Since Januarv 1946 a wade EC Project entitled "Mealiness in fruits Consumers perception and means for detection is being carried out. Mealiness is a sensory attribute that cannot be defined by a single parameter but through a combination of variables (multidimensional structure) Previous studies propose the definition of mealiness as the lack of crispiness of hardness and of juiciness. A destructive instrumental procedure combined with a integration technique has been already developed enabling to identify mealy fruits by destructive instrumental means use other contributions of Barreiro and Ortiz to this Ag Eng 98. Current aims .are focused on establishing non destructive tests for mealiness assessment. Magnetic resonance Imaging (MRI) makes use of the magnetic properties that some atomic nuclei have. especially hidrogen nuclei from water molecules to obtain high quality images in the field of internal quality evaluation the MRI has been used to assess internal injury due to conservation as o treatments as chilling injury un Persimmons Clark&Forbes (1994) and water-core in apples (Wang et al. 1998. In the case of persimmons the chilling injury is described as an initial tissue breakdown and lack of cohesion between cells followed by formation of a firm gel and by a lack of juiciness without changes in the total amount ol water content. Also a browning of the flesh is indicated (Clark&Forhes 1994). This definition fits into the previous description of mealiness.

European Universities are involved in series of great changes regarding teaching and education organization during the last few years. The origin of these changes is the creation of the so-called European Higher Education Area (EHEA), which main target is to harmonize the different University studies throughout Europe. As a consequence, most of the programs of studies in all degrees are suffering changes in order to converge to common structures. Taking advantage of the actual process, some European universities are moving from traditional Agricultural Engineering programs to a more wide discipline named recently as Biosystems Engineering, which is a science- based engineering discipline that integrates engineering science and design with applied biological, environmental and agricultural sciences, broadening in this way the area of application of Engineering sciences not strictly to agricultural sciences, but to the biologic al sciences in general, including the agricultural sciences. This paper presents a comparative study of different Bachelor of Science degrees offered by American and European Universities in the field of Agricultural/Biosystems Engineering. To carry out the analysis 40 programs accredited by ABET in American Universities and 50 European programs. Among other questions, the total number of credits, the number of semesters, the kind of modules and the distribution of subjects in groups (Basic Sciences, Engineering Fundamentals, Agricultural/Biological Sciences, Humanities & Economic Sciences, Applied Agricultural/Biological Engineering and electives) are discussed in the paper. The information provided can be an useful starting point in future definitions of new or renewed degrees with the aim of advancing in internationalization of the programs and helping student’s mobility.

Dynamics of binary mixtures such as polymer blends, and fluids near the critical point, is described by the model-H, which couples momentum transport and diffusion of the components [1]. We present an extended version of the model-H that allows to study the combined effect of phase separation in a polymer blend and surface structuring of the film itself [2]. We apply it to analyze the stability of vertically stratified base states on extended films of polymer blends and show that convective transport leads to new mechanisms of instability as compared to the simpler diffusive case described by the Cahn- Hilliard model [3, 4]. We carry out this analysis for realistic parameters of polymer blends used in experimental setups such as PS/PVME. However, geometrically more complicated states involving lateral structuring, strong deflections of the free surface, oblique diffuse interfaces, checkerboard modes, or droplets of a component above of the other are possible at critical composition solving the Cahn Hilliard equation in the static limit for rectangular domains [5, 6] or with deformable free surfaces [6]. We extend these results for off-critical compositions, since balanced overall composition in experiments are unusual. In particular, we study steady nonlinear solutions of the Cahn-Hilliard equation for bidimensional layers with fixed geometry and deformable free surface. Furthermore we distinguished the cases with and without energetic bias at the free surface. We present bifurcation diagrams for off-critical films of polymer blends with free surfaces, showing their free energy, and the L2-norms of surface deflection and the concentration field, as a function of lateral domain size and mean composition. Simultaneously, we look at spatial dependent profiles of the height and concentration. To treat the problem of films with arbitrary surface deflections our calculations are based on minimizing the free energy functional at given composition and geometric constraints using a variational approach based on the Cahn-Hilliard equation. The problem is solved numerically using the finite element method (FEM).

