Abstract. We resumed mowing in two plots of ca. 100 m2 in an abandoned meadow dominated by Brachypodium pinnatum on the slope of Monte Generoso (Switzerland). We monitored species composition and hay yield using point quadrats and biomass samples. Species frequencies changed little during 10 yr (1988–1997) while hay yields showed large fluctuations according to mean relative humidity in April-June. We performed a seed-addition experiment to test whether the establishment of meadow species is limited by lack of diaspores or favourable microsites for germination and recruitment from the seed bank. We sowed ca. 12 000 seeds of 12 species originating from a nearby meadow individually in plots of a 4 × 6 unbalanced Latin square with four treatments, burning, mowing, mowing and removal of a layer of decayed organic matter, and a control. We monitored the fate of seedling individuals for 24 months. Seedlings of all species were established and survived for 12 months, 10 species survived during at least 24 months, some reached a reproductive stage. Species responded to different qualities of microsites provided by the different treatments thus required different regeneration niches. Spontaneous long-distance immigration was insignificant. We conclude that the former species composition of abandoned meadows cannot easily be restored by mowing alone because many plant species of meadows do not have persistent seed banks and immigration over distances of more than 25 m and successful establishment is very unlikely.

Flemish Pass, located at the western subpolar margin, is a passage (sill depth 1200 m) that is constrained by the Grand Banks and the underwater plateau Flemish Cap. In addition to the Deep Western Boundary Current (DWBC) pathway offshore of Flemish Cap, Flemish Pass represents another southward transport pathway for two modes of Labrador Sea Water (LSW), the lightest component of North Atlantic Deep Water carried with the DWBC. This pathway avoids potential stirring regions east of Flemish Cap and deflection into the interior North Atlantic. Ship-based velocity measurements between 2009 and 2013 at 47°N in Flemish Pass and in the DWBC east of Flemish Cap revealed a considerable southward transport of Upper LSW through Flemish Pass (15-27%, -1.0 to -1.5 Sv). About 98% of the denser Deep LSW were carried around Flemish Cap as Flemish Pass is too shallow for considerable transport of Deep LSW. Hydrographic time series from ship-based measurements show a significant warming of 0.3°C/decade and a salinification of 0.03/decade of the Upper LSW in Flemish Pass between 1993 and 2013. Almost identical trends were found for the evolution in the Labrador Sea and in the DWBC east of Flemish Cap. This indicates that the long-term hydrographic variability of Upper LSW in Flemish Pass as well as in the DWBC at 47°N is dominated by changes in the Labrador Sea, which are advected southward. Fifty years of numerical ocean model simulations in Flemish Pass suggest that these trends are part of a multidecadal cycle.

