Due to the enormous variety of phytochemicals present in plants, their extracts have been used for centuries in the treatment of innumerous diseases, being perceived as an invaluable source of medicines for humans. Furthermore, the combination of different plants was reported as inducing an improved effect (synergism) in comparison to the additive activity of the plants present in those mixtures. Nevertheless, information regarding the effects of plant infusions added with honey is still rather scarce. Accordingly, the aim of this study was evaluating the interaction between chestnut honey, a natural product with well-reported beneficial properties, and three medicinal plants (either as single plant or as combinations of two and three plants), with regard to their antioxidant activity and hepatotoxicity. Antioxidant activity was evaluated by comparing the results from four different assays; the hepatotoxicity was assessed in two different cell lines. Results were compared by analysis of variance and linear discriminant analysis. The addition of honey to the infusions had a beneficial result in both cases, producing a synergistic effect in all samples, except beta-carotene bleaching inhibition for artichoke+milk thistle+honey preparation and also preparations with lower hepatotoxicity, except in the case of artichoke+honey. Moreover, from discriminant linear analysis output, it became obvious that the effect of honey addition overcame that resulting from using single plant or mixed plants based infusions. Also, the enhanced antioxidant activity of infusions containing honey was convoyed by a lower hepatotoxicity.

Secondary metabolites from plants are important sources of high-value chemicals, many of them being pharmacologically active. These metabolites are commonly isolated through inefficient extractions from natural biological sources and are often difficult to synthesize chemically. Therefore, their production using engineered organisms has lately attracted an increased attention. Curcuminoids, an example of such metabolites, are produced in Curcuma longa and exhibit anti-cancer and anti-inflammatory activities. Herein we report the construction of an artificial biosynthetic pathway for the curcuminoids production in Escherichia coli. Different 4-coumaroyl-CoA ligases (4CL) and polyketide synthases (diketide-CoA synthase (DCS), curcumin synthase (CURS) and curcuminoid synthase) were tested. The highest curcumin production (70 mg/L) was obtained by feeding ferulic acid and with the Arabidopsis thaliana 4CL1 and C. longa DCS and CURS enzymes. Other curcuminoids (bisdemethoxy- and demethoxycurcumin) were also produced by feeding coumaric acid or a mixture of coumaric and ferulic acids, respectively. Curcuminoids, including curcumin, were also produced from tyrosine through the caffeic acid pathway. To produce caffeic acid, tyrosine ammonia lyase and 4-coumarate 3-hydroxylase were used. Caffeoyl-CoA O-methyltransferase was used to convert caffeoyl-CoA to feruloyl-CoA. This pathway represents an improvement of the curcuminoids heterologous production. The construction of this pathway in another model organism is being considered, as well as the introduction of alternative enzymes.

This study aims to (a) identify and profile groups of infants according to their behavioral and physiological characteristics, considering their neurobehavioral organization, social withdrawal behavior, and endocrine reactivity to stress, and to (b) analyze group differences in the quality of mother–infant interaction. Ninety seven 8-week-old infants were examined using the Neonatal Behavioral Assessment Scale and the Alarm Distress Baby Scale. Cortisol levels were measured both before and after routine inoculation between 8 and 12 weeks. At 12 to 16 weeks mother–infant interaction was assessed using the Global Rating Scales of Mother–Infant Interaction. Three groups of infants were identified: (a) ‘‘withdrawn’’; (b) ‘‘extroverted’’; (c) ‘‘underaroused.’’ Differences between them were found regarding both infant and mother behaviors in the interaction and the overall quality of mother–infant interaction. The identification of behavioral and physiological profiles in infants is an important step in the study of developmental pathways.

