There are four chapters in this dissertation. The first chapter briefly synthesizes the basic theories, methods and present-day applying situation of environmental magnetism. The second chapter probes into the magnetic mineral diagenesis in the post-glacial muddy sediments from the southeastern South Yellow Sea and its response to marine environmental changes, using the muddy sediment of Core YSDP103 formed in the shelf since about 13 ka BP. The third chapter illustrates the high-resolution early diagenetic processes by investigating the rapidly deposited muddy sediments during the last 6 ka in Cores SSDP-102 and SSDP-103 from the near-shore shelf of Korea Strait. The fourth chapter presents the results of detailed rock magnetic investigation of the surface sediments from the fine-grained depositional area on the outer shelf of the East China Sea in an attempt to provide environmental magnetic evidence for the provenance of the fine-grained deposit. Core YSDP103 was retrieved in the muddy deposit under the cold eddy of the southeastern South Yellow Sea, and the uppermost 29.79 m core represents the muddy sediments formed in the shelf since about 13 ka BP. The lower part from 29.79 to 13.35 m, called Unit A2, was deposited during the period from the post-glacial transgression to the middle Holocene (at about 6 ~(14)C ka BP) when the rising sea level reached its maximum, while the upper part above 13.35 m (called Unit Al) was deposited in a cold eddy associated with the formation of the Yellow Sea Warm Current just after the peak of post-glacial sea level rise. For the the uppermost 29.79 m core, detailed investigation of rock-magnetic properties and analyses of grain sizes and geochemistry were made. The experimental results indicate that the magnetic mineralogy of the core is dominated by magnetite, maghemite and hematite and that, except for the uppermost 2.35 m, the magnetic minerals were subject to reductive diagenesis leading to significant decline of magnetic mineral content and the proportion of low-coercivity component. More importantly, ferrimagnetic iron sulphide (greigite) is found in Unit A2 but absent in Unit Al, suggesting the control of marine environmental conditions on the magnetic mineral diagenesis. Magnetic parameters show abrupt changes across the boundary between the Unit Al and A2, which reflects a co-effect of environmental conditions and primary magnetic components of the sediments on the diagenesis. Alternating zones of high and low magnetic parameters are observed in Unit A2 of Core YSDP103, which is presumably due to periodic changes of the concentration and/or grain size of magnetic minerals carried into the study area. Cores SSDP-102 and SSDP-103, two studied sediment cores from the Korea Strait contain mud sequences (14 m and 32.62 m in thickness) that were deposited during the last 6,000 years. Analyses of grain sizes and geochemistry of the cores have demonstrated that the sediments have uniform lithology and geochemical properties, however, marked down-core changes in magnetic properties suggest that diagenesis has significantly impacted the magnetic properties. An expanded view of early diagenetic reactions that affect magnetic mineral assemblages is evident in these rapidly deposited continental shelf sediments compared to deep-sea sediments. The studied sediments can be divided into four descending intervals, based on magnetic property variations. Interval 1 is least affected by diagenesis and has the highest concentrations of detrital magnetite and hematite, and the lowest solid-phase sulfur contents. Interval 2 is characterized by the presence of paramagnetic pyrite and sharply decreasing magnetite and hematite concentrations, which suggest active reductive dissolution of detrital magnetic minerals, the absolute minimum abundance of magnetite is reached at the end of this interval. Interval 3 is marked by a progressive loss of hematite with depth, and at the base of this interval, 82% to 88% of the hematite component was depleted and the bulk magnetic mineral concentration was reduced to the lowest value in the entire studied mud section. Interval 4 has an increasing down-core enhancement of authigenic greigite, which is interpreted to have formed due to arrested pyritization resulting from consumption of pore water sulfate with depth. This is the first clear demonstration from an active depositional environment for a delay of thousands of years for acquisition of a magnetization carried by greigite. This detailed view of diagenetic processes in continental shelf sediments suggests that studies of geomagnetic field behavior from such sediments should be conducted with care. Paleoceanographic and paleoclimatic studies based on the magnetic properties of shelf sediments with high sedimentation rates like those in the Korea Strait are also unlikely to provide a meaningful signature associated with syn-depositional environmental processes. The rock magnetic properties of the surface sediments from the fine-grained depositional area on the outer shelf of the East China Sea, an area surrounded by sands, are investigated with a view to providing information on the sediment provenance. Multiple magnetic parameters such as magnetic susceptibility (%), anhysteretic remanent magnetization (ARM), saturation rernanent magnetization (SIRM), coercivities of SIRM (Her), and S ratios (relative abundance of low-coercivity magnetic minerals) are measured for all 179 surface samples, and partial representative samples are examined for their magnetic hysteresis parameters, temperature-dependence of magnetic susceptibility and x-ray diffraction spectra. Our research indicates that the magnetic mineralogy is dominated by magnetite with a small amount of hematite and is primarily of pseudo-single domain (PSD) to multidomain (MD) nature with a detrital origin. In the surface sediments, the granulometry of magnetic fractions is basically independent of grain sizes of the sediment containing the magnetic grains, and the composition of magnetic minerals remains almost homogeneous, that is, with a relatively constant ratio of low to high coercivity fraction throughout the area. The magnetic concentration in the study area generally decreases to the east or southeast accompanied by magnetic-particle fining to the east or to the northeast. The geographic pattern of magnetic properties is most reasonably explained by a major source of sediment jointly from the erosion of the old Huanghe River deposit and the discharge of the Changjiang River. The rock magnetic data facilitate understanding of the transport mechanism of fine-grained sediments in the outer shelf of the East China Sea.

