Genetic Variants of Serum Uric Acid and Gout: An Analysis of > 170,000 Individuals

Background/Purpose: Gout is a common and excruciatingly painful inflammatory arthritis caused by hyperuricemia. In addition to various lifestyle risk factors, a substantial genetic predisposition to gout has long been recognized. The Global Urate Genetics Consortium (GUGC) has aimed to comprehensively investigate the genetics of serum uric acid and gout using data from _ 140,000 individuals of European-ancestry, 8,340 individuals of Indian ancestry, 5,820 African-Americans, and 15,286 Japanese. Methods: We performed discovery GWAS meta-analyses of serum urate levels (n_110,347 individuals) followed by replication analyses (n_32,813 different individuals). Our gout analysis involved 3,151 cases and 68,350 controls, including 1,036 incident gout cases that met the American College of Rheumatology Criteria. We also examined the association of gout with fractional excretion of uric acid (n_6,799). A weighted genetic urate score was constructed based on the number of risk alleles across urate-associated loci, and their association with the risk of gout was evaluated. Furthermore, we examined implicated transcript expression in cis (expression quantitative trait loci databases) for potential insights into the gene underlying the association signal. Finally, in order to further identify urate-associated genomic regions, we performed functional network analyses that incorporated prior knowledge on molecular interactions in which the gene products of implicated genes operate. Results: We identified and replicated 28 genome-wide significant loci in association with serum urate (P 5_10_8), including all previously-reported loci as well as 18 novel genetic loci. Unlike the majority of previouslyidentified loci, none of the novel loci appeared to be obvious candidates for urate transport. Rather, they were mapped to genes that encode for purine production, transcription, or growth factors with broad downstream responses. Besides SLC2A9 and ABCG2, no additional regions contained SNPs that differed significantly (P _ 5_10_8) between sexes. Urateincreasing alleles were associated with an increased risk of gout for all loci. The urate genetic risk score (ranging from 10 to 45) was significantly associated with an increased odds of prevalent gout (OR per unit increase, 1.11; 95% CI, 1.09-1.14) and incident gout (OR, 1.10; 95% CI, 1.08-1.13). Associations for many of the loci were of similar magnitude in individuals of non-European ancestry. Detailed characterization of the loci revealed associations with transcript expression and the fractional excretion of urate. Network analyses implicated the inhibins-activins signaling pathways and glucose metabolism in systemic urate control. Conclusion: The novel genetic candidates identified in this urate/gout consortium study, the largest to date, highlight the importance of metabolic control of urate production and urate excretion. The modulation by signaling processes that influence metabolic pathways such as glycolysis and the pentose phosphate pathway appear to be central mechanisms underpinned by the novel GWAS candidates. These findings may have implications for further research into urate-lowering drugs to treat and prevent gout.

Efeito da natureza do eletrólito e da força iônica na energia livre da reação de adsorção de níquel em solos

A adsorção é o principal processo responsável pelo acúmulo de metais pesados na superfície dos colóides do solo. O conhecimento detalhado desse fenômeno pode fornecer subsídios para o aprimoramento das práticas de remediação de solos contaminados. Avaliou-se a energia livre (deltaG0) de adsorção de Ni em amostras superficiais (0,0-0,2 m) e subsuperficiais (na maior expressão do horizonte B) de um Latossolo Vermelho acriférrico típico textura argilosa (LVwf) e de um Nitossolo Vermelho eutroférrico textura muito argilosa (NVef), utilizando-se soluções de NaCl e CaCl2 em três forças iônicas (1,0, 0,1 e 0,01 mol L-1). As amostras de solo receberam 2, 5, 10, 20, 30, 40, 50 e 70 mg dm-3 de Ni, na proporção solo:solução de 1:10. A adsorção de Ni pelos solos foi espontânea, visto que a deltaG0 apresentou valores negativos em todas as concentrações estudadas. Os valores de deltaG0 diminuíram com o aumento da dose de Ni adicionada. O NVef apresentou maior deltaG0 que o LVwf devido, principalmente, às suas características químicas e mineralógicas. Os horizontes superficiais apresentaram, em geral, maior deltaG0 em relação aos subsuperficiais, em razão do elevado teor de matéria orgânica encontrado em superfície. A deltaG0 foi maior para as menores forças iônicas do meio, tanto para CaCl2 quanto para NaCl.

Trends in food availability in Switzerland, 1961-2007.

Trends in food availability in Switzerland were assessed using the Food and Agricultural Organization food balance sheets for the period 1961-2007. A relatively stable trend in the daily caloric supply was found: 3545 kcal/day in 1961 and 3465 kcal/day in 2007. Calories associated with carbohydrates decreased (slope±s.e.: -1.1±0.2 kcal/day/year), namely regarding cereals (-2.9±0.6 kcal/day/year) and fruit (-1.5±0.1 kcal/day/year), while the availability of sugars increased (1.2±0.5 kcal/day/year). In 1961, protein, fat, carbohydrates and alcohol represented 10.6, 33.5, 50.0 and 5.9% of total caloric supply, respectively; in 2007, the values were 10.8, 40.3, 43.7 and 5.2%. In 1961, palm, groundnut and sunflowerseed oil represented 3.4, 30.7 and 5.3% of total vegetable oils, respectively; in 2007, the values were 10.4, 3.7 and 31.6%. We conclude that between 1961 and 2007 total caloric availability remained relatively stable in Switzerland; the health effects of the increased and differing fat availability should be evaluated.

Avaliação espaço-temporal das armazenagens gravimétrica e volumétrica da água num Latossolo com citros

A estabilidade temporal é descrita como a persistência temporal de um padrão espacial e é avaliada pela análise de correlação de medidas em datas sucessivas. Neste trabalho, foi avaliada a armazenagem de água no solo em duas transeções de 76 m cada, contendo 20 pontos em cada uma. em um Latossolo Vermelho-Amarelo argissólico cultivado com citros com 11 anos em Piracicaba/SP. A umidade do solo para determinação da armazenagem, na camada de 0,00-1,10 m, ao longo do tempo, foi obtida com uma sonda de nêutrons, calibrada para o solo em estudo, realizando leituras semanais às profundidades de 0,20, 0,40, 0,60, 0,80, 1,0 e 1,10 m. A armazenagem de água no solo foi determinada utilizando-se o método de Simpson. Os dados da armazenagem média de água no solo nos três anos de estudo mostraram que sua distribuição, ao longo do tempo, apresentou comportamento semelhante e que os coeficientes de correlação linear e de posição de Spearman foram altos entre o ano 1 e o ano 2, o ano 2 e o ano 3 e o ano 1 e o ano 3, indicando existência de estabilidade temporal e persistência temporal significativa do padrão espacial observado. Foi verificada dependência espacial moderada de 17,10 m pelo semivariograma exponencial. Pela técnica da diferença relativa, os pontos que apresentaram comportamento estável no tempo foram o ponto 29, para os anos 1 e 2, e os pontos 16, 29 e 39, para o ano 3, que podem ser usados para estimar com segurança a armazenagem média de água no solo. O ponto 29 foi o que se apresentou estável para os três anos de estudo, sendo, portanto, o mais recomendável tanto para a armazenagem gravimétrica como para a volumétrica.
