Ziel der Arbeit war es, die physiologische Funktion von 2-Adaptin zu charakterisieren. 2 Adaptin wurde 1998 erstmals von Takatsu et al. und Lewin et al. als mögliches Mitglied der Clathrin-Adapter-Proteinfamilie beschrieben. Seine genaue physiologische Funktion ist aber bis heute noch unklar. Bisherige Ergebnisse deuten darauf hin, das 2-Adaptin unabhängig von den AP-Komplexen wirkt. rnIn der HBV-Morphogenese ist eine spezielle Funktion von 2-Adaptin bekannt, da es dort nach seiner Ubiquitinierung durch Nedd4 als Adapter zwischen dem HBV L- und Core-Protein fungiert und Änderungen in der 2 Konzentration die HBV-Freisetzung blockieren.rn2-Adaptin besitzt neben den für die Clathrin-Adapter Proteine typischen Clathrin-bindenden Eigenschaften auch die Fähigkeit, Ubiquitin über sein UIM zu binden. Darüberhinaus wird 2-Adaptin durch seine Interaktion mit der Ubiquitin-Ligase Nedd4 selbst ubiquitiniert. Damit besitzt 2-Adaptin typische Eigenschaften eines Ubiquitin-Adapters. 2-Adaptin ist an MVBs lokalisiert und Abweichungen in der 2 Konzentration verändern die MVB-Morphologie. Zudem führt die Überexpression von 2-Adaptin zur Blockade der Freisetzung retroviraler VLPs und die 2 Depletion blockiert den lysosomalen Abbau von EGF, einem Substrat des endo-lysosomalen Proteintransports. Dies alles deutet auf eine mögliche Funktion von 2-Adaptin in diesem Transportsystem hin, welche in dieser Arbeit näher untersucht wurde.rnEs konnte gezeigt werden, dass die Depletion von 2-Adaptin den Abbau von endogenen (z.B. EGF, ubiquitinierte Proteine) und exogenen (z.B. das retrovirale MLV.gag-Polyprotein) Substraten des endo-lysosomalen Weges inhibiert, während sie bei 2 Überexpression verstärkt abgebaut werden. Alle bisher identifizierten „Substrate“ von 2 Adaptin, also Proteine, die durch überschüssiges 2-Adaptin abgebaut werden, besitzen eine Verbindung zum endo-lysosomalen System und / oder zur Ubiquitin-Maschinerie der Zelle. Weitere Hinweise auf eine Rolle von 2 Adaptin im MVB-Weg lieferte die Identifikation von Vps28 und Chmp2A als spezifische Interaktionspartner von 2-Adaptin. Über Vps28 erhält -Adaptin direkten Zugang zum ESCRT-I- und über Chmp2A zum ESCRT-III-Komplex. rnZudem konnte neben dem UIM eine PH-Domäne in 2-Adaptin als wichtige funktionelle Domäne identifiziert werden. Sie stellt das Modul für die Interaktion mit Rab7 dar, welche erstmals gezeigt werden konnte. Auch die Interaktion mit Rab7 deutet auf eine Rolle von 2 Adaptin im endo-lysosomalen Transportsystem hin, da Rab7 an späten Endosomen lokalisiert ist und u.a. die Fusion der MVBs mit den Lysosomen vermittelt. Da die Auswirkungen der Rab7-Überexpression und Depletion auf MLV.gag denen der 2 Überexpression bzw. Depletion entsprechen, liegt die Vermutung nahe, dass 2-Adaptin an einem ähnlich späten Schritt im endo-lysosomalen Transportsystem wirkt wie Rab7. Jedoch blockiert überschüssiges 2 Adaptin die ESCRT-abhängige VLP-Ausschleusung an der Plasmamembran und fungiert daher möglicherweise als negativer Regulator der ESCRT-Kaskade. Da die Überexpression von -Adaptin aber gleichzeitig zum vermehrten lysosomalen Abbau führt, ist eine Funktion von 2-Adaptin bei der MVB-Lysosomen-Fusion wenig wahrscheinlich. Einer solchen Funktion widerspricht auch, dass die intrazelluläre Konzentration von Rab7 und Vps28 durch überschüssiges 2-Adaptin reduziert werden. rnAls dritte funktionell wichtige Domäne in 2-Adaptin konnte ein LIR-Motiv identifiziert werden, über welches -Adaptin mit dem Autophagie-Markerprotein LC3 interagieren kann. Die Interaktion mit LC3, und damit die Verbindung zur Autophagie-Machinerie, liefert eine mögliche Erklärung für den vermehrten Abbau bei 2-Überexpression und den Abbau von Proteinen auf der MVB-Oberfläche. Dabei induziert 2-Adaptin nicht die Autophagie per se, sondern scheint als Autophagie-Adapter zu wirken, der seine Substrate, z.B. MVBs, selektiv dem Abbau durch Autophagie zuführt. rnrnEine mögliche Rolle von 2-Adaptin im zum Lysosom hin gerichteten zellulären Transport konnte bestätigt werden, wobei 2-Adaptin dabei verschiedene Funktionen übernimmt: rn als Ubiquitin-Adapter im endo-lysosomalen System, rn als negativer Regulator der ESCRT-Kaskadern und / oder als Autophagie-Adapter.rn

