We evaluated the effects of vincristine on the gastrointestinal (GI) motility of awake rats and correlated them with the course of vincristine-induced peripheral neuropathy. Vincristine or saline was injected into the tail vein of male Wistar rats (180-250 g) on alternate days: 50 µg/kg (5 doses, N = 10), 100 µg/kg (2, 3, 4 and 5 doses, N = 49) or 150 µg/kg (1, 2, or 5 doses, N = 37). Weight and stool output were measured daily for each animal. One day after completing the vincristine treatment, the animals were fasted for 24 h, gavage-fed with a test meal and sacrificed 10 min later to measure gastric emptying (GE), GI transit and colon weight. Sensory peripheral neuropathy was evaluated by hot plate testing. Chronic vincristine treatments with total cumulative doses of at least 250 µg/kg significantly decreased GE by 31-59% and GI transit by 55-93%. The effect of 5 doses of vincristine (150 µg/kg) on GE did not persist for more than 1 week. Colon weight increased after 2 and 5 doses of vincristine (150 µg/kg). Fecal output decreased up to 48 h after the fifth dose of vincristine (150 µg/kg). Vincristine decreased the heat pain threshold 1 day after 5 doses of 50-100 µg/kg or after 3-5 doses of 150 µg/kg. This effect lasted for at least 2 weeks after the fifth dose. Chronic intravenous vincristine treatment delayed GE and GI transit of liquid. This effect correlated with the peak increase in colon weight but not with the pain threshold changes.

The objective of this study was to investigate the occurrence of vancomycin-resistant Enterococcus (VRE) cross-transmission between two patient groups (long-term dialysis and kidney transplant patients). Molecular typing, by automated ribotyping with the RiboPrinter Microbial Characterization System (Qualicon, USA), was used to analyze VRE isolates from 31 fecal samples of 320 dialysis patients and 38 fecal samples of 280 kidney transplant patients. Clonal spread of E. faecalis and E. casseliflavus was observed intragroup, but not between the two groups of patients. In turn, transmission of E. gallinarum and E. faecium between the groups was suggested by the finding of vancomycin-resistant isolates belonging to the same ribogroup in both dialysis and transplant patients. The fact that these patients were colonized by VRE from the same ribogroup in the same health care facility provides evidence for cross-transmission and supports the adoption of stringent infection control measures to prevent dissemination of these bacteria.

We investigated whether fibrin glue (FG) could promote urethral sphincter restoration in muscle-derived stem cell (MDSC)-based injection therapies in a pudendal nerve-transected (PNT) rat, which was used as a stress urinary incontinence (SUI) model. MDSCs were purified from the gastrocnemius muscles of 4-week-old inbred female SPF Wistar rats and labeled with green fluorescent protein. Animals were divided into five groups (N = 15): sham (S), PNT (D), PNT+FG injection (F), PNT+MDSC injection (M), and PNT+MDSC+FG injection (FM). Each group was subdivided into 1- and 4-week groups. One and 4 weeks after injection into the proximal urethra, leak point pressure (LPP) was measured to assess urethral resistance function. Histology and immunohistochemistry were performed 4 weeks after injection. LPP was increased significantly in FM and M animals after implantation compared to group D (P < 0.01), but was not different from group S. LPP was slightly higher in the FM group than in the M group but there was no significant difference between them at different times. Histological and immunohistochemical examination demonstrated increased numbers of surviving MDSCs (109 ± 19 vs 82 ± 11/hpf, P = 0.026), increased muscle/collagen ratio (0.40 ± 0.02 vs 0.34 ± 0.02, P = 0.044), as well as increased microvessel density (16.9 ± 0.6 vs 14.1 ± 0.4/hpf, P = 0.001) at the injection sites in FM compared to M animals. Fibrin glue may potentially improve the action of transplanted MDSCs to restore the histology and function of the urethral sphincter in a SUI rat model. Injection of MDSCs with fibrin glue may provide a novel cellular therapy method for SUI.

