In this thesis I treat various biophysical questions arising in the context of complexed / ”protein-packed” DNA and DNA in confined geometries (like in viruses or toroidal DNA condensates). Using diverse theoretical methods I consider the statistical mechanics as well as the dynamics of DNA under these conditions. In the first part of the thesis (chapter 2) I derive for the first time the single molecule ”equation of state”, i.e. the force-extension relation of a looped DNA (Eq. 2.94) by using the path integral formalism. Generalizing these results I show that the presence of elastic substructures like loops or deflections caused by anchoring boundary conditions (e.g. at the AFM tip or the mica substrate) gives rise to a significant renormalization of the apparent persistence length as extracted from single molecule experiments (Eqs. 2.39 and 2.98). As I show the experimentally observed apparent persistence length reduction by a factor of 10 or more is naturally explained by this theory. In chapter 3 I theoretically consider the thermal motion of nucleosomes along a DNA template. After an extensive analysis of available experimental data and theoretical modelling of two possible mechanisms I conclude that the ”corkscrew-motion” mechanism most consistently explains this biologically important process. In chapter 4 I demonstrate that DNA-spools (architectures in which DNA circumferentially winds on a cylindrical surface, or onto itself) show a remarkable ”kinetic inertness” that protects them from tension-induced disruption on experimentally and biologically relevant timescales (cf. Fig. 4.1 and Eq. 4.18). I show that the underlying model establishes a connection between the seemingly unrelated and previously unexplained force peaks in single molecule nucleosome and DNA-toroid stretching experiments. Finally in chapter 5 I show that toroidally confined DNA (found in viruses, DNAcondensates or sperm chromatin) undergoes a transition to a twisted, highly entangled state provided that the aspect ratio of the underlying torus crosses a certain critical value (cf. Eq. 5.6 and the phase diagram in Fig. 5.4). The presented mechanism could rationalize several experimental mysteries, ranging from entangled and supercoiled toroids released from virus capsids to the unexpectedly short cholesteric pitch in the (toroidaly wound) sperm chromatin. I propose that the ”topological encapsulation” resulting from our model may have some practical implications for the gene-therapeutic DNA delivery process.

Il processo di riforma a carattere europeo che ha condotto alla nascita di uno Spazio Europeo dell’Istruzione Superiore ha conosciuto diverse tappe fondamentali, fra cui, per esempio, la ripartizione in cicli e l’adozione dei crediti ECTS. Una di queste tappe, ossia il passaggio da una pianificazione della didattica basata sui contenuti ad una basata sui risultati dell’apprendimento, ricopre un ruolo di primo piano nel presente progetto di dottorato. Lo studio qui descritto ha esaminato, da un punto di vista sintattico e semantico, un campione di obiettivi e risultati dell’apprendimento di insegnamenti di alcuni corsi di laurea di primo e secondo ciclo, delle scienze umanistiche, in Austria, Germania, Italia e Regno Unito. L’obiettivo del progetto è proporre uno schema per una classificazione a faccette di obiettivi dell’apprendimento denominato FLOC. Tale schema è adottato per classificare gli obiettivi dell’apprendimento di quattro contesti linguistico-culturali (nei paesi summenzionati), dando vita a FLOC-AT, FLOC-DE, FLOC-IT e FLOC-EN. Queste quattro classificazioni forniscono inoltre il contesto per un’analisi contrastiva multilingue fra unità di obiettivi di apprendimento.

