Le récepteur V2 (V2R) de la vasopressine est un récepteur couplé aux protéines G (RCPG), jouant un rôle fondamental dans le maintien de l’homéostasie hydrosodique. À l’instar de nombreux RCPGs, il est capable d’interagir avec plusieurs types de protéines G hétérotrimériques et possède des voies de signalisation peu explorées aux mécanismes mal compris. Ces voies non canoniques font l’objet des travaux exposés dans ce mémoire. Il s’agit d’explorer les caractéristiques et mécanismes de la signalisation de V2R via G12, et de la voie d’activation d’ERK 1/2 par transactivation du récepteur de l’insulin-like growth factor 1, IGF1R. Par des études de transfert d’énergie de résonance de bioluminescence (BRET), nous exposons la capacité de V2R à interagir avec la sous-unité Gα12 ainsi que la modulation de la conformation de l’hétérotrimère G12 par l’agoniste de V2R, l’arginine-vasopressine. Ces travaux dévoilent également la modulation de l’interaction entre Gα12 et son effecteur classique RhoA, suggérant un engagement de RhoA, ainsi que la potentialisation via Gα12 de la production d’AMP cyclique. À l’aide de diverses méthodes d’inhibition sélective, nos résultats précisent les mécanismes de la transactivation. Ils supportent notamment le rôle initiateur de l’activation de Src par V2R et l’absence d’implication des ligands connus d’IGF1R dans la transactivation. La métalloprotéase MMP 3 apparaît par ailleurs comme un bon candidat pour réguler la transactivation. Ce projet met en lumière des modes de signalisation peu explorés de V2R, dont l’implication physiologique et physiopathologique pourrait s’avérer significative, au-delà d’un apport fondamental dans la compréhension de la signalisation des RCPGs.

Les leucémies myéloïdes aigües résultent d’un dérèglement du processus de l’hématopoïèse et regroupent des maladies hétérogènes qui présentent des profils cliniques et génétiques variés. La compréhension des processus cellulaires responsables de l’initiation et du maintien de ces cancers permettrait de développer des outils thérapeutiques efficaces et ciblés. Au cours des dernières années, une quantité croissante d’anomalies génétiques reliées au développement de leucémies ont été corrélées à une expression anormale des gènes HOX et de leurs cofacteurs MEIS et PBX. Des modèles expérimentaux murins ont confirmé le rôle direct joué par ces protéines dans le développement de leucémies. En effet, la protéine MEIS1 collabore avec HOXA9 dans la leucémogenèse et requiert pour ce faire trois domaines distincts. Deux de ces domaines sont conservés chez PREP1, un membre de la même classe d’homéoprotéine que MEIS1. En utilisant une approche de gain-de-fonction, j’ai confirmé l’importance du rôle joué par le domaine C-terminal de MEIS1 dans l’accélération des leucémies induites par HOXA9. J’ai également montré que l’activité de ce domaine était corrélée avec une signature transcriptionnelle associée à la prolifération cellulaire. J’ai ensuite réalisé un criblage à haut débit afin d’identifier des antagonistes de l’interaction MEIS-PBX, également essentielle à l’accélération des leucémies HOX. À cette fin, j’ai développé un essai de transfert d’énergie de résonance de bioluminescence (BRET) permettant de détecter la dimérisation MEIS-PBX dans les cellules vivantes. Plus de 115 000 composés chimiques ont été testés et suite à une confirmation par un essai orthogonal, une vingtaine de molécules ont été identifiées comme inhibiteurs potentiels. Ces composés pourront être rapidement testés sur la prolifération de cellules leucémiques primaires dans un contexte d’étude préclinique. Finalement, deux approches protéomiques complémentaires ont permis d’identifier des partenaires potentiels de MEIS1 et PREP1. La catégorisation fonctionnelle de ces candidats suggère un nouveau rôle pour ces homéoprotéines dans l’épissage de l’ARN et dans la reconnaissance de l’ADN méthylé.

CXCR4, a chemokine receptor involved in metastasis and homing of hematopoietic stem cells, signals through two major pathways: Gαi and β-arrestin2. β-arrestin2 terminates G-protein signaling and targets the receptor to endocytosis. This project proposed to study the effect of a previously described set of CXCR4 mutants on both these signaling pathways, as well as their localization. These mutants were assayed by different Bioluminescence Resonance Energy Transfer (BRET) systems. Using these systems, we confirmed that N119S is a constitutively active mutant (CAM), spontaneously activating Gαi. As well, we found that R134A is a constitutively inactive mutant (CIM), devoided of G-protein signaling, but spontaneously recruiting β-arrestin2. In addition, we studied the dependency of β-arrestin2 recruitment on the Gαi activity. By targeting R134A and N119S with pertussis toxin, an inhibitor of the Gαi activation, we showed efficient blocking of the Gαi pathway, while maintaining the constitutive recruitment of β-arrestin2. This demonstrated that for CXCR4, β-arrestin2 recruitment is independent of the Gαi pathway. Finally, two synthetic ligands of CXCR4, AMD3100 and TC14012 were tested for their ability to recruit β-arrestin2. AMD3100 is a clinically approved drug used for stem cell transplantation, with considerable side effects. We found it to be an antagonist on both Gαi and β-arrestin2 recruitment. On the other hand, TC14012 was found to be an inverse agonist on Gαi and an antagonist on β-arrestin2 recruitment. Based on this finding, it would be preferable to use of TC14012 as it will further reduce any basal Gαi activity, without affecting β-arrestin2 recruitment. These results support the development of TC14012 for stem cell mobilization trials.