Intestinal tuberculosis (ITB) and Crohn's disease (CD) are granulomatous disorders with similar clinical manifestations and pathological features that are often difficult to differentiate. This study evaluated the value of fluorescent quantitative polymerase chain reaction (FQ-PCR) for Mycobacterium tuberculosis (MTB) in fecal samples and biopsy specimens to differentiate ITB from CD. From June 2010 to March 2013, 86 consecutive patients (38 females and 48 males, median age 31.3 years) with provisional diagnoses of ITB and CD were recruited for the study. The patients' clinical, endoscopic, and histological features were monitored until the final definite diagnoses were made. DNA was extracted from 250 mg fecal samples and biopsy tissues from each patient. The extracted DNA was amplified using FQ-PCR for the specific MTB sequence. A total of 29 ITB cases and 36 CD cases were included in the analysis. Perianal disease and longitudinal ulcers were significantly more common in the CD patients (P<0.05), whereas night sweats, ascites, and circumferential ulcers were significantly more common in the ITB patients (P<0.05). Fecal FQ-PCR for MTB was positive in 24 (82.8%) ITB patients and 3 (8.3%) CD patients. Tissue PCR was positive for MTB in 16 (55.2%) ITB patients and 2 (5.6%) CD patients. Compared with tissue FQ-PCR, fecal FQ-PCR was more sensitive (X2=5.16, P=0.02). We conclude that FQ-PCR for MTB on fecal and tissue samples is a valuable assay for differentiating ITB from CD, and fecal FQ-PCR has greater sensitivity for ITB than tissue FQ-PCR.

Clostridium difficile is the most common cause of hospital-acquired diarrhea in patients treated with antibiotics, chemotherapeutic agents, and other drugs that alter the normal equilibrium of the intestinal flora. A better understanding of the risk factors for C. difficile-associated disease (CDAD) could be used to reduce the incidence of CDAD and the costs associated with its treatment. The aim of this study was to identify the risk factors for CDAD in a cohort of Chinese patients in a Beijing hospital. Medical charts of a total of 130 inpatients (62 males and 68 females) with hospital-acquired diarrhea (45 with CDAD; 85 without CDAD) were retrospectively reviewed. C. difficile toxins A and B were detected in fecal samples using enzyme-linked fluorescence assays. The drugs used by patients with and without CDAD before the onset of diarrhea were compared. Factors that differed significantly between the two groups by univariate analysis were analyzed by multivariate analysis using a logistic regression model. Multivariate analysis showed that cephalosporin treatment was associated with a significantly higher risk of CDAD in hospitalized patients, while treatment with glycopeptides was significantly associated with a reduction in CDAD (P<0.001 for cephalosporin; P=0.013 for glycopeptides). Our data confirmed previous findings that empirical treatment with cephalosporins is positively associated with CDAD compared to individuals using other CDAD-related drugs. Additionally, we showed that treatment with glycopeptides was negatively associated with CDAD, compared to individuals using other CDAD-related drugs.

Rotaviruses are the main cause of infantile acute diarrhea, and a monovalent (G1P[8]) vaccine against the virus was introduced into the Brazilian National Immunization Program for all infants in March 2006. The objectives of this study were to determine the rate and genotype distribution of rotavirus causing infantile diarrhea in the Triângulo Mineiro region of Brazil during 2011-2012 and to assess the impact of local vaccination. Fecal specimens were analyzed for detection and characterization of rotavirus using polyacrylamide gel electrophoresis, reverse transcription followed by polymerase chain reaction (PCR), and PCR-genotyping assays. Overall, rotavirus was diagnosed in 1.7% (6/348) of cases. Rotavirus positivity rates decreased 88% [95% confidence intervals (CI)=15.2, 98.3%; P=0.026] in 2011 and 78% (95%CI=30.6, 93.0%; P=0.007) in 2012 when compared with available data for baseline years (2005/2006) in Uberaba. In Uberlândia, reductions of 95.3% (95%CI=66.0, 99.4%; P=0.002) in 2011, and 94.2% (95%CI=56.4, 99.2%; P=0.004) in 2012 were also observed compared with data for 2008. The circulation of rotavirus G2P[4] strains decreased during the period under study, and strains related to the P[8] genotype reemerged in the region. This study showed a marked and sustained reduction of rotavirus-related cases, with a lack of rotavirus in the 2011 and 2012 seasons, suggesting a positive impact of the vaccination program.