Das aus wissenschaftlicher und ökonomischer Sicht wichtigste Pflanzenpathogen M. oryzae entwickelte im Laufe der Evolution konservierte aber auch einzigartige Mechanismen zur Signaltransduktion. Das Erforschen dieser Mechanismen und Prozesse ist essenziell für das Verständnis von Differenzierungsprozessen bei der Pathogen-Wirt-Interaktion.rnIm ersten Teil der vorliegenden Arbeit wurde der Signalweg zur Osmoregulation, der „High Osmolarity Glycerol“ (HOG)-Signalweg, erstmals anhand physiologischer Experimente in entsprechenden Mutantenstämmen in M. oryzae untersucht. Dabei konnten klare Unter-schiede zum HOG-Signalweg von S. cerevisiae aufgezeigt werden. rnDas in M. oryzae bisher noch nicht beschriebene Gen MoYPD1, welches das Phosphotransferprotein MoYpd1p kodiert, wurde erfolgreich inaktiviert. Diese Inaktivierung ist in S. cerevisiae und vielen anderen Pilzen letal und resultierte bei M. oryzae in einer apathoge¬nen Albinomutante, deren Konidiogenese gestört ist. Insbesondere die Funktion des Phosphotransferproteins MoYpd1p, sowohl im Phosphorelaysystem des HOG-Signal¬wegs als auch im Wirkmechanismus des Fungizids Fludioxonil, konnte eindeutig mittels Y2H- und Western Blot-Analysen nachgewiesen werden.rnEs wurden entscheidende Fortschritte für das Verständnis des Aufbaus und der Funktion des HOG-Signalwegs sowohl als physiologisches Regulationssystem für Umweltreize als auch als Fungizidtarget im Pflanzenschutz erzielt. Dabei konnte gezeigt werden, dass die Zweikompo-nenten-Hybrid-Histidinkinase (HIK) MoSln1p als Signalsensor für Salzstress und MoHik1p als Signalsensor für Zuckerstress fungiert. Die Beteiligung der Histidinkinasen MoHik5p und MoHik9p als Sensorproteine für Hypoxie im HOG-Signalweg ist durchaus denk¬bar und wurde durch erste Ergebnisse bekräftigt. rnSo konnte der HOG-Signalweg in mehreren Modellen dargestellt werden. Die Modelle der Signalerkennung und –transduktion von osmotischem Stress, von Hypoxie und der Wirkmecha¬nismus von Fludioxonil wurden erstmals in diesem Umfang für M. oryzae ausgearbei¬tet.rnDer zweite Teil dieser Arbeit repräsentiert die erste umfassende Untersuchung aller zehn HIK-codierender Gensequenzen, die im Genom von M. oryzae identifiziert werden konnten. Diese Signalproteine waren bisher noch nicht Gegenstand wissenschaftlicher Studien. Die Untersuchung beginnt mit einer phylogenetischen Einordnung aller untersuchten Proteinsequen¬zen in die verschiedenen Gruppen von Histidinkinasen in Pilzen. Eine ausführli-che phänotypische Charakterisierung aller HIK-codierender Gene folgt und wurde anhand von Mutanten durchgeführt, in denen diese Gene einzeln inaktiviert wurden.rnDie Beteiligung von MoHik5p und MoHik9p als mögliche Sauerstoffsensoren im HOG-Signal-weg konnte dokumentiert werden und die anschließenden Western Blot-Analysen bestätig¬ten erstmals die Aktivierung des HOG-Signalwegs bei hypoxieähnlichen Zuständen.rnDes Weiteren wurden mit MoHik5p und MoHik8p zwei neue Pathogenitätsfaktoren in M. oryzae identifiziert. Die apathogenen Mutantenstämme ΔMohik5 und ΔMohik8 sind in der Konidiogenese gestört und nicht in der Lage Appressorien zu differenzieren. Der Einsatz dieser Proteine als Fungizidtarget im protektiven Pflanzenschutz in der Zukunft ist somit denk-bar.rn

