Analyses of blood and liver samples from live captured sea otters and liver samples from beachcast sea otter carcasses off the remote Washington coast indicate relatively low exposure to contaminants, but suggest that even at the low levels measured, exposure may be indicated by biomarker response. Evidence of pathogen exposure is noteworthy - infectious disease presents a potential risk to Washington sea otters, particularly due to their small population size and limited distribution. During 2001 and 2002, 32 sea otters were captured, of which 28 were implanted with transmitters to track their movements and liver and blood samples were collected to evaluate contaminant and pathogen exposure. In addition, liver samples from fifteen beachcast animals that washed ashore between 1991 and 2002 were analyzed to provide historical information and a basis of reference for values obtained from live otters. The results indicate low levels of metals, butyltins, and organochlorine compounds in the blood samples, with many of the organochlorines not detected except polychlorinated biphenyls (PCBs), and a few aromatic hydrocarbons detected in the liver of the live captured animals. Aliphatic hydrocarbons were measurable in the liver from the live captured animals; however, some of these are likely from biogenic sources. A significant reduction of vitamin A storage in the liver was observed in relation to PCB, dibutyltin and octacosane concentration. A significant and strong positive correlation in vitamin A storage in the liver was observed for cadmium and several of the aliphatic hydrocarbons. Peripheral blood mononuclear cell (PBMC) cytochrome P450 induction was elevated in two of 16 animals and may be potentially related to aliphatic and aromatic hydrocarbon exposure. Mean concentration of total butyltin in the liver of the Washington beach-cast otters was more than 15 times lower than the mean concentration reported by Kannan et al. (1998) for Southern sea otters in California. Organochlorine compounds were evident in the liver of beach-cast animals, despite the lack of large human population centers and development along the Washington coast. Concentrations of PCBs and chlordanes (e.g., transchlordane, cis-chlordane, trans-nonachlor, cis-nonachlor and oxychlordane) in liver of Washington beach-cast sea otters were similar to those measured in Aleutian and California sea otters, excluding those from Monterey Bay, which were higher. Mean concentrations of 1,1,1,- trichloro-2,2-bis(p-chlorophyenyl)ethanes (DDTs) were lower, and mean concentrations of cyclohexanes (HCH, e.g., alpha BHC, beta BHC, delta BHC and gamma BHC) were slightly higher in Washington beach-cast otters versus those from California and the Aleutians. Epidemiologically, blood tests revealed that 80 percent of the otters tested positive for morbillivirus and 60 percent for Toxoplasma, the latter of which has been a significant cause of mortality in Southern sea otters in California. This is the first finding of positive morbillivirus titers in sea otters from the Northeast Pacific. Individual deaths may occur from these diseases, perhaps more so when animals are otherwise immuno-compromised or infected with multiple diseases, but a population-threatening die-off from these diseases singly is unlikely while population immunity remains high. The high frequency of detection of morbillivirus and Toxoplasma in the live otters corresponds well with the cause of death of stranded Washington sea otters reported herein, which has generally been attributable to infectious disease. Washington’s sea otter population continues to grow, with over 1100 animals currently inhabiting Washington waters; however, the rate of growth has slowed over recent years. The population has a limited distribution and has not yet reached its carrying capacity and as such, is still considered at high risk to catastrophic events. (PDF contains 189 pages)

