The influence of restricting feed intake of young bulls in feedlots was evaluated in terms of structure of muscular fibers and respective areas, sampled by biopsy on the semitendinoso muscle. Sixty six crossbred Simental-Nelore bulls, 8 months old, averaging 220±34.03 kg were submited for 84 days in phase 1 (growing period), to three treatments: ad libitum (AL), restriction + whole soybean (RWS) and restriction + toasted whole soybean (RTS). The level of restriction of feed intake was 23%. Phase 2 was performed by splitting the animals in each treatment in phase 1 in two groups, feeding one with a diet containing soybeans and the other with poultry litter. The results showed that the animals AL presented more white fibers (FG), compared to the RWS and RST and a larger area of these fibers. A greater frequency of red fibers was observed in treatments RWS and RST. The conclusion was that the restriction of feed intake and consequent compensatory growth contributed for modulation of the muscular fibers increasing the frequency of the fast oxidative glycolitic (FOG) in 10.88% and decreasing of the slow oxidative (SO) and FG in 4.81 and 6.90%, respectively, with possible alteration on meat quality.

With the objective of identifying predictors of fertility in bulls, the correlation coefficients of the midpiece length of bovine spermatozoa with semen traits and fertility were estimated. Data were obtained from semen samples of 50 crossbred bulls submitted to a progeny test. The midpiece length was determined in 40 midpieces of each bull through a image analysis system (Videoplan). Besides the physical traits before and after semen freezing, the oxygen consumption rate and the cytochemical activity index were observed. The correlation coefficients were low and non-significant (P> 0.05), indicating that the midpiece length is not a good predictor of fertility.

Objective: In view of the considerable importance of venereal transmission of bovine leptospirosis, the objective of the present study was to compare the polymerase chain reaction (PCR), culture/isolation and serology to detect leptospire infection in bovine semen. Design: Blood for serologic examination and semen for bacterial culture and PCR were collected from 20 bulls at artificial insemination centres in Brazil. Each animal was sampled twice for serology. Result: Forty-five percent (9/20) of the serum samples collected showed agglutinin titers to serovar hardjo in the first sample and 25% (5/20) had agglutinin titers to serovar hardjo in the second sample. Eighty percent (16/20) of semen samples were positive by PCR. Leptospires could not be isolated from any of the semen samples examined. Conclusion: Polymerase chain reaction can be a method of great potential for the detection of leptospires at artificial insemination centres.

The objective of this study was to estimate the relative effects of genetic and phenotypic factors on the efficacy and efficiency of superovulation for Holstein-Friesian cows reared in Brazil. A database, established by the Associacao Brasileira de Criadores de Bovinos da Raca Holandesa, consisting of a total of 5387 superovulations of 2941 cows distributed over 473 herds and sired by 690 bulls was used for the analysis. The records were analyzed by MTDFREML (Multiple Trait Derivative-Free Restricted Maximum Likelihood), using a repeatability animal model. The fixed effects included in the model were contemporaneous group (veterinarian, herd, year and season of the superovulation); number of semen doses; cow age; and superovulation order. The estimated repeatability of the number of the transferable embryos was low (0.13), and the estimated heritability was 0.03. These results indicate that environmental factors play a critical role in the response of a cow to a superovulation treatment. There is little evidence that future responses to superovulation by individual females can be predicted by previous treatment(s) or that superovulation response is an heritable trait.

Thirty 3/4 Canchim + 1/4 Nelore young bulls with 417 kg of body weight and 15 months of age, were confined during 84 days. The animals were fed with diets composed with corn silage, corn grain, cottonseed meal, soybean meal, whole soybean and mineral mix, adjusted in agreement with the recommendations of the Metabolizable Protein System (MP), Cornell Net Carbohydrate and Protein System (CNCPS) and Intestine Digestible Protein System (PDI), for predicted body weight gain of 1.3 kg/head/day. The daily body weight gain did not differ among treatments CNCPS, MP and PDI, with 1.51; 1.48; and 1.13 kg/head, respectively. The economic analysis revealed net profit of R$116.25; R$148.30; and R$108.51/head for CNCPS, MP and PDI systems, respectively. The diets adjusted by CNCPS and MP systems provided superior animal performance than that expected, while the diet adjusted by PDI system did not allow the predicted body weight gain.