In holometabolous insects such as Drosophila melanogaster, neuroblasts produce an initial population of diverse neurons during embryogenesis and a much larger set of adult-specific neurons during larval life. In the ventral CNS, many of these secondary neuronal lineages differ significantly from one body segment to another, suggesting a role for anteroposterior patterning genes. Here we systematically characterize the expression pattern and function of the Hox gene Ultrabithorax (Ubx) in all 25 postembryonic lineages. We find that Ubx is expressed in a segment-, lineage-, and hemilineage-specific manner in the thoracic and anterior abdominal segments. When Ubx is removed from neuroblasts via mitotic recombination, neurons in these segments exhibit the morphologies and survival patterns of their anterior thoracic counterparts. Conversely, when Ubx is ectopically expressed in anterior thoracic segments, neurons exhibit complementary posterior transformation phenotypes. Our findings demonstrate that Ubx plays a critical role in conferring segment-appropriate morphology and survival on individual neurons in the adult-specific ventral CNS. Moreover, while always conferring spatial identity in some sense, Ubx has been co-opted during evolution for distinct and even opposite functions in different neuronal hemilineages.

Scarring or detachment of the hip abductors, particularly of the gluteus medius, from their insertion may lead to severe abductor weakness, recurrent dislocations, pain, and diminished quality of life. We performed a retrospective study to evaluate whether vastus lateralis shift is associated with satisfactory results and low rate of complications. Eleven adults underwent vastus lateralis shift to bridge a well-documented abductor muscles' insertion defect. Preoperative and postoperative hip functions were assessed applying the Merle d'Aubigne score, British Medical Council scale, and Visual Analog Scale. Significant postoperative improvement was noted in mean Merle d'Aubigne score, gluteus medius muscle force, and quality of life. Vastus lateralis shift represents a viable treatment option for hip abductor deficiency, significantly improving abductor strength and overall quality of life.

The aim of this study is to develop a new simple method for analyzing one-dimensional transcranial magnetic stimulation (TMS) mapping studies in humans. Motor evoked potentials (MEP) were recorded from the abductor pollicis brevis (APB) muscle during stimulation at nine different positions on the scalp along a line passing through the APB hot spot and the vertex. Non-linear curve fitting according to the Levenberg-Marquardt algorithm was performed on the averaged amplitude values obtained at all points to find the best-fitting symmetrical and asymmetrical peak functions. Several peak functions could be fitted to the experimental data. Across all subjects, a symmetric, bell-shaped curve, the complementary error function (erfc) gave the best results. This function is characterized by three parameters giving its amplitude, position, and width. None of the mathematical functions tested with less or more than three parameters fitted better. The amplitude and position parameters of the erfc were highly correlated with the amplitude at the hot spot and with the location of the center of gravity of the TMS curve. In conclusion, non-linear curve fitting is an accurate method for the mathematical characterization of one-dimensional TMS curves. This is the first method that provides information on amplitude, position and width simultaneously.