La escasez del agua en las regiones áridas y semiáridas se debe a la escasez de precipitaciones y la distribución desigual en toda la temporada, lo que hace de la agricultura de secano una empresa precaria. Un enfoque para mejorar y estabilizar el agua disponible para la producción de cultivos en estas regiones es el uso de tecnologías de captación de agua de lluvia in situ y su conservación. La adopción de los sistemas de conservación de la humedad del suelo in situ, tales como la labranza de conservación, es una de las estrategias para mejorar la gestión de la agricultura en zonas áridas y semiáridas. El objetivo general de esta tesis ha sido desarrollar una metodología de aplicación de labranza de depósito e investigar los efectos a corto plazo sobre las propiedades físicas del suelo de las diferentes prácticas de cultivo que incluyen labranza de depósito: (reservoir tillage, RT), la laboreo mínimo: (minimum tillage, MT), la no laboreo: (zero tillage, ZT) y laboreo convencional: (conventional tillage, CT) Así como, la retención de agua del suelo y el control de la erosión del suelo en las zonas áridas y semiáridas. Como una primera aproximación, se ha realizado una revisión profunda del estado de la técnica, después de la cual, se encontró que la labranza de depósito es un sistema eficaz de cosecha del agua de lluvia y conservación del suelo, pero que no ha sido evaluada científicamente tanto como otros sistemas de labranza. Los trabajos experimentales cubrieron tres condiciones diferentes: experimentos en laboratorio, experimentos de campo en una región árida, y experimentos de campo en una región semiárida. Para investigar y cuantificar el almacenamiento de agua a temperatura ambiente y la forma en que podría adaptarse para mejorar la infiltración del agua de lluvia recolectada y reducir la erosión del suelo, se ha desarrollado un simulador de lluvia a escala de laboratorio. Las características de las lluvias, entre ellas la intensidad de las precipitaciones, la uniformidad espacial y tamaño de la gota de lluvia, confirmaron que las condiciones naturales de precipitación son simuladas con suficiente precisión. El simulador fue controlado automáticamente mediante una válvula de solenoide y tres boquillas de presión que se usaron para rociar agua correspondiente a diferentes intensidades de lluvia. Con el fin de evaluar el método de RT bajo diferentes pendientes de superficie, se utilizaron diferentes dispositivos de pala de suelo para sacar un volumen idéntico para hacer depresiones. Estas depresiones se compararon con una superficie de suelo control sin depresión, y los resultados mostraron que la RT fue capaz de reducir la erosión del suelo y la escorrentía superficial y aumentar significativamente la infiltración. Luego, basándonos en estos resultados, y después de identificar la forma adecuada de las depresiones, se ha diseñado una herramienta combinada (sistema integrado de labranza de depósito (RT)) compuesto por un arado de una sola línea de chisel, una sola línea de grada en diente de pico, sembradora modificada, y rodillo de púas. El equipo fue construido y se utiliza para comparación con MT y CT en un ambiente árido en Egipto. El estudio se realizó para evaluar el impacto de diferentes prácticas de labranza y sus parámetros de funcionamiento a diferentes profundidades de labranza y con distintas velocidades de avance sobre las propiedades físicas del suelo, así como, la pérdida de suelo, régimen de humedad, la eficiencia de recolección de agua, y la productividad de trigo de invierno. Los resultados indicaron que la RT aumentó drásticamente la infiltración, produciendo una tasa que era 47.51% más alta que MT y 64.56% mayor que la CT. Además, los resultados mostraron que los valores más bajos de la escorrentía y pérdidas de suelos 4.91 mm y 0.65 t ha-1, respectivamente, se registraron en la RT, mientras que los valores más altos, 11.36 mm y 1.66 t ha-1, respectivamente, se produjeron en el marco del CT. Además, otros dos experimentos de campo se llevaron a cabo en ambiente semiárido en Madrid con la cebada y el maíz como los principales cultivos. También ha sido estudiado el potencial de la tecnología inalámbrica de sensores para monitorizar el potencial de agua del suelo. Para el experimento