2002年12月,国家环保总局颁布了《城镇污水处理厂污染物排放标准》(GB18918-2002),对我国城镇污水处理厂的出水水质,尤其是对出水的氮、磷指标提出了更加严格的要求。这不仅对污水处理厂的工艺设计提出了挑战,而且对其运行工艺提出了更高要求,因此污水处理厂面临着进行工艺优化及达标改造以满足新的污水排放标准的问题。 本文以沈阳北部污水处理厂为研究对象,结合国际水质协会(IWA)开发推荐的活性污泥模型(ASM2D),按照设备评估和分析、数据收集和分析、活性污泥模型的设计和校正、方案评估及改造建议4个阶段对该污水处理厂进行了工艺优化分析及模拟优化技术研究,并运用收集到的原始数据对活性污泥模型(ASM2D)进行了校正。随后,在模型校正成功基础上,对北部污水处理厂三种可能的污水设计处理工艺(A2/O工艺、5段式 Bardenpho工艺、组合工艺)进行了预测及筛选,选定了组合工艺作为北部污水处理厂的最佳脱氮除磷改造方案,确保了沈阳北部污水处理厂污水排放稳定达标。同时,为沈阳市其它城市污水处理厂的污水排放达标改造提供了借鉴。 该研究对提高城市污水处理厂设计和运行管理水平,减少投资和运行成本,提高处理效果,逐渐缩小我国在模型研究和应用方面与国外的差距,在实际应用方面及理论研究方面都具有重要意义。

湖南芙蓉超大型锡矿田位于湘南千里山-骑田岭地区南西部,骑田岭花岗岩体南部。该矿田是郴州-蓝山北东向W-Sn-Pb-Zn成矿代的重要组成部分。本论文在深入细致的野外地质工作基础上,选取有代表性的样品,首先进行了系统的显微鉴定、流体包裹体观察等,然后通过同位素年代学、微量元素、稀土元素和稳定同位素同位素(S、Pb、稀有气体)对比研究,探讨了矿田的成矿物质来源、成矿流体来源及骑田岭A型花岗岩岩浆活动与成矿的关系。最后初步建立了矿田的成矿理论机制。论文取得的主要认识如下: (1)芙蓉锡矿田是新近发现的一个与骑田岭花岗岩有关的超大型锡矿田。该矿田具有有利的成矿地质背景,其规模超大、Sn品位高、找矿潜力大。该矿田所赋存的骑田岭花岗岩样品投影点都落入A2亚类花岗岩范围内,是至今为止发现的与A型花岗岩有关(目前对该种新型锡矿类型研究还较少)的一个罕见的超大型锡矿田。 (2)芙蓉超单元岩体成岩同位素年龄大都在151~160Ma左右,该单元与成矿有关的岩体同位素年龄大都在136~160Ma,本文最新研究则表明芙蓉锡矿田主成矿期为150~160Ma。该矿区成岩与成矿时间比较吻合,正处于我国东南部中生代大规模的陆内岩石圈拉张和伸展,南北向构造体制向东西向的另一个构造体制转换,从而造成我国东南部成矿大爆发的独特地质背景时期。 (3)成矿物质、成矿流体多来源:矿田矿石硫化物流体包裹体的He同位素地球化学研究结果表明,该矿田成矿流体具壳幔两端员混合的特征,其中壳源组分主要由矿区骑田岭花岗岩提供,而幔源组分则可能与华南中生代大规模的地幔物质上涌、岩石圈发生拉张、伸展作用有关;Pb同位素组成表明,芙蓉超大型锡矿田成矿流体中的铅主要来源于上地壳,主要由矿区骑田岭A型花岗岩的岩浆热液提供;S同位素组成表明,该矿田成矿流体中的硫非单一的骑田岭花岗岩岩浆来源,同时有地层硫、生物硫及地幔硫等多种来源硫的混入;岩石、矿石及矿石硫化物的REE地球化学研究结果也进一步表明,在骑田岭花岗岩的岩浆热液作用阶段,岩石圈发生拉张、伸展作用产生的多期次的高温热液参与了矿田的成矿作用。 (4)成矿流体非均一化:矿石的REE地球化学以及S、He同位素组成分布特征表明,该矿田成矿流体均一化程度不完全。 (5)构造、岩浆岩及骑田岭花岗岩与成矿存在密切的关系:芙蓉锡矿田的构造控矿模式是邵阳-郴州断裂及炎陵-郴州-蓝山断裂为深源成矿物质的上涌提供了通道,郴州-临武大断裂为含矿流体的贯入提供了通道,而骑田岭矿区内发育的大小褶皱及断裂为矿质提供了储存空间,并直接控制了矿体的形成和分布;燕山期大规模、多期次的岩浆活动,及区内广泛分布的中酸性花岗岩类与该区Sn、Pb、Zn等有色金属成矿关系密切;区域大面积骑田岭A型花岗岩不仅在时空上与该区成矿密切相关,而且在成矿过程中起到了提供主要成矿物质、成矿流体等重要作用。 (6)初步建立了矿田成矿机制:在骑田岭岩体的岩浆作用阶段,由于中生代华南地幔物质上涌、岩石圈发生拉张、伸展作用,使幔源岩浆上侵,并在这个过程中导致对地壳物质的同化,形成了壳幔混合流体。骑田岭花岗岩岩体冷却,高的K、U、Th含量导致高的热产率,岩体内及围岩中发育的大断裂和小裂隙及围岩中丰富的水,都十分有利于岩浆侵位后数百万年的热液对流循环,形成广泛的蚀变和成矿作用,而芙蓉锡矿田的成矿流体可能就是这种在岩浆作用阶段已发生过壳幔混合的岩浆流体所分异出来的可能混合了浅源流体(包括雨水源、地下水源等)的热液流体。

锡的分布和成矿作用通常与花岗岩浆作用具有十分密切的联系。以往研究表明锡矿化与高度分异的S型花岗岩或陆壳改造型花岗岩具有密切的成因联系,但近年来随着大量与A型花岗岩有关的锡矿床的发现,人们开始关注A型花岗岩与锡成矿关系的研究。相对于与S型花岗岩有关的锡矿床来说,与A型花岗岩有关的锡矿床成成矿机理的研究积累少,研究程度相对较低。 湘南地区位于南岭多金属成矿带中部,是我国华南地区重要的有色金属成矿带。近年来在该成矿带上新发现的芙蓉超大型锡多金属矿床为世界瞩目,该矿床的形成与骑田岭花岗岩具有密切的时空关系。近年来研究显示骑田岭花岗岩具有A型花岗岩的特征。本文以芙蓉超大型锡多金属矿床和相关的骑田岭岩体为研究对象,在前人研究的基础上,运用岩石学、矿物学、流体包裹体、微量元素和稳定同位素地球化学等理论和方法,对芙蓉锡矿成矿流体的地球化学特征及其演化机制进行了系统的研究,并在此基础上探讨了骑田岭花岗岩体与芙蓉锡矿间的成因联系和芙蓉锡矿的成因机制。论文取得的主要认识包括以下几个方面: 1. 运用矿物学、岩石化学、微量元素地球化学以及同位素地球化学方法,进一步证实了骑田岭花岗岩体具有A型花岗岩的特征,总体具有偏铝质-弱过铝质、高硅富碱高钾的地球化学特征,早晚两期花岗岩具有同源岩浆演化特征,属于A2型花岗岩。同位素地球化学数据显示花岗岩体具有EMII型富集地幔的特征,形成于华南大陆地壳拉张减薄的构造环境,成岩过程中有地幔物质加入。 2. 通过对矿石矿物组构和成分的岩矿鉴定、扫描电镜和电子探针分析,确定了芙蓉锡多金属矿床原生夕卡岩形成于较氧化的环境,成岩作用主要与早期侵入的角闪石黑云母花岗岩密切相关,锡主要以Sn(IV)进入夕卡岩的造岩矿物晶体内。退蚀变夕卡岩、云英岩和蚀变花岗岩矿化为锡成矿主阶段,三种矿化类型的成矿流体具有相似的地球化学性质,即富Cl、Ti和Sn的特征,而锡石硫化物型矿石形成于成矿晚阶段。 3. 运用流体包裹体地球化学理论和方法以及激光拉曼分析技术,揭示了芙蓉锡矿的成矿流体组成、形成的物理化学条件和演化特征。芙蓉锡多金属矿田成矿流体为CO2-CH4-CaCl2- NaCl-KCl不混溶体系,成矿过程中发生流体不混溶作用。芙蓉锡矿成矿流体盐度为0~50.63 wt%NaCl eq.,密度为0.31~1.12g/cm3,主成矿阶段热液流体的均一温度主要集中在300-450℃,流体压力为179-1800bar,成矿晚阶段锡石硫化物型矿石中均一温度主要集中在150~300℃,流体压力为400-600bar。成矿流体特别是主成矿阶段的流体成矿过程中普遍发生了沸腾现象。从主成矿阶段到成矿晚阶段、矿化期后,热液流体盐度呈降低的趋势,流体成分也从含CO2、CH4的CaCl2-NaCl-KCl-H2O水溶液体系转化为不含CO2的简单NaCl-KCl-H2O水溶液体系。 4. 通过分析主要矿化类型矿石中脉石矿物的稀土元素和稳定同位素特征,揭示了成矿流体来源。研究表明芙蓉矿床成矿期热液脉石矿物的稀土元素地球化学和稳定同位素地球化学显示了与本区花岗岩具明显的相似性,骑田岭黑云母花岗岩形成过程中分异出的岩浆期后热液应是芙蓉矿床成矿流体的主要来源,成矿过程中有少量经过深循环的大气降水加入。 