Les opioïdes sont les analgésiques les plus puissants mais leur utilisation prolongée peut entraîner le développement d’une tolérance analgésique. La tolérance serait en partie associée à l’inhibition prolongée de l’adénosine monophosphate cyclique (AMPc) entraînant des changements compensatoires dans la voie de l’adénylate cyclase. Pour cette étude, nous avons eu recours à un biosenseur basée sur la technologie de Bioluminescence Resonnance Energy Transfer (BRET) et qui fournit des mesures de l’AMPc en fonction du temps réel. Durant les 15 premières minutes de stimulation, la réponse de l’AMPc est bi-phasique. Cette progression de la réponse à l’AMPc n’est pas la même pour tous les ligands. Par exemple, la deltorphine II qui induit l’internalisation du récepteur opioïde delta (DOR) affiche une baisse de l’inhibition de l’AMPc. À l’inverse la morphine qui n’induit pas l’internalisation du DOR affiche une réponse stable à l’inhibition de l’AMPc. Ainsi le profil d’internalisation permet de prédire la progression de l’inhibition de l’AMPc à court terme (15 minutes). Nous avons aussi mesuré la réponse à l’AMPc durant 30, 60 et 120 min, étant donné qu’un traitement chronique aux opioïdes induit une tolérance analgésique. Selon les résultats obtenus, le profil d’internalisation du DOR induits par les ligands ne permet pas d’expliquer l’inhibition persistante de l’AMPc.

Le récepteur de chimiokine CXCR3 est un récepteur couplé à la protéine G (RCPG) exprimé, entre autre, sur les cellules T activées lors d’une réponse immune. CXCR3 est activé par trois ligands inductibles par l’interféron-γ (CXCL9, 10, 11) et, plus récemment, il a été découvert que CXCL4 liait CXCR3. Nous savons que CXCR3 joue un rôle dans la chimiotaxie des leucocytes, mais peu d’attention a été portée sur la signalisation biaisée induite par ces quatre ligands. Alors que l’homodimérisation entre récepteurs de chimiokine est un concept grandement observé, l’hétéromérisation entre deux récepteurs reste un domaine de recherche active. La signalisation biaisée et l’hétéromérisation ont été testées grâce à la technique de bioluminescene resonance energy transfer (BRET) dans des cellules HEK293E. Nous présentons une caractérisation pharmacologique des quatre ligands de CXCR3 et démontrons l’hétéromérisation de CXCR3 avec CXCR4 et avec CXCR7. Nos résultats suggèrent que les ligands de CXCR3 n’agissent pas de manière redondante.

Laser induced photoacoustic (PA) technique is used in the study of photostability of polymethyl methacrylate (PMMA) films doped with Rhodamine 6G -Rhodamine B dye system. Energy transfer from a donor molecule to an acceptor molecule in a dye mixture affects the output of the dye system. Details of investigations on the role of laser power, modulation frequency and the irradiation wavelength on the photosensitivity of the dye mixture doped PMMA films are presented.

The present work aims at deciphering the processes that control the nutrient distribution along the EEZ (Exclusive Economic Zone of India) of the west coast of India and to bring out its linkage with primary and secondary productivity. This work assume utmost importance as very few studies have hitherto focused entirely on the EEZ of the west coast of India to address the biochemical responses brought about by monsoons. The present study examines the seasonal variations in physicochemical parameters and associated primary biological responses along the west coast of India. This study targets to measure and understand the shelf ocean exchange in a typical coastal upwelling region of the southeast Arabian Sea, and the influence of convective mixing along the northern part of the west coast of India. The study focuses more directly on coastal upwelling along the southwest coast of India, within the EEZ. The effects of coastal upwelling, eddy formation and the offshore advection are apparent in the present investigation. This has consequences to fisheries and climate, in energy transfer to the food chain and the increased sequestering of carbon in the ocean. The study also focuses on the Oxygen Minimum Zone (OMZ) and dentrification observed along the EEZ of the west coast of India on a seasonal scale. In the study, an attempt is also made to demarcate the geographical boundaries of the denitrification zone in the EEZ of India and on the nature and magnitude of these variations, on a seasonal and inter annual scales

