Customer knowledge management (CKM) practices enable organizations to create customer competence with systematic use of customer information that is integrated throughout the organization. Nonetheless, organizations are not able to fully exploit the vast amount of data available. Previous research on use of customer information is limited especially in a multichannel environment. The aim of this study was to identify the main obstacles for utilizing customer information efficiently across multiple sales channels. The study was conducted as a single case study in order to gain deeper understanding of the research problem. The empirical findings indicate that lack of CKM practices and a common goal are major challenges obstructing effective utilization of customer information. Furthermore, decentralized organizational structure and insufficient analytical skills create obstacles for information sharing and capabilities to process information and create new knowledge. The implications of the study suggest that in order to create customer competence organizations should shift their focus from technology to the organizational factors affecting use of information and implement CKM practices throughout the organization.

Tämän pro gradu -tutkimuksen tavoitteena oli tutkia osaamisen johtamisen käytäntöjä ja kehittämistarpeita kohdeorganisaatiossa henkilöstön näkökulmasta. Tutkimuksen tarkoituksena oli selvittää, miten tehtyjä henkilöstötutkimuksia on hyödynnetty kohdeorganisaatiossa ja näkyykö tämä hyödyntäminen käytännön johtamisessa henkilöstön mielestä. Tutkimus suoritettiin laadullisena tapaustutkimuksena ja empiirinen aineisto kerättiin kuudella teemahaastattelulla, jotka analysoitiin sisällönanalyysimenetelmällä. Tutkimuksessa kartoitettiin, millaisia osaamisen johtamisen käytäntöjä kohdeorganisaatiossa on. Tutkimuksen tuloksien perusteella osaamisen johtamisen peruselementit ovat olemassa: säännölliset palaverit koettiin tärkeiksi, mutta koulutuksien ja kehityskeskusteluiden järjestämisessä sekä jälkiseurannassa henkilöstö näki parannettavaa. Myös hiljaisen tiedon siirtäminen ja hyödyntäminen koettiin merkittäväksi tekijäksi organisaation onnistuneen toiminnan ja jatkuvuuden kannalta. Tutkimuksessa todettiin osaamisen johtamisen kehittämisen ja osaamisen kehittämisen menetelmien linkittyvän vahvasti toisiinsa ja tutkimuksen tuloksena esitettiin konkreettisia toimenpide-ehdotuksia osaamisen johtamisen kehittämisestä kohdeorganisaation johdolle. Tutkimuksen mukaan osaamisen johtaminen ja henkilöstöjohtaminen sulautuvat toisiinsa, eikä niitä voida tarkastella erikseen. Johdon ja henkilöstön välisellä vuorovaikutuksella on suuri merkitys onnistuneelle osaamisen johtamiselle. Keskeistä on kytkeä osaamisen johtaminen organisaation käytäntöihin ja varata sen implementointiin riittävästi resursseja.

Consumers’ increasing awareness of healthiness and sustainability of food presents a great challenge to food industry to develop healthier, biologically active and sustainable food products. Bioactive peptides derived from food proteins are known to possess various biological activities. Among the activities, the most widely studied are antioxidant activities and angiotensin I converting enzyme (ACE) inhibitory activity related to blood pressure regulation and antihypertensive effects. Meanwhile, vast amounts of byproducts with high protein content are produced in food industry, for example potato and rapeseed industries. The utilization of these by-products could be enhanced by using them as a raw material for bioactive peptides. The objective of the present study was to investigate the production of bioactive peptides with ACE inhibitory and antioxidant properties from rapeseed and potato proteins. Enzymatic hydrolysis and fermentation were utilized for peptide production, ultrafiltration and solid-phase extraction were used to concentrate the active peptides, the peptides were fractionated with liquid chromatographic processes, and the peptides with the highest ACE inhibitory capacities were putified and analyzed with Maldi-Tof/Tof to identify the active peptide sequences. The bioavailability of the ACE inhibitory peptides was elucidated with an in vitro digestion model and the antihypertensive effects in vivo of rapeseed peptide concentrates were investigated with a preventive premise in 2K1C rats. The results showed that rapeseed and potato proteins are rich sources of ACE inhibitory and antioxidant peptides. Enzymatic hydrolysis released the peptides effectively whereas fermentation produced lower activities.The native enzymes of potato were also able to release ACE inhibitory peptides from potato proteins without the addition of exogenous enzymes. The rapeseed peptide concentrate was capable of preventing the development of hypertension in vivo in 2K1C rats, but the quality of rapeseed meal used as raw material was found to affect considerably the antihypertensive effects and the composition of the peptide fraction.

