We know from the CMB and observations of large-scale structure that the universe is extremely flat, homogenous, and isotropic. The current favored mechanism for generating these characteristics is inflation, a theorized period of exponential expansion of the universe that occurred shortly after the Big Bang. Most theories of inflation generically predict a background of stochastic gravitational waves. These gravitational waves should leave their unique imprint on the polarization of the CMB via Thompson scattering. Scalar perturbations of the metric will cause a pattern of polarization with no curl (E-mode). Tensor perturbations (gravitational waves) will cause a unique pattern of polarization on the CMB that includes a curl component (B-mode). A measurement of the ratio of the tensor to scalar perturbations (r) tells us the energy scale of inflation. Recent measurements by the BICEP2 team detect the B-mode spectrum with a tensor-to-scalar ratio of r = 0.2 (+0.05, −0.07). An independent confirmation of this result is the next step towards understanding the inflationary universe.

This thesis describes my work on a balloon-borne polarimeter called SPIDER, which is designed to illuminate the physics of the early universe through measurements of the cosmic microwave background polarization. SPIDER consists of six single-frequency, on-axis refracting telescopes contained in a shared-vacuum liquid-helium cryostat. Its large format arrays of millimeter-wave detectors and tight control of systematics will give it unprecedented sensitivity. This thesis describes how the SPIDER detectors are characterized and calibrated for flight, as well as how the systematics requirements for the SPIDER system are simulated and measured.

A leptospirose humana é uma doença infecciosa aguda de amplo espectro clínico e que cursa com alterações metabólicas e dislipidêmicas envolvendo colesterol total e frações, triglicerídeos e ácidos graxos não esterificados (AGNEs). Dentre os mecanismos celulares envolvidos na sua fisiopatologia encontram-se a inibição da enzima Na, K ATPase pela endotoxina GLP e a lipotoxicidade, ambos agravados pela redução dos níveis circulantes da albumina, molécula que exerce um papel fundamental na adsorção de moléculas lipídicas. Neste estudo observacional, determinamos as concentrações séricas de bilirrubina, creatinina e albumina e, pela técnica de cromatografia líquida de alta performance, a concentração sérica dos AGNEs de cadeia longa (C16: C18) de 27 pacientes com síndrome de Weil durante o período de internação hospitalar, dos quais cinco vieram a falecer. Verificamos correlações significantes (p<0,05) ao longo da internação hospitalar, nas concentrações séricas de marcadores bioquímicos de gravidade da doença (bilirrubina, creatinina e albumina), AGNEs, ácido oléico e ácido linoléico, e relação molar ácido oléico/ albumina, com r (Pearson) de -0,7981, -0,7699, 0,9014, -0,8795 -0,9816, -0,9694, -0,9821, respectivamente. A relação molar ácido oléico/ albumina e ácido oléico+ linoléico/albumina foi significantemente mais elevada nos pacientes que faleceram (p<0,001), retornando aos valores semelhantes aos do grupo controle nos pacientes que evoluíram para a cura. Na análise por Curva Roc, a relação molar ácido oléico/albumina se mostrou um bom teste preditivo, com valor de corte 0,705 associado com maior especificidade e sensibilidade prognóstica. Nossos resultados sugerem que a utilização parenteral da albumina humana em pacientes com leptospirose pode ser uma potente ferramenta terapêutica nos casos mais graves ao interferir positivamente no resgate do equilíbrio bioquímico das relações molares ácido oléico/ albumina e ácido oléico+linoléico/albumina.