Foram analisadas 96 amostras de carne-de-sol, das quais 48 foram coletadas em estabelecimentos inspecionados e 48 em estabelecimentos não-inspecionados. Com o objetivo de avaliar as condições higiênico-sanitárias desse produto, durante o processamento e a comercialização, pesquisou-se a incidência de bactérias mesófilas, bolores e leveduras, Staphylococcus aureus, coliformes fecais e Escherichia coli. Foram analisados ainda, alguns parâmetros físico-químicos como atividade de água e teor de cloreto de sódio. Nos dois grupos os resultados obtidos nas análises microbiológicas foram bastante elevados. Nas amostras provenientes de estabelecimentos inspecionados as médias da contagem total de bactérias mesófilas, bolores e leveduras, Staphylococcus spp e coliformes fecais foram 6,2Log UFC/g, 3,8Log UFC/g, 7,4Log UFC/g e 1,3Log NMP/g, respectivamente, e, nas comercializadas sem inspeção foram 7,4Log UFC/g, 4,4Log UFC/g, 6,8Log UFC/g e 2,6Log NMP/g, respectivamente. Esses resultados sugerem que houve falhas tanto no processamento como na manipulação desses produtos, sendo que a contaminação fecal foi bem mais elevada nas amostras coletadas nos estabelecimentos sem inspeção. A variação dos teores de cloreto de sódio e atividade de água das amostras indicou falta de padronização de qualidade da carne-de-sol, havendo a necessidade de se estabelecer parâmetros legais tanto microbiológicos como físico-químicos, capazes de oferecer ao consumidor um alimento seguro.

Quarenta e quatro amostras de água mineral envasadas, de diferentes marcas, foram examinadas quanto à contaminação por coliformes totais, E.coli, Pseudomonas aeruginosa, Enterococos, Clostrídios sulfito redutores a 46ºC, de acordo com o preconizado pela RDC 54/00 do Ministério da Saúde. Empregou-se a metodologia da membrana filtrante. Em nenhuma das amostras foi detectada a presença de clostrídios sulfito redutores a 46ºC, Pseudomonas aeruginosa e enterococos. A contaminação por coliformes totais e E.coli, detectada em 25% e 20,4% das amostras, respectivamente, sugere falhas higiênicas ao longo do processo e contaminação fecal recente. Tais amostras apresentam-se em desacordo com os padrões microbiológicos legais estabelecidos pela Legislação Brasileira. Devem-se adotar práticas higiênicas rigorosas em todo processamento, com o objetivo de obter-se produtos seguros, já que tratamentos não podem ser utilizados visando a redução/eliminação da contaminação.

The postharvest losses of horticultural products justify the use of preservation techniques. The processing not only adds value to the products, but also makes the products more convenient to the consumers. The objective of this research was to define the methodologies for the minimal processing of carrot and green pepper as to the type and intensity of the adoption of conservation techniques, and to monitor the products after processing through microbiological, physicochemical and nutritional analysis. The vegetables were washed and they were immersed in cold (7ºC) water with 100 mg L-1 free chlorine for sanitation, followed by centrifugation for 5 min. The product was put into BOPP/LDPE (biaxially orientated polypropylene/low-density polyethylene) plastic bags, which were sealed under atmospheric air, vacuum and modified atmosphere (2% O2, 10% CO2, 88% N2) and stored at 1ºC±1ºC. The approximate composition of the vegetables stayed stable during the storage period, in the three tested treatments. The contents of vitamin C for the samples of minimally processed carrot and green pepper did not present differences among treatments. The contents of beta-carotene decreased slightly during the storage period for the minimally processed carrot and green pepper. After processing, carrot and green pepper had psychrotrophic counts of 10²-10(5) and 10³-10(6) CFU g-1, respectively. Anaerobic mesophiles and total coliforms were found in green peppers, representing 1.6x10³ - 7.4x10(5) and <10.g-1 - 7.4x10(5), respectively. Total and fecal coliforms, anaerobic mesophiles and Salmonella were not found in carrots. Salmonella was not found in green pepper.