Fatigue life in metals is predicted utilizing regression analysis of large sets of experimental data, thus representing the material’s macroscopic response. Furthermore, a high variability in the short crack growth (SCG) rate has been observed in polycrystalline materials, in which the evolution and distributionof local plasticity is strongly influenced by the microstructure features. The present work serves to (a) identify the relationship between the crack driving force based on the local microstructure in the proximity of the crack-tip and (b) defines the correlation between scatter observed in the SCG rates to variability in the microstructure. A crystal plasticity model based on the fast Fourier transform formulation of the elasto-viscoplastic problem (CP-EVP-FFT) is used, since the ability to account for the both elastic and plastic regime is critical in fatigue. Fatigue is governed by slip irreversibility, resulting in crack growth, which starts to occur during local elasto-plastic transition. To investigate the effects of microstructure variability on the SCG rate, sets of different microstructure realizations are constructed, in which cracks of different length are introduced to mimic quasi-static SCG in engineering alloys. From these results, the behavior of the characteristic variables of different length scale are analyzed: (i) Von Mises stress fields (ii) resolved shear stress/strain in the pertinent slip systems, and (iii) slip accumulation/irreversibilities. Through fatigue indicator parameters (FIP), scatter within the SCG rates is related to variability in the microstructural features; the results demonstrate that this relationship between microstructure variability and uncertainty in fatigue behavior is critical for accurate fatigue life prediction.

Collinear laser spectroscopy has been used as a tool for nuclear physics for more than 30 years. The unique possibility to extract nuclear properties like spins, radii and nuclear moments in a model-independent manner leads to important physics results to test the predictive power of existing nuclear models. rnThis work presents the construction and the commissioning of a new collinear laser spectroscopy experiment TRIGA-LASER as a part of the TRIGA-SPEC facility at the TRIGA research reactor at the University of Mainz. The goal of the experiment is to study the nuclear structure of radioactive isotopes which will be produced by neutron-induced fission near the reactor core and transported to an ion source by a gas jet system. rnThe versatility of the collinear laser spectroscopy technique will be exploited in the second part of this thesis. The nuclear spin and the magnetic moment of the neutron-deficient isotope Mg-21 will be presented, which were measured by the detection of the beta-decay asymmetry induced by nuclear polarization after optical pumping. A combination of this detection method with the classical fluorescence detection is then used to determine the isotope shifts of the neutron-rich magnesium isotopes from Mg-24 through Mg-32 to study the transition to the ''island of inversion''.

Rett's Syndrome (RTT) is a severe neurodevelopmental disorder, characterized by cognitive disability that appears in the first months/years of life. Recently, mutations in the X-linked cyclin-dependent kinase-like 5 (CDKL5) gene have been detected in RTT patients characterized by early-onset seizures. CDKL5 is highly expressed in the brain starting from early postnatal stages to adulthood, suggesting the importance of this kinase for proper brain maturation and function. However, the role/s of CDKL5 in brain development and the molecular mechanisms whereby CDKL5 exerts its effects are still largely unknown. In order to characterize the role of CDKL5 on brain development, we created a mice carrying a targeted conditional knockout allele of Cdkl5. A first behavioral characterization shows that Cdkl5 knockout mice recapitulate several features that mimic the clinical features described in CDKL5 patients and are a useful tool to investigate phenotypic and functional aspects of Cdkl5 loss. We used the Cdkl5 knockout mouse model to dissect the role of CDKL5 on hippocampal development and to establish the mechanism/s underlying its actions. We found that Cdkl5 knockout mice showed increased precursor cell proliferation in the hippocampal dentate gyrus. Interestingly, this region was also characterized by an increased rate of apoptotic cell death that caused a reduction in the final neuron number in spite of the proliferation increase. Moreover, loss of Cdkl5 led to decreased dendritic development of new generated granule cells. Finally, we identified the Akt/GSK3-beta signaling as a target of Cdkl5 in the regulation of neuronal precursor proliferation, survival and maturation. Overall our findings highlight a critical role of CDKL5/AKT/GSK3-beta signaling in the control of neuron proliferation, survival and differentiation and suggest that CDKL5-related alterations of these processes during brain development underlie the neurological symptoms of the CDKL5 variant of RTT.