O objetivo deste estudo foi avaliar o efeito do fator C na resistência adesiva à dentina humana de um compósito à base de silorano, comparando-o com um compósito à base de metacrilato. Foram utilizados 40 molares humanos hígidos. Em metade da amostra foi realizado um preparo do tipo Classe I, com dimensões de 4x4 mm e 2 mm de profundidade, enquanto na outra metade foi realizado um preparo virtual (flat) com auxílio de uma matriz de teflon branco, com as mesmas dimensões da cavidade (4x4x2 mm), apoiada na superfície de dentina exposta. Cada grupo foi divido em dois, de acordo com o sistema restaurador: Sistema Adesivo P90 e resina Filtek P90; Adper Scotchbond Multiuso e resina Filtek Z350 XT (3M ESPE). Após a aplicação do sistema adesivo segundo as especificações do fabricante, as cavidades (real e virtual) foram preenchidas com as resinas (Z350 XT e P90) em dois incrementos. Foi realizada a fotoativação de cada incremento durante 25s. Após uma semana, as coroas foram seccionadas, obtendo-se espécimes constituídos de resina/dente na forma de um paralelepípedo (palitos). Estes palitos foram utilizados para o teste de microtração (Emic), onde a resistência adesiva na unidade MPa foi calculada. Os resultados foram avaliados estatisticamente pelo teste de análise de variância ANOVA One-Way (p = 0,05), e pelo teste de múltiplas comparações Student Newman Keuls. Foi observada uma diferença estatística entre a resina P90 (flat= 16,137,7 MPa e Cavidade= 16,759,0 MPa) e a Z350 XT (flat= 25,8211,6 MPa e Cavidade=24,4713,5 MPa), independentemente do tipo de cavidade restaurada. E não foi possível observar diferença estatística entre os tipos de cavidades (real ou virtual) restauradas com a mesma resina. Através dos testes realizados, foi possível concluir que: o fator C não afetou a resistência adesiva dos dois sistemas restauradores utilizados, e o tipo de sistema restaurador afeta a resistência adesiva, sendo que o sistema à base de silorano apresenta resultados inferiores ao de metacrilato.

Trawling and dredging on Georges Bank (northwest Atlantic Ocean) have altered the cover of colonial epifauna, as surveyed through in situ photography. A total of 454 photographs were analyzed from areas with gravel substrate between 1994 and 2000 at depths of 40–50 m and 80–90 m. The cover of hydroids, bushy bryozoans, sponges, and tubeworms was generally higher at sites undisturbed by fishing than at sites classified as disturbed. The magnitude and significance of this effect depended on depth and year. Encrusting bryozoans were the only type of colonial epifauna positively affected by bottom fishing. Species richness of noncolonial epifauna declined with increased bottom fishing, but Simpson’s index of diversity typically peaked at intermediate levels of habitat disturbance. Species that were more abundant at undisturbed sites possessed characteristics that made them vulnerable to bottom fishing. These characteristics include emergent growth forms, soft body parts, low motility, use of complex microhabitats, long life spans, slow growth, and larval dispersal over short distances. After the prohibition of bottom fishing at one site, both colonial and noncolonial species increased in abundance. Populations of most taxa took two years or more to increase after the fishing closure. This finding indicates that bottom fishing needs to be reduced to infrequent intervals to sustain the benthic species composition of Georges Bank at a high level of biodiversity and abundance.

Variation in the allele frequencies of five microsatellite loci was surveyed in 1256 individual spotted seatrout (Cynoscion nebulosus) obtained from 12 bays and estuaries from Laguna Madre, Texas, to Charlotte Harbor, Florida, to St. John’s River on the Florida Atlantic Coast. Texas and Louisiana collection sites were resampled each year for two to four years (1998−2001). Genetic differentiation was observed. Spotted seatrout from Florida waters were strongly differentiated from spotted seatrout collected in Louisiana and Texas. The greatest genetic discontinuity was observed between Tampa Bay and Charlotte Harbor, and Charlotte Harbor seatrout were most similar to Atlantic Coast spotted seatrout. Texas and Louisiana samples were not strongly structured within the northwestern Gulf of Mexico and there was little evidence of temporal differentiation within bays. These findings are contrary to those of earlier analyses with allozymes and mitochondrial DNA (mtDNA) where evidence of spatial differentiation was found for spotted seatrout resident on the Texas coast. The differences in genetic structure observed among these markers may reflect differences in response to selective pressure, or may be due to differences in underlying genetic processes.

