The routine semen evaluation assessing sperm concentration, motility and morphology, does not identify subtle defects in sperm chromatin architecture. Bulls appear to have stable chromatin, with low levels of DNA fragmentation. However, the nature of fragmentation and its impact on fertility remain unclear and there are no detailed reports characterizing the DNA organization and damage in this species. The intensive genetic selection, the use of artificial insemination and in vitro embryo production associated to the cryopreservation process can contribute to the chromatin damage and highlights the importance of sperm DNA integrity for the success of these technologies. Frozen-thawed semen samples from three ejaculates from a Nellore bull showed high levels of morphological sperm abnormalities (55.8±5.1%), and were selected for complementary tests. Damage of acrosomal (76.9±8.9%) and plasma membranes (75.7±9.3%) as well as sperm DNA strand breaks (13.8±9.5%) and protamination deficiency (3.7±0.6%) were significantly higher compared to the values measured in the semen of five Nellore bulls with normospermia (24.3±3.3%; 24.5±6.1%; 0.6±0.5%; 0.4±0.6% for acrosome, plasma membrane, DNA breaks and protamine deficiency, respectively) (P<0.05). Motility and percentage of spermatozoa with low mitochondrial potential showed no differences between groups. This study shows how routine semen analyses (in this case morphology) may point to the length and complexity of sperm cell damage emphasizing the importance of sperm function testing.

Futsal is one of the most popular sports in Brazil, played on five continents, in hundreds of countries, but is still a poorly studied sports science. Among the types of training used in the preparation of soccer players, the collective training is one of the most used because it is a simulation game. The application of the training load is often made in large quantities (volume) with exercise duration and intensity greater than what the athlete can handle. Thus, this study aimed to evaluate the intensity of physical exertion of futsal players of basic category during collective training. We evaluated twenty (N=20) soccer players from two teams with different levels of training. There were ten players on a team federated (FED) and ten players on a team for a project extension (EXT). Samples were collected at the respective locations of training. An evaluation of anthropometric and during collective training heart rate was measured and at the end of each training time blood samples were collected for analysis of blood lactate concentration and perceived exertion of the players. There was significant difference between the two teams only in fat percentage (FED=13,4 e EXT=21,2). The average heart rate during training was 174 ± 12.2 bpm, which corresponds to 84% HRmax. There was significant difference between heart rate and the percentage of HRmax in the two parts of training in both teams. The Federated team players remained most of the time in more intense intensity zone (above 85% HRmax), while the EXT players were most of the time zone of moderate intensity (65-85% HRmax). The rating of perceived exertion (FED=3,5 e EXT=3,2) and blood lactate concentration (FED=2.4 mmol/L and EXT=2.9 mmol/L) showed no significant difference between the two teams and between the two times. High throughput sequencing (HTS) provides new research opportunities for work on non-model organisms, such as differential expression studies between populations exposed to different environmental conditions. However, such transcriptomic studies first require the production of a reference assembly. The choice of sampling procedure, sequencing strategy and assembly workflow is crucial. To develop a reliable reference transcriptome for Triatoma brasiliensis, the major Chagas disease vector in Northeastern Brazil, different de novo assembly protocols were generated using various datasets and software. Both 454 and Illumina sequencing technologies were applied on RNA extracted from antennae and mouthparts from single or pooled individuals. The 454 library yielded 278 Mb. Fifteen Illumina libraries were constructed and yielded nearly 360 million RNA-seq single reads and 46 million RNA-seq paired-end reads for nearly 45 Gb. For the 454 reads, we used three assemblers, Newbler, CAP3 and/or MIRA and for the Illumina reads, the Trinity assembler. Ten assembly workflows were compared using these programs separately or in combination. To compare the assemblies obtained, quantitative and qualitative criteria were used, including contig length, N50, contig number and the percentage of chimeric contigs. Completeness of the assemblies was estimated using the CEGMA pipeline. The best assembly (57,657 contigs, completeness of 80 %, < 1 % chimeric contigs) was a hybrid assembly leading to recommend the use of (1) a single individual with large representation of biological tissues, (2) merging both long reads and short paired-end Illumina reads, (3) several assemblers in order to combine the specific advantages of each.

The use of legumes intercropped with forage in the Brazilian savannah (Cerrado) region is an alternative to reverse the process of pasture degradation. The natural phosphorus deficiency in this region and the high retention capacity of this nutrient in the soil are factors that directly affect the sustainability of the production system. The objective of this study was to evaluate the effect of a phosphate with medium solubility (Arad) compared to more soluble sources such as triple superphosphate and mono-ammonium phosphate, on the formation and maintenance of pastures of Andropogon gayanus with or without the introduction of Stylosantes guianensis in pasture already established in the Brazilian savannah. Two experiments were carried out under field conditions in an Oxisol using a randomized block design with split plots, plots with two sources of phosphorus (Arad and super-triple or MAP) and subplots with different levels of phosphorus. The study showed that andropogon grass, even though characterised as forage requiring low fertilization, responded to increased fertilization, especially with the use of more soluble sources of P at planting. From the second year on, with the introduction of Stylosantes guianensis, verified effects were seen on legumes in their efficiency of utilization of residual fertilization from the first year; however, this didn't affect legume dry matter production resulting from the P sources used for the andropogon grass. The mixed stands of andropogon grass and Stylosantes guianensis made it more advantageous to increase the P sources using phosphate fertilizer with lower solubility (Arad), when compared to MAP.

Incubation temperature manipulation during fetal development reduces adiposity of broiler hatchlings

Broilers are known as an efficient source of lean meat. Genetic selection resulted in broiler strains with large body size and fast growth, but a concomitant increase in fat deposition also occurred. Other than reducing nutrient intake, there is a lack of alternative methods to control body fat composition of broilers. The present study assessed whether incubation temperature (machine temperatures: 36ºC, 37.5ºC, and 39ºC; eggshell temperatures: 37.4 ± 0.08°C, 37.8 ± 0.15ºC, and 38.8 ± 0.33°C, respectively.) from d 13 affects broiler hatchling fat deposition. We analyzed adipocyte hypertrophy and proliferation in 3 body regions; weight and chemical composition of yolk-free chicks and yolk sacs; and serum lipid profile. Increased incubation temperature reduced abdominal and cervical adipocyte size. Independently of temperature, cervical adipocytes were smaller and showed higher proliferation than adipocytes in the abdominal and thigh regions. Smaller cervical adipocytes were observed in birds from eggs incubated at 36ºC and 39ºC. With regard to weight and composition of chicks, ash content as a percentage of dry matter was the only variable affected by temperature; it was higher in chicks from eggs incubated at 36ºC than at 39ºC and showed no significant difference between chicks incubated at 39ºC and 37.5ºC. Absolute and relative weights of yolk sacs were higher from eggs incubated at 39ºC than at 36ºC, and these two treatments did not differ from the 37.5ºC control. Absolute measures of yolk sac lipids, moisture, dry matter, and crude protein content were lower in chicks from eggs incubated at 36ºC, and no significant differences were found for these variables between chicks from eggs incubated at 37.5ºC and 39ºC. Hatchlings from eggs incubated at 36°C had significantly higher cholesterol levels than chicks incubated at the other 2 temperatures, but no additional effects on blood lipids were detected. Incubation temperature manipulation during fetal development altered cervical and abdominal adipocyte size in broiler hatchlings and could become a tool in hatcheries to manipulate chick quality, although further studies are needed to evaluate its long-term effects.