La tesi tematizza come proprio oggetto di indagine i percorsi di partecipazione politica e civica dei giovani nei contesti di transizione alla vita adulta, concentrandosi sull’influenza delle relazioni tra generazioni su tali espressioni di coinvolgimento. L’approfondimento empirico consiste in una ricerca qualitativa condotta presso il quartiere Navile di Bologna nel 2012. Basandosi sull’approccio metodologico della grounded theory, essa ha coinvolto un campione di giovani e un campione di adulti per loro significativi attraverso interviste semistrutturate. Dall’analisi emerge una rilevante disaffezione giovanile nei confronti della politica che, tuttavia, non traduce in un rifiuto del coinvolgimento, ma in una “partecipazione con riserva” espressa attraverso atteggiamenti tutt’altro che passivi nei confronti della politica formale - basati sulla logica della riforma, della resistenza o della ribellione - e mediante un forte investimento in attività partecipative non convenzionali (associazionismo e coinvolgimento). A fare da sfondo all’interesse partecipativo dei giovani si colloca una lettura negativa della propria condizione presente ed un conseguente conflitto intergenerazionale piuttosto manifesto, che si riflette sulle stesse modalità di attivazione. La politica, nelle sue espressioni più strettamente formali, viene interpretata come un ‘territorio adulto’, gestito secondo logiche che lasciano poco spazio ai giovani i quali, per tale ragione, scelgono di attivarsi secondo modalità alternative in cui il confronto con l’altro, quando presente, avviene prevalentemente tra pari o su basi avvertite come più paritarie. Il distanziamento dei giovani dalla politica formale riflette quindi una parallela presa di distanza dagli adulti, i quali risultano smarriti nello svolgimento delle loro funzioni di modello e di riconoscimento. La loro ambivalenza rispetto ai giovani - ossia il continuo oscillare tra il profondo pessimismo e il cieco ottimismo, tra la guida direttiva e la deresponsabilizzazione - si traduce in un riconoscimento parziale delle reali potenzialità ed esigenze dei giovani come cittadini ed adulti emergenti.

La ricerca si propone di mostrare come il pensiero gramsciano sia stato riferimento prioritario di due intellettuali argentini in esilio in Messico dal 1976 al 1983: Juan Carlos Portantiero e José Maria Aricó. In quel periodo incentrarono le loro elaborazioni teorico-politiche sull’analisi della relazione tra Stato, società civile, democrazia e socialismo, partendo da una prospettiva gramsciana. Il fallimento della guerra di movimento in Argentina nei primi anni settanta li condusse a riflettere su strategie alternative di transizione al socialismo, il cui punto focale fu il concetto di "Egemonia". A partire dal 1975 indirizzarono la ripresa del pensiero di Gramsci alla creazione di un progetto politico adatto ad un contesto sempre più "occidentale", caratterizzato dalla presenza di una "società civile complessa", in cui risultava necessario combattere "guerre di posizione" e non "guerre di movimento". La prospettiva che connotò questo approccio alle riflessioni gramsciane rappresenta il culmine di un percorso che iniziarono negli anni ’50, quando sorsero i primi studi del pensiero gramsciano in Argentina. Sin da allora, Aricó e Portantiero si occuparono di Gramsci insieme al dirigente del PC argentino Agosti e continuarono a farlo anche durante gli anni sessanta e i primi anni settanta sulla rivista Pasado y Presente. Fu, però, nel periodo dell’esilio che ne ripresero il pensiero considerandolo nella sua totalità, a partire dagli scritti giovanili sino ai Quaderni del Carcere, rielaborandolo in maniera originale e costruendo una propria proposta di cammino verso socialismo nell' "occidente periferico" dell'Argentina, influenzati dall'azione del Partito Comunista Italiano.