No presente estudo, proponho-me a mostrar narrativas de/sobre mulheres negras nascidas e/ou radicadas na cidade do Rio de Janeiro que, por se manifestarem publicamente por intermédio do exercício de práticas artístico-culturais, são contempladas pela visibilidade midiática. Trata-se de uma pesquisa qualitativa situada nos campos dos estudos pós-coloniais e dos estudos do cotidiano. Nega Gizza e Tati Quebra Barraco protagonizam a investigação. As narrativas foram coletadas em artefatos culturais midiatizados disponibilizados ao público em geral cotidianamente, ao longo do século XXI. Achados apontam que a cidade do Rio de Janeiro, o lugar do estudo, é um centro propulsor de narrativas, discursos e dispositivos atuantes nacionalmente sobre mulheres negras, os quais mantêm aspectos essencializantes e subalternizantes que sobrepujam supostos fatores positivos que seriam decorrentes de visibilidades, bem como limitam processos de ressignificações identitárias e processos empoderadores. Destacam-se: 1) o discurso sobre a feiura, que se contrapõe ao discurso estético corporal hegemônico; 2) o discurso do corpo negro suspeito. Sendo assim, o dispositivo de transformar negra/o em branca/o embranquecimento continua operando com muita potência na cidade do Rio de Janeiro, em especial tendo em vista aquilo que se espera das mulheres. Tais achados inspiraram a construção da seguinte trilogia: protagonistas impensáveis; muralha do quase; prestígio subalterno. O prestígio subalterno se manifesta por intermédio da mobilidade social restrita a lugares marcados racializados. A muralha do quase é a responsável pela ausência de redes de poder com as quais a população negra possa contar para galgar a mobilidade social. As protagonistas impensáveis são as sujeitas que, por não disporem de atributos ditados pelo discurso da legitimidade estética que merece ser representada, estão interditadas para ocupar o papel principal do espetáculo. Paradoxalmente, no entanto, em algum momento inusitado, elas levantam e falam.

In July 2006, a mandatory observer program was implemented to characterize the commercial reef fish fishery operating in the U.S. Gulf of Mexico. The primary gear types assessed included bottom longline and vertical line (bandit and handline). A total of 73,205 fish (183 taxa) were observed in the longline fishery. Most (66%) were red grouper, Epinephelus morio, and yellowedge grouper, E. flavolimbatus. In the vertical line fishery, 89,015 fish (178 taxa) were observed of which most (60%) were red snapper, Lutjanus campechanus, and vermilion snapper, Rhomboplites aurorubens. Based on surface observations of discarded under-sized target and unwanted species, the majority of fish were released alive; minimum assumed mortality was 23% for the vertical line and 24% for the bottom longline fishery. Of the individuals released alive in the longline fishery, 42% had visual signs of barotrauma stress (air bladder expansion/and or eyes protruding). In the vertical line fishery, 35% of the fish were released in a stressed state. Red grouper and red snapper size composition by depth and gear type were determined. Catch-per-unit-effort for dominant species in both fisheries, illustrated spatial differences in distribution between the eastern and western Gulf. Hot Spot Analyses for red grouper and red snapper identified areas with significant clustering of high or low CPUE values.

Pacific hake, Merluccius productus, the most abundant groundfish in the California Current Large Marine Ecosystem (CCLME), is a species of both commercial significance, supporting a large international fishery, and ecological importance, connecting other species as both predator and prey. Coastal Pacific hake migrations are characterized by movements between northern summer feeding areas and southern winter spawning areas, with variations in annual abundance, distribution, and the extent of these movements associated with varying climate-ocean conditions. In general, warm (cool) years with enhanced (reduced) stratification and poleward (equatorward) transport are often related to good (poor) recruitment, increased (decreased) northward distribution, and reduced (enhanced) growth. However, the classic periodic pattern of annual migration and distribution may no longer be fully representative. Based on recent advances in the understanding of climate-ocean variability off the U.S. west coast, we hypothesize that the annual movements of Pacific hake are more responsive to climate-ocean variability than previously thought, and further, that changes observed in Pacific hake distributions may reflect long-term changes in climate-ocean conditions in the CCLME. Therefore, an updated model of these relations is key to effective monitoring and management of this stock, as well as to devising scenarios of future change in the CCLME as a result of climate variations. The current state of knowledge of the relationship between the Pacific hake and its environment is reviewed, highlighting emerging ideas compared to those of the past, and priorities for future research are suggested.

Fishery observers collected data from 307 tows during 96 trips aboard skimmer trawl vessels in Louisiana’s coastal waters from September 2004 through June 2005 to estimate catch rates of target and nontarget species, including sea turtles (Cheloniidae and Dermochelyidae), by area and season during commercial shrimping operations. About 16,965.7 kg of total catch were recorded during 517.0 hours of fishing operations. Based on weight extrapolations from species composition samples, penaeid shrimp (Penaeidae) dominated the catch at 66%, followed by finfish at 19%, nonpenaeid shrimp crustaceans at 7%, discarded penaeid shrimp at 6%, and debris at 3%. Noncrustacean invertebrates comprised less than 1%. Catch rates in kilograms per hour by category was 21.6 for penaeid shrimp, 6.2 for finfish, 2.2 for nonpenaeid crustaceans, 1.8 for discarded penaeid shrimp, and 0.9 for debris. White shrimp, Litopenaeus setiferus, other penaeid shrimp, and Gulf menhaden, Brevoortia patronus, were the top three dominant species by weight. Seasonally, a higher catch rate was observed from May through August 2005 for penaeid shrimp as compared with the September through December 2004 period. Conversely, the September through December 2004 period experienced a higher catch rate for finfish than during May through August 2005. No sea turtle interactions were documented.