Noting that maternal depression is common during a baby's first year, this study examined the interaction of depressed and non-depressed mother-child dyads. A sample of 26 first-time mothers with postpartum depression at the third month after birth and their 3-month-old infants was compared to a sample of 25 first-time mothers with no postpartum depression at the third month after birth and their 3-month-old infants. The observations were repeated at 6 months and again at 12 months postpartum. The samples were compared for differences in mother interaction behavior, mother's infant care, mother's concern with the baby, infant behavioral difficulties, infant mental and motor development, and infant behavior with the observer. Among the findings are the following: (1) depressed mothers' interaction behavior and care of their infants are less adequate than the non-depressed mothers' interaction behavior and care of their infants at 3, 6, and 12 months postpartum; (2) infants' interaction behaviors during feeding and face-to-face interaction with depressed mothers are less adequate than infants' interactions with non-depressed mothers at 3, 6, and 12 months postpartum; (3) mother-infant interactions are less adequate in the depressed mother dyads than the non-depressed dyads at 3, 6, and 12 months postpartum; (4) depressed mothers are less concerned about their infants than non-depressed mothers at 3, 6, and 12 months postpartum; (5) infants of depressed mothers have more behavioral difficulties at 3, 6, and 12 months postpartum than infants of non-depressed mothers; (6) infants of depressed mothers had lower mental and motor development rates at 6 and 12 months postpartum than infants of non-depressed mothers; and (7) infants of non-depressed mothers behaved in a more positive way with the observer than the infants of depressed mothers. As Escalas de Avaliação da Interação Mãe-Bebé constituem a versão portuguesa das Interaction Rating Scales, propostas por Field (1980), e têm por objetivo avaliar a interação mãe-bebé, aos 3 meses de idade do bebé. As Escalas de Avaliação da Interação Mãe-Bebé foram administradas a 51 díades mãe-bebé aos 3, 6 e 12 meses pós-parto. A versão portuguesa das escalas mostrou elevados índices de consistência interna – Alfa de Cronbach 0,85 (IRSff bebé), 0,91 (IRSff mãe), 0,87 (IRSal bebé), 0,82 (IRSal mãe), assim como elevada fidelidade e validade concorrente e preditiva. As Escalas de Avaliação da Interação Mãe-Bebé assume-se, assim, como um instrumento robusto na avaliação da interação mãe-bebé, na situação de interação face-a-face e na situação de interação alimentar, podendo ser utilizadas em diferentes amostras e contextos, clínicos e de investigação.

Las bacterias que habitan la rizosfera y que poseen la capacidad de provocar un efecto positivo sobre las plantas son denominadas en su conjunto como Rizobacterias Promotoras del Crecimiento Vegetal (PGPR). Estas bacterias han desarrollado diferentes estrategias para adaptarse a diversas condiciones ambientales. La capacidad para responder a variaciones en la disponibilidad nutricional permite la persistencia de la bacteria en el suelo y mejora sus posibilidades para colonizar la planta hospedadora. En la naturaleza, a menudo las bacterias se encuentran en estructuras de comunidades de microorganismos interconectados denominados biofilms, con un estilo de vida diferente al de la vida en forma planctónica. La formación del biofilm podría representar una estrategia de supervivencia de la rizobacteria a condiciones adversas del suelo. Por Microscopía Confocal de Barrido Láser (CLSM), hemos observado que Rhizobium leguminosarum desarrolla un biofilm característico sobre una superficie abiótica. Hemos identificado algunos de los factores genéticos que influyen en su formación. El presente proyecto propone avanzar en el conocimiento de los factores ambientales y genéticos que influyen sobre la capacidad de las rizobacterias para formar biofilms y su impacto en la interacción con las plantas. A través de enfoques genéticos (mutacionales y de expresión génica) y análisis por CLSM nos proponemos acercarnos a un modelo de los factores de superficie, extracelulares y regulatorios propios de la bacteria que influyen en las propiedades de adhesión y la formación de biofilms. Por último, se intentará correlacionar la emisión de compuestos orgánicos volátiles por las bacterias rizosféricas con ciertos aspectos de la promoción del crecimiento de las plantas.