In recent decade customer loyalty programs have become very popular and almost every retail chain seems to have one. Through the loyalty programs companies are able to collect information about the customer behavior and to use this information in business and marketing management to guide decision making and resource allocation. The benefits for the loyalty program member are often monetary, which has an effect on the profitability of the loyalty program. Not all the loyalty program members are equally profitable, as some purchase products for the recommended retail price and some buy only discounted products. If the company spends similar amount of resources to all members, it can be seen that the customer margin is lower on the customer who bought only discounted products. It is vital for a company to measure the profitability of their members in order to be able to calculate the customer value. To calculate the customer value several different customer value metrics can be used. During the recent years especially customer lifetime value has received a lot of attention and it is seen to be superior against other customer value metrics. In this master’s thesis the customer lifetime value is implemented on the case company’s customer loyalty program. The data was collected from the customer loyalty program’s database and represents year 2012 on the Finnish market. The data was not complete to fully take advantage of customer lifetime value and as a conclusion it can be stated that a new key performance indicator of customer margin should be acquired in order to profitably drive the business of the customer loyalty program. Through the customer margin the company would be able to compute the customer lifetime value on regular basis enabling efficient resource allocation in marketing.

As technology has developed it has increased the number of data produced and collected from business environment. Over 80% of that data includes some sort of reference to geographical location. Individuals have used that information by utilizing Google Maps or different GPS devices, however such information has remained unexploited in business. This thesis will study the use and utilization of geographically referenced data in capital-intensive business by first providing theoretical insight into how data and data-driven management enables and enhances the business and how especially geographically referenced data adds value to the company and then examining empirical case evidence how geographical information can truly be exploited in capital-intensive business and what are the value adding elements of geographical information to the business. The study contains semi-structured interviews that are used to scan attitudes and beliefs of an organization towards the geographic information and to discover fields of applications for the use of geographic information system within the case company. Additionally geographical data is tested in order to illustrate how the data could be used in practice. Finally the outcome of the thesis provides understanding from which elements the added value of geographical information in business is consisted of and how such data can be utilized in the case company and in capital-intensive business.

Nascent procollagen peptides and other secretory proteins are transported across the endoplasmic reticulum (ER) membrane through a protein-conducting channel called translocon. Sec61alpha, a multispanning membrane translocon protein, has been implicated as being essential for translocation of polypeptide chains into the cisterns of the ER. Sec61alpha forms a protein complex with collagen and Hsp47, an ER-resident heat shock protein that binds specifically to collagen. However, it is not known whether Sec61alpha is ubiquitously produced in collagen-producing F9 teratocarcinoma cells or under heat shock treatment. Furthermore, the production and utilization of Sec61alpha may depend on the stage of cell differentiation. Cultured F9 teratocarcinoma cells are capable of differentiation in response to low concentrations of retinoic acid. This differentiation results in loss of tumorigenicity. Mouse F9 cells were grown in culture medium at 37ºC and 43ºC (heat shock treatment) treated or not with retinoic acid, and labeled in certain instances with 35S-methionine. Membrane-bound polysomes of procollagen IV were then isolated. Immunoprecipitation and Western blot analysis were performed using polyclonal antibodies against collagen IV, Hsp47 and Sec61alpha. Under retinoic acid-untreated conditions, F9 cells produced undetectable amounts of Sec61alpha. Sec61alpha, Hsp47 and type IV collagen levels were increased after retinoic acid treatment. Heat shock treatment did not alter Sec61alpha levels, suggesting that Sec61alpha production is probably not affected by heat shock. These data indicate that the enhanced production of Sec61alpha in retinoic acid-induced F9 teratocarcinoma cells parallels the increased synthesis of Hsp47 and collagen type IV.