ENGLISH: This study shows how the catch and effort statistics, from 1951 to 1956, of the fishery for yellowfin tuna, Neothunnus macropterus, in the Eastern Tropical Pacific Ocean, have been used to compute: (i) two indices of average population density; (ii) an index of concentration of effort on areas of greatest density of available yellowfin. These three indices were then used to determine: (i) quarterly and annual variation in each of them; (ii) the relationship between the two indices of density; (iii) the relationship of each of the indices to the number of exploited one-degree rectangles. To remove extreme sampling variation at low levels of effort, the data from all one-degree rectangles subjected to less than five logged days' fishing in a quarter were eliminated, and the computations were repeated for comparison with those of the original data. SPANISH: Este estudio da a conocer cómo las estadísticas sobre la pesca y el esfuerzo de pesca de la pesquería del atún aleta amarilla, Neothunnus macropterus, en el Océano Pacífico Oriental Tropical, durante 1951 a 1956, han servido para computar: (i) dos índices del promedio de la densidad de la población; (ií) un índice de la concentración del esfuerzo en las áreas de mayor densidad de atún aleta amarilla disponible. Estos tres índices han sido luego usados para determinar: (i) la variación trimestral y anual en cada uno de ellos; (ií) la relación entre los dos índices de densidad; (iii) la relación de cada uno de los índices con el número de rectángulos de un grado explotados. Para evitar la extrema variación del muestreo a bajos niveles de esfuerzo, se eliminaron los datos de todos los rectángulos de un grado sujetos a menos de cinco días de actividad pesquera durante un trimestre según los registros de los cuadernos de bitácora, y las computaciones se repitieron para compararlas con las de los datos originales.

ENGLISH: The staff of the Inter-American Tropical Tuna Commission for several years has been investigating the life history, population structure, behavior and ecology of the yellowfin tuna, Neothunnus macropterus, and the skipjack, Katsuwonus pelamis, in the Eastern Tropical Pacific Ocean. The tagging and subsequent recovery of these tropical tunas, to provide information on population structure, migrations, mortality rates and growth rates, are important aspects of these investigations. Broadhead (1959) and Schaefer, Chatwin and Broadhead (1961) emphasize the many difficulties involved in tagging these extremely active yet delicate fish and give considerable evidence to suggest that tagging mortality is high, perhaps as great as 60 to 80 per cent. The latter authors suggest that the rather high mortality at tagging is related to the effects of hyperactivity brought about by the tagging operation. SPANISH: El personal de la Comisión Interamericana del Atún Tropical ha estado investigando durante varios años la historia natural, la estructura de la población, los hábitos y la ecología del atún aleta amarilla, Neothunnus macropterus, y del barrilete, Katsuwonus pelamis, en el Océano Pacífico Oriental Tropical. La marcación y el subsiguiente recobro de estos atunes tropicales, lo que da información sobre la estructura de la población, los movimientos migratorios y las tasas de crecimiento y de mortalidad, son importantes aspectos de estas investigaciones. Broadhead (1959) y Schaefer, Chatwin y Broadhead (1961) destacan las muchas dificultades que hay para marcar estos peces activos en extremo pero delicados, y proporcionan considerable evidencia que sugiere que la mortalidad por la marcación es bastante alta, siendo quizás de 60 a 80 por ciento. Los autores citados sugieren que esta elevada mortalidad por la marcación está relacionada con los efectos de la hiperactividad producida por la operación de marcación.

ENGLISH: In a previous Bulletin of this Commission, Griffiths (1960) discussed two indices of population density and an index of concentration of fishing effort of bait boats for yellowfin tuna in the Eastern Tropical Pacific for the 1951-1956 period. Yellowfin and skipjack tuna occur in the same general fishing areas and many of the commercial catches are composed of a mixture of the two species. It is desirable, therefore, to extend the investigation to skipjack and to the two species combined. SPANISH:En un Boletín anterior de esta Comisión, Griffiths (1960) se refiere a dos índices de la densidad de la población y a un índice de la concentración del esfuerzo de pesca de los barcos de carnada sobre el atún aleta amarilla en el Pacífico Oriental Tropical, correspondientes al período 1951-1956. Los atunes aleta amarilla y barrilete se encuentran en las mismas áreas generales de pesca y muchas de las pescas comerciales están compuestas de una mezcla de las dos especies. Es deseable, por lo tanto, ampliar la investigación en lo que se refiere al barrilete y a las dos especies combinadas.

