In 1975 two Cambridge scientists published a short article in Nature which announced the discovery of monoclonal antibodies. The article concluded ‘Such cultures could be valuable for medical and industrial use’. The interest which developed by the end of the decade in the industrial and financial possibilities of the new prospects opening up in biotechnology was to throw the apparent ‘failure’ to follow‐up the potentialities of this discovery into a public prominence rarely achieved by scientific discoveries. By the time Mrs Thatcher came to power it had become a scandal, another example of Britain's apparent inability to exploit effectively the brilliance of its scientific base. It was to explore both the process of scientific discovery and the conditions in Cambridge which nurtured it, and the issues which this particular discovery raised in the area of technology transfer (and the changes of policy that ensued), that the Wellcome Trust's History of Twentieth Century Medicine Group and the Institute of Contemporary British History organised this special witness seminar. It was held at the Wellcome Trust in London on 24 September 1993. The seminar was chaired by Sir Christopher Booth and introduced by Dr Robert Bud of the Science Museum. Those participating included the two authors of the Nature article, Dr César Milstein and Dr Georges Köhler, who received a Nobel Prize for their research, Dr Basil Bard (National Research Development Corporation [NRDC] 1950–74), Sir James Gowans (Secretary of the Medical Research Council [MRC] 1977–87), Sir John Gray (Secretary of the MRC 1968–77), John Newell (BBC World Service science correspondent 1969–79), Dr David Owen (MRC), and Dr David Secher (Laboratory of Molecular Biology [LMB], Cambridge). There were also contributions from Dr Ita Askonas (former head of immunology at the National Institute for Medical Research), Dr John Galloway (former member of MRC headquarters staff), Dr David Tyrrell (former Director, MRC Common Cold Unit), Professor Miles Weatherall (head of Therapeutic Research Division, Wellcome Research Laboratories 1967–75), Dr Guil Winchester (post‐doctoral fellow, Wellcome Institute for the History of Medicine), and Dr Peter Williams (former Director of the Wellcome Trust). Distribution systems are the first volunteers experiencing the benefits of smart grids. The smart grid concept impacts the internal legislation and standards in grid-connected and isolated distribution systems. Demand side management, the main feature of smart grids, acquires clear meaning in low voltage distribution systems. In these networks, various coordination procedures are required between domestic, commercial and industrial consumers, producers and the system operator. Obviously, the technical basis for bidirectional communication is the prerequisite of developing such a coordination procedure. The main coordination is required when the operator tries to dispatch the producers according to their own preferences without neglecting its inherent responsibility. Maintenance decisions are first determined by generating companies, and then the operator has to check and probably modify them for final approval. In this paper the generation scheduling from the viewpoint of a distribution system operator (DSO) is formulated. The traditional task of the DSO is securing network reliability and quality. The effectiveness of the proposed method is assessed by applying it to a 6-bus and 9-bus distribution system.

Knowing exactly where a mobile entity is and monitoring its trajectory in real-time has recently attracted a lot of interests from both academia and industrial communities, due to the large number of applications it enables, nevertheless, it is nowadays one of the most challenging problems from scientific and technological standpoints. In this work we propose a tracking system based on the fusion of position estimations provided by different sources, that are combined together to get a final estimation that aims at providing improved accuracy with respect to those generated by each system individually. In particular, exploiting the availability of a Wireless Sensor Network as an infrastructure, a mobile entity equipped with an inertial system first gets the position estimation using both a Kalman Filter and a fully distributed positioning algorithm (the Enhanced Steepest Descent, we recently proposed), then combines the results using the Simple Convex Combination algorithm. Simulation results clearly show good performance in terms of the final accuracy achieved. Finally, the proposed technique is validated against real data taken from an inertial sensor provided by THALES ITALIA.