Tutkimus sijoittuu varhaiskasvatuksen hajautetun organisaation kontekstiin, mutta tulokset ovat siirrettävissä muihinkin suomalaisiin kasvatus- ja opetustoimen organisaatioihin. Hajautettujen organisaatioiden tutkimus on ollut varhaiskasvatuksen kentällä vielä vähäistä, vaikka organisaatiomallin vaikutukset johtajuuden toteuttamiselle ovat merkittävät. Hajautetulla organisaatiolla varhaiskasvatuksessa tarkoitetaan sitä, että yhden johtajan alaisuudessa on monta eri päiväkotia tai erilaisia päivähoitomuotoja. Tämä organisaatiomalli on yhä enenevässä määrin kasvava suomalaisessa varhaiskasvatuksessa. Varhaiskasvatuksen hajautettujen organisaatioiden tutkimuksessa on aiemmin tarkasteltu johtajan ja työntekijöiden ja työntekijöiden keskinäisiä ammatillisia suhteita. Tässä tutkimuksessa näkökulma painottuu johtamiseen ja työskentelyyn hajautetuissa organisaatiossa sinänsä sekä myös laadunarviointiin sekä pedagogiikkaan. Viitekehyksenä tutkimuksessa on LMX-teoria (leader-member-exchange, johtajuuden vaihtoteoria), jossa tarkastellaan esimies-alaissuhdetta ja siihen kiinteästi liittyvää luottamuksen käsitettä. Luottamuksen merkitys hajautetuissa organisaatioissa korostuu, koska esimies ei ole fyysisesti päivittäin läsnä työntekijöiden arjessa. Tutkimuksessa tarkastellaan hajautetuissa varhaiskasvatuksen organisaatioissa työskentelyä seuraavien tutkimuskysymysten avulla: 1) Mitkä ovat varhaiskasvatuksen hajautettujen organisaatioiden johtamisen erityispiirteet? 2) Miten eri työntekijäryhmät kokevat hajautetussa organisaatiossa työskentelyn? 3) Millaisia kokemuksia esimiehillä ja työntekijöillä on heidän yksiköissään toteutetusta laadunarvioinnista? 4) Millaiseksi työntekijät ja esimiehet kokevat esimieheltään saadun tuen? Tutkimuksessa oli kolme eri aineistoa. Ensimmäinen aineisto koostui 11 hajautetun organisaation johtajan haastattelusta. Toinen aineisto (n = 223) sisälsi haastateltujen esimiesten lomakevastausten lisäksi heidän alaisuudessaan toimivien työntekijöiden, 10 esimieskoulutukseen osallistuneen johtajan sekä kolmen erillisyksikön työntekijöiden vastaukset. Kolmas aineisto oli kerätty pääkaupunkiseudulta varhaiskasvatuksen johtajilta lomakekyselynä (n = 112). Aineistoa on analysoitu teorialähtöisen ja aineistolähtöisen sisällönanalyysin ja tilastollisten analyysien avulla Tulokset osoittavat, että johtajat kokivat hallinnollisten töiden vievän paljon aikaa. Esimiehen kanssa eri työpaikassa työskentelevät työntekijät hahmottivat koko organisaation selkeämmin kuin esimiehen kanssa fyysisesti samassa paikassa työskentelevät. Esimiesten käsitysten mukaan laadunarviointia suoritettiin enemmän kuin mitä työntekijöiden mukaan. Työntekijät kaipasivat esimiehiltään tukea yhteistyöhön ja vuorovaikutukseen, pedagogiseen ohjaukseen, kehittämiseen ja toiminnan resursseihin liittyen. Erillisyksikössä työskentelevät kokivat saavansa enemmän tukea kuin esimiehen kanssa fyysisesti samassa yksikössä työskentelevät työntekijät. Sekä esimieheltä saadun pedagogisen tuen että luottamuksen kokemukset kiinnittävät tämän tutkimuksen tulosten mukaan huomion rakenteiden merkitykseen hajautetuissa organisaatioissa. Arviointiin, pedagogiseen tukeen ja tiedonkulkuun liittyvien rakenteiden huomioiminen helpottaa hajautetussa organisaatiossa johtamista. Edellisten lisäksi johtajan selkeä visio omasta johtamistyöstään ja jaetun johtajuuden hyödyntäminen edesauttavat työn hallinnan kokemuksia.