5. 在总结前人研究成果的基础上,综合上述研究,探讨了骑田岭花岗岩体与芙蓉锡矿间的成因联系和芙蓉锡矿的成因机制。本文认为骑田岭岩体中黑云母花岗岩与Sn成矿具有密切的成因联系,芙蓉锡矿田的成矿流体主要来源于黑云母花岗岩岩浆结晶期后分异出的富Cl和Sn的热液流体。芙蓉锡矿成矿流体中锡主要呈Sn(II)与氯离子形成亚锡氯络合物进行迁移,低温的大气降水与高温的岩浆热液流体混合,导致流体体系温度、盐度、压力的降低和富CO2相流体的分离(CO2去气作用),流体的氧逸度升高,使得Sn(II)与氯离子形成亚锡氯络合物解体,Sn(II)被氧化成SnO2并发生沉淀作用。这种流体的混合作用是导致锡石沉淀的最有效的机制。

华南地区中生代地壳拉张、岩石圈伸展减薄与大规模成岩成矿作用的时代分布格局和地球动力学背景一直是国内外关注的重要科学问题。富碱侵入岩及其有关的成矿作用是伸展构造的直接表现之一。近年来的研究发现,南岭中段花山-西山-香花岭-骑田岭NE向的花岗岩带为一中晚侏罗世形成的具有高εNd(t)和低Nd模式年龄的富碱侵入岩带,并伴有大规模的钨锡多金属成矿作用,是研究大规模地壳拉张、岩石圈伸展减薄与该区大规模的钨锡多金属成矿作用的耦合关系及其深部动力学机制的理想对象。本文以香花岭地区花岗岩及其有关的钨锡多金属矿床为研究对象,运用岩石学、元素地球化学、同位素地球化学等方法手段,对香花岭矿区花岗岩的成因和形成构造环境、成矿物质与成矿流体的来源、成矿年代学及成岩成矿的地球动力学背景等方面进行了系统研究,并初步建立了该矿的成矿模式。论文主要获得以下认识: (1)通过对香花岭矿区癞子岭和尖峰岭岩体的主、微量元素及Sr、Nd同位素研究,查明上述两个岩体均具有高硅、富碱及成矿元素,富LILE及Zr、Ga等部分高场强元素(HFSE),贫Ca、Mg,P,Eu,较高的εNd及较低的Nd模式年龄等特征,具有相同的岩浆源区,属于后造山背景下形成的铝质A型花岗岩(A2型),具有壳、幔混合来源。 (2)利用白云母Ar-Ar同位素定年手段对香花岭锡多金属矿床、香花铺钨多金属矿床及尖峰岭云英岩型锡多金属矿床进行了成矿年代学研究,并首次对该矿的矿石矿物-锡石进行了U-Pb同位素定年。结果表明,香花岭地区钨锡多金属矿床的成矿时限为154-161 Ma,与矿区花岗岩黑云母K-Ar年龄相一致,并与区域主要钨锡矿床的形成时间一致。 (3)微量元素、稀土元素及同位素地球化学的研究表明,香花岭锡多金属矿床与矿区花岗岩具有密切的成因联系,矿区花岗岩不仅为该区成矿作用提供热动力和介质条件,而且为成矿作用提供了主要的物源。此外,矿区内的赋矿地层可能提供了部分成矿物质。早期成矿流体以岩浆水为主,晚期有大量大气降水的加入。 (4)对香花岭锡多金属矿床锡的超常富集机制作了初步探讨,认为该区锡的超常富集可能归功于如下几个因素:首先,富含F等挥发分的花岗岩浆高度分异演化出富含挥发分及成矿元素的超临界流体,成矿流体在流经赋矿地层时萃取了其中的部分成矿元素;其次,伴随着围压的不断降低,成矿流体发生了沸腾及连续的去气作用,导致了成矿流体的高度浓缩,当流体达到饱和或过饱和时,成矿元素沉淀成矿;最后,早期形成的矿床进一步受到后期的叠加改造,从而形成该区高品位的锡矿石。 (5)根据上述研究,香花岭锡多金属矿床与矿区花岗岩具有密切的时间、空间及成因联系,二者应为同一构造背景下形成的产物。结合区域上的研究,香花岭矿区的成岩成矿作用与南岭中段同时代的其他钨锡多金属矿床具有相同的地球动力学背景。这种构造背景可能与该区中-晚侏罗世构造体制转换,地壳拉张-岩石圈伸展减薄背景下,软流圈地幔上涌及发生壳、幔相互作用密切相关。 (6)初步建立了香花岭锡多金属矿床的成因模式:在该区中晚侏罗世大规模地壳拉张、岩石圈伸展减薄背景下,地幔物质上涌到中下地壳,发生壳幔相互作用,形成壳幔混合的花岗岩浆。花岗岩浆经充分分异演化,形成富含挥发分和成矿元素的一种具超临界性质的成矿流体。成矿流体在上升过程中随着围压的不断降低,发生了连续的去气作用,使得成矿流体高度浓缩并沿有利的构造部位成矿,并在后期受到改造、叠加等成矿作用,以至于形成了该区最富的锡矿床。

