NACP, a 140-amino acid presynaptic protein, is the precursor of NAC [the non-amyloid beta/A4 protein (A beta) component of Alzheimer disease (AD) amyloid], a peptide isolated from and immunologically localized to brain amyloid of patients afflicted with AD. NACP produced in Escherichia coli bound to A beta peptides, the major component of AD amyloid. NACP bound to A beta 1-38 and A beta 25-35 immobilized on nitrocellulose but did not bind to A beta 1-28 on the filter under the same conditions. NACP binding to A beta 1-38 was abolished by addition of A beta 25-35 but not by A beta 1-28, suggesting that the hydrophobic region of the A beta peptide is critical to this binding. NACP-112, a shorter splice variant of NACP containing the NAC sequence, bound to A beta, but NACP delta, a deletion mutant of NACP lacking the NAC domain, did not bind A beta 1-38. Furthermore, binding between NACP-112 and A beta 1-38 was decreased by addition of peptide Y, a peptide that covers the last 15 residues of NAC. In an aqueous solution, A beta 1-38 aggregation was observed when NACP was also present in an incubation mixture at a ratio of 1:125 (NACP/A beta), whereas A beta 1-38 alone or NACP alone did not aggregate under the same conditions, suggesting that the formation of a complex between A beta and NACP may promote aggregation of A beta. Thus, NACP can bind A beta peptides through the specific sequence and can promote A beta aggregation, raising the possibility that NACP may play a role in the development of AD amyloid.

Stimulation of muscarinic m1 or m3 receptors can, by generating diacylglycerol and activating protein kinase C, accelerate the breakdown of the amyloid precursor protein (APP) to form soluble, nonamyloidogenic derivatives (APPs), as previously shown. This relationship has been demonstrated in human glioma and neuroblastoma cells, as well as in transfected human embryonic kidney 293 cells and PC-12 cells. We now provide evidence that stimulation of metabotropic glutamate receptors (mGluRs), which also are coupled to phosphatidylinositol 4,5-bisphosphate hydrolysis, similarly accelerates processing of APP into nonamyloidogenic APPs. This process is demonstrated both in hippocampal neurons derived from fetal rats and in human embryonic kidney 293 cells transfected with cDNA expression constructs encoding the mGluR 1 alpha subtype. In hippocampal neurons, both an mGluR antagonist, L-(+)-2-amino-3-phosphonopropionic acid, and an inhibitor of protein kinase C, GF 109203X, blocked the APPs release evoked by glutamate receptor stimulation. Ionotropic glutamate agonists, N-methyl-D-aspartate or S(-)-5-fluorowillardiine, failed to affect APPs release. These data show that selective mGluR agonists that initiate signal-transduction events can regulate APP processing in bona fide primary neurons and transfected cells. As glutamatergic neurons in the cortex and hippocampus are damaged in Alzheimer disease, amyloid production in these regions may be enhanced by deficits in glutamatergic neurotransmission.

Nuclear-encoded proteins targeted to the chloroplast are typically synthesized with N-terminal transit peptides which are proteolytically removed upon import. Structurally related proteins of 145 and 143 kDa copurify with a soluble chloroplast processing enzyme (CPE) that cleaves the precursor for the major light-harvesting chlorophyll a/b binding protein and have been implicated in the maturation of the small subunit of ribulose-1,5-bisphosphate carboxylase/oxygenase and acyl carrier protein. The 145- and 143-kDa proteins have not been found as a heterodimer and thus may represent functionally independent isoforms encoded by separate genes. Here we describe the primary structure of a 140-kDa polypeptide encoded by cDNAs isolated by using antibodies raised against the 145/143-kDa doublet. The 140-kDa polypeptide contains a transit peptide, and strikingly, a His-Xaa-Xaa-Glu-His zinc-binding motif that is conserved in a recently recognized family of metalloendopeptidases, which includes Escherichia coli protease III, insulin-degrading enzyme, and subunit beta of the mitochondrial processing peptidase. Identity of 25-30%, concentrated near the N terminus of the 140-kDa polypeptide, is found with these proteases. Expression of CPE in leaves is not light dependent. Indeed, transcripts are present in dark-grown plants, and the 145/143-kDa doublet and proteolytic activity are both found in etioplasts, as well as in root plastids. Thus, CPE appears to be a necessary component of the import machinery in photosynthetic and nonphotosynthetic tissues, and it may function as a general stromal processing peptidase in plastids.