O presente estágio foi desenvolvido na Britafiel. Um dos projectos em que a Empresa se encontra envolvida é o STOCO, pretendendo implementar à escala industrial um processo de coloração de pedra granítica natural para fins decorativos. Foi neste projecto que se enquadrou o estágio. O tema do estágio centra-se no processo de coloração de granito tendo como principal foco a implementação de um processo industrial de produção de granito colorido. O projecto STOCO nasce da necessidade de complementar a actividade da empresa com produtos de maior valor acrescentado para valorização da matéria-prima de base, o granito. STOCO, resultante de Stone Color, é o nome dado ao projecto e ao novo produto que é granito colorido, sob a forma de brita. Pretende-se obter um produto amigo do ambiente e com boas características: manter a textura natural da pedra granítica e assegurar uma boa resistência a factores agressivos. Estudos prévios de qualidade e de toxicidade mostraram que o produto STOCO desenvolvido até então apresenta um bom comportamento face a agressões climatéricas e que não compromete a vida das espécies usadas nos testes (peixes). Em relação aos lixiviados e resíduos da pedra colorida STOCO, estes não apresentaram qualquer problema ambiental, sendo considerado um produto amigo do ambiente. À data de início do presente trabalho estava em funcionamento um equipamento protótipo de produção de granito colorido (100 kg/partida), sendo a instalação e o arranque da unidade industrial (3 ton/h) concretizados no início de 2014, já no decorrer deste trabalho. Os objectivos cumpridos no âmbito deste trabalho foram então a implementação de uma linha industrial de produção de brita colorida, avaliação técnica do processo e do custo industrial de produção associado às matérias-primas. Neste relatório é descrito o processo inicial adoptado e apresentam-se as alterações efectuadas para melhoria do processo produtivo. Resolveram-se problemas como: definição e instalação de equipamentos complementares para a entrada e a saída da brita no equipamento industrial; pó excessivo na brita; cheiro intenso a gás e elevado ruído; adequação do sistema de pintura; e secagem incompleta da brita. Alguns destes problemas não foram totalmente resolvidos, mas sim minimizados. O equipamento industrial necessita ainda de alterações em diversas áreas, que foram identificadas e para as quais são feitas sugestões de melhoria. Conseguiu-se ainda fazer alguns testes para uma possível substituição de alguns constituintes da tinta. Os componentes que entram na composição base da tinta aquosa, são de modo simplificado: ligante, pigmento, solvente e aditivos. Os constituintes que mais encarecem a tinta, e consequentemente o processo em causa, são o ligante e o pigmento. Os estudos efectuados precisam de ser aprofundados, na tentativa de melhorar o processo minimizando os custos de produção. Formalizaram-se os procedimentos escritos de produção STOCO tanto para o protótipo como para o processo industrial e elaborou-se uma ficha técnica de produto para a brita colorida STOCO.

A presente dissertação visa aferir a dimensão e a relevância das alterações operadas por Nikita Khrushchev, enquanto líder da União Soviética, no Complexo Militar e Industrial deste país, no período da Guerra Fria. Neste contexto, as mesmas serão analisadas e proceder-se-á, paralelamente, ao estudo do impacto das mesmas a nível interno, bem como a nível externo, na interacção da União Soviética com os restantes actores da comunidade internacional, nomeadamente os Estados Unidos. Mormente, como forma de contextualizar as referidas alterações, proceder-se-á também ao apuramento dos motivos que estiveram subjacentes à transmutação de uma componente relevante nas Relações Internacionais, o Complexo Militar e Industrial Soviético.

Wireless mesh networks present an attractive communication solution for various research and industrial projects. However, in many cases, the appropriate preliminary calculations which allow predicting the network behavior have to be made before the actual deployment. For such purposes, network simulation environments emulating the real network operation are often used. Within this paper, a behavior comparison of real wireless mesh network (based on 802.11s amendment) and the simulated one has been performed. The main objective of this work is to measure performance parameters of a real 802.11s wireless mesh network (average UDP throughput and average one-way delay) and compare the derived results with characteristics of a simulated wireless mesh network created with the NS-3 network simulation tool. Then, the results from both networks are compared and the corresponding conclusion is made. The corresponding results were derived from simulation model and real-worldtest-bed, showing that the behavior of both networks is similar. It confirms that the NS-3 simulation model is accurate and can be used in further research studies.