Guam's nearshore reef fishery is a multi-gear, multispecies fishery that has undergone major changes through the years. Methods have evolved and become more modern. This, along with the changing economic status of Guam, has severely stressed the fishery. Top targeted species are being overexploited and "growth overharvesting" is occurring; the more serious form of "recruitment overharvesting," is happening to some of the key species. Major management concerns are discussed with respect to overfishing and habitat destruction. Management recommendations for this fishery include gear restrictions, size restrictions, and the establishment of marine conservation areas.

O presente trabalho investiga os diferentes processos sociais relacionados ao surgimento do transtorno de estresse pós-traumático (TEPT) no contexto brasileiro. Categoria diagnóstica norte-americana instituída na década de 1980 pela terceira edição do Manual Diagnóstico e Estatístico de Transtornos Mentais (Diagnostic and Statistical Manual of Mental Disorders DSM-III) uma publicação da Associação Americana de Psiquiatria (American Psychiatric Association APA) o TEPT tornou-se, desde a sua aparição, uma das categorias nosológicas mais difundidas, estudadas e diagnosticadas da psiquiatria contemporânea. A partir do cotidiano de um laboratório de pesquisa e tratamento do TEPT, de análises conversacionais dos atendimentos médicos, de um estudo das diferentes escalas psiquiátricas utilizadas no acompanhamento dos pacientes e de pesquisas sobre a mídia relacionada às experiências traumáticas buscou-se entender os entrelaçamentos entre os processos de difusão e a construção da legitimidade da categoria diagnóstica do TEPT. A abordagem aqui proposta pretende ir além do aparente dilema entre uma concepção medicalizada que assumem a existência o TEPT como um fenômeno natural e as abordagens sócio-antropológicas que veem o TEPT como uma experiência culturalmente construída. Por fim, pretendo mostrar, pela investigação dos alicerces políticos e culturais dos denominados transtornos mentais, que o estatuto social dos diagnósticos e dos tratamentos da moderna psiquiatria só pode ser compreendido tendo como referência as dinâmicas de longo prazo nas sociedades contemporâneas.

In July 2007, a mandatory Federal observer program was implemented to characterize the U.S. Gulf of Mexico penaeid shrimp (Farfantepenaeus aztecus, F. duorarum, and Litopenaeus setiferus) fishery. In June 2008, the program expanded to include the South Atlantic penaeid and rock shrimp, Sicyonia spp., fisheries. Data collected from 10,206 tows during 5,197 sea days of observations were analyzed by geographical area and target species. The majority of tows (~70%) sampled were off the coasts of Texas and Louisiana. Based on total hours towed, the highest concentrated effort occurred off South Texas and southwestern Florida. Gear information, such as net characteristics, bycatch reduction devices, and turtle excluder devices were fairly consistent among areas and target species. By species categories, finfish comprised the majority (≥57%) of the catch composition in the Gulf of Mexico and South Atlantic penaeid shrimp fisheries, while in the South Atlantic rock shrimp fishery the largest component (41%) was rock shrimp. Bycatch to shrimp ratios were lower than reported in previous studies for the Gulf of Mexico penaeid shrimp fishery. These decreased ratios may be attributed to several factors, notably decreased shrimp effort and higher shrimp catch per unit of effort (CPUE) in recent years. CPUE density surface plots for several species of interest illustrated spatial differences in distribution. Hot Spot Analyses for shrimp (penaeid and rock) and bycatch species identified areas with significant clustering of high or low CPUE values. Spatial and temporal distribution of protected species interactions were documented.

The Ecological Society of America and NOAA's Offices of Habitat Conservation and Protected Resources sponsored a workshop to develop a national marine and estuarine ecosystem classification system. Among the 22 people involved were scientists who had developed various regional classification systems and managers from NOAA and other federal agencies who might ultimately use this system for conservation and management. The objectives were to: (1) review existing global and regional classification systems; (2) develop the framework of a national classification system; and (3) propose a plan to expand the framework into a comprehensive classification system. Although there has been progress in the development of marine classifications in recent years, these have been either regionally focused (e.g., Pacific islands) or restricted to specific habitats (e.g., wetlands; deep seafloor). Participants in the workshop looked for commonalties across existing classification systems and tried to link these using broad scale factors important to ecosystem structure and function.

