During the last years, the emission of heavy metals to the environment has increased, causing a severe negative impact to the ecosystems and seriously compromising human health due to their mutagenic potential. Tri- (III) and hexavalent (VI) chromium (Cr) constitute the oxidative states of the metal chromium that are active in living organisms. These two oxidation states of the chromium differ with regards to their cellular effects, mainly due to the different abilities they possess in relation to easy of transport through biological membranes. Cr VI is transported into the cell through transference channels of endogenous anions that are isostructural and isoelectronical to Cr VI, such as SO 4 -2 and HPO 4 -2. On the other hand, Cr III is unable to diffuse through the cell membrane. Its existence inside the cells is generally due to the reduction of Cr VI, the endocytosis, or the absortion by the cells via phagocytosis. Cr III acts directly on the DNA molecule, while Cr VI reacts little with this molecule. In the ecosystem, however, Cr VI is more dangerous since this is the form that presents greater reactivity with biological membranes, crossing them and being easily incorporated into the cell. In the cell it is biotransformed to Cr III, a potentially mutagenic molecule. In vivo and in vitro studies have shown that organisms exposed to Cr VI present greater induction to a variety of damages to the DNA molecule. Among the damages induced by Cr, changes in the structure of the DNA molecule have been reported, with breaks of the major chain and base oxidation. In the organisms, these alterations generate chromosomal aberrations, micronucleus formation, sister chromatid exchanges, and errors in DNA synthesis.

Agaricus blazei Murrill, popularly known as the sun mushroom, is a native mushroom in SP, Brazil, that has been widely used in the treatment of cancer and many other pathologies in different parts of the world. A water-soluble protein-polysaccharide complex (1 → 6)β-D-glucan has been isolated from its fruiting body that showed immune-modulation activity. From organic extracts, linoleic acid has been isolated and determined to be the main substance with antimutagenic activity. Using both the micronucleus (MN) and comet (single cell microgel electrophoresis) assays, this study determined the genotoxic and antigenotoxic potential of A. blazei (AB) obtained from commercial sources or the following strains: a) strains AB 97/29 (young and sporulated phases); b) a mixture taken from AB 96/07, AB 96/09 and AB 97/ 11 strains; and c) commercial mushrooms from Londrina, PR and Piedade, SP, designated as AB PR and AB SP, respectively. The extracts from these mushrooms were isolated in chloroform:methanol (3:1) and used in vitro at three different concentrations. V79 cells (Chinese hamster lung cells) were exposed to the extracts under pre-, simultaneous and post-treatment conditions, combined with methyl methanesulfonate (MMS). Under the circumstances of this study, these organic extracts did not show any genotoxic or mutagenic effects, but did protect cells against the induction of micronuclei by MMS. The biotechnology, biochemical characterization, and protective effects of the himematsutake and shiitake mushrooms were studied for the Thematic Project, from 1998 to 2003. A new species, Agaricus Brasiliens is Wasser et al. (= A. blazei Murrill ss. Heinem.), was proposed for the cultivated lineages in Brazil. Interactions among lineages, substrates, casing layers, and fructification inductions (temperature alternations) significantly increased the productivity of A. brasiliensis in Brazil (from 40 g to 200 g of fresh mushroom kg -1 moist compost). However, pests and diseases (mainly Sciaridae flies and false truffle), drastically reduced the productivity of A. brasiliensis (below 50 g kg -1). Biochemically for each mushroom species, the polar extracts, no polars, and medium polarity presented the same organic substances; however, their concentrations differed as a result of the lineages, maturation of the fruit bodies, and cultivation conditions. In this aspect, concentration of linoleic acid in A. brasiliensis (added to animal food) was related directly to the chemical protection against carcinogenic drugs in mice. Aqueous extracts of Lentinus edodes (= Lentinula edodes (Berk.) Pegler) and A. brasiliensis may be preventive chemical protectors against mutagenic and carcinogenic drugs, depending on the lineage and extraction method (tea or juice). However, immunomodulator effects and tumor reduction were only observed with concentrated fractions (hexanic, methanolic, and ATF extracts). Aqueous extracts of A. brasiliensis and L. edodes have antibiotic-like substances and substances able to act as elicitors of resistance responses in some plants (local and systemic) and show a potential to be used in the alternative control of plant pathogens. © 2005 by Begell House, Inc.
Antimicrobial activity of coffee-based solutions and their effects on Streptococcus mutans adherence