ENGLISH: In the eastern Pacific Ocean nearly all of the commercial catches of yellowfin tuna (Thunnus albacares) and skipjack (Katsuwonus pelamis) are taken by two types of vessels, baitboats, which use pole and line in conjunction with live-bait, and purse-seiners. From its inception until very recently (1959), this fishery was dominated by baitboats. This method of fishing has been described by Godsil (1938) and Shimada and Schaefer (1956). From 1951 through 1958 baitboats caught between 66.4 and 90.8 per cent of the yellowfin and between 87.2 and 95.3 per cent of the skipjack landed by the California-based fleet. These vessels fished for tuna throughout the year and covered virtually all of the area from southern California to northern Chile. The purse-seine fishery for tunas developed out of the round-haul net fisheries for California sardines and other species. Scofield (1951) gives a detailed description of the development of gear and fishing methods. Prior to 1959 many of the seiners engaged in other fisheries during the fall and early winter months and consequently most of the fishing effort for tuna occurred in the period February-August. The vessels were quite small, averaging approximately 120 tons carrying capacity (Broadhead and Marshall, 1960), in comparison to the baitboats, of which the most numerous size-class was 201-300 tons. The seiners were naturally more restricted in range than the baitboats and most of their effort was restricted to the northern grounds. During the period 1959-61 most of the large baitboats were converted for purse-seining and the existing seiner fleet was modernized. These developments increased the range of the seiner fleet and resulted in a wider and more nearly even spatial and temporal distribution of effort. By the early part of 1961, the purse-seine fleet approximated the level of the preconversion baitboat fleet in amount of effort applied and area covered. The changes in the purse-seine fishery and the fishing methods employed in the modernized fleet are described by Orange and Broadhead (1959), Broadhead and Marshall (1960), McNeely (1961) and Broadhead (1962). The change in the relative importance of the two gears is illustrated by the decline in the proportion of the total logged tonnage landed by California-based baitboats, in comparison to the proportion landed by seiners. In 1959 baitboats landed 49.5 per cent of the yellowfin and 87.8 per cent of the skipjack. In 1960 these percentages were 22.9 and 74.7 respectively and in 1961 the decline continued to 12.6 per cent of the yellowfin and 30.0 per cent of the skipjack (Schaefer, 1962). In previous Bulletins of this Commission (Griffiths, 1960; Calkins, 1961) the baitboat catch and effort statistics were used to compute two indices of population density and an index of concentration of fishing effort and the fluctuations of these indices were analyzed in some detail. Due to the change in the relative importance of the two gears it is appropriate to extend this investigation to include the purse-seine data. The objectives of this paper are to compute two indices of population density and an index of concentration of fishing effort and to examine the fluctuations in these indices before and after the changes in the fishery. A further objective is to compare the purse-seine indices with those of the baitboats for the same time periods. SPANISH: En el Océano Pacífico Oriental casi todas las capturas comerciales del atún aleta amarilla (Thunnus albacares) y del barrilete (Katsuwonus pelamis) son efectuadas por dos tipos de barcos, los barcos de carnada que emplean la caña y el anzuelo en conjunto con la carnada viva, y los barcos rederos. Desde su comienzo hasta hace poco tiempo (1959), esta pesquería estaba dominada por los barcos de carnada. El método de pesca usado por estos barcos ha sido descrito por Godsil (1938) y por Shimada y Schaefer (1956). De 1951 a 1958, los barcos de carnada pescaron entre el 66.4 y el 90.8 por ciento del atún aleta amarilla y entre el 87.2 y el 95.3 por ciento del barrilete descargados por la flota que tiene su base en California. Estos barcos pescaron atún durante todo el año y cubrieron virtualmente toda el área de California meridional hasta la parte norte de Chile. Debido al cambio en la importancia relativa de los dos sistemas de pesca, es conveniente extender esta investigación para incluir los datos correspondientes a los barcos rederos. Los objetivos del presente estudio son de computar dos índices de la densidad de población y un índice de la concentración del esfuerzo de pesca, y examinar las fluctuaciones en estos índices, antes y después de los cambios en la pesquería. Otro objetivo es de comparar los índices de los barcos rederos, con aquellos de los clipers en los mismos períodos de tiempo.

