This population study, which evaluated two parasitological methods for the diagnosis of schistosomiasis mansoni, was performed in a low-transmission area in Pedra Preta, Montes Claros, Minas Gerais, Brazil. A total of 201 inhabitants of the rural area participated in this research. Four stool samples were obtained from all participants and analysed using the Kato-Katz method (18 slides) and a commercial test, the TF-Test®, which was performed quantitatively. The data were analysed to determine prevalence, the sensitivity of the diagnostic methods, the worm burden and the definition of the "gold standard", which was obtained by totalling the results of all samples examined using the Kato-Katz technique and the TF-Test®. The results showed that the prevalence obtained from the examination of one Kato-Katz slide (the methodology adopted by the Brazilian control programme) was 8% compared to 35.8% from the "gold standard", which was a 4.5-fold difference. This result indicates that the prevalence of schistosomiasis in so-called low-transmission areas is significantly underestimated.

Advances in clinical virology for detecting respiratory viruses have been focused on nucleic acids amplification techniques, which have converted in the reference method for the diagnosis of acute respiratory infections of viral aetiology. Improvements of current commercial molecular assays to reduce hands-on-time rely on two strategies, a stepwise automation (semi-automation) and the complete automation of the whole procedure. Contributions to the former strategy have been the use of automated nucleic acids extractors, multiplex PCR, real-time PCR and/or DNA arrays for detection of amplicons. Commercial fully-automated molecular systems are now available for the detection of respiratory viruses. Some of them could convert in point-of-care methods substituting antigen tests for detection of respiratory syncytial virus and influenza A and B viruses. This article describes laboratory methods for detection of respiratory viruses. A cost-effective and rational diagnostic algorithm is proposed, considering technical aspects of the available assays, infrastructure possibilities of each laboratory and clinic-epidemiologic factors of the infection.

Nucleic acid amplification techniques are commonly used currently to diagnose viral diseases and manage patients with this kind of illnesses. These techniques have had a rapid but unconventional route of development during the last 30 years, with the discovery and introduction of several assays in clinical diagnosis. The increase in the number of commercially available methods has facilitated the use of this technology in the majority of laboratories worldwide. This technology has reduced the use of some other techniques such as viral culture based methods and serological assays in the clinical virology laboratory. Moreover, nucleic acid amplification techniques are now the methods of reference and also the most useful assays for the diagnosis in several diseases. The introduction of these techniques and their automation provides new opportunities for the clinical laboratory to affect patient care. The main objectives in performing nucleic acid tests in this field are to provide timely results useful for high-quality patient care at a reasonable cost, because rapid results are associated with improvements in patients care. The use of amplification techniques such as polymerase chain reaction, real-time polymerase chain reaction or nucleic acid sequence-based amplification for virus detection, genotyping and quantification have some advantages like high sensitivity and reproducibility, as well as a broad dynamic range. This review is an up-to-date of the main nucleic acid techniques and their clinical applications, and special challenges and opportunities that these techniques currently provide for the clinical virology laboratory.

A methodology of exploratory data analysis investigating the phenomenon of orographic precipitation enhancement is proposed. The precipitation observations obtained from three Swiss Doppler weather radars are analysed for the major precipitation event of August 2005 in the Alps. Image processing techniques are used to detect significant precipitation cells/pixels from radar images while filtering out spurious effects due to ground clutter. The contribution of topography to precipitation patterns is described by an extensive set of topographical descriptors computed from the digital elevation model at multiple spatial scales. Additionally, the motion vector field is derived from subsequent radar images and integrated into a set of topographic features to highlight the slopes exposed to main flows. Following the exploratory data analysis with a recent algorithm of spectral clustering, it is shown that orographic precipitation cells are generated under specific flow and topographic conditions. Repeatability of precipitation patterns in particular spatial locations is found to be linked to specific local terrain shapes, e.g. at the top of hills and on the upwind side of the mountains. This methodology and our empirical findings for the Alpine region provide a basis for building computational data-driven models of orographic enhancement and triggering of precipitation. Les textes inédits traduits et présentés dans ce volume constituent un témoignage exceptionnel quant à l'évolution de la pensée de Carnap, l'un des fondateurs de la philosophie analytique. Des premières ébauches d'une construction logique du monde à l'analyse logique des propositions du langage, ils nous font en effet assister aux différents moments d'un programme philosophique inscrit dans le débat du physicalisme et de l'unité de la science. La réductibilité des termes de la psychologie puis de la biologie aux termes de la physique implique-t-elle la dérivabilité de leurs lois aux siennes ? Y a-t-il bonne conséquence d'une épistémologie de la réduction à une réduction ontologique ? Le monde, en somme, est-il entièrement explicable par le langage, et si oui, lequel ?