The aim of this study was to evaluate the antimicrobial activity of different coffee solutions and their effects on the adherence of Streptococcus mutans to glass surface. Coffee solutions were prepared with three commercial products (Pilao, Mellita and Café do Ponto) by two different methods (simple and boiled) (n=15). A control group was also included in the study. For antimicrobial activity testing, tubes containing coffee solution and culture medium were inoculated with a suspension of S. mutans ATCC 35688 and incubated for 1 min 1h, 2h and 4h. Serial dilutions and plating on BHI agar were performed. S, mutans adherence to glass in presence of different coffee solutions was also tested. The number of adhered bacteria (CFU/mL) was determined by plating method. The results were statistically analyzed by ANOVA and Turkey's test. The tested coffee solutions did not reduce the number of colony forming units of S. mutans in relation to the control at all evaluation periods. All the solutions reduced significantly the adherence of S. mutans to the glass surface in relation to control. The tested coffee solutions did not present any antimicrobial effect on Streptococcus mutans, however, all the coffee solutions reduced significantly the adherence of S mutans to the glass surface.

The Brazilian National Institute for Space Research (INPE) is operating the Brazilian Environmental Data Collection System that currently amounts to a user community of around 100 organizations and more than 700 data collection platforms installed in Brazil. This system uses the SCD-1, SCD-2, and CBERS-2 low Earth orbit satellites to accomplish the data collection services. The main system applications are hydrology, meteorology, oceanography, water quality, and others. One of the functionalities offered by this system is the geographic localization of the data collection platforms by using Doppler shifts and a batch estimator based on least-squares technique. There is a growing demand to improve the quality of the geographical location of data collection platforms for animal tracking. This work presents an evaluation of the ionospheric and tropospheric effects on the Brazilian Environmental Data Collection System transmitter geographic location. Some models of the ionosphere and troposphere are presented to simulate their impacts and to evaluate performance of the platform location algorithm. The results of the Doppler shift measurements, using the SCD-2 satellite and the data collection platform (DCP) located in Cuiabá town, are presented and discussed.

Different thermal treatments for the synthesis of BaTiO3 powder obtained through the Pechini method were studied. The synthesis of BaTiO3 starts at 150 °C by the thermal dehydration of organic precursors. The usual inevitable formation of barium carbonate during the thermal decomposition of the precursor could be retarded at lower calcination temperatures and optimized heating rates. The organic precursors were treated at temperatures between 200 and 400 °C. Then, the samples were calcined at 700 and 800 °C for 4 and 2 h, respectively. The resulting ceramic powders were characterized by gravimetric and differential thermal analyses, X-ray powder diffraction and infrared spectroscopy. It was found that depending on the heating rate and final temperature of the thermal treatment, high amounts of BaCO3 and TiO2 could be present due to the high concentration of organics in the final calcination step. © 2007 Elsevier B.V. Cytogenetic and random amplified polymorphic DNA analyses carried out in the species Leptodactylus podicipinus, L. ocellatus, L. labyrinthicus, and L. fuscus from rural and urban habitats of the northwest region of São Paulo State, Brazil, showed that the karyotypes (2n = 22), constitutive heterochromatin distribution and nucleolus organizer region (NOR) location did not differ between the populations from the two environments. The in situ hybridization with an rDNA probe confirmed the location of the NORs on chromosome 8 revealing an in tandem duplication of that region in one of the chromosomes of L. fuscus. DAPI showed that part of the C-band-positive heterochromatin is rich in AT, including that in the proximity the NORs in L. podicipinus and L. ocellatus. The molecular analyses showed that the two populations (urban and rural) of L. podicipinus and L. fuscus are similar from a genetic point of view. The urban and rural populations of species L. ocellatus and L. labyrinthicus showed differences in genetic structures, probably due to urbanization which interferes with the dispersion of those frogs. The marked differences observed between the two populations of L. ocellatus can be representing the cryptic condition of the species. Unweighted pair-group method of analysis and genetic distance analysis detected the genetic proximity between L. ocellatus and L. fuscus. The results indicate that there was no reduction in the genetic diversity in the populations from the urban environment; however, the survival of these frogs would not be guaranteed in the case of an increase in human impact especially for populations of L. labyrinthicus and L. ocellatus. ©FUNPEC-RP.

