TTHA complexes with diamagnetic rare earth ions (La3+, Y3+ and LU(3+)) were studied by H-1 and C-13 NMR spectroscopy. A symmetric structural model was suggested for La(TTHA) complex and an asymmetric model for Y(TTHA) and Lu(TTHA) complexes. The complex formation was dependent on the pH value of the solution. The interactions of La(TTHA) with the additional metal ions (La3+, Y3+ and Ca2+) were relatively weak, but relatively strong for that of Lu(TTHA) with the additional Lu3+.

The dmrt (doublesex and mab-3 related transcription factor) gene family comprises several transcription factors that share a conserved DM domain. Dmrt1 is considered to be involved in sexual development, but the precise function of other family members is unclear. In this study, we isolated genomic DNA and cDNA sequences of dmrt4, a member of the dmrt gene family, from olive flounder, Paralichthys olivaceus, through genome walking and real-time reverse transcriptase (RT)-PCR. Sequence analysis indicated that its genomic DNA contains two exons and one intron. A transcriptional factor binding sites prediction program identified a sexual development-related protein, Sox9 (Sry-like HMG box containing 9) in its 5' promoter. Protein alignment and phylogenetic analysis suggested that flounder Dmrt4 is closely related to tilapia Dmo (DM domain gene in ovary). The expression of dmrt4 in adult flounder was sexually dimorphic, as shown by real-time RT-PCR analysis, with strong expression in the testis but very weak expression in the ovary. Its expression was also strong in the brain and gill, but there was only weak or no expression at all in some of the other tissues tested of both sexes. During embryogenesis, its expression was detected in most developmental stages, although the level of expression was distinctive of the various stages. Whole mount in situ hybridization revealed that the dmrt4 was expressed in the otic placodes, forebrain, telencephalon and olfactory placodes of embryos at different developmental stages. These results will improve our understanding of the possible role of flounder dmrt4 in the development of the gonads, nervous system and sense organs.

A highly repetitive satellite sequence was previously identified in the Pacific oyster Crassostrea gigas Thunberg. The sequence has 168 bp per unit, present in tandem repeats, and accounts for 1% to 4% of the genome. We studied the chromosomal location of this satellite sequence by fluorescence in situ hybridization (FISH), A probe was made by polymerase chain reaction and incorporation of digoxigenin-11-dUTP. Hybridization was detected with fluorescein-labeled antidigoxigenin antibodies. FISH signals were located at centromeric regions of 7 pairs of the Pacific oyster chromosomes. No interstitial site was found. Signals were strong and consistent on chromosomes 1, 2, 4, and 7, but weak or variable oil chromosomes 5, 8, and 10. No signal was observed on chromosomes 3, 6, and 9. Our results showed that this sequence is clearly a centromeric satellite, disputing its previous assignment to the telomeric and submetacentric regions of 2 chromosomes. No signal was detected in the American oyster (Crassostrea virginica Gmelin).

Peptidoglycan recognition protein (PGRP) is an essential molecule in innate immunity for both invertebrates and vertebrates, owing to its prominent ability in detecting and eliminating the invading bacteria. Several PGRPs have been identified from mollusk, but their functions and the underlined mechanism are still unclear. In the present study, the mRNA expression profiles, location, and possible functions of PGRP-S1 from Zhikong scallop Chlamys farreri (CfPG RP-St) were analyzed. The CfPGRP-S1 protein located in the mantle, gill, kidney and gonad of the scallops. Its mRNA expression in hemocytes was up-regulated extremely after PGN stimulation (P < 0.01), while moderately after the stimulations of LPS (P < 0.01) and beta-glucan (P < 0.05). The recombinant protein of CfPGRP-S1 (designated as rCfPGRP-S1) exhibited high affinity to PGN and moderate affinity to LPS, but it did not bind beta-glucan. Meanwhile, rCfPGRP-S1 also exhibited strong agglutination activity to Gram-positive bacteria Micrococcus luteus and Bacillus subtilis and weak activity to Gram-negative bacteria Escherichia coli. More importantly, rCfPGRP-S1 functioned as a bactericidal amidase to degrade PGN and strongly inhibit the growth of E. coli and Staphyloccocus aureus in the presence of Zn2+. These results indicated that CfPGRP-S1 could not only serve as a pattern recognition receptor recognizing bacterial PGN and LPS, but also function as a scavenger involved in eliminating response against the invaders. AMS(14)C dating and analysis of grain size, major elements and clay minerals were applied to Core MZ01 from the mud area on the inner shelf of the East China Sea. Based on the environmentally sensitive grain size, clay mineral and major element assemblages, the history of the East Asia winter monsoon since the mid-Holocene could be reconstructed. These three proxies, mean grain size (>9.71 mu m), chemical index of alteration (CIA) and ratio of smectite to kaolinite in particular, show similar fluctuation patterns. Furthermore, 10 extreme values corresponding to the contemporary cooling events could be recognized since the mid-Holocene; these extreme values are likely to have been caused by the strengthening of the East Asia winter monsoon. The cooling events correlated well with the results of the delta O-18 curves of the Dunde ice core and GISP2, which therefore revealed a regional response to global climate change. Four stages of the East Asia winter monsoon were identified, i.e. 8300-6300 a BP, strong and unstable; 6300-3800 a BP, strong but stable; 3800-1400 a BP, weak and unstable; after 1400 a BP, weak but stable.