Objective of the thesis is to create a value based pricing model for marine engines and study the feasibility of implementing such model in the sales organization of a specific segment in the case company’s marine division. Different pricing strategies, concept of “value”, and how perceptions of value can be influenced through value based marketing are presented as theoretical background for the value based pricing model. Forbis and Mehta’s Economic Value to Customer (EVC) was selected as framework to create the value based pricing model for marine engines. The EVC model is based on calculating and comparing life-cycle costs of the reference product and competing products, thus showing the quantifiable value of the company’s own product compared to competition. In the applied part of the thesis, the components of the EVC model are identified for a marine diesel engine, the components are explained, and an example calculation created in Excel is presented. When examining the possibilities to implement in practice a value based pricing strategy based on the EVC model, it was found that the lack of precise information on competing products is the single biggest obstacle to use EVC exactly as presented in the literature. It was also found that sometimes necessary communication channels are missing and that there is simply a lack of interest from some clients and product end-users part to spend time on studying the life-cycle costs of the product. Information on the company’s own products is however sufficient and the sales force is capable to communicate to sufficiently high executive levels in the client organizations. Therefore it is suggested to focus on quantifying and communicating the company’s own value proposition. The dynamic nature of the business environment (variance in applications in which engines are installed, different clients, competition, end-clients etc.) means also that each project should be created its own EVC calculation. This is demanding in terms of resources needed, thus it is suggested to concentrate on selected projects and buyers, and to clients where the necessary communication channels to right levels in the customer organization are available. Finally, it should be highlighted that as literature suggests, implementing a value based pricing strategy is not possible unless the whole business approach is value based.

Cardiac troponins (cTn) I and T are the current golden standard biochemical markers in the diagnosis and risk stratification of patients with suspected acute coronary syndrome. During the past few years, novel assays capable of detecting cTn‐concentrations in >50% of apparently healthy individuals have become readily available. With the emerging of these high sensitivity cTn assays, reductions in the assay specificity have caused elevations in the measured cTn levels that do not correlate with the clinical picture of the patient. The increased assay sensitivity may reveal that various analytical interference mechanisms exist. This doctoral thesis focused on developing nanoparticle‐assisted immunometric assays that could possibly be applied to an automated point‐of‐care system. The main objective was to develop minimally interference‐prone assays for cTnI by employing recombinant antibody fragments. Fast 5‐ and 15‐minute assays for cTnI and D‐dimer, a degradation product of fibrin, based on intrinsically fluorescent nanoparticles were introduced, thus highlighting the versatility of nanoparticles as universally applicable labels. The utilization of antibody fragments in different versions of the developed cTnI‐assay enabled decreases in the used antibody amounts without sacrificing assay sensitivity. In addition, the utilization of recombinant antibody fragments was shown to significantly decrease the measured cTnI concentrations in an apparently healthy population, as well as in samples containing known amounts of potentially interfering factors: triglycerides, bilirubin, rheumatoid factors, or human anti‐mouse antibodies. When determining the specificity of four commercially available antibodies for cTnI, two out of the four cross‐reacted with skeletal troponin I, but caused crossreactivity issues in patient samples only when paired together. In conclusion, the results of this thesis emphasize the importance of careful antibody selection when developing cTnI assays. The results with different recombinant antibody fragments suggest that the utilization of antibody fragments should strongly be encouraged in the immunoassay field, especially with analytes such as cTnI that require highly sensitive assay approaches.

