The science and technology interact with the art in several ways. Biotechnological coupled with analytical approaches can play an important role in protecting and preserving cultural heritage for future generations. Many microorganisms influenced by environmental conditions are the main responsible for biological contamination in built heritage. Biocides based on chemical compounds have been used to mitigate this problem. Thus, it is vitally important to develop proper remediation actions based on environmentally innocuous alternative. Bacillus specie is emerging as an optimistic alternative for built heritage treatment due to their capacity to produce secondary metabolites with antagonistic activities against many fungal pathogens. Therefore, the intent of this work was to access a rapid evaluation of antifungal potential of bioactive metabolites produced by Bacillus strains and simultaneously their characterization using spectroscopic (NMR) and chromatographic techniques (LCESI- MS). The high antifungal activity obtained for Bacillus sp. active compounds produced in this study confirms the great potential to suppress biodeteriogenic fungi growth on historical artworks. Additionally, the proposed methodology allowed to access bioactive metabolites produced without need of the laborious total previous isolation and could be used as a viable alternative to be employed for screening and production of new green biocides.

The damaging of buildings and monuments by biological contamination is a cause of serious concern. Biocides based on chemical toxic compounds have been used to mitigate this problem. However, in the past decade many of the most effective biocides have been banned due to their environmental and health hazards. Therefore, proper remediation actions for microbiologically contaminated historic materials based on environmentally safe solution is of vital importance. Bacillus species are emerging as a promising alternative for built heritage treatment. They produce a great diversity of secondary metabolites with biological activity, well known to possess antagonistic activities against many fungal pathogens. In order to evaluate the antifungal activity of the novel biocides produced in our laboratory by cultures of selected bacterial strains, liquid interaction assays using four biodeteriogenic fungi were achieved, revealing a nearly 100% of inhibitory capacity to fungal proliferation. To confirm their effective safe toxicological properties, in vivo tests using two different biological models were performed. The lyophilized supernatant of the Bacillus culture broth showed no lethality against brine shrimp and also no toxicological effects in Swiss mice through administration of acute dose of 5000 mg/kg by oral gavage. In fact, the bioactive compounds were no lethal at the tested dose unlike Preventol® (commercial biocide) that induced acute toxicity with 10 times minor concentration dose administrated in the same conditions. Therefore, the new bioactive compounds that suppress growth of biodeteriogenic fungi on historical artworks, presenting at the same time no toxicity against other living organisms, constituting an efficient and green safe solution for biodegradation/biodeterioration treatment of Cultural Heritage.

A remediação de locais contaminados com metais pesados usando plantas hiperacumuladoras aparenta ser uma alternativa bastante viável. Neste trabalho comparou-se a acumulação e tolerância ao cádmio (Cd), ambas baseadas nas respostas ao stress oxidativo em três espécies de plantas diferentes: Brassica juncea (L.) Czem., Nicotiana tabacum L. e Solanum nigrum L., descritas na literatura como plantas bastante tolerantes ou até com características híper acumuladoras. As plantas cresceram num solo contaminado com diferentes concentrações de Cd (O- 35 mg kg-1) durante um período de 90 dias. O factor de translocação (FT), utilizado para medir a translocação efectiva do Cd da raiz para a parte aérea, variou consideravelmente entre as espécies desenvolvidas. A N. tabacum foi a planta que apresentou os maiores valores de FT. Neste trabalho foi a única planta que preencheu todas as condições para ser considerada hiperacumuladora para todos os níveis de contaminação do solo. Por outro lado, a S. nigrum apresentou os maiores valores de concentração de Cd nos tecidos, com um FT > 1, na presença de 5 mg Cd kg·1 de solo. Apesar da B. juncea ter apresentado um resultado de FT inferior às restantes, foi a única planta com valores crescentes de FT com o aumento da contaminação de Cd. O stress oxidativo nas plantas desenvolvidas foi avaliado pela peroxidação lipídica e pelas actividades da catalase (CAT), ascorbato peroxidase (APX), guaiacol peroxidase (GPX) e superóxido dismutase (SOO), quer na raiz quer na parte aérea. Foi observado um aumento significativo (versus controlo) na peroxidação lipídica e actividade enzimática da CATe APX na parte aérea da B. juncea, N. tabacum e S. nigrum para os níveis de contaminação mais elevados, 15 e/ou 35 mg Cd kg-1 A B. juncea apresentou maior sensibilidade na resposta da GPX, para todas as concentrações de Cd no solo. A peroxidação lipídica e a actividade da CAT foram superiores na parte aérea em relação à raiz para todas as plantas em todas as contaminações de Cd presentes no solo. A actividade da SOO não apresentou respostas consistentes para nenhuma das plantas. ABSTRACT: Remediation of sites contaminated with heavy metals using hyper accumulators seems a promising alternative to engineering approaches. ln this work, we compared cadmium (Cd) accumulation and tolerance (based on responses to oxidative stress) in three different species, Brassica juncea (L) Czem., Nicotiana tabacum L. and Solanum nigrum L., described in the literature as very tolerant or even as hyper accumulators. The plants were grown in soil spiked with different Cd concentrations (O- 35 mg kg- 1) over a period of 90 days. The translocation factor (TF), used to measure the effectiveness of translocating Cd from roots to shoots, depended greatly on the species. N. tabacum was the plant which exhibited the highest TF values. lt was the only plant under study that fulfilled the conditions of a hyper accumulator for all levels of soil contamination. On the other hand, S. nigrum presented the highest Cd concentration in plant tissues, with TF > 1 in the presence of 5 mg Cd kg-1 of soil. Although B. juncea had presented the lowest TF and Cd concentrations, it was the only plant with TF values increasing with the level of cadmium. Oxidative stress in plants was evaluated by lipid peroxidation and activities of catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (GPX) and superoxide dismutase (SOO), both in roots and shoots. A significant enhancement (versus control) on lipid peroxidation and enzymatic activity of CAT and APX in shoots of B. juncea, N. tabacum and S. nigrum was observed for the highest levels of Cd in soil, 15 and/or 35 mg Cd kg-1. B. juncea presented the most sensitive response of GPX, for all levels of Cd in soil. Lipid peroxidation and CAT activity were greater in shoots than in roots for all plants and soil Cd concentrations. SOO activity did not present consistent trends for any plant.