During the past few decades, numerous plasmid vectors have been developed for cloning, gene expression analysis, and genetic engineering. Cloning procedures typically rely on PCR amplification, DNA fragment restriction digestion, recovery, and ligation, but increasingly, procedures are being developed to assemble large synthetic DNAs. In this study, we developed a new gene delivery system using the integrase activity of an integrative and conjugative element (ICE). The advantage of the integrase-based delivery is that it can stably introduce a large DNA fragment (at least 75 kb) into one or more specific sites (the gene for glycine-accepting tRNA) on a target chromosome. Integrase recombination activity in Escherichia coli is kept low by using a synthetic hybrid promoter, which, however, is unleashed in the final target host, forcing the integration of the construct. Upon integration, the system is again silenced. Two variants with different genetic features were produced, one in the form of a cloning vector in E. coli and the other as a mini-transposable element by which large DNA constructs assembled in E. coli can be tagged with the integrase gene. We confirmed that the system could successfully introduce cosmid and bacterial artificial chromosome (BAC) DNAs from E. coli into the chromosome of Pseudomonas putida in a site-specific manner. The integrase delivery system works in concert with existing vector systems and could thus be a powerful tool for synthetic constructions of new metabolic pathways in a variety of host bacteria.

The water quality and fish populations of the Welland River were observed to decline with distance downstream. This coincided with increased agricultural , domestic and industrial waste loadings. The river upstream of the City of Welland received considerable loadings from agricultural sources. Centrarchids, sciaenids, ictalurids, cyprinids and esocids characterized this upper section of the river. Most of these species were tolerant of low dissolved oxygen concentrations and the high turbidity which prevailed there . The river near Port Robinson receives many industrial and domestic wastes as evidenced by the water quality data. The fish in this section were less abundant and the observed population was comprised almost solely of cyprinids. Further downstream, near Montrose, the Welland River received shock loads of chemical wastes that exceeded a specific conductance of ISiOOO ;umhos/cm. Few fish were captured at this site and those that were captured were considered to be transients. A review of the literature revealed that none of the common indices of water quality in use today could adequately predict the observed distributions. In addition to the above, the long-term trend (l3 yrs) of water quality of the lower Welland River revealed a gradual improvement. The major factor thought to be responsible for this improvement was the operation of the Welland Sewage Treatment Plant. The construction of the New Welland Ship Canal coincided with large fluctuations of the total solids and other parameters downstream. These conditions prevailed for a maximum of three years (1972- 1975)' Furthermore, spawning times and temperatures, geographic distributions, length-weight regressions and many other descriptive aspects of the ecology of some 26 species/ taxa of fish were obtained. Several of these species are rare or new to southern Ontario.

In light of the heavy reliance of the people of the Niagara Peninsula on the T\\'elve Mile Creek (TMC) watershed for recreational activities and for municipal and industrial uses ( e.g., drinking water, shipping and discharge of effluents), it was deemed prudent to assess the envirol1tnental health of the system by analysing the sediments total and exchangeable metal, and TPH contents. The MOEE has set guidelines with limits for the protection and management of aquatic sediments, and the sediments from the headwaters of the TMC have total metal and TPH (subset of O&G) contents well below the lower provincial limits. Areas of environmental concern where total metal contents in sediments, either individually or collectively, exceed the guideline, are the south side of Lake Gibson, the Old WeIland Canal, a segment of TMC just south of the QEW and Martindale Pond. The total metal content of sediments does not in all instances identify areas of biological concern. Instead, it has been found that the exchangeable metal fraction of sediments is a better indicator of metal availability and thus potential accumulation in organisms. In some instances, the exchangeable metal fraction agrees with the total metal fraction defining areas of environmental concern, but it does vary from site to site reflecting the natural variability of the ambient environment. Overall, the exchangeable metal fraction of sediments appears to be a better indicator of anthropogenic pollution and ecosystem impact. A histochemical study of Anodon.ta sp., Elliptio sp. and zebra mussels (Dreissena polyn'101pha) was done in conjunction with passive biomonitoring of zebra and quagga mussels (Dreissena bugensis) from the Twelve Mile Creek watershed and Lake 51. Clair (Jeanette's Creek, Chatham, Ontario). The highest concentrations of divalent metals such as Cu, Ni, Cd, and Zn, and trivalent Al