Tutkimus kuvaa tiedon, osaamisen ja teknologian siirtoa Suomesta ja Itävallasta Puolaan, Romaniaan ja Slovakiaan hajautetussa puupohjaisessa sähkö- ja lämpöenergian tuotannossa. Metsävaroiltaan rikas ja bioenergia-asioissa toimintatavoiltaan edistynyt Suomi toimi Metsäntutkimuslaitoksen (nykyisin Luonnonvarakeskus) johdolla tutkimuksen empiirisen aineiston tuottaneen kehittäjäverkostohankkeen pääkoordinaattorina vuosina 2011-2014. Tutkimusmenetelmänä käytettiin hankkeen dokumentaation sisällönanalyysiä. Itävalta on tunnettu edistyksellisistä bioenergia-alan tuki- ja ohjausjärjestelmistä. Suomi ja Itävalta kuuluvat EU:n viiden edistyneimmän maan joukkoon uusiutuvien energialähteiden hyödyntämisessä. Tämän työn tavoitteena oli selvittää, miten Suomessa ja Itävallassa hyvin toimivia liiketoimintamalleja voidaan siirtää kohdemaihin puupohjaisen uusiutuvan energian tuotannossa hyödynnettäviksi. Työssä kuvataan tiedonsiirtäjämaiden eli Suomen ja Itävallan ydinosaaminen kiinteän biomassan energiantuotannossa politiikkatasolta käytännön liiketoiminnan tasolle. Lisäksi työssä analysoidaan poliittisen ohjauksen merkitystä alan kehittämisessä, missä käydään läpi maakohtaiset uusiutuvan energian toimintasuunnitelmat vuodelta 2010. Lopuksi arvioidaan tiedonsiirtäjä- ja tiedonhyödyntäjämaiden välisiä eroavaisuuksia kyseisellä liiketoiminta-alueella, ja onko olemassa sellaisia tekijöitä, mitkä estävät tiedonsiirtoa tai hyväksi todettujen liiketoimintamallien soveltamista kohdemaissa. Kussakin maassa metsänomistusolosuhteet ratkaisevat kiinteän biomassan tarjonnan toimivuuden eli tilanteen toimitusketjun alkupäässä. Tuotannon ohjauksen pyrkimys on myös hyödyntää mekaanisen puunjalostuksen sivutuotevirrat mahdollisimman tarkoin energiantuotannon tarpeisiin. Yleiset taloudelliset suhdanteet vaikuttavat ketjun toimivuuteen ja tehokkuuteen. Yksin energiantuotannon tarpeisiin puunkorjuuta ei kannata suunnitella, koska se harvemmin erikseen toteutettuna on kannattavaa liiketoimintaa. Puun käyttö energiantuotantoon tarvitsee hyvin suunniteltua tuki- ja ohjausjärjestelmää, joista kansalliset hallitukset vastaavat. Suomalainen tuotannon suunnittelun ja ohjauksen ajattelutapa sekä itävaltalainen energiapuun varastointi biomassan logistiikkakeskuksiin koettiin kohdemaiden yrittäjäkunnassa varteenotettavimpina liiketoiminnan kehittämisvaihtoehtoina paikallisissa toimintaympäristöissä. Lean-tuotantoajatteluun kuuluva hukan poistaminen toimitusketjusta liittyy mm. varastonhallinnan järjestelyihin ja sivutuotevirtojen hyödyntämiseen. Näitä piirteitä oli myös löydettävissä tiedonhyödyntäjämaiden yritysten toiminnassa hankkeen toteutuksen aikana.

cDNA microarray is an innovative technology that facilitates the analysis of the expression of thousands of genes simultaneously. The utilization of this methodology, which is rapidly evolving, requires a combination of expertise from the biological, mathematical and statistical sciences. In this review, we attempt to provide an overview of the principles of cDNA microarray technology, the practical concerns of the analytical processing of the data obtained, the correlation of this methodology with other data analysis methods such as immunohistochemistry in tissue microarrays, and the cDNA microarray application in distinct areas of the basic and clinical sciences.