以往研究表明锡成矿与S型花岗岩具有密切的成因联系。近年来随着大量与A型花岗岩有关的锡矿床的发现,有关锡成矿与A型花岗岩关系的研究成为地学界关注的热点。 芙蓉超大型锡多金属矿床位于我国著名的南岭钨锡多金属成矿带上,锡矿体位于骑田岭A型花岗岩体的内部或者岩体与围岩的内外接触带。成岩成矿年代学研究表明,成岩与成矿为前后相继的地质事件,具有密切的时空关系。本论文以与芙蓉超大型锡多金属矿床有密切时空关系的骑田岭A型花岗岩为研究对象,在详细野外地质调查的基础上,运用岩石学、矿物学、矿物化学、同位素地球化学、流体地球化学等学科的理论和方法,对骑田岭花岗岩的岩石学特征、岩石成因、成岩物理化学条件、岩浆分异的流体特征、挥发性组分特征以及成岩与成矿的关系等方面进行详细的分析,探讨骑田岭花岗岩成岩过程中流体聚集的机制及其对锡成矿的制约,初步揭示A型花岗岩与锡成矿之间的本质联系。本论文主要取得以下成果和认识: (1)通过对与锡矿有关的骑田岭花岗岩体的主量、微量、稀土元素、同位素和花岗岩中黑云母的微量、稀土元素分析研究发现:骑田岭角闪石黑云母花岗岩和黑云母花岗岩为高度分异演化的花岗岩,具有高硅、富铝、富碱、高钾的特征。随着岩体分异演化程度的增加,花岗岩总体向富硅、富碱的方向演化。岩体轻重稀土分异明显,表现为右倾型模式,Eu负异常明显,表现为中等-强烈的负Eu异常。岩体明显富集Rb、Th等大离子亲石元素及Zr、Hf等高场强元素,而亏损Ba、Nb、Sr、P、Ti。骑田岭花岗岩两个阶段岩石有着相似的Sr、Nd同位素特征,揭示其具有相同的物质来源,是同源岩浆演化的产物,为具壳幔混合特征的A2型花岗岩。 (2)对骑田岭花岗岩体矿物学和矿物化学特征、全岩Sn含量分析研究发现:角闪石黑云母花岗岩的结晶温度为774~796℃,氧逸度(logfO2)为-15.30~-15.0。黑云母花岗岩的结晶温度为714~784℃,氧逸度(logfO2)为-17.5~-20.0。随着岩浆的演化,从角闪石黑云母花岗岩到黑云母花岗岩随着结晶温度的降低,氧逸度也随之减小。随着岩浆的演化,岩体中Cl含量不断的减少,而F含量有所增加,Cl趋向分配进入流体相。随着岩浆分异演化程度的增加,岩体成岩温度降低,氧逸度减小,岩体中Sn含量不断的减少,Sn趋向分配进入富Cl流体,表明岩浆演化过程中分异出富Cl、富Sn的流体。 (3)骑田岭花岗岩石英斑晶中的包裹体研究表明:骑田岭角闪石黑云母花岗岩和黑云母花岗岩在岩浆演化过程中经历了两个阶段,即岩浆阶段和岩浆-热液阶段,分别以出现熔融包裹体、流体-熔融包裹体为特征,其中流体-熔融包裹体的出现是岩浆分异流体的直接证据。结合矿物的结构、构造特征,研究发现骑田岭花岗岩浆演化过程分异出流体。骑田岭花岗岩原生流体包裹体地球化学研究表明,岩浆分异出的流体为H2O-CO2-NaCl-KCl-CaCl2不混溶体系,具有盐度高(32.98~52.04Wt%NaCleq.),密度低(0.27~0.95g/cm3),均一温度较高(190~ 494℃)的特征,压力为600~800bar,成岩过程中发生了沸腾现象。 (4)对芙蓉超大型锡矿床和骑田岭花岗岩研究表明,锡矿与花岗岩有着密切的时间、空间和成因联系。矿体产在花岗岩体内部或者岩体与围岩的接触带,成岩与成矿时限一致,随着岩浆分异演化程度的增加,岩体成岩温度降低,氧逸度降低,岩体中的挥发性组分Cl含量减小,而F含量增加,Cl趋向分配进入流体相,这种流体萃取熔体中的成矿元素Sn,并以氯络合物形式迁移。可以认为,随着岩浆的演化,骑田岭花岗岩岩浆结晶期后分异出的热液流体具有富Cl和Sn的特征。芙蓉超大型锡多金属矿床的成矿流体应主要来源于黑云母花岗岩岩浆结晶期后分异出的岩浆热液。