The C4 repressor of the temperate bacteriophages P1 and P7 inhibits antirepressor (Ant) synthesis and is essential for establishment and maintenance of lysogeny. C4 is an antisense RNA acting on a target, Ant mRNA, which is transcribed from the same promoter. The antisense-target RNA interaction requires processing of C4 RNA from a precursor RNA. Here we show that 5' maturation of C4 RNA in vivo depends on RNase P. In vitro, Escherichia coli RNase P and its catalytic RNA subunit (M1 RNA) can generate the mature 5' end of C4 RNA from P1 by a single endonucleolytic cut, whereas RNase P from the E. coli rnpA49 mutant, carrying a missense mutation in the RNase P protein subunit, is defective in the 5' maturation of C4 RNA. Primer extension analysis of RNA transcribed in vivo from a plasmid carrying the P1 c4 gene revealed that 5'-mature C4 RNA was the predominant species in rnpA+ bacteria, whereas virtually no mature C4 RNA was found in the temperature-sensitive rnpA49 strain at the restrictive temperature. Instead, C4 RNA molecules carrying up to five extra nucleotides beyond the 5' end accumulated. The same phenotype was observed in rnpA+ bacteria which harbored a plasmid carrying a P7 c4 mutant gene with a single C-->G base substitution in the structural homologue to the CCA 3' end of tRNAs. Implications of C4 RNA processing for the lysis/lysogeny decision process of bacteriophages P1 and P7 are discussed.

The beta-amyloid precursor protein (beta-APP), from which the beta-A4 peptide is derived, is considered to be central to the pathogenesis of Alzheimer disease (AD). Transgenic mice expressing the 751-amino acid isoform of human beta-APP (beta-APP751) have been shown to develop early AD-like histopathology with diffuse deposits of beta-A4 and aberrant tau protein expression in the brain, particularly in the hippocampus, cortex, and amygdala. We now report that beta-APP751 transgenic mice exhibit age-dependent deficits in spatial learning in a water-maze task and in spontaneous alternation in a Y maze. These deficits were mild or absent in 6-month-old transgenic mice but were severe in 12-month-old transgenic mice compared to age-matched wild-type control mice. No other behavioral abnormalities were observed. These mice therefore model the progressive learning and memory impairment that is a cardinal feature of AD. These results provide evidence for a relationship between abnormal expression of beta-APP and cognitive impairments.

Cells from transgenic mice expressing a human mini-gene for collagen I were used as markers to follow the fate of mesenchymal precursor cells from marrow that were partially enriched by adherence to plastic, expanded in culture, and then injected into irradiated mice. Sensitive PCR assays for the marker collagen I gene indicated that few of the donor cells were present in the recipient mice after 1 week, but 1-5 months later, the donor cells accounted for 1.5-12% of the cells in bone, cartilage, and lung in addition to marrow and spleen. A PCR in situ assay on lung indicated that the donor cells diffusely populated the parenchyma, and reverse transcription-PCR assays indicated that the marker collagen I gene was expressed in a tissue-specific manner. The results, therefore, demonstrated that mesenchymal precursor cells from marrow that are expanded in culture can serve as long-lasting precursors for mesenchymal cells in bone, cartilage, and lung. They suggest that cells may be particularly attractive targets for gene therapy ex vivo.