Nell’ambito della riflessione sui processi di riproduzione e trasmissione intergenerazionale delle disuguaglianze sociali, la presente tesi dottorale propone un’analisi in prospettiva intersezionale dei percorsi verso l’indipendenza abitativa dei giovani bolognesi di origine popolare, e il ruolo svolto dalle loro famiglie in questi percorsi. Nell’approfondimento teorico si offre una ricostruzione del dibattito sociologico nazionale e internazionale sul tema delle disuguaglianze sociali e abitative delle giovani generazioni, nonché dello studio della classe sociale e i principali approcci presenti in letteratura. La ricerca sul campo si concentra, in particolare, sui modi in cui le famiglie appartenenti ad una determinata classe sociale, intesa nel senso conferito al termine da Bourdieu, negoziano e sostengono la transizione all’indipendenza abitativa dei loro figli. L’approfondimento empirico consiste in una ricerca qualitativa longitudinale retrospettiva, condotta nel bolognese nel 2013-2014 su un campione di famiglie working class. Ai giovani-adulti coinvolti e ai loro genitori è stato chiesto di ricostruire le loro biografie attraverso lo strumento delle life histories. La ricerca evidenzia il delinearsi di “micro sistemi economici” familiari specifici e creativi. Le molteplici forme di sostegno genitoriale nei percorsi di autonomizzazione dei figli, individuate nel corso dell’analisi del materiale raccolto, si innestano quindi all’interno di un più ampio sistema di supporto intergenerazionale, che continua anche dopo l’uscita dalla famiglia di origine, a conferma dello stretto legame che caratterizza le famiglie italiane. Nello studio una particolare attenzione viene rivolta alle logiche di legittimazione del sostegno genitoriale, adottate dalle famiglie per orientare i propri interventi di aiuto. Infine, mettendo a confronto la concezione di indipendenza dei giovani adulti con quella dei loro genitori, l’indagine rileva l’esistenza di una apparente contraddizione tra le rappresentazioni dell’indipendenza e dell’autonomia abitativa e i comportamenti attuati nella vita quotidiana, contraddizione che trova una risoluzione nella negoziazione, tra le due diverse generazioni, del concetto stesso di indipendenza.

Il fine principale della tesi è di dimostrare che il concetto di economia verde, pur non avendo natura giuridica, ha un rilievo notevole nel diritto dell’Unione europea, in particolare rispetto al mercato dei servizi. L’indagine concerne innanzitutto lo sviluppo sostenibile, del quale l’economia verde rappresenta uno strumento, per poi incentrarsi sul concetto di economia verde, al fine di ricostruire la sua veste giuridica e la sua collocazione nel diritto (primario) dell’Unione europea. In secondo luogo, si analizza come l’Unione stia utilizzando il proprio diritto per favorire la transizione verso un’economia verde e per stimolare la circolazione dei relativi servizi. Da ultimo, vengono prospettate le varie relazioni che si ritiene possano intercorrere tra l’economia verde (nella sua dimensione giuridica a livello sovranazionale) e la libera prestazione dei servizi.

In the present work, a detailed analysis of a Mediterranean TLC occurred in January 2014 has been conducted. The author is not aware of other studies regarding this particular event at the publication of this thesis. In order to outline the cyclone evolution, observational data, including weather-stations data, satellite data, radar data and photographic evidence, were collected at first. After having identified the cyclone path and its general features, the GLOBO, BOLAM and MOLOCH NWP models, developed at ISAC-CNR (Bologna), were used to simulate the phenomenon. Particular attention was paid on the Mediterranean phase as well as on the Atlantic phase, since the cyclone showed a well defined precursor up to 3 days before the minimum formation in the Alboran Sea. The Mediterranean phase has been studied using different combinations of GLOBO, BOLAM and MOLOCH models, so as to evaluate the best model chain to simulate this kind of phenomena. The BOLAM and MOLOCH models showed the best performance, by adjusting the path erroneously deviated in the National Centre for Environmental Prediction (NCEP) and ECMWF operational models. The analysis of the cyclone thermal phase shown the presence of a deep-warm core structure in many cases, thus confirming the tropical-like nature of the system. Furthermore, the results showed high sensitivity to initial conditions in the whole lifetime of the cyclone, while the Sea Surface Temperature (SST) modification leads only to small changes in the Adriatic phase. The Atlantic phase has been studied using GLOBO and BOLAM model and with the aid of the same methodology already developed. After tracing the precursor, in the form of a low-pressure system, from the American East Coast to Spain, the thermal phase analysis was conducted. The parameters obtained showed evidence of a deep-cold core asymmetric structure during the whole Atlantic phase, while the first contact with the Mediterranean Sea caused a sudden transition to a shallow-warm core structure. The examination of Potential Vorticity (PV) 3-dimensional structure revealed the presence of a PV streamer that individually formed over Greenland and eventually interacted with the low-pressure system over the Spanish coast, favouring the first phase of the cyclone baroclinic intensification. Finally, the development of an automated system that tracks and studies the thermal phase of Mediterranean cyclones has been encouraged. This could lead to the forecast of potential tropical transition, against with a minimum computational investment.