Data concerning daily milk yield (MY), percentage of milk fat (%F), protein (%P), lactose (%LT), and total solids (%TS), and somatic cell counts (SCC) for a herd of 222 Murrah buffalo reared in the state of São Paulo, Brazil, were collected monthly from 1997 to 2000 in order to study the factors affecting SCC and their relation to milk production and constituents during lactation. SCC decreased in the second month of lactation and increased thereafter, up to the ninth month of lactation. The interaction of month of lactation x order of calving was significant. Mean MY observed during the first month of lactation was 6.87 kg, which increased to 7.65 kg during the second month, and then decreased until the ninth month of lactation (3.83 kg). During the different months of lactation, %F, %P, %LT, and %TS ranged from 6.28 to 8.38%, 4.05 to 4.59%, 4.96 to 5.34%, and 16.94 to 18.55%, respectively. Calving year, calving order, and order of month of lactation significantly affected MY, %F, %P, %LT, and %TS. The regression coefficients of transformed SCC on MY and %LT were negative and significant during all months of lactation, showing that milk and lactose yield decreased with increased transformed SCC, causing losses to buffalo milk producers.

Caseins comprise make up about 80% of the total protein content of milk and present polymorphism with change in the amino acid sequence. Within this abundance of proteins, kappa-casein is noteworthy, since it has been associated with differences in milk yield, composition and processing. The objective of this study was to observe the existence of polymorphism in the kappa-casein gene in female buffaloes. For this purpose, blood samples from 115 female buffaloes, collected with vacutainer by needle punctionure of the jugular vein, were used. for genomic DNA extraction was done from blood samples. The PCR-RFLP and SSCP techniques demonstrated that the studied animals were monomorphic for the kappa-casein gene. Only allele B was observed in these animals, which was present in homozygosis. Therefore, it was not possible to quantify the gene action on milk yield and its constituents. The monomorphism observed in the population studied would allow the development of a method to identify mixtures of cow and buffalo milk in mozzarella cheese production, especially because, in cattle, the kappa-casein gene is polymorphic. The objective of this study was to evaluate the LH surge after last hormonal injection of synchronization of ovulation protocols in buffalo. Fifteen multiparous buffaloes received 25 mg of Lecirelin in Day 0, and 150 mg of D-Cloprostenol on Day 7. On Day 8, estradiol benzoate was injected in Group 1 (0.5 mg, n = 5) and Group 2 (1.0 mg, n = 5). On Day 9, five buffaloes received 25 mg of Lecirelin (Control). Blood samples were collected for measure the LH concentrations on Day 7 and then every 3 hours until 72 hours after the PGF 2a injection. For evaluation of LH surge were compared the interval between PGF 2a injection to LH surge, duration, amplitude and area under the LH peak. The LH surge occurred 51.0 + 0.0 hours, 47.3 + 2.7 hours and 47.0 + 3.8 hours after PGF 2a injection for Control, Group 1 and Group 2, respectively (P > 0.05). The duration of LH peak in Control (7.8 + 1.5 hours) was shorter than Groups 1 and 2 (10.5 + 1.5 hours vs. 10.8 + 2.4 hours, respectively; P < 0.05). The amplitudes of LH peak were 4.5 + 0.4 ng/mL, 4.0 + 0.4 ng/mL and 4.3 + 0.8 ng/mL for Control, Group 1 and Group 2, respectively (P > 0.05). The area under LH peak for Control (4.8 ± 0.7) was smaller than the areas of the Groups 1 and 2 (8.8 ± 2.5 vs. 8.7 2.2, respectively; P < 0.05). In summary, the estradiol benzoate injection provided higher duration and area of LH peak than GnRH injection in Ovsynch protocol in buffalo.