en el que se cultivaba la cebada en secano, se realizaron dos prácticas de labranza (RT y MT). Los resultados mostraron que el potencial del agua del suelo aumentó de forma constante y fue consistentemente mayor en MT. Además, con independencia de todo el período de observación, RT redujo el potencial hídrico del suelo en un 43.6, 5.7 y 82.3% respectivamente en comparación con el MT a profundidades de suelo (10, 20 y 30 cm, respectivamente). También se observaron diferencias claras en los componentes del rendimiento de los cultivos y de rendimiento entre los dos sistemas de labranza, el rendimiento de grano (hasta 14%) y la producción de biomasa (hasta 8.8%) se incrementaron en RT. En el experimento donde se cultivó el maíz en regadío, se realizaron cuatro prácticas de labranza (RT, MT, ZT y CT). Los resultados revelaron que ZT y RT tenían el potencial de agua y temperatura del suelo más bajas. En comparación con el tratamiento con CT, ZT y RT disminuyó el potencial hídrico del suelo en un 72 y 23%, respectivamente, a la profundidad del suelo de 40 cm, y provocó la disminución de la temperatura del suelo en 1.1 y un 0.8 0C respectivamente, en la profundidad del suelo de 5 cm y, por otro lado, el ZT tenía la densidad aparente del suelo y resistencia a la penetración más altas, la cual retrasó el crecimiento del maíz y disminuyó el rendimiento de grano que fue del 15.4% menor que el tratamiento con CT. RT aumenta el rendimiento de grano de maíz cerca de 12.8% en comparación con la ZT. Por otra parte, no hubo diferencias significativas entre (RT, MT y CT) sobre el rendimiento del maíz. En resumen, según los resultados de estos experimentos, se puede decir que mediante el uso de la labranza de depósito, consistente en realizar depresiones después de la siembra, las superficies internas de estas depresiones se consolidan de tal manera que el agua se mantiene para filtrarse en el suelo y por lo tanto dan tiempo para aportar humedad a la zona de enraizamiento de las plantas durante un período prolongado de tiempo. La labranza del depósito podría ser utilizada como un método alternativo en regiones áridas y semiáridas dado que retiene la humedad in situ, a través de estructuras que reducen la escorrentía y por lo tanto puede resultar en la mejora de rendimiento de los cultivos. ABSTRACT Water shortage in arid and semi-arid regions stems from low rainfall and uneven distribution throughout the season, which makes rainfed agriculture a precarious enterprise. One approach to enhance and stabilize the water available for crop production in these regions is to use in-situ rainwater harvesting and conservation technologies. Adoption of in-situ soil moisture conservation systems, such as conservation tillage, is one of the strategies for upgrading agriculture management in arid and semi-arid environments. The general aim of this thesis is to develop a methodology to apply reservoir tillage to investigate the short-term effects of different tillage practices including reservoir tillage (RT), minimum tillage (MT), zero tillage (ZT), and conventional tillage (CT) on soil physical properties, as well as, soil water retention, and soil erosion control in arid and semi-arid areas. As a first approach, a review of the state of the art has been done. We found that reservoir tillage is an effective system of harvesting rainwater and conserving soil, but it has not been scientifically evaluated like other tillage systems. Experimental works covered three different conditions: laboratory experiments, field experiments in an arid region, and field experiments in a semi-arid region. To investigate and quantify water storage from RT and how it could be adapted to improve infiltration of harvested rainwater and reduce soil erosion, a laboratory-scale rainfall simulator was developed. Rainfall characteristics, including rainfall intensity, spatial uniformity and raindrop size, confirm that natural rainfall conditions are simulated with sufficient accuracy. The simulator was auto-controlled by a solenoid valve and three pressure nozzles were used to spray water corresponding to different rainfall intensities. In order to assess the RT method under different surface slopes, different soil scooping devices with identical volume were used