ENGLISH: Tagging and the recovery of tagged yellowfin (Thunnus albacares) and skipjack (Katsuwonus pelamis) tunas are important aspects of the investigations conducted by the Inter-American Tropical Tuna Commission in the Eastern Tropical Pacific Ocean. The results of the tagging program provide information on population structures, migrations, mortality rates and growth rates of these two species. The present experimental program was undertaken to study the relationship between muscular fatigue and high tagging mortalities in yellowfin and skipjack. SPANISH: La marcación del atún aleta amarilla (Thunnus albacares) y del barrilete (Katsuwonus pelamis), y el recobro de estos atunes marcados, son aspectos importantes de la investigación que efectúa la Comisión Interamericana del Atún Tropical en el Océano Pacífico Oriental Tropical. Los resultados del programa de marcación proporcionan información sobre la estructura de las poblaciones, migraciones, tasas de mortalidad y tasas de crecimiento de estas dos especies. El programa experimental presente fue emprendido para estudiar la relación entre la fatiga muscular y la alta mortalidad causada por la marcación en el atún aleta amarilla y el barrilete. Background: Implantation and growth of metastatic cancer cells at distant organs is promoted by inflammation-dependent mechanisms. A hepatic melanoma metastasis model where a majority of metastases are generated via interleukin-18-dependent mechanisms was used to test whether anti-inflammatory properties of resveratrol can interfere with mechanisms of metastasis. Methods: Two experimental treatment schedules were used: 1) Mice received one daily oral dose of 1 mg/kg resveratrol after cancer cell injection and the metastasis number and volume were determined on day 12. 2) Mice received one daily oral dose of 1 mg/kg resveratrol along the 5 days prior to the injection of cancer cells and both interleukin-18 (IL-18) concentration in the hepatic blood and microvascular retention of luciferase-transfected B16M cells were determined on the 18(th) hour. In vitro, primary cultured hepatic sinusoidal endothelial cells were treated with B16M-conditioned medium to mimic their in vivo activation by tumor-derived factors and the effect of resveratrol on IL-18 secretion, on vascular cell adhesion molecule-1 (VCAM-1) expression and on tumor cell adhesion were studied. The effect of resveratrol on melanoma cell activation by IL-18 was also studied. Results: Resveratrol remarkably inhibited hepatic retention and metastatic growth of melanoma cells by 50% and 75%, respectively. The mechanism involved IL-18 blockade at three levels: First, resveratrol prevented IL-18 augmentation in the blood of melanoma cell-infiltrated livers. Second, resveratrol inhibited IL-18-dependent expression of VCAM-1 by tumor-activated hepatic sinusoidal endothelium, preventing melanoma cell adhesion to the microvasculature. Third, resveratrol inhibited adhesion-and proliferation-stimulating effects of IL-18 on metastatic melanoma cells through hydrogen peroxide-dependent nuclear factor-kappaB translocation blockade on these cells. Conclusions: These results demonstrate multiple sites for therapeutic intervention using resveratrol within the prometastatic microenvironment generated by tumor-induced hepatic IL-18, and suggest a remarkable effect of resveratrol in the prevention of inflammation-dependent melanoma metastasis in the liver.