Résumé Cette thèse de doctorat se fixe comme objectif général de définir la spécificité d'un espace urbain situé en centre-ville à travers sa dynamique sociale et interethnique. Il s'agit de relever les différentes formes et manifestations ethniques, mais également de comprendre la manière dont est vécue cette socialité, tout en tenant compte du contexte urbain dans lequel elles se situent. La particularité de ce travail est le fait de proposer une démarche alliant deux disciplines à savoir la géographie et la sociologie, dans le but de proposer une recherche transdisciplinaire, avec comme conséquence la mise en oeuvre d'une démarche commune nourrie des deux disciplines. Les quartiers ethniques situés en centre-ville ne se présentent pas comme des espaces figés, puisque des changements fréquents sont observés aussi bien au niveau de leur population que de l'affectation des locaux commerciaux et des logements. Ils semblent davantage se caractériser comme des « espaces à l'avenir indéterminé » dans le sens où leur avenir aussi bien au niveau de leur structure urbaine que de leurs rapports sociaux ne peut être prévisible et qu'ils sont par ailleurs susceptibles d'être soumis à de nombreux changements, d'où l'idée également d' « espaces en mutations ». Pour aborder ces thématiques de recherche, nous avons opté pour une étude fondée sur le principe de la représentation et de l'une des formes essentielles à travers lesquelles elles se produisent et se manifestent, à savoir les images. Cette recherche privilégie ainsi une analyse dynamique du phénomène interethnique, l'inscrivant dans le courant constructiviste qui s'intéresse notamment à l'étude des pratiques des acteurs sociaux et leurs représentations, dans le sens qu'elles sont le fruit d'une construction sociale. Dans ce type de démarche analytique, il s'agit de prendre en considération à la fois l'analyse du chercheur et la réalité objective et subjective du vécu des acteurs urbains qui regroupent les riverains, les commerçants, les membres associatifs, les politiques et les médias. C'est justement l'association de ces deux approches qui permet de définir un quartier urbain. Abstract "The spatial and social stakes of an interethnic dynamics in transition in the downtown area. The construction of the various forms of representations in order to define an urban quarter in Nice" The general objective of this PHD is to define the specificity of an urban space located in the downtown area with social and interethnic dynamics. Specifically the diverse ethnic form and sign to include the understanding mariner where society has lived in a specific urban context. The particularity of this work is to advance an analytic method using two disciplines: geography and sociology, in order to bring up a transdisciplinarian research with a common method. The ethnic quarters in the inner-city area are not presented as fixed spaces, since frequent changes were observed in the population level and at the allotment level of the commercial premises and the lodgings. Defined as "undetermined future spaces" as their future in the urban structure or the social relations can not be foresee and are supposed to be subjective to many changes. Therefore speaking of "mutations spaces". The option of this type of thematic research is the study of the principle of representation and essential form which shows and produces the images. Research promotes a dynamic analyses of the interethnic phenomena, inside the constructivist which is focused the practices of social actors, their own representations, and products of a social construction. The production of analytical approach is necessary in order to consider the analyses of the research worker and the objective and subjective reality of the life of the urban actors. It gathers the residents, tradesmen, members of associations, politicians and the press. The association of these types of approach define a urban quarter.

In Niterói, state of Rio de Janeiro, dengue virus type 4 (DENV-4) was isolated for the first time in March 2011. We analysed the laboratory findings of the first cases and evaluated the use of molecular techniques for the detection of DENV-4 in Aedes aegypti that were field-caught. Conventional reverse transcriptase-polymerase chain reaction (RT-PCR) and SimplexaTM Dengue real-time RT-PCR confirmed DENV-4 infection in all cases. Additionally, DENV-4 was confirmed in a female Ae. aegypti with 1.08 x 10³ copies/mL of virus, as determined by quantitative real-time RT-PCR. This is the first time the SimplexaTM Dengue real-time assay has been used for the classification of cases of infection and for entomological investigations. The use of these molecular techniques was shown to be important for the surveillance of dengue in humans and vectors.