Satellite and in situ observations in the equatorial Atlantic Ocean during 2002-03 show dominant spectral peaks at 40-60 days and secondary peaks at 10-40 days in sea level and thermocline within the intraseasonal period band (10-80 days). A detailed investigation of the dynamics of the intraseasonal variations is carried out using an ocean general circulation model, namely, the Hybrid Coordinate Ocean Model (HYCOM). Two parallel experiments are performed in the tropical Atlantic Ocean basin for the period 2000-03: one is forced by daily scatterometer winds from the Quick Scatterometer (QuikSCAT) satellite together with other forcing fields, and the other is forced by the low-passed 80-day version of the above fields. To help in understanding the role played by the wind-driven equatorial waves, a linear continuously stratified ocean model is also used. Within 3 degrees S-3 degrees N of the equatorial region, the strong 40-60-day sea surface height anomaly (SSHA) and thermocline variability result mainly from the first and second baroclinic modes equatorial Kelvin waves that are forced by intraseasonal zonal winds, with the second baroclinic mode playing a more important role. Sharp 40-50-day peaks of zonal and meridional winds appear in both the QuikSCAT and Pilot Research Moored Array in the Tropical Atlantic (PIRATA) data for the period 2002-03, and they are especially strong in 2002. Zonal wind anomaly in the central-western equatorial basin for the period 2000-06 is significantly correlated with SSHA across the equatorial basin, with simultaneous/ lag correlation ranging from-0.62 to 0.74 above 95% significance. Away from the equator (3 degrees-5 degrees N), however, sea level and thermocline variations in the 40-60-day band are caused largely by tropical instability waves (TIWs). On 10-40-day time scales and west of 10 degrees W, the spectral power of sea level and thermocline appears to be dominated by TIWs within 5 degrees S-5 degrees N of the equatorial region. The wind-driven circulation, however, also provides a significant contribution. Interestingly, east of 10 W, SSHA and thermocline variations at 10 40- day periods result almost entirely from wind-driven equatorial waves. During the boreal spring of 2002 when TIWs are weak, Kelvin waves dominate the SSHA across the equatorial basin (2 degrees S-2 degrees N). The observed quasi-biweekly Yanai waves are excited mainly by the quasi-biweekly meridional winds, and they contribute significantly to the SSHA and thermocline variations in 1 degrees-5 degrees N and 1 degrees-5 degrees S regions.

An assimilation data set based on the GFDL MOM3 model and the NODC XBT data set is used to examine the circulation in the western tropical Pacific and its seasonal variations. The assimilated and observed velocities and transports of the mean circulation agree well. Transports of the North Equatorial Current (NEC), Mindanao Current (MC), North Equatorial Countercurrent (NECC) west of 140degreesE and Kuroshio origin estimated with the assimilation data display the seasonal cycles, roughly strong in boreal spring and weak in autumn, with a little phase difference. The NECC transport also has a semi-annual fluctuation resulting from the phase lag between seasonal cycles of two tropical gyres' recirculations. Strong in summer during the southeast monsoon period, the seasonal cycle of the Indonesian throughflow (ITF) is somewhat different from those of its upstreams, the MC and New Guinea Coastal Current (NGCC), implying the monsoon's impact on it.