In this paper, the fluorescence behaviour of nano colloids of ZnO has been studied as a function of the excitation wavelength. We have found that excitation at the tail of the absorption band gives rise to an emission that shifts with the change of the excitation wavelength. The excitation wavelength dependent shift of the fluorescence maximum is measured to be between 60 and 100 nm. This kind of excitation wavelength dependent fluorescence behaviour, which may appear to be in violation of Kasha’s rule of excitation wavelength independence of the emission spectrum, has been observed for nano ZnO colloids prepared by two different chemical routes and different capping agents. It is shown that the existence of a distribution of energetically different molecules in the ground state coupled with a low rate of the excited state relaxation processes, namely, solvation and energy transfer, are responsible for the excitation wavelength dependent fluorescence behaviour of the systems.

The subject of Photonics is concerned with the generation,control and utilization of photons for performing a variety of tasks.It came to existence as a consequence of the harmonious fusion of optical methods with electronic technology.Wide spread use of laser based methods in electronics is slowly replacing elecrtons with photons in the field of Communication,Control and Computing .Therefore,there is a need to promote the R & D activities in the area of Photonics and to generate well trained manpower in laser related fields.Development and characterization of photonic materials is an important subject of research in the field of Photonics.Optical and thermal characterization of photonic materials using thermal lens technique is a PhD thesis in the field of Photonics in which the author describes how thermal lens effect can be used to characterize themal and optical properties of photonic materials.Plausibility of thermal lens based logic gates is also presented in this thesis.

A sensitive method based on the principle of photothermal phenomena to study the energy transfer processes in organic dye mixtures is presented. A dual beam thermal lens method can be very effectively used as an alternate technique to determine the molecular distance between donor and acceptor in fluorescein–rhodamine B mixture using optical parametric oscillator.

Two-photon excited (TPE) side illumination fluorescence studies in a Rh6G-RhB dye mixture doped polymer optical fiber (POF) and the effect of energy transfer on the attenuation coefficient is reported. The dye doped POF is pumped sideways using 800 nm, 70 fs laser pulses from a Ti:sapphire laser, and the TPE fluorescence emission is collected from the end of the fiber for different propagation distances. The fluorescence intensity of RhB doped POF is enhanced in the presence of Rh6G as a result of energy transfer from Rh6G to RhB. Because of the reabsorption and reemission process in dye molecules, an effective energy transfer is observed from the shorter wavelength part of the fluorescence spectrum to the longer wavelength part as the propagation distance is increased in dye doped POF. An energy transfer coefficient is found to be higher at shorter propagation distances compared to longer distances. A TPE fluorescence signal is used to characterize the optical attenuation coefficient in dye doped POF. The attenuation coefficient decreases at longer propagation distances due to the reabsorption and reemission process taking place within the dye doped fiber as the propagation distance is increased.

Multimode laser emission is observed in a polymer optical fiber doped with a mixture of Rhodamine 6G (Rh 6G) and Rhodamine B (Rh B) dyes. Tuning of laser emission is achieved by using the mixture of dyes due to the energy transfer occurring from donor molecule (Rh 6G) to acceptor molecule (Rh B). The dye doped poly(methyl methacrylate)-based polymer optical fiber is pumped axially at one end of the fiber using a 532 nm pulsed laser beam from a Nd:YAG laser and the fluorescence emission is collected from the other end. At low pump energy levels, fluorescence emission is observed. When the energy is increased beyond a threshold value, laser emission occurs with a multimode structure. The optical feedback for the gain medium is provided by the cylindrical surface of the optical fiber, which acts as a cavity. This fact is confirmed by the mode spacing dependence on the diameter of the fiber.

The photosensitivity of dye mixture-doped polymethyl methacrylate (PMMA) films are investigated as a function of laser power, concentration of the dyes, modulation frequency and the irradiation wavelength. Energy transfer from a donor molecule to an acceptor molecule affects the emission output of the dye mixture system. Photosensitivity is found to change with changes in donor–acceptor concentrations. PMMA samples doped with the dye mixture are found to be more photosensitive when the dyes are mixed in the same proportion.

Dynamics of Nd:YAG laser with intracavity KTP crystal operating in two parallel polarized modes is investigated analytically and numerically. System equilibrium points were found out and the stability of each of them was checked using Routh–Hurwitz criteria and also by calculating the eigen values of the Jacobian. It is found that the system possesses three equilibrium points for (Ij, Gj), where j = 1, 2. One of these equilibrium points undergoes Hopf bifurcation in output dynamics as the control parameter is increased. The other two remain unstable throughout the entire region of the parameter space. Our numerical analysis of the Hopf bifurcation phenomena is found to be in good agreement with the analytical results. Nature of energy transfer between the two modes is also studied numerically.