L-amino acid oxidases are widely found in snake venoms and are thought to contribute to the toxicity upon envenomation. The mechanism of these toxic effects and whether they result from the enzymatic activity are still uncertain although many papers describing the biological and pharmacological effects of L-amino acid oxidases have appeared recently, which provide more information about their action on platelets, induction of apoptosis, haemorrhagic effects, and cytotoxicity. This review summarizes the physiochemical properties, structural characteristics and various biological functions of snake venom L-amino acid oxidases (SV-LAAOs). In addition, the putative mechanisms of SV-LAAO-induced platelet aggregation and apoptosis of cells are discussed in more detail.

The transmembrane ligand ephrinB2 and its cognate Eph receptor tyrosine kinases are important regulators of vascular morphogenesis. EphrinB2 may have an active signaling role, resulting in bi-directional signal transduction downstream of both ephrinB2 and Eph receptors. To separate the ligand and receptor-like functions of ephrinB2 in mice, we replaced the endogenous gene by cDNAs encoding either carboxyterminally truncated (ephrinB2(DeltaC)) or, as a control, full-length ligand (ephrinB2(WT)). While homozygous ephrinB2(WT/WT) animals were viable and fertile, loss of the ephrinB2 cytoplasmic domain resulted in midgestation lethality similar to ephrinB2 null mutants (ephrinB2(KO)). The truncated ligand was sufficient to restore guidance of migrating cranial neural crest cells, but ephrinB2(DeltaC/DeltaC) embryos showed defects in vasculogenesis and angiogenesis very similar to those observed in ephrinB2(KO/KO) animals. Our results indicate distinct requirements of functions mediated by the ephrinB carboxyterminus for developmental processes in the vertebrate embryo.

The water spider Argyroneta aquatica (Clerck) is the only spider that spends its whole life under water. Water spiders keep an air bubble around their body for breathing and build under-water air bells, which they use for shelter and raising offspring, digesting and consuming prey, moulting, depositing eggs and sperm, and copulating. It is unclear whether these bells are an important oxygen reservoir for breathing under water, or whether they serve mainly to create water-free space for feeding and reproduction. In this study, we manipulated the composition of the gas inside the bell of female water spiders to test whether they monitor the quality of this gas, and replenish oxygen if required. We exchanged the entire gas in the bell either with pure O(2), pure CO(2), or with ambient air as control, and monitored behavioural responses. The test spiders surfaced and replenished air more often in the CO(2) treatment than in the O(2) treatment, and they increased bell building behaviour. In addition to active oxygen regulation, they monitored and adjusted the bells by adding silk. These results show that water spiders use the air bell as an oxygen reservoir, and that it functions as an external lung, which renders it essential for living under water permanently. A. aquatica is the only animal that collects, transports, and stores air, and monitors its property for breathing, which is an adaptive response of a terrestrial animal to the colonization of an aquatic habitat. Neuropeptide Y (NPY), which is found in high concentrations in several regions of the brain including nuclei of the brain stem and in nerve fibers surrounding cerebral vessels, has been proposed to play a role in regulating cerebral blood flow (CBF) and systemic vegetative functions. Since CBF is altered during meningitis, we examined whether NPY concentrations changed in various regions of the rabbit brain in response to experimental pneumococcal meningitis. Changes were most pronounced in the medulla, where NPY concentration increased threefold after 48 h of infection. Concomitantly, there was an increase in NPY immunoreactive fibers surrounding small vessels in the dorsolateral medulla, especially in the nucleus tractus solitarius. These results suggest that NPY may play a role in inducing some of the hemodynamic changes seen during pneumococcal meningitis.