O gênero Phytophthora apresenta várias espécies patogênicas a hortaliças, entre elas P. nicotianae destaca-se como agente causal de podridões de frutos em tomate, berinjela e jiló. É importante gerar conhecimento sobre o quão agressivo determinado organismo é, a qual grupo de compatibilidade ele pertence e seu nível de sensibilidade aos fungicidas empregados no seu controle, no sentido obter o máximo de informações sobre o patógeno, a fim de buscar formas eficientes no manejo das doenças. Este trabalho teve o objetivo de caracterizar isolados de P. nicotianae, obtidos de frutos de tomate berinjela e jiló. Para isto foram realizados três experimentos distintos no Laboratório de Fitopatologia da Embrapa Hortaliças, com os marcadores biológicos grupos de compatibilidade, agressividade e resistência ao metalaxyl. Dos 39 isolados, caracterizados quanto ao grupo de compatibilidade, 23 apresentaram o grupo A1 e 16 o grupo A2, sendo que em apenas de um campo de cultivo (localizado em Cristalina - GO) ocorreram isolados de ambos os grupos. Quanto à agressividade foi verificado neste trabalho que P. nicotianae não demonstrou especificidade por nenhum dos seus hospedeiros de origem. Foram testados 32 isolados de P. nicotianae para verificar a resistência ao metalaxyl e, destes, apenas um apresentou resistência, oito foram intermediários e os outros 23 foram sensíveis à ação do fungicida. Tais resultados demonstraram uma alta eficiência do metalaxyl no controle de P. nicotianae fato que pode ser explicado pelo não uso deste fungicida em doenças causadas por este patógeno.