Durch den Straßenbau an der Bundesstraße B3a veranlasst, wurden im Zeitraum vom 13.08.2007 bis zum 07.12.2007 archäologische Untersuchungen im Bereich der Streckenkilometer 19 bis 22 durch die Firma Archbau Essen, unter Kontrolle des Landesamtes für Denkmalpflege Hessen und der Kreisarchäologie Wetteraukreis durchgeführt. Bei km 19 wurde dabei eine Siedlung mit angrenzendem Gräberfeld aus der Linienbandkeramik (im Folgenden als LBK bezeichnet) festgestellt. Daneben ergaben sich eine Bestattung und ein Erdwerk der Michelsberger Kultur, Grubenkomplexe und ein Bronzehort aus der Urnenfelderkultur sowie eine mittelalterliche Straße.Heute liegt die Fundstelle Friedberg B3a km 19 in der südlichen Wetterau 30 km nördlich von Frankfurt am Main in Hessen an der Wetter 140 m über NN und gehört zum Wetteraukreis. rnDie 21 ha messende Ausgrabungsfläche umfasste insgesamt 344 Befunde. Die Identifikation der bandkeramischen Strukturen erfolgte hierbei vorwiegend durch die vergesellschaftete Keramik. Von zentraler Bedeutung sind die fünf Hausgrundrisse A bis E im nördlichen Sektor der Fundstelle, wobei die Präsenz zahlreicher „Dreipfostenriegel“ sowie die Nordost-Südwest Ausrichtung der Strukturen als Belege für eine Zeitstellung innerhalb der LBK angesehen werden. Über die Korrespondenzanalyse der bei den Siedlungsstrukturen angetroffenen Keramik konnte darüber hinaus eine Abfolge von Hausgenerationen erstellt werden. Daneben existierten eine Reihe weiterer Befunde von LBK-zeitlichen Pfostenstellungen, bei denen es sich um Zäune oder Palisaden gehandelt haben könnte. Südwestlich dieser Hausgrundrisse wurde bei den Ausgrabungen eine Grabenstruktur geschnitten, für die eine Funktion als Einfassung der bandkeramischen Siedlungsstrukturen möglich ist. Südlich dieser Grabenstruktur konnten sechs Hockerbestattungen der LBK festgestellt werden. Die räumliche Anordnung im Bereich der westlichen Grabungsgrenze lässt den Schluss zu, dass es sich hierbei um ein Gräberfeld handelt, welches bisher nur zu einem geringen Teil ergraben wurde. Ein Zusammenhang zu den Hausgrundrissen A bis E kann erwogen werden. Der nördliche Sektor der Fundstelle umfasste außerdem zahlreiche Grubenbefunde der LBK sowie einige Grubenkomplexe, wobei letztere als Lehmentnahmegruben anzusprechen sind. Abschließend soll hier noch auf die Existenz zweier bandkeramischer Öfen, darunter ein Grubenofen, im Südosten von Friedberg B3a km 19 hingewiesen werden. rnDen mit Abstand größten Anteil am bandkeramischen Fundmaterial aus Friedberg B3 km 19 hatte die Keramik mit 3428 Elementen. An Steingeräten konnten nur 12 Silices sowie 4 Beile und 14 Mahlsteine über die Typologie und die vergesellschaftete Keramik in die LBK eingeordnet werden. Bei den Knochengeräten zeigte sich ein einzelner beschädigter Kamm. Der Fokus der Analysen des bandkeramischen Fundmaterials aus Friedberg B3a km 19 lag auf der Keramik und deren Auswertung innerhalb von Seriation und Korrespondenzanalyse. Um den Untersuchungen mehr Validität zu verleihen und eine optimale Einordnung zu erreichen, wurden die Keramik-Daten aus Friedberg B3a km 19 mit anderen Datensätzen aus Südhessen kombiniert. Dabei war nicht nur die räumliche Nähe der Fundstellen entscheidend, sondern auch die stilistisch-typologische Nähe der Inventare. In Friedberg B3a km 19 waren die Phasen nach Meier-Arendt von einem frühen III bis zum Ende der Phase V anwesend. Phase II kann nur als marginal angesprochen werden. Es ließ sich kein Übergang zum Mittelneolithikum fassen. Nach der Interpretation aller relativchronologischer Daten könnte es sich in Friedberg B3a km 19 um eine Hausentwicklung mit sieben Phasen à 25 Jahren über 175 bis zu 200 Jahren handeln, die eine kontinuierliche Belegung des Platzes von der mittleren bis zum Ende der jüngsten LBK beschreibt. Insgesamt pflegt sich die Fundstelle Friedberg B3a km 19 so in die bandkeramische Siedlungslandschaft der südlichen Wetterau ein.