Hyaluronan (HA) plays an important role in lung pathophysiology. For this reason it has attracted great attention both as active ingredient and as excipient in treating lung diseases by direct pulmonary HA administration. The aim was the production of highly respirable and flowable HA powders either as a potential carrier for drug delivery or for being delivered directly by inhalation. Engineered sodium hyaluronate powders were produced by spray-drying technique. All the spray-dried powders were characterised in terms of particle size distribution, drug content, morphology and in vitro respirability. HA was successfully formulated with salbutamol sulphate in combination with leucine and highlighted remarkable aerodynamic performance (emitted dose equal to 83 % and FPF % equal to 97.1%). Moreover, HA colloidal solutions were designed and they were spray-dried. In order to improve particle aerodynamic characteristics, different types of excipients were investigated. In particular, stearylamine (5% w/w) allowed to obtain the best performance throughout the experimental set. Finally, in vitro biocompatibility was carried out by MTT assay and High Content Analysis for selected dry powder formulations and starting materials. The assays demonstrated the same outcome by confirming the HA biocompatibility and by producing the same rank of toxicity for the surfactants. The general conclusion of the project is that formulation containing HA and stearyl alcohol represents the best performing formulation.

O material Y2O3:Eu3+ vem sendo usado comercialmente como luminóforo vermelho desde da década de 1960, em uma grande variedade de aplicações devido ao seu elevado rendimento quântico (próximo de 100 %), elevada pureza de cor e boa estabilidade. Portanto, este trabalho propõe um novo método de síntese baseado nos complexos benzenotricarboxilatos (BTC) de terras raras trivalentes (RE3+) dopados com íons Eu3+. O objetivo principal é produzir materiais luminescente RE2O3:Eu3+ a temperatura mais baixa (500 °C) e em escala nanométrica. Os complexos precursores [RE(BTC):Eu3+] e [RE(TLA)·n(H2O):Eu3+], onde RE3+: Y, Gd e Lu; BTC: ácido trimésico (TMA) e ácido trimelítico (TLA) foram calcinados em diferentes temperaturas de 500 a 1000 °C, a fim de obter os materiais luminescentes RE2O3:Eu3+. Os complexos foram caracterizados por análise elementar de carbono e hidrogênio, analise térmica (TG), espectroscopia de absorção no infravermelho (FTIR), difração de raios-X - método do pó (XPD) e microscopia eletrônica de varredura (SEM). Todos os complexos são cristalinos e termo estáveis até 460 °C. Dados de fosforescência dos complexos de Y, Gd e Lu mostram que o nível T1 do aníon BTC3- tem energia acima do nível emissor 5D0 do íon Eu3+, indicando que os ligantes podem atuar como sensibilizadores de energia intramolecular. O estudo das propriedades fotoluminescentes dos complexos dopados foi baseado nos espectros de excitação e emissão e curvas de decaimento de luminescência. Ademais, foram determinados os parâmetros de intensidades experimentais (Ωλ), tempos de vida (τ), taxas de decaimentos radiativo (Arad) e não-radiativo (Anrad). Os materiais luminescentes RE2O3:Eu3+ foram sintetizados de forma bem sucedida por meio do método benzenotricarboxilatos calcinados a 500, 600, 700, 800, 900 e 1000 °C, apresentando alta homogeneidade química e controle de tamanho de cristalito. Os nanomateriais foram caracterizados pelas técnicas de FTIR, XPD SEM e TEM revelando a obtenção dos materiais C-RE2O3:Eu3+ mesmo a 500 °C. Os dados de XPD dos materiais confirmaram um aumento do tamanho dos cristalitos de 5 até 52 nm (equação de Scherrer) de em função da temperatura de calcinação de 500 a 1000 °C, respectivamente, corroborados pelas técnicas de SEM e TEM. Os espectros de emissão de RE2O3:Eu3+ mostram uma banda larga atribuída a transição interconfiguracional de transferência de carga ligante-metal (LMCT) em 260 nm, i.e. O2-(2p)→Eu3+(4f6). Além disso, foram observadas linhas finas de absorção devido as transições intraconfiguracionais 4f do íon európio (7F0,1𔾹LJ; J: 0, 1, 2, 3 e 4), como esperado. As propriedades fotoluminescentes dos luminóforos foram baseadas nos espectros (excitação e emissão) e curvas de decaimento luminescente. Os parâmetros de intensidade experimental, tempos de vida, assim como as taxas de decaimentos radiativos e não radiativos foram calculados. As propriedades fotônicas dos nanomateriais são consistentes com o sítio de baixa simetria C2 ocupado pelo íon Eu3+ no C-RE2O3:Eu3+, produzindo emissão vermelha dominada pela transição hipersensível 5D0𔾻F2 do íon Eu3+ no sitio C2, ao invés do sítio centrossimétrico S6. Além disso, os nanomateriais Y2O3:Eu3+ exibem características espectroscópicas semelhantes e elevados valores de eficiência quântica (η~91 %), compatível com os luminóforos comerciais disponíveis no mercado. Este novo método pode ser utilizado para o desenvolvimento de novos nanomateriais contendo íons terras raras, assim como outros íons metálicos.