to create depressions. The performance of the soil with these depressions was compared to a control soil surface (with no depression). Results show that RT was able to reduce soil erosion and surface runoff and significantly increase infiltration. Then, based on these results and after selecting the proper shape of depressions, a combination implement integrated reservoir tillage system (integrated RT) comprised of a single-row chisel plow, single-row spike tooth harrow, modified seeder, and spiked roller was developed and used to compared to MT and CT in an arid environment in Egypt. The field experiments were conducted to evaluate the impact of different tillage practices and their operating parameters at different tillage depths and different forward speeds on the soil physical properties, as well as on runoff, soil losses, moisture regime, water harvesting efficiency, and winter wheat productivity. Results indicated that the integrated RT drastically increased infiltration, producing a rate that was 47.51% higher than MT and 64.56% higher than CT. In addition, results showed that the lowest values of runoff and soil losses, 4.91 mm and 0.65 t ha-1 respectively, were recorded under the integrated RT, while the highest values, 11.36 mm and 1.66 t ha -1 respectively, occurred under the CT. In addition, two field experiments were carried out in semi-arid environment in Madrid with barley and maize as the main crops. For the rainfed barley experiment, two tillage practices (RT, and MT) were performed. Results showed that soil water potential increased quite steadily and were consistently greater in MT and, irrespective of the entire observation period, RT decreased soil water potential by 43.6, 5.7, and 82.3% compared to MT at soil depths (10, 20, and 30 cm, respectively). In addition, clear differences in crop yield and yield components were observed between the two tillage systems, grain yield (up to 14%) and biomass yield (up to 8.8%) were increased by RT. For the irrigated maize experiment, four tillage practices (RT, MT, ZT, and CT) were performed. Results showed that ZT and RT had the lowest soil water potential and soil temperature. Compared to CT treatment, ZT and RT decreased soil water potential by 72 and 23% respectively, at soil depth of 40 cm, and decreased soil temperature by 1.1 and 0.8 0C respectively, at soil depth of 5 cm. Also, ZT had the highest soil bulk density and penetration resistance, which delayed the maize growth and decreased the grain yield that was 15.4% lower than CT treatment. RT increased maize grain yield about 12.8% compared to ZT. On the other hand, no significant differences among (RT, MT, and CT) on maize yield were found. In summary, according to the results from these experiments using reservoir tillage to make depressions after seeding, these depression’s internal surfaces are consolidated in such a way that the water is held to percolate into the soil and thus allowing time to offer moisture to the plant rooting zone over an extended period of time. Reservoir tillage could be used as an alternative method in arid and semi-arid regions and it retains moisture in-situ, through structures that reduce runoff and thus can result in improved crop yields.

Recent advances in coherent optical receivers is reviewed. Digital-Signal-Processing (DSP) based phase and polarization management techniques make coherent detection robust and feasible. With coherent detection, the complex field of the received optical signal is fully recovered, allowing compensation of linear and nonlinear optical impairments including chromatic dispersion (CD) and polarization-mode dispersion (PMD) using digital filters. Coherent detection and advanced optical modulation formats have become a key ingredient to the design of modern dense wavelength-division multiplexed (DWDM) optical broadband networks. In this paper, firstly we present the different subsystems of a digital coherent optical receiver, and secondly, we will compare the performance of some multi-level and multi-dimensional modulation formats in some physical impairments and in high spectral-efficiency (SE) and high-capacity DWDM transmissions, simulating the DSP with Matlab and the optical network performance with OptiSystem software.