The problem of re-sampling spatially distributed data organized into regular or irregular grids to finer or coarser resolution is a common task in data processing. This procedure is known as 'gridding' or 're-binning'. Depending on the quantity the data represents, the gridding-algorithm has to meet different requirements. For example, histogrammed physical quantities such as mass or energy have to be re-binned in order to conserve the overall integral. Moreover, if the quantity is positive definite, negative sampling values should be avoided. The gridding process requires a re-distribution of the original data set to a user-requested grid according to a distribution function. The distribution function can be determined on the basis of the given data by interpolation methods. In general, accurate interpolation with respect to multiple boundary conditions of heavily fluctuating data requires polynomial interpolation functions of second or even higher order. However, this may result in unrealistic deviations (overshoots or undershoots) of the interpolation function from the data. Accordingly, the re-sampled data may overestimate or underestimate the given data by a significant amount. The gridding-algorithm presented in this work was developed in order to overcome these problems. Instead of a straightforward interpolation of the given data using high-order polynomials, a parametrized Hermitian interpolation curve was used to approximate the integrated data set. A single parameter is determined by which the user can control the behavior of the interpolation function, i.e. the amount of overshoot and undershoot. Furthermore, it is shown how the algorithm can be extended to multidimensional grids. The algorithm was compared to commonly used gridding-algorithms using linear and cubic interpolation functions. It is shown that such interpolation functions may overestimate or underestimate the source data by about 10-20%, while the new algorithm can be tuned to significantly reduce these interpolation errors. The accuracy of the new algorithm was tested on a series of x-ray CT-images (head and neck, lung, pelvis). The new algorithm significantly improves the accuracy of the sampled images in terms of the mean square error and a quality index introduced by Wang and Bovik (2002 IEEE Signal Process. Ethanol-gasoline fuel blends are increasingly being used in spark ignition (SI) engines due to continued growth in renewable fuels as part of a growing renewable portfolio standard (RPS). This leads to the need for a simple and accurate ethanol-gasoline blends combustion model that is applicable to one-dimensional engine simulation. A parametric combustion model has been developed, integrated into an engine simulation tool, and validated using SI engine experimental data. The parametric combustion model was built inside a user compound in GT-Power. In this model, selected burn durations were computed using correlations as functions of physically based non-dimensional groups that have been developed using the experimental engine database over a wide range of ethanol-gasoline blends, engine geometries, and operating conditions. A coefficient of variance (COV) of gross indicated mean effective pressure (IMEP) correlation was also added to the parametric combustion model. This correlation enables the cycle combustion variation modeling as a function of engine geometry and operating conditions. The computed burn durations were then used to fit single and double Wiebe functions. The single-Wiebe parametric combustion compound used the least squares method to compute the single-Wiebe parameters, while the double-Wiebe parametric combustion compound used an analytical solution to compute the double-Wiebe parameters. These compounds were then integrated into the engine model in GT-Power through the multi-Wiebe combustion template in which the values of Wiebe parameters (single-Wiebe or double-Wiebe) were sensed via RLT-dependence. The parametric combustion models were validated by overlaying the simulated pressure trace from GT-Power on to experimentally measured pressure traces. A thermodynamic engine model was also developed to study the effect of fuel blends, engine geometries and operating conditions on both the burn durations and COV of gross IMEP simulation results.