Trihalomethanes are organic compounds formed in drinking water distribution systems as a result of disinfection. This capstone project researched and evaluated the statistical correlation of trihalomethanes in finished drinking water and total organic carbon in source water using data generated by Denver area utilities. Results of the study conclude that some drinking water supply systems show a slight correlation between source water total organic carbon levels and trihalomethane levels in finished water. Results of the study also verify the assertion that changes to treatment for the reduction of trihalomethanes, for the protection of human health under the Safe Drinking Water Act should be determined by each utility, using information from gathered data, seasonal trends, and small scale batch testing.

Results show that it is possible to activate a low softening point isotropic petroleum pitch, without intermediate pre-treatments, by chemical activation with KOH. The chemical activation is carried out by direct heat treatment of a mixture of the isotropic pitch and KOH. It produces activated carbons (ACs) with micropore volumes as high as 1.12 cm3/g, and BET surface areas around 3000 m2/g. The activating agent/precursor ratios studied (from 1/1 to 4/1; wt./wt.) show, as expected, that increasing the ratio enhances the adsorption characteristics of the resulting AC.

Palladium, platinum, and ruthenium supported on activated carbon were used as catalysts for the selective hydrogenation of 1-heptyne, a terminal alkyne. All catalysts were characterized by temperature programmed reduction, X-ray diffraction, transmission electron microscopy, and X-ray photoelectron spectroscopy. TPR and XPS suggest that the metal in all catalysts is reduced after the pretreatment with H2 at 673 K. The TPR trace of the PdNRX catalyst shows that the support surface groups are greatly modified as a consequence of the use of HNO3 during the catalyst preparation. During the hydrogenation of 1-heptyne, both palladium catalysts were more active and selective than the platinum and ruthenium catalysts. The activity order of the catalysts is as follows: PdClRX > PdNRX > PtClRX ≫ RuClRX. This superior performance of PdClRX was attributed in part to the total occupancy of the d electronic levels of the Pd metal that is supposed to promote the rupture of the H2 bond during the hydrogenation reaction. The activity differences between PdClRX and PdNRX catalysts could be attributed to a better accessibility of the substrate to the active sites, as a consequence of steric and electronic effects of the superficial support groups. The order for the selectivity to 1-heptene is as follows: PdClRX = PdNRX > RuClRX > PtClRX, and it can be mainly attributed to thermodynamic effects.

Advanced porous materials with tailored porosity (extremely high development of microporosity together with a narrow micropore size distribution (MPSD)) are required in energy and environmental related applications. Lignocellulosic biomass derived HTC carbons are good precursors for the synthesis of activated carbons (ACs) via KOH chemical activation. However, more research is needed in order to tailor the microporosity for those specific applications. In the present work, the influence of the precursor and HTC temperature on the porous properties of the resulting ACs is analyzed, remarking that, regardless of the precursor, highly microporous ACs could be generated. The HTC temperature was found to be an extremely influential parameter affecting the porosity development and the MPSD of the ACs. Tuning of the MPSD of the ACs was achieved by modification of the HTC temperature. Promising preliminary results in gas storage (i.e. CO2 capture and high pressure CH4 storage) were obtained with these materials, showing the effectiveness of this synthesis strategy in converting a low value lignocellulosic biomass into a functional carbon material with high performance in gas storage applications.

