classical Water Footprint (WF) approach‐ that includes an assessment of the water productivity from an economic and social perspective. Compared to the classical WF, the EWF allows for a more complete overview of the sector, providing new insights for policy decisions (e.g. to define options and possibilities on water re‐allocation in order to achieve both better ecosystem conservation and social equity). The second objective is to use the EWF to compare the existing authorized and non‐authorized or illegal use of water. This allows us to discuss current initiatives by public authorities in relation to the existing frame of water rights

In arid countries worldwide, social conflicts between irrigation-based human development and the conservation of aquatic ecosystems are widespread and attract many public debates. This research focuses on the analysis of water and agricultural policies aimed at conserving groundwater resources and maintaining rurallivelihoods in a basin in Spain's central arid region. Intensive groundwater mining for irrigation has caused overexploitation of the basin's large aquifer, the degradation of reputed wetlands and has given rise to notable social conflicts over the years. With the aim of tackling the multifaceted socio-ecological interactions of complex water systems, the methodology used in this study consists in a novel integration into a common platform of an economic optimization model and a hydrology model WEAP (Water Evaluation And Planning system). This robust tool is used to analyze the spatial and temporal effects of different water and agricultural policies under different climate scenarios. It permits the prediction of different climate and policy outcomes across farm types (water stress impacts and adaptation), at basin's level (aquifer recovery), and along the policies’ implementation horizon (short and long run). Results show that the region's current quota-based water policies may contribute to reduce water consumption in the farms but will not be able to recover the aquifer and will inflict income losses to the rural communities. This situation would worsen in case of drought. Economies of scale and technology are evidenced as larger farms with cropping diversification and those equipped with modern irrigation will better adapt to water stress conditions. However, the long-term sustainability of the aquifer and the maintenance of rurallivelihoods will be attained only if additional policy measures are put in place such as the control of illegal abstractions and the establishing of a water bank. Within the policy domain, the research contributes to the new sustainable development strategy of the EU by concluding that, in water-scarce regions, effective integration of water and agricultural policies is essential for achieving the water protection objectives of the EU policies. Therefore, the design and enforcement of well-balanced region-specific polices is a major task faced by policy makers for achieving successful water management that will ensure nature protection and human development at tolerable social costs. From a methodological perspective, this research initiative contributes to better address hydrological questions as well as economic and social issues in complex water and human systems. Its integrated vision provides a valuable illustration to inform water policy and management decisions within contexts of water-related conflicts worldwide.

Aquaponics is the science of integrating intensive fish aquaculture with plant production in recirculating water systems. Although ion waste production by fish cannot satisfy all plant requirements, less is known about the relationship between total feed provided for fish and the production of milliequivalents (mEq) of different macronutrients for plants, especially for nutrient flow hydroponics used for strawberry production in Spain. That knowledge is essential to consider the amount of macronutrients available in aquaculture systems so that farmers can estimate how much nutrient needs to be supplemented in the waste water from fish, to produce viable plant growth. In the present experiment, tilapia (Oreochromis niloticus L.) were grown in a small-scale recirculating system at two different densities while growth and feed consumption were noted every week for five weeks. At the same time points, water samples were taken to measure pH, EC25, HCO3 – , Cl – , NH4 + , NO2 – , NO3 – , H2PO4 – , SO4 2– , Na + , K+ , Ca 2+ and Mg 2+ build up. The total increase in mEq of each ion per kg of feed provided to the fish was highest for NO3 - , followed, in decreasing order, by Ca 2+ , H2PO4 – , K+ , Mg 2+ and SO4 2– . The total amount of feed required per mEq ranged from 1.61- 13.1 kg for the four most abundant ions (NO3 – , Ca 2+ , H2PO4 – and K+ ) at a density of 2 kg fish m–3 , suggesting that it would be rather easy to maintain small populations of fish to reduce the cost of hydroponic solution supplementation for strawberries.