appear to accumulate in gill and kidney tissues. Metal contents of organ tissues in Anodonta sp. vary with size class. Organ metal content varies among size classes, thus requiring consideration of size in biomonitoring studies. Shucked zebra and quagga mussel tissues, exhibited similar size class to Al content trends. In addition they reflected the Al content trends of top (approximately 10 cm) most sediments in the Twelve Mile Creek watershed. Quagga mussels appear to have higher Al concentrations than zebra mussels, thus suggesting that quagga mussels may be better passive biomonitors of AI. Cd content in zebra mussel tissues, seemed to increase with size class trends. This was not demonstrated in the quagga mussel tissues. This suggests that Cd may be regulated by quagga mussels and not by zebra mussels, and that zebra mussels may be better passivebiomonitors of Cd than are quagga mussels. Zebra mussel, quagga mussel, Anodonta sp., and Elliptio sp. were used in a two part, active (translocated) biomonitoring study of the Twelve Mile Creek watershed. There was no statistical difference in death rates between zebra and quagga mussels after 65 days of biomonitoring. However there does appear to be a difference of death rates between sites. Unfortunately the data base did not permit us to differentiate between sites. Relative to Port Colborne Harbour (Port Colborne, Ontario), the Twelve Mile Creek watershed appears to be elevated in bioavailable AI. An area near the terminus of the Twelve Mile Creek appears to be an area of environmental concern since mussels seemed to have accumulated relatively large concentrations of Cd, Zn, and Pb. In addition to possible metal loading from a nearby outfalls, or possible upstream outfalls, road salt runoff from storm sewers may have contributed to metal accumulation through cation exchanges processes. Similar trends in cumulative quagga mussel metal concentrations during the two time periods (65 and 159 days), suggest that quagga mussels may reach equilibrium within 65 days of translocation. Differences in bioaccumulated metal concentrations of the two dreissenid species demonstrate that active biomonitoring studies must use a variety of organisms to adequately assess the environmental situation of specific waterways and/or bodies.

In light of the fact that literature on toxicity of heavy metals in non-acidified
freshwater systems is sparse, this project was initiated to conduct an environmental
assessment of Lake Gibson. Chemistry of soils from adjacent areas and vineyards in the
region provide a comparative background database. Water quality determinations were used
to identify and highlight areas of environmental concern within the Lake Gibson watershed.
A Shelby Corer was used to obtain 66 sediment cores from Lake Gibson. These were
sectioned according to lithology and color to yield 298 samples. A suite of 122 soil samples
was collected in the region and vicinity of Lake Gibson. All were tested for metals and
some for Total Petroleum Hydrocarbons (TPH). Evaluation of the results leads to the
following conclusions:
1. Metal concentrations ofAI, Cd, Cu, Cr, Pb, Ni, Fe and Zn in soils from the Niagara
Region are well below background limits set by the Ministry of the Environment
and Energy (MOEE) for provincial soils.
2. There is a spatial and depth difference for some of the metals within the various

Wine produced using an appassimento-type process represents a new and exciting innovation for the Ontario wine industry. This process involves drying grapes that have already been picked from the vine, which increases the sugar content due to dehydration and induces a variety of changes both within and on the surface of the grapes. Increasing sugar contents in musts subject wine yeast to conditions of high osmolarity during alcoholic fermentations. Under these conditions, yeast growth can be inhibited, target alcohol levels may not be attained and metabolic by-products of the hyperosmotic stress response, including glycerol and acetic acid, may impact wine composition. The further metabolism of acetic acid to acetylCoA by yeast facilitates the synthesis of ethyl acetate, a volatile compound that can also impact wine quality if present in sufficiently high concentrations. The first objective of this project was to understand the effect of yeast strain and sugar concentration on fermentation kinetics and metabolite formation, notably acetic acid and ethyl acetate, during fermentation in appassimento-type must. Our working hypotheses were that (1) the natural isolate Saccharomyces bayanus would produce less acetic acid and ethyl acetate compared to Saccharomyces cerevisiae strain EC-1118 fermenting the high and low sugar juices; (2) the wine produced using the appassimento process would contain higher levels of acetic acid and lower levels of ethyl acetate compared to table wine; (3) and the strains would be similar in the kinetic behavior of their fermentation performances in the high sugar must. This study determined that the S. bayanus strain produced significantly less acetic acid and ethyl acetate in the appassimento wine and table wine fermentations. Differences in acetic acid and ethyl acetate production were also observed within strains fermenting the two sugar