A matéria orgânica é um importante elemento para a separação e classificação de horizontes dos solos nos diversos sistemas taxonômicos, devido à relação com a morfologia, atributos químicos, físicos e mineralógicos. Este trabalho teve como objetivo avaliar os teores de carbono orgânico e as rações húmicas dos horizontes superficiais e subsuperficiais dos solos da X Reunião Brasileira de Classificação e Correlação. Foram selecionados perfis e nestes, horizontes superficiais e subsuperficias que identificam as classes de solo. Em amostras de terra, foram determinados os teores do carbono orgânico do solo e das frações húmicas. A fração humina predominou, seguido da fração ácidos húmicos nos pedoambientes Salina e Capão (sedimentos arenosos e calcário), enquanto, para os demais pedoambientes a fração humina é seguida da fração ácidos fúlvicos. Exceção com o predomínio das frações alcalinosolúveis no horizonte A2 do perfil MS03 (Capão) e horizontes A2 e Bw1 do perfil MS17 (Planalto). Com base na distribuição das frações húmicas não foi detectado acúmulo das frações alcalinosolúveis para os horizontes subsuperficiais caracterizados como Bh dos perfis MS01 e MS12 (Salina).

This thesis has been focused on the proteomic characterization of human saliva from donors of different ages, starting from birth up to adult age, and pediatric brain tumor tissues. The first study has been performed in order to compare the acid-insoluble fraction of saliva from preterm with at-term newborns and adults and establish if differences exist. In the second study medulloblastoma and pilocytic astrocytoma pediatric brain tumor extracts have been compared. In both studies 2- DE analysis was coupled with high resolution tandem mass spectrometry (MS/MS). The proteomic characterization of the acid-insoluble fractions of saliva from preterm newborns allowed to integrate data previously obtained on the acid-soluble fraction by HPLC-electrospray ionization (ESI)-mass spectrometry (MS), and to evidence several differences between preterm newborns, at-term newborns and adults. Spots differentially expressed between the three groups, according to image analysis of the gels, were submitted to in-gel tryptic digestion and the peptide mixture analyzed by high performance HPLC-ESI-MS/MS for their characterization. By this strategy, we identified three over-expressed proteins in atterm newborns with respect to preterm newborns and adults (BPI fold-containing family A member 1, two proteoforms of annexin A1, and keratin type 1 cytoskeletal 13), and several over-expressed proteins in adults (fatty acid-binding protein, S100A6, S100A7, two proteoforms of S100A9, several proteoforms of prolactin-inducible protein, Ig kappa chain, two proteoforms of cystatin SN, one proteoform of cystatin S and several proteoforms of α-amylase 1). Moreover, for the first time, it was possible to assign by MS/MS four spots of human saliva 2-DE, already detected by other authors, to different proteoforms of S100A9. The strategy applied used a sequential staining protocol to the 2-DE gels, first with Pro-Q Diamond, that allows specific detection of phosphoproteins, and successively with total protein SYPRO Ruby stain. In the second study, proteomic analysis of two pediatric brain tumor tissues pointed out differences between medulloblastoma, the prevalent malignant tumor in childhood, and pilocytic astrocytoma, the most common, that only rarely shows a malignant progression. Due to the limited availability of bioptic tissue, the study was performed on pooled tumor tissues, and was focused on acid-insoluble fraction to integrate the characterization performed by a group of colleagues in Rome on the acid-soluble fraction by high performance HPLC-ESI-MS/MS. The results indicated that the two tumors exhibit different proteomic profiles and evidenced interesting differential expression of several proteins. Among them, peroxiredoxin- 1, peptidyl-prolyl cis–trans isomerase A, heterogeneous nuclear ribonucleoproteins A2/B1, mitochondrial isoform of malate dehydrogenase, nucleoside diphosphate kinase A, glutathione S-transferase P and fructose bisphosphate aldolase A resulted significantly over-expressed in medulloblastoma while glial fibrillary acidic protein, serotransferrin, α crystallin B chain, ferritin light chain, annexin A5, fatty acid-binding protein (brain), sorcin and apolipoprotein A-I resulted significantly over-expressed in pilocytic astrocytoma. In conclusion, the work done allowed to evidence the usefulness of using an integrated bottom-up/top-down approach, based on 2-DE-MS analysis and high performance MS in order to obtain a complete characterization of the proteome under investigation, revealing and identifying, not only peptides and small proteins, but also proteins with higher MW, that often it is not possible to identify by using exclusively a top-down ESI-MS approach.