This study uses PCR-derived marker systems to investigate the extent and distribution of genetic variability of 53 Garnacha accessions coming from Italy, France and Spain. The samples studied include 28 Italian accessions (named Tocai rosso in Vicenza area; Alicante in Sicily and Elba island; Gamay perugino in Perugia province; Cannonau in Sardinia), 19 Spanish accessions of different types (named Garnacha tinta, Garnacha blanca, Garnacha peluda, Garnacha roja, Garnacha erguida, Garnacha roya) and 6 French accessions (named Grenache and Grenache noir). In order to verify the varietal identity of the samples, analyses based on 14 simple sequence repeat (SSR) loci were performed. The presence of an additional allele at ISV3 locus (151 bp) was found in four Tocai rosso accessions and in a Sardinian Cannonau clone, that are, incidentally, chimeras. In addition to microsatellite analysis, intravarietal variability study was performed using AFLP, SAMPL and M-AFLP molecular markers. AFLPs could discriminate among several Garnacha samples; SAMPLs allowed distinguishing few genotypes on the basis of their geographic origin, whereas M-AFLPs revealed plant-specific markers, differentiating all accessions. Italian samples showed the greatest variability among themselves, especially on the basis of their different provenance, while Spanish samples were the most similar, in spite of their morphological diversity.

Th e CERES-Maize model is the most widely used maize (Zea mays L.) model and is a recognized reference for comparing new developments in maize growth, development, and yield simulation. Th e objective of this study was to present and evaluate CSMIXIM, a new maize simulation model for DSSAT version 4.5. Code from CSM-CERES-Maize, the modular version of the model, was modifi ed to include a number of model improvements. Model enhancements included the simulation of leaf area, C assimilation and partitioning, ear growth, kernel number, grain yield, and plant N acquisition and distribution. Th e addition of two genetic coeffi cients to simulate per-leaf foliar surface produced 32% smaller root mean square error (RMSE) values estimating leaf area index than did CSM-CERES. Grain yield and total shoot biomass were correctly simulated by both models. Carbon partitioning, however, showed diff erences. Th e CSM-IXIM model simulated leaf mass more accurately, reducing the CSM-CERES error by 44%, but overestimated stem mass, especially aft er stress, resulting in similar average RMSE values as CSM-CERES. Excessive N uptake aft er fertilization events as simulated by CSM-CERES was also corrected, reducing the error by 16%. Th e accuracy of N distribution to stems was improved by 68%. Th ese improvements in CSM-IXIM provided a stable basis for more precise simulation of maize canopy growth and yield and a framework for continuing future model developments

Los pasados días 4y 5 de junio tuvimos la oportunidad de probar en Lleida el modelo de pulverizador Mercury 3500 de Hardi, equipado con el nuevo sistema Iris-2. El objetivo de la prueba fue caracterizar no solo la máquina y sus especificaciones técnicas, sino la calidad de su trabajo, verificando asimismo el consumo de combustible.

The evapotranspiration (ET c) of a table grape vineyard (Vitis vinifera, cv. Red Globe) trained to a gable trellis under netting and black plastic mulching was determined under semiarid conditions in the central Ebro River Valley during 2007 and 2008. The netting was made of high-density polyethylene (pores of 12 mm2) and was placed just above the ground canopy about 2.2 m above soil surface. Black plastic mulching was used to minimize soil evaporation. The surface renewal method was used to obtain values of sensible heat flux (H) from high-frequency temperature readings. Later, latent heat flux (LE) values were obtained by solving the energy balance equation. For the May–October period, seasonal ET c was about 843 mm in 2007 and 787 mm in 2008. The experimental weekly crop coefficients (K cexp) fluctuated between 0.64 and 1.2. These values represent crop coefficients adjusted to take into account the reduction in ET c caused by the netting and the black plastic mulching. Average K cexp values during mid- and end-season stages were 0.79 and 0.98, respectively. End-season K cexp was higher due to combination of factors related to the precipitation and low ET o conditions that are typical in this region during fall. Estimated crop coefficients using the Allen et al. (1998) approach adjusting for the effects of the netting and black plastic mulching (K cFAO) showed a good agreement with the experimental K cexp values.