Based on the latest seismic and geological data, tectonic subsidence of three seismic lines in the deepwater area of Pearl River Mouth Basin (PRMB), the northern South China Sea (SCS), is calculated. The result shows that the rifting process of study area is different from the typical passive continental margin basin. Although the seafloor spreading of SCS initiated at 32 Ma, the tectonic subsidence rate does not decrease but increases instead, and then decreases at about 23 Ma, which indicates that the rifting continued after the onset of seafloor spreading until about 23 Ma. The formation thickness exhibits the same phenomenon, that is the syn-rift stage prolonged and the post-rift thermal subsidence delayed. The formation mechanisms are supposed to be three: (1) the lithospheric rigidity of the northern SCS is weak and its ductility is relatively strong, which delayed the strain relaxation resulting from the seafloor spreading; (2) the differential layered independent extension of the lithosphere may be one reason for the delay of post-rift stage; and (3) the southward transition of SCS spreading ridge during 24 to 21 Ma and the corresponding acceleration of seafloor spreading rate then triggered the initiation of large-scale thermal subsidence in the study area at about 23 Ma.

An ocean general circulation model (OGCM) is used to study the roles of equatorial waves and western boundary reflection in the seasonal circulation of the equatorial Indian Ocean. The western boundary reflection is defined as the total Kelvin waves leaving the western boundary, which include the reflection of the equatorial Rossby waves as well as the effects of alongshore winds, off-equatorial Rossby waves, and nonlinear processes near the western boundary. The evaluation of the reflection is based on a wave decomposition of the OGCM results and experiments with linear models. It is found that the alongshore winds along the east coast of Africa and the Rossby waves in the off-equatorial areas contribute significantly to the annual harmonics of the equatorial Kelvin waves at the western boundary. The semiannual harmonics of the Kelvin waves, on the other hand, originate primarily from a linear reflection of the equatorial Rossby waves. The dynamics of a dominant annual oscillation of sea level coexisting with the dominant semiannual oscillations of surface zonal currents in the central equatorial Indian Ocean are investigated. These sea level and zonal current patterns are found to be closely related to the linear reflections of the semiannual harmonics at the meridional boundaries. Because of the reflections, the second baroclinic mode resonates with the semiannual wind forcing; that is, the semiannual zonal currents carried by the reflected waves enhance the wind-forced currents at the central basin. Because of the different behavior of the zonal current and sea level during the reflections, the semiannual sea levels of the directly forced and reflected waves cancel each other significantly at the central basin. In the meantime, the annual harmonic of the sea level remains large, producing a dominant annual oscillation of sea level in the central equatorial Indian Ocean. The linear reflection causes the semiannual harmonics of the incoming and reflected sea levels to enhance each other at the meridional boundaries. In addition, the weak annual harmonics of sea level in the western basin, resulting from a combined effect of the western boundary reflection and the equatorial zonal wind forcing, facilitate the dominance by the semiannual harmonics near the western boundary despite the strong local wind forcing at the annual period. The Rossby waves are found to have a much larger contribution to the observed equatorial semiannual oscillations of surface zonal currents than the Kelvin waves. The westward progressive reversal of seasonal surface zonal currents along the equator in the observations is primarily due to the Rossby wave propagation.