Aqueous solutions of amphiphilic polymers usually comprise of inter- and intramolecular associations of hydrophobic groups often leading to a formation of a rheologically significant reversible network at low concentrations that can be identified using techniques such as static light scattering and rheometry. However, in most studies published till date comparing water soluble polymers with their respective amphiphilic derivatives, it has been very difficult to distinguish between the effects of molecular mass versus hydrophobic associations on hydrodynamic (intrinsic viscosity [g]) and thermodynamic parameters (second virial coefficient A2), owing to the differences between their degrees of polymerization. This study focuses on the dilute and semi-dilute solutions of hydroxyethyl cellulose (HEC) and its amphiphilic derivatives (hmHEC) of the same molecular mass, along with other samples having a different molecular mass using capillary viscometry, rheometry and static light scattering. The weight average molecular masses (MW) and their distributions for the nonassociative HEC were determined using size exclusion chromatography. Various empirical approaches developed by past authors to determine [g] from dilute solution viscometry data have been discussed. hmHEC with a sufficiently high degree of hydrophobic modification was found to be forming a rheologically significant network in dilute solutions at very low concentrations as opposed to the hmHEC with a much lower degree of hydrophobic modification which also enveloped the hydrophobic groups inside the supramolecular cluster as shown by their [g] and A2. The ratio A2MW/[g], which takes into account hydrodynamic as well as thermodynamic parameters, was observed to be less for associative polymers compared to that of the non-associative polymers.

We studied the pattern of BCR involvement in 52 patients with chronic myeloid leukemia by Southern blotting. Of 33 Philadelphia (Ph)-positive patients, 30 had evidence of M-BCR rearrangement, two cases were difficult to interpret, and one clearly lacked evidence of M-BCR rearrangement. Of 19 Ph-negative patients, nine showed M-BCR rearrangement, nine showed no rearrangement, and one result was uncertain. We selected for more detailed study eight patients (three Ph-positive and five Ph-negative). Two of the Ph-positive patients, whose Southern blots were difficult to interpret, had rearranged bands when the BCR gene was studied by pulsed field gel electrophoresis (PFGE). Results of PFGE studies and in situ hybridization to metaphase chromosomes in the third Ph-positive patient, whose DNA clearly lacked M-BCR rearrangement on Southern analysis, were consistent with a breakpoint on chromosome 22 located 3' of all known exons of the BCR gene. However, mRNA studied with the polymerase chain reaction showed evidence of a classical b2-a2 linkage. The findings in this patient may be explained by an unusual genomic breakpoint downstream of the BCR gene associated with long range splicing that excluded all of the 3' BCR exons. Of the five patients with Ph-negative M-BCR non-rearranged CML studied by PFGE for BCR gene rearrangement, none had evidence of rearranged bands. We conclude that PFGE is a valuable adjunct to standard molecular techniques for the study of atypical cases of CML. Occasional patients with Ph-positive CML have breakpoints outside M-BCR. The BCR gene is probably not involved in patients with Ph-negative, M-BCR non-rearranged CML.

Nicotinic acid is one of the most effective agents for both lowering triglycerides and raising HDL. However, the side effect of cutaneous flushing severely limits patient compliance. As nicotinic acid stimulates the GPCR GPR109A and Gi/Go proteins, here we dissected the roles of G proteins and the adaptor proteins, beta-arrestins, in nicotinic acid-induced signaling and physiological responses. In a human cell line-based signaling assay, nicotinic acid stimulation led to pertussis toxin-sensitive lowering of cAMP, recruitment of beta-arrestins to the cell membrane, an activating conformational change in beta-arrestin, and beta-arrestin-dependent signaling to ERK MAPK. In addition, we found that nicotinic acid promoted the binding of beta-arrestin1 to activated cytosolic phospholipase A2 as well as beta-arrestin1-dependent activation of cytosolic phospholipase A2 and release of arachidonate, the precursor of prostaglandin D2 and the vasodilator responsible for the flushing response. Moreover, beta-arrestin1-null mice displayed reduced cutaneous flushing in response to nicotinic acid, although the improvement in serum free fatty acid levels was similar to that observed in wild-type mice. These data suggest that the adverse side effect of cutaneous flushing is mediated by beta-arrestin1, but lowering of serum free fatty acid levels is not. Furthermore, G protein-biased ligands that activate GPR109A in a beta-arrestin-independent fashion may represent an improved therapeutic option for the treatment of dyslipidemia.

The objective the study was to determine the levels of glucose and triglycerides in seminal plasma of 10 guinea pigs, which were fed for a period of 2 months with a diet containing 10% more ED. The level of glucose found in seminal plasma was 11.59 ± 0.5 mg/dL and triglyceride value was 55.95 ± 3.2 mg/dL, while the motility was 97% on average. We conclude that in guinea pigs the levels both glucose and triglycerides were increased by major level of ED in feed, but the spermatic motility was not.