The evapotranspiration (ETc) of sprinkler-irrigated rice was determined for the semiarid conditions of NE Spain during 2001, 2002 and 2003. The surface renewal method, after calibration against the eddy covariance method, was used to obtain values of sensible heat flux (H) from high-frequency temperature readings. Latent heat flux values were obtained by solving the energy balance equation. Finally, lysimeter measurements were used to validate the evapotranspiration values obtained with the surface renewal method. Seasonal rice evapotranspiration was about 750–800 mm. Average daily ETc for mid-season (from 90 to 130 days after sowing) was 5.1, 4.5 and 6.1 mm day−1 for 2001, 2002 and 2003, respectively. The experimental weekly crop coefficients fluctuated in the range of 0.83–1.20 for 2001, 0.81–1.03 for 2002 and 0.84–1.15 for 2003. The total growing season was about 150–160 days. In average, the crop coefficients for the initial (Kcini), mid-season (Kcmid) and late-season stages (Kcend) were 0.92, 1.06 and 1.03, respectively, the length of these stages being about 55, 45 and 25 days, respectively.

This study states the potential trace elements (TE’s) content of red soils located at the centre region of Spain, characterized by low rainfall and slight acidity over prolonged weathering periods. For this purpose, three soil profiles from a catena were described, sampled and analyzed. The most notable characteristics are the low organic matter content and the predominantly acidic pH. Illite and kaolinite are the predominant clay minerals. The fertility of the soils is sufficient to provide most of the nutrients required, with very suitable potassium levels. The geochemical characters of this soil are: only few elements remain almost invariable across the profiles and over time, however the majority of them were directly linked with the clay content. These soils are characterized by relatively low levels of some trace elements such as Sr (64.35 mg?kg–1), Ba (303.67 mg?kg–1) and Sc (13.14 mg?kg–1); high levels of other trace elements such as V (103.92 mg?kg–1), Cr (79.9 mg?kg–1), Cu (15.18 mg?kg–1), Hf (10.26 mg?kg–1), Ni (38 mg?kg–1) and Zr (337 mg?kg–1); while the levels for rare earth elements (REE’s) such as La (48.36 mg?kg–1), Ce (95.07 mg?kg–1), Th (13.33 mg?kg–1) and Nd (42.65 mg?kg–1) are significantly high. The distribution of mayor and trace elements was directly re- lated to weathering processes, parent material and anthropogenic activities.

In this work we propose an image acquisition and processing methodology (framework) developed for performance in-field grapes and leaves detection and quantification, based on a six step methodology: 1) image segmentation through Fuzzy C-Means with Gustafson Kessel (FCM-GK) clustering; 2) obtaining of FCM-GK outputs (centroids) for acting as seeding for K-Means clustering; 3) Identification of the clusters generated by K-Means using a Support Vector Machine (SVM) classifier. 4) Performance of morphological operations over the grapes and leaves clusters in order to fill holes and to eliminate small pixels clusters; 5)Creation of a mosaic image by Scale-Invariant Feature Transform (SIFT) in order to avoid overlapping between images; 6) Calculation of the areas of leaves and grapes and finding of the centroids in the grape bunches. Image data are collected using a colour camera fixed to a mobile platform. This platform was developed to give a stabilized surface to guarantee that the images were acquired parallel to de vineyard rows. In this way, the platform avoids the distortion of the images that lead to poor estimation of the areas. Our preliminary results are promissory, although they still have shown that it is necessary to implement a camera stabilization system to avoid undesired camera movements, and also a parallel processing procedure in order to speed up the mosaicking process.