English Summary: Both in the remote origin of the city and the more recent one of urbanism as a remedial discipline conceived as a means of tackling with the urban and territorial impacts of industrialisation, the relatively balanced relation between town and country was a constant which endured until the progressive fragmentation and separation among the different areas of knowledge and action contributed to create a gap between them both, with disastrous consequences in environmental, social and economical terms. The tasks at hands from the perspective of integral sustainability is to promote a reunion throughout a new culture of territory. Resumen Tanto en los orígenes remotos de la ciudad como en los recientes del urbanismo como disciplina surgida para mitigar el impacto urbano y territorial del industrialismo, la relación relativamente equilibrada entre campo y ciudad fue una constante que se mantuvo hasta que la progresiva fragmentación entre áreas de conocimiento y de intervención contribuyó a un paulatino desencuentro entre ambos, de desastrosas consecuencias en términos sociales, ambientales y económicos. La tomografía axial computerizada (TAC) es la modalidad de imagen médica preferente para el estudio de enfermedades pulmonares y el análisis de su vasculatura. La segmentación general de vasos en pulmón ha sido abordada en profundidad a lo largo de los últimos años por la comunidad científica que trabaja en el campo de procesamiento de imagen; sin embargo, la diferenciación entre irrigaciones arterial y venosa es aún un problema abierto. De hecho, la separación automática de arterias y venas está considerado como uno de los grandes retos futuros del procesamiento de imágenes biomédicas. La segmentación arteria-vena (AV) permitiría el estudio de ambas irrigaciones por separado, lo cual tendría importantes consecuencias en diferentes escenarios médicos y múltiples enfermedades pulmonares o estados patológicos. Características como la densidad, geometría, topología y tamaño de los vasos sanguíneos podrían ser analizados en enfermedades que conllevan remodelación de la vasculatura pulmonar, haciendo incluso posible el descubrimiento de nuevos biomarcadores específicos que aún hoy en dípermanecen ocultos. Esta diferenciación entre arterias y venas también podría ayudar a la mejora y el desarrollo de métodos de procesamiento de las distintas estructuras pulmonares. Sin embargo, el estudio del efecto de las enfermedades en los árboles arterial y venoso ha sido inviable hasta ahora a pesar de su indudable utilidad. La extrema complejidad de los árboles vasculares del pulmón hace inabordable una separación manual de ambas estructuras en un tiempo realista, fomentando aún más la necesidad de diseñar herramientas automáticas o semiautomáticas para tal objetivo. Pero la ausencia de casos correctamente segmentados y etiquetados conlleva múltiples limitaciones en el desarrollo de sistemas de separación AV, en los cuales son necesarias imágenes de referencia tanto para entrenar como para validar los algoritmos. Por ello, el diseño de imágenes sintéticas de TAC pulmonar podría superar estas dificultades ofreciendo la posibilidad de acceso a una base de datos de casos pseudoreales bajo un entorno restringido y controlado donde cada parte de la imagen (incluyendo arterias y venas) está unívocamente diferenciada. En esta Tesis Doctoral abordamos ambos problemas, los cuales están fuertemente interrelacionados. Primero se describe el diseño de una estrategia para generar, automáticamente, fantomas computacionales de TAC de pulmón en humanos. Partiendo de conocimientos a priori, tanto biológicos como de características de imagen de CT, acerca de la topología y relación entre las distintas estructuras pulmonares, el sistema desarrollado es capaz de generar vías aéreas, arterias y venas pulmonares sintéticas usando métodos de crecimiento iterativo, que posteriormente se unen para formar un pulmón simulado con características realistas. Estos casos sintéticos, junto a imágenes reales de TAC sin contraste, han sido usados en el desarrollo de un método completamente automático de segmentación/separación AV. La estrategia comprende una primera extracción genérica de vasos pulmonares usando partículas espacio-escala, y una posterior clasificación AV de tales partículas mediante el uso de Graph-Cuts (GC) basados en la similitud con arteria o vena (obtenida con algoritmos de aprendizaje automático) y la inclusión de información de conectividad entre partículas. La validación de los fantomas pulmonares se ha llevado a cabo mediante inspección visual y medidas cuantitativas relacionadas con las distribuciones de intensidad, dispersión de estructuras y relación entre arterias y vías aéreas, los cuales muestran una buena correspondencia entre los pulmones reales y los generados sintéticamente. La evaluación del algoritmo de segmentación AV está basada en distintas estrategias de comprobación de la exactitud en la clasificación de vasos, las cuales revelan una adecuada diferenciación entre arterias y venas tanto en los casos reales como en los sintéticos, abriendo así un amplio abanico de posibilidades en el estudio clínico de enfermedades cardiopulmonares y en el desarrollo de metodologías y nuevos algoritmos para el análisis de imágenes pulmonares. ABSTRACT Computed tomography (CT) is the reference image modality for the study of lung diseases and pulmonary vasculature. Lung vessel segmentation has been widely explored by the biomedical image processing community, however, differentiation of arterial from venous irrigations is still an open problem. Indeed, automatic separation of arterial and venous trees has been considered during last years as one