Coliformes fecais são definidos como coliformes capazes de fermentar a lactose com produção de gás em 48 h a 45ºC. Escherichia coli, juntamente com algumas cepas de Enterobacter e Klebsiella, podem apresentar essas características. Entretanto, apenas a presença de Escherichia coli em alimentos indica contaminação fecal por ser encontrada em grande quantidade no trato gastrointestinal do homem e animais de sangue quente, não sendo isolada normalmente em outros nichos. A denominação clássica de coliformes fecais foi alterada para coliformes a 45ºC, na Resolução nº 12 da Agência Nacional de Vigilância Sanitária. O objetivo deste trabalho foi avaliar a presença de E. coli entre os coliformes a 45ºC e comparar a eficiência das técnicas dos tubos múltiplos e Petrifilm EC na detecção de coliformes totais e E. coli em queijo Minas, lingüiça frescal, hortaliças e fubá. Petrifilm EC mostrou-se mais sensível na detecção de E. coli em relação ao método de tubos múltiplos, o qual apresentou resultados falso-negativos ou contagens subestimadas de E. coli, principalmente para amostras de alimentos de origem animal. Petrifilm EC foi o mais eficiente e prático, sendo um método alternativo adequado para a enumeração de coliformes totais e E. coli em alimentos.

The purpose of this research was to determine the best gas mixture in controlled atmosphere conditions to store fresh-cut ‘Pérola’ pineapple, particularly in relation to the maintenance of visual quality and reduction of microbial growth. After sanitation, fruit was manually peeled, sliced and dipped in 20 mg.L-1 NaOCl solution for 30 seconds. Then, the excess liquid was drained and the slices were placed in sealed glass jars connected to a flowboard installed in a cold room (5 ± 1 °C). Desired gas mixtures were supplied continuously for 12 days from cylinders connected to the flowboard. Controlled atmospheres of 2:5, 2:10, 2:15, 5:5, 5:10, 5:15, 8:5, 8:10 and 8:15 (O2:CO2, %) and air were used. The product was evaluated for pulp color, total and fecal coliforms, mesophilic and psychrotrophic aerobic, mold and yeast counts. Total and fecal coliforms were not detected. The fresh-cut ‘Pérola’ pineapple was not very sensitive to storage in controlled atmosphere, considering that the slices had little browning and were free of contamination, that would affect the food safety at the end of the storage period

A healthy life style and a balanced diet, associated with a high fruit and vegetable intake, are linked to good health and the prevention of diseases. Apples contain bioactive compounds that help in the prevention and control of hyperlipidemia. One of the Public Health concerns in Brazil is to reduce cardiovascular diseases. Therefore, the objective of this work was to analyze the chemical composition of Gala apples and to study the effect of their consumption on weight gain, food intake, serum levels of total cholesterol, HDL-C, LDL-C, triglycerides, hepatic cholesterol and fecal cholesterol in male albino Wistar rats fed a hypercholesterolemic diet. Six animals were utilized for each treatment (control, 5, 15 and 25% apple diet), during 30 and 60 days. This study showed that one apple (200 g) can provide 14.5% of recommended total fiber and 55% of recommended vitamin C, besides supplying considerable quantities of phenolic compounds (0.38 g.100 g-1) and tannins (0.16 g.100 g-1). All animals showed a non-significant reduction in their weight gain and food intake with an increase in the concentration of apple in the diets. At the end of 30 days, all of the diets provided a significant reduction in the levels of triglycerides compared to the control group. The 15 and 25% apple diets showed significant reductions in the serum levels of total cholesterol and LDL-C and an increase in the level of fecal cholesterol in relation to the control group. The 25% apple diet provided a significant reduction in the hepatic cholesterol levels compared to the control group. After 60 days, the serum levels of total cholesterol, LDL-C, HDL-C and triglycerides in rats fed with 5, 15 and 25% apple diets were similar to the control group. This probably happened due to a revertion of the process. These results show the importance of Gala apples in the control of hyperlipidemia in rats. A diet rich in vegetables and fruits, including apples, associated to a healthy life style, over time, could prevent or reduce the risk of heart disease.