温度跃层是反映海洋温度场的重要物理特性指标,对水下通讯、潜艇活动及渔业养殖、捕捞等有重要影响。本文利用中国科学院海洋研究所“中国海洋科学数据库”在中国近海及西北太平洋(110ºE-140ºE,10ºN-40ºN)的多年历史资料(1930-2002年,510143站次),基于一种改进的温跃层判定方法,分析了该海域温跃层特征量的时空分布状况。同时利用Princeton Ocean Model(POM),对中国近海,特别是东南沿海的水文结构进行了模拟,研究了海洋水文环境对逆温跃层的影响。最后根据历史海温观测资料,利用EOF分解统计技术,提出了一种适于我国近海及毗邻海域,基于现场有限层实测海温数据,快速重构海洋水温垂直结构的统计预报方法,以达到对现场温跃层的快速估计。 历史资料分析结果表明,受太阳辐射和风应力的影响,20°N以北研究海域,温跃层季节变化明显,夏季温跃层最浅、最强,冬季相反,温跃层厚度的相位明显滞后于其他变量,其在春季最薄、秋季最厚。12月份到翌年3月份,渤、黄及东海西岸,呈无跃层结构,西北太平洋部分海域从1月到3月份,也基本无跃层结构。在黄海西和东岸以及台湾海峡附近的浅滩海域,由于风力搅拌和潮混合作用,温跃层出现概率常年较低。夏季,海水层化现象在近海陆架海域得到了加强,陆架海域温跃层强度季节性变化幅度(0.31°C/m)明显大于深水区(约0.05°C/m),而前者温跃层深度和厚度的季节性变化幅度小于后者。20°N以南研究海域,温跃层季节变化不明显。逆温跃层主要出现在冬、春季节(10月-翌年5月)。受长江冲淡水和台湾暖流的影响,东南沿海区域逆温跃层持续时间最长,出现概率最大,而在山东半岛北及东沿岸、朝鲜半岛西及北岸,逆温跃层消长过程似乎和黄海暖流有关。多温跃层结构常年出现于北赤道流及对马暖流区。在黑潮入侵黄、东、南海的区域,多温跃层呈现明显不同的季节变化。在黄海中部,春季多温跃层发生概率高于夏季和秋季,在东海西部,多跃层主要出现在夏季,在南海北部,冬季和春季多温跃层发生概率大于夏季和秋季。这些变化可能主要受海表面温度变化和风力驱动的表层流的影响。 利用Princeton Ocean Model(POM),对中国东南沿海逆温跃层结构进行了模拟,模拟结果显示,长江冲淡水的季节性变化以及夏季转向与实际结果符合较好,基本再现了渤、黄、东海海域主要的环流、温盐场以及逆温跃层的分布特征和季节变化。通过数值实验发现,若无长江、黄河淡水输入,则在整个研究海域基本无逆温跃层出现,因此陆源淡水可能是河口附近逆温跃层出现的基本因素之一。长江以及暖流(黑潮和台湾暖流)流量的增加,均可在不同程度上使逆温跃层出现概率及强度、深度和厚度增加,且暖流的影响更加明显。长江对东南沿海逆温跃层的出现,特别是秋季到冬季初期,有明显的影响,使长江口海域逆温跃层位置偏向东南。暖流对于中国东南沿海的逆温跃层结构,特别是初春时期,有较大影响,使长江口海域的逆温跃层位置向东北偏移。 通过对温跃层长期变化分析得出,黄海冷水团区域,夏季温跃层强度存在3.8年左右的年际变化及18.9年左右的年代际变化,此变化可能主要表现为对当年夏季和前冬东亚地区大气气温的热力响应。东海冷涡区域,夏季温跃层强度存在3.7年的年际变化,在El Nino年为正的强度异常,其可能主要受局地气旋式大气环流变异所影响。谱分析同时表明,该海域夏季温跃层强度还存在33.2年的年代际变化,上世纪70年代中期,温跃层强度由弱转强,而此变化可能与黑潮流量的年代际变化有关。 海洋水温垂直结构的统计预报结果显示,EOF分解的前四个主分量即能够解释原空间点温度距平总方差的95%以上,以海洋表层附近观测资料求解的特征系数推断温度垂直结构分布的结果最稳定。利用东海陆架区、南海深水区和台湾周边海域三个不同区域的实测CTD样本廓线资料,对重构模型的检验结果表明,重构与实测廓线的相关程度超过95%的置信水平。三个区重构与实测温度廓线值的平均误差分别为0.69℃,0.52℃,1.18℃,平均重构廓线误差小于平均气候偏差,统计模式可以很好的估算温度廓线垂直结构。东海陆架海区温度垂直重构廓线与CTD观测廓线获得的温跃层结果对比表明,重构温跃层上界、下界深度和强度的平均绝对误差分别为1.51m、1.36m和0.17℃/m,它们的平均相对误差分别为24.7%、8.9%和22.6%,虽然温跃层深度和强度的平均相对误差较大,但其绝对误差量值较小。而在南海海区,模型重构温跃层上界、下界和强度的平均绝对预报误差分别为4.1m、27.7m和0.007℃/m,它们的平均相对误差分别为16.1%、16.8%和9.5%,重构温跃层各特征值的平均相对误差都在20%以内。虽然南海区温跃层下界深度平均绝对预报误差较大,但相对于温跃层下界深度的空间尺度变化而言(平均温跃层下界深度为168m),平均相对误差仅为16.8%。因此说模型重构的温度廓线可以达到对我国陆架海域、深水区温跃层的较好估算。 基于对历史水文温度廓线观测资料的分析及自主温跃层统计预报模型,研制了实时可利用微机简单、快捷地进行温跃层估算及查询的可视化系统,这是迄今进行大范围海域温跃层统计与实时预报研究的较系统成果。