This work reports the synthesis of nanoTiC–graphite composites using mesophase pitch containing titanium as TiC or TiO2 nanoparticles. NanoTiC–graphite composites have been prepared using Ti-doped self-sintering mesophase powders as starting materials without using any binders or a metal carbide-carbon mixing stage. The effect of manufacture variables on the graphite compacts properties was studied. Graphites were characterised using XRD and Raman spectroscopy, SEM and TEM, as well as by their mechanical, electrical and thermal properties. The presence of TiC promotes graphitisation producing materials with larger crystal sizes. The kind of titanium source and mesophase content of the starting pitch affects to the final properties. Mesophase pitch with higher amount of mesophase content produces graphites with higher degree of graphitisation. The incorporation of TiC nanoparticles to the graphites composites improved thermal conductivity more than four times, and mechanical properties are not significantly modified by the presence of TiC.

Supported metals are traditionally prepared by impregnating a support material with the metal precursor solution, followed by reduction in hydrogen at elevated temperatures. In this study, a polymeric support has been considered. Polypyrrole (PPy) has been chemically synthesized using FeCl3 as a doping agent, and it has been impregnated with a H2PtCl6 solution to prepare a catalyst precursor. The restricted thermal stability of polypyrrole does not allow using the traditional reduction in hydrogen at elevated temperature, and chemical reduction under mild conditions using sodium borohydride implies environmental concerns. Therefore, cold RF plasma has been considered an environmentally friendly alternative. Ar plasma leads to a more effective reduction of platinum ions in the chloroplatinic complex anchored onto the polypyrrole chain after impregnation than reduction with sodium borohydride, as has been evidenced by XPS. The increase of RF power enhanced the effectiveness of the Ar plasma treatment. A homogeneous distribution of platinum nanoparticles has been observed by TEM after the reduction treatment with plasma. The Pt/polypyrrol catalyst reduced by Ar plasma at 200 watts effectively catalyzed the aqueous reduction of nitrates with H2 to yield N2, with a very low selectivity to undesired nitrites and ammonium by-products.

The effect of a severe steaming treatment on the physicochemical properties and catalytic performance of H-SAPO-34 molecular sieves during the methanol-to-hydrocarbons (MTH) reaction has been investigated with a combination of scanning transmission X-ray microscopy (STXM), catalytic testing, and bulk characterization techniques, including ammonia temperature programmed desorption and 27Al and 29Si magic angle spinning nuclear magnetic resonance. For this purpose, two samples, namely a calcined and a steamed H-SAPO-34 catalyst powder, have been compared. It has been found that calcined H-SAPO-34 displays a high selectivity towards light olefins, yet shows a poor stability as compared to a zeolite H-ZSM-5 catalyst. Moreover, in situ STXM at the carbon K-edge during the MTH reaction allows construction of nanoscale chemical maps of the hydrocarbon species formed within the H-SAPO-34 aggregates as a function of reaction time and steam post-treatment. It was found that there is an initial preferential formation of coke precursor species within the core of the H-SAPO-34 aggregates. For longer times on stream the formation of the coke precursor species is extended to the outer regions, progressively filling the entire H-SAPO-34 catalyst particle. In contrast, the hydrothermally treated H-SAPO-34 showed similar reaction selectivity, but decreased activity and catalyst stability with respect to its calcined counterpart. These variations in MTH performance are related to a faster and more homogeneous formation of coke precursor species filling up the entire steamed H-SAPO-34 catalyst particle. Finally, the chemical imaging capabilities of the STXM method at the Al and Si K-edge are illustrated by visualizing the silicon islands at the nanoscale before and after steaming H-SAPO-34.