Despite widespread use of species-area relationships (SARs), dispute remains over the most representative SAR model. Using data of small-scale SARs of Estonian dry grassland communities, we address three questions: (1) Which model describes these SARs best when known artifacts are excluded? (2) How do deviating sampling procedures (marginal instead of central position of the smaller plots in relation to the largest plot; single values instead of average values; randomly located subplots instead of nested subplots) influence the properties of the SARs? (3) Are those effects likely to bias the selection of the best model? Our general dataset consisted of 16 series of nested-plots (1 cm(2)-100 m(2), any-part system), each of which comprised five series of subplots located in the four corners and the centre of the 100-m(2) plot. Data for the three pairs of compared sampling designs were generated from this dataset by subsampling. Five function types (power, quadratic power, logarithmic, Michaelis-Menten, Lomolino) were fitted with non-linear regression. In some of the communities, we found extremely high species densities (including bryophytes and lichens), namely up to eight species in 1 cm(2) and up to 140 species in 100 m(2), which appear to be the highest documented values on these scales. For SARs constructed from nested-plot average-value data, the regular power function generally was the best model, closely followed by the quadratic power function, while the logarithmic and Michaelis-Menten functions performed poorly throughout. However, the relative fit of the latter two models increased significantly relative to the respective best model when the single-value or random-sampling method was applied, however, the power function normally remained far superior. These results confirm the hypothesis that both single-value and random-sampling approaches cause artifacts by increasing stochasticity in the data, which can lead to the selection of inappropriate models.

Campylobacter, a major zoonotic pathogen, displays seasonality in poultry and in humans. In order to identify temporal patterns in the prevalence of thermophilic Campylobacter spp. in a voluntary monitoring programme in broiler flocks in Germany and in the reported human incidence, time series methods were used. The data originated between May 2004 and June 2007. By the use of seasonal decomposition, autocorrelation and cross-correlation functions, it could be shown that an annual seasonality is present. However, the peak month differs between sample submission, prevalence in broilers and human incidence. Strikingly, the peak in human campylobacterioses preceded the peak in broiler prevalence in Lower Saxony rather than occurring after it. Significant cross-correlations between monthly temperature and prevalence in broilers as well as between human incidence, monthly temperature, rainfall and wind-force were identified. The results highlight the necessity to quantify the transmission of Campylobacter from broiler to humans and to include climatic factors in order to gain further insight into the epidemiology of this zoonotic disease.

Estrogens are known to play a role in both reproductive and non-reproductive functions in mammals. Estrogens and their receptors are involved in the development of the central nervous system (brain development, neuronal survival and differentiation) as well as in the development of the peripheral nervous system (sensory-motor behaviors). In order to decipher possible functions of estrogens in early development of the zebrafish sensory system, we investigated the role of estrogen receptor beta(2) (ERbeta(2)) by using a morpholino (MO) approach blocking erbeta(2) RNA translation. We further investigated the development of lateral line organs by cell-specific labeling, which revealed a disrupted development of neuromasts in morphants. The supporting cells developed and migrated normally. Sensory hair cells, however, were absent in morphants' neuromasts. Microarray analysis and subsequent in situ hybridizations indicated an aberrant activation of the Notch signaling pathway in ERbeta(2) morphants. We conclude that signaling via ERbeta(2) is essential for hair cell development and may involve an interaction with the Notch signaling pathway during cell fate decision in the neuromast maturation process.

Responses of many real-world problems can only be evaluated perturbed by noise. In order to make an efficient optimization of these problems possible, intelligent optimization strategies successfully coping with noisy evaluations are required. In this article, a comprehensive review of existing kriging-based methods for the optimization of noisy functions is provided. In summary, ten methods for choosing the sequential samples are described using a unified formalism. They are compared on analytical benchmark problems, whereby the usual assumption of homoscedastic Gaussian noise made in the underlying models is meet. Different problem configurations (noise level, maximum number of observations, initial number of observations) and setups (covariance functions, budget, initial sample size) are considered. It is found that the choices of the initial sample size and the covariance function are not critical. The choice of the method, however, can result in significant differences in the performance. In particular, the three most intuitive criteria are found as poor alternatives. Although no criterion is found consistently more efficient than the others, two specialized methods appear more robust on average.