O presente trabalho teve por objetivo avaliar o efeito fisiológico da Farinha de Semente de Abóbora (FSA) no trato intestinal de ratos Wistar machos recém desmamados, submetidos a dietas experimentais contendo Farinhas de Semente de Abóbora (FSAs) integral, peneirada e residual, por 10 dias. As dietas experimentais foram obtidas substituindo-se 30% do valor total de amido e dextrina, da ração controle, pela FSA correspondente. As farinhas e rações controle e experimentais foram caracterizadas quimicamente. Os animais divididos em grupos foram avaliados quanto ao crescimento, consumo, material fecal e pH cecal. As FSAs e rações experimentais apresentaram relevantes teores de fibras alimentares, proteínas e lipídios. Dietas contendo FSAs integral, peneirada e residual levaram a um ganho ponderal e ingestão similar entre todos os grupos. Em relação ao grupo controle, os animais experimentais submetidos a dietas contendo FSAs apresentaram maior volume e peso fecais (p < 0,05), além de significativa excreção de fibra insolúvel, em especial o grupo FSA residual. Apenas a FSA residual proporcionou queda (p < 0,05) do pH cecal. Os resultados obtidos indicam o potencial da FSA como fonte de fibra e sua capacidade em atuar aumentando peso e volume fecal, promovendo laxação.

A industrialização da maçã, em particular do suco, gera, no processamento, como principal resíduo ou subproduto, o bagaço, e como gerenciá-lo adequadamente está entre as prioridades das indústrias processadoras. A composição físico-química da farinha de bagaço de maçã apresentou, neste estudo, 43% de fibras em base seca. Os estudos sobre as fibras demonstram que elas exercem uma ação hipocolesterolêmica, reduzindo a digestão e a absorção dos lipídios dietéticos, aumentando a excreção fecal dos ácidos biliares e esteróis neutros, aumentando a produção de ácidos graxos de cadeia curta no cólon e diminuindo a porcentagem de ácidos biliares primários na bile. O objetivo deste estudo foi avaliar sensorialmente produtos panificados, adicionados de farinha de bagaço de maçã, e comprovar que a farinha de bagaço de maçã pode constituir fonte alternativa potencial de fibra alimentar para a formulação de alimentos panificados.

The effectiveness of different types of rice in relation to their ability to accelerate diarrhea recovering was evaluated in a rat model of osmotic diarrhea (OD). Animals (90-100 g) received protein free diet until reaching up to 20% weight loss, followed by lactose rich diet (LRD) to induce osmotic diarrhea. Rats presenting osmotic diarrhea were divided into 4 groups, which received lactose rich diet for 4 days from 8 am to 8 pm, and one of three experimental products containing 6% rice flour differing in amylose content during the night: high (HA), intermediate (IA), and low (LA). A group fed stock diet containing equivalent amount of lactose was taken as control and allowed to recover spontaneously. Amylose and viscosity (cp at 25 °C, 10 rpm) of final products were determined. Effectiveness was expressed as the ratio between percentages of normal vs. diarrheic stools during the treatment. Fecal characteristics in this rat model improved only as result of feeding high amylose content (HA) type of rice. In this experimental model of osmotic diarrhea in young rats, the antidiarrheal effects of rice were strongly dependent on the type of diet used and appear to be related to its amylose content.