Percutaneous needle intervention based on PET/CT images is effective, but exposes the patient to unnecessary radiation due to the increased number of CT scans required. Computer assisted intervention can reduce the number of scans, but requires handling, matching and visualization of two different datasets. While one dataset is used for target definition according to metabolism, the other is used for instrument guidance according to anatomical structures. No navigation systems capable of handling such data and performing PET/CT image-based procedures while following clinically approved protocols for oncologic percutaneous interventions are available. The need for such systems is emphasized in scenarios where the target can be located in different types of tissue such as bone and soft tissue. These two tissues require different clinical protocols for puncturing and may therefore give rise to different problems during the navigated intervention. Studies comparing the performance of navigated needle interventions targeting lesions located in these two types of tissue are not often found in the literature. Hence, this paper presents an optical navigation system for percutaneous needle interventions based on PET/CT images. The system provides viewers for guiding the physician to the target with real-time visualization of PET/CT datasets, and is able to handle targets located in both bone and soft tissue. The navigation system and the required clinical workflow were designed taking into consideration clinical protocols and requirements, and the system is thus operable by a single person, even during transition to the sterile phase. Both the system and the workflow were evaluated in an initial set of experiments simulating 41 lesions (23 located in bone tissue and 18 in soft tissue) in swine cadavers. We also measured and decomposed the overall system error into distinct error sources, which allowed for the identification of particularities involved in the process as well as highlighting the differences between bone and soft tissue punctures. An overall average error of 4.23 mm and 3.07 mm for bone and soft tissue punctures, respectively, demonstrated the feasibility of using this system for such interventions. The proposed system workflow was shown to be effective in separating the preparation from the sterile phase, as well as in keeping the system manageable by a single operator. Among the distinct sources of error, the user error based on the system accuracy (defined as the distance from the planned target to the actual needle tip) appeared to be the most significant. Bone punctures showed higher user error, whereas soft tissue punctures showed higher tissue deformation error.