The lift force on a spherical nanoparticle near a wall in micro/nanofluidics has not received sufficient attention so far. In this letter the concentration of 200 nm particles is measured at 0.25–2.0 m to a wall in a microchannel with pressure-driven de-ionized water flow pressure gradient 0–2000 kPa/m . The measured data show the influence of the lift force on the nanoparticle concentration distribution. By introducing the Saffman lift force into the Nernst–Planck equation near a wall, we find that the lift force is dominant at the range of 2

The growth responses and feed utilization of Sarotherodon niloticus held in metal cages in a pond and fed diets containing fish-meal, cow blood-meal or sorghum was studied. Results indicate that the best growth, feed conversion and protein efficiency ratio were obtained with the diet containing 60% fish-meal. The growth performance of fish on 40% fish-meal, and 40% and 60% blood meal were not significantly different, and were quite close to the performance with 60% fish-meal. The growth and food utilization of fish on 84% sorghum diet was significantly lower than the rest. The Caged fish without supplemental feeding had a light gain in weight. All fish with supplemental feeding appeared healthy. It is concluded that cow blood meal at 40% or 60% inclusion in diet can adequately replace fish-meal in S. niloticus supplemental diet in pond culture

Fingerling of Clarias gariepinus have been reported to have an optimal protein requirement of 40%. Not much is known about the effect that varying this protein level has in the haematological characteristics (i.e packed cell volume, red blood cell count, haemoglobin concentration and mean corpuscular haemoglobin concentration) and on some landmarks measured along the fish's body. The haematological parameter are useful in assessing the effect of dietary treatment on leanness or robustness in the fish. The results of these experiment reveals that of the 17 landmarks measured on the bodies of the fish species fed dietary protein levels of no protein 11% (low protein), 29% (sub optimal) and 40% (optimal), only four of the landmarks show significant difference. Also, analysis of the haematological characteristics show significant difference in haematoant (PCV) and erthroycte count (RBC) in all the treatments

Micro-fabrication technology has substantial potential for identifying molecular markers expressed on the surfaces of tissue cells and viruses. It has been found in several conceptual prototypes that cells with such markers are able to be captured by their antibodies immobilized on microchannel substrates and unbound cells are flushed out by a driven flow. The feasibility and reliability of such a microfluidic-based assay, however, remains to be further tested. In the current work, we developed a microfluidic-based system consisting of a microfluidic chip, an image grabbing unit, data acquisition and analysis software, as well as a supporting base. Specific binding of CD59-expressed or BSA-coupled human red blood cells (RBCs) to anti-CD59 or anti-BSA antibody-immobilized chip surfaces was quantified by capture efficiency and by the fraction of bound cells. Impacts of respective flow rate, cell concentration, antibody concentration and site density were tested systematically. The measured data indicated that the assay was robust. The robustness was further confirmed by capture efficiencies measured from an independent ELISA-based cell binding assay. These results demonstrated that the system developed provided a new platform to effectively quantify cellular surface markers effectively, which promoted the potential applications in both biological studies and clinical diagnoses.

Fingerlings of Clarias anguillaris obtained from a homogenous source through induce breeding and each with a mean weight of 2.8g were stocked in ten hapas each measuring 1.0x1.0m in outdoor concrete tank and were fed for eight (8) weeks. Results shows that the best growth rate was recorded among fingerlings fed fish meal as the only protein source (TD5) while DT2 containing soya bean, groundnut cake (40%), blood meal as the protein sources came next. The growth rate of fingerlings fed DT2 (40 % groundnut cake, 10% soyabean meal and 10% blood meal) was higher than those fed DT4 containing 10% fish meal but lower than those fed DT5 which has fish meal as its sole source of protein (53.0%). Analysis of various growth parameters like SGR, FCR and PER. shows that DT5 was the overall best diet but there was no significant statistical difference in weight gained by fish fed the five diets (P <0.05)

Blood meal and full fat soyabean meal were mixed in different proportions to give 0%, 10%, 25%, 50%, 75%, 90% and 100% meal in the protein fraction of the diet and fed to Claria anguillaris fingerlings in floating hapas. The growth performance of the fingerlings were monitored for 84 days. At the end of the experiment the mean weight of the fingerlings increased in the level of blood meal up to 50% blood meal in the diet after which there was a decline in the mean weight of the fish. This same level of blood meal gave the best specific growth rate, feed conversion efficiency and protein efficiency ratio. Thus the nutritive value of blood meal was enhanced by the addition of an equal level of full fat soyabean meal in the diet