Alternative methods to the utilization of laboratory animal blood and its by-products are particularly attractive, especially regarding hamsters due to their small size and difficulties in obtaining serial blood samples. Steroid hormone metabolite quantification in feces, widely used in studies of free-ranging or intractable animals, is a non-invasive, non-stressor, economical, and animal saving technique which allows longitudinal studies by permitting frequent sampling of the same individual. The present study was undertaken to determine the suitability of this method for laboratory animals. Estradiol and progesterone metabolites were quantified by radioimmunoassay in feces of intact, sexually mature female Syrian hamsters during the estrous cycle (control) and in feces of superovulated females. Metabolites were extracted by fecal dilution in ethanol and quantified by solid phase radioimmunoassay. Median estrogen and progesterone concentrations were 9.703 and 180.74 ng/g feces in the control group, respectively. Peaks of estrogen (22.44 ± 4.54 ng/g feces) and progesterone (655.95 ± 129.93 ng/g feces) mean fecal concentrations respectively occurred 12 h before and immediately after ovulation, which is easily detected in this species by observation of a characteristic vaginal postovulatory discharge. Median estrogen and progesterone concentrations (28.159 and 586.57 ng/g feces, respectively) were significantly higher in superovulated animal feces (P < 0.0001). The present study demonstrated that it is possible to monitor ovarian activity in Syrian hamsters non-invasively by measuring fecal estradiol and progesterone metabolites. This technique appears to be a quite encouraging method for the development of new endocrinologic studies on laboratory animals.

Today’s healthcare organizations are under constant pressure for change, as hospitals should be able to offer their patients the best possible medical care with limited resources and, at the same time, to retain steady efficiency level in their operation. This is challenging, especially in trauma hospitals, in which the variation in the patient cases and volumes is relatively high. Furthermore, the trauma patient's care requires plenty of resources as most the patients have to be treated as single cases. Occasionally, the sudden increases in demand causes congestion in the operations of the hospital, which in Töölö hospital appears as an increase in the surgery waiting times within the yellow urgency class patients. An increase in the surgery waiting times may cause the diminution of the patient's condition, which also raises the surgery risks. The congestion itself causes overloading of the hospital capacity and staff. The aim of this master’s thesis is to introduce the factors contributing to the trauma process, and to examine the correlation between the different variables and the lengthened surgery waiting times. The results of this study are based on a three-year patient data and different quantitative analysis. Based on the analysis, a daily usable indicator was created in order to support the decision making in the operations management. By using the selected indicator, the effects of congestion can be acknowledged and the corrective action can also be taken more proactively.

An electric system based on renewable energy faces challenges concerning the storage and utilization of energy due to the intermittent and seasonal nature of renewable energy sources. Wind and solar photovoltaic power productions are variable and difficult to predict, and thus electricity storage will be needed in the case of basic power production. Hydrogen’s energetic potential lies in its ability and versatility to store chemical energy, to serve as an energy carrier and as feedstock for various industries. Hydrogen is also used e.g. in the production of biofuels. The amount of energy produced during hydrogen combustion is higher than any other fuel’s on a mass basis with a higher-heating-value of 39.4 kWh/kg. However, even though hydrogen is the most abundant element in the universe, on Earth most hydrogen exists in molecular forms such as water. Therefore, hydrogen must be produced and there are various methods to do so. Today, the majority hydrogen comes from fossil fuels, mainly from steam methane reforming, and only about 4 % of global hydrogen comes from water electrolysis. Combination of electrolytic production of hydrogen from water and supply of renewable energy is attracting more interest due to the sustainability and the increased flexibility of the resulting energy system. The preferred option for intermittent hydrogen storage is pressurization in tanks since at ambient conditions the volumetric energy density of hydrogen is low, and pressurized tanks are efficient and affordable when the cycling rate is high. Pressurized hydrogen enables energy storage in larger capacities compared to battery technologies and additionally the energy can be stored for longer periods of time, on a time scale of months. In this thesis, the thermodynamics and electrochemistry associated with water electrolysis are described. The main water electrolysis technologies are presented with state-of-the-art specifications. Finally, a Power-to-Hydrogen infrastructure design for Lappeenranta University of Technology is presented. Laboratory setup for water electrolysis is specified and factors affecting its commissioning in Finland are presented.