To explore typhoon effects on dissolved organic carbon (DOC) dynamics, field investigations (tributary and dam site) and laboratory experiments (bioassay and DOC consumption) were conducted in a subtropical reservoir. A tributary survey indicated that after typhoon disruption, upstream areas were the sources of phosphate (P) but not DOC for the dam site located downstream. Bioassay experiments verified P-limitation on bacteria and phytoplankton during summer stratification, and bacteria showed a faster response than algae to added P. Experiments indicated that DOC consumption was determined by the availability of P. The 4 yr typhoon period (June-September) data of the dam site denoted that DOC concentration (27 to 270 mu M C) and its rate of change (-13 to 24 mu M C d(-1)) varied more dramatically in the weak (2006 and 2007) than in the strong (2004 and 2005) typhoon years. The negative correlation of DOC with the ratio of bacterial production (BP) to primary production (PP) in the euphotic zone (0 to 10 m) signified the interactive effects of auto- and heterotrophic processes on DOC variation. In the aphotic zone, the variation of DOC could be ascribed to the change of BP, which showed a positive correlation with P concentrations. This study documents that DOC concentration in the studied system varied at multiple time scales. Such variation can be explained by the decoupling between BP and PP, which is believed to be a function of the limiting nutrient's availability. More importantly, this study suggests that the P supply introduced by strong typhoons might have substantiated a tighter coupling between BP and PP, so that the amplitude of DOC oscillation during the summer period was effectively reduced.

Two biological aerated filters (BAF) were setup for ammonia removal treatment of the circulation water in a marine aquaculture. One of the BAFs was bioaugmented with a heterotrophic nitrifying bacterium, Lutimonas sp. H10, where the ammonia removal was not improved and the massive inoculation was even followed by a nitrification breakdown from day 9 to 18. The nitrification was remained stable in control BAF operated under the same conditions. Fluorescent in situ hybridization (FISH) with rRNA-targeted probes and cultivable method revealed that Lutimonas sp. H10 almost disappeared from the bioaugomented BAF within 3 d, and this was mainly due to the infection of a specific phage as revealed by flask experiment, plaque assay and transmission electron observation. Analyses of 16S rRNA gene libraries showed that bacterial groups from two reactors evolved differently and an overgrowth of protozoa was observed in the bioaugmented BAR Therefore, phage infection and poor biofilm forming ability of the inoculated strain are the main reasons for bioaugmentation failure. In addition, gazing by protozoa of the bacteria might be the reason for the nitrification breakdown in bioaugmented BAF during day 9-18.

Roles and distributions of various forms of nitrogen in biogeochemical cycling in the southern Yellow Sea surface sediments were investigated. The southern Yellow Sea could be divided into three regions (I, II and III) according to the proportion of fine-grained sediment in > 65%, 35-65% and < 35%, respectively. The ratios of different forms of nitrogen contents between each two of the three regions indicated that the nitrogen contents increased with the proportion of fine-grained sediment increasing. The quanta of exchangeable forms of nitrogen were similar in the three regions, while their releasing time increased from regions I to III, indicating that the cycle of nitrogen in fine-grained sediments was shorter than that in coarse-grained sediments. Nitrogen burial fluxes were also similar in these regions, while the burial efficiency increased from regions I to III. The highest burial efficiency was 30.21% in region III, indicating that more than 70% of nitrogen in the southern Yellow Sea surface sediments could be released to take part in biogeochemical recycling. When all the four forms of exchangeable nitrogen (nitrogen in ion exchangeable form (IEF-N), nitrogen in weak acid extractable form (WAEF-N), nitrogen in strong alkali extractable form (SAEF-N) and nitrogen in strong oxidant extractable form (SOEF-N)) were released to take part in recycling, their potential contributions were 80% (SOEF-N), 11% (IEF-N), 6% (SAEF-N), 3% (WAEF-N) respectively, which showed that SOEF-N was the predominant one, and its contribution to biogeochemical cycling was the highest.