conditions. Acetic acid production was higher in table wine fermented by S. bayanus as no acetic acid was produced in appassimento-style wine, and 1.4-times higher in appassimento wine fermented by EC-1118 over that found in table wine. Ethyl acetate production was 27.6-times higher in table wine fermented by S. bayanus, and 5.2-times higher by EC-1118, compared to that in appassimento wine. Sugar utilization and ethanol production were comparable between strains as no significant differences were determined. The second objective of this project was to bring a method in-house for measuring the concentration of pyridine nucleotides, NAD+, NADP+, NADH and NADPH, in yeast cytosolic extract. Development of this method is of applicative interest for our lab group as it will enable the redox balance of the NAD+/ NADH and NADP+/ NADPH systems to be assessed during high sugar fermentations to determine their respective roles as metabolic triggers for acetic acid production. Two methods were evaluated in this study including a UV-endpoint method using a set of enzymatic assay protocols outlined in Bergmeyer (1974) and a colorimetric enzyme cycling method developed by Sigma-Aldrich® using commercial kits. The former was determined to be limited by its low sensitivity following application to yeast extract and subsequent coenzyme analyses, while the latter was shown to exhibit greater sensitivity. The results obtained from the kits indicated high linearity, accuracy and precision of the analytical method for measuring NADH and NADPH, and that it was sensitive enough to measure the low coenzyme concentrations present in yeast extract samples. NADtotal and NADPtotal concentrations were determined to be above the lower limit of quantification and within the range of the respective calibration curves, making this method suitable for our research purposes.

Un objectif principal du génie logiciel est de pouvoir produire des logiciels complexes, de grande taille et fiables en un temps raisonnable. La technologie orientée objet (OO) a fourni de bons concepts et des techniques de modélisation et de programmation qui ont permis de développer des applications complexes tant dans le monde académique que dans le monde industriel. Cette expérience a cependant permis de découvrir les faiblesses du paradigme objet (par exemples, la dispersion de code et le problème de traçabilité). La programmation orientée aspect (OA) apporte une solution simple aux limitations de la programmation OO, telle que le problème des préoccupations transversales. Ces préoccupations transversales se traduisent par la dispersion du même code dans plusieurs modules du système ou l’emmêlement de plusieurs morceaux de code dans un même module. Cette nouvelle méthode de programmer permet d’implémenter chaque problématique indépendamment des autres, puis de les assembler selon des règles bien définies. La programmation OA promet donc une meilleure productivité, une meilleure réutilisation du code et une meilleure adaptation du code aux changements. Très vite, cette nouvelle façon de faire s’est vue s’étendre sur tout le processus de développement de logiciel en ayant pour but de préserver la modularité et la traçabilité, qui sont deux propriétés importantes des logiciels de bonne qualité. Cependant, la technologie OA présente de nombreux défis. Le raisonnement, la spécification, et la vérification des programmes OA présentent des difficultés d’autant plus que ces programmes évoluent dans le temps. Par conséquent, le raisonnement modulaire de ces programmes est requis sinon ils nécessiteraient d’être réexaminés au complet chaque fois qu’un composant est changé ou ajouté. Il est cependant bien connu dans la littérature que le raisonnement modulaire sur les programmes OA est difficile vu que les aspects appliqués changent souvent le comportement de leurs composantes de base [47]. Ces mêmes difficultés sont présentes au niveau des phases de spécification et de vérification du processus de développement des logiciels. Au meilleur de nos connaissances, la spécification modulaire et la vérification modulaire sont faiblement couvertes et constituent un champ de recherche très intéressant. De même, les interactions entre aspects est un sérieux problème dans la communauté des aspects. Pour faire face à ces problèmes, nous avons choisi d’utiliser la théorie des catégories et les techniques des spécifications algébriques. Pour apporter une solution aux problèmes ci-dessus cités, nous avons utilisé les travaux de Wiels [110] et d’autres contributions telles que celles décrites dans le livre [25]. Nous supposons que le système en développement est déjà décomposé en aspects et classes. La première contribution de notre thèse est l’extension des techniques des spécifications algébriques à la notion d’aspect. Deuxièmement, nous avons défini une logique, LA , qui est utilisée dans le corps des spécifications pour décrire le comportement de ces composantes. La troisième contribution consiste en la définition de l’opérateur de tissage qui correspond à la relation d’interconnexion entre les modules d’aspect et les modules de classe. La quatrième contribution concerne le développement d’un mécanisme de prévention qui permet de prévenir les interactions indésirables dans les systèmes orientés aspect.