of the main future challenges in the field. Artery-Vein (AV) segmentation would be useful in different medical scenarios and multiple pulmonary diseases or pathological states, allowing the study of arterial and venous irrigations separately. Features such as density, geometry, topology and size of vessels could be analyzed in diseases that imply vasculature remodeling, making even possible the discovery of new specific biomarkers that remain hidden nowadays. Differentiation between arteries and veins could also enhance or improve methods processing pulmonary structures. Nevertheless, AV segmentation has been unfeasible until now in clinical routine despite its objective usefulness. The huge complexity of pulmonary vascular trees makes a manual segmentation of both structures unfeasible in realistic time, encouraging the design of automatic or semiautomatic tools to perform the task. However, this lack of proper labeled cases seriously limits in the development of AV segmentation systems, where reference standards are necessary in both algorithm training and validation stages. For that reason, the design of synthetic CT images of the lung could overcome these difficulties by providing a database of pseudorealistic cases in a constrained and controlled scenario where each part of the image (including arteries and veins) is differentiated unequivocally. In this Ph.D. Thesis we address both interrelated problems. First, the design of a complete framework to automatically generate computational CT phantoms of the human lung is described. Starting from biological and imagebased knowledge about the topology and relationships between structures, the system is able to generate synthetic pulmonary arteries, veins, and airways using iterative growth methods that can be merged into a final simulated lung with realistic features. These synthetic cases, together with labeled real CT datasets, have been used as reference for the development of a fully automatic pulmonary AV segmentation/separation method. The approach comprises a vessel extraction stage using scale-space particles and their posterior artery-vein classification using Graph-Cuts (GC) based on arterial/venous similarity scores obtained with a Machine Learning (ML) pre-classification step and particle connectivity information. Validation of pulmonary phantoms from visual examination and quantitative measurements of intensity distributions, dispersion of structures and relationships between pulmonary air and blood flow systems, show good correspondence between real and synthetic lungs. The evaluation of the Artery-Vein (AV) segmentation algorithm, based on different strategies to assess the accuracy of vessel particles classification, reveal accurate differentiation between arteries and vein in both real and synthetic cases that open a huge range of possibilities in the clinical study of cardiopulmonary diseases and the development of methodological approaches for the analysis of pulmonary images.

Strategies for delaying pest resistance to genetically modified crops that produce Bacillus thuringiensis (Bt) toxins are based primarily on theoretical models. One key assumption of such models is that genes conferring resistance are rare. Previous estimates for lepidopteran pests targeted by Bt crops seem to meet this assumption. We report here that the estimated frequency of a recessive allele conferring resistance to Bt toxin Cry1Ac was 0.16 (95% confidence interval = 0.05–0.26) in strains of pink bollworm (Pectinophora gossypiella) derived from 10 Arizona cotton fields during 1997. Unexpectedly, the estimated resistance allele frequency did not increase from 1997 to 1999 and Bt cotton remained extremely effective against pink bollworm. These results demonstrate that the assumptions and predictions of resistance management models must be reexamined.

Theories of image segmentation suggest that the human visual system may use two distinct processes to segregate figure from background: a local process that uses local feature contrasts to mark borders of coherent regions and a global process that groups similar features over a larger spatial scale. We performed psychophysical experiments to determine whether and to what extent the global similarity process contributes to image segmentation by motion and color. Our results show that for color, as well as for motion, segmentation occurs first by an integrative process on a coarse spatial scale, demonstrating that for both modalities the global process is faster than one based on local feature contrasts. Segmentation by motion builds up over time, whereas segmentation by color does not, indicating a fundamental difference between the modalities. Our data suggest that segmentation by motion proceeds first via a cooperative linking over space of local motion signals, generating almost immediate perceptual coherence even of physically incoherent signals. This global segmentation process occurs faster than the detection of absolute motion, providing further evidence for the existence of two motion processes with distinct dynamic properties.

The measurement of dipolar contributions to the splitting of 15N resonances of 1H-15N amide pairs in multidimensional high-field NMR spectra of field-oriented cyanometmyoglobin is reported. The splittings appear as small field-dependent perturbations of normal scalar couplings. Assignment of more than 90 resonances to specific sequential sites in the protein allows correlation of the dipolar contributions with predictions based on the known susceptibility and known structure of the protein. Implications as an additional source of information for protein structure determination in solution are discussed.