Las casas del siglo XX construidas al borde del mar –escenario único y origen de su expresión- siguen la topografía del suelo que habitan en su descenso al agua, y organizan espacios que obtienen miradas al horizonte marino. El horizonte nos confronta a lo desconocido. La vista del mar incita al deseo de recorrerlo, al deseo de viajar. Con origen en el otium romano, la presencia del agua define un modo de vida apacible –epicúreo- que los viajeros de la arquitectura -que nos acompañan en la tesis- traducen en sus refugios más íntimos. Experimentan con los cambios en los conceptos y en las técnicas, que se trasladan fácilmente a la arquitectura de la casa al borde del agua desde los inicios del Movimiento Moderno. Sus espacios del habitar frente al mar nos permiten descubrir estrategias comunes en sus respuestas más modernas. El arquitecto ante el proyecto frente al mar llega a la ubicación elegida, mira hacia el horizonte, y desplazándose ladera arriba se coloca en un punto estratégico que elige; después, delante de su tablero, recorre el camino inverso, coloca el lugar y sobre él dibuja los elementos que configurarán los espacios de la casa buscando esa mirada al mar. Las situaciones y ubicaciones posibles son infinitas; se definen algunas consonancias espaciales comunes en los suelos que se ocupan debidas a la presencia del mar que asociamos entre sí. De la comparación entre todas las casas -emblemas del XX-, surgen múltiples variantes de la mirada y de espacios al abierto, y modos de fabricar entornos con criterios comunes para dominar la visión del mar. Interiores que se abren al panorama, espacios cuyas ventanas buscan su mirada en la extensión del horizonte, rescisiones y aperturas. Reconocemos condicionantes en el territorio a los que las villas responden, categorías arquitectónicas que dan respuesta frente al mar en la búsqueda del moderno, la topografía, la mirada y el espacio al abierto. Las casas comparten la idea del dominio del paisaje desde el punto más alto, y en algunos suelos se invierte la tipología por la topografía, confirmando así un criterio común basado en la lectura del suelo como consecuencia de la búsqueda del espacio de la mirada. Los espacios al abierto se significan en todas ellas, son espacios al -aire libre- abiertos, unos envueltos, otros porticados, puertas del horizonte que se abren al exterior, en el techo de la casa, otros cubiertos y abiertos, espacios entre interior y exterior, en plataformas con bancales o patios envolventes, recintos o habitaciones abiertas. Descubrimos un logro del XX en los espacios positivos o negativos que traducen o juegan con el entorno, que ocupan o sustraen de los contornos construidos y que obtienen espacios intermedios en la búsqueda de la relación con el mar. Las herramientas que se utilizan son los dibujos de los autores, de las casas visitadas, el elenco de viajeros y sus viajes, el conocimiento desde el estudio de los proyectos. A través de la comparación por aproximaciones parciales, los dibujos nos definen la mirada al mar, el modo de ocupación y la forma de relación con el paisaje. La arquitectura del habitar frente al mar en el XX, hecha para y por arquitectos, topografía el suelo y construye la mirada, fabricando espacios al abierto en la relación entre la casa y el entorno marítimo. ABSTRACT Houses of the 20th century built by the sea – a unique setting which gives rise to their expression – follow the topography of the land they occupy in its descent towards the sea, and they organize spaces which give views of the maritime horizon. The horizon brings us face to face with the unknown. The sea view provokes a desire to cross it, to travel. The presence of the sea defines a peaceful, epicurean way of life, with origins in the Roman otium, which architectural travellers – who accompany us through the thesis – translate into their most intimate retreats. They experiment with changes in concepts and techniques, which are easily transferred to the architecture of the seaside house since the beginnings of the Modern Movement. Their living spaces allow us to discover common strategies in the most modern responses. The architect with a seaside project arrives at the site, looks towards the horizon, then walks uphill and chooses a strategic point; then with his drawing board he retraces his steps, he sets the position and then draws in the elements that make up the house that seeks a sea view. The number of potential situations and locations is infinite; certain common spatial accordances are defined in land which is occupied due to the presence of the sea. Comparison of all the houses – 20th century emblems – throws up multiple variations of view and open spaces, and ways of creating settings with common criteria so as to command the vision of the sea. Interiors which open up to the panorama, spaces whose windows seek their view in the expanse of the horizon, openings and closures. We recognise determinant factors in the territory to which the villas respond, architectural categories which give a seaside solution to the search for the modern, the topography, the view, and the open space. The houses share the idea of dominating the landscape from the highest point, and in some areas typology and topography are inverted, thus confirming a common criteria based on the reading of the ground as a conse quence of the search for the view space. Open spaces stand out in all the villas – spaces open to the outdoor air - some are wrapped, some arcaded, doors to the horizon which open up to the exterior, on the roof of the house. There are open and covered spaces, spaces between the exterior and interior, on platforms with banks and surrounding patios, enclosures and open rooms. We discover an achievement of the 20th century in the positive and negative spaces which translate and play with the setting, which occupy or are extracted from built contours and which obtain intermediate spaces in the search for the relationship with the sea. The tools used are the author’s drawings of the houses visited, the cast of travelling companions and their travels, the knowledge gained from study of the projects. Through comparison by means of partial approaches, the drawings define the view of the sea, the occupation mode and the way of relating to the landscape. Architecture for living by the sea in the 20th century, carried out both by and for the architects, shapes the land and constructs the view, creating open spaces in the relationship between the house and the sea surroundings.

Cross-contamination between cell lines is a longstanding and frequent cause of scientific misrepresentation. Estimates from national testing services indicate that up to 36% of cell lines are of a different origin or species to that claimed. To test a standard method of cell line authentication, 253 human cell lines from banks and research institutes worldwide were analyzed by short tandem repeat profiling. The short tandem repeat profile is a simple numerical code that is reproducible between laboratories, is inexpensive, and can provide an international reference standard for every cell line. If DNA profiling of cell lines is accepted and demanded internationally, scientific misrepresentation because of cross-contamination can be largely eliminated.

An automated oligonucleotide synthesizer has been developed that can simultaneously and rapidly synthesize up to 96 different oligonucleotides in a 96-well microtiter format using phosphoramidite synthesis chemistry. A modified 96-well plate is positioned under reagent valve banks, and appropriate reagents are delivered into individual wells containing the growing oligonucleotide chain, which is bound to a solid support. Each well has a filter bottom that enables the removal of spent reagents while retaining the solid support matrix. A seal design is employed to control synthesis environment and the entire instrument is automated via computer control. Synthesis cycle times for 96 couplings are < 11 min, allowing a plate of 96 20-mers to be synthesized in < 5 hr. Oligonucleotide synthesis quality is comparable to commercial machines, with average coupling efficiencies routinely > 98% across the entire 96-well plate. No significant well-to-well variations in synthesis quality have been observed in > 6000 oligonucleotides synthesized to date. The reduced reagent usage and increased capacity allow the overall synthesis cost to drop by at least a factor of 10. With the development of this instrument, it is now practical and cost-effective to synthesize thousands to tens of thousands of oligonucleotides.