The growing population on earth along with diminishing fossil deposits and the climate change debate calls out for a better utilization of renewable, bio-based materials. In a biorefinery perspective, the renewable biomass is converted into many different products such as fuels, chemicals, and materials, quite similar to the petroleum refinery industry. Since forests cover about one third of the land surface on earth, ligno-cellulosic biomass is the most abundant renewable resource available. The natural first step in a biorefinery is separation and isolation of the different compounds the biomass is comprised of. The major components in wood are cellulose, hemicellulose, and lignin, all of which can be made into various end-products. Today, focus normally lies on utilizing only one component, e.g., the cellulose in the Kraft pulping process. It would be highly desirable to utilize all the different compounds, both from an economical and environmental point of view. The separation process should therefore be optimized. Hemicelluloses can partly be extracted with hot-water prior to pulping. Depending in the severity of the extraction, the hemicelluloses are degraded to various degrees. In order to be able to choose from a variety of different end-products, the hemicelluloses should be as intact as possible after the extraction. The main focus of this work has been on preserving the hemicellulose molar mass throughout the extraction at a high yield by actively controlling the extraction pH at the high temperatures used. Since it has not been possible to measure pH during an extraction due to the high temperatures, the extraction pH has remained a “black box”. Therefore, a high-temperature in-line pH measuring system was developed, validated, and tested for hot-water wood extractions. One crucial step in the measurements is calibration, therefore extensive efforts was put on developing a reliable calibration procedure. Initial extractions with wood showed that the actual extraction pH was ~0.35 pH units higher than previously believed. The measuring system was also equipped with a controller connected to a pump. With this addition it was possible to control the extraction to any desired pH set point. When the pH dropped below the set point, the controller started pumping in alkali and by that the desired set point was maintained very accurately. Analyses of the extracted hemicelluloses showed that less hemicelluloses were extracted at higher pH but with a higher molar-mass. Monomer formation could, at a certain pH level, be completely inhibited. Increasing the temperature, but maintaining a specific pH set point, would speed up the extraction without degrading the molar-mass of the hemicelluloses and thereby intensifying the extraction. The diffusion of the dissolved hemicelluloses from the wood particle is a major part of the extraction process. Therefore, a particle size study ranging from 0.5 mm wood particles to industrial size wood chips was conducted to investigate the internal mass transfer of the hemicelluloses. Unsurprisingly, it showed that hemicelluloses were extracted faster from smaller wood particles than larger although it did not seem to have a substantial effect on the average molar mass of the extracted hemicelluloses. However, smaller particle sizes require more energy to manufacture and thus increases the economic cost. Since bark comprises 10 – 15 % of a tree, it is important to also consider it in a biorefinery concept. Spruce inner and outer bark was hot-water extracted separately to investigate the possibility to isolate the bark hemicelluloses. It was showed that the bark hemicelluloses comprised mostly of pectic material and differed considerably from the wood hemicelluloses. The bark hemicelluloses, or pectins, could be extracted at lower temperatures than the wood hemicelluloses. A chemical characterization, done separately on inner and outer bark, showed that inner bark contained over 10 % stilbene glucosides that could be extracted already at 100 °C with aqueous acetone.