Les défis conjoints du changement climatique d'origine anthropique et la diminution des réserves de combustibles fossiles sont le moteur de recherche intense pour des sources d'énergie alternatives. Une avenue attrayante est d'utiliser un processus biologique pour produire un biocarburant. Parmi les différentes options en matière de biocarburants, le bio-hydrogène gazeux est un futur vecteur énergétique attrayant en raison de son efficacité potentiellement plus élevé de conversion de puissance utilisable, il est faible en génération inexistante de polluants et de haute densité d'énergie. Cependant, les faibles rendements et taux de production ont été les principaux obstacles à l'application pratique des technologies de bio-hydrogène. Des recherches intensives sur bio-hydrogène sont en cours, et dans les dernières années, plusieurs nouvelles approches ont été proposées et étudiées pour dépasser ces inconvénients. À cette fin, l'objectif principal de cette thèse était d'améliorer le rendement en hydrogène moléculaire avec un accent particulier sur l'ingénierie métabolique et l’utilisation de bioprocédés à variables indépendantes. Une de nos hypothèses était que la production d’hydrogène pourrait être améliorée et rendue plus économiquement viable par ingénierie métabolique de souches d’Escherichia coli producteurs d’hydrogène en utilisant le glucose ainsi que diverses autres sources de carbone, y compris les pentoses. Les effets du pH, de la température et de sources de carbone ont été étudiés. La production maximale d'hydrogène a été obtenue à partir de glucose, à un pH initial de 6.5 et une température de 35°C. Les études de cinétiques de croissance ont montré que la μmax était 0.0495 h-1 avec un Ks de 0.0274 g L-1 lorsque le glucose est la seule source de carbone en milieu minimal M9. .Parmi les nombreux sucres et les dérivés de sucres testés, les rendements les plus élevés d'hydrogène sont avec du fructose, sorbitol et D-glucose; 1.27, 1.46 et 1.51 mol H2 mol-1 de substrat, respectivement. En outre, pour obtenir les interactions entre les variables importantes et pour atteindre une production maximale d'hydrogène, un design 3K factoriel complet Box-Behnken et la méthodologie de réponse de surface (RSM) ont été employées pour la conception expérimentale et l'analyse de la souche d'Escherichia coli DJT135. Le rendement en hydrogène molaire maximale de 1.69 mol H2 mol-1 de glucose a été obtenu dans les conditions optimales de 75 mM de glucose, à 35°C et un pH de 6.5. Ainsi, la RSM avec un design Box-Behken était un outil statistique utile pour atteindre des rendements plus élevés d'hydrogène molaires par des organismes modifiés génétiquement. Ensuite, l'expression hétérologue de l’hydrogénases soluble [Ni-Fe] de Ralstonia eutropha H16 (l'hydrogénase SH) a tenté de démontrer que la mise en place d'une voie capable de dériver l'hydrogène à partir de NADH pourrait surpasser le rendement stoechiométrique en hydrogène.. L’expression a été démontrée par des tests in vitro de l'activité enzymatique. Par ailleurs, l'expression de SH a restaurée la croissance en anaérobie de souches mutantes pour adhE, normalement inhibées en raison de l'incapacité de réoxyder le NADH. La mesure de la production d'hydrogène in vivo a montré que plusieurs souches modifiées métaboliquement sont capables d'utiliser l'hydrogénase SH pour dériver deux moles d’hydrogène par mole de glucose consommé, proche du maximum théorique. Une autre stratégie a montré que le glycérol brut pourrait être converti en hydrogène par photofermentation utilisant Rhodopseudomonas palustris par photofermentation. Les effets de la source d'azote et de différentes concentrations de glycérol brut sur ce processus ont été évalués. À 20 mM de glycérol, 4 mM glutamate, 6.1 mol hydrogène / mole de glycérol brut ont été obtenus dans des conditions optimales, un rendement de 87% de la théorie, et significativement plus élevés que ce qui a été réalisé auparavant. En prolongement de cette étude, l'optimisation des paramètres a également été utilisée. Dans des conditions optimales, une intensité