O presente trabalho teve como objectivo a implementação de estratégias de remediação de duas minas abandonadas na região do Alto Alentejo, a mina da Tinoca e a mina da Mostardeira. Neste trabalho foram realizados ensaios de fitoremediação com as espécies Brassica juncea (L.) Czern, Solanum nigrum L., Piptatherum miliaceum (L.)Coss. e Spergularia purpurea (Persoon)G. Don fil. As misturas utilizadas como suporte para desenvolvimento das plantas resultaram da mistura (1:1) de resíduos de escombreira e solo. Estas misturas foram realizadas para as duas minas independentemente. A estas misturas foram adicionados correctivos minerais e um correctivo orgânico. Dos estudos realizados observou-se que medidas de atenuação/diluição não se mostraram suficientemente eficazes para permitir uma estabilização da zona superficial em causa, pelas espécies de plantas estudadas. Por outro lado, a adição de um correctivo orgânico, juntamente com a subida do pH do solo, conduziu a um melhoramento das características físicas e químicas das misturas originais facilitando a sua revegetação. ABSTRACT: This study was designed to implement remediation strategies in two abandoned mines in the region of Alto Alentejo, the Tinoca mine and Mostradeira mine. Phytoremediation tests were carried out with the species Brassica juncea (L.) Czern, Solanum nigrum L., Piptatherum miliaceum (L.) Coss. and Spergularia purpurea (Persoon) G. Don fil. The mixtures used to support plant growth resulted from the admixture (1:1) of waste tailings and soil and were performed independently for the two mines. Soil mineral correctives and organic fertilizers were added to these mixtures. The study showed that mitigation/dilution measures were not sufficiently effective to allow the stabilization of the area with the plant species studied. However, the addition of an organic corrective, along with the rise of soil pH, led to an improvement in the physical and chemical properties of the original amendment soil mixtures, hence facilitating its phytorestoration.

Laboratory incubation experiments were carried out to estimate the mineralisation of metalaxyl 14C {N-(2-6 dimethyphenyl)-N-(methoxyacetyl) alanine methyl ester} in four Brazilian soils with different physico-chemical properties, at 3 and 30 ?g a.i. g-1. In the Petrolina sandy soil the mineralisation presented higher 14CO2 production rates, at two essayed concentrations, after 70 days. Microbiological studies were done to determine the numbers of bacteria, actinobacteria and fungi (CFU g-1 soil). In relation with other microbial community, bacterial population demonstrated to be a major component of the cultivable heterotrophic community after the application.of the compound. No detectable metabolites were found in this study. The results suggest that soil properties and application history may have a strong influence on the fungicide behavior in these soil samples.

Phytoremediation studies have been conducted in an area contaminated by heavy metals, located in Piracicaba - SP, Brazil. This area was contaminated accidentally by the addition of auto scrap shredding to the soil and was limed later to reduce heavy metal mobility in the environment. Previous characterization showed that it also presents high concentration of boron, which has limited the initial plant development of some species. As sunflower plants require a high boron supply and the literature describes its use in the phytoremediation of soils contaminated with heavy metals under some conditions, the aim of this work was to evaluate its potential for the remediation of this area. In the present study, the results of preliminary tests are presented, aiming at the evaluation of sunflower plant germination and its initial development when cultivated in the contaminated soil described. Two sunflower hybrids were sown in soils treated with different rates of boron and in the soil from the contaminated area in study. The results showed that sunflower plants had a normal initial development, even in the soil from the contaminated area. Therefore, sunflower is a promising crop and further studies will be developed to evaluate the sunflower efficiency in phytoextraction or phytostabilization of heavy metals in areas where boron contamination also occurs, as is the case in the study area.

Pseudomonas may use as bioremediator and as biopesticide. The use of soil enzymatic assays as biological indicator of possible negative effects in soil functioning was evaluated after P.putida AF7 inoculation. For that, AF7 was originally isolated from the rizosphere of rice and was inoculated on three soils: Rhodic Hapludox (RH), Typic Hapludox (TH); and Arenic Hapludult (AH). Soil characteristics were measured in each plot. Acid phosphatase, ?-glucosidase and protease activities were measured at 7, 14 and 21 days. The enzyme activity waved during the experimental period but there is a significant reduction of ?-glucosidase activity in RH soil on day 14. Corg was positively correlated to the activities of ?-glucosidase and protease. The presented data indicate that soil biochemical properties may be useful as indicator of soil perturbations.

In the semi-arid region of northeastern Brazil, the Atriplex plant genus has been efficient in removing salts from soils irrigated with saline wastewater. However, this removal might not be significant compared with the amount of salts added to the soil by the wastewater irrigation. Considering this aspect, the aim of this work was to evaluate the effectiveness of Atriplex nummularia Lindl plants in the remediation of a soil submitted to saline wastewater irrigation. Despite the known inhibition effect of saline wastewater on soil enzyme activity, the cultivation of Atriplex nummularia Lindl maintained the treated soil enzyme activity levels similar to the ones found in natural soils.