A obtenção de genótipos superiores no melhoramento de plantas depende da existência de variabilidade genética. A existência de coleções de germoplasma representativas e a utilização de um tamanho adequado de amostra são fundamentais para a preservação das frequências alélicas e genotípicas, diminuindo a perda de variabilidade genética e postergando o aparecimento dos efeitos da deriva genética. Assim, teve-se como objetivo avaliar os efeitos da deriva genética em caracteres quantitativos em subpopulações de milho. Este estudo foi realizado a partir das populações originais BR-105 e BR-106, das quais 10 subpopulações foram obtidas em cada um dos cinco ciclos sucessivos de amostragem com tamanho efetivo reduzido, totalizando 50 subpopulações para cada população original, as quais foram posteriormente autofecundadas, gerando um nível a mais de endogamia. Os tratamentos foram constituídos de 10 amostras da população original sem autofecundação, 10 amostras com autofecundação, 50 subpopulações obtidas da população original e 50 subpopulações autofecundadas, totalizando 120 tratamentos para cada população, avaliados separadamente. Utilizou-se o delineamento em blocos casualizados no esquema de parcelas subdivididas em faixas hierárquico, em quatro ambientes com duas repetições por ambiente. Os caracteres avaliados foram produção de grãos (PG), prolificidade (PROL), comprimento e diâmetro de espigas (CE e DE), número de fileiras por espiga (NFE), número de grãos por fileira (NGF), altura de planta e espiga (AP e AE), florescimento masculino e feminino (FM e FF) e número de ramificações do pendão (NRP). Foram estimados os efeitos da deriva genética entre as médias das subpopulações nos dois níveis de endogamia e os efeitos da depressão por endogamia nas subpopulações dentro dos ciclos. Posteriormente, realizaram-se análises de regressão linear para as subpopulações nos dois níveis de endogamia, separadamente, e em conjunto. Foi verificada uma grande variação nas médias das subpopulações ao longo dos ciclos, indicando que a deriva genética causou diferenciação entre as mesmas e que estas se diferenciaram das populações originais. Detectaram-se efeitos significativos da deriva genética nas populações não autofecundadas para todos os caracteres avaliados, em maior número para PG, já que este caráter é mais sensível à deriva genética por possuir maior grau de dominância que os demais. Houve diminuição no número de estimativas de deriva significativas para as populações autofecundadas, incluindo mudanças na magnitude e no sinal das mesmas em relação às populações não autofecundadas. Para as estimativas de depressão por endogamia, os caracteres PG, NGF, FM e FF apresentaram maior quantidade de estimativas significativas que os demais. Para a maioria dos caracteres, a regressão linear explicou a maior parte da variação encontrada com o aumento dos coeficientes de endogamia. As populações BR-105 e BR-106, por terem estruturas genéticas distintas, apresentaram performances diferentes quanto aos efeitos da deriva genética. Enfim, como a deriva genética interfere na integridade genética das populações, torna-se importante considerar seus efeitos na coleta e manutenção dos bancos de germoplasma e nas populações utilizadas no melhoramento genético de plantas.

Urban researchers and planners are often interested in understanding how economic activities are distributed in urban regions, what forces influence their special pattern and how urban structure and functions are mutually dependent. In this paper, we want to show how an algorithm for ranking the nodes in a network can be used to understand and visualize certain commercial activities of a city. The first part of the method consists of collecting real information about different types of commercial activities at each location in the urban network of the city of Murcia, Spain. Four clearly differentiated commercial activities are studied, such as restaurants and bars, shops, banks and supermarkets or department stores, but obviously we can study other. The information collected is then quantified by means of a data matrix, which is used as the basis for the implementation of a PageRank algorithm which produces a ranking of all the nodes in the network, according to their significance within it. Finally, we visualize the resulting classification using a colour scale that helps us to represent the business network.

En la última década, la organización del sistema financiero español ha tenido una convulsión de un alcance inimaginable y puede decirse sin ningún género de dudas que el cambio ha sido histórico. De un sistema en el que el reparto de las instituciones financieras estaba representado por las Cajas de Ahorro y la banca en un 50% por cada grupo, se ha pasado a un escenario en el que las Cajas de Ahorro han desaparecido casi en su totalidad y la banca ha ocupado completamente todo el sistema financiero español. Difícilmente se puede explicar este cambio por factores exclusivamente atribuibles a unas u otras instituciones ya que, en buena lógica, una crisis económica afecta con distinta intensidad a cada una de las empresas y, por lo tanto, hubiese cabido esperar que el mantenimiento de algunas cajas de ahorros ante el embate de la crisis hubiese significado que quedasen algunas en funcionamiento. Pero ¿todas? También hubiera cabido esperar que la crisis hubiera afectado a más bancos, pero ¿tan pocos? ¿Tan bien estaban o sabían más que las demás entidades financieras? En esta comunicación vamos a tratar de analizar el comportamiento del Banco de España en este proceso y ver hasta qué punto sus decisiones o falta de decisiones han contribuido a profundizar en la crisis económica y en el hundimiento de todo el sector de cajas de ahorro españolas.