Cooperative energy-transfer upconversion luminescence in Tb 3+/Yb 3+-codoped PbGeO 3-PbF 2-CdF 2 vitroceramic and its precursor glass under resonant and off-resonance infrared excitation, is investigated. Bright UV-visible emission signals around 384, 415, 438 nm, and 473-490, 545, 587, and 623 nm, identified as due to the 5D 3( 5G 6 → 7F J(J=6,5,4) and 5D 4 → 7F J(J=6,5,4,3) transitions, respectively, were readily observed. The results indicate that cooperative energy-transfer between ytterbium and terbium ions followed by excited-state absorption are the dominant upconversion excitation mechanisms herein involved. The comparison of the upconversion process in a vitroceramic sample and its glassy precursor revealed that the former present much higher upconversion efficiency. The dependence of the upconversion emission upon pump power, temperature, and doping content is also examined.

To address daily fluctuations in electricity demands, the quantities of water passing through the turbines of hydropower plants can vary significantly (up to fourfold) during a 24-h cycle. This study evaluates the effects of hourly variations in water discharges on the limnological conditions observed in two below-dam river stretches. The study reservoirs, Capivara and Taquaruçu, are the 9th and 10th reservoirs in a cascade of dams in the Paranapanema River in south-east Brazil. The reservoirs exhibit different trophic conditions, water retention times, thermal regimes and spillway positions. Capivara Reservoir is deeper, meso-eutrophic, with a high water retention time and hypolimnetic discharges (32 m) varying between 500 and 1400 m3 s-1. In contrast, Taquaruçu Reservoir is relatively shallow, oligo-mesotrophic, and has a low retention time, with water discharges varying between 500 and 2000 m3 s-1. Its turbine water intake zone also is more superficial (7 m). For two periods of the year, winter and summer, profiles of limnological measurements were developed in the lacustrine (above-dam site) zones of the reservoirs, as well as in the downstream river stretches (below-dam site). In both cases, the sampling was carried out at 4-h intervals over a complete nictemeral cycle. The results demonstrated that the reservoir operating regime (water discharge variations) promoted significant differences in the conditions of the river below the dams, especially for water velocity, turbidity, and nutrient and suspended solids concentrations. The reservoir physical characteristics, including depth, thermal stratification and outlet structure, are also key factors influencing the limnology and water quality at the below-dam sampling sites. In the case of Capivara Reservoir, for example, the low dissolved oxygen concentration (<5.0 mg L-1) in its bottom water layer was transferred to the downstream river stretches during the summer. These study results demonstrated that it is important to continue such investigations as a means of verifying whether or not these high-amplitude/low-frequency variations could negatively affect the downstream river biota. © 2009 The Authors. The current study compared the toxicity of different concentrations of boric acid in adult workers of Atta sexdens rubropilosa Forel (Hymenoptera: Formicidae), with toxicological bioassays, and examining the dose-dependent and time-dependent histopathological changes, of the midgut, Malpighian tubules, and postpharyngeal glands. Our results revealed the importance of conducting toxicological bioassays combined with morphological analyses of the organs of ants chronically exposed to insecticides used in commercial ant baits. In vitro bioassays showed that boric acid significantly decreases the survivorship of workers regardless of concentration, whereas the morphological data suggested progressive dose-dependent and time-dependent changes in the organs examined, which were evident in the midgut. The midgut is the first organ to be affected, followed by the postpharyngeal gland and Malpighian tubules. This sequence is in agreement with the absorption pathway of this chemical compound in the midgut, its transference to the hemolymph, possibly reaching the postpharyngeal glands, and excretion by the Malpighian tubules. These progressive changes might be due to the cumulative and delayed effect of boric acid. Our findings provide important information for the understanding of the action of boric acid in ant baits in direct and indirect target organs. © 2010 Entomological Society of America.