Marine mammals, such as dolphins, can serve as key indicator species in coastal areas by reflecting the effects of natural and anthropogenic stressors. As such they are often considered sentinels of environmental and ecosystem health (Bossart 2006; Wells et al. 2004; Fair and Becker 2000). The bottlenose dolphin is an apex predator and a key component of many estuarine environments in the southeastern United States (Woodward-Clyde Consultants 1994; SCDNR 2005). Health assessments of dolphins are especially critical in areas where populations are depleted, show signs of epidemic disease and/or high mortality and/or where habitat is being altered or impacted by human activities. Recent assessments of environmental conditions in the Indian River Lagoon, Florida (IRL) and the estuarine waters surrounding Charleston, South Carolina (CHS) highlight the need for studies of the health of local bottlenose dolphins. While the condition of southeastern estuaries was rated as fair in the National Coastal Condition Report (U.S. EPA 2001), it was noted that the IRL was characterized by poorer than expected benthic communities, significant sediment toxicity and increased nutrient concentrations. Similarly, portions of the CHS estuary have sediment concentrations of aliphatic aromatic hydrocarbons, select inorganic metals, and some persistent pesticides far in excess of reported bioeffect levels (Hyland et al. 1998). Long-term trends in water quality monitoring and recent scientific research suggest that waste load assimilation, non-point source runoff impacts, contaminated sediments, and toxic pollutants are key issues in the CHS estuary system. Several ‘hot spots’ with high levels of heavy metals and organic compounds have been identified (Van Dolah et al. 2004). High concentrations of anthropogenic trace metals, polychlorinated biphenyls (PCB’s) and pesticides have been found in the sediments of Charleston Harbor, as well as the Ashley and Cooper Rivers (Long et al. 1998). Two superfund sites are located within the CHS estuary and the key contaminants of concern associated with these sites are: polycyclic aromatic hydrocarbons (PAH), lead, chromium, copper, arsenic, zinc and dioxin. Concerns related to the overall health of IRL dolphins and dermatologic disease observed in many dolphins in the area (Bossart et al. 2003) initiated an investigation of potential factors which may have impacted dolphin health. From May-August 2001, 35 bottlenose dolphins died in the IRL during an unusual mortality event (MMC 2003). Many of these dolphins were diagnosed with a variety of skin lesions including proliferative ulcerative dermatitis due to protozoa and fungi, dolphin pox and a vesicular dermatopathy of unknown etiology (Bossart et al. 2003). Multiple species from fish to dolphins in the IRL system have exhibited skin lesions of various known and unknown etiologies (Kane et al. 2000; Bossart et al. 2003; Reif et al. 2006). On-going photo-identification (photo-ID) studies have documented skin diseases in IRL dolphins (Mazzoil et al. 2005). In addition, up to 70% of green sea turtles in the IRL exhibit fibropapillomas, with the highest rates of occurrence being seen in turtles from the southern IRL (Hirama 2001).

Karlodinium veneficum (syn. Karlodinium micrum, Bergholtz et al. 2006; J Phycol 42:170–193) is a small athecate dinoflagellate commonly present in low levels in temperate, coastal waters. Occasionally, K. veneficum forms ichthyotoxic blooms due to the presence of cytotoxic, hemolytic compounds, putatively named karlotoxins. To evaluate the anti-grazing properties of these karlotoxins, we conducted food removal experiments using the cosmopolitan copepod grazer Acartia tonsa. Wild-caught, adult female A. tonsa were exposed to 6 monoalgal or mixed algal diets made using bloom concentrations of toxic (CCMP 2064) and non-toxic (CSIC1) strains of K. veneficum. Ingestion and clearance rates were calculated using the equations of Frost (1972). Exposure to the toxic strain of K. veneficum did not contribute to an increased mortality of the copepods and no significant differences in copepod mortality were found among the experimental diets. However, A. tonsa had significantly greater clearance and ingestion rates when exposed to a monoalgal diet of the non-toxic strain CSIC1 than when exposed to the monoalgal diet of toxic strain CCMP 2064 and mixed diets dominated by this toxic strain. These results support the hypothesis that karlotoxins in certain strains of K. veneficum deter grazing by potential predators and contribute to the formation and continuation of blooms.