Arterial hypertension in childhood is less frequent as compared to adulthood but is more likely to be secondary to an underlying disorder. After ruling out more obvious causes, some patients still present with strongly suspected secondary hypertension of yet unknown etiology. A number of these children have hypertension due to single gene mutations inherited in an autosomal dominant or recessive fashion. The finding of abnormal potassium levels (low or high) in the presence of suppressed renin secretion, and metabolic alkalosis or acidosis should prompt consideration of these familial diseases. However, mild hypertension and the absence of electrolyte abnormalities do not exclude hereditary conditions. In monogenic hypertensive disorders, three distinct mechanisms leading to the common final pathway of increased sodium reabsorption, volume expansion, and low plasma renin activity are documented. The first mechanism relates to gain-of-function mutations with a subsequent hyperactivity of renal sodium and chloride reabsorption leading to plasma volume expansion (e.g., Liddle's syndrome, Gordon's syndrome). The second mechanism involves deficiencies of enzymes that regulate adrenal steroid hormone synthesis and deactivation (e.g., subtypes of congenital adrenal hyperplasia, apparent mineralocorticoid excess (AME)). The third mechanism is characterized by excessive aldosterone synthesis that escapes normal regulatory mechanisms and leading to volume-dependent hypertension in the presence of suppressed renin release (glucocorticoid remediable aldosteronism). Hormonal studies coupled with genetic testing can help in the early diagnosis of these disorders.

The protozoan parasite Plasmodium is transmitted by female Anopheles mosquitoes and undergoes obligatory development within a parasitophorous vacuole in hepatocytes before it is released into the bloodstream. The transition to the blood stage was previously shown to involve the packaging of exoerythrocytic merozoites into membrane-surrounded vesicles, called merosomes, which are delivered directly into liver sinusoids. However, it was unclear whether the membrane of these merosomes was derived from the parasite membrane, the parasitophorous vacuole membrane or the host cell membrane. This knowledge is required to determine how phagocytes will be directed against merosomes. Here, we fluorescently label the candidate membranes and use live cell imaging to show that the merosome membrane derives from the host cell membrane. We also demonstrate that proteins in the host cell membrane are lost during merozoite liberation from the parasitophorous vacuole. Immediately after the breakdown of the parasitophorous vacuole membrane, the host cell mitochondria begin to degenerate and protein biosynthesis arrests. The intact host cell plasma membrane surrounding merosomes allows Plasmodium to mask itself from the host immune system and bypass the numerous Kupffer cells on its way into the bloodstream. This represents an effective strategy for evading host defenses before establishing a blood stage infection.

Glucose supply markedly changes during the transition to extrauterine life. In this study, we investigated diet effects on glucose metabolism in neonatal calves. Calves were fed colostrum (C; n = 7) or milk-based formula (F; n = 7) with similar nutrient content up to d 4 of life. Blood plasma samples were taken daily before feeding and 2 h after feeding on d 4 to measure glucose, lactate, nonesterified fatty acids, protein, urea, insulin, glucagon, and cortisol concentrations. On d 2, additional blood samples were taken to measure glucose first-pass uptake (FPU) and turnover by oral [U-(13)C]-glucose and i.v. [6,6-(2)H(2)]-glucose infusion. On d 3, endogenous glucose production and gluconeogenesis were determined by i.v. [U-(13)C]-glucose and oral deuterated water administration after overnight feed deprivation. Liver tissue was obtained 2 h after feeding on d 4 and glycogen concentration and activities and mRNA abundance of gluconeogenic enzymes were measured. Plasma glucose and protein concentrations and hepatic glycogen concentration were higher (P < 0.05), whereas plasma urea, glucagon, and cortisol (d 2) concentrations as well as hepatic pyruvate carboxylase mRNA level and activity were lower (P < 0.05) in group C than in group F. Orally administered [U-(13)C]-glucose in blood was higher (P < 0.05) but FPU tended to be lower (P < 0.1) in group C than in group F. The improved glucose status in group C resulted from enhanced oral glucose absorption. Metabolic and endocrine changes pointed to elevated amino acid degradation in group F, presumably to provide substrates to meet energy requirements and to compensate for impaired oral glucose uptake.