Thyroid hormone receptors (TRs) are ligand-gated transcription factors with critical roles in development and metabolism. Although x-ray structures of TR ligand-binding domains (LBDs) with agonists are available, comparable structures without ligand (apo-TR) or with antagonists are not. It remains important to understand apo-LBD conformation and the way that it rearranges with ligands to develop better TR pharmaceuticals. In this study, we conducted hydrogen/deuterium exchange on TR LBDs with or without agonist (T 3) or antagonist (NH3). Both ligands reduce deuterium incorporation into LBD amide hydrogens, implying tighter overall folding of the domain. As predicted, mass spectroscopic analysis of individual proteolytic peptides after hydrogen/ deuterium exchange reveals that ligand increases the degree of solvent protection of regions close to the buried ligand-binding pocket. However, there is also extensive ligand protection of other regions, including the dimer surface at H10-H11, providing evidence for allosteric communication between the ligand-binding pocket and distant interaction surfaces. Surprisingly, Cterminal activation helix H12, which is known to alter position with ligand, remains relatively protected from solvent in all conditions suggesting that it is packed against the LBD irrespective of the presence or type of ligand. T 3, but not NH3, increases accessibility of the upper part of H3-H5 to solvent, and we propose that TR H12 interacts with this region in apo-TR and that this interaction is blocked by T 3 but not NH3.Wepresent data from site-directed mutagenesis experiments and molecular dynamics simulations that lend support to this structural model of apo-TR and its ligand-dependent conformational changes. Structural Health Monitoring (SHM) denotes a system with the ability to detect and interpret adverse changes in a structure. One of the critical challenges for practical implementation of SHM system is the ability to detect damage under changing environmental conditions. This paper aims to characterize the temperature, load and damage effects in the sensor measurements obtained with piezoelectric transducer (PZT) patches. Data sets are collected on thin aluminum specimens under different environmental conditions and artificially induced damage states. The fuzzy clustering algorithm is used to organize the sensor measurements into a set of clusters, which can attribute the variation in sensor data due to temperature, load or any induced damage.

Some wild species of the genus Arachis have demonstrated potential for improvement of peanuts. This work was performed to evaluate the occurrence and symptoms of Enneothrips flavens and Stegasta bosquella and its effects on agronomic traits of wild Arachis accessions. Nine accessions of wild Arachis species and a commercial A. hypogaea variety were studied in a split plot statistical scheme with a completely randomized block design and four replications. The main plots consisted of plants sprayed or not sprayed for insect control, while the subplots comprised the peanut accessions. Accessions GKP10017 (A. cardenasii) and V7639 (A. kuhlmannii) showed the lowest percentages of leaflets with E. flavens and S. bosquella. Accessions V9912, V7639 and V8979 (all three A. kuhlmannii) and V13250 (A. kempff-mercadoi) showed the lowest responses to insecticide application among the various plant traits evaluated. These accessions are of interest for further studies to identify mechanisms of resistance, to be used in breeding programs for resistance to these insects.