lumineuse de 175 W/m2, 30 mM glycérol et 4.5 mM de glutamate, 6.69 mol hydrogène / mole de glycérol brut ont été obtenus, soit un rendement de 96% de la valeur théorique. La détermination de l'activité de la nitrogénase et ses niveaux d'expression ont montré qu'il y avait relativement peu de variation de la quantité de nitrogénase avec le changement des variables alors que l'activité de la nitrogénase variait considérablement, avec une activité maximale (228 nmol de C2H4/ml/min) au point central optimal. Dans la dernière section, la production d'hydrogène à partir du glucose via la photofermentation en une seule étape a été examinée avec la bactérie photosynthétique Rhodobacter capsulatus JP91 (hup-). La méthodologie de surface de réponse avec Box-Behnken a été utilisée pour optimiser les variables expérimentales de façon indépendante, soit la concentration de glucose, la concentration du glutamate et l'intensité lumineuse, ainsi que d'examiner leurs effets interactifs pour la maximisation du rendement en hydrogène moléculaire. Dans des conditions optimales, avec une intensité lumineuse de 175 W/m2, 35 mM de glucose, et 4.5 mM de glutamate,, un rendement maximal d'hydrogène de 5.5 (± 0.15) mol hydrogène /mol glucose, et un maximum d'activité de la nitrogénase de 246 (± 3.5) nmol C2H4/ml/min ont été obtenus. L'analyse densitométrique de l'expression de la protéine-Fe nitrogenase dans les différentes conditions a montré une variation significative de l'expression protéique avec un maximum au point central optimisé. Même dans des conditions optimales pour la production d'hydrogène, une fraction significative de la protéine Fe a été trouvée dans l'état ADP-ribosylée, suggérant que d'autres améliorations des rendements pourraient être possibles. À cette fin, un mutant amtB dérivé de Rhodobacter capsulatus JP91 (hup-) a été créé en utilisant le vecteur de suicide pSUP202. Les résultats expérimentaux préliminaires montrent que la souche nouvellement conçue métaboliquement, R. capsulatus DG9, produit 8.2 (± 0.06) mol hydrogène / mole de glucose dans des conditions optimales de cultures discontinues (intensité lumineuse, 175 W/m2, 35 mM de glucose et 4.5 mM glutamate). Le statut d'ADP-ribosylation de la nitrogénase-protéine Fe a été obtenu par Western Blot pour la souche R. capsulatus DG9. En bref, la production d'hydrogène est limitée par une barrière métabolique. La principale barrière métabolique est due au manque d'outils moléculaires possibles pour atteindre ou dépasser le rendement stochiométrique en bio-hydrogène depuis les dernières décennies en utilisant les microbes. À cette fin, une nouvelle approche d’ingénierie métabolique semble très prometteuse pour surmonter cette contrainte vers l'industrialisation et s'assurer de la faisabilité de la technologie de la production d'hydrogène. Dans la présente étude, il a été démontré que l’ingénierie métabolique de bactéries anaérobiques facultatives (Escherichia coli) et de bactéries anaérobiques photosynthétiques (Rhodobacter capsulatus et Rhodopseudomonas palustris) peuvent produire de l'hydrogène en tant que produit majeur à travers le mode de fermentation par redirection métabolique vers la production d'énergie potentielle. D'autre part, la méthodologie de surface de réponse utilisée dans cette étude représente un outil potentiel pour optimiser la production d'hydrogène en générant des informations appropriées concernant la corrélation entre les variables et des producteurs de bio-de hydrogène modifiés par ingénierie métabolique. Ainsi, un outil d'optimisation des paramètres représente une nouvelle avenue pour faire un pont entre le laboratoire et la production d'hydrogène à l'échelle industrielle en fournissant un modèle mathématique potentiel pour intensifier la production de bio-hydrogène. Par conséquent, il a été clairement mis en évidence dans ce projet que l'effort combiné de l'ingénierie métabolique et la méthodologie de surface de réponse peut rendre la technologie de production de bio-hydrogène potentiellement possible vers sa commercialisation dans un avenir rapproché.