Increases in plasma cholesterol are associated with progressive increases in the risk of atherosclerotic cardiovascular disease. In humans plasma cholesterol is contained primarily in apolipoprotein B-based low density lipoprotein (LDL). Cells stop making the high-affinity receptor responsible for LDL removal as they become cholesterol replete; this slows removal of LDL from plasma and elevates plasma LDL. As a result of this delayed uptake, hypercholesterolemic individuals not only have more LDL but have significantly older LDL. Oxidative modification of LDL enhances their atherogenicity. This study sought to determine whether increased time spent in circulation, or aging, by lipoprotein particles altered their susceptibility to oxidative modification. Controlled synchronous production of distinctive apolipoprotein B lipoproteins (yolk-specific very low density lipoproteins; VLDLy) with a single estrogen injection into young turkeys was used to model LDL aging in vivo. VLDLy remained in circulation for at least 10 days. Susceptibility to oxidation in vitro was highly dependent on lipoprotein age in vivo. Oxidation, measured as hexanal release from n-6 fatty acids in VLDLy, increased from 13.3 +/- 5.5 nmol of 2-day-old VLDLy per ml, to 108 +/- 17 nmol of 7-day-old VLDLy per ml. Oxidative instability was not due to tocopherol depletion or conversion to a more unsaturated fatty acid composition. These findings establish mathematically describable linkages between the variables of LDL concentration and LDL oxidation. The proposed mathematical models suggest a unified investigative approach to determine the mechanisms for acceleration of atherosclerotic cardiovascular disease risk as plasma cholesterol rises.

The receptor tyrosine kinases (RTKs) c-kit and platelet-derived growth factor receptor alpha chain (PDG-FRa) are encoded at the white spotting (W) and patch (Ph) loci on mouse chromosome 5. While W mutations affect melanogenesis, gametogenesis, and hematopoiesis, the Ph mutation affects melanogenesis and causes early lethality in homozygotes. W-sash (Wsh) is an expression mutation and blocks c-kit expression in certain cell types and enhances c-kit expression in others, including at sites important for early melanogenesis. We have determined the effect of Ph on c-kit expression during embryogenesis in Ph heterozygotes. Immunohistochemical analysis revealed enhanced c-kit expression in several cell types, including sites important for early melanogenesis. We propose that in both Wsh and Ph mutant mice c-kit misexpression affects early melanogenesis and is responsible for the pigment deficiency. Moreover, we have defined the organization of the RTKs in the W/Ph region on chromosome 5 and characterized the Wsh mutation by using pulsed-field gel electrophoresis. Whereas the order of the RTK genes was determined as Pdgfra-c-kit-flk1, analysis of the Wsh mutation revealed that the c-kit and Pdgfra genes are unlinked in Wsh, presumably because of an inversion of a small segment of chromosome 5. The Ph mutation consists of a deletion including Pdgfra and the 3' deletion endpoint of Ph lies between Pdgfra and c-kit. Therefore, positive 5' upstream elements controlling c-kit expression in mast cells and some other cell types are affected by the Wsh mutation and negative elements are affected by both the Wsh and the Ph mutation.

La ricerca di nuove strategie per la rigenerazione ossea rappresenta un focus di interesse centrale per migliorare la gestione di casi clinici complessi nell’ambito della chirurgia orale e maxillo-facciale. Uno degli approcci più utilizzati in tale contesto si basa sull’utilizzo di molecole con proprietà osteoinduttive e molte sostanze sono state fino ad oggi sperimentate. E’ noto in letteratura che gli androgeni svolgono un ruolo chiave nella regolazione della morfogenesi ossea e nel mantenimento della sua omeostasi durante il corso della vita. Questo lavoro di tesi nasce dall’ipotesi che la somministrazione locale di tali ormoni, eventualmente combinata a materiali da innesto, possa favorire la guarigione di difetti ossei. Stando a questa premessa, sono stati valutati gli effetti dello steroide sintetico Stanozololo sulla rigenerazione ossea in diversi settings sperimentali. La tesi è strutturata secondo un percorso che segue le fasi della ricerca, attraverso sperimentazioni in vitro e in vivo; ogni capitolo può essere approcciato come uno studio a sé stante, corrispondente ad una determinata tappa dell’iter sperimentale. Sulla base di questi intenti, viene fornito inizialmente un quadro d’insieme circa gli effetti degli androgeni sull’osso. A seguire, è presentata una sperimentazione in vitro nella linea cellulare SaOS-2. Infine, è proposta un’innovativa metodologia di analisi per lo studio della rigenerazione ossea nel modello di ratto, ove viene testata la somministrazione locale di Stanozololo combinato a materiale da innesto.

We investigate the critical properties of the four-state commutative random permutation glassy Potts model in three and four dimensions by means of Monte Carlo simulations and a finite-size scaling analysis. By using a field programmable gate array, we have been able to thermalize a large number of samples of systems with large volume. This has allowed us to observe a spin-glass ordered phase in d=4 and to study the critical properties of the transition. In d=3, our results are consistent with the presence of a Kosterlitz-Thouless transition, but also with different scenarios: transient effects due to a value of the lower critical dimension slightly below 3 could be very important.

The last two decades have been marked by a growing public awareness of family violence. Research by social scientists has suggested that family violence is widespread (Gelles and Straus, 1988). It is estimated that every year 1.8 to 4 million women are physically abused by their partners (Novello, 1992). In fact, more women are abused by their husbands or boyfriends than are injured in car accidents, muggings, or rapes (Jaffe, Wolfe, and Wilson, 1990). A recent prevalence study by Fantuzzo, Boruch, Beriama, Atkins, and Marcus (1997) found that children were disproportionately present in households where there was a substantial incident of adult female assault. Experts estimate that 3.3 to 10 million children are exposed to marital violence each year (Carlson, 1984; Straus, 1991). Until recently, most researchers did not consider the impact of parental conflict on the children who witness this violence. The early literature in this field primarily focused on the incidence of violence against women and the inadequate response of community agencies (Jaffe et al, 1990). The needs of children were rarely considered. However, researchers have become increasingly aware that children exposed to marital violence are victims of a range of psychological maltreatment (e.g., terrorizing, isolation;Hart, Brassared & Karlson, 1996) and are at serious risk for the development of psychological problems (Fantuzzo, DePaola, Lambert, Martino, Anderson, and Sutton, 1991). Jouriles, Murphy and O'Leary (1989) found that children of battered women were four times more likely to exhibit psychopathology as were children living in non-violent homes. Further, researchers have found associations between childhood exposure to parental violence and the expression of violence in adulthood (Carlson, 1990). Existing research suggests that children who have witnessed marital violence manifest numerous emotional, social, and behavioral problems (Sternberg et al., 1993; Fantuzzo et al., 1991; Jaffe et al, 1990). Studies have found that children of battered women exhibit more internalizing and externalizing behavior problems than non-witnesschildren (Hughes and Fantuzzo, 1994; McCloskey, Figueredo, and Koss, 1995). In addition, children exposed to marital violence have been found to exhibit difficulties with social problem-solving, and have lower levels of social competence than nonwitnesses (Rosenberg, 1987; Moore, Pepler, Weinberg, Hammond, Waddell, & Weiser, 1990). Other reported difficulties include low self esteem (Hughes, 1988), poor school performance (Moore et al., 1990) and problems with aggression (Holden & Ritchie, 1991; Jaffe, Wolfe, Wilson, & Zak, 1986). Further, within the last decade, researchers have found that some children are traumatized by the witnessing experience, showing elevated levels of posttraumatic stress symptoms (Devoe & Graham-Bermann, 1997; Rossman, Bingham, & Emde, 1996; Kilpatrick, Litt, & Williams, 1997). These findings corroborate clinical reports that describe many exposed children as experiencing trauma reactions. It appears that the negative effects of witnessing marital violence are numerous and varied, ranging from mild emotional and behavioral problems to clinically significant levels of posttraumatic stress symptoms. These incidence figures and research findings indicate that children's exposure to violence is a significant problem in our nation today and has serious implications for the future.
